Antibacterial substances active against Corynebacterium fascians have been shown to be present in the coats of various seeds. There were considerable differences in activity between the seeds of different genera and species and between individual seeds of a single species.
C. fascians also showed strain variation in sensitivity to the antibacterial substances of the seeds of any one species. An inverse relationship was demonstrated between the sensitivity of a strain to the sweet-pea seed substances and its ability to infect sweet-pea seedlings under laboratory conditions.  相似文献

Two types of organism are known to be present in strawberry plants suffering from cauliflower disease, namely, eelworms, Aphelenchoides ritzema-bosi and/or Aphelenchoides fragariae , and a bacterium, Corynebacterium fascians. Neither type, inoculated separately, has consistently reproduced the disease.
An experimental investigation of a possible obligate etiological relationship between the eelworm and the bacterium is described, in which Aphelenchoides ritzema-bosi and several strains of Corynebacterium fascians were inoculated together and separately.
No abnormalities appeared in the absence of the eelworm. Typical cauliflower symptoms were reproduced only by a combination of the eelworm and strains of C. fascians originally isolated from cauliflower strawberries. Small alaminate leaves and enations appeared in all treatments where the eelworm was introduced, including those where no bacteria were co-inoculated. Subsequent isolations, however, demonstrated the presence of contaminant strains of C. fascians in the latter plants. The severity of the enation symptom was related to the presence of a particular strain of C. fascians , but the alaminate leaves showed no such correlation. Evidence on the source of the contaminant strains found in the experimental plants is discussed.
In parallel field investigations it was established that eelworm-infested strawberries showing no cauliflower symptom frequently bore alaminate leaves, with which C. fascians was consistently associated. It is suggested therefore that cauliflower is simply the least frequent of a range of symptoms of a bacterial disease endemic in strawberry plants infested with Aphelenchoides spp.  相似文献

Laboratory experiments showed that gram ( Cicer arietinum ) could be infected by Corynebacterium fascians. Parallel tests on sweet peas and gram confirmed the existence of host specificity among strains of the pathogen, and of differences in host susceptibility towards a given strain. Adventitious root formation appeared as a new, though rare, sign of infection. Inoculation of aerial dormant buds invariably caused infection whereas cotyledonary bud inoculation often failed. Sporing bacteria antagonistic to C. fascians in culture media were isolated from soil, but they failed to exert their influence in infection experiments in sand.  相似文献

Abstract The sacB gene of Bacillus subtilis was successfully applied in various Arthrobacter, Brevibacterium, Corynebacterium and Rhodococcus strains for the isolation of transposable elements. Three different insertion sequence (IS) elements entrapped in sacB were isolated. The IS elements IS- Bl and IS- Cg isolated from Brevibacterium lactofermentum and Corynebacterium glutamicum , respectively, were found to be similar in size (1.45 kb) and generated target duplications of 8 bp. Their inverted repeats showed homology. In contrast, the IS element IS- Rf isolated from Rhodococcus fascians was only 1.3 kb long and generated a 3-bp target duplication. IS- Cg and IS- Rf were not restricted to their original host strains, and we also found strains harbouring more than one element.  相似文献

All avirulent mutants isolated by growth on rich media at 37 degrees C were unable to grow on media containing agmatine or proline as sole sources of nitrogen. Until recently, the presence in transfer ribonucleic acid (tRNA) of the hydroxylated cytokinin ribosylzeatin [N6-(4-hydroxy-3-methylbut-2-enyl)adenosine]was thought to be unique to higher plants. This extension of work from several laboratories indicates the presence of 2-methylthioribosylzeatin in the tRNA of the plant-associated bacteria Rhizobium leguminosarum, Agrobacterium tumefaciens, and Corynebacterium fascians, but not in that of Erwinia amylovora. This cytokinin has the cis configuration, as is normally found in the tRNA's of plants. The tRNA thionucleotide patterns in these bacteria are different from those of Escherichia coli, Bacillus subtilis, and Salmonella typhimurium, which contain the unhydroxylated analogs of ribosylzeatin or 2-methylthioribosylzeatin.  相似文献

