The efficacy of submicron chitosan dispersions (SCD) on Alternaria alternata rot of the ‘Lingwu changzao’ jujube (Zizyphus jujuba Mill. cv. Lingwu changzao) fruit and possible mechanisms involved were investigated in this study. When jujube fruit at the preharvest stage were sprayed twice with SCD (10 mg/ml) water solution and then natural infections were allowed to develop, there was a significant reduction in disease incidence and lesion index compared to control fruit during storage period, and similar results were obtained from postharvest fruit inoculated with the Alternaria rot pathogen. Scanning electron microscopy of the pathogen revealed that hyphae and spores were damaged and deformed when growing on the surface of fruit pretreated with SCD or CHO. The slowed softening of jujube fruit resulted from the protopectin content increasing compared with the control. Furthermore, SCD sprays inhibited cell wall hydrolysing enzymes, pectinase activity was decreased, and the increase in cellulase activity was postponed (from 30 days to 45 days).It was concluded that preharvest application of SCD was a more effective alternative to conventional chitosan for preventing postharvest from Alternaria rot of ‘Lingwu changzao’ jujube fruit.  相似文献

The potential of using an antagonistic yeast alone or in combination with microwave treatment for controlling blue mould rot of jujube fruit, and its effect on postharvest quality of fruit, was investigated. The results showed that the growth of Penicillium citrinum was completely inhibited by a 2450‐MHz microwave heating for 2 or more minutes in vitro. The population density of P. citrinum in surface wounds of fruit treated with microwave treatment for 2–3 min was significantly lower than that of controls. When tested on jujube fruit, antagonistic yeast or microwave treatment, as stand‐alone treatment, the disease incidence of infected wounds was reduced from 100% to 45.0% and 36.0%, and lesion diameters were reduced from 1.92 cm to 1.50 cm and 1.38 cm, respectively. However, in fruit treated with a combination of Metschnikowia pulcherrima and microwave treatment, the disease incidence of infected wounds and lesion diameters was only 21.0% and 1.00 cm, respectively. The natural decay incidence on jujube fruit treated with the combination of microwave treatment and M. pulcherrima was 6.2% after storage at 2 ± 1°C for 45 days and at 22°C for 7 days. None of the treatments impaired quality parameters of fruits. Thus, the combination of microwave treatment and M. pulcherrima could provide an alternative to synthetic fungicides for controlling postharvest blue mould rot of jujube fruit.

The causal agent of husk rot of macadamia is often attributed to Colletotrichum gloeosporioides sensu lato. However, in recent husk rot outbreaks, the characteristic concentric ring of pycnidia of C. gloeosporioides that is associated with the disease was often absent. Due to its sporadic occurrence, the importance of husk rot is often underrated and attributed to environmental and physiological factors. In order to determine the significance, prevalence and factors that influence husk rot in macadamia, this study examined the aetiology of husk rot in Australia. The relative incidence and severity of husk rot was evaluated in several macadamia orchards over eight consecutive years. Pathogenicity assays were developed to confirm the identity of the causal agent. A range of fungi from several genera including: Diaporthe, Lasiodiplodia, Colletotrichum, Pestalotiopsis, Aspergillus, Alternaria, Nigrospora and Epicoccum were isolated from samples of macadamia pericarps with husk rot symptoms from different orchards. Fungi in the genus Diaporthe were most frequently isolated, often from symptomatic fruit. Results from pathogenicity trials showed the characteristic soft or spongy black lesions characteristic of husk rot symptoms in wounded fruits that were incubated with the diseased fruit or inoculated with a conidial suspension of Diaporthe spp. Our results suggest that injury to the macadamia fruit pericarp not only predisposes the pericarp to pathogen infection but it is a prerequisite for infection. Large variations in husk rot severity were observed over years. Husk rot severity was linked to days after anthesis and was associated with mean weekly relative humidity and minimum temperatures. This study confirmed that Diaporthe species cause husk rot in macadamia, hence, a rationale for adopting Phomopsis husk rot as the name of the disease is discussed.  相似文献

