A total of 60 maize genotype samples from different agroclimatic regions of India were collected. Fresh harvest of these maize samples comprising some commercial maize genotypes and some land races were kept under ambient storage conditions for 9 months duration at grain moisture ranges from 14% to 10.5% with a view to identifying the least contaminated maize genotype with aflatoxin. The purpose of this study was to identify the maize genotypes which can survive in ambient storage conditions with minimum spoilage. The response of various maize genotypes for AFB1 accumulation was variable in similar storage conditions. Promising genotypes which showed lower accumulation of AFB1 were identified: Shaktiman-1 (A QPM variety) by showing the lowest concentration of AFB1 (0.30 ppb) followed by KMH-1701 (0.40 ppb); HQPM-1 (0.50 ppb); and QPM-2-136 (0.60 ppb), whereas the most highly toxic sample was Mon - 4 (62.42 ppb) at grain moisture ranges from 12.6 to 11.1%. During the study it has been observed that Shaktiman-1 and other QPM genotypes showed minimum levels of AFB1. Further, it was confirmed by western blot analysis by comparing the resistant and susceptible genotype under artificially inoculated grains with Aspergillus flavus and uninoculated maize grains. It was observed that more chitinase activity was found in shaktiman-1, when the grains were artificially inoculated with A. flavus. The thickness of the seed coat and Aleuron layer was maximum in (90–100 μm) in Shaktiman compared to that of Pro-311 (80–85 μm). It was observed that the thickness of seed coat may act as a barrier for mold contamination and as a result the storage spoilage is minimised.  相似文献

These approaches have proved their merits and have been successfully applied to a wide range of fungal species belonging in the Ascomycetes and Basidiomycetes. Species in the class Zygomycetes have been rather neglected from this aspect. One of the methods for measuring the efficiency of N utilization by plants is the index of physiological efficiency of absorbed N (PEN) which for cereals is defined as the ratio of grain produced to the total N absorbed by the above-ground plant parts (grain and straw) at maturity. This index indicates how this absorbed N is used by the plant to produce grain. The objective of this work was to study the genotypic variability of PEN in oats (Avena sativa L.) and to what extent grain yield is related to PEN. Seven selected oat genotypes were studied under greenhouse conditions with 5 levels of added N including control (no additional N). At maturity the grain and straw were harvested separately and analyzed for total N. The results show that there was a highly significant variation among genotypes in both yield per pot and PEN. Grain yield was positively and significantly related to PEN (r=0.95xx). The total N absorbed by plants was much less correlated with grain yield than PEN. The results suggest that PEN may be used in a plant breeding program to detect the potentially high yielding oat genotypes and to evaluate those capable of exploiting N input most efficiently.  相似文献

Fusion to form a chimera has been documented in many marine invertebrate taxa, including poriferans, cnidarians, bryozoans, and colonial ascidians. Allogenic interactions in chimeric ascidian colonies vary widely across taxonomic groups but are poorly characterized in the invasive colonial ascidian Didemnum vexillum. The moderate level of discrimination expressed in the fusion–rejection response of D. vexillum suggests that there is some integration of cells beyond the fusion line in a chimeric colony. We tracked the shifts in representation of microsatellite alleles between fused colonies of D. vexillum to elucidate the extent of genotypic integration in fused colonies and the patterns of changes to the genotypic composition of colonies immediately following chimera formation. By genotyping colonies before and after fusion, we found that allogeneic fusion in D. vexillum may lead to genotypic changes beyond the visible fusion line. Alleles from one colony were found in multiple tissue samples in the chimera 7–10 days after fusion had occurred. In some instances, alleles that were in a single colony prior to fusion were lost following fusion. We observed multiple patterns of allelic change, including both the unidirectional transfer and reciprocal exchange of alleles between fused colonies. Our findings suggest that tissue or cells are exchanged following allogeneic fusion between colonies of D. vexillum and that the genotypic composition of chimeric colonies may be fluid.  相似文献

