This work illustrates the feasibility of vermitechnology to stabilize sludge from an agro-industry. To achieve the goal, industrial sludge (IS) was mixed with three different bulky agents, i.e. cow dung (CD), biogas plant slurry (BGS) and wheat straw (WS), in different ratios to produce nine different feed mixtures for earthworm Eisenia fetida. Vermicomposting bedding material was analyzed for its different physic-chemical parameters after 15 weeks of experimentations. In all waste mixtures, a decrease in pH, organic C and C:N ratio, but increase in total N, available P, exchangeable K, exchangeable Ca and trace elements (Mg, Fe and Zn) was recorded. IS (40%) + CD (60%) and IS (40%) + BGS (60%) vermibeds showed the highest mineralization rate and earthworm growth patterns during vermicomposting process. Vermicompost contains (dry weight basis) a considerable range of plant available forms of P (17.5–28.9 g kg?1), K (13.8–21.4 g kg?1), Ca (41.1–63.4 g kg?1), Mg (262.4–348.3 mg kg?1), Fe (559.8–513.0 mg kg?1) and Zn (363.1–253.6 mg kg?1). Earthworm growth parameters, i.e. biomass gain, total cocoon production, individual growth rate (mg wt. worm?1 day?1), natality rate, total fecundity were optimum in bedding containing 20–40% industrial sludge. C:N ratio of worm-processed material was within the agronomic acceptable or favorable limit (<15–20). The results clearly suggested that vermitechnology can be a potential technology to convert industrial sludges into vermifertilizer for sustainable land restoration practices.  相似文献

In this work, straw hydrolysates were used to produce succinic acid by Actinobacillus succinogenes CGMCC1593 for the first time. Results indicated that both glucose and xylose in the straw hydrolysates were utilized in succinic acid production, and the hydrolysates of corn straw was better than that of rice or wheat straw in anaerobic fermentation of succinic acid. However, cell growth and succinic acid production were inhibited when the initial concentration of sugar, which was from corn straw hydrolysate (CSH), was higher than 60 g l?1. In batch fermentation, 45.5 g l?1 succinic acid concentration and 80.7% yield were attained after 48 h incubation with 58 g l?1 of initial sugar from corn straw hydrolysate in a 5-l stirred bioreactor. While in fed-batch fermentation, concentration of succinic acid achieved 53.2 g l?1 at a rate of 1.21 g l?1 h?1 after 44 h of fermentation. Our work suggested that corn straw could be utilized for the economical production of succinic acid by A. succinogenes.  相似文献

The impact of conservation tillage practices on soil carbon has been of great interest in recent years. Conservation tillage might have the potential to enhance soil carbon accumulation and alter the depth distribution of soil carbon compared to conventional tillage based systems. Changes in the soil organic carbon (SOC) as influenced by tillage, are more noticeable under long-term rather than short-term tillage practices. The objective of this study was to determine the impacts of long-term tillage on SOC and dissolved organic carbon (DOC) status after 19 years of four tillage treatments in a Hydragric Anthrosol. In this experiment four tillage systems included conventional tillage with rotation of rice and winter fallow system (CTF), conventional tillage with rotation of rice and rape system (CTR), no-till and ridge culture with rotation of rice and rape system (NT) and tillage and ridge culture with rotation of rice and rape system (TR). Soils were sampled in the spring of 2009 and sectioned into 0–10, 10–20, 20–30, 30–40, 40–50 and 50–60 cm depth, respectively.Tillage effect on SOC was observed, and SOC concentrations were much larger under NT than the other three tillage methods in all soil depths from 0 to 60 cm. The mean SOC concentration at 0–60 cm soil depth followed the sequence: NT (22.74 g kg?1) > CTF (14.57 g kg?1) > TR (13.10 g kg?1) > CTR (11.92 g kg?1). SOC concentrations under NT were significantly higher than TR and CTR (P < 0.01), and higher than CTF treatment (P < 0.05). The SOC storage was calculated on equivalent soil mass basis. Results showed that the highest SOC storage at 0–60 cm depth presented in NT, which was 158.52 Mg C ha?1, followed by CTF (106.74 Mg C ha?1), TR (93.11 Mg C ha?1) and CTR (88.60 Mg C ha?1). Compared with conventional tillage (CTF), the total SOC storage in NT increased by 48.51%, but decreased by 16.99% and 12.77% under CTR and TR treatments, respectively. The effect of tillage on DOC was significant at 0–10 cm soil layer, and DOC concentration was much higher under CTF than the other three treatments (P < 0.01). Throughout 0–60 cm soil depth, DOC concentrations were 32.92, 32.63, 26.79 and 22.10 mg kg?1 under NT, CTF, CTR and TR, and the differences among the four treatments were not significant (P > 0.05). In conclusion, NT increased SOC concentration and storage compared to conventional tillage operation but not for DOC.  相似文献

