Surfactant protein-A (SP-A) is the best studied and most abundant of the protein components of lung surfactant and plays an important role in host defense of the lung. It has been shown that ozone-induced oxidation of SP-A protein changes its functional and biochemical properties. In the present study, eight plant polyphenols (three flavonoids, three hydroxycinnamic acids, and two hydroxybenzoic acids) known as strong antioxidants, were tested for their ability to inhibit ozone-induced SP-A oxidation as a mechanism for chemoprevention against lung damage. SP-A isolated from alveolar proteinosis patients was exposed to ozone (1 ppm) for 4 h. The flavonoids protected SP-A from oxidation in a dose dependent manner. ( - )-Epicatechin was the most potent flavonoid and exhibited inhibition of ozone-induced formation of carbonyls by 35% at a concentration as low as 5 μM. Hydroxybenzoic acids inhibited SP-A oxidation in a dose-dependent manner although they were less potent than flavonoids. On the other hand, hydroxycinnamic acids exhibited a different inhibitory pattern. Inhibition was observed only at medium concentrations. The results indicate that inhibition of SP-A oxidation by plant polyphenols may be a mechanism accounting for the protective activity of natural antioxidants against the effects of ozone exposure on lungs.  相似文献

Ozone Quenching Properties of Isoprene and Its Antioxidant Role in Leaves   总被引:24,自引:0,他引:24  
Isoprene is formed in and emitted by plants and the reason for this apparent carbon waste is still unclear. It has been proposed that isoprene stabilizes cell and particularly chloroplast thylakoid membranes. We tested if membrane stabilization or isoprene reactivity with ozone induces protection against acute ozone exposures. The reduction of visible, physiological, anatomical, and ultrastructural (chloroplast) damage shows that clones of plants sensitive to ozone and unable to emit isoprene become resistant to acute and short exposure to ozone if they are fumigated with exogenous isoprene, and that isoprene-emitting plants that are sensitive to ozone do not suffer damage when exposed to ozone. Isoprene-induced ozone resistance is associated with the maintenance of photochemical efficiency and with a low energy dissipation, as indicated by fluorescence quenching. This suggests that isoprene effectively stabilizes thylakoid membranes. However, when isoprene reacts with ozone within the leaves or in a humid atmosphere, it quenches the ozone concentration to levels that are less or non-toxic for plants. Thus, protection from ozone in plants fumigated with isoprene may be due to a direct ozone quenching rather than to an induced resistance at membrane level. Irrespective of the mechanism, isoprene is one of the most effective antioxidants in plants.  相似文献

Oxidative stress has been identified as the root cause of the development and progression of several diseases. Supplementation of exogenous antioxidants or boosting endogenous antioxidant defenses of the body is a promising way of combating the undesirable effects of reactive oxygen species (ROS) induced oxidative damage. Plants have an innate ability to biosynthesize a wide range of non-enzymatic antioxidants capable of attenuating ROS- induced oxidative damage. Several in vitro methods have been used to screen plants for their antioxidant potential, and in most of these assays they revealed potent antioxidant activity. However, prior to confirming their in vivo therapeutic efficacy, plant antioxidants have to pass through several physiopharmacological processes. Consequently, the findings of in vitro and in vivo antioxidant potential assessment studies are not always the same. Nevertheless, the results of in vitro assays have been irrelevantly extrapolated to the therapeutic application of plant antioxidants without undertaking sufficient in vivo studies. Therefore, we have briefly reviewed the physiology and redox biology of both plants and humans to improve our understanding of plant antioxidants as therapeutic entities. The applications and limitations of antioxidant activity measurement assays were also highlighted to identify the precise path to be followed for future research in the area of plant antioxidants.  相似文献

Current elevated concentrations of ozone in the atmosphere, as they are observed during summer seasons, can cause severe effects on plant vegetation. This study was initiated to analyze whether ozone-stressed plants also transfer signals below ground and thereby alter the bacterial community composition in their rhizospheres. Herbaceous plants, native to Germany, with tolerance (Anthoxanthum odoratum, Achillea millefolium, Poa pratensis, Rumex acetosa, and Veronica chamaedrys) and sensitivity (Matricaria chamomilla, Sonchus asper, and Tanacetum vulgare) to ozone, raised in the greenhouse, were exposed in open-top chambers to two different ozone regimes, i.e., "summer stress" and a normal ozone background. DNA of bacterial cells from the rhizospheres was directly extracted, and partial sequences of the 16S rRNA genes were PCR amplified with primers targeting the following phylogenetic groups: Bacteria, alpha-Proteobacteria, Actinobacteria, and Pseudomonas, respectively. The diversity of the amplified products was analyzed by genetic profiling based on single-strand conformation polymorphism (SSCP). Neither the tolerant nor the sensitive plants, the latter with visible above-ground damage, showed ozone-induced differences in any of the SSCP profiles, with the single exception of Actinobacteria-targeted profiles from S. asper. To increase the stress, S. asper was germinated and raised in the continuous presence of an elevated level of ozone. SSCP profiles with Bacteria-specific primers combined with gene probe hybridizations indicated an ozone-related increase in a Xanthomonas-related 16S rRNA gene and a decrease in the respective gene from the plant plastids. The fact that only this latter unrealistic scenario caused a detectable effect demonstrated that ozone stress has a surprisingly small effect on the structural diversity of the bacterial community in rhizospheres.  相似文献

