The antimicrobial properties of acetone, methanol, and aqueous extracts of the lichens Lasallia pustulata, Parmelia sulcata, Umbilicaria crustulosa, and Umbilicaria cylindrica were studied comparatively in vitro. Antimicrobial activities of the extracts of different lichens were estimated by the disk diffusion test for Gram-positive bacteria, Gram-negative bacteria, and fungal organisms, as well as by determining the MIC (minimal inhibitory concentration). The obtained results showed that the acetone and methanol extracts of Lasallia pustulata, Parmelia sulcata, and Umbilicaria crustulosa manifest antibacterial activity against the majority of species of bacteria tested, in addition to selective antifungal activity. The MIC of lichen extracts was lowest (0.78 mg/ml) for the acetone extract of Lasallia pustulata against Bacillus mycoides. Aqueous extracts of all of the tested lichens were inactive. Extracts of the lichen Umbilicaria cylindrica manifested the weakest activity, inhibiting only three of the tested organisms.  相似文献

The present work demonstrates the screening of extracts of the rare medicinal herb Euphorbia fusiformis for antifungal activity. The main aim was to investigate its antifungal properties against Candida albicans and Cryptococcus neoformans, the causative agents of human candidiasis and cryptococcosis, respectively. Aqueous and organic solvent extracts from the leaves and rootstock of the plant were tested against the fungi by the well-in-agar method. Almost all the organic solvent extracts exhibited an inhibitory effect against C. albicans and to some extent on C. neoformans, except for the aqueous extracts, which had no effect. The combined formulations of the extracts also had better activity against C. albicans than C. neoformans. This study thus concludes by demonstrating the antifungal properties of E. fusiformis and also the potential research in identifying the active principles, which may have future therapeutic value.  相似文献

Synergistic combinations of antimicrobial agents with different mechanisms of action have been introduced as more successful strategies to combat infections involving multidrug resistant (MDR) bacteria. In this study, we investigated synergistic antimicrobial activity of Camellia sinensis and Juglans regia which are commonly used plants with different antimicrobial agents. Antimicrobial susceptibility of 350 Gram-positive and Gram-negative strains belonging to 10 different bacterial species, was tested against Camellia sinensis and Juglans regia extracts. Minimum inhibitory concentrations (MICs) were determined by agar dilution and microbroth dilution assays. Plant extracts were tested for synergistic antimicrobial activity with different antimicrobial agents by checkerboard titration, Etest/agar incorporation assays, and time kill kinetics. Extract treated and untreated bacteria were subjected to transmission electron microscopy to see the effect on bacterial cell morphology. Camellia sinensis extract showed higher antibacterial activity against MDR S. Typhi, alone and in combination with nalidixic acid, than to susceptible isolates.” We further explore anti-staphylococcal activity of Juglans regia that lead to the changes in bacterial cell morphology indicating the cell wall of Gram-positive bacteria as possible target of action. The synergistic combination of Juglans regia and oxacillin reverted oxacillin resistance of methicillin resistant Staphylococcus aureus (MRSA) strains in vitro. Marine biofilms are a virtually untapped source of bioactive molecules that may find application as novel antifoulants in the marine paint industry. This study aimed at determining the potential of marine biofilm bacteria to produce novel biomolecules with potential application as natural antifoulants. Nine representative strains were isolated from a range of surfaces and were grown in YEB medium and harvested during the late exponential growth phase. Bacterial biomass and spent culture medium were extracted with ethanol and ethyl acetate, respectively. Extracts were assayed for their antifouling activity using two tests: (1) antimicrobial well diffusion test against a common fouling bacterium, Halomonas marina, and (2) anti-crustacean activity test using Artemia salina. Our results showed that none of the ethanolic extracts (bacterial biomass) were active in either test. In contrast, most of the organic extracts had antimicrobial activity (88%) and were toxic towards A. salina (67%). Sequencing of full 16 S ribosomal DNA analysis showed that the isolates were related to Bacillus mojavensis and Bacillus firmus. Matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF-MS) profiling of ethyl acetate extracts of culture supernatants showed that these species produce the bioactive lipopeptides surfactin A, mycosubtilin and bacillomycin D.  相似文献

