Various nitrate concentrations (0, 1, 2, 4, 8, 20, 50 mol m–3)were applied at weekly intervals for 10 weeks to the caesalpinioidlegume Chamaecrista fasciculata. Microscopic techniques andgeneral growth studies showed that nitrate affected both theplant and its rhizobial symbiosis. As the nitrate concentrationwas increased, nodule structure became increasingly disruptedeven though nitrate remained limiting to plant growth until8 mol m–3. Hydrogenase activity (Tritium uptake activity) in cell-free extracts of bacteroids from soybean and lupin nodules was separated into two parts, whose activities were different from each other in responsiveness to ATP, by differential centrifugation, Sephadex gel filtration or DEAE-cellulose column chromatography. In a study of the inhibitory effect of sulphanilamide on thegrowth of flax seedlings and its reversal byp-aminobenzoic acid(PABA) and related compounds, two distinct types of reversalmechanism have been observed. The results, derived from growth-ratemeasurements, are interpreted as indicating that, in the flaxseedling, sulphanilamide interferes with the conversion of PABAto pteroyl-glutamic acid. The influence of protein synthesis and nitrate reductase activity on nitrate absorption by barley (Hordeum vulgare L.) was investigated. Cycloheximide decreased nitrate absorption. Influence of various saline media on Chlorella fusca growth, contents of photosynthetic pigments, and activity of the nitrate reductase (NRA) was determined. KCl, CaCl2, and MgCl2 in concentrations relative to NaCl as 1:1, 2:1, and 3:1 promote cell number, dry mass, and contents of photosynthetic pigments. The activity of NR was enhanced by Mg2+ and K+ and in some cases by Ca2+ at all ratios applied. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The regulation and nitrate inhibition of nodule formation insoybean, Glycine max (L.) Merr., was further examined usingthe nodulation mutants of cv. Enrei. The non-nodulating mutantsEn115, Enl282, and En1314 produced extremely few markedly-curledroot hairs which were all devoid of infection threads, and invariablyfailed to initiate sub-epidermal cell divisions (SCDs) in theroot cortex. A considerable number of arrested SCDs was foundbefore nodule emergence in Enrei, but not in En6500 which hadsignificantly more SCDs that progressively increased at moreadvanced stages of nodule ontogeny. These observations indicatethat autoregulation acts by blocking the developmental stagebefore nodule emergence. In both Enrei and En65OO, the maturationof emerged nodules was restricted by a late-acting nodulationcontrol mechanism that is apparently unrelated to autoregulation.Reciprocal wedge-grafts of plants inoculated at sowing showedthat the control of the supernodulating phenotype resides inthe shoot, while the non-nodulating phenotype is strictly root-controlled.The nodulation phenotype of the current non-nodulating mutantsresults not from an alteration of the autoregulatory mechanism,but from mutation that exerts a root-localized effect that blocksSCDs which trigger the autoregulatory mechanism. Reciprocalgrafting experiments on Enrei and En6500 seedlings grown undervarious nitrate levels suggest that nitrate inhibition of nodulation,like autoregulation, is shoot-controlled. Since these two processesare invariably expressed together, they are probably causallyrelated, acting synergistically to regulate nodule formationin soybean. These results indicate that the regulation and nitrateinhibition of nodulation in the nodulation mutants of cv. Enreiare similar to those of cv. Bragg nodulation mutants. A procedure is described for obtaining a large number of nodules(approximately 1000) on a hydroponically grown soybean plantafter a brief exposure to Rhizobium japonicum. This procedurewas used to study the effect of medium nitrate upon nodulationof the soybean plant. It was found that nitrate, at concentrationsequal to or greater than 2.0 mM, restricted nodule developmentbut did not inhibit appreciably the activity or synthesis ofnitrogenase in those few nodules that were formed. The influence of NH4+, in the external medium, on fluxes of NO3 and K+ were investigated using barley (Hordeum vulgare cv Betzes) plants. NH4+ was without effect on NO3 (36ClO3) influx whereas inhibition of net uptake appeared to be a function of previous NO3 provision. Plants grown at 10 micromolar NO3 were sensitive to external NH4+ when uptake was measured in 100 micromolar NO3. By contrast, NO3 uptake (from 100 micromolar NO3) by plants previously grown at this concentration was not reduced by NH4+ treatment. Plants pretreated for 2 days with 5 millimolar NO3 showed net efflux of NO3 when roots were transferred to 100 micromolar NO3. This efflux was stimulated in the presence of NH4+. NH4+ also stimulated NO3 efflux from plants pretreated with relatively low nitrate concentrations. It is proposed that short term effects on net uptake of NO3 occur via effects upon efflux. By contrast to the situation for NO3, net K+ uptake and influx of 36Rb+-labeled K+ was inhibited by NH4+ regardless of the nutrient history of the plants. Inhibition of net K+ uptake reached its maximum value within 2 minutes of NH4+ addition. It is concluded that the latter ion exerts a direct effect upon K+ influx.  相似文献

Q10 values for 13NO3 influx were determined in `uninduced' (NO3-starved) and `induced' (NO3-pretreated) roots of barley (Hordeum vulgare L.) plants at various concentrations of external NO3 ([NO3]0). At 0.02 mole per cubic meter [NO3]0, Q10 values for influx were from 3 to 4 between 5 and 10°C. As [NO3]0 increased Q10 values decreased, reaching values of 1.2 and 2.0, respectively, at 20 moles per cubic meter in uninduced and induced plants. The metabolic dependence of 13NO3 influx at low and high [NO3]0 (0.1 and 20.0 moles per cubic meter, respectively) in uninduced and induced plants was probed by the use of various inhibitors. These experiments confirmed the findings of the Q10 studies, demonstrating that at low [NO3]013NO3 influx was extremely sensitive to metabolic inhibition. By contrast, at high [NO3]0, influx was relatively insensitive to the presence of inhibitors.  相似文献

