The pollen of Ginkgo biloba is one of the allergens that cause pollen allergy symptoms. The plant complex type N-glycans bearing beta1-2 xylose and/or alpha1-3 fucose residue(s) linked to glycoallergens have been considered to be critical epitopes in various immune reactions. In this report, the structures of N-glycans of total glycoproteins prepared from Ginkgo biloba pollens were analyzed to confirm whether such plant complex type N-glycans occur in the pollen glycoproteins. The glycoproteins were extracted by SDS-Tris buffer. N-Glycans liberated from the pollen glycoprotein mixture by hydrazinolysis were labeled with 2-aminopyridine and the resulting pyridylaminated (PA-)N-glycans were purified by a combination of size-fractionation HPLC and reversed-phase HPLC. The structures of the PA-sugar chains were analyzed by a combination of two-dimensional sugar chain mapping, IS-MS, and MS/MS. The plant complex type structures (GlcNAc2Man3Xyl1Fuc1GlcNAc2 (31%), GlcNAc2Man3Xyl1GlcNAc2 (5%), Man3Xyl1Fuc1GlcNAc2 (13%), GlcNAc1Man3Xyl1Fuc1GlcNAc2 (8%), and GlcNAc1Man3Xyl1GlcNAc2 (17%)) have been found among the N-glycans of the glycoproteins of Ginkgo biloba pollen, which might be candidates for the epitopes involved in Ginkgo pollen allergy. The remaining 26% of the total pollen N-glycans have the typical high-mannose type structures: Man8GlcNAc2 (11%) and Man6GlcNAc2 (15%).  相似文献

In this study, the seasonally averaged intradiurnal patterns of four different pollen types (Fraxinus, Betula, Poaceae and Artemisia) and the role of traffic volume, air pollution and selected weather parameters were investigated. Measurements were carried out with a 7-day recording volumetric spore trap (Hirst type) near a congested city motorway (the A 100) in Berlin, Germany, in 2012, 2013 and partly 2011. Both Poaceae and Artemisia pollen showed distinct patterns which were similar across the years. The main period of grass pollen concentrations in the air was from 8 a.m. to 10 p.m. with peaks about midday or in the afternoon. Mugwort pollen mainly occurred between 6 a.m. and 2 p.m. with a clear maximum from 8 to 10 a.m. With regard to Fraxinus and Betula pollen, the patterns were not as clear and showed differences throughout the years. The intradiurnal patterns of traffic volume and pollen load, mainly of Poaceae in the afternoon and Artemisia in the morning, were partly coincident. The combination of both a high pollen count and air pollution, due to exhaust emissions, represents a special health threat which could result in a double burden for allergy sufferers. In the case of the daily means of Betula and Poaceae, relative humidity had a significantly negative effect on pollen concentrations on the same and/or next day/s, sunshine duration (Poaceae) and air temperature (Artemisia) a positive one.  相似文献

The pollen of oil palm (Elaeis guineensis Jacq.) is a strong allergen and causes severe pollinosis in Malaysia and Singapore. In the previous study (Biosci. Biotechnol. Biochem., 64, 820-827 (2002)), from the oil palm pollens, we purified an antigenic glycoprotein (Ela g Bd 31 K), which is recognized by IgE from palm pollinosis patients. In this report, we describe the structural analysis of sugar chains linked to palm pollen glycoproteins to confirm the ubiquitous occurrence of antigenic N-glycans in the allergenic pollen. N-Glycans liberated from the pollen glycoprotein mixture by hydrazinolysis were labeled with 2-aminopyridine followed by purification with a combination of size-fractionation HPLC and reversed-phase HPLC. The structures of the PA-sugar chains were analyzed by a combination of two-dimensional sugar chain mapping, electrospray ionization mass spectrometry (ESI-MS), and tandem MS analysis, as well as exoglycosidase digestions. The antigenic N-glycan bearing alpha1-3 fucose and/or beta1-2 xylose residues accounts for 36.9% of total N-glycans: GlcNAc2Man3Xyl1Fuc1GlcNAc2 (24.6%), GlcNAc2Man3Xyl1GlcNAc2 (4.4%), Man3Xyl1Fuc1-GlcNAc2 (1.1%), GlcNAc1Man3Xyl1Fuc1GlcNAc2 (5.6%), and GlcNAc1Man3Xyl1GlcNAc2 (1.2%). The remaining 63.1% of the total N-glycans belong to the high-mannose type structure: Man9GlcNAc2 (5.8%), Man8GlcNAc2 (32.1%), Man7GlcNAc2 (19.9%), Man6GlcNAc2 (5.3%).  相似文献

