Localization of a heartwood norlignan, agatharesinol, in Sugi (Japanese cedar, Cryptomeria japonica D. Don, Taxodiaceae) was investigated by immunohistochemistry. Immuno light microscopy showed that the contents of ray parenchyma cells were immunostained in heartwood but not in sapwood. The staining of the heartwood tissue was competitively inhibited by agatharesinol but not by other Sugi heartwood extractives, and was, furthermore, markedly reduced by pre-extraction of the tissue with MeOH. These results indicated that the staining can be ascribed to the immunolabeling of agatharesinol in situ. The accumulations over the inner surface of some tracheid cell walls adjacent to the ray parenchyma cells were also immunolabeled, while the contents in axial parenchyma cells were not. In conclusion, agatharesinol was localized in the ray parenchyma cells in Sugi heartwood, and differences between the chemical structure of the contents of ray and axial parenchyma cells were also suggested.  相似文献

In vitro hydroxylation of the norlignan agatharesinol to sequirin C and metasequirin C was demonstrated for the first time. After incubating agatharesinol with a microsomal preparation from the heartwood side of the intermediate wood of Cryptomeria japonica in the presence of cofactors, the aromatic ring-monohydroxylated derivatives of agatharesinol, sequirin C and metasequirin C, were formed. Although hydroxylation hardly occurred in the absence of cofactors, it could be initiated by adding NADPH or NADH, and was enhanced by further adding FAD or FMN. When microsomal preparations from the sapwood or from the sapwood side of the intermediate wood were used, hydroxylation did not occur. This in vitro conversion of the norlignans indicates that the hydroxylation of agatharesinol to sequirin C and metasequirin C is part of the in vivo biosynthetic pathway of norlignans. Another C. japonica norlignan, sugiresinol, which is a side chain-cyclized isomer of agatharesinol, does not seem to be accepted as a substrate, because hardly any hydroxysugireinol was formed after similar incubation with the enzyme.  相似文献

The biosynthetic relationship between the two norlignans agatharesinol and trans-hinokiresinol was investigated. Fresh sapwood sticks of Cryptomeria japonica were fed with stable isotope-labeled compounds, namely p-coumaryl alcohol-[9,9-2H], p-coumaryl alcohol-[9-18O] and trans-hinokiresinol-[1-2H], and then incubated under high-humidity for approximately 20 days, during which the two norlignans were produced simultaneously. While trans-hinokiresinol was strongly deuterium-labeled after feeding with p-coumaryl alcohol-[9,9-2H], agatharesinol was only lightly labeled after feeding with either p-coumaryl alcohol-[9,9-2H] or -[9-18O]. These results suggest that p-coumaryl alcohol, which is a precursor of hinokiresinol, is not involved in the biosynthesis of agatharesinol. Therefore, the norlignan carbon skeleton of agatharesinol must be framed from different types of phenylpropanoid monomers compared to those utilized by the trans-hinokiresinol pathway. The biosynthesis of these two norlignans seems to branch at an early stage, i.e., before the framing of the norlignan carbon skeleton. Furthermore, agatharesinol was not labeled with deuterium after feeding with 2H-labeled trans-hinokiresinol, which has the simplest norlignan structure. This result strongly supports the suggestion that the conversion of trans-hinokiresinol to agatharesinol is not part of the biosynthesis of norlignans and that early branching occurs instead.  相似文献

Time-of-flight secondary ion mass spectrometry (TOF-SIMS) was applied to the investigation of heartwood extractives in Sugi (Cryptomeria japonica). Sugi heartwood tissue generated secondary ions that were not produced from sapwood tissue by TOF-SIMS. Among the peculiar ions generated from heartwood, two positive ions of m/z 285 and 301 were remarkable due to their appearance in a larger mass range and with a high intensity. These two ions were not generated from heartwood tissue preextracted with n-hexane, and the n-hexane extract of Sugi heartwood produced both ions. Gas chromatography-mass spectrometry of the n-hexane extract demonstrated that ferruginol, a diterpene phenol, the molecular weight of which is 286, constituted one of the predominant constituents of the extract. Authentic ferruginol also generated both ions by TOF-SIMS. The molecular formula of the m/z 285 ion generated from Sugi heartwood tissue was estimated to be C20H29O, which corresponds well with that of ferruginol, i.e. C20H30O, by peak identification. All these results strongly suggest that the m/z 285 ion generated from Sugi heartwood tissue originated significantly from ferruginol in Sugi heartwood. By TOF-SIMS imaging, the m/z 285 ion was detected uniformly in the tracheid cell walls, in the cell walls of the axial parenchyma cells and ray parenchyma cells, and also inside these parenchyma cells. These results indicate that ferruginol was distributed almost evenly in Sugi heartwood tissue.  相似文献