Many doubts still persist even today when it comes to selection of the solvents for extracting the active constituents from various Indian medicinal plants. This study was aimed at assessing and establishing the best solvent for extracting the active constituents from 10 plant extracts. Thin layer chromatography (TLC) was used to separate and establish the active constituents present in each of the medicinal plants. Active constituents from each plant were extracted by using three different solvent systems namely diethyl ether, chloroform and hexane and were tested against three species of gram negative and three species of gram positive bacteria (Escherichia coli, Pseudomonas aureginosa, Streptococcus pneumoniae, Aeromonas hydrophila, Staphylococcus aereus, Bacillus cereus) by means of agar well diffusion assay. Studies on the antioxidant activity studies were also carried out for these plant extracts by using Diphenylpicryl-hydrazyl (DPPH) method. For the antimicrobial activity, the study revealed that among the selected plants, Azadiracta indica, Pongamia pinnata, Aloe barbadensis had the maximum antibacterial activity. Among the extraction procedures diethyl ether was found to be the best solvent that could be used for the extraction procedure. On the antioxidant activity part, Coleus amboinicus and Calotropis procera were found to have high antioxidant activity of 91.64% and 88.72% respectively and the further results are reported and discussed.  相似文献

We investigated seasonal variations in allelopathic potential ofArtemisia princeps var.orientalis. Aqueous and meth-anol extracts and volatile substances were prepared in the laboratory from samples collected monthly (April through October). Their impacts were then assessed on the germination and seedling growth ofLactuca sativa andAchyranthes japonica. The allelopathic potential varied with the time of sample collection and the concentration tested. For example, germination ofL. sativa was not inhibited by the aqueous extract but seedling growth (shoots and roots) was, with its seasonal effect being significant. ForA. japonica, seed germination was not inhibited at lower concentrations (except for August samples). However, at higher concentrations and in certain months (especially July), germination was more negatively affected. The degree of seedling growth inhibition also differed by month and by extract concentration, with roots being impacted more than shoots. Volatile substances also had a time-dependent influence on the germination and seedling elongation ofA. japonica. In a separate experiment, the ethyl-acetate and water fractions of a crude methanol extract were prepared monthly fromA. princeps var.orientalis. Here, we examined their antimicrobial activities against three gram-positive bacteria (Bacillus cereus, Bacillus subtilis, andStaphylococcus aureus), two gramnegative bacteria (Escherichia coli andPseudomonas fluorescens), and one lactic acid bacterium,Lactobacillus plantar urn. The ethyl-acetate fraction that was sampled in September was remarkably potent againstB. cereus andB. subtilis, whereas the water fraction collected in August and September showed great antimicrobial activity against the grampositive and -negative bacteria. In contrast,L. plantarum was not inhibited by the water fraction, regardless of the sampling month. Likewise, the ethyl-acetate and water fractions collected in April and October had the lowest levels of antimicrobial activity.  相似文献

Summary The Juglans regia x Juglans nigra hybrid cannot be obtained because of stable interspecific barriers. We have analyzed and described various morphological and biochemical characters in J. regia and J. nigra pollen: differences in the mineral composition, polysaccharide reserves, wall ultrastructure and protein pattern of soluble cytoplasm, membranes, and cell-wall fractions. The absence of a glycoprotein fraction in the wall of J. nigra suggests that the regulation of critical steps in the interspecific matching with female tissues could be affected.  相似文献

