Immunoreactive beta-endorphin (IR-beta-ENDO) was compared in the plasma, pituitary and hypothalamus of young female rats on the day of estrus and old constant estrous (CE) female rats, and in intact and chronically castrated old CE female rats. The concentration of IR-beta-ENDO in the plasma and the content and concentration of IR-beta-ENDO in the neurointermediate lobe of the pituitary were significantly greater in the old CE female rats than in the young female rats on the day of estrus. The content and concentration of IR-beta-ENDO in the anterior pituitary and hypothalamus were similar in the two age groups. To determine if estrogen contributed to the increase in plasma and pituitary levels of IR-beta-ENDO observed in the old animals, a group of old CE female rats were castrated and compared to sham operated control CE rats. Thirty days after castration, levels of plasma, pituitary and hypothalamic IR-beta-ENDO were comparable in the intact and the chronically castrated old female rats. These data indicate that in old CE female rats, plasma and pituitary IR-beta-ENDO are significantly increased in comparison to young female rats on the day of estrus, and that these increased levels of IR-beta-ENDO observed in old female rats do not appear to be influenced by gonadal estrogen.  相似文献

Administration of pertussis vaccine to hamsters markedly affected their lipid metabolism. Four days after the administration of the vaccine a severe fatty liver was observed. Concomitantly, a rise in the serum levels of free fatty acids, triacylglycerols and ketone bodies was detected. It is suggested that an altered regulation of adipose tissue lipolysis might be at least partially responsible for the observed effects.  相似文献

The Bio 14.6 hamster has a well-documented cardiomyopathy which leads to congestive heart failure. Previous work demonstrated that hearts from these hamsters have depressed fatty acid oxidation and depressed carnitine concentrations compared to those of normal hamsters. Analyses of tissue carnitine concentrations from 40 to 464 days of age demonstrate that the cardiomyopathic hamsters have a cardiac carnitine deficiency throughout life. Therefore, the carnitine deficiency is not a secondary effect of an advanced stage of the cardiomyopathy. Both the observation that other tissues of the cardiomyopathic hamster have normal or markedly elevated carnitine concentrations and the observation that oral carnitine treatment could not increase the cardiac carnitine concentrations to those of normal hamsters are consistent with the hypothesis that the cardiac carnitine deficiency is the result of a defective cardiac transport mechanism. Cardiac carnitine-binding protein (which may function in the cardiac carnitine transport mechanism) prepared from hearts of cardiomyopathic hamsters had a lower maximal carnitine binding and an increased dissociation constant for carnitine compared to the cardiac carnitine-binding protein prepared from normal hamsters. Thus, several types of data indicate that the cardiomyopathic hamster has an altered cardiac carnitine transport mechanism.  相似文献

Two strains of rats (S3 and WEzob), which show different levels of aggression in the laboratory, were tested in repeated heterosexual confrontations. Daily 15-min observations were made of the interactions between a female throughout a complete estrous cycle and the same male partner. In both strains the topography of aggression was similar in males and females, but the frequency of specific parameters varied. Males showed more offensive and females more defensive patterns. The overall level of aggression was very low on the day of estrous, when the female was sexually receptive. There were no differences in any elements of female or male behavior between the other 3 days of the cycle. The results support previous conclusions from single-sex encounters that in rats there is no sexual dimorphism in the ability to show aggression.  相似文献

The urine of intact, adult male mice elicits more investigatory sniffing from female mice than does the urine of castrated males. When either of two androgen-dependent urinary compounds, 2-sec-butyl dihydrothiazole or dehydro-exo-brevicomin are added to castrate urine, its relative attractiveness remains the same. When both compounds are added to castrate urine, however, its activity is enhanced and the castrate urine becomes as attractive to females as whole, intact male urine. Females exposed to the reconstituted ‘normal’ urine for 3 min per day, displayed more frequent oestrus cycles. The two synthetic compounds are synergistic in the context of castrate urine, producing an olfactory message that behaviourally and physiologically mimics the activity of the normal biological signal.  相似文献

