A rigorous theoretical investigation has been performed on the propagation of cylindrical and spherical Gardner solitons (GSs) associated with dust-ion-acoustic (DIA) waves in a dusty plasma consisting of inertial ions, negatively charged immobile dust, and two populations of kappa distributed electrons having two distinct temperatures. The well-known reductive perturbation method has been used to derive the modified Gardner (mG) equation. The basic features (amplitude, width, polarity, etc.) of nonplanar DIA modified Gardner solitons (mGSs) have been thoroughly examined by the numerical analysis of the mG equation. It has been found that the characteristics of the nonplanar DIA mGSs significantly differ from those of planar ones. It has been also observed that kappa distributed electrons with two distinct temperatures significantly modify the basic properties of the DIA solitary waves and that the plasma system under consideration supports both compressive and rarefactive DIA mGSs. The present investigation should play an important role for understanding localized electrostatic disturbances in space and laboratory dusty plasmas where stationary negatively charged dust, inertial ions, and superthermal electrons with two distinct temperatures are omnipresent ingredients.  相似文献

Two distinct classes of dust ion acoustic (DIA) solitary waves based on relativistic ions and electrons, dust charge Z d and ion-to-dust mass ratio Q’ = m i /m d are established in this model of multicomponent plasmas. At the increase of mass ratio Q’ due to increase of relativistic ion mass and accumulation of more negative dust charges into the plasma causing decrease of dust mass, relativistic DIA solitons of negative potentials are abundantly observed. Of course, relativistic compressive DIA solitons are also found to exist simultaneously. Further, the decrease of temperature inherent in the speed of light c causes the nonlinear term to be more active that increases the amplitude of the rarefactive solitons and dampens the growth of compressive solitons for relatively low and high mass ratio Q’, respectively. The impact of higher initial streaming of the massive ions is observed to identify the point of maximum dust density N d to yield rarefactive relativistic solitons of maximum amplitude.  相似文献

Mucosal crude microsomes, prepared from proximal rat small intestine, exhibited significant Mg-dependent, Zn-ATPase activity; V max = 23 μmoles Pi/mg protein/hr, K m = 160 nm, and Hill Coefficient, n= 1.5. Partial purification (∼10-fold) was achieved by detergent extraction, and centrifugation through 250 mm sucrose: V max = 268 units, K m = 1 nm, and n= 6. In partially purified preparations, the assay was linear with time to 60 min, and with protein concentration to 1 μg/300 μl. Activities at pH 8 and 8.5 were higher than at pH 7.2. The ATP K m was 0.7 mm, with an optimal ATP/Mg ratio of ∼2. Ca elicited ATPase activity but did not augment the Zn-dependent activity. In partially purified preparations, the homologous salts of Co, Cd, Cu, and Mn exhibited no detectable activity. Vanadate inhibition studies yielded two component kinetics with a K i of 12 μm for the first component, and 96 μm for the second component, in partially purified preparations. Tissue distribution analyses revealed gradients of activity. In the proximal half of the small intestine, Mg/Zn activity increased progressively from crypt to villus tip. Two approaches to determine the fraction (μ) of mitochondrial respiration sustained during illumination by measuring CO2 gas exchange are compared. In single leaves, the respiration rate in the light (`day respiration' rate Rd) is determined as the ordinate of the intersection point of A–ci curves at various photon flux densities and compared with the CO2 evolution rate in darkness (`night respiration' rate Rn). Alternatively, using leaves