植物根际促生菌的筛选及鉴定   总被引:15,自引:0,他引:15  
【目的】植物根际促生菌(PGPR)和植物的互作关系往往不稳定,PGPR菌群有可能提高菌株对野外环境的适应性。为此,本文根据PGPR促生机制的多样性,从不同植物根际土壤进行了PGPR的筛选及鉴定。【方法】首先,按照固氮、解磷、解钾、拮抗6种常见病原真菌,同时能在植物根际定殖为基本初筛标准,然后在实验室条件下测定初筛菌株的多项促生能力(PGP),最后通过生理生化试验和16SrRNA基因序列分析对所筛菌株进行鉴定。【结果】从江苏扬州、盐城等地土壤样品筛选出14株PGPR,具有体外抑菌、产NH3、产IAA、产HCN、产嗜铁素、解磷、溶钾、固氮以及产抗生素等促生能力。分类鉴定结果显示:7株属于假单胞菌属(Pseudomonas)、3株属于类芽孢杆菌属(Paenibacillus)、2株为芽孢杆菌属(Bacillus)、1株为布克霍尔德氏菌属(Burkholderia)、1株为欧文氏菌属(Erwinia)。【结论】所筛细菌具有多种促生能力,且能在根际定殖,为进一步构建多功能PGPR广适菌群提供菌株资源。  相似文献

The investigation evaluated the role of plant growth promoting rhizobacteria (PGPR) and Ag-nano particle on the growth and metabolism of maize irrigated with municipal wastewater (MW). Three PGPR isolated from MW were identified on the basis of 16S-rRNA gene sequence analyses as Pseudomonas sp., Pseudomonas fluorescence, and Bacillus cereus. The municipal waste water was used to irrigate the maize seeds inoculated with 3 isolated PGPR. The isolated PGPR had catalase and oxidase enzymes, solubilize insoluble bound phosphate and exhibit antifungal and antibacterial activities. The colony forming unit (cfu) of the PGPR was inhibited by Ag-nano particle, but was stimulated by the municipal wastewater. The Ag-nano particles augmented the PGPR induced increase in root area and root length. The root-shoot ratio was also changed with the Ag-nano particles. The plants irrigated with municipal wastewater had higher activities of peroxidase and catalase which were further augmented by Ag-nano particle. The Ag- nano particle application modulated level of ABA (34%), IAA (55%), and GA (82%), increased proline production (70%) and encountered oxidative stress and augmented the bioremediation potential of PGPR for Pb, Cd, and Ni. Municipal wastewater needs to be treated with PGPR and Ag nano particle prior to be used for irrigation. This aims for the better growth of the plant and enhanced bioremediation of toxic heavy metals.  相似文献

A plant growth-promoting isolate of a fluorescent Pseudomonas sp. EM85 and two bacilli isolates MR-11(2) and MRF, isolated from maize rhizosphere, were found strongly antagonistic to Fusarium moniliforme, Fusarium graminearum and Macrophomina phaseolina, causal agents of foot rots and wilting, collar rots/stalk rots and root rots and wilting, and charcoal rots of maize, respectively. Pseudomonas sp. EM85 produced antifungal antibiotics (Afa+), siderophore (Sid+), HCN (HCN+) and fluorescent pigments (Flu+) besides exhibiting plant growth promoting traits like nitrogen fixation, phosphate solubilization, and production of organic acids and IAA. While MR-11(2) produced siderophore (Sid+), antibiotics (Afa+) and antifungal volatiles (Afv+), MRF exhibited the production of antifungal antibiotics (Afa+) and siderophores (Sid+). Bacillus spp. MRF was also found to produce organic acids and IAA, solubilized tri-calcium phosphate and fixed nitrogen from the atmosphere. All three isolates suppressed the diseases caused by Fusarium moniliforme, Fusarium graminearum and Macrophomina phaseolina in vitro. A Tn5:: lacZ induced isogenic mutant of the fluorescent Pseudomonas EM85, M23, along with the two bacilli were evaluated for in situ disease suppression of maize. Results indicated that combined application of the two bacilli significantly (P = 0.05) reduced the Macrophomina-induced charcoal rots of maize by 56.04%. Treatments with the MRF isolate of Bacillus spp. and Tn5:: lacZ mutant (M23) of fluorescent Pseudomonas sp. EM85 significantly reduced collar rots, root and foot rots, and wilting of maize caused by Fusarium moniliforme and F. graminearum (P = 0.05) compared to all other treatments. All these isolates were found very efficient in colonizing the rhizotic zones of maize after inoculation. Evaluation of the population dynamics of the fluorescent Pseudomonas sp. EM85 using the Tn5:: lacZ marker and of the Bacillus spp. MRF and MR-11(2) using an antibiotic resistance marker revealed that all the three isolates could proliferate successfully in the rhizosphere, rhizoplane and endorhizosphere of maize, both at 30 and 60 days after seeding. Four antifungal compounds from fluorescent Pseudomonas sp. EM85, one from Bacillus sp. MR-11(2) and three from Bacillus sp. MRF were isolated, purified and tested in vitro and in thin layer chromatography bioassays. All these compounds inhibited R. solani, M. phaseolina, F. moniliforme, F. graminearum and F. solani strongly. Results indicated that antifungal antibiotics and/or fluorescent pigment of fluorescent Pseudomonas sp. EM85, and antifungal antibiotics of the bacilli along with the successful colonization of all the isolates might be involved in the biological suppression of the maize root diseases.  相似文献