Six new norlignans, named sequosempervirins B-G (1-6), together with three known norlignans, agatharesinol (7), agatharesinol acetonide (8), and sugiresinol (9), were isolated from the branches and leaves of Sequoia sempervirens. Their structures were determined mainly by high-resolution mass spectroscopy (HR-MS), and various 1D- and 2D-NMR methods, as well as, in the case of 1, by means of X-ray diffraction. Compound 8 showed anticancer activity towards the A549 non-small-cell lung-cancer cell line (IC50 = 27.1 microM). The acetone extract of S. sempervirens was found to be antifungal towards Candida glabrata (IC50 = 15.98 microg/ml), and both the acetone and MeOH extracts inhibited the proteolytic activity of cathepsin B (IC50 = 4.58 and 5.49 microg/ml, resp.).  相似文献

During our studies on the components in Japanese cedar, Cryptomeria japonica, we found that the crude methanol extract of C. japonica showed intense antifeeding activity against one snail species, Acusta despesta, which is well-known as a pest of many vegetables and crops. The active components in the extract were separated into the hexane and ethyl acetate soluble fractions. From the active ethyl acetate soluble fraction, two norlignans, sequirin-C and agatharesinol, were isolated and identified as the active compounds. Both compounds inhibited feeding behavior of A. despesta at 30 microg/cm2 and 40 microg/cm2 concentrations, respectively, when applied by an eggplant leaf or filter paper containing 20 microl of 5% sucrose solution.  相似文献

In order to study the biosynthesis of agatharesinol, a norlignan, l-phenylalanine-[ring-2,3,4,5,6-2H] and trans-cinnamic acid-[ring-13C6] were administered to fresh sapwood sticks of Cryptomeria japonica (sugi, Japanese cedar), that is, the labeled precursors were allowed to be absorbed through the tangential section of the wood sticks. The wood sticks were then maintained in high humidity desiccators for approximately 20 d after which ethyl acetate (EtOAc) extracts of the wood sticks were analyzed by gas chromatography-mass spectrometry (GC-MS). Native agatharesinol (trimethylsilylated) produces an m/z 369 ion and an m/z 484 ion that are characteristic of its structure. Agatharesinol formed in the sapwood sticks treated with the deuterium-labeled l-phenylalanine generated both of these ions together with m/z 373 and 377 ions (m/z 369+4 and +8, respectively), and also m/z 488 and 492 ions (m/z 484+4 and +8, respectively). Generation of m/z 373 and 488 ions is attributed to the substitution by deuterium of the four hydrogen atoms of either of the p-hydroxyphenyl rings of agatharesinol, and that of m/z 377 and 492 ions is attributed to the substitution by deuterium of the eight hydrogen atoms of both p-hydroxyphenyl rings. In the administration of the 13C-labeled trans-cinnamic acid, m/z 375 and 381 ions (m/z 369+6 and +12, respectively), and also m/z 490 and 496 ions (m/z 484+6 and +12, respectively) were found, indicating that either aromatic ring or both aromatic rings of agatharesinol were 13C-labeled. Consequently, assimilation of the labeled precursors into agatharesinol was clearly detected, and an experimental procedure for studies on the biosynthesis was developed.  相似文献

