The major constrains for practical exploitation of the somatic hybrids between eggplant and its wild relatives have been their sterility and tetraploidy which prevented their incorporation into breeding programs. Here we demonstrate that anther culture was successfully utilized to bring back the ploidy level to the diploid status in tetraploid interspecific hybrids between eggplant and the allied species S. integrifolium and S. aethiopicum gr. gilo. Both the relative species are resistant to Fusarium oxysporum f. sp. melongenae and to some strains of bacterial wilt (Ralstonia solanacearum) which are very destructive diseases of eggplant. Dihaploid androgenetic plants were obtained from the somatic hybrids, from the “double somatic hybrid” obtained by sexual cross of the two somatic hybrids [(eggplant + S. aethiopicum) × (eggplant + S. integrifolium)], and from tetraploid backcrossed plants between the somatic hybrid with S. aethiopicum and eggplant. Phenotypical, molecular, biological and biochemical characterization, and also artificial inoculation with Fusarium oxysporum are consistent with a recombination between the genomes of the species involved in the hybridizations. Dihaploids resistant to Fusarium were successfully backcrossed with eggplant. Besides their utility as potential valuable breeding materials, the introgressed lines obtained may be utilized in genetic and molecular studies about the resistance to Fusarium from S. integrifolium and S. aethiopicum gr. gilo.  相似文献

We studied (a) the extent adhesion of Penicillium oxalicum conidia to tomato roots after application of P. oxalicum conidial formulations with or without stickers, (b) the relationship between the extent of conidial adhesion to roots and biocontrol of the conidial formulations against tomato wilt, and (c) colonisation of roots by P. oxalicum. Adhesion of P. oxalicum conidia to tomato roots occurred within the first minute of contact between the root and the conidial formulation and the bonding strength was sufficiently strong to prevent conidial removal from the roots. In addition, some formulations with stickers that increased conidial adhesion to roots improved the biocontrol of tomato wilt, when compared to that of formulations without stickers. A “dried conidia without stickers” with 0.025% Nu-Film 17 had no effect on the biocontrol of tomato wilt, despite good adherence of the conidia to the roots. The numbers of P. oxalicum conidia that adhered to the roots was constant for 60 days after application of a “dried conidia without stickers” conidial formulation. The significance of these results (speed of adhesion, number of adhered conidia, and variability of conidial external surface) are discussed in relation to the biocontrol success of tomato wilt using different types of conidial formulations with and without stickers.  相似文献

Summary Eggplant (Solanum melongena L. cv. Violetta lunga 2) cotyledon expiants grown on hormone-free medium (controls) or on medium containing either naphthaleneacetic acid alone (root forming) or in combination with zeatin riboside (shoot forming) showed minor differences in free polyamine titres during culture. In contrast, conjugated polyamines (particularly those in the trichloroacetic acid-soluble fraction) accumulated only in hormonetreated explants, but not in controls. The extent and the temporal changes in soluble-conjugate levels differed between root-forming and shoot-forming expiants; in the former, accumulation began earlier (within 1 day of culture) and reached the highest levels. In both organogenic programmes, maximum conjugate accumulation occurred just before and during organ emergence. Adventitious roots and shoots were formed along the cut surfaces. The regions closest to these (borders) displayed a significantly higher ratio of conjugated to free spermidine and/or putrescine than the nonorganogenic regions (centres) of the explant. Ornithine decarboxylase activity was higher than arginine decarboxylase activity both in control and hormone-treated explants. However, both activities increased markedly on day 2 of culture in the presence of hormones. Thereafter ornithine decarboxylase activity remained high in shoot-forming explants, but not in root-forming ones. Putrescine oxidising activity was also enhanced by exogenously supplied hormones starting from day 4 of culture. This activity remained high up to day 12 in the presence of auxin plus cytokinin, whereas it peaked on day 6 in auxin-treated explants. Spermidine oxidising activity was the only enzyme activity which was consistently higher in controls than in hormone-treated tissue. Differences between the two organogenic programmes with respect to temporal changes in polyamine content, and putrescine biosynthetic and oxidative activities are discussed in relation to the timing of organ formation. Two strains of xylose-containing and Q-10-having ballistoconidiogenous yeasts isolated from plant leaves collected in Taiwan were found to represent two new species of the genus Bullera. In the phylogenetic trees based on the sequence analysis of 18S rDNA and D1/D2 domain of 26S rDNA, these species are located in the Bullera variabilis (Bulleribasidum) cluster in Hymenomycetes. They are described as Bullera begoniae sp. nov. and Bullera setariae sp. nov., respectively.  相似文献