Bacteria were either isolated from leaf surfaces of Hedera helix or obtained from a culture collection in order to analyse their effect on barrier properties of isolated Hedera and Prunus laurocerasus cuticles. On the basis of the 16S rDNA sequences the genera of the six bacterial isolates from Hedera were identified as Pseudomonas sp., Stenotrophomonas sp. and Achromobacter. Water permeability of cuticles isolated from H. helix was measured before and after inoculation with the six bacterial strains. In addition water permeability of cuticles isolated from P. laurocerasus was measured before and after inoculation with the three bacterial strains Pseudomonas aeruginosa, Xanthomonas campestris and Corynebacterium fascians. Rates of water diffusing across isolated cuticles of both species significantly increased by up to 50% after inoculation with all bacterial strains. Obtained results show that epiphytic bacteria have the ability of increasing water permeability of Hedera and Prunus cuticles, which in turn should increase the availability of water and dissolved compounds in the phyllopshere. Consequently, living conditions in the habitat phyllosphere are improved. It can be concluded that the ability to change leaf surface properties will improve epiphytic fitness of leaf surface bacteria.  相似文献

A strain identified as Corynebacterium fascians was found to accumulate extracellular D-alanine from glycerol. Cultural conditions for the accumulation of D-alanine were investigated and, as a result, a yield of 7 g of D-alanine per liter was obtained after a 96-h incubation in a medium containing 5% glycerol, 4% (NH(4))(2)HPO(4), and 0.3% corn steep liquor. Optical purity of D-alanine was dependent upon the concentration of corn steep liquor. At the optimal condition, almost optically pure D-alanine was formed and readily isolated (5 g/liter) from the fermentation broth. The product was not contaminated with any detectable amount of other amino acids, except for glycine which was present at a concentration of less than 1 percent.  相似文献

A simple method for PCR-based plant clinical diagnosis of bacterial blight of geraniums caused by Xanthomonas campestris pv. pelargonii is described. The method entails maceration of infected tissues in water or 10mM Tris- HCI, pH 8.0 buffer, followed by treatment of the macerate with a commercially-available extraction matrix (GeneReleaserTM) in which nucleic acid is released by brief microwave heating. Nucleic acid prepared in this manner served directly as template for PCR amplification with primers targeting a sequence in the genome of the bacterium. Using this protocol, it was possible to quickly identify X. campestris pv. pelargonii in infected geraniums, whereas amplification products were not obtained with nucleic acid preparations from noninfected plants, or from plants infected with the bacterial pathogens, Corynebacterium fascians or Pseudomonas cichorii .  相似文献

Rhodococcus fascians is a nocardiform bacteria that induces leafy galls (fasciation) on dicotyledonous and several monocotyledonous plants. The wild-type strain D188 contained a conjugative, 200 kb linear extrachromosomal element, pFiD188. Linear plasmid-cured strains were avirulent and reintroduction of this linear element restored virulence. Pulsed field electrophoresis indicated that the chromosome might also be a linear molecule of 4 megabases. Three loci involved in phytopathogenicity have been identified by insertion mutagenesis of this Fi plasmid. Inactivation of the fas locus resulted in avirulent strains, whereas insertions in the two other loci affected the degree of virulence, yielding attenuated (att) and hypervirulent (hyp) bacteria. One of the genes within the fas locus encoded an isopentenyltranferase (IPT) with low homology to analogous proteins from Gram-negative phytopathogenic bacteria. IPT activity was detected after expression of this protein in Escherichia coli cells. In R.fascians, ipt expression could only be detected in bacteria induced with extracts from fasciated tissue. R.fascians strains without the linear plasmid but containing this fas locus alone could not provoke any phenotype on plants, indicating additional genes from the linear plasmid were also essential for virulence. These studies, the first genetic analysis of the interaction of a Gram-positive bacterium with plants, suggest that a novel mechanism for plant tumour induction has evolved in R.fascians independently from the other branches of the eubacteria.  相似文献