Antibacterial effects of knotwood extractives on paper mill bacteria   总被引:2,自引:0,他引:2  
Hydrophilic knotwood extracts from 18 wood species were assessed in disc diffusion and liquid culture tests for antibacterial effects against three species of paper mill bacteria. The Pinus sylvestris, P. resinosa, P. contorta, and P. banksiana extracts decreased or inhibited bacterial growth. The susceptibility order was P. sylvestris > P. resinosa > P. contorta > P. banksiana, correlating with the concentrations of pinosylvin and pinosylvin monomethyl ether in these wood species. Also, Pseudotsuga menziesii and Thuja occidentalis extracts had a small inhibitory effect. The Gram-positive Bacillus coagulans was more susceptible to the extracts than the Gram-negative Burkholderia multivorans and Alcaligenes xylosoxydans. The main components in the Pinus knotwood extracts were pinosylvin monomethyl ether and pinosylvin, suggesting these to be the active components. Therefore, pure pinosylvin, pinosylvin monomethyl ether, and dihydro-pinosylvin monomethyl ether were also tested. All compounds showed antibacterial effects. However, higher concentrations were needed for these pure compounds than for the knotwood extracts. Pinosylvin had stronger antibacterial effects than pinosylvin monomethyl ether. This work shows that knotwood extracts, especially from Pinus species, have a potential for use as natural biocides in papermaking.  相似文献

为考察中草药抗菌物质基础筛选出活性提取物,该研究通过80%乙醇冷浸和95%乙醇回流提取制备23种中草药的提取物,采用琼脂扩散法测量抑菌圈直径,用微量液体培养基倍比稀释法测定最低抑菌浓度(minimum inhibitory concentration,MIC)和最低杀菌浓度(minimum bactericidal/fungicidal concentration,MBC/MFC),并测定了提取物对临床4种常见病原菌体外抗菌活性。结果表明:紫珠草、千斤拔、黄龙尾等9种中草药对金黄色葡萄球菌有较强的抑菌活性,其MIC/MBC值除个别菌是12.5 mg·mL~(-1)外,其他都在0.09~3.12 mg·mL~(-1)之间;千斤拔、大红袍、过江龙等5种中草药对铜绿假单胞菌有较强抑菌活性,其MIC/MBC值在3.12~12.5 mg·mL~(-1)之间;紫珠草、千里光、石楠等13种中草药对大肠埃希菌有较强的抑菌活性,其MIC/MBC值在0.09~6.25 mg·mL~(-1)之间;八角对白色念珠菌有较强抑菌活性,其MIC/MFC值在0.78~12.5 mg·mL~(-1)之间。23种中草药的抗细菌活性较好,尤其是千斤拔、大红袍、过江龙、八角、黄药子对金黄色葡萄球菌、大肠埃希菌、铜绿假单胞菌都具有较好的抑菌活性,具有广谱抗菌活性;但对真菌抑菌效果不明显,仅有八角对白色念珠菌有抑菌活性。此外,提取溶剂浓度、提取温度和提取时间对中草药的提取率和活性均有影响,冷提稍优于热提。  相似文献

The aqueous, ethyl acetate, methanolic and Total Oligomer Flavonoids (TOF) enriched extracts, obtained from the aerial parts of Cyperus rotundus, were investigated for their contents in phenolic compounds. Antioxidative activity using the NBT/riboflavin assay system, antimicrobial activity against Gram positive and Gram negative bacterial reference strains as well as antigenotoxic activity tested with the SOS chromotest assay were also studied. Significant antibacterial activity against reference strains; Staphylococcus aureus, Enterococcus faecalis, Salmonella enteritidis and Salmonella typhimurium, was detected in the presence of ethyl acetate and TOF enriched extracts. In addition to their antimicrobial activity, the same extracts showed a significant ability to inhibit nitroblue tetrazolium reduction by the superoxide radical in a non enzymatic O2.− generating system, and were also able to reduce significantly the genotoxicity induced by nifuroxazide and Aflatoxin B1. The antioxidant, antimicrobial and antigenotoxic activities exhibited by C. rotundus depend on the chemical composition of the tested extracts.  相似文献

