Studies of phenolic compounds were performed during cell suspension cultures in relation with the induction of embryogenic structures in two cultivars of cotton. Coker 312 produced embryogenic structures, unlike R405-2000 which was found to be a non-embryogenic cultivar. Embryogenesis induction in Coker 312 was strongly linked to a higher content of caffeic, ferulic and salicylic acids and to the appearance of p-coumaric acid, benzoic acid, trans-resveratrol, catechin and naringenin.  相似文献

The first method using high-performance liquid chromatography (HPLC) has been developed for the determination of trans-resveratrol in human plasma. The method involves a liquid–liquid extraction followed by reversed-phase HPLC with UV detection. The detection limit of trans-resveratrol in human plasma was 5.0 ng/ml. Standard curves are linear over the concentration range of 5.0–5000.0 ng/ml. Intra-assay variability ranged from 1.9 to 3.7% and inter-assay variability ranged from 2.5 to 4.0% at the concentration range of 15.0–4000.0 ng/ml.  相似文献

Biotransformation of piceid in Polygonum cuspidatum to resveratrol by Aspergillus oryzae was investigated in this study. Resveratrol is widely used in medicine, food, and cosmetic because of its pharmacological properties. However, it has a much lower content in plants compared with its glucoside piceid, which has a much lower bioavailability. Traditionally, the aglycone is acquired by acid or enzymatic hydrolysis of its glucoside, but the violent condition and the acid pollution in hydrolytic reaction and the high cost of the enzyme limit their industrial development. In this paper, fermentation of P. cuspidatum by A. oryzae was successfully performed, during which, piceid was converted to resveratrol with the highest yield of trans-resveratrol 1.35%, 3.6 times higher than that obtained from raw herb by microwave-assisted extraction. Scale-up production was also performed and the yield of trans-resveratrol was 3.1 times higher after 24 h incubation. Therefore, biotransformation is a better method to increase the yield of resveratrol because of its high yield and mild conditions.  相似文献

The polyphenol trans-resveratrol (3,5,4′-trihydroxy-trans-stilbene) is one of the best known plant secondary metabolites. The number of articles devoted to trans-resveratrol has been steadily increasing. Trans-resveratrol is a molecule that is beneficial to human health; this explains the high level of interest in trans-resveratrol among different research groups. Therefore, it is important to develop an effective method to produce this compound commercially. The applicability of biotechnology for trans-resveratrol extraction is still uncertain. This review describes and compares the available biotechnological methods of trans-resveratrol production, focusing on their advantages and disadvantages.  相似文献

Summary Genetic variability for virulence of the bacterial blight pathogen [Xanthomonas campestris pv malvacearum (Smith) Dye] on cotton (Gossypium hirsutum L.) has been shown by the identification of 19 races of the pathogen based on disease reactions of a set of ten host differentials. This study was conducted to determine the inheritance of host resistance to three recently identified isolates of X. campestris pv malvacearum, which are virulent on the entire set of differentials. True leaves of Tamcot CAMD-E, LEBOCAS-3-80, Stoneville 825, and their f1, F2, and backcross progenies were wound-inoculated in the field with separate bacterial suspensions of the virulent HV3, HV7, and Sudan isolates of the pathogen. LEBOCAS-3-80 was replaced with S295, a new immune cultivar, for a greenhouse study in which both cotyledons and true leaves were inoculated. Disease reactions were rated on a scale of 1–10, and genetic models were proposed utilizing generation means analysis. Dominance, when significant, was in the direction of resistance in all but one cross-isolate combination. Digenic interaction components indicated a duplicate type. Narrow-sense heritability for resistance ranged from 0.59 to 0.68; therefore, primarily additive-genetic variability among the selected cutlivars was detected, indicating that breeding for improved resistance to these isolates is a practical goal.Contribution of the Department of Soil and Crop Sciences and the Texas Agricultural Experiment Station  相似文献

The presence of resveratrol (3,5,4′-trihydroxystilbene) and its β-glucoside, piceid (resveratrol-3-β-D-glucopyranoside), together with their isomers in wine appears to be one of the beneficial factors conferring a protective effect against cardiovascular disease through red wine ingestion. A total of 42 red and white wines was collected in arears from Hokkaido to Kyushu in Japan. The wines were fractionated with a C18 Sep-pak cartridge, and the active principles were eluted with ethyl acetate. Crude trans- and cis-piceid were extracted from a Chinese medicine, ‘Kojohkon’ (Polygonum cuspidatum), and their retention times and UV absorption were confirmed by HPLC. trans- and cis-Resveratrol, and trans- and cis-piceid were analyzed in a short C18 HPLC column, and cis-resveratrol was quantified from the amount of cis-isomer converted from authentic trans-resveratrol that had been treated by UV irradiation. The content of piceid is shown as the resveratrol equivalent. The average content of total stilbene compounds was 4.37mg/liter in red wines, while only 0.68 mg/liter in white wines. Red wines made from Pinot noir, Merlot, and Zweigeltrebe grapes all had a high resveratrol content.  相似文献