Crown rust of oats (Puccinia coronata f. sp. avenae) was investigated as a biocontrol agent for wild oats (Avena fatua) on San Clemente Island, California. Successful restoration of the native habitats of this island will involve the reduction of wild oats and revegetation with native grasses such as stipa (Nassella pulchra). Determination of the outcome of interference between wild oats and stipa is important in the prediction of the success of the biocontrol agent. An addition series design was used to investigate these interactions, with densities of each species ranging from 0 to 2000 seeds per m2. Eight replicates were established, four of which were randomly chosen and infected with the pathogen. After 75 days, plant height, dry weight, and seed production were measured. The data were fit to a hyperbola surface model by use of a nonlinear regression procedure. Results indicate that wild oat is the superior competitor at the seedling stage; however, early rust infection greatly reduces fitness of wild oats, causing an increased fitness for stipa. Simulations with a plant community model constructed from the results of the greenhouse experiment and information in the literature indicated that an equilibrium may be established between wild oat and stipa if high initial seeding rates of stipa are used in revegetation.  相似文献

Plant cell walls play a major role in the outcome of host-parasite interactions. Wall fragments released from the plant, and/or the fungal pathogen, can act respectively as endogenous and exogenous elicitors of the defence response, and other wall components, such as callose, lignin, or hydroxyproline-rich glycoproteins, can inhibit pathogen penetration and/or spreading. We have previously demonstrated that calli from tomato cultivars resistant in vivo to Fusarium oxysporum f.sp. lycopersici show a high amount of polysaccharides in vitro. The aim of the present work was to assess the possible role of polysaccharide content and/or synthetic capacity in determining the competence of plant cells for active defence. For this purpose, tomato cell clones with increased and decreased polysaccharide (FL+, FL-) and callose (A+, A-) content have been selected by means of specific stains as visual markers and tested for the effect of these changes on the extent of response to Fusarium. The analysis of several parameters known to be indicative of active defence (cell browning after elicitor treatment, peroxidase and -glucanase induction and inhibition of fungal growth in dual culture) clearly shows that FL+ and A+ clones have acquired an increased competence for the activation of defence response. The results are thoroughly discussed in terms of an evaluation of the relative importance of constitutive and/or inducible polysaccharide synthetic capacity for plant response to pathogens, and their possible regulation by plant physiological backgrounds.  相似文献

The leaf oils of both subspecies of Atherosperma moschatum have been investigated to determine if there were any chemical differences between the two subspecies. While both subspecies of A. moschatum contained methyl eugenol (55–87%), safrole (5–20%) and linalool (1–6%), camphor (5–10%) was found to be only in the oils of A. moschatum subsp. moschatum and to be either absent or, at most, less than 0.5% in the oils of A. moschatum subsp. integrifolium. There was also a range of monoterpene hydrocarbons and 1,8-cineole, all in the range of 0.1–5%, that were present in subspecies moschatum but virtually absent in subspecies integrifolium.  相似文献