Shrimp head waste is a major byproduct of crustacean processing in North-eastern Brazil and represents an interesting source of bioactive molecules. Additionally, its use increases the sustainability of processing fishery products. The present study reports a process developed for recovering bioactive molecules from shrimp heads through autolysis. A protein hydrolysate (120 ± 0.4 g) formed by a 9% (w/v) solution was recovered and lyophilized from 1 kg of shrimp heads. Approximately 195 ± 0.5 mg of carotenoids was recovered as an ethanolic extract. The recovery of chitin and chitosan were 25 ± 2 g kg?1 and 17 ± 4 g kg?1 wet processing waste, respectively. Chitosans were characterized by 13C NMR, and FT-IR analysis and exhibited a variable degree of deacetylation (60–80%). Sulfated glycosaminoglycans that exhibited electrophoretic migration similar to mammalian standards were also recovered (79 ± 2 mg kg?1 wet processing waste), and their degradation products suggested the presence of C6-sulfated heparan sulfate. These data point to the feasibility of an integrated process for isolating highly bioactive molecules, such as sulfated- and amino-polysaccharides, with a broad spectrum of applications from shrimp processing waste.  相似文献

Mine tailings are an environmental problem in Southern Spain because wind and water erosion of bare surfaces results in the dispersal of toxic metals over nearby urban or agricultural areas. Revegetation with tolerant native species may reduce this risk. We grew two grasses, Lygeum spartum and Piptatherum miliaceum, and the crop species Cicer arietinum (chickpea) under controlled conditions in pots containing a mine tailings mixed into non-polluted soil to give treatments of 0%, 25%, 50%, 75% and 100% mine tailings. We tested a neutral (pH 7.4) mine tailings which contained high concentrations of Cd, Cu, Pb and Zn. Water-extractable metal concentrations increased in proportion to the amount of tailings added. The biomass of the two grasses decreased in proportion to the rate of neutral mine-tailing addition, while the biomass of C. arietinum only decreased in relation to the control treatment. Neutron radiography revealed that root development of C. arietinum was perturbed in soil amended with the neutral tailings compared to those of the control treatment, despite a lack of toxicity symptoms in the shoots. In all treatments and for all metals, the plants accumulated higher concentrations in the roots than in shoots. The highest concentrations occurred in the roots of P. miliaceum (2500 mg kg?1 Pb, 146 mg kg?1 Cd, 185 mg kg?1 Cu, 2700 mg kg?1 Zn). C. arietinum seeds had normal concentrations of Zn (70–90 mg kg?1) and Cu (6–9 mg kg?1). However, the Cd concentration in this species was ~1 mg kg?1 in the seeds and 14.5 mg kg?1 in shoots. Consumption of these plant species by cattle and wild fauna may present a risk of toxic metals entering the food chain.  相似文献