Alternaria rot of Pingguoli pear occurred after latent infection. Fruit surfaces were asymptomatic within 60 days storage under cold condition (0°C, RH 85–90%), but black‐grey hyphae could be seen in the lenticels or calyx tube of Pingguoli pear after 90 days of storage. The tissue collapsed and resulted in visible black spots as the hyphae spread over the fruit. Average incidence of Alternaria rot of fruits from an orchard in Gansu was 28.86% at 100 days of storage. The main fungus isolated from the Alternaria rot on stored Pingguoli pear was identified as Alternaria alternata (Fr. : Fr.) Keissl. This pathogen was able to initially infect the fruit via two pathways during the growing season, and then remain in a latent state. The fungus first colonized the styles at the full‐blossoming stage, and then grew into the carpel cavities progressively after 50 days from petal fall. The percentage latent infection of A. alternata was up to 45% in the carpel cavity until the harvest time. The fungus also attacked fruit surfaces and remained latent in the fruit peel during fruit development. The percentage of A. alternata latent infection at the calyx end, middle part and stem end of the fruit peel was 40%, 24% and 42.8%, respectively, at harvest time.  相似文献

In the year 2000, a fungus was isolated from rotting lemon (Citrus limon) fruit in the Kahnouj region, south‐eastern Iran and subsequently identified as Ceratocystis radicicola (anamorph: Chalara sp.). A pathogenicity test was carried out to fulfil Koch's postulates. The pathogen could only enter the ripen lemon fruit and cause the rot disease through the wounds and cracks. The fungus was also pathogenic on fruit of other citrus species as well as date palm (Phoenix dactylifera). Symptom development was slow on fruits on the tree but accelerated after fruit harvest.  相似文献

We examined the phytopathological and biological characters ofBotryosphaeria spp. isolated from apples and other deciduous fruit trees, and determined the nucleotide sequences of their rDNA ITS regions. TheBotryosphaeria isolates from deciduous fruit trees can be divided into three groups based on their production of warts on twigs, size of the conidia, and nucleotide sequences of rDNA ITS 1, ITS 2 and 5.8S rDNA. Isolates ofBotryosphaeria in ITS group A produced conidia of intermediate size and showed warts on infected twigs prior to the development of ring rot on fruit. This group was common on deciduous fruit trees in Japan as a causal agent of ring rot and wart bark diseases of apples and pears; and it appears similar to theB. dothidea from the US that was isolated from apple exhibiting white rot. The ITS group BBotryosphaeria produced small conidia and induced shoot blight without wart development prior to the development of ring rot on fruit. This group was localized on pear, persimmon, and kiwi fruit in restricted areas of Japan. The ITS group CBotryosphaeria consisted ofB. obtusa, the causal agent of apple black rot in the US, which produced large dark brown conidia.  相似文献

Observations in West Midlands orchards in 1947–50 on the apple varieties Cox's Orange Pippin and Laxton's Superb showed that brown rot was often associated with shallow holes in the fruit. Laboratory experiments with undamaged apples proved that earwigs ( Forficula auricularia ) could cause this type of injury.
Bands of sacking soaked in BHC placed around trunks of Laxton's Superb trees in 1949 and 1950 trapped many earwigs; less brown rot occurred in the banded than on control trees. A survey of picked fruit in 1950 showed close correlation between degree of earwig damage and amount of brown rot.
Earwig damage was appreciable in fruit from ten out of thirteen grass orchards, but was negligible in fruit from nine arable plantations.  相似文献

