Biomass in the form of agricultural residues is becoming popular among new renewable energy sources, especially given its wide potential and abundant usage. Pyrolysis is the most important process among the thermal conversion processes of biomass. In this study, the various characteristics of bio-oils acquired under different pyrolysis conditions from safflower seed press cake (SPC) were identified. The elemental analyses and calorific values of the bio-oils were determined, and then the chemical compositions of the bio-oils were investigated using chromatographic and spectroscopic techniques such as column chromatography, 1H NMR, FTIR and GC. The fuel properties of the bio-oil such as kinematic viscosity, flash point, density, water content and ASTM distillation were also determined. Chemical compositions of bio-oils showed that some quantities of hydrocarbons were present, while oxygenated and polar fractions dominated. The bio-oils obtained from safflower seed press cake were presented as an environmentally friendly feedstock candidate for biofuels and chemicals.  相似文献

Ultrasound complex enzymes (UCE) were used to extract crude polysaccharides (CEP) from Epimedium leaves. The response surface methodology (RSM) based on Box–Behnken Design (BBD) was employed to further optimize the extraction conditions. The optimum conditions were: complex enzyme dose of 50, 250, 200, 100 U/g (papain, pectase, cellulase and α-amylase), extraction temperature 46.8 °C, ultrasound time 42.3 min, pH 4.28, and ultrasound power 311 W. Under these conditions, the experimental yield of CEP was 5.98%, which was well matched with the value predicted by the model. Three major polysaccharide fractions (EP-1, EP-2 and EP-3) were obtained by ultrafiltration, chromatography on DEAE-Sepharose Fast Flow and Sephadex G-100 column. Their chemical–physical characteristics were investigated using chemical methods, gas chromatography (GC) and high-performance size-exclusion chromatography (HPSEC). Biochemical characterization of the polysaccharide fractions was evaluated on the basis of free radical scavenging, ferric-reducing antioxidant power (FRAP), lymphocyte proliferation, macrophage phagocytosis and NK cell activity assay in vitro. The results showed that all fractions contained higher galacturonic acid (GalA) and galactose (Gal), possessed considerable antioxidant activity, and could promote proliferation of splenic lymphocytes, activate macrophages and enhance NK cells’ activity. UCE as a more effective and environment-friendly technique could be employed for extracting active ingredients from plant materials, extensively applied in pharmaceutical and food industries.  相似文献

Headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography–mass spectrometry (GC–MS) analysis was used to study volatile and semi-volatile compounds emitted by the Eupatorium odoratum (E. odoratum) extract. Variables of HS-SPME such as the type of SPME fiber, extraction time and temperature, incubation time, desorption time and temperature have been optimized. Optimized conditions were obtained by the use of divinylbenzene/carboxen/polydimethylsiloxane (DVB/CAR/PDMS) fiber, 5 min/20 min incubation/extraction time at 65 °C, 5 min desorption time at 260 °C. Using three different polar chromatographic columns to get retention index and mass spectrometry data, 99 volatile and semi-volatile compounds were tentatively identified in the E. odoratum extract. This study has identified the promising source of E. odoratum oviposition repellent.  相似文献

Poplar leaves, poplar bark and poplar wood were deoxy-liquefied directly in an air-proof stainless steel reactor at different temperatures. The oils from leaves at 350 °C, from bark at 400 °C and from wood at 450 °C, at which the liquid product yields were the maximum, were analyzed by GC–MS. The oils obtained from three parts of poplar tree were quite different from each other in the relative contents of their compositions. The oil from leaves was rich in hydrocarbons (alkanes: C7–C29; aromatics) and poor in phenolics, while oil from wood was rich in phenolics and poor in hydrocarbons. The oil from bark was moderate. Relative contents of hydrocarbons in the leaves oil were as high as 60.01% but decreased to 29.71% in bark oil and 11.43% in wood oil. GC analysis of gases and FT-IR, GC–MS and elemental analysis of oils were performed in this study.  相似文献