Urealytic strains of coryneform bacteria that are designated Corynebacterium group D2 and are isolated from human urine are a cause of urinary tract infections. Cell wall and lipid analyses confirmed that these organisms are members of the genus Corynebacterium but can be separated from other species in the genus on the basis of DNA base composition and DNA-DNA hybridization values. Biochemically, strains in this taxon can be distinguished from other Corynebacterium spp. by their failure to produce acid from carbohydrates, by their failure to reduce nitrates, and by their ability to hydrolyze urea. We regard these bacteria as a new species of the genus Corynebacterium and propose the name Corynebacterium urealyticum. The type strain is strain NCTC 12011 (= ATCC 43042).  相似文献

The role and metabolism of indole-3-acetic acid in gram-negative bacteria is well documented, but little is known about indole-3-acetic acid biosynthesis and regulation in gram-positive bacteria. The phytopathogen Rhodococcus fascians, a gram-positive organism, incites diverse developmental alterations, such as leafy galls, on a wide range of plants. Phenotypic analysis of a leafy gall suggests that auxin may play an important role in the development of the symptoms. We show here for the first time that R. fascians produces and secretes the auxin indole-3-acetic acid. Interestingly, whereas noninfected-tobacco extracts have no effect, indole-3-acetic acid synthesis is highly induced in the presence of infected-tobacco extracts when tryptophan is not limiting. Indole-3-acetic acid production by a plasmid-free strain shows that the biosynthetic genes are located on the bacterial chromosome, although plasmid-encoded genes contribute to the kinetics and regulation of indole-3-acetic acid biosynthesis. The indole-3-acetic acid intermediates present in bacterial cells and secreted into the growth media show that the main biosynthetic route used by R. fascians is the indole-3-pyruvic acid pathway with a possible rate-limiting role for indole-3-ethanol. The relationship between indole-3-acetic acid production and the symptoms induced by R. fascians is discussed.  相似文献

Rhodococcus fascians is a plant-pathogenic bacterium that causes malformations on aerial plant parts, whereby leafy galls occur at axillary meristems. The colonization behavior on Nicotiana tabacum and Arabidopsis thaliana plants was examined. Independent of the infection methods, R. fascians extensively colonized the plant surface where the bacteria were surrounded by a slime layer. R. fascians caused the collapse of epidermal cells and penetrated intercellularly into the plant tissues. The onset of symptom development preceded the extensive colonization of the interior. The meristematic regions induced by pathogenic strain D188 were surrounded by bacteria. The nonpathogenic strain, D188-5, colonized the exterior of the plant equally well, but the linear plasmid (pFiD188) seemed to be involved in the penetration efficiency and colonization of tobacco tissues.  相似文献

The effect of Rhodococcus fascians, the causal agent of leafy gall disease, on the mitotic behavior of synchronized tobacco Bright Yellow-2 (BY-2) cells was investigated. Incubation of aphidicolin-synchronized BY-2 cells with R. fascians cells specifically resulted in a broader mitotic index peak, an effect that was linked to an intact and expressed fas virulence locus. The obtained results pointed towards an effect of R. fascians on the prophase of mitosis. The relevance of these results to the virulence of the bacterium is discussed.  相似文献

The morphological, cytochemical (acid phosphatase activity) and cytophysiological (phagocytosis) features of mouse peritoneal macrophages activated in vivo by two bacterial agents. Corynebacterium parvum parvum and Polidin, were investigated in vitro. The presence of a 138-kilobase plasmid (pD188) correlated with increased resistance to cadmium in Rhodococcus fascians D188. This plasmid could be transferred by a conjugation-like system in matings between R. fascians strains. Transconjugants expressed the cadmium resistance and could be used as donors in subsequent matings. Four other R. fascians strains (NCPPB 1488, NCPPB 1675, NCPPB 2551, and ATCC 12974) could also be used as donors for cadmium resistance in matings. Strain NCPPB 1675 showed a 100% cotransfer of cadmium and chloramphenicol resistance markers.  相似文献