Plant growth promoting rhizobacteria (PGPR) are an attractive eco-friendly alternative to chemicals in agriculture. While the rhizospheres of crop plants have been well studied with the objective of screening PGPR, weeds, which play an important role in maintaining ecological balance, have largely been ignored. The rhizosphere of a luxuriantly growing, medicinal weed, Cassia occidentalis was analysed by enumerating PGPR on N free media from the most diverse stage of plant (determined by profiles obtained on denaturing gradient gel electrophoresis). Each isolate was tested for other plant growth promotion assays including production of cellulase, indole acetic acid (IAA), ammonia, HCN, siderophore and chitinase to select for ones possessing multi-trait plant growth promoting (PGP) properties. Selected isolates were used for bacterization of Vigna radiata and Vigna mungo to evaluate their efficacy in promoting plant's growth in seedling germination and axenic pot conditions. Thirty five isolates were analysed further for the array of PGP properties they exhibit. A total of 6 isolates were shortlisted on the basis of maximum traits positive, amount of phosphate solubilized and IAA produced. V. radiata responded well to seed bacterization during seedling germination. A maximum increase of approximately 36 and 60?% was observed for shoot and root length, respectively in V. radiata in axenic pot culture over control plants. Extensive branching of roots was also observed with isolate NL, which produced the maximum amount of IAA. Present study investigated the plant growth promoting isolates obtained on N free media in the rhizosphere of C. occidentalis, which have the potential to be used as inoculants for other crops. This provides a new dimension to the significance of weeds in agricultural ecosystems. The study opens up possibilities for utilization of this property of weeds in plant growth promotion, and subsequent enhancement of yield for agricultural crops.  相似文献

The effects of plant growth promoting rhizobacteria (PGPR) on the rooting and root growth of semi-hardwood and hardwood kiwifruit stem cuttings were investigated. The PGPR used were Bacillus RC23, Paenibacillus polymyxa RC05, Bacillus subtilis OSU142, Bacillus RC03, Comamonas acidovorans RC41, Bacillus megaterium RC01 and Bacillus simplex RC19. All the bacteria showed indole-3-acetic acid (IAA) producing capacity. Among the PGPR used, the highest rooting ratios were obtained at 47.50% for semi-hardwood stem cuttings from Bacillus RC03 and Bacillus simplex RC19 treatments and 42.50% for hardwood stem cuttings from Bacillus RC03. As well, Comamonas acidovorans RC41 inoculations indicated higher value than control treatments. The results suggest that these PGPR can be used in organic nursery material production and point to the feasibility of synthetic auxin (IBA) replacement by organic management based on PGPR.  相似文献