with varying values of CO2 compensation concentration (Γ), intracellular resistance (ri) and Rn, an average number for μ can be derived from the linear regression between Γ and the product riċRn. Both methods also result in a number c* for that intercellular CO2 concentration at which net CO2 uptake rate is equal to –Rd. c* is an approximate value of the photocompensation point Γ* (Γ in the absence of mitochondrial respiration), which is related to the CO2/O2 specificity factor of Rubisco Sc/o. The presuppositions and limitations for application of both approaches are discussed. In leaves of Nicotiana tabacum, at 22 °C, single leaf measurements resulted in mean values of μ = 0.71 and c* = 34 μmol mol−1. At the photosynthetically active photon flux density of 960 μmol quanta m−2 s−1, nearly the same numbers were derived from the linear relationship between Γ and riċRn. c* and Rd determined by single leaf measurements varied between 31 and 41 μmol mol−1 and between 0.37 and 1.22 μmol m−2 s−1, respectively. A highly significant negative correlation between c* and Rd was found. From the regression equation we obtained estimates for Γ* (39 μmol mol−1), Sc/o (96.5 mol mol−1) and the mesophyll CO2 transfer resistance (7.0 mol−1 m2 s). d-Aspartate (d-Asp) uptake by suspensions of cerebral rat brain astrocytes (RBA) maintained in long-term culture was studied as a means of characterizing function and regulation of Glutamate/Aspartate (Glu/Asp) transporter isoforms in the cells. d-Asp influx is Na+-dependent with K m = 5 μm and V max= 0.7 nmoles · min−1· mg protein−1. Influx is sigmoidal as f[Na+] with Na+ K m ∼ 12 μm and Hill coefficient of 1.9. The cells establish steady-state d-Asp gradients >3,000-fold. Phorbol ester (PMA) enhances uptake, and gradients near 6,000-fold are achieved due to a 2-fold increase in V max, with no change in K m . At initial [d-Asp] = 10 μm, RBA take up more than 90% of total d-Asp, and extracellular levels are reduced to levels below 1 μm. Ionophores that dissipate the ΔμNa+ inhibit gradient formation. Genistein (GEN, 100 μm), a PTK inhibitor, causes a 40% decrease in d-Asp. Inactive analogs of PMA (4α-PMA) and GEN (daidzein) have no detectable effect, although the stimulatory PMA response still occurs when GEN is present. Further specificity of action is indicated by the fact that PMA has no effect on Na+-coupled ALA uptake, but GEN is stimulatory. d-Asp uptake is strongly inhibited by serine-O-sulfate (S-O-S), threohydroxy-aspartate (THA), l-Asp, and l-Glu, but not by d-Glu, kainic acid (KA), or dihydrokainate (DHK), an inhibition pattern characteristic of GLAST and EAAC1 transporter isoforms. mRNA for both isoforms was detected by RT-PCR, and Western blotting with appropriate antibodies shows that both proteins are expressed in these cells. CDP840     
We present the in vitro characterization of a novel phosphodiesterase type 4 inhibitor, CDP840 (R-[+]-4-[2-{3-cyclopentyloxy-4-methoxyphenyl}-2-phenylethyl]pyridine), which has shown efficacy in a phase II allergen challenge study in asthmatics without adverse effects. CDP840 potently inhibits PDE-4 isoenzymes (IC50 2–30 nM) without any effect on PDE-1, 2, 3, 5, and 7 (IC50>100 μM). It exhibited no significant selectivity in inhibiting human recombinant isoenzymes PDE-4A, B, C or D and was equally active against the isoenzymes lacking UCR1 (PDE-4B2 and PDE-4D2). In contrast to rolipram, CDP840 acted as a simple competitive inhibitor of all PDE-4 isoenzymes. Studies with rolipram indicated a heterogeneity within all the preparations of PDE-4 isoenzymes, indicative of rolipram inhibiting the catalytic activity of PDE-4 with both a low or high affinity. These observations were confirmed by the use of a PDE-4A variant, PDE-4A330–886, which rolipram inhibited with low affinity (IC50=1022 nM). CDP840 in contrast inhibited this PDE-4A variant with similar