Isolation of bioactive actinomycetes from marine sediments using rifampicin   总被引:4,自引:0,他引:4  
Summary Bioactive actinomycetes were isolated from marine sediments using rifampicin. Plating the sediments on starch-casein agar, supplemented with rifampicin, eliminated the occurrence of contaminating microorganisms. Total counts, however, were reduced in the presence of rifampicin. Most of the isolates contained ll-2,6-diaminopimelic acid (DAP), whereas 37% contained meso-DAP. The use of increasing concentrations of rifampicin tended to yield a higher proportion of strains with cell extracts positive for meso-DAP. Streptomyces and Micromonospora represented the major genera identified. Antimicrobial activity was exhibited by 46% of the isolates, primarily against Gram-positive bacteria. Inhibition of Gram-negative bacteria was minimal, but antimycotic activity was displayed by 28% of the actinomycetes. Most of the latter activity was attributable to polyenes, particularly hexanenes. The results obtained indicate that rifampicin, added to starch-casein agar, is effective for the isolation of bioactive actinomycetes from marine sediments.  相似文献

Summary A total of 94 actinomycete strains were isolated from the marine sediments of a shrimp farm, 87.2% belonged to the genus Streptomyces, others were Micromonospora spp. Fifty-one percent of the actinomycete strains showed activity against the pathogenic Vibrio spp. strains. Thirty-eight percent of marine Streptomyces strains produced siderophores on chrome azurol S (CAS) agar plates. Seven strains of Streptomyces were found to produce siderophores and to inhibit the growth of Vibrio spp. in vitro. Two of them belonged to the Cinerogriseus group, the most frequently isolated group of Streptomyces. The results showed that streptomycetes could be a promising source for biocontrol agents in aquaculture.  相似文献

本研究采用均匀实验设计优化预处理条件来分离黄海海泥和庐山土壤样品中的放线菌。通过均匀实验得到最优的预处理条件为湿热50℃处理20 min,不进行超声处理以及添加苯酚。经过形态排重后,从海泥和庐山样品中分离得到86株和11株不同表型的放线菌。通过进一步的分子生物学鉴定,海泥中的86株放线菌分别属于3个不同的属,而庐山样品的11株分别属于4个不同的属。对这97株放线菌进行抗菌实验发现,18.5%的菌株对大肠杆菌有抑菌活性,7.2%的菌株对枯草芽孢杆菌有抑菌活性,但对酿酒酵母都无抑菌活性。  相似文献

Actinomycetes were isolated from near-shore marine sediments collected at 15 island locations throughout the Bahamas. A total of 289 actinomycete colonies were observed, and all but 6 could be assigned to the suprageneric groups actinoplanetes and streptomycetes. A bimodal distribution in the actinomycete population in relation to depth was recorded, with the maximum numbers occurring in the shallow and deep sampling sites. This distribution can be accounted for by a rapid decrease in streptomycetes and an increase in actinoplanetes with increasing depth and does not conform to the theory that actinomycetes isolated from marine sources are of terrestrial origin. Sixty-three of the isolated actinomycetes were tested for the effects of seawater on growth. Streptomycete growth in nonsaline media was reduced by 39% compared with that in seawater. The actinoplanetes had a near obligate requirement of seawater for growth, and this is presented as evidence that actinomycetes can be physiologically active in the marine environment. Problems encountered with the enumeration of actinomycetes in marine sediments are also discussed.  相似文献

Summary Actinomycetes were isolated from marine sediments collected in Sandy Hook Bay, New Jersey. A variety of pretreatments, including heat and phenol, were employed to improve the recovery of these microorganisms. In addition, plating of sediment samples on chitin agar, and filtration through cellulose membrane filters were also utilized. These pretreatments eliminated or severely curtailed the growth of contaminating microorganisms thereby facilitating the isolation of actinomycetes. A total of 120 isolates was obtained, of which 19 displayed significant antimicrobial activity. Most of the activity was directed against gram-positive bacteria, but inhibition of gram-negative species and a yeast were also evident.  相似文献

Marine sediments from Newfoundland, Canada were explored for biotechnologically promising Actinobacteria using culture-independent and culture-dependent approaches. Culture-independent pyrosequencing analyses uncovered significant actinobacterial diversity (H′—2.45 to 3.76), although the taxonomic diversity of biotechnologically important actinomycetes could not be fully elucidated due to limited sampling depth. Assessment of culturable actinomycete diversity resulted in the isolation of 360 actinomycetes representing 59 operational taxonomic units, the majority of which (94 %) were Streptomyces. The biotechnological potential of actinomycetes from NL sediments was assessed by bioactivity and metabolomics-based screening of 32 representative isolates. Bioactivity was exhibited by 41 % of isolates, while 11 % exhibited unique chemical signatures in metabolomics screening. Chemical analysis of two isolates resulted in the isolation of the cytotoxic metabolite 1-isopentadecanoyl-3β-d-glucopyranosyl-X-glycerol from Actinoalloteichus sp. 2L868 and sungsanpin from Streptomyces sp. 8LB7. These results demonstrate the potential for the discovery of novel bioactive metabolites from actinomycetes isolated from Atlantic Canadian marine sediments.  相似文献

