 Microtubules in pollen tubes are evident within the vegetative and generative cell cytoplasm. This observation led to the formulation of several hypotheses regarding the role of microtubules in cytoplasmic movement and the migration of the vegetative nucleus/generative cell along the pollen tube. The study of microtubular motor proteins in pollen tubes followed the discovery and characterization of an immunoreactive homolog of mammalian kinesin in tobacco pollen tubes. Recent identification of dynein-related polypeptides in pollen tubes of Nicotiana tabacum and pollen of Ginkgo biloba is a significant step in the definition of the role of microtubule function within pollen and pollen tubes. Patterns of pollen spectra formation in the tundra zone of Eurasia were considered. Changes in total pollen concentration were traced in subfossil pollen samples of the tundra zone. The data on subfossil pollen spectra were used to evaluate the proportion between local and regional plus extra-local components of tundra pollen samples as well as the changes in concentration of pollen of Scots and Siberian stone pines as well as of tree and shrub birches. The diameter of dwarf birch pollen was determined in different tundra subzones of Western Siberia. The events and processes occurring between pollen maturation, opening of the anther and presentation of pollen to dispersing agents are described. In the final phases of pollen development, starch is always stored; this occurs before the anther opens. Depending on the species, this starch may be totally or partially transformed into: (a) other types of polysaccharides (fructans and rarely callose); (b) disaccharides (sucrose); (c) monosaccharides (glucose and fructose, all situated in the cytoplasm. While awaiting dispersing agents and during dispersal, polysaccharides, especially fructans, and sucrose may be interconverted to control osmotic pressure and prevent loss and uptake of water. Opening of the anther is preceded by disappearance of the locular fluid and in many cases by partial dehydration of the pollen. Pollen generally has a water content between 5 and 50%. Pollen with a high water content may or may not be able to control water retention during pollen exposure and dispersal. Pollen may be dispersed in monads or grouped in pollen dispersing units by the following mechanisms: (i). tangling of filamentous pollen; (ii). adhesion by viscous substances (pollenkitt, tryphine, elastoviscin) derived from the tapetum; (iii). common walls. When the anther opens, the pollen may be dispersed immediately, remain until dispersed (primary presentation), or be presented to pollinators in another part of the flower (secondary presentation).  相似文献

Pollen spectra of 23 Cundill pollen traps from 23 different sampling sites in Southwest Turkey are compared with the corresponding pollen spectra of moss cushions from the same sites. The Cundill pollen traps represent the modern pollen rain data from one year whereas the moss cushions represent the pollen rain of several years. The comparative study reveals some main differences between the two pollen entrapment media. The one-year entrapment medium (pollen trap) appears to be more sensitive for local (releve area 10x10 m) and regional (100-500 m, or a few kilometres for Olea europaea) vegetation. Pollen spectra of moss cushions are dominated by high pine pollen percentage values and hardly sense fine vegetation structures. The conclusion of this comparative modern pollen study allows to interpret fossil sediment spectra from the Near East in a more critical way. It is concluded that one should preferably sample rapidly deposited sediments for palynological analyses, as the resulting highresolution pollen diagrams will be most informative about the former vegetation patterns.  相似文献

The air that we inhale contains simultaneously a multiple array of allergenic pollen. It is well known that such allergens cause allergic reactions in some 15 of the population of the Western World. However little is known about the quantitative aspect of this phenomenon. What is the lowest concentration of pollen that might trigger allergic responses? As people are exposed to heterogeneous and variable environments, clarification of the partial contribution of each of the major airborne pollen allergens and determination of its role in invoking allergy are of prime importance. Objectives: (1) Assessment of a possible correlation between the concentration of airborne pollen and incidence of allergy. (2) Estimation of the lowest average concentrations for various species of airborne pollen that elicit allergic symptoms when exceeded. (3) Determination of the extent of the variations in manifestation of allergy symptoms that can be explained by fluctuations in the concentration of individual species of airborne pollen. Methods: The study was conducted during 14?months with a rural population in Israel. The participants completed a detailed questionnaire and were skin prick tested with the common airborne allergens. The appearance of clinical symptoms, i.e. nasal, bronchial, ocular or dermal, were reported daily by the patients. Concentrations of the airborne pollen and spores were monitored in the center of activity of the residents during one day every week, using three ‘Rotorod’ pollen traps. The pollen grains were identified by light microscopy. Results: The pollen spectrum was divided into time-blocks presenting the main pollination periods of the investigated species. The correlation between the concentration of airborne pollen of the relevant species and the clinical symptoms of the patients was determined for each time block. The correlation differed for different clinical symptoms and for different pollen allergens. Highest correlation with airborne pollen counts was found for patients with nasal and bronchial symptoms. The onset of the clinical symptoms by sensitive patients started, in each of the relevant groups, once the weekly average concentration of the airborne pollen crossed a threshold level. Under the limitations of the present study, this level was estimated to be 2–4 pollen m?3 air for olive, 3–5 pollen m?3 air for grasses, 4–5 pollen m?3 air for Artemisia, 10–20 pollen m?3 air for pecan and 50–60 pollen m?3 air for cypress. Conclusions: Fluctuations in specific airborne pollen grains explained up to 2/3 of the variation in clinical allergy responses. Those were: 69 of the variation for cypress (March–April), 66 for the grasses (March–April), 49 for the pecan (May–June) and 62 for Artemisia (Autumn).  相似文献