Two hundred and six phosphate-solubilizing rhizobacteria (PSB) were isolated from rhizosphere soil (RS) and root endosphere (ER) of apple trees from different sites of four locations viz., Chamba, Shimla, Kinnaur and Kullu of Himachal Pradesh, Northern India, and were screened for plant growth promoting traits (PGPTs) by using culture dependent procedures. Indole acetic acid (IAA) production was detected in 50 isolates (24.2 %), siderophore synthesis in 53 isolates (25.7 %), hydrocyanic acid (HCN) in 40 isolates (19.4 %) and percentage growth inhibition against Dematophora necatrix in 61 isolates (29.6 %). Overall, 54.3 % of PSB isolates from RS and 64.4 % from ER showed none of the PGPTs tested. Among the PSB showing PGPTs, 10.6 % had single trait and 30.6 % had multiple traits showing two (10.7 %), three (14.1 %) and four (5.8 %) types of PGPTs. The Shannon–Weaver diversity index (H′) revealed that PGPT-possessing PSBs in RS were more abundant than ER. Clustering analysis by principal component analysis showed that ER was most important factor influencing the ecological distribution and physiological characterization of PGPT-possessing PSB. There was a positive correlation (0.94, p < 0.05) between HCN and antifungal activity producers, and IAA and antifungal activity producers (0.99, p < 0.05). Significant positive correlation (0.42, p < 0.05) between HCN producers and altitude was also noted.  相似文献

[目的]以不同植物根及根际土壤为研究材料,进行植物根际促生菌(PGPR)的筛选,并探索其植物促生作用机制.[方法]以解磷、固氮、产氨、产IAA和拮抗3种常见病原真菌为筛选标准,测定了初筛菌株的多项促生能力,并通过对这些菌分别单独回接和多菌混接的玉米盆栽试验,测定了其对玉米的促生效应.[结果]从渭南、成阳、安康、商洛和榆林5地分离得到的158株菌中有17株茵具有上述多种植物促生作用的菌株.盆栽试验的测定结果表明:单独接种和多菌混合接种在玉米株高、根长、茎长、茎平均直径和干重方面与对照组相比较都有所增加,尤其是在多个指标上,多菌混合接种所显示出的促生效应均明显优于单菌接种.[结论]所筛选到的具有多种促生能力的菌株,可以为进一步构建植物根际促生菌(PGPR)菌群提供良好的种质资源.  相似文献

Bacteria-mediated plant growth promotion is a well-established and complex phenomenon that is often achieved by the activities of more than one plant growth-promoting (PGP) trait, which may not always be present in a single organism. Biofilms developed using a combination of two organisms with useful plant growth-promoting rhizobacteria (PGPR) traits may provide a definite advantage. In this context, in vitro studies were conducted evaluating the PGP traits of novel biofilms developed using Trichoderma as matrix and agriculturally important bacteria (Azotobacter chroococcum, Pseudomonas fluorescens and Bacillus subtilis) as partners. Such biofilms exhibited higher values for various biochemical attributes as compared to the individual organisms and dual cultures. TrichodermaBacillus and TrichodermaPseudomonas biofilms exhibited enhanced antifungal activity, ammonia, indole acetic acid (IAA) and siderophore production, as compared to the other treatments. TrichodermaAzotobacter biofilm recorded the highest nitrogenase activity and 1-aminocyclopropane-1-carboxylic (ACC) deaminase activity. The synergism in terms of the PGP traits in the biofilms revealed their promise as superior PGP inoculants.  相似文献

Plant growth‐promoting rhizobacteria (PGPR) affect growth of host plants through various direct and indirect mechanisms. Three native PGPR (Pseudomonas putida) strains isolated from rhizospheric soil of a Mentha piperita (peppermint) crop field near Córdoba, Argentina, were characterised and screened in vitro for plant growth‐promoting characteristics, such as indole‐3‐acetic acid (IAA) production, phosphate solubilisation and siderophore production, effects of direct inoculation on plant growth parameters (shoot fresh weight, root dry weight, leaf number, node number) and accumulation and composition of essential oils. Each of the three native strains was capable of phosphate solubilisation and IAA production. Only strain SJ04 produced siderophores. Plants directly inoculated with the native PGPR strains showed increased shoot fresh weight, glandular trichome number, ramification number and root dry weight in comparison with controls. The inoculated plants had increased essential oil yield (without alteration of essential oil composition) and biosynthesis of major essential oil components. Native strains of P. putida and other PGPR have clear potential as bio‐inoculants for improving productivity of aromatic crop plants. There have been no comparative studies on the role of inoculation with native strains on plant growth and secondary metabolite production (specially monoterpenes). Native bacterial isolates are generally preferable for inoculation of crop plants because they are already adapted to the environment and have a competitive advantage over non‐native strains.  相似文献