Quantification of airborne pollen is an important tool in scientific research and patient care in allergy. The currently available method relies on microscopic examination of pollen slides, performed by qualified researchers. Although highly reliable, the method is labor intensive and requires extensive training of the researchers involved. In an approach to develop alternative detection methods, we performed a feasibility study on the automated recognition of the allergenic relevant pollen, grass, birch, and mugwort, by utilizing digital image analysis and pattern recognition tools. Of a total of 254 pollen samples (including 79 of grass, 79 of birch and 96 of mugwort), 97.2% were recognized correctly. This encouraging result provides a promising prospect for future developments.  相似文献

The present article deals with the efficacy of seed hairs of poplar trees (Populus spp.) as a potent natural airborne pollen trap. Different species of Populus are commonly found planted along the streets in the cities of North China. The seed hairs and pericarp of poplar trees were collected from the trees and on the ground in Beijing Botanical Garden of Chinese Academy of Sciences and around Miyun Reservoir during May 2005 for pollen analysis. Different pollen spectra are recorded from different samples and are characterised by dominant occurrence of pollen grains of arboreal and anemophilous plants. In addition, pollen grains of non‐arboreal plants including grasses are also found trapped. Among the 46 trapped pollen grains, 26 are known to be allergenic. This study suggests that poplar seed hairs possibly make people feel uncomfortable due to the presence of allergenic pollen trapped in the hairs.  相似文献

A pollen calendar has been constructed for the area of Thessaloniki and relationships between pollen transport and meteorological parameters have been assessed. Daily airborne pollen records were collected over a 15-year period (1987-2001), using a Burkard continuous volumetric pollen trap, located in the centre of the city. Sixteen allergenic pollen types were identified. Simultaneously, daily records of five main meteorological parameters (mean air temperature, relative humidity, rainfall, sunshine, wind speed) were made, and then correlated with fluctuations of the airborne pollen concentrations. For the first time in Greece, a pollen calendar has been constructed for 16 pollen types, from which it appears that 24.9% of the total pollen recorded belong to Cupressaceae, 20.8% to Quercus spp., 13.6% to Urticaceae, 9.1% to Oleaceae, 8.9% to Pinaceae, 6.3% to Poaceae, 5.4% to Platanaceae, 3.0% to Corylus spp., 2.5% to Chenopodiaceae and 1.4% to Populus spp. The percentages of Betula spp., Asteraceae (Artemisia spp. and Ambrosia spp.), Salix spp., Ulmaceae and Alnus spp. were each lower than 1%. A positive correlation between pollen transport and both mean temperature and sunshine was observed, whereas usually no correlation was found between pollen and relative humidity or rainfall. Finally, wind speed was generally found to have a significant positive correlation with the concentrations of 8 pollen types. For the first time in the area of Thessaloniki, and more generally in Greece, 15-year allergenic pollen records have been collected and meteorological parameters have been recorded. The airborne pollen concentration is strongly influenced by mean air temperature and sunshine duration. The highest concentrations of pollen grains are observed during spring (May).  相似文献

Airborne concentrations of pollen from Betula (birch), Poaceae (grasses) and Artemisia (mugwort) are compared during a seven year period (90–96) with respect to both quantitative and seasonal aspects, at three different sampling sites, one in Estonia (Tartu) and two in Sweden (Stockholm and Roma on the island of Gotland). All three taxa occur in the region and are well‐known causes of allergic sensitisation. The annual total and peak values of birch, grass and mugwort pollen were found to be much higher in Tartu than in Stockholm and Roma. Both the birch and the grass pollen seasons ended later in Stockholm than in Roma and Tartu. The mugwort flowering season always began earlier in Stockholm than at the other sites, and more days elapsed between start day and peak day in Stockholm than in Tartu.  相似文献

Knowledge of airborne pollen concentrations and the weather conditions influencing them is important for air quality forecasters, allergists and allergy sufferers. For this reason, a 7-day recording volumetric spore trap of the Hirst design was used for pollen monitoring between January 2006 and December 2007 in Kastamonu, Turkey. A total of 293,427 pollen grains belonging to 51 taxa were recorded during the study period. In the 2?years of study, the period March–August was identified as the main pollination season for Kastamonu. The highest monthly pollen counts were observed in May in both years. Six taxa made up 86.5% of the total amount of pollen recorded in the atmosphere of Kastamonu. These were as follows: Pinaceae (42.9%), Cupressaceae (20.6%), Poaceae (9.7%), Quercus (5.5%) Betula (5.3%) and Carpinus (2.6%). Four of these are considered to be highly allergenic (Betula, Carpinus, Cupressaceae and Poaceae). There were also a greater percentage of highly allergenic taxa found within the city, including Betula pendula that is not part of the local flora. This shows that through urban planting, the public and municipalities can unconsciously create a high risk for allergy sufferers. Daily average pollen counts from the six most frequently recorded pollen types were entered into Spearman’s correlation analysis with meteorological data. Mean daily temperature, relative humidity, daily rainfall and wind speed were found to significantly (p?<?0.05) affect atmospheric pollen concentrations, but the relationships between pollen concentrations and meteorological variables can vary and so there is a need for more local studies of this nature.  相似文献