Several species of marine polychaetes reside in individual protective tubes. These tubes may be agglomerated in patches of varied sizes with very high densities, or form massive reef-like mounds which can be stretch over several kilometers. These polychaetes can thus be considered as the most important building organisms after corals in coastal environments. We especially focused on several species belonging to the families of Sabellariidae, Terrebeliidae and Pectinariidae. Tubes grains are glued together with biomineralized cement secreted from a building organ connected to specialized glands. Different methods of micro-analysis were used to analyze the biomineral components of these cements for four varyingly gregarious tube-building polychaetes: Lanice conchilega, Pectinaria koreni, Sabellaria alveolata and Phragmatopoma caudata. The aim of this study is to describe and compare the inter- and the intraspecific variability in the main biomineral components of cement at local, regional and continental scales. Scanning Electron Microscopy confirms an identical structure of the cement within species of Sabellariidae in Europe and America. Electron Probe Micro-Analysis (EPMA) confirms the presence of calcium, phosphorus, and magnesium plus traces of manganese in all cements, with varying concentrations of these elements at different locations for the same species or for different species at the same location. Information on heritability and predicted gains from selection for increased biomass yield for ethanol production in switchgrass is limited and may vary among breeding populations. The purpose of this study was to estimate heritability and predicted gains from selection for higher biomass yield within a lowland ecotype switchgrass population, Southern Lowland 93 (SL‐93), and two upland ecotype switchgrass populations, Southern Upland Northern Upland Early Maturing (SNU‐EM) and Southern Upland Northern Upland Late Maturing (SNU‐LM). Narrow‐sense heritabilities (hn2) for biomass yield in each of the three populations were estimated via progeny–parent regression analysis. Half‐sib (HS) progeny families from 130 randomly selected plants from the SL‐93 population were evaluated for biomass yield in replicated trials in 2002 and 2003. Clonal parent plants were evaluated for biomass yield in separate environments to provide unbiased hn2 estimates from progeny–parent regression. Yield differences were highly significant among SL‐93 HS progenies within and over years. For the SL‐93 population, hn2 estimates were 0.13 and 0.12 based on individual plant and phenotypic family mean (PFM) selection, respectively. Predicted genetic gains (ΔG) per selection cycle were 0.15 kg dry matter (dm) plant?1 and 0.10 kg dm plant?1 for PFM and individual plant selection methods, respectively. For the SNU‐EM and SNU‐LM populations, year and year × HS family effects were highly significant (P < 0.01) and the HS family effect over years was nonsignificant (P < 0.05). However, HS family effects were highly significant within respective years (P < 0.01). Estimates of hn2 for the SNU‐EM and SNU‐LM populations based on PFM and individual plant selection were similar, ranging from 0.44 to 0.47; ΔG per selection cycle ranged from 0.22 to 0.33 kg dm plant?1. The magnitudes of the estimates of additive genetic variation suggest that selection for higher biomass yield should be possible. The substantial effect of environment on biomass yields in the upland populations and the failure of families to respond similarly over years stress the importance of adequately testing biomass yield over years to assess yield.  相似文献

Abstract

Phyto-recurrent selection is an established method for selecting tree genotypes for phytoremediation. To identify promising Populus (poplar) and Salix (willow) genotypes for phytotechnologies, our objectives were to (1) evaluate the genotypic variability in survival, height, and diameter of poplar and willow clones established on soils heavily contaminated with nitrates; and (2) assess the genotypic stability in survival and diameter of selected poplar clones after one and eleven growing seasons. We tested 27 poplar and 10 willow clones planted as unrooted cuttings, along with 15 poplar genotypes planted as rooted cuttings. The trees were tested at an agricultural production facility in the Midwestern, United States. After 11 growing seasons, using phyto-recurrent selection, we surveyed survival and measured the diameter of 27 poplar clones (14 unrooted, 13 rooted) that were selected based on superior survival and growth throughout plantation development. Overall, willow exhibited the greatest survival, while poplar had the greatest height and diameter. At 11 years after planting, superior clones were identified that exhibited above-average diameter growth at the establishment- and rotation-age, most of which had stable genotypic performance over time. Selection of specific clones was favorable to genomic groups, based on the geographic location and soil conditions of the site.  相似文献