The postharvest development of crown rot of bananas depends notably on the fruit susceptibility to this disease at harvest. It has been shown that fruit susceptibility to crown rot is variable and it was suggested that this depends on environmental preharvest factors. However, little is known about the preharvest factors influencing this susceptibility. The aim of this work was to evaluate the extent to which fruit filling characteristics during growth and the fruit development stage influence the banana susceptibility to crown rot. This involved evaluating the influence of (a) the fruit position at different levels of the banana bunch (hands) and (b) changing the source–sink ratio (So–Si ratio), on the fruit susceptibility to crown rot. The fruit susceptibility was determined by measuring the internal necrotic surface (INS) after artificial inoculation of Colletotrichum musae. A linear correlation (r = −0.95) was found between the hand position on the bunch and the INS. The So–Si ratio was found to influence the pomological characteristics of the fruits and their susceptibility to crown rot. Fruits of bunches from which six hands were removed (two hands remaining on the bunch) proved to be significantly less susceptible to crown rot (INS = 138.3 mm 2) than those from bunches with eight hands (INS = 237.9 mm 2). The banana susceptibility to crown rot is thus likely to be influenced by the fruit development stage and filling characteristics. The present results highlight the importance of standardising hand sampling on a bunch when testing fruit susceptibility to crown rot. They also show that hand removal in the field has advantages in the context of integrated pest management, making it possible to reduce fruit susceptibility to crown rot while increasing fruit size.  相似文献

Two bacterial strains (ZJ01 and ZJ02) isolated from the phyllosphere of Chinese jujube were tested for antifungal activities and morphological mechanisms against Phoma destructiva, Alternaria alternata and Fusicoccum spp., three key pathogens of jujube fruits. Based on their physiological characteristics and 16S rDNA gene sequences, both strains were identified as Bacillus amyloliquefaciens. The disease incidence of jujube fruits sprayed with the fermentation liquor of ZJ01 and ZJ02 on the puncture wounds before inoculation with the spores of those three key pathogenic fungi was 10% or 66.7%, significantly lower than that of Nutrient Broth medium treatment (100%). These results suggest that strains ZJ01 may be a potential biological control agent for Chinese jujube fruit diseases. The antagonistic bacteria could cause five kinds of hyphal alterations of the causal agents, the maximum number ever observed. Of them, hyphal swelling and excessive branching were the dominant phenomenon, the other three varied with the pathogens. The diverse morphological malformation might be helpful to understand the broad antifungal activity of Bacillus spp..  相似文献

The effect of chitosan (0.1 mol/L) combined with sodium silicate (100 mmol/L) treatment on Alternaria rot caused by Alternaria alternata in postharvest jujube fruit (Ziziphus jujuba Mill. cv. Dongzao) was studied. The results showed that chitosan combined with sodium silicate treatment significantly reduced the lesion diameter, decay incidence, red index and weight loss of jujube fruit compared with control samples. Combining treatment increased the ascorbic acid, flavonoids, total phenolic compounds and lignin content. The level of superoxide anion () and hydrogen peroxide of treated samples was also increased compared with the control samples. Meanwhile, the activities of phenylalanine ammonia lyase, polyphenol oxidase, superoxide dismutase, peroxidase, chitinase and β‐1,3‐glucanase were also accumulated in treated jujube samples, while the activity of catalase markedly decreased. These results indicated that chitosan combined with sodium silicate treatment could induce the disease resistance of postharvest jujube. Therefore, coating postharvest jujube using chitosan combined with sodium silicate could promise as a novel method for preventing the disease infection of postharvest jujube.  相似文献

? Premise of the study: A total of 38 polymorphic microsatellite loci of the Indian jujube (Ziziphus mauritiana), an economically important fruit tree, were developed to evaluate genetic diversity and aid in the identification of cultivars. ? Methods and Results: The 38 microsatellite markers were isolated from the Indian jujube using a magnetic bead enrichment method, and polymorphisms were identified in 24 Indian jujube cultivars. The number of alleles ranged from two to 13, with expected heterozygosity ranging from 0.261 to 0.898. The polymorphism information content ranged from 0.248 to 0.889, with a mean of 0.616. Of these 38 simple sequence repeat loci, 20 loci from Z. jujuba (Chinese jujube) were successfully amplified using the simple sequence repeat primer sets. ? Conclusions: These polymorphic loci should be useful in further studies of the genetic diversity and the identification of cultivars of both the Indian jujube and the Chinese jujube.  相似文献

