Normal human peripheral blood granulocytes which are tagged with 1-fluoro-2,4-dinitrobenzene (DNFB) are agglutinated by concanavalin A (ConA) in a way which resembles the pattern of reactivity displayed by leukemic cells. The present study further defines this reaction. The binding of ConA to untagged and DNP-tagged granulocytes, treated with DNFB at a ratio of 1011 molecules/cell, was quantified by isotopic dilution experiments employing [3H]ConA. Similar amounts of the lectin were bound to untagged and DNP-tagged cells following incubation for 5 min at 4 °C or 30 min at 24 °C: 1.1 × 105 molecules/cell, 4.6 × 1022 of surface area, and 1.6 × 103/μg of protein. The binding of [3H]ConA to both untagged and DNP-tagged cells was inhibited to the same degree by α-methylglucopyranoside (α-MG). Fixation with either glutaraldehyde or formaldehyde, which immobilizes ConA receptor sites, completely inhibited the agglutination of both untagged and DNP-tagged cells although lectin binding was unchanged. This suggests that the inhibition of agglutination was not due to the blocking of ConA-binding sites by aldehyde groups but rather to the immobilization of lectin receptors. We conclude that dinitrophenylation of normal granulocytes facilitates the rearrangement of lectin receptors in a way which resembles the ConA-induced clustering of sites which have been observed with malignant and transformed cells.  相似文献

L5178 mouse lymphoma cells were treated with the mismatching agent 6-hydroxy-aminopurine (HAP), a base analogue known to produce forward and reverse mutations in bacteria. Mutants with the phenotype deficient in hypoxanthine guanine phosphoribosyl transferase (HPRT) were selected in 6-thioguanine (TG)-containing medium and isolated. Reverse mutations to Hhe HPRT-proficient phenotype oc occuredd both spontaneously and after treatment with ethyl nitrosourea (ENU), which suggested that the initial HAP treatment had induced point mutations at the HPRT locus.

Reconstruction experiments, in which a small number of wild-type cells together with different numbers of mutant cells were seeded in HAT medium, indicated that densities up to 106 cells per ml can be used for the selection of revertants. Optimal expression of induced revertants was obtained two days after treatment.

The dose-response relationship for induction of reverse mutations by ENU appears not to deviate from linearity. The highest revertant frequency observed was 3.3 × 10−5 at an ENU concentration of 1 mM. The spontaneous reversion frequency per generation — based on 3 spontaneous revertants — was estimated to be 1.3 × 10−9. All revertants were indistinguishable from the parental wild-type line with respect to the activity as well as the electrophoretic mobility of HPRT.  相似文献

Chinese hamster ovary cells cultured in the presence of sodium butyrate decrease their growth rate, increase in length, and tend to grow in a monolayer. The addition of the same concentrations of sodium isobutyrate are completely ineffective. The growth alterations produced by 5 × 10−4 M sodium butyrate are similar to those caused by dibutyryl adenosine 3′:5′-cyclic monophosphate treatment.  相似文献

KB cell cultures exposed to 10−4 M dibutyryl cyclic AMP were significantly inhibited and exhibited contact inhibition of growth at cell densities of 8 × 104/cm2 irrespective of the initial plating density. Control cultures reached densities of 2.5 × 105/cm2. Inhibition of growth did not occur in KB cells when the density was below 1 × 104 cells/cm2. When dibutyryl cyclic AMP was removed from KB cells in the contact-inhibited state, growth resumed with DNA synthesis beginning in about 6 h. Labeled metaphases increased rapidly after 22 h without the appearance of an early rise in unlabeled metaphases. This suggests that the inhibitory effect of dibutyryl cyclic AMP is on the G1 phase of the cell cycle.  相似文献