Free-living nitrogen fixing bacteria were isolated from rhizosphere of seven different plant namely sesame, maize, wheat, soybean, lettuce, pepper and rice grown in Chungbuk Province, Korea. Five isolates with nitrogenase activity above 150nmol(-1) mg(-1) protein were identified based on, phenotypic and 16S rDNA sequences analysis. The strains were identified as Stenotrophomonas maltophilia (PM-1, PM-26), Bacillus fusiformis (PM-5, PM-24) and Pseudomonas fluorescens (PM-13), respectively. All the isolates produced indole-3-acetic acid (IAA), in the presence of tryptophan, ranging from 100.4 microg ml(-1) (PM-13) to 255 microg ml(-1) (PM-24). The isolate PM-24 (Bacillus fusiformis) exhibiting highest nitrogenase activity (3677.81 nmol h(-1) mg(-1) protein) and IAA production (255microg ml(-1)) has a promising potential for developing as a plant growth promoting rhizobacteria.  相似文献

In order to explore salt tolerant plant growth promoting rhizobacteria (PGPR), 121 isolates were recovered from Tsunami affected area of Andaman and Nicobar Islands, India, out of which 23 were grown at 10% NaCl. Out of total isolates grown at 10% NaCl concentration, 14 solubilized phosphate, 13 showed siderophore production, five showed Indole acetic acid (IAA) production and 16 produced one or more extra cellular enzymes. Isolates SJ10 and CR5 showed the highest enzyme production and PGP properties and five isolates showed antagonistic activity against Sclerotium rolfsii. Screening of potential isolates for nitrogen fixing gene (nifH) showed only three isolates (MM3, MM6 and MRC11) possessed nifH gene. Salt tolerant Bacillus PGPR could be used as a bio-inoculant to enhance the crop growth under Tsunami affected soils.  相似文献

[目的]获得具有产ACC、IAA,铁载体,能固氮或解磷的潜在促生菌株.[方法]通过稀释涂布的方法,从麻疯树根际土壤中分离得到98株细菌,从中选取28株以产l-氨基环丙烷-1-羧酸(ACC)脱氨酶为主要促生指标进行筛选,同时检测了其产吲哚乙酸(IAA)、固氮、解磷及铁载体等促生指标的能力.[结果]结果显示,46%的菌株能产ACC脱氨酶,其含量最高可达到128.308 μmol α-KA/(mg.h),68%的菌株能产生IAA,54%的菌株有固氮的能力,32%的菌株有解磷的能力.少量菌株同时具有产ACC脱氨酶、IAA,固氮,解磷等能力.挑选代表性菌株进行16S rRNA序列分析,这些菌株属于芽孢杆菌属(Bacillus)、节杆菌属(Arthrobacter)、假单胞菌属(Pseudomonas)和产碱杆菌属(Advenella)等8个属,其中多数菌株(50%)属于芽孢杆菌属,系统发育分析表明菌株KLBMP 4817、KLBMP 4821和KLBMP 4824为窄食单胞菌属(Stenotrophomonas)和类芽孢杆菌属(Paenibacillus)的潜在新种.[结论]攀枝花麻疯树根际土壤细菌中含有丰富的遗传多样性,且存在大量的促生菌株.其中,菌株KLBMP 4804产ACC脱氨酶含量最高.菌株KLBMP4820产IAA含量最显著.  相似文献