Numerous studies have demonstrated that endotoxin plays an important role in the development and progression of hepatic cirrhosis. However, there is no effective remedy for the prevention and treatment of intestinal endotoxemia. Taurine has been reported to have beneficial effects on endotoxemia. Oats have been shown to absorb intestinal toxins and increase excretion of intestinal toxins. The present study was to investigate whether a combination of taurine and oat has an additive inhibitory effect on endotoxin release in a rat liver ischemia/reperfusion model. Our results showed that the combination of taurine (300 mg kg?1 d?1) and oat fiber (15 g kg?1 d?1) significantly reduced endotoxin levels in the portal vein by 36.3% when compared to the control group (0.168 ± 0.035 Eu/ml in the treatment group vs 0.264 ± 0.058 Eu/ml in the control group, P < 0.01). The treatment of taurine (300 mg kg?1 d?1) and oat fiber (15 g kg?1 d?1) induced 21.5% and 18.4% reduction in endotoxin levels, respectively, when compared to the control group (P < 0.05). We conclude that the combination of taurine and oat fiber achieved an additive inhibitory effect on intestinal endotoxin release, which might be an effective approach for the treatment of intestinal endotoxemia.  相似文献

Uptake and release of nutrients from ponds used for lotus cultivation were measured in ponds under short-term (1 yr) cultivation with compost application (pond I) and under long-term (20 yr) cultivation without compost application (pond II). Total inflow loads of TN (irrigation water, rainfall and compost) during lotus cultivation period in ponds I and II were 72.3 and 34.3 kg ha?1 182 day?1, respectively. TN removal rates in ponds I and II were 77.3 and 49.8% of total inflow load, respectively. Major removal mechanisms of TN were attributed to microbial processes and uptake by lotus. The total outflow loads (infiltration and runoff) of TN during the lotus cultivation period were 13.9 kg ha?1 182 day?1 (19.2% of total inflow TN load) for pond I, and 11.3 kg ha?1 182 day?1 (32.9% of total inflow TN load) for pond II. For TP the total inflow loads (irrigation water, rainfall and compost) during lotus cultivation in ponds I and II were 80.8 and 1.9 kg ha?1 182 day?1, respectively. TP removal rates in ponds I and II were 84.9 and ?274.1% of total input, respectively. Phosphorus removal was attributed to lotus uptake and soil adsorption. The total outflow loads (infiltration and runoff) of TP during lotus cultivation period were 10.1 kg ha?1 182 day?1 (12.5% of total inflow TP load) for pond I, and 6.6 kg ha?1 182 day?1 (355.6% of total inflow TP load) for pond II. TN and TP in runoff from pond I (with compost) was higher than that in pond II (without compost), showing that TN and TP in runoff were strongly influenced by compost addition. Therefore, in order to satisfy established water-quality standards, the amount of compost used in lotus cultivation should be evaluated.  相似文献

Biodegradation rate and the high molecular weight hydrocarbons are among the important concerns for bioremediation of crude oil. Inoculation of a non-oil-degrading bacterium as supplementary bacteria increased oil biodegradation from 57.1% to 63.0% after 10 days of incubation. Both the oil-degrading bacteria and the non-oil-degrading bacteria were isolated from Malaysian marine environment. Based on the 16S rDNA sequences, the oil-degrading bacteria was identified as Pseudomonas pseudoalcaligenes (99% similarity) while the non-oil-degrading bacterium was Erythrobacter citreus (99% similarity). E. citreus does not grow on crude oil enriched medium under present experimental condition but it withstands 5000 mg kg?1 Tapis blended crude oil in sediment. Under optimal condition, the oil-degrading bacterium; P. pseudoalcaligenes, alone utilized 583.3 ± 3.8 mg kg?1 (57.1%) at the rate of 3.97 × 10?10 mg kg?1 cell?1 day?1 Tapis blended crude oil from 1000 mg kg?1 oil-contaminated sediment. Inoculation of E. citreus as the supplementary bacteria to P. pseudoalcaligenes enhanced biodegradation. The bacterial consortium degraded 675.8 ± 18.5 mg kg?1 (63.0%) Tapis blended crude oil from the 1000 mg kg?1 oil-contaminated sediment. Biodegradation rate of the bacterial consortium increased significantly to 4.59 × 10?10 mg kg?1 cell?1 day?1 (p = 0.02). Improvement of the oil degradation by the bacterial consortium was due to the synergetic reaction among the bacterial inoculants. There are two implications: (1) E. citreus may have a role in removing self-growth-inhibiting compounds of P. pseudoalcaligens. (2) P. pseudoalcaligenes degraded Tapis blended crude oil while E. citreus competes for the partially degraded hydrocarbons by P. pseudoalcaligenes. P. pseudoalcaligenes forced to breakdown more hydrocarbons to sustain its metabolic requirement. The bacterial consortium degraded 78.7% of (C12–C34) total aliphatic hydrocarbons (TAHs) and 74.1% of the 16 USEPA prioritized polycyclic aromatic hydrocarbons.  相似文献

