In this study crude laccases from the white‐rot fungi Cerrena unicolor and Trametes hirsuta were tested for their ability to decolorize simulated textile dye baths. The dyes used were Remazol Brilliant Blue R (RBBR) (100 mg/L), Congo Red (12.5 mg/L), Lanaset Grey (75 mg/L) and Poly R‐478 (50 mg/L). The effect of redox mediators on dye decolorization by laccases was also assessed. C. unicolor laccase was able to decolorize all the dyes tested. It was especially effective towards Congo Red and RBBR with 91 and 80% of color removal in 19.5 h despite the fact that simulated textile dye baths were used. Also Poly R‐478 and Lanaset Grey were partially decolorized (69 and 48%, respectively). C. unicolor laccase did not need any mediators for removing the dyes. However, T. hirsuta laccase was only able to decolorize simulated Congo Red and RBBR dye baths (91 and 45%, respectively) in 19.5 h without mediators. When using mediators the decolorization capability was enhanced substantially, e.g. Poly R‐478 was decolorized by 78% in 25.5 h. The effect of copper (II) ions on the growth of three brown-rot fungi, six white-rot fungi and one blue-stain fungus in solid
medium was evaluated. The fungi were grown in malt extract agar with different concentrations of copper added, and the radial
growth rate was determined. At the end of the incubation period, the mycelial biomass and the media pH were determined. The
white-rot and blue-stain fungus grew up to 3 mM and 6 mM copper, respectively and the brown-rot fungi were the only ones that
grew up to 10 mM, with higher growth rates than those shown by the other fungi. In general, the brown-rot fungi produced greater
acidification in the culture media than the white-rot fungi and blue-stain fungus, and the acidification increased when the
amount of copper was increased. The biomass production for the different species, in the absence or presence of copper, was
not related to the radial growth rate, and the fungal species that produced the greatest biomass amounts did not correspond
to those that presented the highest growth rates. The brown-rot fungi Wolfiporia cocos and Laetiporus sulfureus and blue-stain fungus Ophiostoma sp. demonstrated greater tolerance to high copper concentrations in solid medium than the white-rot fungi, determined as
radial growth rate. On the other hand, the highest biomass producers in solid medium with copper added were the white-rot
fungi Ganoderma australe and Trametes versicolor and the brown-rot fungus Gloeophyllum trabeum. 相似文献

以平皿培养方式对采集自中国和芬兰的白腐真菌菌株降解6种不同结构的人工染料的能力进行了筛选研究。在40株菌株中,黑管孔菌Bjerkandera adusta Y5012,一色齿毛菌Cerrena unicolor Y5002,硬毛粗盖孔菌Funalia trogii Y4997,香栓孔菌Trametes suaveolens D8325和云芝栓孔菌Trametes versicolor Y4946对刚果红、橙黄G、茜素红、结晶紫、中性红和亚甲基蓝均显示出较强的脱色能力。对一色齿毛菌Cerrena unicolor Y5002的液体培养脱色条件进行了研究,其最适碳源和氮源分别为蔗糖和麦芽浸粉;在不同橙黄G浓度下均获得较高的脱色率,因此浓度为500mg/L的橙黄G未对该菌的脱色能力产生抑制作用,而浓度为400mg/L茜素红则对其脱色作用产生明显抑制。对菌丝生物量和染料脱色率的研究表明,在不同碳源和氮源条件下,两者之间具有明显的正相关性。 相似文献

Selective purification still poses a challenge in the downstream processing of biomolecules such as proteins and especially enzymes. In this study a polyethylene glycol 3000 (PEG 3000)–phosphate aqueous two-phase system at 25 °C and pH 7 was successfully used for laccase purification and separation. Initially, the effect of phase forming components on enzyme activities in homogenous systems was studied. In the course of the extraction experiments tie lines, enzyme source, initial enzyme activities, phase ratio and sodium chloride concentrations were varied and their influence on the activity partitioning was determined. Partitioning results were validated using clear-native-PAGE and isoelectric focusing. Based on these results, the separation of laccases from Trametes versicolor and Pleurotus sapidus was investigated using the principle of superposition. Sodium chloride was used to adjust laccase partitioning in the applied aqueous two-phase system (ATPS). Finally, two modes of operation are proposed depending on the aim of the purification task. One mode with 0.133 g g−1 of PEG3000, 0.063 g g−1 of phosphate and without sodium chloride separates P. sapidus laccases from T. versicolor laccases with clearance factors of 5.23 and 6.45, respectively. The other mode of operation with 0.124 g g−1 of PEG3000, 0.063 g g−1 of phosphate and 0.013 g g−1 of sodium chloride enables a partitioning of both laccases into the bottom phase of the ATPS resulting in a purification factor of 2.74 and 96% activity recovery. 相似文献

