—The incorporation of [2-14C]mevalonate into nonsaponifiable lipids by rat brain homogenates is inhibited by phenolic acids derived from tyrosine. The phenyl acids derived from phenylalanine are inhibitory only at very high concentrations compared with phenolic acids. The brain is more sensitive to inhibition by the phenolic acids than the liver. These studies indicate a possible role for phenolic acids in the impairment of cerebral sterol metabolism in phenylketonuria.  相似文献

The synthesis of 1,3,5-13C3- and 2,4-13C2-labeled 5-O-bromobenzyl-2-deoxyribonolactones 2, precursors to 13C-enriched nucleoside phosphoramidites for solid-phase synthesis of DNA oligonucletides, is described. An equimolar combination of these two multiply labeled lactones affords a “population-labeled” mixture of isotopomers which exhibits an approximately 50-fold increase in the sensitivity of 13C-NMR compared to natural abundance measurements. The 13C-13C 2-bond and 4-bond coupling constants are reported for the lactones; all are < 2 Hz, confirming that this labeling scheme should be especially useful for NMR-relaxation measurements.  相似文献

1. A tissue culture method was devised in which suspensions of osteoblasts, obtained directly from frontal bones of fowl embryos, were grown in a fluid, fibrin-free medium. 2. Maximum growth of the tissue, as measured by dry weight, with the formation of collagen protein, based on the estimation of hydroxyproline, was obtained in periods of up to 6 days. 3. Appreciable amounts of protein-bound hydroxyproline were formed during the first 24 hour growth period, but electron microscopy of portions of the same cultures failed to demonstrate the presence of any typical collagen fibrils. 4. The subsequent formation of many characteristic collagen fibrils was not associated with a significant rise in the mean hydroxyproline content of the tissue. 5. The cytoplasmic granules of the osteoblasts stained intensely with the P.A.S. technique when the collagen fibrils were being formed. 6. It is suggested that collagen-forming cells synthesise and secrete a hydroxyproline-rich precursor of protein or large peptide nature, which subsequently becomes directly transformed into typical collagen fibrils.  相似文献

The uptake of C14-labeled proteins (lysozyme, hemoglobin, lactoglobulin, and ovalbumin) from solution by tomato plants with sterile roots was studied. It was found that C14-compounds (proteins and/or protein-degradation products) were translocated to the foliage if the roots had undergone minor mechanical injury or if the plants were subjected to temporary wilting, i.e., physiological damage. C14-lysozyme was not transported to foliar tissue in healthy plants; C14-hemoglobin showed radioactivity in leaves of both healthy and injured plants, but there was evidence of a breakdown of the molecule; C14-ovalbumin gave a faint labeling of foliar tissues of some plants in which wilting or mechanical damage was below the threshold of detection. It is concluded, however, that translocation of proteins from roots in nutrient solution to tomato leaves does not occur in significant amounts in healthy plants in spite of the large uptake of proteins by root cortex, as found in earlier studies.  相似文献

The major C14-labeled substance in sieve tube exudate of M. pyrifera is D-mannitol, comprising 3.6% (w/v). No sugars are detectable. Certain amino acids also possess some C14-labeling and occur in significantly high concentrations in exudate. The exudate contains negligible ether-soluble lipid, but has a large amount of protein and a high concentration of K+ Neither protein nor lipid become labeled significantly in sieve tubes during short-term translocation experiments with C14. In general the chemical composition of the assimilate stream is comparable to that of vascular plants and does not, consequently, necessitate a different mechanism for translocation.  相似文献

1. The deposition of casein on a platinum anode which takes place on the passage of a direct current through solutions of alkali caseinates was quantitatively studied, and it was found that: (a) the amount of casein which is deposited is directly proportional to the current, i.e. it obeys Faraday''s law; (b) the amount of casein deposited is inversely proportional (within the limits studied) to the amount of alkali which is combined with the casein. 2. A method of determining the transport numbers of proteins insoluble at their isoelectric point has been developed. 3. A titration method for determining the amount of alkali in a casein solution is given. 4. Data from the results of transference experiments on sodium caseinate, potassium caseinate, cesium caseinate, and rubidium caseinate solutions are given. It is shown that the data are best explained on the assumption that in these solutions the carriers of the current are alkali metal cations and casein anions. 5. On the basis of our transference results an explanation is given of the results which were obtained by Robertson and by Haas in their migration experiments.  相似文献

O'Leary, William M. (Cornell University Medical College, New York, N.Y.). S-adenosylmethionine in the biosynthesis of bacterial fatty acids. J. Bacteriol. 84:967-972. 1962.-The fatty-acid contents of a wild strain of Aerobacter aerogenes and of three auxotrophic mutants were determined by the use of gas-liquid chromatography. All strains studied exhibited essentially identical fatty-acid spectra. This organism was shown to contain large amounts of a C(17) cyclopropane fatty acid, and it is suggested that it may be a convenient natural source of this compound. By use of the auxotrophic mutants which have varying requirements for exogenous sources of methionine and S-adenosylmethionine, it was shown by means of radioisotope tracer techniques that S-adenosylmethionine functions in this organism as a donor of a one-carbon unit that is used to form the ring structure in the biosynthesis of cyclopropane fatty acids.  相似文献
