The first specimen of platypus (Ornithorhynchus anatinus) that reached Britain in the late 18th century was regarded a scientific hoax. Over decades the anatomical characteristics of these unique mammals, such as egg laying and the existence of mammary glands, were hotly debated before they were accepted. Within the last 40 years, more and more details of monotreme physiology, histology, reproduction and genetics have been revealed. Some show similarities with birds or reptiles, some with therian mammals, but many are very specific to monotremes. The genome is no exception to monotreme uniqueness. An early opinion was that the karyotype, composed of a few large chromosomes and many small ones, resembled bird and reptile macro- and micro-chromosomes. However, the platypus genome also features characteristics that are not present in other mammals, such as a complex translocation system. The sex chromosome system is still not resolved. Nothing is known about dosage compensation and, unlike in therian mammals, there seems to be no genomic imprinting. In this article we will recount the mysteries of the monotreme genome and describe how we are using recently developed technology to identify chromosomes in mitosis, meiosis and sperm, to map genes to chromosomes, to unravel the sex chromosome system and the translocation chain and investigate X inactivation and genomic imprinting in monotremes.  相似文献

Immunohistochemical studies on the pituitary of Anolis carolinensis detected ACTH-like, β-endorphin-like, and 16K fragment-like immunoreactivity in distinct clusters of cells in the anterior lobe; ACTH-like, αMSH-like, β-endorphin-like, and 16K fragment-like immunoreactivity was detected in all the cells of the intermediate lobe. Crude acid extracts of both lobes, when alayzed by radioimmunoassay, gave displacement curves in ACTH and β-endorphin assays which were parallel to the appropriate synthetic standard. Only extracts of the intermediate lobe gave parallel displacement curves in an αMSH radioimmunoassay. Extracts of both lobes crossreacted with antiserum to 16K fragment, but the displacement curves were not parallel to that of mouse 16K fragment standard. The levels of immunoreactive ACTH and β-endorphin in the intermediate lobe were approximately 8-fold higher than in the anterior lobe. Fractionation of anterior lobe and intermediate lobe extracts by either gel filtration on Sephadex G-75 in 10% formic acid or sodium dodecyl sulfate polyacrylamide gel electrophoresis revealed multiple forms of ACTH-related and β-endorphin-related substances in both lobes. In the anterior lobe the major forms of immunoreactivity were, respectively, ACTH-sized and β-endorphin-sized. In the intermediate lobe the major forms of immunoreactivity were αMSH-sized, CLIP-sized, and β-endorphin-sized. In both lobes, antisera directed against ACTH and β-endorphin detected high molecular weight material with an apparent molecular weight slightly less than that of mouse pro-ACTH/endorphin; this material probably represents the putative common precursor for ACTH and β-endorphin in this species.  相似文献

Natural disturbances are key processes in the vast majority of ecosystems and a range of ecological theories have been developed in an attempt to predict biotic responses to them. However, empirical support for these theories has been inconsistent and considerable additional work remains to be done to better understand the response of biodiversity to natural disturbance. We tested predictions from the intermediate disturbance hypothesis and the habitat accommodation model of succession for reptile responses to fire history and a single major fire event. We focused our work on a broad range of vegetation types spanning sedgeland to temperate rainforest located within a national park in south‐eastern Australia. We found no significant relationships between reptile species richness and the number of fires over the past 35 years, the time since the last fire, or the severity of a major fire in 2003. Thus, we found no strong evidence to support the intermediate disturbance hypothesis. A correspondence analysis of reptile assemblages revealed a gradient in species responses to fire history. However, we found limited evidence for an ordered succession of reptiles. Nor could the responses of individual species be readily predicted from life history attributes. Thus, our findings were generally not consistent with predictions from the habitat accommodation model of succession. A possible explanation for the absence of a predictable sequence of recovery following disturbance might be the rapidity of post‐fire recovery of many components of native vegetation cover that were found to be important for reptiles (e.g. the extent of grass cover). This would have limited the time for early successional conditions to prevail and limited opportunities for species associated with such conditions. We found that most reptile species responses were much more strongly linked to vegetation type than fire variables, emphasizing a need to understand relationships with vegetation before being able to understand possible fire effects (if and where they exist). We found the disturbance concepts we examined were limited in their ability to accurately predict reptile responses to past fire history or the impacts of a single major fire in 2003. Practical management might be best guided not by disturbance theory, but by carefully setting objectives to meet conservation goals for particular individual species of reptiles.  相似文献