Data on pollen and pistil traits from 14 Argentinean Nyctaginaceae species with starch or lipids as pollen reserves are presented. We expect differences in the traits between these two groups of species, but the same pattern within each group for (a) the relationship between pollen size and pistil length assuming that pollen tube length is predetermined by provisions in the pollen independently of the pollen reserve type, and for (b) a trade-off between size and number because available resources for male function are not unlimited. In particular we expect that (a) species with longer pistils will have larger pollen grains, (b) pollen grain size and the number of pollen grains per flower will be negatively correlated. Significant differences in the mean pollen number per flower and mean pollen size were observed between species with different pollen reserve type but not for pistil length. Then, correlation analyses were performed for species with starchy pollen or with pollen with lipids separately. Pollen size - pistil length correlation was positive and significant for species with starchy pollen but not for species with pollen with lipids. On the other hand, pollen size - number correlation was not significant for both starchy and oil-rich species. Results suggest that pollen reserve type would be a relevant factor that constraint pollen size in species of Nyctaginaceae, and that this pollen trait should be considered when studying pollen-pistil relationships, mainly between species of those families with mixed pollen reserves.  相似文献

以蜂蜜为主料,加入花粉,经过调配、发酵、过滤、陈酿后获得氨基酸含量丰富的酒精饮料。花粉破壁分别采用超声破碎法、酶法和温差破壁法这几种不同方式处理,并添加于蜂蜜发酵液中,经酒精发酵,制得富含活性成分的花粉蜂蜜酒。最后利用毛细管电泳法来测定成品酒中的氨基酸,通过分析优化出最佳的花粉破壁方法。研究结果表明花粉经过温差破壁法处理后加入发酵液中,其成品的氨基酸含量最高。该产品不仅营养价值高,风味独特,而且还具有良好的保健作用。  相似文献

The sugar composition of pollen grain and pollen tube cell walls was studied for Camellia japonica, C. sasanqua, C. sinensis, Tulipa gesneriana and Lilium longiflorum. In all species, the main components of pollen grain walls were arabinose, galactose, glucose and uronic acid. On the other hand, the pollen tube walls consisted mostly of glucose. The pollen tube wall of C. japonica was fractionated into hemicellulose, α-cellulose and pectic substance fractions in yields of 61, 19 and 3 %, respectively. The hemicellulose fraction was composed essentially of glucose. The sugar composition of the pollen tube wall was not influenced by the nature of exogenously supplied sugars. Rapid growth of the pollen tube seemed to correlate with the synthesis of hemicellulosic glucan.  相似文献

Summary The permeability of pollen grains, pollen tubes and generative cells of Helleborus foetidus and Galanthus nivalis has been investigated using four probes spanning a wide range of molecular weights: 4,6-diamidino-2-phenyl indole (DAPI; mol.wt. 350). Evans blue (mol.wt. 960), FITC-dextran (average mol.wt. 19400) and FITC-albumin (average mol.wt. 67000). DAPI penetrated into the vegetative cells of desiccated and hydrated pollen, and also entered growing pollen tubes. In contrast, the generative cells of hydrated pollen and of pollen tubes were highly resistant to penetration, as they were when isolated in osmotically balancing medium. Evans blue failed to enter intact generative cells under any of the conditions tested. The dye ultimately entered the vegetative cells of some pollen grains, but these were non-germinable. Growing pollen tubes invariably resisted penetration. Neither of the high molecular weight conjugates entered germinable pollen grains or intact pollen tubes. The results suggest that it is highly unlikely that DNA fragments of high molecular weight can enter viable pollen, pollen tubes or generative cells under any normal conditions.  相似文献