potency (IC50=3.9 nM), which was in good agreement with theK d of 4.8 nM obtained from [3H]-CDP840 binding studies. Both CDP840 and rolipram inhibited the high-affinity binding of [3H]-rolipram binding to PDE-4A, B, C and D with similarK d app (7–19 nM and 3–5 nM, respectively). Thus, the activity of CDP840 at the [3H]-rolipram binding site was in agreement with the inhibitor’s activity at the catalytic site. However, rolipram was ∼100-fold more potent than CDP840 at inhibiting the binding of [3H]-rolipram to mouse brain in vivo. These data clearly demonstrate that CDP840 is a potent selective inhibitor of all PDE-4 isoenzymes. In contrast to rolipram, CDP840 was well-tolerated in humans. This difference, however, cannot at present be attributed to either isoenzyme selectivity or lack of activity in vitro at the high-affinity rolipram binding site (Sr).  相似文献

Studies on biogeochemical cycling of carbon in the Chilka Lake, Asia’s largest brackish lagoon on the east coast of India, revealed, for the first time, strong seasonal and spatial variability associated with salinity distribution. The lake was studied twice during May 2005 (premonsoon) and August 2005 (monsoon). It exchanges waters with the sea (Bay of Bengal) and several rivers open into the lake. The lake showed contrasting levels of dissolved inorganic carbon (DIC) and organic carbon (DOC) in different seasons; DIC was higher by ∼22% and DOC was lower by ∼36% in premonsoon than in monsoon due to seasonal variations in their supply from rivers and in situ production/mineralisation. The DIC/DOC ratios in the lake during monsoon were influenced by physical mixing of end member water masses and by intense respiration of organic carbon. A strong relationship between excess DIC and apparent oxygen utilisation showed significant control of biological processes over CO2 production in the lake. Surface partial pressure of CO2 (pCO2), calculated using pH–DIC couple according to Cai and Wang (Limnol and Oceanogr 43:657–668, 1998), exhibited discernable gradients during monsoon through northern (1,033–6,522 μatm), central (391–2,573 μatm) and southern (102–718 μatm) lake. The distribution pattern of pCO2 in the lake seems to be governed by pCO2 levels in rivers and their discharge rates, which were several folds higher during monsoon than premonsoon. The net CO2 efflux, based on gas transfer velocity parameterisation of Borges et al. (Limnol and Oceanogr 49(5):1630–1641, 2004), from entire lake during monsoon (141 mmolC m−2 d−1 equivalent to 2.64 GgC d−1 at basin scale) was higher by 44 times than during premonsoon (9.8 mmolC m−2 d−1 ≈ 0.06 GgC d−1). 15% of CO2 efflux from lake in monsoon was contributed by its supply from rivers and the rest was contributed by in situ heterotrophic activity. Based on oxygen and total carbon mass balance, net ecosystem production (NEP) of lake (−308 mmolC m−2 d−1 ≈ −3.77 GgC d−1) was found to be almost in consistent with the total riverine organic carbon trapped in the lake (229 mmolC m−2 d−1 ≈ 2.80 GgC d−1) suggesting that the strong heterotrophy in the lake is mainly responsible for elevated fluxes of CO2 during monsoon. Further, the pelagic net community production represented 92% of NEP and benthic compartment plays only a minor role. This suggests that Chilka lake is an important region in biological transformation of organic carbon to inorganic carbon and its export to the atmosphere.  相似文献