In this study, we found that oxalic acid (OA) at the concentrationof 5 mM could delay jujube fruit sene-scence by reducing ethyleneproduction, repressing fruit reddening and reducing alcoholcontent, which consequently increased fruit resistance againstblue mold caused by Penicillium expansum. In order to gain afurther understanding of the mechanism by which OA delays senescenceand increases disease resistance of jujube fruit, we used aproteomics approach to compare soluble proteome of jujube fruitstreated with water or 5 mM OA for 10 min. A total of 25 differentiallyexpressed proteins were identified by using electrospray ionizationquadrupole time-of-flight tandem mass spectrometry (ESI-Q-TOF-MS/MS).Among these proteins, alcohol dehydrogenase 1, which plays adirect role in ethanol metabolism, was repressed, and the abundancesof three photosynthesis-related proteins was enhanced in jujubefruit after OA treatment. The protein identified as a cystathionineβ-synthase domain-containing protein, which can regulateethylene precursors, was also induced by OA treatment. The activityof 1-aminocyclopropane-1-carboxylic acid synthase was significantlysuppressed in OA-treated jujube fruit. In addition, three proteinsrelated to the defense/stress response were up-regulated byOA, and contributed to the establishment of systemic resistanceinduced by OA in jujube fruits. These results indicated thatOA treatment might affect ethanol and ethylene metabolism, resultingin delaying senescence, and increase resistance of jujube fruitsagainst fungal pathogens.  相似文献

AIMS: To investigate the aetiology of seed and boll rot of cotton grown in South Carolina (SC). METHODS AND RESULTS: Bacteria were isolated from diseased locules of cotton bolls collected in a field in SC, USA and tested for the ability to cause comparable disease symptoms in greenhouse grown cotton fruit. Spontaneously generated rifampicin-resistant (Rif(r)) mutants of the isolates were used in confirmatory pathogenicity tests. Resistance to the antibiotic was both stable and effective in differentiating between an inoculated Rif(r) strain, rifampicin-sensitive contaminants and/or endophytes. A series of inoculation methods was tested at various boll developmental stages and at different fruiting nodes on the plant. Field disease symptoms were reproduced by inoculating bolls at 2 weeks postanthesis with bacterial suspensions ranging from 10(3) to 10(6) CFU ml(-1). Pathogenic isolates were categorized as Pantoea agglomerans on the basis of phenotype testing, fatty acid profiling (similarity index = 0.94), and 16s ribosomal DNA sequence analysis (99% nucleotide identity). CONCLUSIONS: Pantoea agglomerans isolates from field-collected immature, diseased cotton caused comparable infection symptoms in greenhouse produced cotton fruit. SIGNIFICANCE AND IMPACT OF THE STUDY: In 1999, significant yield losses in SC cotton resulted from a previously unobserved seed and boll rot that has since been reported in other southeastern states. This study demonstrated a role of P. agglomerans in producing opportunistic bacterial seed and boll rot of cotton.  相似文献

The crown rot pathogens isolated from banana samples collected from 12 localities in Sri Lanka were Lasiodiplodia theobromae, Fusarium proliferatum and Colletotrichum musae. Fungal pathogens isolated were able to cause crown rot disease alone or in combination. Disease severity was higher when combinations of virulent pathogens were used. Cymbopogon nardus and Ocimum basilicum oils displayed fungicidal activity against C. musae and F. proliferatum between 0.2-0.6% (v/v) in a Poisoned food bioassay. Slightly lower concentrations of the test oils were needed for similar activity during liquid bioassays. The combination of Cymbopogon nardus and O. basilicum oils demonstrated synergistic action during both in-vivo bioassays.  相似文献

Sclerotium rolfsii, a destructive soil pathogen common in tropical soils, was found to cause stipe rot of the fruit bodies ofPleurotus tuber-regium. The rot occurs only when the pathogen is inoculated into the soil before seeding and before primordial emergence. It caused 100% inhibition of primordia and sporophore formation when the soil was inoculated before seeding and highly reduced yield when the soil was inoculated with the pathogen just before primordial emergence. However, no fruit body rot occurred on the mushroom when the soil was inoculated with the pathogen after primordial emergence from the soil.  相似文献
