唾液酸广泛存在于生物体内,常存在于糖复合物的糖链末端,对组织器官特别是脑神经的发育至关重要,并且与多种疾病的发生和发展过程密切相关.唾液酸被唾液酸酶水解后,可改变糖复合物的构象从而调控相关因子的生物学功能.在目前发现的4种唾液酸酶中,对Neu1的研究较为深入.研究表明,Neu1的剪切底物具有多样性,其与细胞表面受体的结构和功能调节密切相关.随着研究的深入,Neu1逐渐被认为是唾液酸介导的调控疾病发生过程中的重要因子,Neu1在人类疾病中的作用比预想的还要深远.本篇综述了在已有总结的唾液酸酶性质和生理病理学功能的基础上,概述了近年来Neu1的研究进展,并对其作用及与不同细胞表面受体的相互作用机制做了总结.  相似文献

非人源性唾液酸N-羟乙酰神经氨酸(N-glycolylneuraminic acid, Neu5Gc)是红肉中潜在的致癌性因子。唾液酸转移酶是涉及转运其前体物质的关键酶之一。大鼠用不同浓度的山奈酚(kaempferol,KA)和槲皮素(quercetin,Qu)灌胃,并模拟宰前对Neu5Gc合成影响的结果表明,不同浓度的KA、Qu对大鼠体内肌肉组织、肝和肾中Neu5Gc的含量均有一定的影响,最大抑制率分别为36.44%±0.11%和33.37%±0.08%。为探究其抑制机制,采用唾液酸转移酶(sialyltransferase,ST)与KA、Qu进行分子对接和分子动力学模拟分析。分子对接复合物二维相互作用图表明:KA和Qu能有效占据ST特异性抑制剂5′-胞苷单磷酸的活性位点氨基酸残基;对最佳复合物进行200 ns的动力学模拟结果显示,ST和KA产生稳定氢键作用的氨基酸残基主要是His301和Gly298,与Qu产生氢键作用的氨基酸残基主要是Gly293、Glu324、Thr272和Ser276。结合自由能分析表明,范德华力、静电吸引作用对抑制过程具有重要作用。本研究为合成和筛选高效ST抑制剂提供了一定实验依据。  相似文献

为了研究乳腺癌发生与发展过程中突变型p53和Neu基因之间的相互作用,我们构建了p53/Neu双转基因鼠。用5只成年雄性Neu转基因鼠与10只成年雌性p53转基因鼠交配,对150只雌性杂交后代鼠进行p53/Neu双转基因鼠的筛选。为此,本实验建立了PCR-一步鉴定法。即:在同一个反应管内能同时满足小鼠β-酪蛋白基因、p53基因和Neu基因上三个片段(分别为0.5kb、1.2kb、1.7kb)的扩增反应者被判定为双转基因鼠(Fig.1)。传统的Southern分析(Fig.2)与PCR一步法鉴定结果相吻合。证明:该法的准确率为100%,进一步、随机取样,Northern分析(Fig.3)表明:双转基因鼠(2号)在哺乳第二天即出现p53基因的高。继而、在姓娠第10、17天,哺乳第10、21天也检测到该基因的表达。另外,免疫组化染色也检测到了p53和Neu蛋白(未展示结果)。  相似文献

Summary The lectin from the elderberry (Sambucus nigra L.) bark, shown to recognize the sequence neuraminic acid (2,6) galactose/N-acetylgalactosamine, was applied for detecting binding sites in Lowicryl K4M sections by light and electron microscopy. The lectin was used either directly complexed to colloidal gold or in a two-step cytochemical affinity technique. The lectin-gold complex proved to be superior and thus was extensively tested on rat liver, kidney and hepatoma cells as well as on sheep and bovine submandibular glands. Controls to establish specificity of lectin-gold binding included sugar and glycoprotein inhibition tests and enzymic removal of sialic acid. In agreement with biochemical data demonstrating the potentiating effect of sialic acid on the binding of the lectin to oligosaccharides, enzymic removal of sialic acid from liver sections resulted in abolition of lectin staining. However, in the submandibular glands, neuraminidase pretreatment of the sections had no effect on the subsequent lectin-gold binding. In rat kidney some structures became negative while others retained the lectin-gold staining due to binding to penultimate.N-acetylgalactosamine exposed after sialic acid removal. In line with this, spot blot analysis demonstrated that the lectin-gold complex reacted with both fetuin and asialofetuin. Taken together, these results suggest that, for cytochemical staining, the sialic acid and the galactose/N-acetylgalactosamine lectin combining subsites ofSambucus nigra L. lectin are equally reactive with cellular glycoconjugates and that neuraminidase predigestion of tissue sections is of utmost importance to ensure specificity of staining for the sequence neuraminic acid (2,6) galactose/N-acetylgalactosamine.  相似文献