【目的】转基因作物的大规模种植,可能会对人类健康和生态环境造成影响。因此,商业化种植之前,评价环境安全性十分必要。【方法】以转基因(RRM2)高产棉为实验品种,受体材料中棉所12为对照品种,分别于2013年和2014年连续2年对2种棉田的苗期蚜虫及其几种主要捕食性天敌的田间种群数量进行系统的田间调查,并比较它们在这2种不同棉田间的差异。【结果】与中12相比,转RRM2基因棉苗期无翅蚜的发生数量显著增加,有翅蚜迁入棉田的数量也有所增加,但二者差异不显著;2个棉花品系间棉蚜的几种主要捕食性天敌发生数量也无明显差异。【结论】与亲本材料相比,转高产棉花苗蚜数量显著增加,但其捕食性天敌数量增加不明显。本研究为转RRM2高产基因棉花环境安全评价技术的完善提供了理论依据。  相似文献

Seven stilbenes, a new cis-ε-viniferin and the six known stilbenes, trans-resveratrol, trans-resveratrol-4′-O-β-D-glucopyranoside, trans-ε-viniferin, gnetin H, and suffruticosol A and B, were isolated and identified from seeds of Paeonia lactiflora. The antioxidative activity of these stilbene derivatives was evaluated against the 2-deoxyribose degradation and rat liver microsomal lipid peroxidation induced by the hydroxyl radical generated via a Fenton-type reaction. Among these stilbenes, trans-ε-viniferin and gnetin H significantly inhibited 2-deoxyribose degradation and lipid peroxidation in rat liver microsomes. In addition, cis-ε-viniferin, and suffruticosol A and B also exhibited moderate antioxidative activity. These results suggest that resveratrol dimers and trimers, together with resveratrol from seeds of Paeonia lactiflora may be useful as potential sources of natural antioxidants.  相似文献

【目的】新型转基因棉花在进入大规模商业化应用前,需对其生态环境安全性进行评价;同时,经基因改造的新型转基因抗虫棉花可能影响抗虫棉的次生代谢,进而导致一些综合的生态学效应,致使棉花生理上发生改变,这也是转基因植物安全性评价研究的重要内容。【方法】比较了不同关键时期新型转Cry1Ac+Cry2Ab基因棉花与转Cry1Ac基因棉花和非转基因棉花叶片的鲜重、干重和干鲜比、主要酶[超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)、抗坏血酸过氧化物酶(APX)和谷胱甘肽还原酶(GR)]活性、营养物质(蛋白质、氨态氮、脯氨酸和可溶性糖)和次生代谢产物(棉酚和单宁)含量的差异及其对棉田不同昆虫营养层昆虫个体总数和物种数的影响。【结果】棉花生长的蕾期、花期和花铃期,转Cry1Ac+Cry2Ab基因棉花、转Cry1Ac基因棉花和非转基因棉花叶片的鲜重、干重和干鲜比呈先升高后降低的趋势;SOD和POD活性在花铃期明显升高,CAT、APX和GR活性无显著变化;蛋白质、氨态氮含量无明显变化,脯氨酸和可溶性糖含量均表现为先升高后下降的趋势;棉酚含量在3个时期无显著变化,而单宁含量呈逐渐升高的趋势。3种棉花叶片中干物质积累、主要酶活性、营养物质和次生代谢产物含量均无显著差异;单株大铃数表现为转Cry1Ac+Cry2Ab基因棉花转Cry1Ac基因棉花非转基因棉花,小铃数则表现为转Cry1Ac基因棉花Cry1Ac+Cry2Ab基因棉花非转基因棉花;昆虫群落和害虫亚群落的昆虫个体总数均表现为转Cry1Ac+Cry2Ab基因棉田转Cry1Ac基因棉田非转基因棉田,天敌亚群落昆虫个体总数无显著变化;3种棉田中昆虫群落、害虫亚群落和天敌亚群落的物种数均未发生显著变化。【结论】转Cry1Ac+Cry2Ab基因棉花叶片干物质积累、产量性状、生化物质含量、酶活性在不同生长期表现不同,但上述参数在3种棉花之间无显著差异;且转Cry1Ac+Cry2Ab基因棉花具有较好的抗虫性,能有效降低棉田害虫数量。  相似文献