Summary. Rapid diffusion of allergenic proteins in isotonic media has been demonstrated for different pollen grains. Upon contact with stigmatic secretion or with the mucosa of sensitive individuals, pollen grains absorb water and release soluble low-molecular-weight proteins, these proteins enter in the secretory pathway in order to arrive at the cell surface. In this study we located allergenic proteins in mature and hydrated-activated pollen grains of Parietaria judaica L. (Urticaceae) and studied the diffusion of these proteins during the first 20 min of the hydration and activation processes. A combination of transmission electron microscopy and immunocytochemical methods was used to locate these proteins in mature pollen and in pollen grains after different periods of hydration and activation processes. Activated proteins reacting with antibodies in human serum from allergic patients were found in the cytoplasm, wall, and exudates from the pollen grains. The allergenic component of these pollen grains changes according to the pollen state; the presence of these proteins in the exine, the cytoplasm, and especially in the intine and in the material exuded from the pollen grains, is significant in the hydrated-activated studied times, whereas this presence is not significant in mature pollen grains. The rapid activation and release of allergenic proteins of P. judaica pollen appears to be the main cause of the allergenic activity of these pollen grains. This work aims to investigate the presence of airborne grass pollen and to identify antigenic and allergenic profiles from eight different grass species collected in the Porto region (Portugal). Poaceae airborne pollen, sampled using a Hirst-type volumetric trap during 2003–2007, was the second most abundant type, and high concentrations were found from April to August. Pollen proteins extracted from the eight grass species collected were separated by SDS-PAGE, being the allergenic profile investigated by immunoblotting using sera from atopic patients and maize profilin polyclonal antibody (ZmPRO3). Pollen extract profiles showed several bands ranging from 10 to 97 kDa. In immunoblotting studies, a low molecular weight protein (12–13 kDa) was recognized by profilin antibody. Also, in all pollen extracts except Zea mays, the IgE binding proteins of 12–13 kDa were detected in sera from the 25 patients with different sensitization profiles presenting high IgE values (>80 kU/l). This protein can be considered as a potential causal agent of the allergic respiratory diseases.  相似文献

Pollen is an important trigger of allergic diseases. Recent studies have shown that ragweed pollen NAD(P)H oxidase generates reactive oxygen species (ROS) and plays a prominent role in the pathogenesis of allergies in mouse models. Here, we demonstrated that allergenic pollen grains showed NAD(P)H oxidase activity that differed in intensity and localization according to the plant families. The activity occurred at the surface or in the cytoplasm in pollen of grasses, birch, and ragweed; in subpollen particles released from ragweed pollen; and at the inner surface or in the cytoplasm but not on the outer wall, which was sloughed off after the rupture, of pollen of Japanese cedar and Japanese cypress. The activity was mostly concentrated within insoluble fractions, suggesting that it facilitates the exposure of tissues to ROS generated by this enzyme. The extent of exposure to pollen-generated ROS could differ among the plant families.  相似文献

Pollinosis caused by several allergenic proteins in pollen grains has been a major problem of healthcare in most parts of the world. Environmental factors such as air pollution are known to alter the release of allergenic pollen proteins from a variety of the plant species. Cassia siamea is commonly planted along roadsides and in industrialised areas in many parts of India. The present study reports the findings of animal experiments demonstrating the effect of air pollution on the allergenicity of Cassia siamea pollen proteins. Total white blood cell (WBC) count and lymphocyte count were significantly higher in animals that received protein extract of pollen collected from a polluted site compared to those that received protein extract of pollen collected from a non-polluted site. This was concomitant with increased production of IgE antibodies; followed by marked degranulation of mast cell leading to heighten type I hypersensitivity in these animals. These results are important for the development of a consensus linking ever-increasing pollution due to industrialisation and an increase in associated pollinosis.  相似文献

The role of pollen grains as a causative agent of respiratory allergic disorders such as asthma and allergic rhinitis is common and very well established. The aim of this study was to assess the frequencies of airborne pollen in the Calcutta metropolis and to identify the taxa which cause significant amounts of sensitization. An aeropalynological survey of the atmosphere of Calcutta was carried out from 2004 to 2006. Skin tests were performed with a panel of the most common pollen types on local patients with clinical features of pollinosis. The meteorological factors responsible for the frequency of the pollen types were analysed. The results of monthly visits to the clinic by these patients were correlated with the monthly pollen counts of three dominant and perennial pollen taxa. The dominant pollen types were Trema (19%), Poaceae (12.98%), Casuarina (5.76%), Cocos (5.7%), Azadirachta (4.65%), Peltophorum (3.71%), Cyperaceae (3.68%), Delonix (3.18%) and Areca (2.56%). Total pollen concentration seems to have a significant positive correlation with temperature and wind speed whereas there was a negative correlation with humidity. Skin tests were most frequently found to be positive with the pollen of Poaceae (49%), Azadirachta (46%), Cocos (47%), Cyperaceae (35%), Peltophorum (33%), Areca (29%), Phoenix (26%), and Borassus (23%). A positive correlation occurred between visits to the clinic and monthly pollen count of Areca, Cocos, and Poaceae. This is the first study to design a pollen calendar for Calcutta city; it will provide useful data for enabling allergologists to achieve accurate diagnosis for patients with pollen hypersensitivity.  相似文献