The residues from the palm oil industry are the main contributors to biomass waste in Malaysia, and these wastes require extra attention with respect to handling. The biomass waste is a renewable resource that can potentially be used to produce absorbents, fuels, and chemical feedstocks through the pyrolysis process. In this study, the wastes of palm shell, empty fruit bunches, and mesocarp fiber were characterized and then pyrolyzed in a fixed-bed reactor under the following conditions: a temperature of 500 °C, a nitrogen flow rate of 2 L/min and reaction time of 60 min. After pyrolysis, characterization of the products with an emphasis on the bio-oil and the bio-char was performed using various approaches (including Karl Fischer water-content tests, FTIR, SEM, TGA and CNH/O analyses). The results showed that the pyrolysis of palm oil wastes yielded more bio-oil than bio-char or non-condensable gases. The results also indicated that all of the bio-oils were acidic and contained high levels of oxygen. The bio-oils heating values were low and varied from 10.49 MJ/kg to 14.78 MJ/kg. The heating values of the bio-chars (20–30 MJ/kg) were higher than those of the bio-oils. Among the biomasses studied in this work, palm shell contained the highest level of lignin and showed the highest levels of bio-char yield and fixed and elemental carbon in the raw and bio-char form.  相似文献

Molasses fermentation performance by both a cryotolerant and a thermophilic yeast (strain AXAZ-1) isolated from grapes in Greece was evaluated in an extremely wide temperature range (3–40 °C). Sequence analysis of the 5.8S internal transcribed spacer and the D1/D2 ribosomal DNA (rDNA) regions assigned isolate to Saccharomyces cerevisiae. Restriction fragment length polymorphism of the mitochondrial DNA showed that strain AXAZ-1 is genetically divergent compared to other wild strains of Greek origin or commercial yeast starters. Yeast cells growing planktonically were capable of fermentation in a wide temperature spectrum, ranging from 3 °C to 38 °C. Immobilization of yeast on brewer’s spent grains (BSG) improved the thermo-tolerance of the strain and enabled fermentation at 40 °C. Time to complete fermentation with the immobilized yeast ranged from 20 days at 3 to 38 h at 40 °C. The daily ethanol productivity reached maximum (58.1 g/L) and minimum (2.5 g/L) levels at 30 and 3 °C, respectively. The aroma-related compounds’ profiles of immobilized cells at different fermentation temperatures were evaluated by using solid phase microextraction (SPME) gas chromatography–mass spectrometry (GC–MS). Molasses fermentation resulted in a high quality fermentation product due to the low concentrations of higher and amyl alcohols at all temperatures tested. Strain AXAZ-1 is very promising for the production of ethanol from low cost raw materials, as it was capable to perform fermentations of high ethanol concentration and productivities in both low and high temperatures.  相似文献

Urinary amino acid analysis is typically done by cation-exchange chromatography followed by post-column derivatization with ninhydrin and UV detection. This method lacks throughput and specificity. Two recently introduced stable isotope ratio mass spectrometric methods promise to overcome those shortcomings. Using two blinded sets of urine replicates and a certified amino acid standard, we compared the precision and accuracy of gas chromatography/mass spectrometry (GC–MS) and liquid chromatography–tandem mass spectrometry (LC–MS/MS) of propyl chloroformate and iTRAQ® derivatized amino acids, respectively, to conventional amino acid analysis. The GC–MS method builds on the direct derivatization of amino acids in diluted urine with propyl chloroformate, GC separation and mass spectrometric quantification of derivatives using stable isotope labeled standards. The LC–MS/MS method requires prior urinary protein precipitation followed by labeling of urinary and standard amino acids with iTRAQ® tags containing different cleavable reporter ions distinguishable by MS/MS fragmentation. Means and standard deviations of percent technical error (%TE) computed for 20 amino acids determined by amino acid analyzer, GC–MS, and iTRAQ®–LC–MS/MS analyses of 33 duplicate and triplicate urine specimens were 7.27 ± 5.22, 21.18 ± 10.94, and 18.34 ± 14.67, respectively. Corresponding values for 13 amino acids determined in a second batch of 144 urine specimens measured in duplicate or triplicate were 8.39 ± 5.35, 6.23 ± 3.84, and 35.37 ± 29.42. Both GC–MS and iTRAQ®–LC–MS/MS are suited for high-throughput amino acid analysis, with the former offering at present higher reproducibility and completely automated sample pretreatment, while the latter covers more amino acids and related amines.  相似文献

