Three novel strains capable of heterotrophic nitrification–aerobic denitrification were isolated from the landfill leachate
treatment system. Based on their phenotypic and phylogenetic characteristics, the isolates were identified as Agrobacterium sp. LAD9, Achromobacter sp. GAD3 and Comamonas sp. GAD4, respectively. Batch tests were carried out to evaluate the growth and the ammonia removal patterns. The maximum
growth rates as determined from the growth curve were 0.286, 0.228, and 0.433 h−1 for LAD9, GAD3 and GAD4, respectively. The maximum aerobic nitrification–denitrification rate was achieved by the strain
GAD4 of 0.381 mmol/l h, followed by LAD9 of 0.374 mmol/l h and GAD3 of 0.346 mmol/l h. Moreover, hydroxylamine oxidase and
periplasmic nitrate reductase were successfully expressed in all the isolates. The relationship between the enzyme activities
and the aerobic nitrification–denitrification rates revealed that hydroxylamine oxidation may be the rate-limiting step in
the heterotrophic nitrification–aerobic denitrification process. The study results are of great significance to the wastewater
treatment systems where simultaneous removal of carbon and nitrogen is desired. 相似文献
Efficient nitrification and denitrification of wastewater containing 1,700 mgl−1 of ammonium-nitrogen was achieved using aerobic granular sludge cultivated at medium-to-high organic loading rates. The cultivated
granules were tested in a sequencing batch reactor (SBR) fed with 6.4 or 10.2 kg NH4+-N m−3 day−1, a loading significantly higher than that reported in literature. With alternating 2 h oxic and 2 h anoxic operation (OA)
modes, removal rate was 45.5 mg NH4+-N g−1 volatile suspended solids−1 h−1 at 6.4 kg NH4+-N m−3 day−1 loading and 41.3 ± 2.0 at 10.2 kg NH4+-N m−3 day−1 loading. Following the 60 days SBR test, granules were intact. The fluorescence in situ hybridization and confocal laser
scanning microscopy results indicate that the SBR-OA granules have a distribution with nitrifers outside and heterotrophs
outside that can effectively expose functional strains to surrounding substrates at high concentrations with minimal mass
transfer limit. This microbial alignment combined with the smooth granule surface achieved nitrification–denitrification of
wastewaters containing high-strength ammonium using aerobic granules. Conversely, the SBR continuous aeration mode yielded
a distribution with nitrifers outside and heterotrophs inside with an unsatisfactory denitrification rate and floating granules

Excess inorganic nitrogen in water poses a severe threat to enviroment. Removal of inorganic nitrogen by heterotrophic nitrifying–aerobic denitrifying microorganism is supposed to be a promising and applicable technology only if the removal rate can be maintained sufficiently high in real wastewater under various conditions, such as high concentration of salt and wide range of different nitrogen concentrations. Here, a new heterotrophic nitrifying–aerobic denitrifying bacterium was isolated and named as Pseudomonas mendocina TJPU04, which removes NH4+-N, NO3−-N and NO2−-N with average rate of 4.69, 5.60, 4.99 mg/L/h, respectively. It also maintains high nitrogen removal efficiency over a wide range of nitrogen concentrations. When concentration of NH4+-N, NO3−-N and NO2−-N was up to 150, 150 and 50 mg/L, 98%, 93%, and 100% removal efficiency could be obtained, respectively, after 30-h incubation under sterile condition. When it was applied under non-sterile condition, the ammonia removal efficiency was slightly lower than that under sterile condition. However, the nitrate and nitrite removal efficiencies under non-sterile condition were significantly higher than those under sterile condition. Strain TJPU04 also showed efficient nitrogen removal performance in the presence of high concentration of salt and nitrogen. In addition, the removal efficiencies of NH4+-N, NO3−-N and TN in real wastewater were 91%, 52%, and 75%, respectively. These results suggest that strain TJPU04 is a promising candidate for efficient removal of inorganic nitrogen in wastewater treatment.