【目的】白腐菌Cerrena unicolor Y-G07是一种不产孢子的丝状真菌,其漆酶合成与生长相偶联。应用于单细胞微生物发酵的搅拌器并不适用于白腐真菌。在此,综合考虑溶氧效果和剪切力作用,研究不同搅拌器类型对白腐菌Cerrena unicolor Y-G07发酵生产漆酶产量的影响。【方法】针对Y-G07菌株生长过程需氧量大,且对剪切力敏感的特性,订制5种不同类型的搅拌器(径向流、轴向流等),在通气已控制在设备最大量程及转速优化的前提下,研究Y-G07在发酵生产过程中使用不同类型搅拌器对菌丝生长形态、生长速度、溶氧情况、糖代谢和漆酶合成的影响。【结果】Cerrena unicolor Y-G07菌株对不同类型搅拌器产生的发酵液流态性质和剪切力敏感,表现在菌丝体的生长形态、细胞浓度差异较大,且生长周期改变,从而影响漆酶的合成。采用六折叶DT602搅拌器最有利于该菌株形成致密度合适的网状菌丝体,菌丝体细胞浓度高,断裂少、生长状况好,漆酶的单位产量可达690 U/m L,相对于普通使用的六叶平直叶搅拌器(448 U/m L)提高了54%。【结论】选择合适的搅拌器类型有利于好氧但对剪切力敏感的微生物发酵。 相似文献

This work presents a preliminary report of a series of studies on the ability of several indigenous wood-rotting fungi from
Chile to produce hydrolytic and ligninolytic enzymes and the evaluation of these native microorganism to future research on
potential applications in bioremediation programs. Wood-rotting Basidiomycete fungi were collected from indigenous hardwood
forest in the South of Chile. Twenty-eight strains were identified and qualitative enzymatic tests for peroxidases, laccase,
tyrosinase, xylanase and cellulase production were performed in solid medium. Eleven selected strains were evaluated in liquid
medium to quantify their ligninolytic enzyme production and their capacity to grow in solid medium supplemented with 2,4-dichlorophenol
(2,4-DCF), 2,4,6-trichlorophenol (2,4,6-TCF) and pentachlorophenol (PCP). PCP degradation and ligninolytic enzymes production
were also evaluated in liquid medium. Results showed that laccase was present in 28 of the selected strains (≈73%). Peroxidase
was present in 40% and cellulase in 37% of the strains. Xilanase and tyrosinase were obtained in a smaller percentage in the
strains (28% and 7%, respectively). The 11 selected strains showed high concentrations of lignin peroxidase (Lip) and manganese
peroxidase (MnP). Anthracophyllum discolor (Sp4), produced LiP and MnP at 90.3 and MnP 125.5 U L−1 respectively, compared to the control fungus Phanerochaete chrysosporium CECT-2798 that produced 58.1 and 118.4 U L−1 of LiP and MnP. Tolerance test showed that native Chilean fungi did not present high tolerance to 2,4,6-TCF and PCP but were
quite tolerant to 25 and 50 mg L−1 of 2,4-DCF. However, pre-acclimatization in 2,4-DCP notably improved the growth in medium with 2,4,6-TCP and PCP. PCP in
liquid medium was efficiently degraded by the fungi Anthracophyllum discolor, Lenzites betulina (Ru-30) and Galerina patagónica (Sp3), and the major MnP activity was produced by A. discolor (Sp4) (67 U L−1). 相似文献

The wood-degrading basidiomycete Cerrena unicolor C-139 has been suggested as a potential producer of the industrially important enzyme laccase. Basic culture parameters influencing
the enzyme synthesis in shaken-flask and aerated bioreactor cultures were evaluated to improve the yields of the process.
Production of extracellular laccase was considerably enhanced by the addition of Cu2+ in the micromolar range to a carbon-sufficient and nitrogen-sufficient culture medium (C/N = 16.69). When an optimised medium
containing glucose (10 g/L) and l-asparagine (1.5 g/L) was used, and enzyme synthesis was stimulated by addition of 10 μM Cu2+ to the culture medium on days 3, 6 and 9, maximal laccase productivity obtained after 17 days’ cultivation in shaken flask
cultures was above 100,000 nkat/L. In fermenter fungal cultures, the influence of stabilisation of medium pH on laccase activity
was additionally studied. The use of a bioreactor with an automatic pH control set at pH 6.5 after 48-h incubation resulted
in the enzyme activity of 65,000 nkat/L after 8 days’ cultivation. 相似文献