哺乳动物细胞内的某些蛋白质或脂类可以被糖基化修饰,而糖链末端往往存在唾液酸化的现象,催化添加唾液酸的酶为糖基转移酶(sialyltransferase,ST),而去除唾液酸的为唾液酸酶(sialidase,SA或称为neuraminidase,NEU).本实验检测了人膀胱正常上皮细胞HCV29、非浸润性膀胱癌细胞KK47和浸润性膀胱癌细胞YTS-1中唾液酸的表达,发现恶性肿瘤细胞中唾液酸的含量高于正常细胞;进一步分析唾液酸酶和唾液酸转移酶的表达,发现唾液酸酶Neu1在正常细胞中表达最高,在良性肿瘤细胞中次之,在恶性肿瘤细胞中表达最低,推测在膀胱癌中Neu1对唾液酸的异常表达起着主要作用.同时,膀胱癌细胞中Toll样受体1,2,3,4(toll-like receptors,TLRs)表达趋势也与Neu1一致.利用TGF-β处理HCV29,使之发生上皮间质转化(epithelial-mesenchymal transition,EMT),细胞中Neu1和TLR3表达明显减少;将Neu1基因沉默后,TLR3表达也明显减少.此外,在YTS-1细胞中过表达Neu1,TLR3表达增高且激活了下游NF-κB通路.这一结果说明膀胱癌中Neu1与TLR3的表达有着密切的关系,这为膀胱癌的分子机理研究提供了工作基础.  相似文献

5乙,5异戊巴比妥酸(Amytal)对鼠肝线粒体胆碱脱氢酶的抑制为竞争性,K_i为0.80×10~(-3)M。当胆碱脱氢酶被Triton X-100从膜上增溶下来后Amytal不再抑制溶解的胆碱脱氢酶。Amytal对鼠肝线粒体胆碱-细胞色素c还原酶的抑制为反竞争性,K_i为1.80×10~(-4)M。Amytal的这种抑制不受线粒体老化的影响,也不被维生素K_3所解除,说明不是继发于胆碱的氧化产物三甲铵乙醛的氧化被抑制的结果。  相似文献

Both the 5α, 6α- and 5β, 6β-dichloromethylene adducts (2a and 2b) of 3β-acetoxy-5-androsten-17-one (1) are produced when the latter is exposed to dichlorocarbene generated from chloroform and base by Phase Transfer Catalysis using ultrasound as a means of agitation. The 1H NMR substituent effects of 5α, 6α- and 5β, 6β-dichloromethylene on the angular methyl groups (Zürcher values) are given. Summary The Kurloff cell (KC), a natural killer lymphocyte, contains a large (10-m diameter) periodic acid-Schiff (PAS)-positive lysosome-like inclusion body called the Kurloff body (KB), which exhibits strong acid phosphatase activity. The presence of Sambucus nigra agglutinin (SNA)-reactive Neu5Ac(2,6)-D-Gal/Gal-NAc(1,4)GlcNAc oligosaccharide sequences and the absence of the corresponding Neu5Ac(2,3) Maackia amurensis agglutinin (MAA)-reactive sequence in the major 35-kDa N-glycosylproteins of the complex or hybrid type extracted from purified KC were established by Western-lectin-blotting of cytosolic extracts from purified KC. Moreover, these SNA-reactive sequences, or at least part of them, were shown to be borne by sialidase-sensitive KC acid isophosphatases. Thymic sections rich in KC, from estrogenized guinea pigs were examined by affino-histochemistry with these sialic acid-reactive lectins. The SNA-reactivity of thymic sections was quasiexclusively confined to KC clusters, whereas the whole thymic section was negative for MAA. KC were not SNA-reactive following preincubation and incubation with 200 mM lactose. When submitted to enzymatic or mild chemical desialylation processes, the SNA-reactivity of the KC clusters was enhanced. The SNA-reactivity of KC clusters was completely abolished following prolonged chemical desialylation, whereas the PAS-positivity of KB remained unchanged. Even after a prolonged sialidase treatment, this SNA-reactivity was only reduced. Moreover, after both these desialylation processes, KC developed a heavier Ricinus communis agglutinin-reactivity, thus confirming the presence of penultimate Gal residues in their abundant SNA-reactive oligosaccharide sequences Neu5Ac(2,6)Gal(1,4)GlcNAc. Such a selective lectin histochemical property provides a marker for detecting KC.  相似文献

Three kinds of diketopiperazines which have retarditive activity for the growth of plant seedlings and plant roots at concentrations ranging from 1 : 2,500 to 1 : 100,000, were isolated from the neutral fraction by extracting the cultured broth of Rosellinia necatrix. These three diketopiperazines have been proved to be l-prolyl-l-leucine anhydride, l-prolyl-l-valine anhydride and l-prolyl-l-phenylalanine anhydride respectively, and the last one seems to be a new diketo-piperazine.

