A new standardized bioassay procedure for testing the chronic toxicity of sediments to Hyalella azteca was developed. Tests were initiated with 0–1 wk old amphipods exposed to sediments from Hamilton Harbour, Toronto Harbour, and Lake Ontario for 4 to 8 weeks. Both survival and growth were significantly reduced in the Hamilton Harbour sediments relative to those from the lake after 4 weeks. Exposures of 8 weeks resulted in greater variability; survival of amphipods in sediments from one of the harbour stations, and growth in sediments from both harbour stations with surviving young were not statistically different from survival and growth in lake sediments. Growth and survival in lake sediments were comparable to cultures grown with cotton gauze and no sediment after 4 weeks, but survival was poorer by week 8. Replication was good in 12 out of 13 tests done in duplicate; the difference in survival between replicates averaged 2.2 animals (20 amphipods/replicate, 4 week exposure). We propose that 4 week exposures of young (0–1 wk old) Hyalella would provide a suitable standardized chronic toxicity test for sediments. Respiration rate measurements were conducted with the marine oligochaete Monopylephorus cuticulatus Baker and Brinkhurst to determine the sublethal toxicity of sediments collected from Puget Sound, Washington. Worms were exposed to elutriates prepared from centrifuged sediment slurries. Standard respiration rates were measured at high dissolved oxygen levels for each sample tested and were compared with control and other test results. A total of 97 sediment samples were tested; 40 samples demonstrated significant respiration effects (elevation or depression). Comparison with results of other tests conducted at the same stations or geographic locations (genotoxicity to fish cells, lethality to sensitive species, reproductive impairment tests) indicated very good agreement on broad scale toxicity patterns at different geographical areas, and that respiration measurements effectively determined sublethal toxicity of sediments. This study represents the first application of respiration measurements as a sublethal toxicity test for field-collected sediments.  相似文献

The River Kolbäcksån system is located in a historical mining and steel works district of central Sweden. Ten years ago, intensive limnological studies indicated that the sediments in many of the lakes of this system were contaminated with metals and oil (grease). More recently a very high toxicity was found in the sediments from some of these lakes in tests with Daphnia magna and Tubifex tubifex. The objective of this study was to determine the toxicity to Daphnia magna of surficial sediments from representative locations (N = 39) within this system of lakes and to look for possible correlations with concentrations of metals (Cd, Cr, Cu, Hg, Ni, Pb, Zn) and oil (total) analysed in parallel samples. These results were also compared with spiked sediment toxicity data generated for these metals and with the bottom fauna at 8 of the 39 locations. Among the metals analyzed, only Cd, Pb and Zn were correlated with whole sediment toxicity. Only for Zn and Cr did the maximum concentrations approach those that were toxic in the spiked sediments. Correlations using single and combined metal contamination and toxicity (additive models) explained 16% of the sediment toxicity (r 2 = 0.16; N = 39). Significant correlations with the bottom fauna in situ (gross abundance, biomass and species richness) were found for whole sediment toxicity, but not for metal contamination (N = 8). The use of the sediment quality triad approach for assessment of sediment quality is exemplified with a numerical normalization procedure for the latter 8 sites.  相似文献

The laboratory testing of bottom sediments (BSs) from the Yenisei River containing different concentrations of technogenic radionuclides, heavy metals, and biogenic elements (N and P) based on aquatic such plants as Elodea canadensis (Canadian waterweed) and Myriophyllum spicatum (Eurasian watermilfoil) has revealed a higher sensitivity of roots to the general quality of BSs than shoots: shoot length (9%) < root length (11%) < root number (15%) in M. spicatum; shoot length (22%) < root length (42%) < root number (44%) in E. canadensis. In contrast to M. spicatum, the growth parameters of roots and shoots in E. canadensis have differed in a significant statistical manner between most BS samples. A reverse correlation has been found between the increase in shoot length and the activity of technogenic radionuclides in BSs, which is mostly significant in E. canadensis (r 2 = 0.90–0.95, p = 0.05). Since the growth of shoots and roots in E. canadensis has turned out to be more sensitive to changes in the quality of BSs than that in M. spicatum, E. canadensis can be considered more prospective for biotesting BSs.  相似文献