Protease enzyme from Bacillus megaterium was successively purified by ammonium sulfate precipitation, ion exchange chromatography on DEAE-cellulose and gel filtration chromatography on Sephadex G-200. The purification steps of protease resulted in the production of two protease fractions namely protease P1 and P2 with specific activities of 561.27 and 317.23 U mg?1 of protein, respectively. The molecular weights of B. megaterium P1 and P2 were 28 and 25 KDa, respectively. The purified fractions P1 and P2 were rich in aspartic acid and serine. Relatively higher amounts of alanine, leucine, glycine, valine, thereonine valine and glutamic acid were also present. The maximum protease activities for both enzyme fractions were attained at 50 °C, pH 7.5, 1% of gelatine concentration and 0.5 enzyme concentrations. P1 and P2 fractions were more stable over pH 7.0–8.5 and able to prolong their thermal stability up to 80 °C. The effect of different inhibitors on the protease activity of both enzyme fractions was also studied. The enzyme was found to be serine active as it had been affected by lower concentrations of phenylmethylsulfonyl fluoride (PMSF). Complete dehairing of the enzyme-treated skin was achieved in 12 h, at room temperature.  相似文献

Cornstalk powders were effectively liquefied in a hot-compressed phenol–water medium (1:4 wt/wt). The optimum liquefaction temperature was around 350 °C, where the liquid yield attained a maximum at about 70 wt%. The addition of sodium carbonate showed negligible effect over the Liquefaction product yields. When liquefied in phenol–water medium, essentially no phenol was combined with the liquid products, and the cornstalk-derived bio-oils were partially degraded monomeric and oligomeric products with a broad molecular distribution. Resol type bio-based phenol formaldehyde resins were readily synthesized from the cornstalk-derived bio-oils catalyzed by sodium hydroxide.  相似文献

A simple and reliable solid phase microextraction/gas chromatography–mass spectrometry (SPME/GC–MS) method was developed for the single-step determination of PCBs 126 and 153 in rat brain and serum, using liquid/liquid and solid phase extraction (SPE) as reference techniques. The multi-factor categorical experimental design used to study simultaneously the main parameters and their interactions affecting the efficiency of the method, showed that the use of an 85 μm PA exposed at 100 °C for 40 min was the optimum sampling condition for both PCBs. SPME was then validated by studying its linear dynamic (over two orders of magnitude), limits of detection (brain: 2 ng/g, serum: 0.2 ng/g) and analytical precision that was within 9% for SPME in both brain and serum. Finally, the method was used to determine the brain and blood target dose in mothers and pups after oral exposure of the mothers.  相似文献

α-Bisabolol is a commercially important aroma chemical currently obtained from the Candeia tree (Vanillosmopsis erythropappa). Continuous unsustainable harvesting of the Candeia tree has prompted the urgent need to identify alternative crops as a source of this commercially important sesquiterpene alcohol. A chemotaxonomic assessment of two Salvia species indigenous to South Africa is presented and recommended as a potential source of α-bisabolol. The essential oil obtained by hydrodistillation of the aerial parts was analysed by gas chromatography coupled to mass spectrometry (GC–MS) and mid-infrared spectroscopy (MIRS). Orthogonal projections to latent structures–discriminant analysis (OPLS–DA) were used for multivariate classification of the oils based on GC–MS and MIRS data. Partial least squares (PLS) calibration models were developed on the MIRS data for the quantification of α-bisabolol using GC–MS as the reference method. A clear distinction between Salvia stenophylla and Salvia runcinata oils was observed using OPLS–DA on both GC–MS and MIRS data. The MIR calibration model showed high coefficient of determination (R2 = 0.999) and low error of prediction (RMSEP = 0.540%) for α-bisabolol content.  相似文献