Hyptis suaveolens L. (Poit.) essential oil was tested in vitro on the growth and morphogenesis of Fusarium oxysporum f.sp. gladioli (Massey) Snyder & Hansen, which causes Fusarium corm rot and yellows in various susceptible cultivars of gladiolus. The fungitoxicity of the oil was measured by percentage radial growth inhibition using the poisoned food technique (PF) and volatile activity assay (VA). The mycelial growth of the test fungus was completely inhibited at 0.998 and 0.748 μg ml−1 concentration of oil in PF and VA, respectively. Essential oil was found to be fungicidal in nature at 1.247 and 0.998 μg ml−1 concentration of oil in PF and VA, respectively. Determination of conidial germination in the presence of oil was also carried out and it was found that the oil exhibited 100% inhibition of conidial germination at 0.450 μg ml−1 concentration. The effect of essential oil on the yield of mycelial weight was observed and it was found that at 0.873 μg ml−1 concentration no mycelium was recorded and 100% inhibition was observed. The fungitoxicity of oil did not change even on exposure to 100°C temperature or to autoclaving, and the oil also retained its fungicidal nature even after storage of 24 months. The main changes observed under light microscopy after oil treatment were a decrease and loss of conidiation and anomalies in the hyphae such as a decrease in the diameter of hyphae and granulation of cytoplasm. The treatment of the oil also showed highly reduced cytoplasm in the hyphae, showing clear retraction of the cytoplasm from the hyphae and ultimately in some areas hyphae without cytoplasm were also found. GC-MS studies of the essential oil revealed that the oil consisted of 24 compounds with 1,8-cineole as major component accounting for 44.4% of the total constituents.  相似文献

Four xylanases were purified, two from the termite Macrotermes bellicosus workers (X1T and X2T) and two from its symbiotic fungus Termitomyces sp. (X1Mc and X2Mc). The analysis of the step required for the purification of X1T and X1Mc and the comparison of their different properties suggested that xylanases X1T and X1Mc were the same enzyme, X1. The determination of the reducing sugars by TLC revealed that X1 was an endoxylanase (EC 3.2.1.8) and X2T and X2Mc were exoxylanases (EC 3.2.1.37). The apparent molecular weights of the three xylanases, determined by SDS-polyacrylamide gel electrophoresis, were 36 kDa for X1, 56 kDa for X2T and 22.5 kDa for X2Mc. The optimal pH of the three xylanases was 5.5, and Km values determined with birchwood xylan as substrate were 0.2% for X1, 0.1% for X2T and 0.3% for X2Mc, showing a high affinity for this substrate. Fungi present the ability to hydroxylate steroids. In some filamentous fungi, progesterone induces an enzyme system which converts the compound into a less toxic hydroxylated product. We investigated the progesterone response in the vascular wilt pathogen Fusarium oxysporum, using mass spectrometry and high performance liquid chromatography (HPLC). Progesterone was mainly transformed into 15α-hydroxyprogesterone, which was found predominantly in the extracellular medium. The role of two conserved fungal signaling cascades in the induction of the progesterone-transforming enzyme system was studied, using knockout mutants lacking the mitogen-activated protein kinase Fmk1 or the heterotrimeric G-protein β subunit Fgb1 functioning upstream of the cyclic adenosine monophosphate (cAMP) pathway. No steroid hydroxylation was induced in the Δfgb1 strain, suggesting a role for the G-protein β subunit in progesterone signaling. Exogenous cAMP restored the induction of progesterone-transforming activity in the Δfgb1 strain, suggesting that steroid signaling in F. oxysporum is mediated by the cAMP-PKA pathway.  相似文献

Two hundred and forty-two actinomycete strains were isolated from the interior of leaves and roots of healthy and wilting banana plants. Most of them were streptomycetes, Streptomyces griseorubiginosus-like strains were the most frequently isolated strains. Community analysis demonstrated increased actinomycete diversity in wilting leaves compared to that in healthy leaves, similar actinomycete communities were found in wilting and healthy roots. Screening of the isolates for antagonistic activity against Fusarium oxysporumf. sp. cubenserevealed that the proportion of antagonistic streptomycetes in healthy roots was higher than that in wilting roots (P < 0.01), but no difference was found between antagonistic strains isolated from healthy and wilting leaves. The potential biological control of Panama disease of banana by endophytic streptomycetes, especially Streptomyces griseorubiginosus-like strains was discussed.  相似文献

The most important factor concerning wild cat populations is the loss of habitat. Therefore, it is necessary to assess the size of the home ranges of wild and domestic cats along with the features of these areas (vegetation, elevation, proximity to human settlement, etc.). A total of 16 wildcats and 19 domestic cats were caught and fitted with radio collars within the period between 1989–1993. It was possible to analyze the radiotelemetry data of 4 wildcats and 3 domestic cats. It resulted that the wildcats occupied larger home ranges than the domestic cats, however, there were exceptions. Home range size variability was extensive in both species. The males occupied larger areas than the females. This was most likely due to the reproductional wandering of males into female home ranges. Also the overlap between the home ranges of males was larger than that of females. However, there were very small overlaps between the core areas. No cats used the same sites at the same time. This indicates that the home ranges of cats exist only in space and time as well. Although these animals are solitary, there was some indication that hierarchy exists between males.  相似文献