To enhance the startup and efficient simultaneous nitrification and denitrification for sewage treatment, sequencing batch biofilm reactors (SBBRs) partially coupled with rice husk were established and operated under various intermittent micro-aeration cycles (IMCs) and COD/N ratios under oxygen-limiting intermittent aeration conditions. Experimental results showed that the increase of IMCs with non-aeration/micro-aeration mode of (8 h/4 h)1 to (2 h/1 h)4 in a 12 h-cycle accelerated the startup performance and improved NH4+–N and COD removal. NH4+–N, TN and COD removal efficiencies were 98.7?±?0.9, 89.2?±?5.2 and 82.9?±?6.7% at COD/N ratio of 7.6 with the highest IMCs in SBBR, respectively. Higher TN removal efficiencies of 87.2?±?4.0 and 58.1?±?3.5% were also achieved at lower COD/N ratio of 5.6 and 2.8, respectively. In SBBRs with various IMCs, facultative denitrifier like genus Acinetobacter and solid-phase denitrifier belonging to Comamonadaceae family were enriched. However, aerobic denitrifiers with function of heterotrophic nitrification like Paracoccus were favored to enrich under higher IMCs condition, and more anoxic denitrifiers like sulfur-based autotrophic denitrifier Thiothrix and heterotrophic denitrifiers like Pseudomonas and Methyloversatilis were observed at lower IMCs condition. Stimulation of nitrification and denitrification by long term (from years to decades) grazing has commonly been reported in different grassland ecosystems. However, grazing generally induces important changes in plant species composition, and whether changes in nitrification and denitrification are primarily due to changes in vegetation composition has never been tested. We compared soil nitrification- and denitrification-enzyme activities (NEA and DEA, respectively) between semi-natural grassland sites experiencing intensive (IG) and light (LG) grazing/mowing regimes for 13 years. Mean NEA and DEA (i.e. observed from random soil sampling) were higher in IG than LG sites. The NEA/DEA ratio was higher in IG than LG sites, indicating a higher stimulation of nitrification. Marked changes in plant species composition were observed in response to the grazing/mowing regime. In particular, the specific phytomass volume of Elymus repens was lower in IG than LG sites, whereas the specific volume of Lolium perenne was higher in IG than LG sites. In contrast, the specific volume of Holcus lanatus, Poa trivialis and Arrhenatherum elatius were not significantly different between treatments. Soils sampled beneath grass tussocks of the last three species exhibited higher DEA, NEA and NEA/DEA ratio in IG than LG sites. For a given grazing regime, plant species did not affect significantly soil DEA, NEA and NEA/DEA ratio. The modification of plant species composition is thus not the primary factor driving changes in nitrification and denitrification in semi-natural grassland ecosystems experiencing long term intensive grazing. Factors such as trampling, N returned in animal excreta, and/or modification of N uptake and C exudation by frequently defoliated plants could be responsible for the enhanced microbial activities. 相似文献
Poultry manure contains high levels of ammonia, which result in a suboptimal bioconversion to methane in anaerobic digesters (AD). A simultaneous process of nitrification, Anammox and denitrification (SNAD) in a continuous granular bubble column reactor to treat the anaerobically digested poultry manure was implemented. Thus, two strategies to achieve high efficiencies were proposed in this study: (1) ammonia overload to suppress nitrite oxidizing bacteria (NOB) and (2) gradual adaptation of the partial nitrification–Anammox (PN–A) biomass to organic matter. During the NOB-suppression stage, microbial and physical biomass characterizations were performed and the NOB abundance decreased from 31.3% to 3.3%. During the adaptation stage, with a nitrogen loading rate of 0.34 g L−1 d−1, a hydraulic retention time of 1.24 d and an influent COD/N ratio of 2.63 ± 0.02, a maximum ammonia and total nitrogen removal of 100% and 91.68% were achieved, respectively. The relative abundances of the aerobic and the anaerobic ammonia-oxidizing bacteria were greater than 35% and 40% respectively, during the study. These strategies provided useful design tools for the efficient removal of nitrogen species in the presence of organic matter. 相似文献
Four samples of natural ecosystems and one sample from an activated sludge treatment plant were mixed together and progressively
adapted to alternating aerobic/anoxic phases in the presence of nitrate in order to enrich the microflora in aerobic denitrifiers.