Phylogenetic diversity of sediment bacteria in the northern Bering Sea   总被引:2,自引:0,他引:2  
The bacterial diversity in sediments from the northern Bering Sea was investigated by culture-independent approaches. Community fingerprint analysis by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) revealed that sediment at two deep stations (DBSE and DBS1, >400 m in depth) harbored a bacterial community distinct from the sediments collected at shallow stations (<150 m in depth) on the continental shelf. Three 16S rRNA gene clone libraries for sediments collected from shallow to deep water stations (NEC5, DBSE and DBS1, respectively) were established. Sediment collected at the deepest station DBS1 showed the highest diversity index value. Sequences fell into 19 major lineages of the domain Bacteria: Alpha-, Beta-, Gamma-, Delta- and Epsilonproteobacteria, Bacteroidetes, Acidobacteria, Actinobacteria, Firmicutes, Planctomycetes, Nitrospirae, Verrucomicrobia, Chloroflexi, Chlorobi, Spirochaetes, Cyanobacteria (or chloroplasts), and candidate divisions OP8, TM6, and WS3. A small fraction of retrieved sequences (1.8%) did not fall into any taxonomic division. Deltaproteobacteria (30%) was the dominant phylum in the three libraries, followed by Gammaproteobacteria (21%) and Acidobacteria (16%). The percentages of cloned sequences with the highest similarity to reported sequences below 97 and 93% were 48.1 and 24.3%, respectively. A large number of phylotypes affiliated with bacteria that play important roles in the carbon, sulfur, and nitrogen cycles suggest an important link of bacteria to the matter cycling in these subarctic sediments.  相似文献

In May 1998, during the fifty-first voyage on board the research vessel Professor Vodyanitskii, a comparative study was conducted of the species diversity of green and purple sulfur bacteria in the water column of the chemocline zone at deep-sea stations and on the bottom surface of the Black Sea shallow regions. At three deep-sea stations, the accumulation of photosynthetic bacteria in the chemocline zone at a depth of 85–115 m was revealed on the basis of the distribution of potential values of carbon dioxide light fixation. The location of the site of potential carbon dioxide light fixation suggests that the photosynthesis may be determined by the activity of the brown Chlorobium sp., earlier revealed at these depths. Enrichment cultures of brown sulfur bacteria were obtained from samples taken at the deep-sea stations. By morphology, these bacteria, assigned to Chlorobium sp., appear as nonmotile straight or slightly curved rods 0.3–0.5 × 0.7–1.2 µm in size; sometimes, they form short chains. Ultrathin sections show photosynthetic antenna-like structures, chlorosomes, typical of Chlorobiaceae. The cultures depended on the presence of NaCl (20 g/l) for growth, which corresponds to the mineralization of Black Sea water. The bacteria could grow photoautotrophically, utilizing sulfide, but the Black Sea strains grew much more slowly than the known species of brown sulfur bacteria isolated from saline or freshwater meromictic lakes. The best growth of the strains studied in this work occurred in media containing ethanol (0.5 g) or sodium acetate (1 g/l) and low amounts of sulfide (0.4 mM), which is consistent with the conditions of syntrophic growth with sulfidogens. The data obtained allow us to conclude that the cultures of brown sulfur bacteria are especially adapted to developing at large depths under conditions of electron donor deficiency owing to syntrophic development with sulfate reducers. The species composition of the photosynthetic bacteria developing in the bottom sediments of shallow stations differed substantially from that observed at deep-sea stations. Pure cultures of the green Chlorobium sp. BS 1C and BS 2C (chlorobactin as the carotenoid), purple sulfur bacteria Chromatium sp. BS 1Ch (containing spirilloxanthine series pigments), and Thiocapsa marina BS 2Tc (containing the carotenoid okenone) were obtained from samples of sediments at shallow-water stations. Marine sediments from 12 shallow water stations in Raritan Bay, New York were tested for the presence of Acanthamoeba. Eight stations were positive for one or more species of Acanthamoeba, A. castellanii, A. comandoni, A. hatchetti, A. lenticulata, A. polyphaga, A. rhysodes, and Acanthamoeba spp. Isolates that grew at 38–40° C were found at four stations (A. comandoni, A. lenticulata, and two unidentified strains). The two unknown strains were characterized on the basis of morphological features, isoenzyme profiles, and mouse pathogenicity tests. One of the two strains was determined to be a new species and is designated herein as Acanthamoeba stevensoni n. sp., ATCC 50388. Mature cysts were most similar to those of morphological Group II of Pussard & Pons (1977). Acanthamoeba stevensoni n. sp. was isolated from inshore coastal sediments where seawater ranged from 20–30%‰ (ppt.). The sediments supported commercially valuable populations of hard clams, Mercenaria mercenaria, that required depuration prior to sale because of contamination by sewage-associated bacteria.  相似文献