The permethyl ethers of three new norlignans (sequirins-E, -F, and -G) from Sequoiadendron gigantea heartwood have been characterized by NMR and MS as 2-(3,4-dimethoxyphenyl)-4-(4-methoxyphenyl-, 2,4-(3,4-dimethoxyphenyl)-, and 2-(3,4-dimethoxyphenyl)-4-(3,4,5-trimethoxyphenyl)-5-hydroxytetrahydropyran respectively, with the 2,5-trans,4,5-trans stereochemistry. Sequirins A-D, characteristic norlignans of Sequoia sempervirens Endl., could not be detected in S. gigantea heartwood, nor could sequirins E-G be found in S. sempervirens heartwood. Wood is of critical importance to humans as a primary feedstock for biofuel, fiber, solid wood products, and various natural compounds including pharmaceuticals. The trunk wood of most tree species has two distinctly different regions: sapwood and heartwood. In addition to the major constituents, wood contains extraneous chemicals that can be removed by extraction with various solvents. The composition and the content of the extractives vary depending on such factors as, species, growth conditions, and time of year when the tree is cut. Despite the great commercial and keen scientific interest, little is known about the tree-specific biology of the formation of heartwood and its extractives. In order to gain insight on the molecular regulations of heartwood and its extractive formation, we carried out global examination of gene expression profiles across the trunk wood of black locust (Robinia pseudoacacia L.) trees. Of the 2,915 expressed sequenced tags (ESTs) that were generated and analyzed in the current study, 55.3% showed no match to known sequences. Cluster analysis of the ESTs identified a total of 2278 unigene sets, which were used to construct cDNA microarrays. Microarray hybridization analyses were then performed to survey the changes in gene expression profiles of trunk wood. The gene expression profiles of wood formation differ according to the region of trunk wood sampled, with highly expressed genes defining the metabolic and physiological processes characteristic of each region. For example, the gene encoding sugar transport had the highest expression in the sapwood, while the structural genes for flavonoid biosynthesis were up-regulated in the sapwood-heartwood transition zone. This analysis also established the expression patterns of 341 previously unknown genes.  相似文献

Epitopes associated with a synthetic hepatitis B surface antigen peptide   总被引:5,自引:0,他引:5  
A synthetic peptide (SP1), corresponding to the amino acid residues 122 through 137 of the major polypeptide derived from hepatitis B surface antigen (HBsAg), subtype ayw, was analyzed for the presence of the major epitopes of HBsAg. Both a cyclic form, produced by introduction of an intrachain disulfide bond, and a linear form of the peptide were characterized. A panel of monoclonal antibodies with defined specificity for the cross-reactive group a antigenic determinant(s) and for the y and w subtype specificities was used for this analysis. The cyclic, but not the linear, form of SP1 reacted with five of 14 anti-a monoclonal antibodies, demonstrating that the cyclic peptide contains a conformation-dependent a epitope. Only one anti-a antibody was found to react with both cyclic and linear forms of SP1. Because SP1 failed to react with the remaining 8 anti-a monoclonal antibodies, it was concluded that the a antigenic reactivity associated with HBsAg contains an additional epitope(s) unrelated to that expressed on SP1. Both cyclic and linear SP1 reacted with three of three anti-y monoclonal antibodies, indicating that a sequential y epitope is also present on SP1; no w reactivity was detected. Analysis of the idiotypes associated with the monoclonal antibodies showed those that combined with cyclic SP1 also inhibited the binding of a common human anti-HBs (CHBs) idiotype with its rabbit anti-idiotype serum, whereas a monoclonal antibody that did not react with the cyclic SP1 epitope failed to inhibit the CHBs idiotype-anti-idiotype reaction. Thus, the conformational a epitope present on cyclic SP1 appears to contain the predominant epitope recognized by humans in response to a natural HBV infection.  相似文献

The aim of this study was to compare natural durability of Siberian larch heartwood grown in Siberia and Sweden as well as European larch and Scots pine heartwood grown in Sweden. The study was based on standard in- and above ground tests lasting 12 years but laboratory decay tests with white and brown rot fungi was also included. Field test results showed that Siberian larch heartwood from Siberia was the most durable among the studied heartwoods with a decay index of 60 after 12 years in Simlångsdalen (Sweden), while European larch heartwood grown in Sweden, was decayed to failure before the end of the test. Scots pine heartwood was found to perform similarly to Siberian larch from Siberia. No relationship could be established between natural durability of examined heartwoods and their water absorption behavior; however, strong correlation to the total amount of extractives was observed. Scots pine and Siberian larch heartwood from Siberia had 12.7 and 19.6% total extractives content respectively but the extractives composition differs. The study revealed also that lignin and monosaccharide content could not explain the variations in decay resistance of the studied heartwoods. No similarities in the natural durability revealed by laboratory and field tests were observed.  相似文献