The aim of this study was to assess the suitability of sewage sludge use for mung bean {Vigna radiata L. cv. Malviya janpriya (HUM 6)} plants by evaluating the growth, and yield responses, nutritional quality and heavy metal accumulation at different sewage sludge amendment (SSA) rates. Sewage sludge amendment modified the physico-chemical properties of soil by decreasing pH and increasing organic carbon, total iron and heavy metals. Plants showed increments in shoot length, leaf area and total biomass at all SSA rates, but root length increased only up to 9 kg m?2 SSA rates. Plants grown at different SSA rates showed higher nutrients and heavy metals in seeds, but protein content declined. Sewage sludge application caused about 39, 76 and 60% more yield at 6, 9 and 12 kg m?2 treatments, respectively. Concentrations of Pb and Ni in grains were higher than the Indian permissible limits at and above 9 kg m?2 and of Cd at 12 kg m?2 SSA rates.The study suggests that SSA at a rate lower than 9 kg m?2 may be recommended due to better fertilizing value for soil and promoting mung bean yield. Higher rate of sewage sludge application leads to elevated accumulation of heavy metals in seeds, which limits the suitability for human consumption.  相似文献

There is a major need to understand the historical condition and chemical/biological functions of the ecosystems following a conversion of wetlands to agricultural functions. To better understand the dynamics of soil total organic carbon (TOC) and phosphorus (P) during beef cattle pastures to wetland reconversion, soil core samples were collected from the beef cattle pasture and from the natural wetland at Plant City, FL, during five summer seasons (2002–2007). The levels of TOC and soil P were significantly affected by changing land use and hydrology. Draining natural wetlands to grazed pastures resulted in very pronounced reduction of TOC from 180.1 to 5.4 g g?1. Cumulative concentrations of total phosphorus (TP) in soils (1134 mg kg?1) under drained condition are two to three times lower than those in soils (2752 mg kg?1) under flooded condition over the periods of land use reconversion. There was a declining trend (r = 0.82**; p  0.01) in total soil P from natural wetland (763 mg kg?1) to altered pastures (340 mg kg?1), largely as organic-bound P (natural wetland, 48%; grazed pastures, 44%; altered pastures, 29%). These results are important in establishing baseline information on soil properties in pasture and wetland prior to restoring and reconverting pasture back to wetland conditions. The results further suggest that changes in soil properties due to changing land use and hydrologic conditions (drying and re-wetting) could be long lasting.  相似文献