(Z)-3-nonenol, (Z,Z)-3,6-nonadienol and (S,S)-(-)-epianastrephin proved active as male-produced pheromones which elicited behavioral responses from virgin female Mexican fruit flies (Anastrepha ludens) (Diptera: Tephritidae) in laboratory bioassays. All three chemicals elicited attraction and/or locomotor arrest when tested individually. When tested together, (Z)-3-nonenol inhibited the behavioral effect of (Z,Z)-3,6-nonadienol but either of the alcohols synergized the effect of (S,S)-(-)-epianastrephin. Quantities of the pheromones per male abdomen were: (Z)-3-nonenol-100 ng; (Z,Z)-3,6-nonadienol-40 ng; and total epianastrephin (R,R and S,S enantiomers)-700 ng. Relative to the quantities in abdominal extract, males released these chemicals during sexual display in a blend containing a higher proportion of the alcohols.
Résumé Le (Z)-3-nonènol, le (Z,Z)-3,6-nonadiènol et le (S,S)-(-)-épianastréphine produits par les mâles de la mouche mexicaine du fruit (Anastrepha ludens) (Diptera: Tephritidae) ont attirés les femelles lors de tests conduits en laboratoire. Le pouvoir attractif du (Z)-3-nonènol est faible comparé à celui des deux autres phéromones. Evalués ensemble, le (Z)-3-nonènol inhibe l'attractivité du (Z,Z)-3,6-nonadiènol mais chacun des deux alcools synergise l'effet attractif de la (S,S)-(-)-épianastréphine. Les quantités recouvrées par mâle dans un extrait abdominal étaient respectivement de 100, 40 et 700 ng pour le (Z)-3-nonènol, le (Z,Z)-3,6-nonadiènol, et l'épianastréphine totale (R,R et S,S) énantiomères. Comparées à ces valeurs, le mélange élange émis par les mâles lors de leur cour sexuelle est plus riche in alcools. Le rapport (Z)-3-nonènol (Z,Z)-3,6-nonadiènol est cependant identique dans l'extrait abdominal et dans le mélange émis par les mâles.

Peristenus sp. pupae collected from Lygus spp. nymphs in 2001 and 2002 were over-wintered in the laboratory. In both years, more than 30% of adults emerging from over-wintering pupae were identified as ichneumonid hyperparasitoids, Mesochorus curvulus Thomson and Meschorus sp. (Hymenoptera: Ichneumonidae). At the end of the over-wintering period, Peristenus sp. males emerged first followed by Peristenus sp. females and finally Mesochorus spp. The male:female ratio in emerging Peristenus sp. adults was skewed towards males. The Internal Transcribed Spacer (ITS) region and the cytochrome oxidase I (COI) gene from Mesochorus spp. were sequenced. ITS sequences were used to develop PCR primers to detect Mesochorus spp. hyperparasitism in the primary host, Lygus spp. PCR analysis of field-collected Lygus spp. nymphs gave similar estimates of Mesochorus spp. hyperparasitism to the rearing protocols (25–28%). Sequence analysis of COI and ITS regions and subsequent restriction endonuclease analysis of ITS PCR products from Mesochorus spp. indicate the presence of two genotypes in the population. The possibility that these two genotypes represent separate or cyrptic species is discussed.  相似文献

A new species, Epistemma neuerburgii is described. This species was discovered in the Nyungwe National Park, Rwanda, Tropical Africa during a field investigation of epiphytes in montane rainforest. It was, however, also found in Uganda, in similar environment. E. neuerburgii is the fourth species described for Epistemma, but the other three species are found about 2000 km to the west in Cameroon, Gabon, Ghana, Ivory Coast and Nigeria. E. neuerburgii is an epiphytic climber distinguished by fairly large lanceolate leaves and conspicuous reddish-purple flowers with corolline white, densely hairy corona lobes.  相似文献