Resveratrol (RES), a component of red wine, possesses anti-inflammatory properties. The studies described in the present work were aimed at evaluating the potential for RES and related stilbene analogs (piceatannol, PIC; pterostilbene, TPS; trans-stilbene, TS; and trans-stilbene oxide, TSO) to exhibit toxicity towards RAW 264.7 mouse macrophages. The effect of TS, TSO, RES and TPS on RAW 264.7 macrophage viability was determined by two standard methods: (a) the MTT assay and (b) the trypan blue dye exclusion test. Whereas macrophages were more sensitive to PIC (LC50 trypan ∼ 1.3 μM) and to TPS (LC50 trypan ∼ 4.0 μM and LC50 MTT ∼ 8.3 μM) than to RES (LC50 trypan ∼ 8.9 μM and LC50 MTT ∼ 29.0 μM), they were relatively resistant to TSO (LC50 trypan ∼ 61.0 μM and LC50 MTT > 100 μM) and to TS (LC50 trypan ≥ 5.0 μM and LC50 MTT ≥ 5.0 μM). The ability of selected stilbenes (RES, TPS and PIC) to exhibit growth inhibitory effects was also examined. Although RES and TPS were observed to inhibit cell proliferation in macrophages (IC50 ≤ 25 μM), these cells were resistant to growth inhibition by PIC (IC50 ≥ 50 μM). The data obtained in the present analysis demonstrate that substituted stilbene compounds such as RES have the capacity to exhibit cytotoxic and anti-proliferative activities in macrophages.  相似文献

The effects of nonextensivity and nonthermality of ions of two distinct temperatures on dustacoustic Gardner solitons (DAGSs) in an unmagnetized dusty plasma system are investigated theoretically. The constituents of the dusty plasma under consideration are negatively charged mobile dust fluid, Boltzmann-distributed electrons, and ions of two distinct temperatures following nonextensive (q) and nonthermal distributions, respectively. The Korteweg-de Vries (KdV), modified KdV, and Gardner equations are derived by using the reductive perturbation technique, and thereby their characteristic features are compared. It is observed that both the nonextensive and nonthermal ions significantly modify the basic properties and polarities of dust-acoustic solitary waves. The present investigation may be of relevance to space and laboratory dusty plasma systems.  相似文献

To determine the photosynthetic characteristics of C3 plants and their sensitivity to CO2 at different altitudes on the Tibetan Plateau, hulless barley (Hordeum vulgare L. ssp. vulgare) was grown at altitudes of 4,333 m and 3,688 m. Using gas-exchange measurements, photosynthetic parameters were simulated, including the maximum net photosynthesis (P max) and the apparent quantum efficiency (α). Plants growing at higher altitude had higher net photosynthetic rates (P N), photosynthesis parameters (P max and α) and sensitivities to CO2 enhancement than plants growing at lower altitude on the Tibetan Plateau. The enhancements of P N, P max, and α for plants growing at higher altitude, corresponding with 10 μmol(CO2) mol−1 increments, were approximately 0.20∼0.45%, 0.05∼0.20% and 0.12∼0.36% greater, respectively, than for plants growing at lower altitude, respectively, where CO2 levels rose from 10 to 170 μmol(CO2) mol−1. Therefore, on the Tibetan Plateau, the changes in the photosynthetic capacities and the photosynthetic sensitivities to CO2 observed in the C3 plants grown above 3,688 m are likely to increase with altitude despite the decreasing CO2 partial pressure.  相似文献

Smooth muscle cells isolated from the secondary and tertiary branches of the rabbit mesenteric artery contain large Ca2+-dependent channels. In excised patches with symmetrical (140 mm) K+ solutions, these channels had an average slope conductance of 235 ± 3 pS, and reversed in direction at −6.1 ± 0.4 mV. The channel showed K+ selectivity and its open probability (P o ) was voltage-dependent. Iberiotoxin (50 nm) reversibly decreased P o , whereas tetraethylammonium (TEA, at 1 mm) reduced the unitary current amplitude. Apamin (200 nm) had no effect. The channel displayed sublevels around 1/3 and 1/2 of the mainstate level. The effect of [Ca2+] on P o was studied and data fitted to Boltzmann relationships. In 0.1, 0.3, 1.0 and 10 μm Ca2+, V 1/2 was 77.1 ± 5.3 (n= 18), 71.2 ± 4.8 (n= 16), 47.3 ± 10.1 (n= 11) and −14.9 ± 10.1 mV (n= 6), respectively. Values of k obtained in 1 and 10 μm [Ca2+] were significantly larger than that observed in 0.1 μm [Ca2+]. With 30 μm NS 1619 (a BKCa channel activator), V 1/2 values were shifted by 39 mV to the left (hyperpolarizing direction) and k values were not affected. TEA applied intracellularly, reduced the unitary current amplitude with a K d of 59 mm. In summary, BKCa channels show a particularly weak sensitivity to intracellular TEA and they also display large variation in V 1/2 and k. These findings suggest the possibility that different types (isoforms) of BKCa channels may exist in this vascular tissue. Received: 22 December 1997/Revised: 27 March 1998  相似文献