As part of the study of the Upper Great Lakes Connecting Channels sponsored by the U.S. Environmental Protection Agency, the U.S. Fish and Wildlife Service examined the occurrence of Hexagenia nymphs and visible oil in sediments at 250 stations throughout the St. Marys River and the St. Clair-Detroit River system from May 14 to June 11, 1985. The mean density of Hexagenia nymphs per square meter averaged 194 for the total study area, 224 in the St. Marys River, 117 in the St. Clair River, 279 in Lake St. Clair, and 94 in the Detroit River. The maximum density of nymphs ranged from 1,081 to 1,164 m-2 in the three rivers and was 3,099 m-2 in Lake St. Clair. A comparison of nymph density at 46 stations where oil was observed in sediments physically suitable for nymphs showed that densities were lower in oiled sediments (61 m-2) than in sediments without oil (224 m-2). Densities of nymphs were relatively high at only four stations where oil was observed in sediments. The Keweenaw Peninsula in northern Michigan was once a major copper mining area and these mining activities were responsible for depositing tons of tailings in and around the Keweenaw Waterway. In recent years there has been concern about possible toxic effects of the contaminated sediments on aquatic communities in the system. In the fall of 1990, sediments were collected from various locations along the Waterway. Ten-day tests were conducted with the samples using three species of benthic invertebrates that have been proposed as suitable for evaluating the toxicity of freshwater sediments: Hyalella azteca (amphipods), Chironomus tentans (chironomids) and Lumbriculus variegatus(oligochaetes). A number of sediments were toxic to one or more of the three species and, in general, there was good agreement among the tests with regard to identifying toxic samples. Unexpectedly, the relative sensitivity of the three species to the test sediments was not accurately predicted from water-only copper exposures. This indicates that factors modifying exposure, such as different lifestyles and/or varying sensitivity to physico-chemical characteristics of sediments can influence results of sediment toxicity tests.  相似文献