Hydro-distilled essential oil from Kenyan Piper capense (Piperaceae) was analysed by gas chromatography mass spectrometry (GC–MS) and evaluated for larvicidal activity against the malaria vector, Anopheles gambiae. The oil consisted mainly of sesquiterpene hydrocarbons which accounted for 43.9% of the oil. The major sesquiterpenes were δ-cadinene (16.82%), β-bisabolene (5.65%), and bicyclogermacrene (3.30%). The oil also had appreciable amounts of monoterpene hydrocarbons (30.64%), including β-pinene (7.24%) and α-phellandrene (4.76%), and arylpropanoids (8.64%), including myristicin (4.26%). The oil showed larvicidal activity against third instar larvae of A. gambiae, with LC50 and LC90 values of 34.9 and 85.0 ppm, respectively. Most of the larvae died within the first few hours. The high larvicidal activity of this oil was indicated by the fact that over 80% mortality was observed at a concentration of 100 ppm after 24 h. These results compared favourably with the commercial larvicide pylarvex® which had LC50 and LC90 values of 3.7 and 7.8 ppm, respectively. Application of this oil or of products derived from it to larval habitats may lead to promising results in malaria and mosquito management programmes.  相似文献

A simple, sensitive, and inexpensive singe-drop microextraction (SDME) followed by gas chromatography and flame-ionization detection (GC-FID) was developed for determination of nicotine, anabasine, and cotinine in human urine and saliva samples. The target compounds were extracted from alkaline aqueous sample solution into an organic acceptor drop suspended on the tip of a 25-μL GC microsyringe in the aqueous sample solution. This microsyringe was also used for direct injection after extraction. Under optimized experimental conditions, calibration plots were found to be linear in the range of 0.5–25.0, 0.5–65.0, and 0.5–45.0 mg L?1 for nicotine, anabasines and cotinine, respectively. The method detection limit values were in the range of 0.33–0.45 mg L?1. Intra-day and inter-day precisions for peak area ratios were in the range of 1.3–9.2% and 2.0–7.0%, respectively. The proposed procedure was successfully applied to the determination of analytes in spiked urine and saliva samples with satisfactory results. The mean relative recoveries of spiked water samples ranged over 71.2–111.0%, with relative standard deviations varying from 2.3% to 10.0%.  相似文献

In this work, catalytic cracking of biomass waste oil fractions separated from food waste leachate was performed using microporous catalysts, such as HY, HZSM-5 and mesoporous Al-MCM-48. The experiments were carried out using pyrolysis gas chromatography/mass spectrometry (Py-GC/MS) to allow the direct analysis of the pyrolytic products. Most acidic components, especially oleic acid, contained in the food waste oil fractions were converted to valuable products, such as oxygenates, hydrocarbons and aromatics. High yields of hydrocarbons within the gasoline-range were obtained when microporous catalysts were used; whereas, the use of Al-MCM-48, which exhibits relatively weak acidity, resulted in high yields of oxygenated and diesel-range hydrocarbons. The HZSM-5 catalyst produced a higher amount of valuable mono aromatics due to its strong acidity and shape selectivity. Especially, the addition of gallium (Ga) to HZSM-5 significantly increased the aromatics content.  相似文献