This study used two different approaches to model changes in biomass composition during microwave‐based pretreatment of switchgrass: kinetic modeling using a time‐dependent rate coefficient, and a Mamdani‐type fuzzy inference system. In both modeling approaches, the dielectric loss tangent of the alkali reagent and pretreatment time were used as predictors for changes in amounts of lignin, cellulose, and xylan during the pretreatment. Training and testing data sets for development and validation of the models were obtained from pretreatment experiments conducted using 1–3% w/v NaOH (sodium hydroxide) and pretreatment times ranging from 5 to 20 min. The kinetic modeling approach for lignin and xylan gave comparable results for training and testing data sets, and the differences between the predictions and experimental values were within 2%. The kinetic modeling approach for cellulose was not as effective, and the differences were within 5–7%. The time‐dependent rate coefficients of the kinetic models estimated from experimental data were consistent with the heterogeneity of individual biomass components. The Mamdani‐type fuzzy inference was shown to be an effective approach to model the pretreatment process and yielded predictions with less than 2% deviation from the experimental values for lignin and with less than 3% deviation from the experimental values for cellulose and xylan. The entropies of the fuzzy outputs from the Mamdani‐type fuzzy inference system were calculated to quantify the uncertainty associated with the predictions. Results indicate that there is no significant difference between the entropies associated with the predictions for lignin, cellulose, and xylan. Genotypic variability in vulnerability of leaf xylem to water-stress-induced cavitation was determined in four sugarcane (Saccharum sp.) clones using detached leaf segments in a hydraulic conductivity apparatus. Vulnerability curves were constructed by plotting the percentage of maximum conductivity versus leaf water potential (ψI) and fitting curves using a Weibull function. The ψI at which each clone lost 10, 50, and 80% of maximum conductivity was determined. Maximum conductivity per unit of leaf width was positively associated with metaxylem vessel diameter. The commercial clone H65-7052 exhibited the highest and the nondomesticated S. spontaneum exhibited the lowest conductivity. All four clones lost substantial conductivity at values of ψI less negative than −1.4 MPa, but H65-7052 was able to maintain 50% conductivity to lower ψI than the other clones. S. spontaneum sustained the most negative ψI (−1.99 MPa) before reaching the 80% conductivity loss point. Clone H69-8235 was consistently the most vulnerable to initial loss of conductivity. These vulnerability functions were used in conjunction with field measurements of ψI to estimate diurnal losses in leaf hydraulic conductivity under irrigated and droughted conditions. H69-8235 lost up to 50% of its conductivity during the day, even when well irrigated, and more than 80% when subjected to drought. The other clones exhibited lower conductivity losses. These losses are apparently reversed overnight by root pressure. Despite their close genetic relationships, these clones exhibited large differences in conductivity, in the vulnerability of their xylem to cavitation, and in gas exchange behavior. The potential for altering water relations by selecting for particular hydraulic characteristics is discussed.  相似文献

Switchgrass (Panicum virgatum L.) is a C4 grass with high biomass yield potential and is now a model species for the Bioenergy Feedstock Development Program. Two distinct ecotypes (e.g., upland and lowland) and a range of plant morphotypes (e.g., leafy and stemmy) have been observed in switchgrass. The objective of this study was to determine the influence of ecotype and morphotype on biomass feedstock quality. Leaf and stem tissues of leafy and stemmy morphotypes from both lowland and upland ecotypes were analyzed for key feedstock traits. The leaf : stem ratio of leafy morphotype was more than 40% higher than the stemmy morphotype in both upland and lowland ecotypes. Therefore, the stemmy morphotype has significant advantages over leafy morphotype during harvesting, storage, transportation and finally the feedstock quality. Remarkable differences in feedstock quality and mineral composition were observed in switchgrass genotypes with distinct ecotypic origins and variable plant morphotypes. Lignin, hemicelluloses and cellulose concentrations were higher in stems than in the leaves, while ash content was notably high in leaves. A higher concentration of potassium was found in the stems compared to the leaves. In contrast, calcium was higher and magnesium was generally higher in the leaves compared to stems. The upland genotypes demonstrated considerably higher lignin (14.4%) compared with lowland genotypes (12.4%), while hemicellulose was higher in lowland compared with upland. The stemmy type demonstrated slightly higher lignin compared with leafy types (P < 0.1). Differences between the ecotypes and morphotypes for key quality traits demonstrated the potential for improving feedstock composition of switchgrass through selection in breeding programs.  相似文献

The world is experiencing increasing climatic variability, an ongoing loss of biodiversity and a growing spread of invasive species. Previous experimental studies demonstrated that the invasibility of plant populations is reduced with increasing resident genetic diversity and is promoted by environmental fluctuations, but their combined effect has so far not been considered. In a growth chamber experiment, we tested whether the genotypic diversity of experimental populations of Arabidopsis thaliana (1, 3 or 6 genotypes) and temperature fluctuations affect population invasion by Senecio vulgaris, and how these factors interact. We found that genotypic diversity tended to increase the invasion resistance of experimental plant populations in terms of the biomass ratio between the species, and that temperature fluctuations strongly favoured Arabidopsis (biomass: +49%) over Senecio (–28%). However, there were no interactions between environmental fluctuations and genotypic diversity. Nevertheless, the magnitude of net diversity effects and transgressive overyielding depended on temperature conditions, indicating that increased environmental variability can influence diversity mechanisms. Our study shows that, although genotypic diversity and environmental variability did not interact, these two factors independently affected the invasibility of plant populations.  相似文献