Clostridium thermoaceticum were determined by equilibrium dialysis. CoA bound to as-isolated native α 2 β 2 enzyme with K D = 10 ± 8 μM and n = 0.2 ± 0.1 moles per αβ dimer, where K D is the thermodynamic dissociation constant and n is the number of CoAs bound per αβ dimer of the enzyme. The enzyme is heterogeneous; for example, only  ∼ 30% of α subunits contain A-clusters with labile Ni ions (the remainder have nonlabile Ni ions and are nonfunctional). The observed n value suggests that CoA binds only to αβ units with Ni-labile A-clusters. The CoA binding properties of enzyme lacking labile Ni was essentially the same, indicating that CoA does not bind directly to the Ni of the A-cluster. This was further evidenced by the observation that bound CoA did not inhibit removal of the labile Ni by 1,10-phenanthroline. CoA did not bind CO-reduced enzyme, and the EPR signal exhibited by the one-electron reduced and CO-bound form of the A-cluster was unaffected by the presence of up to 200 μM CoA. In contrast, CoA did bind Ti(III)-citrate-reduced enzyme (K D = 36 ± 16 μM, n = 0.16 ± 0.08). Implications of these results for the mechanism of catalysis are discussed. The mobilization of dust (i.e., detachment and removal of dust grains from a substrate) and the exfoliation of a film of erosion products in tokamaks have been studied theoretically. The following mechanisms of dust mobilization have been taken into account: (i) sharp heating (thermal shock) as a result of, e.g., plasma disruption and edge instabilities; (ii) substrate vibrations; and (iii) gas and plasma flow (wind) action. The most effective mobilization takes place under the action of sharp heating. Power fluxes that are characteristic of edge instabilities can mobilize dust grains with dimensions within or even greater than 0.1–1 μm. The velocities of detached grains reach ν ∼ 100 m/s for heavy grains and up to ν ∼ 300 m/s for the light ones. Conditions favoring the exfoliation of a film of erosion products are determined. It is shown that exfoliation under the action of edge instabilities can take place at a film thickness of h > 1 μm. Under the action of thermal-shock-induced stresses, the exfoliated film flakes with a size ranging from fractions of a millimeter to several centimeters break into pieces.  相似文献

 DNA binding by trans-[(H2O)(Pyr)(NH3)4RuII]2+ (Pyr=py, 3-phpy, 4-phpy, 3-bnpy, 4-bnpy) is highly selective for G7 with K G=1.1×104 to 2.8×104, with the more hydrophobic Pyr ligands exhibiting slightly higher binding. A strong dependence on ionic strength indicates that ion-pairing with DNA occurs prior to binding. At μ=0.05, d[RuII-DNA]/dt=k[RuII][DNA], where k=0.17–0.21 M–1 s–1 with the various Pyr ligands. The air oxidation of [(py)(NH3)4RuII] n -DNA to [(py)(NH3)4RuIII] n -DNA at pH 6 occurs with a pseudo-first-order rate constant of k obs=5.6×10–4 s–1 at μ=0.1, T=25  °C. Strand cleavage of plasmid DNA appears to occur by both Fenton/Haber-Weiss chemistry and by base-catalyzed routes, some of which are independent of oxygen. Base-catalyzed cleavage is more efficient than O2 activation at neutral pH and involves the disproportionation of covalently bound RuIII and, in the presence of O2, Ru-facilitated autoxidation to 8-oxoguanine. Disproportionation of [py(NH3)4RuIII] n -DNA occurs according to the rate law: d[RuII–GDNA]/dt=k 0[RuIII–GDNA]+k 1[RuIII–GDNA][OH], where k 0=5.4×10–4 s–1 and k 1=8.8 M–1 s–1 at 25  °C, μ=0.1. The appearance of [(Gua)(py)(NH3)4RuIII] under argon, which occurs according to the rate law: d[RuIII–G]/dt=k 0[RuIII–GDNA]+k 1[OH][RuIII–GDNA] (k 0=5.74×10–5 s–1, k 1=1.93×10–2 M–1 s–1 at T=25  °C, μ=0.1), is consistent with lysis of the N-glycosidic bond by RuIV-induced general acid hydrolysis. In air, the ratio of [Ru-8-OG]/[Ru-G] and their net rates of appearance are 1.7 at pH 11, 25  °C. Small amounts of phosphate glycolate indicate a minor oxidative pathway involving C4′ of the sugar. Summary The growth of GH4C1, GH3, GH1, and GH3C15 rat pituitary tumor cell lines was studied in a serum-free medium (designated TRM-1) formulated with 1∶1 (vol/vol) mixture of Ham's F12 nutrient mixture and Dulbecco's modified Eagle's medium (F12-DME) containing 15 mM 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES), 50 μg/ml gentamicin supplemented with 10 μg/ml bovine insulin, 10 μg/ml human transferrin (Tf), 10 ng/ml selenous acid, 10 nM 3,5,3′-triiodothyronine (T3), 50 μM ethanolamine (Etn), and 500 μg/ml bovine serum albumin. Of the lines evaluated, only the GH1 failed to grow in TRM-1. Passage of the GH4C1 and GH3 lines from serum-containing medium into TRM-1 caused an initial selection resulting in cells that grew progressively at higher rates and finally were maintained indefinitely in TRM-1. These populations showed a requirement for supraphysiologic concentrations of T3 (1.0 to 10 nM). After adaptation of the GH4C1 line in TRM-1 for ≤20 generations, removal of components gave a less complex mixture containing 15 mM HEPES, 50 μ/ml gentamicin, 10 μg/ml Tf, 10 nM T3, and 50 μM Etn (designated TRM-2) that supported serial passage of the cells. Under these conditions, thyroid hormone dependence was lost progressively. When T3 was removed from TRM-2 adapted cells, a third population was selected that no longer required thyroid hormones and was only slightly stimulated by T3. These studies demonstrated that the combination of serum-containing and serum-free conditions can be used to select pituitary cell populations that a) required both serum-factor(s) and T3 for optimum growth, b) required supraphysiologic concentrations of T3 without serum proteins other than Tf and albumin, and c) were completely autonomous in that they proliferated in medium supplemented only with Tf and nutrients without necessity of other serum factor(s) or T3. The time evolution of the matter parameters and current distribution in the discharge channel formed during a nanosecond explosion of a 25-μm-diameter 12-mm-long aluminum wire was studied in a series of experiments with the following parameters: the discharge voltage was U 0 = 20 kV, the current amplitude was I max ∼ 8 kA, and the current rise rate was dI/dt ∼ 40 A/ns. Optical shadow and schlieren images of the discharge channel were obtained using the second harmonic of a YAG: Nd+3 laser, and UV images of the discharge channel self-radiation were recorded using a four-frame camera with a microchannel plate. The process of aluminum wire explosion was simulated numerically (including simulations performed from the “cold start”). The numerical results were compared with the experimental data.  相似文献

Summary We have used a combination of chemical labeling and detergent fractionation techniques to locate the divalent cation binding sites on the chloroplast membrane. We determined the Ca2+-binding properties of Triton X-100 subchloroplast particles. Photosystem II (TSFII) particles showed one binding site withn=8.4 moles-mg chl–1 andk d =20 m. Photosystem I (TSFI) particles contained two binding sites. The first had ann=1.5 moles-mg chl–1 andk d =4 m. The second had ann=9.6 moles-mg chl–1 andk d =160 m. We have previously shown (Prochaska & Gross,Biochim. Biophys. Acta 376:126, 1975) that