Worldwide, 400 million people suffer from hay fever and seasonal asthma. The major causative agents of these allergies are pollen specific proteins called the group-1 grass pollen allergens. Although details of their antigenicity have been studied for 40 years with an eye towards immunotherapy, their function in the plant has drawn scant attention. Zea m 1 constitutes a class of abundant grass pollen allergens coded for by several genes that loosen the walls of grass cells, including the maize stigma and style. We have examined the impact of a transposon insertion into one of these genes (EXPB1, the most abundant isoform of Zea m 1) on the production of Zea m 1 protein, pollen viability, and pollen tube growth, both in vitro and in vivo. We also examined the effect of the insertional mutation on the competitive ability of the pollen by experimentally varying the sizes of the pollen load deposited onto stigmas using pollen from heterozygous plants and then screening the progeny for the presence of the transposon using PCR. We found that the insertional mutation reduced the levels of Zea m 1 in maize pollen, but had no effect on pollen viability, in vitro pollen tube growth or the proportion of progeny sired when small pollen loads are deposited onto stigmas. However, when large pollen loads are deposited onto the stigmas, the transposon mutation is vastly underrepresented in the progeny, indicating that this major pollen allergen has a large effect on pollen tube growth rates in vivo, and plays an important role in determining the outcome of the pollen-pollen competition for access to the ovules. We propose that the extraordinary abundance (4% of the extractable protein in maize pollen) of this major pollen allergen is the result of selection for a trait that functions primarily in providing differential access to ovules.  相似文献

Sexual reproduction of flowering plants depends on delivery of the sperm to the egg, which occurs through a long, polarized projection of a pollen cell, called the pollen tube. The pollen tube grows exclusively at its tip, and this growth is distinguished by very fast rates and reaches extended lengths. Thus, one of the most fascinating aspects of pollen biology is the question of how enough cell wall material is produced to accommodate such rapid extension of pollen tube, and how the cell wall deposition and structure are regulated to allow for rapid changes in the direction of growth. This review discusses recent advances in our understanding of the mechanism of pollen tube growth, focusing on such basic cellular processes as control of cell shape and growth by a network of cell wall-modifying enzymes, molecular motor-mediated vesicular transport, and intracellular signaling by localized gradients of second messengers.  相似文献

The mean diameter of viable pollen grains is approximately 13 μm greater than the mean diameter of inviable grains in Mimulus guttatus. We show that this difference is large enough to be detected by particle counters and that these machines can be used to obtain a rapid estimate of pollen viability. While requiring a separate calibration, a size-based statistic is also strongly correlated with pollen viability in Collinsia verna. These results suggest that statistics derived from the size distribution of pollen grains may provide an alternative to more labor-intensive methods for estimating pollen viability, particularly in cases where inviability results from inbreeding depression or hybrid failure.  相似文献

In this study on Viola tricolor pollen, the competitive ability of 16 pollen donors originating from a wild population was analysed in a set of greenhouse and germination temperatures. The aim was to examine the consistency in donor pollen performance across temperatures and to see whether variation in performance was random or due to individual differences in the plastic response to temperature. Pollen tube growth rate in vitro was investigated in two greenhouse temperatures (on the day pollen was collected) and in four germination temperatures. In addition, pollen tube growth rate was assessed in vivo (in one temperature) to examine the relationship between in vivo and in vitro growth. A temperature difference of 5 K - corresponding to natural variation in time and space detected in the field - affected pollen tube growth rate. For both temperature components, significant pollen donor by temperature interactions were found and rank order of pollen donors changed across treatments. Although pollen competitive ability in violets was strongly influenced by both temperature components, the occurrence of pollen donor by temperature interactions indicates that donor siring ability varies with temperature. This, in turn, may suggest a means to maintain pollen competitive ability despite selection for this trait.  相似文献

In this study, the pollen‐active LAT59 promoter from tomato was used to express a green fluorescent protein (GFP) encoding gene in Nicotiana tabacum (tobacco) pollen. This promoter is preferentially expressed in anthers and pollen. Pollen in transgenic plants segregated in a 1 : 1 Mendelian ratio, and the plants were polymerase chain reaction (PCR)‐positive. GFP‐tagged pollen was developed as a tool for tracking the movement of transgenic plant pollen in the environment. Specifically, it should be a useful tool for characterizing the spatial distribution patterns of transgenic pollen, to determine pollination mechanisms, to monitor the effects on nontarget organisms, and to monitor gene flow in field conditions.  相似文献