Meconium fatty acid ethyl esters (FAEEs) are currently used as biomarkers to detect heavy prenatal alcohol exposure. We introduce a novel technique to quantify FAEEs in meconium using headspace-solid-phase microextraction (HS-SPME) coupled with gas chromatography–mass spectrometry (GC–MS). This method improves on previous approaches by decreasing sample preparation time, eliminating the need for organic solvents, and reducing the required sample size. Using 50 mg of meconium, the detection limits of FAEEs ranged from 0.05 to 0.16 nmol/g and had good reproducibility making it ideal for routine analysis of clinical samples.  相似文献

It has been previously demonstrated that both externally generated and internally synthesized nitric oxide (NO) can affect red blood cell (RBC) deformability. Further studies have shown that the RBC has active NO synthesizing mechanisms and that these mechanisms may play role in maintaining normal RBC mechanical properties. However, hemoglobin within the RBC is known to be a potent scavenger of NO; oxy-hemoglobin scavenges NO faster than deoxy-hemoglobin via the dioxygenation reaction to nitrate. The present study aimed at investigating the role of hemoglobin oxygenation in the modulation of RBC rheologic behavior by NO. Human blood was obtained from healthy volunteers, anticoagulated with sodium heparin (15 IU/mL), and the hematocrit was adjusted to 0.4 L/L by adding or removing autologous plasma. Several two mL aliquots of blood were equilibrated at room temperature (22 ± 2 °C) with moisturized air or 100% nitrogen by a membrane gas exchanger, The NO donor sodium nitroprusside (SNP), at a concentration range of 10?7–10?4 M, was added to the equilibrated aliquots which were maintained under the same conditions for an additional 60 min. The effect of the non-specific NOS inhibitor l-NAME was also tested at a concentration of 10?3 M. RBC deformability was measured using an ektacytometer with an environment corresponding to that used for the prior incubation (i.e., oxygenated or deoxygenated). Our results indicate an improvement of RBC deformability with the NO donor SNP that was much more pronounced in the deoxygenated aliquots. SNP also had a more pronounced effect on RBC aggregation for deoxygenated RBC. Conversely, l-NAME had no effect on deoxygenated blood but resulted in impaired deformability, with no change in aggregation for oxygenated blood. These findings can be explained by a differential behavior of hemoglobin under oxygenated and deoxygenated conditions; the influence of oxygen partial pressure on NOS activity may also play a role. It is therefore critical to consider the oxygenation state of intracellular hemoglobin while studying the role of NO as a regulator of RBC mechanical properties.  相似文献

Brilliant green, used extensively to color silk and wool in the commercial textile industry is a hazardous recalcitrant. Aspergillus sp. strain CB-TKL-1 isolated from a water sample from Tsumoriri Lake, Karzok, Ladakh, India, was found to completely decolorize this dye within 72 h when cultured under aerobic conditions at 25 °C. The extent of decolorization was monitored by the decrease in absorbance maxima of the dye by UV–visible spectroscopy. The decolorization was optimum at pH 5 and 35 °C when agitated at 200 rpm. Addition of glucose (2%) as a carbon source and sodium nitrate (0.2%) as a nitrogen source enhanced the decolorization ability of the culture. The culture exhibited maximum extent of decolorization of brilliant green with a C:N ratio of 2.5 after 72 h. Thirteen N-demethylated decolorized products of brilliant green were identified based on UV–visible spectroscopy, Fourier Transform Infrared (FT-IR) spectroscopy and liquid chromatography–electrospray ionization mass spectrometry (LC–ESI-MS) analysis at the end of 72 h before mineralization. The difference of the relative absorption peaks in the decolorized sample indicated a linear release of N-demethylated compounds, indicating a stepwise N-demethylation in the decolorization process.  相似文献