Seventy-seven red deer (Cervus elaphus hippelaphus), 40 roe deer (Capreolus capreolus) from the Northwestern (NW) Alps (Turin Province, NW Italy) and 29 roe deer from the NW Apennines (Alessandria province, NW Italy) were examined for the presence of Mycobacterium avium subsp. paratuberculosis (MAP) by culture, IS900 nested polymerase chain reaction (PCR) and IS1311 PCR restriction endonuclease analysis for strain characterisation. MAP identification (nested PCR and/or culture) allowed us to detect 32.9% MAP-infected red deer and 22.5% infected roe deer in the NW Alps and 41.4% MAP infected roe deer in the NW Apennines. On the basis of the polymorphism present in the IS1311 sequence, all MAP isolates were characterised as cattle strains. Our results show that MAP circulates widely among populations of wild cervids in NW Italy.  相似文献

Stable isotopes in tree rings have widely been used for palaeoclimate reconstructions since tree rings record climatic information at annual resolution. However, various wood components or different parts of an annual tree-ring may differ in their isotopic compositions. Thus, sample preparation and subsequent laboratory analysis are crucial for the isotopic signal retained in the final tree-ring isotope series used for climate reconstruction and must therefore be considered for the interpretation of isotope–climate relationships. This study focuses on wood of Corsican Pine trees (Pinus nigra ssp. laricio) as this tree species allows to reconstruct the long-term climate evolution in the western Mediterranean. In a pilot study, we concentrated on methodological issues of sample preparation techniques in order to evaluate isotope records measured on pooled whole tree-ring cellulose and whole tree-ring bulk wood samples. We analysed 80-year long carbon and oxygen chronologies of Corsican Pine trees growing near the upper tree line on Corsica. Carbon and oxygen isotope records of whole tree-ring bulk wood and whole tree-ring cellulose from a pooled sample of 5 trees were correlated with the climate parameters monthly precipitation, temperature and the self-calibrating Palmer Drought Severity Index (sc-PDSI). Results show that the offsets in carbon and oxygen isotopes of bulk wood and cellulose are not constant over time. Both isotopes correlate with climate parameters from late winter and summer. The carbon and oxygen isotope ratios of cellulose are more sensitive to climatic variables than those of bulk wood. The results of this study imply that extraction of cellulose is a pre-requisite for the reconstruction of high-resolution climate records from stable isotope series of P. nigra ssp. laricio.  相似文献

Major cell wall-bound phenolic compounds were detected and identified in roots of tomato at different stages of growth. Alkaline hydrolysis of the cell wall material of the root tissues yielded ferulic acid as the major bulk of the phenolic compounds. Other phenolic compounds identified were 4-hydroxybenzoic acid, vanillic acid, 4-hydroxybenzaldehyde, vanillin and 4-coumaric acid. All the six phenolic acids were higher in very early stage of plant growth. Ferulic acid, 4-hydroxybenzoic acid and 4-coumaric acid exhibited a decreasing trend up to 60 days and then the content of these phenolic acids increased somewhat steadily towards the later stage of growth. Total phenolics, phenylalanine ammonia-lyase (PAL) activity and peroxidase (POD) activity were in tandem match with the occurrence pattern of the phenolic acids. Ferulic acid showed highest antifungal activity against tomato wilt pathogen Fusarium oxysporum f. sp. lycopersici. The results of this study may be interpreted to seek an explanation for high susceptibility of tomato plants at flowering stage to Fusarium wilt. It may also be concluded that greater amounts of ferulic acid in combination with other phenolics and higher level of PAL and POD activities after 60 days of growth may have a role in imparting resistance against Fusarium wilt at a late stage of plant growth.  相似文献

Summary  A new genus, Cleistesiopsis, was segregated from Cleistes based on morphological and molecular characteristics, and two new species of Cleistes: C. batistana and C. elongata, both occurring in the Brazilian Central Plateau (Central-Western Brazil), are described and illustrated. Furthermore, a key to genera currently recognised within Pogonieae is presented.