the divalent cation binding sites could be blocked using a water-soluble carbodiimide plus a nucleophile. Chlorophylla fluorescence and lightscattering changes were affected at the same carbodiimide concentrations emphasizing the relationship between these processes. The carbodiimide-sensitive sites were found to be located on the Photosystem II particles. A direct correlation between the inhibition of calcium binding and the carbodiimide-mediated incorporation of a (14C)-nucleophile was observed upon varying such parameters as carbodiimide concentration, nucleophile concentration, pH, and time of reaction. The presence of CaCl2 during the carbodiimide plus nucleophile modification procedure decreased the incorporation of (14C)-nucleophile, emphasizing the competition of the CaCl2 and the modification reagents for some of the same sites. Sodium dodecylsulfate gel electrophoresis of chlorophyll protein aggregates suggested that the site of competition of the calcium chloride and the modification reagents was the light-harvesting chlorophylla/b protein.  相似文献

Apoptosis of thyroid follicular cells is induced by high doses of iodide, epidermal growth factor (EGF), transforming growth factor-β (TGF-β), as well as H2O2 and might be attenuated by antioxidants. Therefore, we examined the apoptotic index induced by these substances in selenium-treated vs untreated human thyroid follicular cells. Reconstituted human thyroid follicles were incubated with sodium selenite (10 or 100 nM) for 72 h; controls received none. The follicles were then distributed to 24-well plates and incubated with potassium iodide (5, 10, or 20 nM), EGF (5 ng/mL), TGF-β (5 ng/mL), or H2O2 (100 μM). Apoptosis was determined by a mitochondrial potential assay and the number of apoptotic cells counted by two independent, experienced technicians and the glutathione peroxidase (GPx) activity was determined. A significant increase of apoptic cells was obtained in control thyroid follicles treated with iodine (5, 10, or 20 μM), thyroid-stimulating hormone (TSH) 1, or 10 mU/mL in combination with 5 and 10 μM iodine, EGF (5 ng/mL) and TGF-β (5 ng/mL), or H2O2 (100 μM) (p<0.001). In contrast, in thyroid follicles preincubated with 10 or 100 nM sodium selenite, the apoptototic index was identical to the basal rate. In H2O2-treated follicles, the apoptotic index was still significantly elevated but 50% lower compared to control cells. The GPx activity increased from 1.4±0.2 to 2.25±0.4 mU/μg DNA with 10 nM selenite and 2.6+0.4 mU/μg DNA with 100 nM selenite. Sodium selenite might increase the antioxidative potential in human thyroid follicles in vitro and therefore diminish the apoptosis induced by TGF-β, EGF, iodide, and even H2O2  相似文献

Summary Total retro-inverso (TRI) analogues of bradykinin (BK), the B2a -selective kinin antagonistd-Arg0[Hyp3,d-Phe7,Leu8]BK, angiotensin II (AT II) and the AT II antagonist Saralasin ([Sar1, Val5, Ala8]AT II) were prepared by conventional solid-phase synthesis. Molecular recognition of TRI peptidomimetics by G-protein-coupled receptors was studied by competitive radioligand displacement experiments. TRI analogues ofd-Arg0[Hyp3,d-Phe7,Leu8]BK specifically bound to the kidney medulla B2a bradykinin receptor with affinities (K d ) ranging from 64 μM to 4 μM. Conversely, TRI analogues of BK, AT II and Saralasin did not bind to either the B2a bradykinin receptor or the rat AT1a AT II receptor, respectively. These studies indicate that the TRI strategy is more compatible with the synthesis of antagonists than ‘agonists’. Three TRI peptidomimetics ofd-Arg0[Hyp3,d-Phe7,Leu8]BK were weak inhibitors of angiotensin converting enzyme. All other TRI peptidomimetics had no effect upon ACE activity. These data endorse the utility of the TRI strategy for the synthesis of protease-resistant antagonists of peptide hormones and neuropeptides.  相似文献