Two brain areas behaviorally responsive to progesterone (P) were examined to determine their possible involvement in the control of rat preceptive behavior, i.e., solicitation behavior directed at the male. Progesterone implants were placed in the habenular nuclei and the interpeduncular nucleus-ventral tegmental area of the midbrain reticular formation (MRF). Different testing procedures and levels of priming with estradiol benzoate (EB) were used in order to distinguish the effects of P in either region on proceptive and receptive behavior during exposure to 10 mounts by stimulus males. To test for receptivity, sexually experienced 60-day-old ovariectomized (ovx) rats bearing stereotaxically placed guide cannulas extending to the habenula or MRF were given 10 μg EB subcutaneously. Forty-eight hours later, lordosis quotient (LQ) was determined. Immediately following this test, each animal was implanted with cholesterol (C) or P and was retested 2 hr later. Treatments for the proceptivity test were similar except that the animals received 2.5 μg EB/100 g body wt sc for 7 days before testing on the eighth day; LQ as well as hopping, darting, and ear wiggling were scored. In the receptivity test, P implantation in both the medial portions of the habenula and the MRF significantly increased lordosis above the levels found both in their preimplantation tests and following control implantation of C. Little proceptivity was observed. In the proceptivity test, P implants in both regions also significantly increased proceptive behavior above both types of control tests. All animals were highly receptive, and there was no difference in LQ among the groups. There was no increase of plasma P levels in similarly implanted animals during a 24-hr monitoring period, indicating that systemic leakage of the hormone was not responsible for the observed behavior. The data indicate that both the habenula and MRF are P-sensitive regions. Progesterone's action on the two areas facilitates expression of both proceptive and receptive components of female sexual behavior, indicating that the neural regulation of the two kinds of behavior is integrated at these levels.  相似文献

The purpose of the present study was to determine whether nonsteroidal antiestrogens could act as estrogens by inducing or facilitating estrogen-induced sexual receptivity in a female lizard (Anolis carolinensis). Experiments were conducted to (1) test the ability of enclomiphene (ENC) and zuclomiphene (ZUC) to induce sexual receptivity in estrogen-untreated ovariectomized females; (2) to determine the effect of ENC and ZUC pretreatment on E2B induction of sexual receptivity; and (3) to examine the ability of a variety of antiestrogens to act as estrogens by inducing sexual receptivity in females pretreated with a behaviorally ineffective estrogen treatment regimen.  相似文献

Two experiments were performed to characterize the process of postnatal demasculinization in Japanese quail. In the first experiment, it was shown that estradiol (E2) can complete female demasculinization during the first 4 weeks of life. By contrast, E2 did not demasculinize sexual behavior and cloacal gland in neonatally castrated males. Neonatally gonadectomized females preferentially performed mount attempts when tested in their home cage by comparison to a test arena. In Experiment 2, E2 Silastic implants (40-mm) maintained full copulatory behavior in castrated males but not in females. This large dose of E2 did not demasculinize adult sexually active birds (males or females) even if treatment lasted for 1 month. It is concluded that E2 can demasculinize sexual behavior only in females and only if treatment is performed in very young birds.  相似文献

Lipase has been isolated from sheep pancreas. The lipoprotein complex formed in pancreas homogenates by the enzyme and endogenous lipids is split by treatment with acetone. Lipase is further purified by ion-exchange chromatography and gel filtration. The molecular weight and the amino-acid composition of ovine lipase are very similar to that of the porcine and bovine enzymes. As previously found in bovine lipase, no carbohydrate is covalently bound to the polypeptide chain which has a N-terminal residue of lysine. The study of the catalytic properties of ovine pancreatic lipase indicates that the enzyme is fully activated by colipase from various species in the presence of conjugated bile salt micellar solutions.  相似文献

Rats maintained on tritiated 1,25-dihydroxyvitamin D3 as their sole source of vitamin D and placed on diets differing in calcium content had similar intestinal levels of tritiated 1,25-dihydroxyvitamin D3. Since 1,25-dihydroxyvitamin D3 administration eliminated adaptation of intestinal calcium transport, it appears that increased production of 1,25-dihydroxyritamin D3 is responsible for the stimulation of calcium transport by low dietary calcium. When maintained on tritiated 1,25-dihydroxyvitamin D3, rats fed a low-phosphorus diet had somewhat higher levels of tritiated 1,25-dihydroxyvitamin D3 in the duodenum and plasma than rats on a normal-phosphorus diet. In addition to stimulating 1,25-dihydroxyvitamin D3 synthesis, low dietary phosphorus may increase the accumulation of 1,25-dihydroxyvitamin D3 in both intestine and plasma.  相似文献