以我国南海海域分离的赤潮原因种——球形棕囊藻(Phaeocystis globosa)为材料, 研究了不同硝酸盐浓度下藻细胞生长及硝酸还原酶活性的变化。当培养基中不含硝酸盐时, 藻细胞内硝酸还原酶的活性保持在非常低的水平, 藻细胞的生长受到限制, 不能形成正常的生长曲线: 当培养基中硝酸盐浓度为3.62 mmol.L-1时, 藻细胞的硝酸还原酶活性和比生长速率达到最大。在含有硝酸盐的培养基中, 接种培养后第9天藻细胞硝酸还原酶活性达到最大值, 并且在4种不同硝酸盐浓度下, 藻细胞硝酸还原酶活性的差异性达到极显著水平(P<0.01)。在接种培养第16天藻细胞密度达到最大值, 并且4种不同硝酸盐浓度培养的藻细胞密度之间的差异性也达到极显著水平(P<0.01)。实验结果表明, 在培养基中添加不同浓度的硝酸盐, 对球形棕囊藻细胞硝酸还原酶的活性和藻细胞的生长有极显著的影响, 含有较高硝酸盐的富营养化海域有利于球形棕囊藻细胞的持续生长。  相似文献

Nitrate Supply and the Biophysics of Leaf Growth in Salix viminalis   总被引:2,自引:0,他引:2  
The influence of nitrogen on leaf area development and the biophysicsof leaf growth was studied using clonal plants of the shrubwillow, Salix viminalis grown with either optimal (High N) orsub-optimal (Low N) supplies of nitrate. Leaf growth rate andfinal leaf size were reduced in the sub-optimal treatment andthe data suggest that in young rapidly growing leaves, thiswas primarily due to changes in cell wall properties, sincecell wall extensibility (% plasticity) was reduced in the LowN plants. The biophysical regulation of leaf cell expansion also differedwith nitrogen treatment as leaves aged. In the High N leaves,leaf cell turgor pressure (P) increased with age whilst in theLow N leaves P declined with age, again suggesting that foryoung leaves, cell wall plasticity limited expansion in theLow N plants. Measurements of cell wall properties showed thatcell wall elasticity (%E) was not influenced by nitrogen treatmentand remained constant regardless of leaf age. Seed germination and barley seedling growth in various D2O concentrations have been studied. It was observed that the emergence of root and shoot was delayed, there being greater delay in shoot than in root emergence. A complete block was observed in germination in pure D2O and the germination rate was slowed down significantly in lower concentrations. An initial germination delay by different D2O concentrations seemed to cause a subsequent retardation in the growth measured as shoot and root length. A comparison of root and shoot length with their respective dry weights suggested that the growth by cell division/elongation might have been affected more than the transport of food materials from the endosperm to the embryo. Analysis of the total sugars of the endosperm and the embryo at 8 hour intervals showed that while the hydrolysis of starch to sugars was progressively decreased by increasing D2O concentrations, the transport rate from endosperm to embryo showed a sharp inhibition in 50% D2O and above. This indicated that the inhibition in the transport of materials, besides less hydrolysis of reserve food materials, may also be a causal factor of germination and growth inhibition in D2O.  相似文献

Antiserum was prepared in rabbits against purified alfalfa (Medicago sativa L.) nodule phosphoenolpyruvate carboxylase (PEPC). Immunotitration assays revealed that the antiserum recognized the enzyme from alfalfa nodules, uninoculated alfalfa roots, and from soybean nodules. Tandem-crossed immunoelectrophoresis showed that the PEPC protein from alfalfa roots and nodules was immunologically indistinguishable. The 101 kilodalton polypeptide subunit of alfalfa nodule PEPC was identified on Western blots. The PEPC polypeptide was detected in low quantities in young alfalfa roots and nodules but was present at increased levels in mature nodules. Senescent nodules appeared to contain a reduced amount of the PEPC polypeptide. PEPC was also detected by western blot in some plant- and bacterially-conditioned ineffective alfalfa nodules but was not detected in bacteroids isolated from effective nodules. Alfalfa nodule PEPC is constitutively expressed in low levels in roots. In nodules, expression of PEPC polypeptide increases several-fold, resulting in increased PEPC activity. Antiserum prepared against the C4 PEPC from maize leaves recognized the PEPC enzyme in all legume nodules and roots tested, while the antiserum prepared against alfalfa nodule PEPC also recognized the leaf PEPC of several C4 plant species. Neither antiserum reacted strongly with any C3 leaf proteins. The molecular weight of the PEPC polypeptide from C4 leaves and legume nodules appears to be similar.  相似文献

A computer-controlled multichannel data acquisition system was employed to obtain continuous measurements of net nitrate or chlorate uptake by roots of intact barley plants (Hordeum vulgare cv Betzes) using nitrate-specific electrodes. Plants, previously grown in solutions maintained at 10 or 200 micromolar NO3 (low N or high N conditions, respectively), were provided with 200 micromolar NO3 or ClO3 during the uptake period. Initial rates of NO3 uptake were several times higher in low N plants than in high N plants. Within 10 min, uptake in the former plants declined to a new steady rate which was sustained for the remainder of the experiment. No such time-dependent changes were evident in the high N plants. Rates and patterns of net chlorate uptake exhibited almost identical dependence upon previous nitrate provision. NO3 (36ClO3) influx, by contrast, appeared to be independent of NO3 pretreatment prior to influx determination. Nitrate efflux, estimated by several different methods, was strongly correlated with internal nitrate concentration of the roots.  相似文献