The distribution of acetyl esterase was studied in 30 strains of wood-rotting fungi. A screening test on agar plates using
glucose β-d-pentaacetate as a substrate indicated that all tested fungi produced acetyl esterase to form a clear zone on the culture.
All fungi also showed positive responses in an agar test using carboxymethyl cellulose acetate. Enzyme assay showed that extracellular
acetylxylan esterase activity was present in the filtrates of wood-meal culture of all these fungi. The ratio of fungal acetylxylan
esterase activity to 4-nitrophenyl acetyl esterase activity were higher than that of porcine liver esterase, indicating that
fungal esterases have high affinity for acetylated carbohydrates. Acetyl esterase is suggested to be distributed widely in
wood-rotting fungi for degradation of native acetylated hemicelluloses. 相似文献

Abstract The mycorrhizal fungi Amanita muscaria, Paxillus involutus, Hymenoscyphus ericae, Pisolithus tinctorius, Rhizopogon roseolus , and Suillus bovinus oxidized elemental sulphur to thiosulphate and sulphate in vitro. In some, but not all cases, tetrathionate was also formed. Limited oxidation of elemental sulphur by R. roseolus also occurred when growing in association with Pinus contorta in unsterilized peat. Although yeasts capable of oxidizing sulphur could not be isolated from a wide range of soils, a yeast-like fungus ( Monilia sp.) isolated from deciduous woodland soil oxidized elemental sulphur to sulphate, forming thiosulphate, but not tetrathionate. This fungus also oxidized tetrathionate to sulphate but showed only limited ability to oxidize thiosulphate to tetrathionate. Both Aspergillus niger and Trichoderma harzianum oxidized elemental sulphur in mixed culture with Mucor flavus . Larger amounts of sulphate were initially formed in mixed, compared to single culture; but by week 5 of the incubation period sulphate formation was greatest in single culture. The wood-rotting fungi, Hypholoma fasciculare and Phanerochaete velutina showed a limited ability to oxidize elemental sulphur in vitro but were incapable of oxidizing the element when growing as mycelial cords in non-sterilized soils. The relevance of these results to the possibility that fungi play a role in sulphur oxidation in soils is commented upon. 相似文献

Penicillin acylase obtained from E. Coli (E. C. 3.5.1.11) was covalently bound via glutaric aldehyde to acrylic carriers crosslinked with divinylbenzene or ethylene glycol dimethacrylate. The best enzymatic preparation was obtained by using ethyl acrylate/ ethylene glycol dimethacrylate copolymer. 1 cm3 of the carrier bound 6.4 mg of protein, having 72% activity in relation to the native enzyme. The preparation lost only 10% of its initial activity after 100 d of storage at 4°C. A negligible effect of immobilization on the enzyme activity at different temperatures or pH as well as significant increase of the stability of the immobilized enzyme at elevated temperatures were observed.Abbreviations BA

Dextranase (1,6-α-d-glucan 6-glucanohydrolase, EC 3.2.1.11) from Penicillium aculeatum culture has been immobilized on a bentonite support. The matrix-bound enzyme could be stored as acetone-dried powder or as a suspension in acetate buffer, pH 5.6, for about three weeks at 4°C without any loss of activity. There was no change in the specific activity of the enzyme on immobilization and the enzyme yield was 0.1–0.6 mg/g bentonite matrix. In the presence of sucrose, thermal stability of the immobilized enzyme was high and the bound enzyme could be used for about six cycles. 相似文献

In this study, 2‐hydroxyethyl methacrylate (HEMA) was used as the monomers for surface grafting on electrospun PU/RC nanofiber membrane via atom transfer radical polymerization (ATRP) method, and the PU/RC‐poly(HEMA) nanofiber membrane was investigated as a carrier for LAC. Free and immobilized LAC was characterized, and efficiency of bisphenol A (BPA) removal was determined. The results indicated that the PU/RC‐poly(HEMA)‐LAC showed relatively higher pH stability, temperature stability, and storage stability than free and PU/RC‐LAC; moreover, more than 60% of the PU/RC‐poly(HEMA)‐LAC activity was retained after 10 cycles of ABTS treatment. Notably, the BPA removal efficiency of PU/RC‐poly(HEMA)‐LAC membrane generally ranged from 87.3 to 75.4% for the five cycles. Therefore, the PU/RC‐poly(HEMA) nanofiber membrane has great potential as a carrier for the LAC immobilization for various industrial applications and bioremediation. 相似文献