Arsenic hyperaccumulation by Pteris vittata L. (Chinese brake fern) may serve as a defense mechanism against herbivore attack. This study examined the effects of arsenic exposure (0, 5, 15 and 30 mg kg?1) on scale insect (Saissetia neglecta) infestation of P. vittata. Scale insects were counted as a percentage fallen from the plant to the total number of insects after 1 week of As-treatment. The arsenic concentrations in the fronds ranged from 5.40 to 812 mg kg?1. Greater arsenic concentrations resulted in higher percentage of fallen-scale insects (17.2–55.0%). Lower arsenic concentrations (≤5 mg kg?1) showed significantly lower effect on the population compared to 15–30 mg kg?1 (p < 0.05). Arsenic content in the fallen-scale insects was as high as 194 mg kg?1, which indicated that arsenic has been ingested by the scale insects via plant sap. This study is consistent with the hypothesis that arsenic may help P. vittata defend against herbivore's attack.  相似文献

This study tested the effect of calcium oxide (CaO), sodium hydroxide (NaOH) and NaOH plus hydrogen peroxide (H2O2; AHP) on cell wall composition, digestion and fermentation of wheat straw (straw) in sheep. Treated straws were prepared by mixing straw either with water followed by dusting with CaO at 160 g kg−1 DM or with a NaOH solution alone at 3 l kg−1 DM to supply 80 g NaOH kg−1 DM (Na) or pre-soaked with Na exactly as in the previous treatment for 27 h followed by mixing with 130 g H2O2 kg−1 DM (AHP) for 6 h. After 14 days of storage, the treated straws and an untreated straw (U) were fed automatically every 2 h to four individually housed sheep together with a supplement in a 4×4 latin square experiment. Each kilogram supplement DM contained 422 g CP and 10.8 MJ ME. NDF (p<0.001) and hemicellulose (p<0.01) contents were significantly reduced whereas cellulose was increased (p<0.001) in treated compared to untreated straw. ADL was reduced in Ca (p<0.05) but increased (p<0.05) in Na and AHP compared with U. The rumen and total tract digestibility were significantly (p<0.001) greater in sheep fed treated compared with untreated straw. Significant differences (p<0.05) between treatments for pH, NH3 and VFA were also observed. All treatments improved the nutritive value of straws compared with untreated through modification of cell wall with a subsequent increase in digestibility by sheep. Although the digestibility for Ca was lower than that for Na despite reduction in cell wall, its use to treat straws may be more safe and cost effective than Na. AHP was the most effective and could also improve the energy value of other low quality forages for ruminants. However, the need of AHP for a large amount of NaOH to achieve highly alkaline pH limits its farm scale application. Therefore, further studies should either consider reducing the amount of NaOH or finding alternative alkalis that are cost effective and user-friendly.  相似文献

Hydrocarbon biodegradation in clayed and weathered polluted soils is a challenge; thus the aim of the present study was to determine the best experimental conditions that improve the hydrocarbon biodegradability in clayed and weathered polluted soils, modifying the nitrogen and phosphorous content considering the C:N:P ratio and the water content as a percentage of the water-holding capacity of the soil. Biodegradation tests were performed in microcosms containing 20 g of dry soil at 30 °C. A uniform-precision central composite design of second order with three levels was used to assess the effect of nutrient and water content adjustment on the hydrocarbon degradation rate, total carbon consumption, and microbial activity. The results showed that the water-holding capacity corresponding to 350% and a C:N:P ratio of 100:7.5:0.66 were the best experimental conditions for obtaining the highest hydrocarbon degradation rate (1145 mg TPH kg?1 dry soil day?1), whereas the hydrocarbon degradation rate in a non-stimulated control was only 129 mg TPH kg?1 dry soil day?1. Water content was the factor that showed the highest significant effect (p  0.05) on the hydrocarbon degradation rate. The results of the present study allowed the achievement of the best experimental conditions that enhance hydrocarbon biodegradability in clayed and weathered polluted soils. Also, these conditions are proposed for use as a biodegradability assay.  相似文献