We previously proposed that extractives in highly durable heartwood may protect wood against fungal colonization and subsequent degradation by dual mechanisms: the extractives have some fungicidal activity and are also free radical scavengers (antioxidants). In short-term laboratory decay tests using two different wood species and decay fungi, the antioxidant 2,6-dimethyl-di-tert-butyl-4-methylphenol (BHT) alone had little or no preservative effect. In contrast, the combination of BHT with different organic commercial biocides always showed an increase in efficacy compared to the organic biocide alone. Consequently, we conclude that the combination of a commercial antioxidant and biocide is synergistic. This implies that extractives may protect wood by more than simply being fungicidal.  相似文献

Two monoclonal antibodies to human milk-fat globule membranes, which recognize an epithelial antigen designated MAM-3c, were found to bind strongly to epithelial glycoproteins derived from non-secretors. Further investigations, using purified glycoproteins and structurally defined oligosaccharides, established that the optimal antigenic structure for both antibodies involves the Type 1 based blood group antigen, Lea, in combination with the Type 2 based onco-developmental antigen, SSEA-1, (Formula: see text) as in lacto-N-difucohexaose II. The antibodies may also react with the corresponding monofucosyl structures lacking the 3- or 4- linked fucose residues and to a lesser extent with the afucosyl tetrasaccharide sequence as in lacto-N-tetraose. The Lea and SSEA-1 antigens are known to occur on human epithelial glycoproteins. However, this is the first report of an antigenic specificity involving a combination of the Type 1 and Type 2 based fuco-oligosaccharides and occurring on epithelial glycoproteins.  相似文献

Many monoclonal antibodies that react with the lacto-N-fucopentaose III (LNF III) antigenic determinant, Gal beta 1-4(Fuc alpha 1-3)GlcNAc beta 1-3Gal beta 1-4Glc, have been described recently. The terminal trisaccharide of this determinant, fucosyllactosamine, is present on glycolipids and glycoproteins and on the surface of granulocytes, monocytes, and other cells. To study the structural and genetic diversity of these antibodies, syngeneic anti-idiotypic monoclonal antibodies were produced in BALB/c mice against PMN 6, a monoclonal antibody directed against this sequence. Anti-idiotypic antibodies 6B1 and 6C4 reacted with 50% of a panel of 20 anti-LNF III monoclonal antibodies, whereas 6A3 reacted strongly only with PMN 6. This indicates that the determinants recognized by 6C4 and 6B1 represent major cross-reactive idiotopes of this family of antibodies. The binding of idiotypic antibodies to a glycolipid bearing this antigenic determinant was completely inhibited by the three anti-idiotypic antibodies, 6A3, 6B1, and 6C4. The idiotopes could be demonstrated on the heavy chain of the monoclonal antibodies by an antibody transfer technique when mild reducing conditions were employed, but a high concentration of reducing agent destroyed the idiotypic determinants. This suggests that the anti-idiotypic antibodies recognize conformational structures expressed on the heavy chain molecules. The binding of 18 monoclonal antibodies to two glycolipid antigens and to a fucosyllactosamine-bovine serum albumin conjugate was compared. Antibodies that possessed the 6C4 cross-reactive idiotope bound to fucosyllactosamine-bovine serum albumin more weakly than idiotype-negative antibodies (p = 0.001). This suggests that the 6C4-positive antibodies might represent germline structures.  相似文献

Twenty-three major components were detected in the methanol extractives of the heartwood of Eucalyptus sideroxylon. The components identified include resveratrol, resveratrol-β-glucoside, 3,3′-di- and 3,3′,4-tri-o-methylellagic acids and their glucosides. The 3,3′-di-o-methylellagic acid 4′-glucoside isolated had properties significantly different from those previously reported for this compound. Also present were gailic acid, catechin, ellagic acid, an unidentified stilbene, the ellagitannins D-6 and D-13, polymerized leucocyanidin and an oily material. The sapwood contained gailic acid, small amounts of ellagitannins and ellagic acids and traces of other components. The heartwood extractives of related eucalypt species were also examined.  相似文献

The Sm antigen was isolated and purified from calf thymus nuclear extract by affinity chromatography. The affinity columns were made with serum antibodies from an SLE patient or an anti-Sm monoclonal antibody derived from a hybridoma cell line. Proteins eluted from these two columns had m.w. of 58,000 and 35,000 by SDS polyacrylamide gel electrophoresis. The natural conformation of this antigen appears to be 95,000 in m.w. with the 58,000 particle containing the Sm antigenic determinant. The affinity column-purified antigen detected by the human anti-Sm antibodies is also recognized by anti-Sm antibodies in murine lupus serum, as shown by solid-phase radioimmunoassay. This study 1) demonstrates the molecular and antigenic nature of the Sm antigen and 2) compares the anti-Sm binding capabilities of antibody populations present in sera from SLE patients and from MRL lpr/lpr mice.  相似文献