For many years, the wood decay process by fungi was associated almost exclusively with production of lignocellulolytic enzymes. However, recent studies by electron microscopy have shown that fungal enzymes are too large to penetrate into the cell wall at an early stage of decay. Thus, the hypothesis that low molecular mass agents may initiate the breakdown of both cellulose and lignin was proposed. The purpose of this work was to detect low molecular mass compounds, with metal-chelating capability, from liquid cultures of two wood-rot fungi. The brown-rot fungus Wolfiporia cocos produced the highest chrome azurol S (CAS) reaction, simultaneously reducing the pH of the malt extract medium. In contrast, the white-rot fungus Trametes versicolor did not react with CAS and the pH remained approximately constant during the culture period. The presence of hydroxamate derivatives and oxalic acid was detected in extracts of low molecular mass of both fungi. Moreover, in W. cocos extracts, catecholate derivatives were also detected. Accumulation of oxalic acid was greater in W. cocos than in T. versicolor at the end of the culture period, and this might be responsible for the strong response from W. cocos in the CAS reaction. 相似文献

Abstract: White-rot fungi produce extracellular lignin-modifying enzymes, the best characterized of which are laccase (EC 1.10.3.2), lignin peroxidases (EC 1.11.1.7) and manganese peroxidases (EC 1.11.1.7). Lignin biodegradation studies have been carried out mostly using the white-rot fungus Phanerochaete chrysosporium which produces multiple isoenzymes of lignin peroxidase and manganese peroxidase but does not produce laccase. Many other white-rot fungi produce laccase in addition to lignin and manganese peroxidases and in varying combinations. Based on the enzyme production patterns of an array of white-rot fungi, three categories of fungi are suggested: (i) lignin-manganese peroxidase group (e.g. P. chrysosporium and Phlebia radiata ), (ii) manganese peroxidase-laccase group (e.g. Dichomitus squalens and Rigidoporus lignosus ), and (iii) lignin peroxidase-laccase group (e.g. Phlebia ochraceofulva and Junghuhnia separabilima ). The most efficient lignin degraders, estimated by 14 CO2 evolution from 14 C-[Ring]-labelled synthetic lignin (DHP), belong to the first group, whereas many of the most selective lignin-degrading fungi belong to the second, although only moderate to good [14 C]DHP mineralization is obtained using fungi from this group. 以琼脂粉为基质制备金属螯合载体,并用于固定重组腈水解酶。研究发现:制备金属螯合载体最合适的金属离子为Zn2+。当Zn2+离子浓度0.3 mol/L、给酶量15.6 mg/g、固定化pH 8.0、固定化温度40℃时,制得的固定化酶活性最高。固定化酶最适反应温度为50℃、最适反应pH为7.0。当扁桃腈浓度为10 mmol/L、反应1 h时,固定化酶最大产率为0.041 mmol/(g·h);在反应12 h时,产物e.e.值可达到99%以上。固定化酶重复使用8次以后,酶活力仍保持在45%。 相似文献

Ergosterol contents of six wood-rotting basidiomycetes were analyzed under different cultivation conditions. Four white-rot and two brown-rot fungi were cultivated in liquid synthetic medium with low nutrient nitrogen (2 mM) and 0.1% glucose, and ergosterol in mycelial biomasses were measured weekly for 35 days. The highest ergosterol content per fungal dry mass in the white-rot fungi was found in Phanerochaete chrysosporium being 2100 μg g−1, while in Ceriporiopsis subvermispora it was 1700 μg g−1, Phlebia radiata 700 μg g−1, and Physisporinus rivulosus 560 μg g−1. In brown-rot fungi the ergosterol content was in Poria placenta 2868 μg g−1 and in Gloeophyllum trabeum 3915 μg g−1. On agar media, P. chrysosporium and P. radiata reached the highest ergosterol value in 7 days, while in wood block cultures the ergosterol contents were quite stable. The conversion factors for ergosterol-to-fungal biomass varied from 48 and 243, which were lower than values for ascomycetous soil fungi reported in the literature. 相似文献

Enzyme immobilization is a routine biotechnology of many industries such as pharmaceutical, chemical and food. Among the different techniques of enzyme immobilization, cross-linking methods are often used. Geniposide is a natural product extracted from gardenia and its hydrolysate genipin is one of green cross-linking agent for enzyme immobilization, but the environmental pollution and cost of the genipin extraction process have become the main obstacle to its wide application. Enzyme β-glucosidase was immobilized on chitosan by self-catalysis and further used to hydrolyze geniposide. The laccase was immobilized on Nano-SiO2 through the hydrolysate of geniposide directly acts as cross-linking agent. The simplification of the extraction steps overcomes the obstacles to the widespread use of genipin. Compared with the free laccase, the Nano-SiO2@laccase exhibited better pH stability and thermal stability. The Nano-SiO2@laccase was used to degrade Bisphenol A (BPA) and the biodegradation efficiency of the Nano-SiO2@laccase was 84.3 % after 10 cycles of reusing. 相似文献