Natural product's properties are related to certain classes of compounds such as alkaloids, flavonoids, essential oils and others. Traditionally, separation techniques including thin layer chromatography (TLC), liquid chromatography (LC), gas chromatography (GC) and capillary electrophoresis (CE) even hyphenated to mass spectrometry (MS) were used for the elucidation, qualitative and quantitative analysis of individual compounds.In food industry, spectroscopic investigations using infrared radiation have been used to monitor and evaluate the composition and quality already since the early sixties. During the last four decades near-infrared spectroscopy (NIR; 800–2500 nm; 12,500–4000 cm−1) has become one of the most attractive and used methods for analysis for the following reasons: it represents a non-invasive analytical tool allowing a fast and simultaneous qualitative and quantitative characterization of natural products and their constituents. Additionally, the development of custom-made hand-held instruments enables in-field measurement for determining the optimum harvest time.Attenuated total reflection (ATR) and Fourier transform infrared (FTIR) spectroscopic imaging are suitable not only for the differentiation of different plant species, but also to distinct various ingredients within a plant. FTIR spectroscopic microscopy enables molecular imaging of complex botanical samples and therefore the detection and characterization of the molecular components of biological tissue.In the present contribution, the principle, technique and methodology of the different infrared spectroscopic methods are described followed by a discussion of quantitative and qualitative application possibilities in the field of natural product analysis.  相似文献

Serratia marcescens TKU011, a protease- and chitosanase-producing bacterium, the optimized condition for protease and chitosanase production was found after the media were heated at 121 °C for 120 min and the culture was shaken at 25 °C for 5 days in 100 mL of medium containing 1% squid pen powder (SPP) (w/v), 0.1% K2HPO4, and 0.05% MgSO4. An extracellular metalloprotease with novel properties of solvent stable, and alkaline was purified from the culture supernatant of S. marcescens TKU011 with squid pen wastes as the sole carbon/nitrogen source. The enzyme was a monomeric protease with a molecular mass of 48–50 kDa by SDS–PAGE and gel filtration chromatography. The optimum pH, optimum temperature, pH stability, and thermal stability of TKU011 protease were 8, 50 °C, pH 5–11, and <40 °C, respectively. Besides protease and chitosanase, with this method, deproteinization of squid pen for β-chitin, the production of peptide and reducing sugar may be useful for biological applications.  相似文献

In the present study liver samples (n = 26) of Northeast Arctic cod (Gadus morhua), ranging in total arsenic concentrations from 2.1 to 240 mg/kg liver wet weight (ww), were analysed for their content of total arsenic and arsenic species in the lipid-soluble and water-soluble fractions. The arsenic concentrations in the lipid fractions ranged from 1.8 to 16.4 mg As/kg oil of liver, and a linear correlation (r2 = 0.80, p < 0.001) was observed between the total arsenic concentrations in liver and the total arsenic concentrations in the respective lipid fractions of the same livers. The relative proportion of arsenolipids was considerably lower in liver samples with high total arsenic levels (33–240 mg/kg ww), which contained from 3 to 7% of the total arsenic in the lipid-soluble fraction. In contrast liver samples with low arsenic concentrations (2.1–33 mg/kg ww) contained up to 50% of the total arsenic as lipid-soluble species. Arsenic speciation analysis of the lipid-soluble fractions of the livers, using reversed-phase high performance liquid chromatography coupled to inductively coupled plasma mass spectrometry (HPLC–ICP-MS), revealed the presence of several arsenolipids. Three major arsenic-containing hydrocarbons (C17H39AsO, C19H41AsO and C23H37AsO) and five arsenic-containing fatty acids (C17H35AsO3, C19H39AO3, C19H37AsO3, C23H37AsO3 and C24H37AsO3) were identified using HPLC coupled to quadrupole time-of-flight mass spectrometry (qTOF-MS). Arsenobetaine was the major arsenic species in the water-soluble fraction of the livers, while dimethylarsinate, arsenocholine and inorganic arsenic were minor constituents. Inorganic arsenic accounted for less than 0.1% of the total arsenic in the liver samples.  相似文献