为探明爵床(Justicia procumbens)甲醇提取物对小菜蛾的生物活性,采用室内生测法测定了爵床甲醇提取物对小菜蛾的触杀、拒食、胃毒、生长发育抑制和产卵忌避作用。结果表明,爵床甲醇提取物对小菜蛾幼虫具有较强的触杀、拒食、胃毒和生长发育抑制活性,对小菜蛾成虫具有较强的产卵忌避活性。在触杀试验中,药后1、2 d和3 d爵床甲醇提取物对小菜蛾3龄幼虫的致死中浓度(LC50)分别为5.17、4.05和3.06 mg/m L;在拒食试验中,药后1 d和2 d提取物对3龄幼虫的选择性拒食中浓度(AFC50)分别为2.64和3.13 mg/m L,药后1 d和2 d提取物对3龄幼虫的非选择性拒食中浓度(AFC50)分别为3.70、4.54 mg/m L;在胃毒试验中,药后4、5、6 d和7 d提取物对3龄幼虫的致死中浓度(LC50)分别为8.13、3.65、2.88、2.23 mg/m L;在生长发育抑制试验中,药后1 d和2 d提取物对3龄幼虫的抑制中浓度(IC50)分别为2.02、1.40 mg/m L;在产卵忌避试验中,药后1、2 d和3 d提取物对小菜蛾成虫的选择性产卵忌避中浓度(AOC50)分别为2.61、3.66、4.58 mg/m L,药后1、2和3 d提取物对小菜蛾成虫的非选择性产卵忌避中浓度(AOC50)分别为3.19、4.52、5.65 mg/m L。由此证实,爵床提取物对小菜蛾具有显著的毒杀活性,具有开发为新型高效、低毒植物源农药的潜在价值。  相似文献

为了探究外来红树植物拉关木对乡土红树植物的化感作用,该研究观察了不同浓度(0.1、0.3、0.5g·mL~(-1))的拉关木根、叶水浸提液对乡土红树植物桐花树和正红树的胚轴(种子)萌发、幼苗生长及叶片抗氧化酶活性的影响。结果表明:(1)拉关木水浸提液对桐花树种子的成苗率、萌发指数和根长均存在抑制作用,其中对根长的抑制作用随水浸提液浓度的提高而增强。(2)根水浸提液对桐花树幼苗的根长、苗高、生物量等生长指标的影响总体上均表现为低浓度促进,高浓度抑制。(3)拉关木水浸提液对正红树胚轴的萌发率、萌发指数、生长指标均表现为促进作用,且根水浸提液0.1、0.3 g·mL~(-1)处理组的芽长以及根、叶水浸提液0.1、0.3 g·mL~(-1)处理组的生物量显著大于对照组;拉关木水浸提液对正红树幼苗的生物量也表现为促进作用。(4)抗性生理方面,随着拉关木水浸提液浓度的升高,桐花树和正红树幼苗SOD活性降低,正红树幼苗POD活性在根水浸提液0.3 g·mL~(-1)和叶水浸提液0.1 g·mL~(-1)处理组显著高于对照组。以上结果表明,不同乡土植物对拉关木化感作用的敏感性不同,拉关木水浸提液抑制了桐花树的生长,而对正红树的生长则表现出一定程度的促进作用。  相似文献

以采自农田中自然生长的植物群落中的香丝草为供体,以典型的双子叶植物蚕豆和典型的单子叶植物玉米的幼苗为受体,运用根尖微核试验和染色体畸变试验,研究了香丝草的根、茎、叶和幼果4种器官水浸提液对受体的遗传毒性。结果表明:(1)在香丝草不同器官水浸提液作用下,蚕豆和玉米根尖细胞的有丝分裂各时期均受到明显影响,细胞中出现了微核、染色体桥、染色体断片、染色体环、染色体粘连及染色体滞后等多种染色体畸变。(2)香丝草各器官水浸提液对蚕豆幼苗根尖细胞分裂的抑制作用明显大于玉米。(3)香丝草各器官水浸提液对蚕豆和玉米幼苗根尖的染色体畸变诱导存在显著的浓度效应,即水浸提液浓度越高,受体的微核率和畸变率越高,相应的有丝分裂指数越低,水浸提液的诱导作用与浓度呈正相关关系,但不是简单的加和作用。(4)香丝草各器官水浸提液均具有较强的遗传毒性,但整体化感效应表现为叶>幼果>茎>根,即叶片产生的化感作用最强。因此,香丝草分泌的化感物质可能通过对受体植物生长点的细胞有丝分裂和细胞形态产生影响,造成受体植物染色体的多种畸变和不可逆的遗传损伤,从而成功入侵新的栖息地。  相似文献