Summary The behaviour of eleven Saintpaulia ionantha (H. Wendl.) genotypes in protoplast culture was compared. Isolation of protoplasts from young shootlets regenerated in vitro on leaf explants, yielded 0.7 to 1.8 × 106 protoplasts per gram fresh weight. In all cultivars and breeding lines tested, cell divisions were observed. The mean division frequencies varied between 1.0 and 5.0% after 14 days, and between 6.4 and 13.8% after 24 days of culture. In ten genotypes callussing and shoot regeneration were achieved. The difference between the genotypes in shoot regeneration rate, between 2 and 68%, was more pronounced. A pot experiment confirmed that pigeonpea could efficiently utilize various sources of phosphorus (P) (aluminium phosphate, iron phosphate and apatite), irrespective of genotype. A qualitative assay method for iron (Fe)-P solubilizing activity showed that root exudates collected from P-deficient pigeonpea contained Fe-P solubilizing substances and that they were released mainly from root tips. Citric, malic, malonic, succinic and piscidic acids were identified in root exudates. Citric and piscidic acids release from roots was increased by low-P treatment in all the genotypes tested. The release rates of citric and piscidic acids were affected by the P concentration of shoots rather than that of roots. The pigeonpea roots released approximately 5–100 times more piscidic acid than citric acid depending on P stress status, plant age and genotype. When organic acids were added to Alfisols, citric acid was most capable of mobilizing P from the soil, followed by piscidic acid and malic acid. No correlation was found between genotypic variability in the release rates of citric and piscidic acids from the roots under low-P treatment at hydroponic culture and in the growth and P uptake of plants on Alfisols. Although citric and piscidic acids released from pigeonpea roots may play a partial role in solubilizing unavailable insoluble P in soils, the releases were thought to be an unsatisfactory strategy for explaining genotypic variation in low P availability of pigeonpea.  相似文献

物种丰富的植物群落更能抵抗入侵。近十年来的研究表明,遗传变异具有很多生态效应。本研究旨在检验本地物种遗传多样性的响应模式在不同生长形式的定殖物种之间是否存在差异,以及这种响应是否受到土壤养分的影响。我们设立常见草种紫羊茅(Festuca rubra) 实验样地,在两种土壤养分水平下,设置3个遗传多样性水平(1、6或18个克隆基因型,表征基因型多样性)。在样地设立后第4年,将4种定殖者混合播种到样地中:匍匐性豆科植物白车轴草(Trifolium repens)、阔叶黄花茅(Anthoxanthum odoratum)、大莲座丛杂草长叶车前(Plantago lanceolata)和小莲座丛杂草兴安风铃草(Campanula rotundifolia)。我们观测三年来物种的建立和生长情况,检验定殖成功是否取决于基因型多样性、特定的羊茅属基因型、土壤养分或定殖者的生长形式。本地无性系牧草的基因型多样性和基因型同一性对其定殖成功和生物量有显著影响。然而,定殖物种的反应各异。本地物种对基因型多样性最大的响应是生长形式和结构与本地物种相似的草丛型禾草。大莲座丛植物在生长初期有响应,而匍匐性豆科植物则完全没有响应。种内遗传型多样性和本地物种的基因型同一性在植物群落聚合中起着重要作用。  相似文献

Summary Nine randomly chosen clones of tall fescue (Festuca arundinacea Schreb.) were mated in all possible combinations to determine the nature of genetic variation for Mg, Ca, K, and P concentrations in a broad genetic base population. General combining ability mean squares were significant for most variables, whereas specific combining ability mean squares were not significant in most instances indicating that additive genetic variance was more important. Genotype x year interactions were significant for most variables, suggesting that selection should be evaluated over many environments. Broad-sense heritability estimates based on parental and progeny variance components were generally high for P, K, Ca, and Mg but low for the ratio K/(Ca + Mg). Narrow-sense heritabilities for these minerals were close to the broad sense values since the additive genetic variance was the largest component of the total genetic variation. Correlations between mineral concentrations and herbage dry matter yield were low. It was concluded that adequate genetic variation exists to improve mineral concentration without altering herbage dry matter yields.Journal Series no. 5886  相似文献