It is well recognized that estradiol (E2) is one of the most important hormones supporting the growth and evolution of breast cancer. Consequently, to block this hormone before it enters the cancer cell or in the cell itself, has been one of the main targets in recent years. In the present study we explored the effect of the progestin, nomegestrol acetate, on the estrone sulfatase and 17β-hydroxy-steroid dehydrogenase (17β-HSD) activities of MCF-7 and T-47D human breast cancer cells. Using physiological doses of estrone sulfate (E1S: 5 × 10−9 M), nomegestrol acetate blocked very significantly the conversion of E1S to E2. In the MCF-7 cells, using concentrations of 5 × 10−6 M and 5 × 10−5 M of nomegestrol acetate, the decrease of E1S to E2 was, respectively, −43% and −77%. The values were, respectively, −60% and −71% for the T-47D cells. Using E1S at 2 × 10−6 M and nomegestrol acetate at 10−5 M, a direct inhibitory effect on the enzyme of −36% and −18% was obtained with the cell homogenate of the MCF-7 and T-47D cells, respectively. In another series of studies, it was observed that after 24 h incubation of a physiological concentration of estrone (E1: 5 × 10−9 M) this estrogen is converted in a great proportion to E2. Nomegestrol acetate inhibits this transformation by −35% and −85% at 5 × 10−7 M and 5 × 10−5 M, respectively in T-47D cells; whereas in the MCF-7 cells the inhibitory effect is only significant, −48%, at 5 × 10−5 M concentration of nomegestrol acetate. It is concluded that nomegestrol acetate in the hormone-dependent MCF-7 and T-47D breast cancer cells significantly inhibits the estrone sulfatase and 17β-HSD activities which converts E1S to the biologically active estrogen estradiol. This inhibition provoked by this progestin on the enzymes involved in the biosynthesis of E2 can open new clinical possibilities in breast cancer therapy.  相似文献

Mouse lymphoma cells (L5178Y) exposed to hypertonic media for 1 h behave as osmometers, but in hypotonic media, after initial swelling, they shrink back to normal volume and maintain it for long periods of time. The lower limit of osmolarity at which this “volume adaptation” will occur lies between 140 and 185 mosM. The “volume adaptation” is associated with a loss of cellular K+ probably due to a transient increase in K+ permeability and to loss of associated anions and osmotically obliged water. Partial dissipation of the large gradient of K+ between cells and medium by pre-exposure to ouabain or to K+-free medium results in a diminished capacity to adapt. After the shrinking phase is completed, a new steady state is established with a reduced cellular K+ content, normal Na+, normal K+-permeability, and a reduced activity of the Na+ − K+ transport system. When adapted cells are returned to normal medium, an initial shrinking is followed by a re-swelling to normal size, associated with a gain in K+ content, presumably due to the return to normal activity of the Na+ − K+ transport system.  相似文献

Isolated rat hepatocytes posses a saturable glucocorticoid uptake system with high affinity (Kd value = 2.8 ± 0.7 × 10−8 M; 318,000 ± 80,000 binding sites per cell; 317 fmol/mg protein). The initial rates of uptake decrease by about 30–40% if the cells are incubated simultaneously with [3H]corticosterone and either SH-reagents (N-ethylmaleimide and p-chloromercuriphenylsulphonate, 1 mM), metabolic inhibitors (2,4-dinitrophenol, 1 mM; and antimycin, 0.1 mM) or the Na+/K+-ATPase-inhibitors, ouabain and quercetine. These Na+/K+-ATPase-blockers exert half-maximal inhibition at 3 × 10−7 and 3 × 10−6 M, respectively. A slight increase in K+ concentration and a corresponding decrease in Na+ in the medium leads to a significant reduction in the initial uptake rate. The uptake system from the rat hepatocytes shows a clear steroid specificity, being different from the intracellular receptor. Corticosterone and progesterone are the strongest competitors, cortisol, 5- and 5β-dihydrocorticosterone, 11-deoxycorticosterone, cortisone and testosterone have an intermediate effect and only weak competition is exerted by dexamethasone and by the mineralocorticoid, aldosterone. Estradiol and estrone sulphate as well as the synthetic glucocorticoid triamcinolone acetonide are unable to inhibit initial corticosterone uptake.  相似文献

We have examined the chromosomal radiosensitivities of an ionizing-radiation- and MMS-sensitive mutant (M10), and a UV- and 4NQO-sensitive mutant (Q31), isolated from mouse lymphoma L5178Y cells, with regard to killing effects. In the first mitoses after 100 R γ-irradiations, it was found that M10 cells were highly radiosensitive in terms of chromosomal aberrations accompanying longer mitotic delay (3 h); the frequencies of both chromatid-type and chromosome-type aberrations were, respectively, about 7 and 4 times higher than that of wild-type L5178Y cells. Furthermore, chromatid exchanges, particularly triradials, isochromatid breaks with sister union, and chromatid gaps and breaks were markedly enhanced at G1 phase of M10 cells. In contrast, the chromosomal radiosensitivity of Q31 cells after 100 R irradiation was similar to that of L5178Y cells. On the other hand, spontaneous aberration frequencies (overall breaks per cell) of M10 and Q31 cells were, respectively, 5.1 and 2.2 times higher than that of wild-type L5178Y cells. The chromosomal hypersensitivity to γ-rays in M10 cells is discussed in the light of knowledge obtained from ataxia telangiectasia cells.  相似文献