Previously it was observed that mixture cultures of “nullipotent” and pluripotent embryonal carcinoma (EC) cells failed to differentiate, suggesting that nullipotent cells might restrict the differentiation of pluripotent cells (M. J. Rosenstraus and A. J. Levine, 1979, Cell17, 337–346). This report shows, however, that the differentiation of mixed cultures is dependent on the relative growth rates of the two cell types and that nullipotent cells do not directly affect pluripotent cell development. When the growth rate of nullipotent cells was modulated by taking advantage of drug-resistance markers, mixed cultures, containing pluripotent PSA-1 cells and a genetically marked subclone of nullipotent F9 cells, exhibited extensive differentiation. The differentiated cells were PSA-1 derivatives, whereas, the F9 cells remained as undifferentiated EC cells. Similar results were obtained when a genetically marked PSA-1 subclone was cocultured with a second nullipotent cell line, Nulli SSC1. Thus nullipotent and pluripotent EC cells appear to express their developmental potential autonomously in mixed cultures. This implies that the nullipotent cell lines studied are intrinsically incapable of responding to the conditions that trigger in vitro differentiation of EC cells upon aggregation.  相似文献

Murine bone marrow cells (strain A) have been allowed to differentiate in vivo in syngeneic (A) or semiallogeneic hosts (A × B) to produce mature splenic T lymphocytes. After stimulation of these cells with irradiated allogeneic (C) spleen cells in tissue cultures, the cytotoxic T-cell blasts (CTL) were purified by velocity sedimentation and used to immunize (A × C) F1 hybrid mice, to produce antisera recognizing the receptor structure (for C) on the relevant A cytotoxic cells (and their precursors). Using these sera we have been able to show that the T-cell receptor for alloantigen C on strain A cytotoxic precursor lymphocytes (CTLp) seems to differ according to the host environment in which those T cells differentiate from immature bone marrow precursors.  相似文献

A procedure for the automated assay of purified amylase inhibitors was developed. Samples were analyzed at a rate of 60/h using ferricyanide reagent to monitor the suppression of the release of reducing groups from a solution of starch by a calibrated alpha-amylase reagent. In addition to the sequential analysis of individual samples, the use of gradients permitted the continuous analysis of the effect of substrate and of inhibitor concentration. Also described are some of the effects of starch and inhibitor concentration and time of preincubation on the amylase-inhibitor reaction. The procedure was also suitable for the assay of samples of amylase and should be applicable to the determination of the effects of inhibitors on other enzymes which release reducing sugars.  相似文献

Rat-brain poly(A)-containing RNAs were prepared by affinity chromatography on Poly(U)-Sepharose and translated in the rabbit reticulocyte lysate system in the presence of [35S]-methionine. By using antimyelin proteolipids antibodies, immunoreactive polypeptides of 23.5 K and 19 K were isolated from the translation products : they had electrophoretic mobilities similar to those of the rat brain myelin proteolipids apoproteins.  相似文献

The sexual behavior of 53 beef heifers and 66 beef steers was observed and recorded for 7 days following insertion of subcutaneous ear implants at 2 months of age. Thirty-five of the heifers were implanted with Synovex S (200 mg of progesterone and 20 mg of estradiol benzoate). Forty-four of the steers were implanted with Synovex H (200 mg of testosterone and 20 mg of estradiol benzoate). The remaining 18 heifers and 22 steers served as nonimplanted controls. Synovex S treatment increased the incidence of mounting (P < 0.005) and mounting receptivity (P < 0.005) of heifers, while Synovex H treatment had only slight effect, if any, on the mounting behavior (P < 0.25) and mounting receptivity (P < 0.25) of steers. Both steers and heifers mounted heifers more frequently than steers (P < 0.005). Heifers were mounted with similar frequencies by both heifers and steers (P < 0.90), while steers were mounted only by heifers (P < 0.15).  相似文献

The pH-dependent interaction between the cyclic guanidino analog of biotin, 2-iminobiotin, and avidin has been used in the design of an efficient affinity isolation system for avidin and its fluorescent and iodinated derivatives. Avidin and its derivatives are retained by a column of 2-iminobiotin-6-aminohexyl-Sepharose 4B at pH values between 9 and 11 and are specifically eluted from the column at pH 4. This affinity isolation procedure overcomes the harsh conditions, i.e., 6 m guanidine-HCl, pH 1.5, required to dissociate avidin from an immobilized biotin column.  相似文献

Several chalcone derivatives; e.g. Ro 09-0204, Ro 09-0323 and Ro 09-0501 were found to reduce markedly the revertant increase of Salmonella typhimurium TA100 by benzo(a)pyrene during the incubation with S-9 Mix. The antimutagenic activity was 100 - 700 times stronger than that of L-ascorbic acid. Effect on other mutagens, the structure activity relationship and the possible mechanism of action are briefly discussed.  相似文献