为了明确调水调沙工程长期影响下黄河口近岸沉积物中重金属含量的分布特征及其生态风险,基于2012年黄河口近岸27个站位的表层沉积物样品,通过ICP-MS测定重金属(Zn、Cr、Ni、Pb、Cu、Cd)和砷(As)含量,并运用潜在生态风险指数法(RI)对其进行生态风险评估。结果表明:近岸沉积物中重金属和As的平均含量表现为AsZnCrNiPbCuCd。Cr、Ni、Cu和Pb四种元素的分布规律较为一致,整体呈现出近岸和近黄河口高而远离河口和岸线低的空间分异特征。Ni、Cu、Pb、Zn与粘土均呈极显著或显著正相关(P0.01或P0.05),而Cd、Cr和As与其相关性均未达到显著水平(P0.05)。近岸沉积物中6种重金属和As的平均单项潜在生态风险指数大小顺序整体表现为CdAsNiPbCuCrZn。就潜在生态风险(RI)而言,研究区域18.52%的站位属轻微生态危害,70.07%的站位属中等生态危害,7.41%的站位属强生态危害,Cd和As是造成危害的两种主要元素。近岸沉积物中重金属和As的来源复杂且多样,主要是由于农业化肥使用、海上石油开采和泄漏、化石燃料燃烧以及河口污染物输入所致。对比研究发现,随着调水调沙工程的长期实施,除Cd和As外沉积物中其他重金属含量均呈下降趋势,说明二者的生态风险将会随调水调沙的长期实施呈增加趋势,而其他重金属的生态风险将呈降低趋势,故未来应重点关注近岸沉积物中Cd和As的生态毒理风险。  相似文献

Avian and fish botulism outbreaks have been recorded since 1999 in eastern Lake Erie. These outbreaks are caused by Clostridium botulinum type E, a toxin-producing bacteria that is found in anoxic substrates rich in organic material. We studied the environmental conditions present in eastern Lake Erie during 2002, a year when several botulism outbreaks were observed. We also tested for the presence of C. botulinum type E in lake sediments. Samples were taken at six stations from two sites of different depths in the Dunkirk (New York, USA) area. The depth of the sampling sites influenced physico-chemical and biological processes in the sediments. We used the quantitative polymerase chain reaction (Q-PCR) to quantify the levels of C. botulinum type E in the samples. Sediment samples contained a patchy distribution of type E spore concentrations (from not detectable to 5520 DNA copies/mg). Samples of benthic invertebrates tested positive for C. botulinum type E spores in tissues (Gammarus 2028 DNA copies/mg, oligochaetes 428 DNA copies/mg, chironomids 148 DNA copies/mg and dreissenid mussels 715 DNA copies/mg). Principal components analysis (PCA) from inshore stations indicated that a decrease in dissolved oxygen, pH and redox potential near the sediment was associated to an increase in specific conductance and the type E toxin gene in sediments. We also found that C. botulinum type E spores are present in sediments at different depths and at different times through the ice-free season.  相似文献