Bropirimine (U-54,461) is a novel compound which is being developed as a biological response modifier for use in treatment of neoplastic and viral disease. Compounds of this type exert their therapeutic effects by immuno-stimulation or other non-cytotoxic mechanisms. The purpose of the experiments described in this paper was to evaluate the hazard potential of this drug. Bropirimine was previously reported to be negative in the Ames Salmonella assay (Aaron et al., 1989a) and the in vitro UDS assay (Aaron et al., 1989b). In experiments reported here positive response was observed in a test for clastogenicity in vitro in CHO cells, but bropirimine was negative in the L5178Y mouse lymphoma TK+/− assay. A subsequent experiment demonstrated the ability of bropirimine to induce HPRT mutations in CHO cells. Interestingly, evidence for induction of chromosome aberrations in the L5178Y cells by bropirimine was also obtained. While the reason for the apparent insensitivity of the L5178Y TK+/− assay to bropirimine is unexplained by the experiments, it is clear that at high dose bropirimine is capable of clastogenesis in both CHO and L5178Y cells and can give rise to gene mutations in CHO cells but apparently not in L5178Y cells.  相似文献

The effects of prednisolone and other glucocorticoids were studied in a cell culture system of L5178Y mouse lymphoma. A relatively high level of 8 × 10−6 M prednisolone was required to reduce growth by 50%. The response was due to an increase in generation time and was not a result of the cytolytic effects of the steroid. A synchronous cell population was obtained by successive treatment with excess thymidine and Colcemid. Using this technic it was determined that an exposure to prednisolone for a time longer than a complete cell cycle was required in order to bring about inhibition of cell multiplication. In the course of experiments involving brief exposure to prednisolone, it was determined that prednisolone when added to synchronized cells inhibited incorporation of 14C-choline into the lipid cell fraction. The inhibition of choline incorporation in cells selected for increased resistance to prednisolone was about half that observed in steroid-sensitive cells.  相似文献

Sodium metaperiodate is mitogenic for human peripheral lymphocytes. Evidence of stimulation can be detected as increased thymidine incorporation at 72 h after only 10 sec of exposure to the IO4. The degree of response varies with lymphocytes from different donors, but maximum stimulation for the healthy donors studied was obtained at concentrations of IO4 between 10−3 M and 4 × 10−3 M. Concentrations of 8 × 10−3 M and above are non-stimulatory and toxic. Exposures to optimum concentrations for 1 h or longer result in essentially no stimulation and inincreased cell death. However, a significant response to phytohemagglutinin (PHA) and pokeweed mitogen (PWM) remains. The kinetics of response over a 4 day culture period are similar for IO4, PHA and PWM. The morphology of the blast cells and the degree of response suggest that the IO4 responsive lymphocyte population may be more closely related to the PWM stimulated cells than the PHA responsive lymphocytes.  相似文献

• Beta-adrenergic receptor identification and properties are probed in rat parotid membranes utilizing the high affinity β-adrenergic antagonist(−)-[3H]dihydroalprenolol.
• The binding of (−)-[3H]dihydroalprenolol to membrane preparations of parotid is rapid, equilibrium being reached in 5 min. Strict stereospecificity is observed, (−)-propanolol being 100 times more potent than (+)-propranolol in competing with (−)-[3H]dihydroalprenolol for binding sites. Beta-adrenergic agonists compete for the binding sites with (−)-[3H]dihydroalprenolol with the same characteristics, i.e., much higher concentrations of the (+)-stereoisomers than the (−)-stereoisomers are required to produce 50% inhibition, the range varies from 14-fold for epinephrine to 300-fold for isoproterenol. Among the (−)-stereoisomers, the relative potency of inhibitory action is (−)-propranolol > (−)-isoproterenol > (−)-epinephrine ≡ (−)-norepinephrine. (−)-Isoproterenol is about 20 times as potent as norepinephrine, the least potent agonist among all the catecholamine (−)-stereoisomers.
• The binding of (−)-[3H]dihydroalprenolol is saturable, with a maximum number of binding sites equalling 450 fmol/mg protein and a dissociation constant of 7.9 nM. The Scatchard plots show no significant curvilinear character. Hill plots consistently give a Hill coefficient close to unity (0.92–1.05). Both pieces of evidence suggest a single-component system with no significant cooperativity.
• Dissociation kinetics study after the method of De Metys et al. (1973) Biochem. Biophys. Res. Commun. 155, 154, indicates a lack of site-to-site interactions among the binding sites. The rate of dissociation of bound (−)-[3H]dihydroalprenolol is the same in the presence and absence of 1 · 10−5 M (±)-alprenolol.
• Based on the binding of (−)-[3H]dihydroalprenolol, it is concluded that the beta-adrenergic receptors can be identified in rat parotid and that these binding sites display β1 character. Results of the study indicate a one-component system with no observable site-to-site interactions.