The dynamics of Poaceae pollen season, in particularly that of the Secale genus, in Szczecin (western Poland) 2004–2008 was analysed to establish a relationship between the meteorological variables, air pollution and the pollen count of the taxa studied. Consecutive phases during the pollen season were defined for each taxon (1, 2.5, 5, 25, 50, 75, 95, 97.5, 99% of annual total), and duration of the season was determined using the 98% method. On the basis of this analysis, the temporary differences in the dynamics of the seasons were most evident for Secale in 2005 and 2006 with the longest main pollen season (90% total pollen). The pollen season of Poaceae started the earliest in 2007, when thermal conditions were the most favourable. Correlation analysis with meteorological factors demonstrated that the relative humidity, mean and maximum air temperature, and rainfall were the factors influencing the average daily pollen concentrations in the atmosphere; also, the presence of air pollutants such as ozone, PM10 and SO2 was statistically related to the pollen count in the air. However, multiple regression models explained little part of the total variance. Atmospheric pollution induces aggravation of symptoms of grass pollen allergy.  相似文献

In earlier works we have investigated the correlation between aerobiological and phytogcographical data, assessing the presence and number of the most important allergenic taxa in our area. More recently we have enquired into the relationship between phenological and aerobiological phenomena, checking the flowering period of the allergenic species growing in different ecological stations.

These studies have been further developed by analytical investigations of the phenomena. Presence-abundance of the taxonomic groups has been assessed by field surveys and represented cartographically. An accurate evaluation of the results has made it possible to draw up a series of maps in order to provide (a) a more precise knowledge of the relationship between pollen dispersal and trapping and the pollen sources, (b) useful data for the interpretation and prediction of symptoms in patients.  相似文献

Over the period 2002–2019, air temperature and precipitation significantly increased regionally for Bratislava, which could lead to phenological changes in some plant species. This study aimed to analyse the changes in the intensity, timing, and duration of pollen seasons of three allergological important plant taxa (Alnus, Poaceae, Artemisia) in the study area over 18 years. The pollen sampling was performed using a Hirst-type sampler. Mann–Kendall tau test was used to determine trends in pollen season characteristics, while Spearman’s correlation analysis was used to identify the relationships between the characteristics of pollen seasons and both air temperature and precipitation trends. The notable changes in the pollen-season-related features were observed for all analysed taxa. The Alnus pollen season now reaches the peak earlier and its intensity is rising in line with the summer-autumn temperature increasing trend, while unexpectedly intensity and duration of the Artemisia pollen season are declining in line with the increased precipitation and/or temperature trends. On the other hand, the intensity of the Poaceae pollen season is also declining, however, without statistically significant correlations with recorded increases in meteorological parameters considered. This phenomenon is probably related to both the reduction of the area of grasslands due to urbanization and the implementation of effective maintenance of urban green areas (e.g., timely mowing preventing the repeatedly flowering of grasses).

Historically in the East Midlands, UK, airborne pollen has been monitored in two cities, Derby and Leicester, situated 41 km (25 miles) apart. The aim of the present study was to compare aerobiological data from both sites to determine if a forecast based on data from one site would be sufficient for both, and to address the wider issue of reproducibility between geographically separated sites. Pollen types recorded could be split into two groups according to annual abundance, maximum daily concentration and the number of high count days. Six taxa made up the abundant group; ash, birch, grass, oak, nettle-type and yew-type, representing 90 and 88% of the total air spora for Derby and Leicester, respectively. Three consecutive years of grass and nettle pollen data are presented, supported by one year of abundant tree pollen data. There were highly significant positive correlations between the counts obtained. Line charts showing the average number of pollen grains m−3 air day−1 show similar trends, and Bland–Altman plots show little discrepancy between the amounts of pollen counted on any given day. Each day was classified according to the UK accepted threshold levels for grass. Weighted kappa statistics showed substantial or almost perfect agreement between the forecast classifications. With the caveat that this would not apply in a region with restrictions to air flow such as a mountain range or with extreme fluctuations such as a coastline site, this study suggests that data from a single site is suitable for forecasting a distance of up to 41 km.  相似文献