Superficial (0 to 2 cm) sediments were sampled from 62 sites in Kattegat and Skagerrak during autumn 1989 and spring 1990, tested for toxicity to Daphnia magna and Nitocra spinipes (Crustacea) and analyzed for heavy metals (Cd, Cr, Cu, Hg, N, Pb, Zn), nutrients (N and P) and organic carbon. Whole sediment toxicity to Nitocra spinipes, expressed as 96-h LC50, ranged from 1.8 to > > 32 percent sediment (wet wt), which is equivalent to 0.63 to 53 percent dry wt. Sediment total metal concentrations (mg kg-1 dry wt) ranged from 0.01 to 0.32 for Cd, 8 to 57 for Cr, 3 to 40 for Cu, 0.03 to 0.86 for Hg, 3 to 43 for Ni, 6 to 37 for Pb and 21 to 156 for Zn. Analyzed concentrations of heavy metals were tested for correlation with whole sediment toxicity normalized to dry wt, and significant correlations (Spearman p<0.05) were found for Cd, Cr, Cu, Hg, and Ni. However, the analyzed concentrations of these metals were below the spiked sediment toxicity of these heavy metals to N. spinipes, except for Cr and Zn for which analyzed maximum concentrations approached the 96-h spiked sediment LC50s. There was no improvement in correlation between the sum of heavy metal concentrations normalized to their spiked toxic concentrations (Toxic Unit approach) and the whole sediment toxicity. Calculated heavy-metal-derived toxicity based on toxic units and whole sediment toxicity ranged from 0.1 to 24 (mean value 2.3 and SD 4.2). Theoretically, a value of 1.0 would explain whole sediment toxicity from measured metal concentrations using this approach. Thus, in spite of the fact that the total concentrations of the heavy metals were sufficient to cause toxicity based on an additive model for most of these sediments, the observed toxicity of the sediments from Kattegat and Skagerrak could not exclusively be explained by the concentrations of heavy metals, except for Cr and Zn at their maximum concentrations. The p53 tumor suppressor protein is involved in a complicated regulatory network, mediating expression of ~1000 human genes. Recent studies have shown that many p53 in vivo binding sites (BSs) reside in transposable repeats. The relationship between these BSs and functional p53 response elements (REs) remains unknown, however. Sandy and organic sediments characterized by different heavy metal binding capacities (HMBC), and contaminated with Copper (Cu), mercury (Hg), or zinc (Zn) were treated ex-situ using a remediation approach consisting of (i) sorption onto oxidized zero-valent iron (ZVI) surfaces and (ii) retrieval of formed metal-ZVI complexes from sediment matrices by magnetic separation. The research focused on the reduction/elimination of the bioavailable fractions of metals, and the efficiency of the method assessed by a combination of a bacterial (MetPLATE?) and an invertebrate (the 48-h Ceriodaphnia dubia acute toxicity test) based bioassays. In sandy sediments, characterized by low HMBC (20.8, 23.5, and 39.6 for Hg, Cu, and Zn, respectively), the determined toxicity units (TU) prior to sediment treatment increased in the order Hg < Cu < Zn, regardless of the bioassay used. The use of ZVI and magnetic separation in these sandy sediments resulted in up to 97% TU reduction. In organic-rich sediments, the affinity of the studied metals for organic matter (OM) resulted in much higher HMBC values (83.9, 108.3, and 136.2 for Cu, Zn, and Hg, respectively) and much lower TU values before sediment treatment with ZVI. The use of MetPLATE? on non-treated sediments resulted in TU values increasing in the order Hg < Cu < Zn, with TU removal efficiencies ranging from 83% to 97% after treatment. The TU values measured with the 48-h C. dubia assay were higher than those obtained with MetPLATE?, and in this case, sediments contaminated with Zn exhibited the lowest percentage of TU removal, with only 81.7% and 80.5% TU removal for sediments with contamination levels of 400 and 800 mg/kg, respectively. For organic sediments contaminated with Cu and Hg, the TU removal exceeded 95%. Overall, this study showed that the proposed remediation method has great potentials with regard to the elimination of the bioavailable metal fractions in contaminated sediments.  相似文献

Benthic oligochaetes were sampled on three occasions (June, August and October 1992) in the upper (0–10 cm) and hyporheic (35–45 cm depths) sediments at five sites of the River Moselle, from upstream of the town of Epinal to Velle-sur-Moselle. The first site (upstream from Epinal) is considered as unpolluted and the four remaining sites are polluted by industrial effluents. The most polluted stations were generally dominated by the pollution tolerant taxon Limnodrilus. Numbers of individuals of this taxon decreased at the less polluted last site in recovery zone, and were also scarce in the first unpolluted site. It is noteworthy that these tendencies were observed in both superficial and hyporheic substrates and to the greatest degree in hyporheic ones. At the unpolluted site, the hyporheic habitat is dominated by the groundwater species Propappus volki, Pristina spp., Pristinella spp. At the less polluted site (last site), the deep sediments are dominated by groundwater species and the Tubificidae without hair setae decrease from June to October. As a result of water exchange between superficial and subterranean waters, superficial substrates of the first and the last stations tend to be colonised by a high proportion of hyporheic species that suggests that flow is primarily from subterranean to superficial waters. The contrary is the case at other polluted stations which are characterised by the invasion of hyporheic substrates by the pollution tolerant superficial taxa Limnodrilus. This suggests that water flows from the river to the deeper groundwater. These two stations are located near drinking water plants which utilise groundwater, thus increasing the vulnerability of groundwater to surface contaminants.  相似文献