β-Lactoglobulin is one of the major components of bovine milk and it remains in a dimeric form under physiological conditions. The present contribution elucidates the structural change of β-lactoglobulin at pH 7.4 under the action of guanidine hydrochloride (GnHCl) and heat at the single molecular level. The only free cysteine (Cys-121) of β-lactoglobulin has been tagged with 7-diethylamino-3-(4-maleimidophenyl)-4-methylcoumarin (CPM) for this purpose. The dimeric structure of β-lactoglobulin found to undergoes a monomerization prior to the unfolding process upon being subjected to GnHCl. The hydrodynamic diameter of the native dimer, native monomer and the unfolded monomer has been estimated as ~ 55 Å, ~ 29 Å and ~ 37 Å, respectively. The free energy change for the monomerization and denaturation are respectively 1.57 kcal mol 1 and 8.93 kcal mol 1. With change in temperature, development of two types of aggregates (small aggregates and large aggregates) was observed, which is triggered by the formation of the monomeric structure of β-lactoglobulin. The hydrodynamic diameters of the smaller and larger aggregates have been estimated to be ~ 77 Å and ~ 117 Å, respectively. The formation of small aggregates turns out to be reversible whereas that of larger aggregates is irreversible. The free energy associated with these two steps are 0.69 kcal mol 1 and 9.09 kcal mol 1. Based on the size parameters, the smaller and larger aggregates have been proposed to contain ~twenty and ~sixty monomeric units. It has also been concluded that the monomeric subunits retain their native like secondary structure in these aggregates.  相似文献

Metal hyperaccumulation is of great interest in recent years because of its potential application for phytoremediation of heavy metal contaminated soils. In this study, a field survey and a hydroponic experiment were conducted to study the accumulation characteristics of lead (Pb), zinc (Zn) and cadmium (Cd) in Arabis paniculata Franch., which was found in Yunnan Province, China. The field survey showed that the wild population of A. paniculata was hyper-tolerant to extremely high concentrations of Pb, Zn and Cd, and could accumulate in shoots an average level of 2300 mg kg?1 dry weight (DW) Pb, 20,800 mg kg?1 Zn and 434 mg kg?1 Cd, with their translocation factors (TFs) all above one. Under the hydroponic culture, stimulatory effects of Pb, Zn and Cd on shoot dry biomass were noted from 24 to 193 μM Pb, 9 to 178 μM Cd and all Zn supply levels in nutrient solution, while the effects were not obvious in the roots. Chlorophyll concentrations in Pb, Zn and Cd treatments showed an inverted U-shaped pattern, consistent with the change of plant biomass. Pb, Zn and Cd concentrations in the shoots and roots increased sharply with increasing Pb, Zn and Cd supply levels. They reached > 1000 mg kg?1 Pb, 10,000 mg kg?1 Zn and 100 mg kg?1 Cd DW in the 24 μM Pb, 1223 μM Zn and 9 μM Cd treatment, respectively, in which the plants grew healthy and did not show any symptoms of phytotoxicity. The TFs of Zn were basically higher than one and the amount of Zn taken by shoots ranged from 78.7 to 90.4% of the total Zn. However, the TFs of Pb and Cd were well below one, and 55.0–67.5% of total Pb and 57.8–83.5% of total Cd was accumulated in the shoots. These results indicate that A. paniculata has a strong ability to tolerate and hyperaccumulate Pb, Zn and Cd. Meanwhile, suitable levels of Pb, Zn and Cd could stimulate the biomass production and chlorophyll concentrations of A. paniculata. Thus, it provides a new plant material for understanding the mechanisms of stimulatory effect and co-hyperaccumulation of multiple heavy metals.  相似文献