Actinomycetes are one of the most efficient groups of secondary metabolite producers and are very important from an industrial point of view. Among its various genera, Streptomyces, Saccharopolyspora, Amycolatopsis, Micromonospora and Actinoplanes are the major producers of commercially important biomolecules. Several species have been isolated and screened from the soil in the past decades. Consequently the chance of isolating a novel actinomycete strain from a terrestrial habitat, which would produce new biologically active metabolites, has reduced. The most relevant reason for discovering novel secondary metabolites is to circumvent the problem of resistant pathogens, which are no longer susceptible to the currently used drugs. Existence of actinomycetes has been reported in the hitherto untapped marine ecosystem. Marine actinomycetes are efficient producers of new secondary metabolites that show a range of biological activities including antibacterial, antifungal, anticancer, insecticidal and enzyme inhibition. Bioactive compounds from marine actinomycetes possess distinct chemical structures that may form the basis for synthesis of new drugs that could be used to combat resistant pathogens.  相似文献

Actinomycetes could be isolated efficiently on a defatted wood powder medium from the guts of various species of termites. The actinomycete flora in the termites' guts depended largely on the area in which the termites naturally occur. In termites from the same area, the actinomycete flora changed depending on the taxonomic difference between termite species. Some actinomycetes isolated from termites' guts grew satisfactorily on lignin-related media, and the others grew on cellulose-related media.  相似文献

Recent findings from culture-dependent and culture-independent methods have demonstrated that indigenous marine actinomycetes exist in the oceans and are widely distributed in different marine ecosystems. There is tremendous diversity and novelty among the marine actinomycetes present in marine environments. Progress has been made to isolate novel actinomycetes from samples collected at different marine environments and habitats. These marine actinomycetes produce different types of new secondary metabolites. Many of these metabolites possess biological activities and have the potential to be developed as therapeutic agents. Marine actinomycetes are a prolific but underexploited source for the discovery of novel secondary metabolites.  相似文献

Alachlor (2-cloro-N-(methoxymethyl)-N-(2,6-diethylphenyl)-acetamide) is an extremely toxic and highly mobile herbicide that is widely used for pre-emergence control of grasses and weeds in many commercial crops in Brazil. In order to select soil actinomycetes able to degrade this herbicide, fifty-three actinomycete strains were isolated from soil treated with alachlor using selective conditions and subjected to in vitro degradation assays. Sixteen isolates were shown to be tolerant to high concentrations of the herbicide (up to 720 mg L-1), and six of these were able to grow and degrade 50 alachlor (72 mg L-1) in mineral salts medium. Morphological and phylogenetic analysis enabled the assignment of the alachlor-degrading strains to the genus Streptomyces. Strain LS151 was related to the type strains of Streptomyces capoamus/Streptomyces galbus, whereas strains LS143 and LS153 were related to Streptomyces bikiniensis. The remaining strains, LS166, LS177 and LS182, were similar in morphological features and recovered in a single cluster based on 16S rDNA sequence analysis, but shown to be distinct on the basis of genomic fingerprint data (rep-PCR). Though a definitive taxonomic assignment of alachlor-degrading strains was not possible, these data indicate that ability to degrade this pesticide was detected in different Streptomyces taxa.  相似文献

We have undertaken a study to estimate the species diversity of zoosporogenous actinomycetes that can be isolated from an arid environment. The study site encompassed an area of approximately 22 000 square kilometers of the Mojave Desert along the California-Nevada border. A series of 29 soil samples was collected along two intersecting transects of approximately 190 and 240 km which traversed a number of distinct ecosystems. A0 horizon soils were collected from the rhizosphere of the predominant vegetation at each sampling site and screened for the target genera using selective isolation techniques: chemoattraction (xylose and -collidine) and baiting with hair. Following incubation of primary isolation plates for 28 days at 28°C, all colonies that exhibited filamentous growth, presence of sporangia and/or motile spores upon direct microscopic observation (450 and 1000×) were further characterized by fatty acid analysis (FAME). Most of the isolates fell into three broad clusters that roughly correlated with presumptive genus assignments. Individual isolates could be assigned to 226 FAME biotypes based on chromatographic similarity (85%). The dominant species (514/826 isolates) belong to a previously undescribed taxon that morphologically resemblesGeodermatophilus but possesses unique FAME profiles that include at least three novel lipids. The remainder of the isolates were species ofActinoplanes, indeterminate species or vagrant isolates ofStreptomyces.  相似文献

An improved membrane filtration procedure for use on board ship to enumerate Escherichia coli and Group D faecal streptococci in marine sediments is described. Ultrasonication extraction combined with resuscitation of sublethally-injured cells yielded significantly higher counts of E. coli than sediments shaken by hand. Counts of E. coli were also higher on mFC agar (without rosalic acid) after a period of resuscitation on tryptone-soy agar supplemented with 0.1% yeast extract than on a 4% Teepol-lactose medium. Ultrasonication of sediments made no significant difference to counts of Group D faecal streptococci on KF-streptococcus agar. These improved isolation procedures allowed better discrimination of the area affected by sewage sludge at a disposal site off the northeast coast of England.  相似文献

Ten actinomycetes isolated from various marine sediments were investigated for the production of tetrodotoxin (TTX). Tissue culture assay, high performance liquid chromatography and gas chromatography-mass spectrometry confirmed that the nine strains produced TTX. Actinomycetes may be responsible for TTX accumulation in marine environments.  相似文献