Global temperatures have risen over the last century, and are forecast to continue rising. Ectotherms may be particularly sensitive to changes in thermal regimes, and tropical ectotherms are more likely than temperate species to be influenced by changes in environmental temperature, because they may have evolved narrow thermal tolerances. Keelback snakes (Tropidonophis mairii) are tropical, oviparous reptiles. To quantify the effects of temperature on the morphology and physiology of hatchling keelbacks, clutches laid by wild-caught females were split and incubated at three temperatures, reflecting the average minimum, overall average and average maximum temperatures recorded at our study site. Upon hatching, the performance of neonates was examined at all three incubation temperatures in a randomized order over consecutive days. Hatchlings from the ‘hot’ treatment had slower burst swim speeds and swam fewer laps than hatchlings from the cooler incubation temperatures in all three test temperatures, indicating a low thermal optimum for incubation of this tropical species. There were no significant interactions between test temperature and incubation temperature across performance variables, suggesting phenotypic differences caused by incubation temperature did not acclimate this species to post-hatching conditions. Thus, keelback embryos appear evolutionarily adapted to development at cooler temperatures (relative to what is available in their habitat). The considerable reduction in hatchling viability and performance associated with a 3.5 °C increase in incubation temperature, suggests climate change may have significant population-level effects on this species. However, the offspring of three mothers exposed to the hottest incubation temperature were apparently resilient to high temperature, suggesting that this species may respond to selection imposed by thermal regime.  相似文献

We describe a new lizard taxon, Stefanikia siderea gen. et sp. nov., from the early-middle Eocene locality of Messel in Germany based on a nearly complete skeleton, which we studied using μCT methods. It shares many characters with the Eocene taxon Eolacerta, which is broadly distributed in the Eocene of central and Western Europe, but is much smaller and shows several important anatomical differences. The new discovery sheds light on the paleodiversity of these lizards in the Eocene of Europe, and the new family name Eolacertidae is proposed to encompass Eolacerta and Stefanikia. The relationships of Eolacerta have been intractable. Our phylogenetic analyses confirm that Eolacertidae is a member of the clade Lacertiformes and provide strong support for a sister-group relationship to Lacertidae. In some places, skin impressions are preserved, displaying the body scalation. As such, the exquisitely preserved specimens of Eolacertidae from Messel provide new insight into the morphology and ecology of lizards on the stem of Lacertidae, Europe’s dominant group of living reptiles.

We compared the performances of the candidate loci for moss DNA barcoding and the primers used in amplifying the loci. Primers for three coded loci (matK, rps4 and rbcL a) and four non coded loci (atpB rbcL, atpF H, psbK I and trnH psbA) of the chloroplast genome, one from the mitochondrial genome (nad5), and one from the nucleus genome (ITS2) were evaluated. Seventy four samples representing 14 species belonging to five genera of Trachypodoaceae (or Meteoriaceae) were screened. All primers for matK and a pair of primers for trnH psbA failed. Low successes were encountered with the primers for atpF H and psbK I. The primers for psbK I produced several bands and the PCR products of atpF H were difficult to sequence. The powers of the remaining six loci were compared using the variability, identification success and the resolutions. It was found that ITS2 is the most promising candidate for DNA barcoding for mosses. Among the chloroplast genes, atpB rbcL exhibited the highest resolution. Although trnH psbA is very variable, it is too short to be an ideal barcode alone. Combinations of chloroplast genes were also tried and Ps of both atpB rbcL+trnH psbA and rbcL a++trnH psbA were 64% using NJ method. More additions of loci did not increase the resolution. No barcoding gap exists for all these loci. Phylogenetic analyses were carried out prior to the DNA barcoding evaluation and some taxonomic problems do exist. This study exemplifies the necessity of correct species delimitation and the adoption of both plastid and nuclear loci in plant DNA barcoding.  相似文献

Summary A method is described for large scale routine phenotyping of haptoglobin (Hp) which allows complete subtyping without prior purification of the Hp molecule. The procedure includes polyacrylamide gel isoelectric focusing of reduced, neuraminidase treated serum or plasma samples, and nitrocellulose blots developed with the immunoperoxidase technique. Different variables including sample treatment, electrofocusing, blotting procedures, and immunoperoxidase visualization are discussed.Characteristic -chain patterns allow identification of the common allotypes 2FS, 2SS, 2FF, IS, IF, and Johnson. Isoelectric variations in the -chain may also be recognized. For comparison, two-dimensional Hp-patterns are presented. The results from concurrent typing of 600 samples by ordinary starch gel electrophoresis and by the described isofocusing technique, are evaluated.  相似文献