We have developed two improved methods: (1) a procedure for coupling 125Iodine to ConcanavalinA (ConA) that yields intensely labeled and fully active lectin; (2) a procedure that allows studies of lectin binding to be carried out with a minimum of non-specific binding to reaction vessels. We found that BALB/c 3T3 cells, SV3T3 cells, and human red blood cells have 1.3 × 107, 1.5 × 107, and 2.2 × 106 ConA binding sites/cell. More than 99.5% of the radioactivity in the samples counted was associated with the cells; background radioactivity, in the absence of cells, was negligible. We also found that although α-methylmannopyranoside (α-MM) prevented almost all of the ConA from binding to cells, when ConA had first been allowed to bind, α-MM removed only 60 to 80% of the bound ConA. In addition, even after the removal of a portion of bound lectin by α-MM, most, if not all, of the remaining cell-associated ConA was coupled to the plasma membrane.  相似文献

[3H]norepinephrine binding to isolated rat fat cells was studied as a function of adipose cell age and size. Rats aged from 4 to 78 weeks were used.Scatchard analysis of norepinephrine binding revealed in old fat cells like in young ones the existence of two orders of binding sites with respectively high and low affinity for norepinephrine. The apparent association constants Ka1 and Ka2 associated with these binding sites did not differ consistently in the different groups of fat cells studied (Ka1 = 1.7 to 2.2 × 106 × M−1; Ka2 = 1.9 to 2.5 × 104 × M−1), suggesting that age and cell size do not modify the apparent affinity of norepinephrine-binding sites in rat fat cells.On the contrary, the total amount of norepinephrine bound to each of these sites was dependent upon cell age and size. In fact, maximum binding of norepinephrine to the high affinity sites was 0.9 and 9 pmol/105 cells in small (diameter: 35 μ) and large (diameter: 105 μ) adipocytes, respectively, the values found for the low affinity sites being 13 and 135 pmol/105 cells. When expressed per unit of fat cell area, however, the total binding capacity for these sites appeared practically constant (2.4 — 2.8 pmol × 10−3/mm2 and 34.2 — 38.2 pmol × 10−3/mm2 for the high and low affinity sites respectively). These data suggest that the total norepinephrine binding capacity of the fat cell is directly proportional to its surface.  相似文献

Nitrobenzylthioniosine (NBMPR), a potent and specific inhibitor of nucleoside transport, is bound reversibly by high affinity sites on nucleoside transporter proteins of erythrocyte membranes and, upon photoactivation, NBMPR molecules become covalently bonded to the sites. This study showed that [3H]NBMPR molecules reversibly bound to intact S49 and L5178Y mouse lymphoma cells became covalently bound upon exposure to UV light. Electrophoretic analysis of plasma membrane fractions from the labelled cells showed that 3H was present in polypeptides which migrated as a major band with an apparent Mr of 45000–65000.  相似文献

We have examined the possibility that gangliosides act as the cell surface receptor for fibronectin, as previously suggested by the data of Kleinman et al. (Proc natl acad sci US 76 (1979) 3367), using three different approaches.
1. 1. Gangliosides inhibited the spreading of both CHO and BHK cells on fibronectin-coated substrates. 50% inhibition of cell spreading was produced by 1.0 and 3.5 × 10−5 M di- and trisialogangliosides respectively, although monosialogangliosides were less effective inhibitors. The inhibition was apparently due to an interaction of gangliosides with fibronectin and not due to a direct effect of gangliosides on the cells.
2. 2. Using anti-fibronectin antibodies, 125I-labelled protein A, and gangliosides adsorbed to polystyrene tubes, we have provided direct evidence that fibronectin will bind to gangliosides. However, the interaction is apparently of low affinity compared with binding of cholera toxin to gangliosides.
3. 3. We have compared the ability of BALB/c 3T3 cells to spread on fibronectin with that on substrates coated with a ganglioside-specific ligand, cholera toxin B-subunit. Cells plated onto fibronectin-coated substrates rapidly (within 60 min) adopted a well spread morphology, whereas spreading on substrates coated with the toxin B-subunit was less extensive even after 2 h. In addition, the organization of F-actin within cells spread on the two types of substrate was also quite different.
We conclude that the interaction of cells with fibronectin may well be influenced by membrane-bound gangliosides. It is unlikely, however, that binding of fibronectin to such gangliosides can lead to the cytoskeletal reorganization which is characteristic of cells spread on fibronectin.  相似文献