Furthermore, a crystalline wax having m.p. 52°C, a physiologically inactive substance, was also isolated from the same neutral fraction and presumed to be the saturated hydrocarbon of n-pentacosane C25H52.  相似文献

Oxysterol sulfation plays an important role in regulation of lipid metabolism and inflammatory responses. In the present study, we report the discovery of a novel regulatory sulfated oxysterol in nuclei of primary rat hepatocytes after overexpression of the gene encoding mitochondrial cholesterol delivery protein (StarD1). Forty-eight hours after infection of the hepatocytes with recombinant StarD1 adenovirus, a water-soluble oxysterol product was isolated and purified by chemical extraction and reverse-phase HPLC. Tandem mass spectrometry analysis identified the oxysterol as 5-cholesten-3β, 25-diol, disulfate (25HCDS), and confirmed the structure by comparing with a chemically synthesized compound. Administration of 25HCDS to human THP-1-derived macrophages or HepG2 cells significantly inhibited cholesterol synthesis and markedly decreased lipid levels in vivo in NAFLD mouse models. RT-PCR showed that 25HCDS significantly decreased SREBP-1/2 activities by suppressing expression of their responding genes, including ACC, FAS, and HMG-CoA reductase. Analysis of lipid profiles in the liver tissues showed that administration of 25HCDS significantly decreased cholesterol, free fatty acids, and triglycerides by 30, 25, and 20%, respectively. The results suggest that 25HCDS inhibits lipid biosynthesis via blocking SREBP signaling. We conclude that 25HCDS is a potent regulator of lipid metabolism and propose its biosynthetic pathway.  相似文献

5-Fluoro-2'-deoxyuridine (floxuridine, 5-FdUrd) and 5-fluorouracil (5-FU) are widely used for the treatment of colorectal cancers. The mechanisms of action of 5-FdUrd and 5-FU, as well as the biochemical pathway responsible for their metabolism, are well understood. Identification of every metabolite and achieving mass balance by conventional UV absorption-based HPLC analysis are not feasible because the metabolites beyond 5-FU in the 5-FdUrd metabolic pathway are undetectable by UV light. We therefore established a mass spectrometry method, designed for fast and convenient analysis, for simultaneously measuring 5-FdUrd, 5-FU, and their metabolites. Linearity, precision and accuracy were validated in the concentration ranges studied for each compound. Hydrolysis studies of 5-FdUrd and amino acid mono ester prodrugs of 5-FdUrd in Capan-2 cell homogenates were carried out and the achievement of mass balance was established with this method (recovery of 5'-O-l-leucyl-FdUrd was 96.6-108.2% and that of 5-FdUrd was 79.4-117.4%). This simple LC-MS method achieves reliable quantitation and mass balance of 5-FdUrd, 5-FU, and their metabolites and can be effectively utilized for further kinetic studies.  相似文献

The intestinal pathogen Escherichia coli serotype O104:H4 (ECO104) can cause bloody diarrhea and haemolytic uremic syndrome. The ECO104 O antigen has the unique repeating unit structure [4Galα1–4Neu5,7,9Ac3α2–3Galβ1–3GalNAcβ1-], which includes the mammalian sialyl-T antigen as an internal structure. Previously, we identified WbwC from ECO104 as the β3Gal-transferase that synthesizes the T antigen, and showed that α3-sialyl-transferase WbwA transfers sialic acid to the T antigen. Here we identify the wbwB gene product as a unique α1,4-Gal-transferase WbwB that transfers Gal from UDP-Gal to the terminal sialic acid residue of Neu5Acα2–3Galβ1–3GalNAcα-diphosphate-lipid acceptor. NMR analysis of the WbwB enzyme reaction product indicated that Galα1-4Neu5Acα2–3Galβ1–3GalNAcα-diphosphate-lipid was synthesized. WbwB from ECO104 has a unique acceptor specificity for terminal sialic acid as well as the diphosphate group in the acceptor. The characterization studies showed that WbwB does not require divalent metal ion as a cofactor. Mutagenesis identified Lys243 within an RKR motif and both Glu315 and Glu323 of the fourth EX7E motif as essential for the activity. WbwB is the final glycosyltransferase in the biosynthesis pathway of the ECO104 antigen repeating unit. This work contributes to knowledge of the biosynthesis of bacterial virulence factors.  相似文献

By means of the reaction between VCl3 and benzofuroxan the compound VCl3·2C6H4N2O2 was obtained, while in reaction with TiX4 (XCl, Br) the compounds with stoichiometry TiX4·C6H4N2O2 (X Cl, Br) were obtained.Furthermore, with some benzofuroxan derivatives (5-methyl, 5-chloro and 5-methoxi) the complexes MCl4·2YC6H3N2O2 (MTi, V; YCH3O, Cl, CH3), TiCl4·YC6H3N2O2 (YCH3O, CH3) and TiBr4·YC6H3N2O2 (YCH3O, Cl, CH3) have been isolated.The compounds were characterized by elementary analysis, cryoscopic molecular weight determination in nitrobenzene, magnetic measurements and IR, Vis and EPR spectroscopy.  相似文献