Although plant growth-promoting rhizobacteria (PGPR) have been reported to influence plant growth, yield and nutrient uptake by an array of mechanisms, the specific traits by which PGPR promote plant growth, yield and nutrient uptake were limited to the expression of one or more of the traits expressed at a given environment of plant–microbe interaction. We selected nine different isolates of PGPR from a pool of 233 rhizobacterial isolates obtained from the peanut rhizosphere on the basis of ACC-deaminase activity. The nine isolates were selected, initially, on the basis of germinating seed bioassay in which the root length of the seedling was enhanced significantly over the untreated control. All the nine isolates were identified as Pseudomonas spp. Four of these isolates, viz. PGPR1, PGPR2, PGPR4 and PGPR7 (all fluorescent pseudomonads), were the best in producing siderophore and indole acetic acid (IAA). In addition to IAA and siderophore-producing attributes, Pseudomonas fluorescens PGPR1 also possessed the characters like tri-calcium phosphate solubilization, ammonification and inhibited Aspergillus niger and A. flavus in vitro. P. fluorescens PGPR2 differed from PGPR1 in the sense that it did not show ammonification. In addition to the traits exhibited by PGPR1, PGPR4 showed strong in vitro inhibition to Sclerotium rolfsii. The performances of these selected plant growth-promoting rhizobacterial isolates were repeatedly evaluated for 3 years in pot and field trials. Seed inoculation of these three isolates, viz. PGPR1, PGPR2 and PGPR4, resulted in a significantly higher pod yield than the control, in pots, during rainy and post-rainy seasons. The contents of nitrogen and phosphorus in soil, shoot and kernel were also enhanced significantly in treatments inoculated with these rhizobacterial isolates in pots during both the seasons. In the field trials, however, there was wide variation in the performance of the PGPR isolates in enhancing the growth and yield of peanut in different years. Plant growth-promoting fluorescent pseudomonad isolates, viz. PGPR1, PGPR2 and PGPR4, significantly enhanced pod yield (23–26%, 24–28% and 18–24%, respectively), haulm yield and nodule dry weight over the control in 3 years. Other attributes like root length, pod number, 100-kernel mass, shelling out-turn and nodule number were also enhanced. Seed bacterization with plant growth-promoting P. fluorescens isolates, viz. PGPR1, PGPR2 and PGPR4, suppressed the soil-borne fungal diseases like collar rot of peanut caused by A. niger and PGPR4 also suppressed stem rot caused by S. rolfsii. Studies on the growth patterns of PGPR isolates utilizing the seed leachate as the sole source of C and N indicated that PGPR4 isolate was the best in utilizing the seed leachate of peanut, cultivar JL24. Studies on the rhizosphere competence of the PGPR isolates, evaluated on the basis of spontaneous rifampicin resistance, indicated that PGPR7 was the best rhizoplane colonizer and PGPR1 was the best rhizosphere colonizer. Although the presence of growth-promoting traits in vitro does not guarantee that an isolate will be plant growth promoting in nature, results suggested that besides ACC-deaminase activity of the PGPR isolates, expression of one or more of the traits like suppression of phytopathogens, solubilization of tri-calcium phosphate, production of siderophore and/or nodulation promotion might have contributed to the enhancement of growth, yield and nutrient uptake of peanut.  相似文献

AIMS: Plant growth promoting rhizobacteria (PGPR) are commonly used as inoculants for improving the growth and yield of agricultural crops, however screening for the selection of effective PGPR strains is very critical. This study focuses on the screening of effective PGPR strains on the basis of their potential for in vitro auxin production and plant growth promoting activity under gnotobiotic conditions. METHODS AND RESULTS: A large number of bacteria were isolated from the rhizosphere soil of wheat plants grown at different sites. Thirty isolates showing prolific growth on agar medium were selected and evaluated for their potential to produce auxins in vitro. Colorimetric analysis showed variable amount of auxins (ranging from 1.1 to 12.1 mg l-1) produced by the rhizobacteria in vitro and amendment of the culture media with l-tryptophan (l-TRP), further stimulated auxin biosynthesis (ranging from 1.8 to 24.8 mg l-1). HPLC analysis confirmed the presence of indole acetic acid (IAA) and indole acetamide (IAM) as the major auxins in the culture filtrates of these rhizobacteria. A series of laboratory experiments conducted on two cv. of wheat under gnotobiotic (axenic) conditions demonstrated increases in root elongation (up to 17.3%), root dry weight (up to 13.5%), shoot elongation (up to 37.7%) and shoot dry weight (up to 36.3%) of inoculated wheat seedlings. Linear positive correlation (r = 0.99) between in vitro auxin production and increase in growth parameters of inoculated seeds was found. Based upon auxin biosynthesis and growth-promoting activity, four isolates were selected and designated as plant growth-promoting rhizobacteria (PGPR). Auxin biosynthesis in sterilized vs nonsterilized soil inoculated with selected PGPR was also monitored that revealed superiority of the selected PGPR over indigenous microflora. Peat-based seed inoculation with selected PGPR isolates exhibited stimulatory effects on grain yields of tested wheat cv. in pot (up to 14.7% increase over control) and field experiments (up to 27.5% increase over control); however, the response varied with cv. and PGPR strains. CONCLUSIONS: It was concluded that the strain, which produced the highest amount of auxins in nonsterilized soil, also caused maximum increase in growth and yield of both the wheat cv. SIGNIFICANCE AND IMPACT OF STUDY: This study suggested that potential for auxin biosynthesis by rhizobacteria could be used as a tool for the screening of effective PGPR strains.  相似文献