We examined the effect of Bt-cotton (Event 531) plants expressing the Bacillus thuringiensis δ-endotoxin CryIA(c) on two hymenopteran endoparasitoids, Cotesia marginiventris and Copidosoma floridanum. In the laboratory, parasitized and unparasitized Pseudoplusia includens larvae were reared on foliage from a conventional soybean cultivar (Pioneer 97B61), a conventional cotton cultivar (DPL 5415), or a Bt-cotton cultivar (NuCotn 33B). C. marginiventris developed significantly faster within P. includens larvae feeding on Pioneer 97B61 and DPL 5415 compared to those feeding on NuCotn 33B. C. marginiventris that developed inside P. includens larvae feeding on NuCotn 33B suffered reduced longevity, and females had fewer ova. NuCotn 33B also affected the growth and development of P. includens parasitized with C. floridanum and life history parameters of adult C. floridanum. Parasitized and unparasitized P. includens developed more slowly when they were fed NuCotn 33B and the prepupae weighed less. Survival of parasitized and unparasitized P. includens was lower when larvae were fed NuCotn 33B and some evidence points to higher susceptibility of parasitized caterpillars to intoxication by NuCotn 33B. Fewer C. floridanum adults emerged from hosts fed NuCotn 33B, but pupal weight and adult longevity were unaffected. Analysis comparing the two experiments conducted with C. floridanum suggests that older NuCotn 33B plants (90–120 days after planting) may affect parasitoid development and adult survival less than younger NuCotn 33B plants (60–90 days after planting). Feeding on NuCotn 33B by P. includens affected the survival and development of the two hymenopteran endoparasitoids studied here, and the degree of the effect was similar to that observed with natural resistance found in soybean plants. It remains to be determined if the effects demonstrated here are less than, equal to, or greater than the impact of conventional insecticide applications used in conventional, non-transgenic, cotton.  相似文献

A plant transformation vector, pCLKSCLA25 (EU327498), was developed to contain eight cloning sites and the inducible self-excision system which provided an effective approach to eliminate the selectable marker gene(s) from transgenic plants. Upon induction by salicylic acid, the cre gene produced a recombinase that eliminated sequences encoding the selectable marker neomycin phosphotransferase and cre itself. The excision efficiency was 41% in transgenic tomato regenarants. The stilbene synthase gene (vst1) from Vitis vinifera L. was cloned into pCLKSCLA25. The expression of vst1 gene contributed to the accumulation of trans-reveratrol from 3.4 to 8.7 μg/g fresh wt in different marker-free transgenic tomato lines. Concentrations of trans-resveratrol were analyzed in 36 Korean-grown grape cultivars, including cv. Kyoho. This cultivar has large irregular berries and a deep-black skin and accounts for >15% of all grapevines grown in Korea. The content of trans-resveratrol is an important quality parameter, and its pattern of variability increased gradually until the time of harvest. Its distribution also fluctuated significantly among leaves, seeds, and exocarps, with the highest amount (1,477 μg g−1 dry weight) occurring in the leaves. Among all cultivars evaluated here, trans-resveratrol contents were significantly higher in “Cheongsoo” and “High Bailey,” their levels being 50% above the overall mean (i.e., 2.0-mg g−1 dry weight), whereas 14 cultivars had contents that were 10% less than that mean value  相似文献

广藿香药材以广藿香酮含量较高的酮型广藿香为最优质。而广藿香酮为一种萜类成分,其生物合成途径尚未明确。MVA(甲羟戊酸)途径是萜类化合物生物合成的重要途径。为了分析MVA途经基因表达与化学成分的相关性从而获得促进广藿香酮合成的潜在基因,该文以2种酮型广藿香栽培品种(石牌广藿香、高要广藿香)为材料,通过实时定量PCR分析基因表达和主要成分含量测定,并研究了供试材料不同时期的茎、叶中与甲羟戊酸代谢途径相关的HMGR、MK、MDD基因表达及化学成分。结果表明:(1)HMGR基因在石牌广藿香嫩叶中表达更明显;MK基因在石牌广藿香和高要广藿香中表达模式相似,主要在老茎中表达;MDD基因在石牌广藿香叶中比高要广藿香表达量更高,在两种广藿香的茎中表达模式相似。(2)同属于酮型广藿香,石牌广藿香与高要广藿香的化学成分相似,老叶广藿香醇含量最高,老茎的广藿香酮含量更高。(3)MDD和MK基因与广藿香酮的合成正相关。综上结果所述,酮型广藿香两个栽培种MVA途径的基因表达模式相似,MDD和MK基因可能为酮型广藿香萜类代谢途径的关键基因。  相似文献