Both prostaglandins (PGs) and nitric oxide (NO) have cytoprotective and hyperemic effects in the stomach. However, the effect of NO on PG synthesis in gastric mucosal cells is unclear. We examined whether sodium nitroprusside (SNP), a releaser of NO, stimulates PG synthesis in cultured rabbit gastric mucus-producing cells. These cells did not release NO themselves. Co-incubation with SNP (2 × 10−4, 5 × 10−4, 10−3 M) increased PGE2 synthesis, and SNP (10−3 M) increased PGI2 synthesis in these cells. Hemoglobin, a scavenger of NO, (10−5 M) eliminated the increase in PGE2 synthesis by SNP, but methylene blue, an inhibitor of soluble guanylate cyclase, (5 × 10−5 M) did not affect the increase in PGE2 synthesis by SNP. 8-bromo guanosine 3′ : 5′-cyclic monophosphate (8-bromo cGMP), a cGMP analogue, (10−6, 10−5, 10−4, 10−3 M) did not affect PGE2 synthesis. These findings suggest that NO increased PGE2 and PGI2 synthesis via a cGMP-independent pathway in cultured rabbit gastric cells.  相似文献

The protective effects of dimethyl sulfoxide (DMSO) against cell killing by 137Cs γ-rays were investigated in XRCC4-deficient cell line M10, XRCC4-complemented M10 and the parental mouse leukemia cell line L5178Y. Cell survival was determined by the colony-forming ability. M10 cells were more sensitive to γ-ray-induced cell death than L5178Y and complemented M10 cells. Cell survival was increased in both M10 and L5178Y in the presence of DMSO. However, estimation of the DMSO-protectable fraction revealed a smaller protectable fraction for M10 cells than for L5178Y cells, indicating that indirect effects contributed in a smaller extent to the cytotoxicity in M10 than that in L5178Y. This effect is due to XRCC4 deficiency, since transfection of XRCC4 cDNA into M10 cells restored the radioprotective effects of DMSO to the level seen in L5178Y. In M10 cells, the killing effects of high LET radiation (Auger electrons from 125I-antipyrine, carbon ions with an LET of 166 keV μm−1) were similar to those of low LET radiation (137Cs γ-rays, characteristic X-rays from 125I-bovine serum albumin). We discuss that lethal lesions produced by indirect actions in L5178Y and XRCC4-complemented M10 cells may differ, at least in part, from DNA double-strand breaks repairable by non-homologous end joining.  相似文献

We have previously demonstrated that cystic fibrosis (CF) cells show increased survival compared to normal cells when exposed to ouabain in medium lacking potassium. In this report, we show that the CF cells bind significantly less ouabain than the normal cells in potassium-deficient medium. Using age-matched normal and CF skin fibroblast strains, we show that (1) at ouabain concentrations <20 × 10−9 M binding for both normal and CF cells is linear with time for 1 h and reaches equilibrium after 4 h; (2) at ouabain concentrations between 2 and 20 × 10-9 M, the initial rate of binding for CF cells is approx. 70% of the normal cells; (3) under equilibrium-binding conditions, Scatchard analysis reveals that three different CF strains have 12, 16, and 44% fewer ouabain-binding sites than their matched normal controls. In addition, studies in potassium-free medium of the inhibition of 86Rb flux (a K+ analogue) into the cell by ouabain show no differences between CF and normal cells. We have also previously shown that in a low glucose, potassium-deficient medium the CF cells survived ouabain exposure no better than normal cells. In this report, equilibrium-binding studies with [3H]ouabain clearly show that CF cells bind less ouabain under these conditions than normal cells. These results indicate that ouabain resistance in CF cells is not solely a function of differences in ouabain binding. Furthermore, the differential ouabain killing may not be due to ion transport differences, but rather to as yet unknown mechanisms. CF cells thus appear to be unlike previously characterized ouabain-resistant mutants.  相似文献

Bisulfite had a severe cytotoxic effect on HeLa S-3 cells at a concentration of 10−4 M, causing complete inhibition of colony formation. A concentration of 10M bisulfite had a moderate cytotoxic effect on the cells, reducing their colony-forming activity to one-third of that of untreated control cultures. Addition of vitamin E at a concentration of 10−7 M restored the colony-forming activity of 10−5 M bisulfite-treated cells to 71% of that of control cultures. Addition of 10−6 M vitamin E resulted in almost complete recovery of the colony-forming activity of bisulfite-treated cells to that of the control cultures. The counteraction of vitamin E to the cytotoxic effect of bisulfite on human cells was discussed.  相似文献