AIM: To study the antifungal activity and plant beneficial traits of a broad-spectrum antagonistic fluorescent pseudomonad strain, PUPa3. METHODS AND RESULTS: Strain PUPa3 was isolated from the rhizosphere soil of rice and identified as Pseudomonas aeruginosa on the basis of biochemical tests and by comparison of 16S rDNA sequences. This bacterium exhibits a broad-spectrum antifungal activity towards phytopathogenic fungi. The antifungal metabolite by PUPa3 was extracted, purified and characterized using nuclear magnetic resonance (NMR) and mass spectroscopy (MS). Production of indole-3-acetic acid (IAA), siderophores, phosphatase and protease in PUPa3 was determined. Strain PUPa3 did not produce hydrogen cyanide, cellulase and pectinase. CONCLUSION: The antifungal metabolite produced by PUPa3 has been identified as phenazine-1-carboxamide (PCN) on the basis of NMR and MS data. Strain PUPa3 showed a broad-spectrum antifungal activity towards a range of phytopathogenic fungi. This bacterium also showed several plant growth-promoting traits but did not show the traits attributed to deleterious rhizobacteria. SIGNIFICANCE AND IMPACT OF THE STUDY: Present study reports the production of PCN as well as IAA for the first time by a saprophytic P. aeruginosa strain PUPa3. Because of the production of siderophore, growth hormone, protease and phosphatase and its innate fungicidal potential, this strain can be used as biofertilizer and antagonist against a range of phytopathogenic fungi that infect rice, groundnut, tobacco, chili, mango, sugarcane, tea, cotton and banana.  相似文献

The present study was undertaken to examine the effect of the application of fly ash (FA) into Garden soil (GS), with and without inoculation of plant growth promoting bacteria (PGPB), on the growth and metal uptake by Zea mays plants. Three FA tolerant PGPB strains, Pseudomonas sp. PS5, PS14, and Bacillus sp. BC29 were isolated from FA contaminated soils and assessed for their plant growth promoting features on the Z. mays plants. All three strains were also examined for their ability to solubilize phosphate and to produce Indole Acetic Acid (IAA), siderophores, and hydrogencynide acid (HCN) production. Although inoculation of all strains significantly enhanced the growth of plants at both the concentration of FA but maximum growth was observed in plants inoculated with BC29 and PS14 at low level (25%) of FA concentration. The experimental results explored the plant growth promoting features of selected strains which not only enhanced growth and biomass of plants but also protected them from toxicity of FA.  相似文献

The response of two root associated bacteria Pseudomonas pseudoalcaligenes and Bacillus pumilus were studied in the (salt-sensitive) rice GJ17 cultivar to salinity under controlled environmental growth conditions for protection of plant from adverse effect of salinity. Salinity affects the growth of salt-sensitive cultivar, but inoculation of plant growth promoting rhizobacteria (PGPR) reduces the harmful effect of salinity. The present study states that PGPR helps to reduce lipid peroxidation and superoxide dismutase activity in salt-sensitive GJ17 cultivar under salinity and play an important role in the growth regulation for positive adaptation of plants to salt stress. This study shows that inoculation of paddy (Oryza sativa) with such bacteria could provide salt-tolerant ability by reducing the toxicity of reactive oxygen species by reducing plant cell membrane index, cell caspase-like protease activity, and programmed cell death and hence resulted in increase cell viability. As these isolates remain associated with the roots, the effects of tolerance against salinity are observed here. Results also indicate that isolated PGPR strain help in alleviating up to 1.5 % salinity stress as well as improve tolerance.  相似文献