Among 67 endophytic fungi isolated from Quercus variabilis, 53.7% of endophytic fungal fermentation broths displayed growth inhibition on at least one test microorganism, such as pathogenic fungi (Trichophyton rubrum, Candida albicans, Aspergillus niger, Epidermophyton floccosum, Microsporum canis) and bacteria (Escherichia coli, Bacillus subtilis, Pseudomonas fluorescens). Moreover, 19.4% of strains showed a broader antimicrobial spectrum, such as Aspergillus sp., Penicillium sp., Alternaria sp., 20.9% of strains showed strong inhibition (+++) to pathogenic bacteria, while only 7.5% displayed that to test fungi. The most active antifungal strain I(R)9-2, Cladosporium sp. was selected and fermented. From the broth, a secondary metabolite, brefeldin A was obtained. This is the first report on the antimicrobial potentials of endophytic fungi residing in Q. variabilis and isolation of brefeldin A produced by Cladosporium sp.  相似文献

A total of 48 strains were isolated from the normal tissues of Malus halliana and the EtOAc extracts of their cultures were subjected to primary antimicrobial screening against four test bacteria and three fungi. As a result, 22 strains exhibited antimicrobial activity against at least one test microbe. Among them, Alternaria brassicicola ML-P08 showing strong activity (MICs: 0.31–2.50 mg/ml) was selected for further investigation on its secondary metabolites. Bioassay-guided fractionation of the EtOAc extract of its liquid culture afforded seven compounds, which were identified as alternariol (1), alternariol 9-methyl ether (2), altechromone A (3), herbarin A (4), cerevisterol (5), 3β,5α-dihydroxy-(22E,24R)-ergosta-7,22-dien-6-one (6) and 3β-hydroxy-(22E,24R)-ergosta-5,8,22-trien-7-one (7), respectively, by spectral means (MS, IR, 1H- and 13C-NMR). In vitro antimicrobial assay showed that compound 3 was substantially active against Bacillus subtilis, Escherichia coli, Pseudomonas fluorescens and Candida albicans with the MICs of 3.9, 3.9, 1.8, and 3.9 μg/ml, respectively. Compound 4 also showed pronounced antifungal activity against Trichophyton rubrum and C. albicans with MICs of both 15.6 μg/ml. In addition, compound 1 exhibited strong xanthine oxidase inhibitory activity with the IC50 of 15.5 μM, comparable to that of positive control, allopurinol (IC50: 10.7 μM).  相似文献

Vegetation history for the study region is reconstructed on the basis of pollen, charcoal and AMS14C investigations of lake sediments from Lago del Segrino (calcareous bedrock) and Lago di Muzzano (siliceous bedrock). Late-glacial forests were characterised byBetula andPinus sylvestris. At the beginning of the Holocene they were replaced by temperate continental forest and shrub communities. A special type of temperate lowland forest, withAbies alba as the most important tree, was present in the period 8300 to 4500 B.P. Subsequently,Fagus, Quercus andAlnus glutinosa were the main forest components andA. alba ceased to be of importance.Castanea sativa andJuglans regia were probably introduced after forest clearance by fire during the first century A.D. On soils derived from siliceous bedrock,C. sativa was already dominant at ca. A.D. 200 (A.D. dates are in calendar years). In limestone areas, however,C. sativa failed to achieve a dominant role. After the introduction ofC. sativa, the main trees were initially oak (Quercus spp.) and later the walnut (Juglans regia). Ostrya carpinifolia became the dominant tree around Lago del Segrino only in the last 100–200 years though it had spread into the area at ca. 5000 cal. B.C. This recent expansion ofOstrya is confirmed at other sites and appears to be controlled by human disturbances involving especially clearance. It is argued that these forests should not be regarded as climax communities. It is suggested that under undisturbed succession they would develop into mixed deciduous forests consisting ofFraxinus excelsior, Tilia, Ulmus, Quercus andAcer.  相似文献