We demonstrate the effects of age and genotype on the likelihoodthat an individual worker honeybee (Apis mellifera L.) willbecome a forager. We established experimental colonies thatwere each initially composed of identifiable, nonforaging workersof similar ages (1–5 days old). Workers in each test colonywere the progeny of two queens, providing age and genotypicgroups of workers for comparisons. We then recorded the daythat each worker was first observed foraging. Older workerswere more likely to become foragers under our experimental conditions,even when age differences were just 1 day. At a given age, workersfrom different queens varied in their likelihood of becomingforagers. However, we found that neither age nor genotype (queensource) directly affected the likelihood that a forager wouldrevert to within-nest, larval care activities after the removalof nonforaging bees from colonies. The likelihood of revertingwas only dependent on how long a worker was a forager beforeremoving the nonforagers of the colony.  相似文献

Background

The present study analyses changes in body composition over the course of a working week. The purpose of the study is to identify the size of the changes in the observed parameters by means of typical error of measurement (TE) as the initial value for the interpretation of the detected changes in the repeated measurement in diagnostic practice.

Methods

The researched group consisted of 86 males, aged 21.4 ± 1.0 years. All the participants were free of any medical conditions. The measurement of each participant took place over 1 week from Monday till Friday, in the morning hours. Parameters measured: body mass (BM), total body water (TBW), and body fat (BF). The measurement employed two devices using the bioelectric impedance analysis. These devices were the analyzers Tanita BC 418 MA and Nutriguard MS. In order to assess the differences between the average values, the analysis of repeated measurements was used. To assess the material significance, eta squared was used. TE was used to express the size of the changes in the observed parameters.

Results

A statistically significant difference between the average values of the observed parameters was only detected when using the Tanita BC 418 MA analyzer. Based on the post-hoc tests, these differences in the average values were always detected on Monday and Friday. No material significance was proved, however. The highest TE values were also detected in measurements carried out on Monday and Friday. For BM, the value of TE was 0.6 kg, for TBW 1.0–1.1 %, 0.8–0.9 kg, and for BF 1.2–1.6 %, 1.1–1.3 kg depending on the analyzer used.

Conclusions

The results of the present study demonstrate the stability of parameters of body composition throughout a working week, with the provision that standard measuring conditions are fulfilled. For the purpose of diagnostic practice, when interpreting the results of the repeated measurements, it is advisable to take as provable change caused by the observed factors only the ones whose values exceed the value of a weekly TE or the upper limit of the interval of the measurement reliability.  相似文献

Switchgrass (Panicum virgatum L.) and coastal panic grass (Panicum amarum A.S. Hitchc. & Chase) are perennial grasses indigenous to North America. Switchgrass has been shown to have good potential as a biofuel crop in both the US and Canada. In the study reported here, seven varieties of switchgrass and one panic grass were evaluated for 5 years under the temperate maritime conditions in Southern England. Both species had 0 or 60 kg N ha(-1) applied annually in spring as treatment. Yield was measured after flowering and when stems were dead in the winter. Yield increased annually for 4-5 years except for the variety Dacotah, and in the fifth year dead stem yields ranged from 8.82 to 13.97 t dm ha(-1). There was no response to N except for one variety in one year. Mineral concentration in biomass was higher at flowering than at dead stem harvest and delaying harvesting further provided more time for P, K and Cl to be leached but yield also declined.  相似文献

This work examined cell composition gradients in maturing tillers of switchgrass (Panicum virgatum L.) with the aim of developing baseline information on this important forage and biomass crop. Flowering tillers were collected from plants raised from seeds in a greenhouse and field, harvested at soil level and separated into internodes beginning with the node subtending the peduncle. Internodes were analyzed using microscopy, by fiber digestion, high-performance liquid chromatography and by gas chromatography-mass spectrometry to obtain anatomical and compositional data. Microscopy demonstrated the development and maturation of cortical fibers which eventually became confluent with the fiber sheath surrounding vascular bundles in the lower internodes. Detergent fiber analysis indicated increasing cellulose and lignin contents and decreases in cell solubles and hemicelluloses with increasing distance of the internodes from the top of the plant. Soluble phenolics were greatest in amounts and complexity in top internodes. The lower internodes contained greater levels of wall-bound phenolic acids, principally as 4-coumarate and ferulate.  相似文献