AIM: To isolate and characterize groundnut-associated bacterial isolates for growth promotion of groundnut in field. METHODS AND RESULTS: Three hundred and ninety-three groundnut-associated bacteria, representing the geocarposphere, phylloplane and rhizosphere, and endophytes were applied as seed treatment in greenhouse. Maximum increase in plant biomass (up to 26%) was observed following treatment with a rhizosphere isolate identified as Bacillus firmis GRS 123, and two phylloplane isolates Bacillus megaterium GPS 55 and Pseudomonas aeruginosa GPS 21. There was no correlation between the production of L-tryptophan-derived auxins and growth promotion by the test isolates. Actively growing cells and peat formulations of GRS 123 and GPS 55, and actively growing cells of GPS 21, significantly increased the plant growth and pod yield (up to 19%) in field. Rifampicin-resistant mutants of GRS 123 and GPS 21 colonized the ecto- and endorhizospheres of groundnut, respectively, up to 100 days after sowing (DAS), whereas GPS 55 was recovered from both the habitats at 100 DAS. CONCLUSION: Seed bacterization with phylloplane isolates promoted groundnut growth indicating the possibility of isolating rhizosphere beneficial bacteria from different habitats. SIGNIFICANCE AND IMPACT OF THE STUDY: Identification of phylloplane bacteria as effective plant growth-promoting rhizobacteria (PGPR) broadens the spectrum of PGPR available for field application.  相似文献

获得辣椒根际促生菌(Plant growth promoting rhizobacteria,PGPR)并探究其抗病促生特性。采用固氮、无机磷和有机磷培养基从江苏省徐州市采集的辣椒根际土壤中分离筛选根际促生菌株(PGPR),通过形态特征及16S rDNA序列分析进行菌株鉴定,对菌株的固氮、解磷、分泌3-吲哚乙酸(IAA)能力及对4种辣椒病害病原菌抗病能力进行探究。得到13株辣椒PGPR菌株,经鉴定分别属于Bacillus、Pseudomonas、Lelliottia、Siccibacter、Achromobacter、Microbacterium和Paenibacillus;13株PGPR菌株均有固氮功能;其中7株可解有机磷,分别属于Lelliottia、Bacillus、Siccibacter、Microbacterium、Paenibacillus;5株可解无机磷,分别属于Lelliottia、Bacillus、Siccibacter、Pseudomonas;3株具有分泌IAA能力,分别属于Lelliottia、Siccibacter、Bacillus;5株具有抗病能力,分别属于Bacillus、Lelliottia、Siccibacter。辣椒根际土壤含有在农业生产上具有潜在的应用价值的多功能根际促生菌。  相似文献

Traits involved in plant growth promotion by bacteria are ambiguously decided as phytohormones, siderophores, HCN, proteases, chitinases, cellulases, ammonia, exopolysaccharide production and phosphate solubilization or antagonistic activity. A total of 40 fluorescent Pseudomonas strains were isolated from diverse soils of various agro-ecosystems of India. Among these 7 strains viz, P1, P10, P13, P18, P21, P28 and P38 were selected depending on their character of possessing single or multiple PGPR traits. These isolates individually and in combination with AM fungi (Glomus fasciculatum and Glomus aggregatum) were used for treating sorghum seeds. 25 days after sowing, plants were analysed for different plant growth promoting parameters. It was observed that strains in presence of mycorrhizae performed well compared to the strains devoid of mycorrhizae. Pseudomonas sp. P1 possessing GA3, EPS production and ‘P’ solubilization performed well. Pseudomonas sp. P38 which was a volatile (HCN) producer but a good phosphate solubilizer did not perform well. Strain P28 with multiple PGPR traits did not show the expected out come. Results varied when mycorrhizae was used in combination. P10 and P13 which were good in IAA, GA3, EPS, siderophore and ‘P’ solubilization performed well with mycorrhizae, with an overall increase in plant biomass, leaf area, total chlorophyll and mycorrhizal infection compared to other combinations. Here, strain P1 in combination with mycorrhizae did not show significant increase in plant growth compared to P10 and P13. Hence, mere possession of multiple PGPR traits does not confer fluorescent Pseudomonas strains as PGPR. Further studies have to be carried out in order to evaluate the other traits which may be involved in plant growth promotion.  相似文献