The substrate selectivity of several microbial lipases has been examined in the esterification of the conjugated linoleic acid (CLA) isomers cis-9,trans-11-, cis-9,cis-11-, trans-9,trans-11- and trans-10,cis-12-octadecadienoic acid with n-butanol in n-hexane. Lipases from Candida cylindracea and Mucor miehei had a preference for the cis-9,trans-11-octadecadienoic acid, while Chirazyme L-5, a Candida antarctica lipase A, accepted the trans-9,trans-11-fatty acid with a high selectivity. Moreover, lipase from Candida cylindracea and Chirazyme L-5 catalysed the esterification of the cis-9,trans-11-octadecadienoic acid with n-butanol faster than the corresponding reaction of the trans-10,cis-12-fatty acid.  相似文献

In this work, the effect of different inducing factors on trans-resveratrol extracellular production in Monastrell grapevine suspension cultured cells is evaluated. A detailed analysis provides the optimal concentrations of cyclodextrins, methyljasmonate and UV irradiation dosage, optimal cell density, elicitation time and sucrose content in the culture media. The results indicate that trans-resveratrol production decreases as the initial cell density increases for a constant elicitor concentration in Monastrell suspension cultured cells treated with cyclodextrins individually or in combination with methyljasmonate; the decrease observed in cell cultures elicited with cyclodextrins alone is far more drastic than those observed in the combined treatment. trans-Resveratrol extracellular production observed by the joint use of cyclodextrins and methyljasmonate (1,447.8 ± 60.4 μmol trans-resveratrol g−1 dry weight) is lower when these chemical compounds are combined with UV light short exposure (669.9 ± 45.2 μmol trans-resveratrol g−1 dry weight). Likewise, trans-resveratrol production is dependent on levels of sucrose in the elicitation medium with the maximal levels observed with 20 g l−1 sucrose and the joint action of cyclodextrins and 100 μM methyljasmonate. The sucrose concentration did not seem to limit the process although it affects significantly the specific productivity since the lowest sucrose concentration is 10 g l−1, the highest productivity is reached (100.7 ± 5.8 μmol trans-resveratrol g−1 dry weight g−1 sucrose) using cyclodextrins and 25 μM methyljasmonate.  相似文献

The interaction between Botrytis cinerea Pers. and grapevine (Vitis vinifera L.) was studied in a model system of reduced complexity. Cultured plant cells and fragments of fungal cell wall were used to simulate some of the processes taking place upon infection of grapevine with B. cinerea. A soluble glucan elicitor was prepared from the fungal cell wall by acid hydrolysis. Like the insoluble wall preparation, the soluble fragment derived from the cell wall acted upon plant cells in eliciting stilbene formation. In grapevine cells, the interaction with the fungus led to a dramatic shut-off general protein synthesis and to the selective formation of a small set of proteins involved in induced resistance. The proteins synthesized de novo with highest rates were stilbene synthase (StiSy) and l-phenylalanine ammonia-lyase (PAL). Stilbene synthase was purified to apparent homogeneity and its molecular properties were characterized. The enzyme is a homodimer with subunit Mr 43 000 and pl = 5.4. Although there were indications of the presence of isoenzymes, these were not distinguished by charge differences. In size, the grapevine StiSy shows microheterogeneity and differs from the appreciably larger enzyme prepared from peanut. Prior to induction by fungal attack, virtually no stilbenes are formed in the plant cell. Upon induction of the pathway leading to the stilbene resveratrol, StiSy activity determines the ratelimiting step in the metabolic sequence. Laboratory bioassays were conducted to determine host plant effect on pathogenicity of the entomopathogenic fungus Beauveria bassiana (Balsamo) Vuill. (Ascomycota: Hypocreales) to the sweetpotato whitefly, Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae). Fourth instar B. tabaci reared on cucumber, tomato, melon, green pepper, potato, eggplant, marrow, cabbage, bean or cotton, were treated with 1 × 107 conidia/ml B. bassiana EABb 93/14-Tp isolate. Mortality caused by B. bassiana significantly increased with time and it was significantly affected by the host plant on which the nymphs were reared. Mean mortality of nymphs 8 days after inoculation ranged between 52.3±7.3 for nymphs reared on cotton and 91.8±5.8 for nymphs reared on cucumber. Average survival times of nymphs treated with the fungal suspensions were also significantly influenced by the host plant, with a mean of 4.7±0.1 days for nymphs reared on cucumber, 6.6±0.2 days for cotton and 6.9±0.1 days for green pepper. The production of newly formed conidia was also affected by host plant and varied from 111000±8600 conidia/cadaver for nymphs reared on cotton to 597000±28000 conidia/cadaver for those reared on melon.  相似文献