Aerobic denitrifying performances of this mixed ecosystem at various dissolved oxygen concentrations and various carbon–nitrogen
loads were evaluated and compared to those obtained with the aerobic denitrifier Microvirgula aerodenitrificans. The consortium and the pure strain exhibited an aerobic denitrifying activity at air saturation conditions (7 mg dissolved
oxygen l–1), i.e. there was co-respiration of the two electron acceptors with significant specific nitrate reduction rates. Dissolved
oxygen concentrations had no influence on denitrifying performances above a defined threshold: 0.35 mg l–1 for the consortium and 4.5 mg l–1 for M. aerodenitrificans respectively. Under these thresholds, decreasing the dissolved oxygen concentrations enhanced the denitrifying activity of
each culture. The higher the carbon and nitrogen loads, the higher the performance of the aerobic denitrifying ecosystem.
However, for M. aerodenitrificans, the nitrate reduction percentage was affected more by variations in nitrogen load than in carbon load.

The presence of pathogenic bacteria is a major health risk factor in food samples and the commercial food supply chain is susceptible to bacterial contamination. Thus, rapid and sensitive identification methods are in demand for the food industry. Quantitative polymerase chain reaction (PCR) is one of the reliable specific methods with reasonably fast assay times. However, many constituents in food samples interfere with PCR, resulting in false results and thus hindering the usability of the method. Therefore, we aimed to develop an aptamer-based magnetic separation system as a sample preparation method for subsequent identification and quantification of the contaminant bacteria by real-time PCR. To achieve this goal, magnetic beads were prepared via suspension polymerization and grafted with glycidylmethacrylate (GMA) brushes that were modified into high quantities of amino groups. The magnetic beads were decorated with two different aptamer sequences binding specifically to Escherichia coli or Salmonella typhimurium. The results showed that even 1.0% milk inhibited PCR, but our magnetic affinity system capture of bacteria from 100% milk samples allowed accurate determination of bacterial contamination at less than 2.0 h with limit of detection around 100 CFU/mL for both bacteria in spiked-milk samples. 相似文献
Wheelchair control requires multiple degrees of freedom and fast intention detection, which makes electroencephalography (EEG)-based wheelchair control a big challenge. In our previous study, we have achieved direction (turning left and right) and speed (acceleration and deceleration) control of a wheelchair using a hybrid brain–computer interface (BCI) combining motor imagery and P300 potentials. In this paper, we proposed hybrid EEG-EOG BCI, which combines motor imagery, P300 potentials, and eye blinking to implement forward, backward, and stop control of a wheelchair. By performing relevant activities, users (e.g., those with amyotrophic lateral sclerosis and locked-in syndrome) can navigate the wheelchair with seven steering behaviors. Experimental results on four healthy subjects not only demonstrate the efficiency and robustness of our brain-controlled wheelchair system but also indicate that all the four subjects could control the wheelchair spontaneously and efficiently without any other assistance (e.g., an automatic navigation system). 相似文献

Summary Ammonium fertilizer applied to tea soils is readily converted to nitrate by the nitrifying bacteria in soil. Excess nitrate in soil could undergo rapid leaching losses under high rainfall conditions. Data is presented in this paper to show that waste tea could be effectively used to retard and delay nitrate production and thereby prevent loss of nitrogen as nitrate by leaching. BackgroundNanodiamonds (NDs) provide a unique multitasking system for drug delivery and fluorescent imaging in biological environments. Owing to their quantum properties, NDs are expected to be employed as multifunctional probes in the future for the accurate visualization of biophysical parameters such as temperature and magnetic fields. However, the use of NDs for the selective targeting of the biomolecules of interest within a complicated biological system remains a challenge. One of the most promising solutions is the appropriate surface design of NDs based on organic chemistry and biochemistry. The engineered NDs have high biocompatibility and dispersibility in a biological environment and hence undergo cellular uptake through specific pathways.Scope of reviewThis review focuses on the selective targeting of NDs for biomedical and biophysical applications from the viewpoint of ND surface functionalizations and modifications. These pretreatments make possible the specific targeting of biomolecules of interest on or in a cell by NDs via a designed biochemical route.Major conclusionsThe surface of NDs is covalently or noncovalently modified with silica, polymers, or biomolecules to reshape them, control their size, and enhance the colloidal stability and biomolecular selectivity toward the biomolecules of interest. Electroporation, chemical treatment, injection, or endocytosis are the methods generally adopted to introduce NDs into living cells. The pathway, efficiency, and the cell viability depend on the selected method.General significanceIn the biomedical field, the surface modification facilitates specific delivery of a drug, leading to a higher therapeutic efficacy.