Nitrate loss from drainage tiles across the cornbelt of the upper midwestern US is a result of intensive agriculture with limited crop diversity, extensive periods of fallow soil, and the need for high fertilizer applications to corn, all located on a hydrologically modified landscape. Two methods proposed to reduce tile nitrate export are managed or controlled drainage to limit tile flow and bioreactors to enhance denitrification. Nitrogen budgets and tile flow monitoring were conducted over two- to three-year periods between 2006 and 2009. We estimated N budgets in a seed corn-soybean rotation farming system near DeLand, east-central Illinois, USA, with free (FD) and controlled drainage (CD) patterned tile systems. In addition, wood chip filled trenches (bioreactors) were installed below the CD structures, one lined with plastic and one unlined. We measured daily tile flow and nitrate-N (NO3-N) concentrations and calculated cumulative N loss from the tile water at both FD and CD areas for a period of three cropping years. We also monitored the tile flow and nitrate concentration in inlet and outlet of the bioreactor associated with a CD system and evaluated the efficiency of the bioreactor for two cropping years. Most components of the N balance were unaffected by CD (yields and therefore N harvested, surface soil denitrification), and there was a negative N balance in the soybean cropping year (?165 and ?163 kg N ha?1 at FD and CD areas, respectively), whereas seed corn cropping in the following year resulted in positive N balances (29 and 34 kg N ha?1 at FD and CD areas, respectively). For two years, the overall N balances were ?136 and ?129 kg N ha?1 at FD and CD areas, respectively, consistent with other recent corn belt studies showing a small net depletion of soil organic N. Controlled drainage greatly reduced tile N export, with a three-year average loss of 57.2 kg N ha?1 yr?1 from FD compared to 17 kg N ha?1 yr?1 for CD. There was high uncertainty in denitrification measurements and thus the fate of missing N in the CD system remained unknown. Nitrate reduction efficiency of the bioreactor varied greatly, with periods where nearly 100% of the nitrate was denitrified. The overall efficiency of the bioreactor associated with the CD system in reducing the tile N load was 33%. When nitrate was non-limiting, the nitrate removal rate of the bioreactor was 6.4 g N m?3 d?1. Little N2O emission was found from the bioreactor bed and is not thought to be a problem with these systems. Both the tile bioreactor and controlled drainage greatly reduced tile nitrate export in this leaky seed corn and soybean agricultural field.  相似文献

The enzymatic saccharification of three different feedstocks, rice straw, bagasse and silvergrass, which had been pretreated with different dilute acid concentrations, was studied to verify how enzymatic saccharification was affected by the lignin composition of the raw materials. There was a quantitatively inverse correlation between lignin content and enzymatic digestibility after pretreatment with 1%, 2% and 4% sulfuric acid. The lignin accounted for about 18.8–21.8% of pretreated rice straw, which was less than the 23.1–26.5% of pretreated bagasse and the 21.5–24.1% of pretreated silvergrass. The maximum glucose yield achieved, under an enzyme loading 6.5 FPU g?1 DM for 72 h, was close to 0.8 g glucose/g glucan from the enzymatic hydrolysis of the pretreated rice straw; this was twice that from bagasse and silvergrass. A decrease in initial rate of glucose production was observed in all cases when the raw materials underwent enzymatic saccharification with 4% sulfuric acid pretreatment. It is suggested that the higher acid concentration led to an inhibition of β-glucosidase activity. Fourier transform infrared (FTIR) spectroscopy further indicated the chemical properties of the rice straw and silvergrass become more hydrophilic after pretreatment using 2% of sulfuric acid, but the pretreated bagasse tended to become more hydrophobic. The hydrophilic nature of the pretreated solid residues may increase the inhibitive effects of lignin on the cellulase and this could become very important for raw materials such as silvergrass that contain more lignin.  相似文献