Sediments from four lakes polluted by effuents from steel industries and from two reference lakes were investigated. The sediments contained elevated concentrations of oil, Pb, Cd, Hg, Zn, Ni and Cr in different combinations. The toxicity of the polluted sediments to Daphnia was much higher than reported elsewhere (24-h LC50 0.5–1.5 vol%). Tubifex survived more than 3 months in the polluted sediments, but growth and reproduction were inhibited.  相似文献

In this study, we accessed culturable fungal assemblages present in the sediments of three lakes potentially impacted anthropogenically in the Fildes Peninsula, King George Island, Antarctica and identified 63 taxa. Cladosporium sp. 2, Pseudeurotium hygrophilum, and Pseudogymnoascus verrucosus were recovered from the sampled sediments of all lakes. High concentrations of metals and the lowest fungal diversity indices were detected in the sediments of the Central Lake, which can be influenced by human activities due to their proximity to research stations to those of the other two lakes, which were far from the Antarctic stations. At least one type of biological activity was demonstrated by 40 fungal extracts. Among these, P. hygrophilum, P. verrucosus, Penicillium glabrum, and Penicillium solitum demonstrated strong trypanocidal, herbicidal, and antifungal activities. Our results suggest that an increase of the anthropogenic activities in the region might have affected the microbial diversity and composition. In addition, the fungal diversity in these lakes may be a useful model to study the effect of anthropogenic activities in Antarctica. We isolated a diverse group of fungal taxa from Antarctic lake sediments, which have the potential to produce novel compounds for the both the medical and agriculture sectors.

Aquatic mosses and sediments were monitored along Topolnitsa River Basin (Bulgaria), copper-producing and mining region. Six mosses were applied as biomonitors: Bryum turbinatum, Fontinalis antipyretica, Leptodictyum riparium, Platyhypnidium riparioides, Rhizomnium punctatum and Sciuro-hypnum plumosum. Background levels were determined for both plants and fine fraction of sediments collected from an unpolluted site. Contamination factors (CFs) and metal pollution index at each site were presented. The highest sediment enrichment with toxic elements was found in Zlatishka River. Strong increase above background levels showed all 13 heavy metals and toxic elements in biomonitors from stations at Zlatishka and Pirdopska rivers. The two moss species adopted as biomonitors for the first time – Bryum turbinatum and Rhizomnium punctatum – showed high CFs for several heavy metals and contributed to chemical pollution pattern assessment. The parallel use of mosses and sediments in the monitoring study, as well as the applied statistical tools, led to the conclusion that mosses as higher plants revealed pollution patterns not detected by sediments. Results also showed that in a seriously contaminated aquatic environment, which requires rapid assessment, different moss species can be applied for water-monitoring purposes to present preliminary picture of pollution patterns and to direct further studies.  相似文献

Macrofauna density and bioturbation intensity (measured with X-ray radiography), were studied in the Dutch Wadden Sea near Ameland and in Mok Bay, Texel, in September 1988. The bivalveMacoma balthica and the polychaeteHeteromastus filiformis were widely distributed in the areas studied. The cockleCerastoderma edule and the polychaeteScoloplos armiger did not occur in areas with high clay content, while the molluscsHydrobia ulvae andMya arenaria preferred fine sediments. The polychaeteNereis diversicolor preferred the higher parts of the intertidal. In the Ameland area disturbance of primary sediment structures was highest near the shore and near the middle of the tidal confluence where physical reworking was low. The sheltered Mok Bay sediments were completely reworked by deposit-feeders. Bioturbation intensity and deposit-feeder (bioturbator) density were, however, not highly correlated. A number of stations showed higher bioturbation towards the surface and this may be related to the high abundance of near surface dwelling macrofauna, due to eutrophication and organic enrichment of the sediments in recent years.  相似文献