Various parts of Datura innoxia were examined for potential antibacterial activity by preparing their crude aqueous and organic extracts against Gram-negative bacteria (Escherichia coli and Salmonella typhi) and Gram-positive bacteria (Bacillus cereus, Bacillus subtilis and Staphylococcus aureus). The results of agar well diffusion assay indicated that the pattern of inhibition depends largely upon the plant part, solvent used for extraction and the organism tested. Extracts prepared from leaves were shown to have better efficacy than stem and root extracts. Organic extracts provided potent antibacterial activity as compared to aqueous extracts. Among all the extracts, methanolic extract was found most active against almost all the bacterial species tested. Gram-positive bacteria were found most sensitive as compared to Gram-negative bacteria. Staphylococcus aureus was signifi cantly inhibited by almost all the extracts even at very low MIC followed by other Gram-positives. For Escherichia coli (a Gram-negative bacterium), the end point was not reached for ethyl acetate extract while it was very high for other extracts. The study promises an interesting future for designing a potentially active antibacterial agent from Datura innoxia.  相似文献

以本氏针茅及其优势种大针茅和百里香种子为受体,采用生物检测的方法研究了0(对照)、0.1、0.2和0.5g/mL本氏针茅根际土甲醇浸提液和水浸提液对3种受体种子萌发的影响,以探究草地演替过程中物种间的生态关系和化感作用的表现形式。结果显示:(1)甲醇浸提液对3种受体种子的发芽均有抑制作用;水浸提液对本氏针茅发芽有促进作用,对百里香有部分促进作用,而对大针茅有抑制作用。(2)甲醇浸提液对本氏针茅和大针茅胚根和胚芽生长无显著影响,而抑制百里香胚根和胚芽的生长;水浸提液对本氏针茅胚根生长无显著影响,对本氏针茅胚芽生长具有显著促进作用,而对其它两种受体胚根和胚芽均无显著影响。(3)各种浓度甲醇浸提液对大针茅和百里香生长表现出了化感综合抑制效应,对本氏针茅生长于0.2g/mL时表现出化感综合抑制效应,其它2种浓度(0.1、0.5g/mL)表现为化感综合促进效应;水浸提液对本氏针茅和百里香表现为化感综合促进效应,而对大针茅表现为化感综合抑制效应。研究表明,本氏针茅根际土甲醇浸提液和水浸提液的化感物质存在差异;不同受体对同一浸提液反应不同,同一受体对不同浸提液的反应也不同。  相似文献

为鉴定引起四川盆地地区核桃黑斑病的病原菌,采用组织分离法对病原菌进行分离,利用柯赫氏法则验证其致病性,依据菌株形态学和基于16S rDNA基因序列分析对病原菌进行鉴定;同时,利用分离的菌株对18个栽培品种(无性系)进行抗病性评价。结果表明,分离菌株的菌落形态与黄单胞杆菌属(Xanthomonas)相似,其16S rDNA序列与树生黄单胞杆菌(X. arboricola)的(登录号为KP340804.1)同源性高达99%,因此,将引起四川盆地地区核桃黑斑病的病原菌鉴定为树生黄单胞杆菌。18个核桃栽培品种(无性系)的田间侵染发病率和病情指数分别为35.07%~78.57%和17.71%~51.96%,变异系数分别为17.62%和28.78%,并以此为基础评价出5个高抗病品种(无性系)。这为核桃黑斑病致病机理研究和抗病新品种的选育奠定基础。  相似文献