The concept of a pollen tube attractant was proposed in the late nineteenth century when pollen tubes were found to grow toward excised pistil tissues on medium. Since then, for about 140 years, plant biologists have tried to identify the pollen tube attractants. However, no molecule has been convincingly demonstrated to be the true attractant that actually controls the navigation of pollen tubes in the pistil. The past decade has seen substantial progress in this field in terms of our understanding of the various mechanisms of pollen tube guidance. It was suggested that diffusible pollen tube attractants might provide localized signals that affect the directional growth of the pollen tube, especially in the last phase of guidance by the target female gametophyte. Here, we review the mechanisms of pollen tube guidance, with special focus on the gametophytic guidance and the attractant. The necessary and appropriate conditions required by the true attractant will be discussed. Environmental monitoring of pollen grains in the atmosphere of Melbourne has been achieved using Burkard volumetric traps. Twenty-two families of flowering plants and confiers were identified in the pollen counts. About 62% of these pollen grains belonged to trees, 20% to grasses and 9% to herbs and weedy plants. During spring and summer, the atmosphere contained about 70% of the total annual pollen count. Tree pollen, predominantly elm and cypress, occurred abundantly in late winter and spring, with grass pollen predominantly in spring and early summer. These three types of pollen grains occurred in significant amounts, together accounting for more than 60% of the total annual catch. A seasonal incidence chart (pollen calendar) for Melbourne based on 2 years observation has been constructed. This pollen calendar is useful in identifying sources of allergies against particular seasonal airborne pollen types. Comparison of the time of occurrence of a particular pollen type using the pollen calendar and the time of allergic symptoms, can lead to accurate diagnosis and preventive measures being taken. This study has confirmed that grass pollen is the major source of allergenic pollen in the external environment triggering hay fever and allergic asthma in spring and early summer in Melbourne, Australia.  相似文献

Cell wall hydrolases are well documented to be present on pollen, but their roles on the stigma during sexual reproduction have not been previously demonstrated. We explored the function of the tapetum-synthesized xylanase, ZmXYN1, on maize (Zea mays L.) pollen. Transgenic lines (xyl-less) containing little or no xylanase in the pollen coat were generated with use of an antisense construct of the xylanase gene-coding region driven by the XYN1 gene promoter. Xyl-less and wild-type plants had similar vegetative growth. Electron microscopy revealed no appreciable morphological difference in anther cells and pollen between xyl-less lines and the wild type, whereas immunofluorescence microscopy and biochemical analyses indicated an absence of xylanase on xyl-less pollen. Xyl-less pollen germinated as efficiently as wild-type pollen in vitro in a liquid medium but less so on gel media of increasing solidity or on silk, which is indicative of partial impaired water uptake. Once germinated in vitro or on silk, the xyl-less and wild-type pollen tubes elongated at comparable rates. Tubes of germinated xyl-less pollen on silk did not penetrate into the silk as efficiently as tubes of wild-type pollen, and this lower efficiency could be overcome by the addition of xylanase to the silk. For wild-type pollen, coat xylanase activity on oat spelled xylan in vitro and tube penetration into silk were inhibited by xylose but not glucose. The overall findings indicate that maize pollen coat xylanase facilitates pollen tube penetration into silk via enzymatic xylan hydrolysis.  相似文献

This is a study of the acid-fast staining characteristics of pine pollen and an investigation of factors causing loss of acid-fastness after pine pollen has been in contact with tisues or mucous membranes. Intact loblolly pine pollen was readily stained with cold carbol-fuchsin, and retained its acid-fastness after decolorization with 3% HC1 in 95% ethyl alcohol for 2 min, followed by methylene blue counterstain. Pine pollen resembles spermatozoa in ease of staining and resistance to decolorization. Acid-fastness was destroyed by crushing or by germination of the pollen grain, and by contact for several hours with serum or saline solutions, but was unchanged by exposure to 0.1% solution of streptomycin in water. Nonviable pine pollen did not lose acid-fastness after suspension for several days in serum or water. When counterstain was omitted, crushed or germinated pollen appeared red to pink after staining with carbol-fuchsin and decolorization with acid alcohol, thus indicating that lipids of an acid-fast nature were still present. The large size and biologic properties of pine pollen provide a unique means of studying the chemical and physical aspects of the acid-fast phenomenon.  相似文献