Straighthead disease is a physiological disorder of rice (Oryza sativa L.) characterized by sterility of the florates/spikelets leading to reduced grain yield. Though the exact cause of straighthead is unknown, a glass house experiment was conducted to investigate the effect of inorganic arsenic on straighthead disease in rice (Oryza sativa L.). BRRI dhan 29, a popular Bangladeshi rice strain, was grown in soils spiked with arsenic (prepared from sodium arsenate, Na2HAsO4·7H2O) at the rate of 10, 20, 30, 40, 50, 60, 70, 80 and 90 mg of As kg?1 and one control treatment was also run to compare the results. Although there may be some other soil physico-chemical factors involved, arsenic concentration was found to be closely associated with straighthead of rice. With the increase of soil arsenic concentration, the severity of straighthead increased significantly. Up to the 50 mg of As kg?1 soil treatments, the severity of straighthead incidences were not prevalent. Straighthead resulted in sterile florets with distorted lemma and palea, reduced plant height, tillering, panicle length and grain yield. Straighthead caused approximately 17–100% sterile florates/spikelets formation and about 16–100% loss of grain yield. Straighthead also causes the reduction of panicle formation and panicle length significantly (p < 0.01). In the present study, panicle formation was found to be reduced by 21–95% by straighthead.  相似文献

There are currently few successful examples of using straw hemicellulose as a carbon source in the fermentation industry. In this paper, hemicellulose hydrolysates were recovered from steam-exploded wheat straw (SEWS) and used to produce microbial oil. The effects of the steam explosion treatment conditions, the elution temperature and the ratio of elution water to SEWS on sugar recovery were examined. A broth with 3.8 g l?1 of reducing sugar and 22.3 g l?1 of total soluble sugars was obtained with a 10-fold excess (w/w) of water at 40 °C to wash the SEWS treated under steam explosion conditions at 200 °C for 5 min. This broth was used to produce microbial oil by the oleaginous fungus Microsphaeropsis sp., which was able to secrete xylanase to degrade oligosaccharides from straw hemicellulose and accumulate microbial oil. Under optimized conditions, the oil concentration was 2.6 g l?1. The yield of oil from sugar consumed was 0.14 g g?1. The microbial oil produced by this research could be used as feedstock for biodiesel production because the microbial oil was primarily composed of neutral lipids. This research establishes a novel protocol for microbial oil production from straw hemicellulose.  相似文献

Restoration of the Chesapeake Bay poses significant challenges because of increasing population pressure, conversion of farmland to urban/suburban development, and the expense of infrastructure needed to achieve significant and sustained nutrient reductions from agricultural and urban sources. One radical approach for removing non-point source nutrients before they reach the bay is to deploy large-scale algal turf scrubbers along its tributaries. The objective of this study was to determine rates of nutrient removal and algal fatty acid production using small ATS units located along three Chesapeake Bay rivers. Small-scale ATS units (each containing 1 m2 growing area) were operated for 5–10 months from April 2007 to April 2008 on three western shore tributaries of the Chesapeake Bay in Maryland: the Bush River, the Patapsco River and the Patuxent River. Total nitrogen (TN) and total phosphorus (TP) removal rates at the Patuxent site fluctuated considerably but averaged 250 mg TN, 45 mg TP m?2 day?1 from May to October 2007, then decreased to 16 mg TN, 3 mg TP m?2 day?1 from December 2007 to February 2008. Nutrient removal rates at the Bush river site also fluctuated but averaged only 85 mg TN, 10 mg TP m?2 day?1 from May to June 2007, before decreasing to <10 mg TN, <1 mg TP m?2 day?1 from July to September 2007. The Patapsco River unit began operation in August 2007, reached its maximum removal values of 150 mg TN, 18 mg TP m?2 day?1 from mid-October to late-November 2007, then decreased to values of 45 mg TN, 4 mg TP m?2 day?1 from November 15, 2007 to mid-April 2008. In the best case (Patuxent site from May to October 2007), daily removal rates of 250 mg N and 45 mg P m?2 are equivalent to removal rates of 380 kg N and 70 kg P ha?1 over a 150-day season in Maryland. Fatty acid (FA) content of the harvested material was consistently low (0.3–0.6% of dry weight) and varied little between sites. Mean algal FA production rates (23–54 mg FA m?2 day?1) are equivalent to rates of 34–81 kg FA ha?1 year?1 based on a 150-day operational season in Maryland.  相似文献