 Radial distribution of soluble phenolics was investigated at different heights in stems of Juglans nigra, J. regia and hybrids J. nigra 23 × J. regia. Four major phenolic compounds were studied: hydrojuglone glucoside (HJG), quercitrin (QUER) and two unknown compounds characterized as two ellagic acid derivatives E1 and E2. HJG and E1 content increased gradually in the sapwood, peaked in the sapwood-heartwood transition zone, and decreased drastically in the heartwood. QUER was accumulated preferentially around the transition zone, and its content was relatively low in the outer part of the sapwood and in the inner part of the heartwood. E2 content was low in the sapwood and increased in the heartwood. The heartwood formation was marked by the accumulation of new soluble compounds. The relationship between wood extractives and wood colour were evaluated and discussed. HJG was considered to be a major precursor of heartwood colour providing chromophores through hydrolysis (deglucosylation), oxidation and polymerization processes. Paracoccidioides brasiliensis components interact with host cells and can influence the pathogenesis of paracoccidioidomycosis (PCM). Among the components released by P. brasiliensis, gp 43 and a heavily glycosylated antigen with MM>160 kDa are the most recognized by serum antibodies from patients with PCM. In order to isolate the high MM glycoconjugate, we carried out affinity chromatography of a crude exoantigen preparation on immobilized jacalin. The bound fraction (JBE, jacalin binding exoantigen) consisted of a major antigen of high MM and frequently of an additional 70-kDa minor protein. This protein, designated paracoccin, exhibited selective binding to immobilized GlcNAc, a property that was used for its purification. The structural data of paracoccin obtained by mass spectrometry of tryptic peptides did not match any known protein. Anti-paracoccin serum localized the lectin on the surface of P. brasiliensis yeasts, especially in the budding regions. Paracoccin was able to interact with laminin in a dose-dependent manner. This interaction was inhibited by GlcNAc, followed by D-glucose and D-mannose, but not by D-galactose, N-acetyl-galactosamine or L-fucose. Interestingly, paracoccin induced both resident and elicited mouse peritoneal cavity macrophages to release high and persistent levels of TNF-alpha in vitro, a fact that was associated with high nitric oxide production in elicited cells. Because binding to laminin can favor yeast adhesion and invasion of host tissues, and overproduction of NO has been associated with suppression of cell immunity, paracoccin is suggested to play an important role in PCM pathogenesis.  相似文献

In order to assess the relevance of conformation for the antigenic site A of the hemagglutinin of influenza virus we synthesized two peptides, comprising two variant sequences of the central part of site A (amino acids 140 - 146 of subunit HA1) inserted into an artificial peptide skeleton, which imposes a loop-like structure on the respective sequence stretch. Assuming that the loop structure in the synthetic peptides would roughly approximate to the structure of the cognate protein sequence we tried to raise protein-reactive anti-peptide antibodies. The antibodies obtained indeed showed reactivity against influenza virus, although the discriminating specificity with regard to a mutation at position 144 was lost for virus binding in contrast to the highly specific peptide binding. Considering the failures in raising anti-hemagglutinin antibodies against the site A by immunization with short flexible peptide our results support the hypothesis that conformation makes a major contribution to the immunogenic and antigenic characteristics of site A in influenza hemagglutinin.  相似文献

Extractable materials from some timber species have been identified which prevent wood decay; however, little has been reported on the effect(s) of such materials against mould species that colonize timber. With increasing interest in the use of Trichoderma species, both as agents of permeability enhancement and biological control, more information is required on how chemical components within fresh and processed timber influence growth of Trichoderma. Fresh and dried samples of Scots pine and Sitka spruce sapwood and heartwood were leached in a Soxhlet apparatus and the resulting extract was combined with malt extract agar and inoculated with Trichoderma. Trichoderma isolates were inhibited to varying degrees by extractives removed from fresh and dried heartwood of the two timbers. Growth on sapwood extractives, however, showed a lesser degree of inhibition. The implications of the results for applications of Trichoderma in timber are discussed.  相似文献