Crop yields may be threatened by increases in UV-B radiation resulting from depletion of the ozone layer. In higher plants, the presence of flavonols provides a protective mechanism, and we report a novel staining procedure for the visualization of such protectants in plant tissue. It is shown that the proposed technique provides sensitive and specific fluorescence of flavonoids in chlorophyll-bleached tissue of Arabidopsis thaliana.  相似文献

The objective of this study was to determine whether acutely exposing rats to ozone would result in the loss of antioxidants from plasma and bronchoalveolar lavage fluid (BALF). Additional goals were to compare analyses of the same antioxidant concentration between different laboratories, to investigate which methods have the sensitivity to detect decreased levels of antioxidants, and to identify a reliable measure of oxidative stress in ozone-exposed rats. Male Fisher rats were exposed to either 2.0 or 5.0 ppm ozone inhalation for 2 h. Blood plasma and BALF samples were collected 2, 7, and 16 h after the exposure. It was found that ascorbic acid in plasma collected from rats after the higher dose of ozone was lower at 2 h, but not later. BALF concentrations of ascorbic acid were decreased at both 2 and 7 h postexposure. Tocopherols (α, δ, γ), 5-nitro-γ-tocopherol, tocol, glutathione (GSH/GSSG), and cysteine (Cys/CySS) were not decreased, regardless of the dose or postexposure time point used for sample collection. Uric acid was significantly increased by the low dose at 2 h and the high dose at the 7 h point, probably because of the accumulation of blood plasma in the lung from ozone-increased alveolar capillary permeability. We conclude that measurements of antioxidants in plasma are not sensitive biomarkers for oxidative damage induced by ozone and are not a useful choice for the assessment of oxidative damage by ozone in vivo.  相似文献

Elevated levels of tropospheric ozone and their effects on plants have been studied for a great number of years. Ozone is a gaseous pollutant and acts as a phytotoxin. Even though ozone is known to change the physiology of plants, little attention has been given to the indirect effects of ozone on plant-insect interactions. This paper addresses this question by investigating the interactive effects of ozone and plant genotype on insects. Lines of rapid-cycling Brassica rapa (L.) selected for their contrasting sensitivity to ozone and Pieris brassicae (L.) (Lepidoptera: Pieridae) were used as a model system. The effect of differences in ozone sensitivity and ozone fumigation on the plant's carbon and nitrogen pools, the feeding preference, and behaviour of P. brassicae larvae were investigated. The results show that the plant's susceptibility to ozone interacts in a complex way with ozone induced alterations in the suitability of the plant for the insect. Only the larval performance on the sensitive line was affected by ozone exposure. Biochemical changes in the resistant B. rapa line made the plant a better food source for the insects, since the digestibility of this plant was significantly higher than that of the sensitive line, and the larvae pupated more quickly and were heavier.  相似文献

Ozonation of either human whole blood or saline-washed erythrocytes causes considerable damage to the latter and this result has opened a controversy. With the benefit of hindsight, it appears logical that once erythrocytes are deprived of the potent antioxidants of plasma, they become very sensitive to the oxidant effects of ozone. The aim of the present work was to perform a physical–chemical evaluation of some critical parameters able to clarify this issue. We have ascertained that when whole blood is exposed to the appropriate ozone doses used in human therapy, no damage ensues while saline-washed erythrocytes undergo conspicuous haemolysis. The dogma that ozone is always toxic is incorrect because its reactivity below the concentration of 80 μg/mL can be controlled by the plasmatic antioxidant system.  相似文献

The influence of long-term chronic ozone exposure on carbon fluxes from young beech trees (Fagus sylvatica L.) into the phospholipid fraction of microbial communities (PLFA) in the rhizosphere and into the dissolved organic carbon (DOC) fraction was studied in a lysimeter experiment using 13C depleted CO2 over one vegetation period to identify possible changes in below ground carbon translocation processes due to the plant stress. It could be shown that microbial biomass as well as individual microbial communities and their activity pattern in the rhizosphere of young beech trees are mainly driven by the vegetation period. An increase in total microbial biomass as well as individual microbial communities was detected during the vegetation period from June to September. However, also a clear ozone effect was visible mainly at the end of the vegetation period. Enzyme activities and PLFA data indicated earlier induced plant senescence as a response to the elevated ozone treatment. Furthermore higher microbial biomass and abundance of plant C utilizing microbes was observed in elevated ozone treatments over the whole vegetation period.  相似文献

Many reactive oxygen species such as ozone, singlet oxygen, hydroxyl radical, and organic oxyradicals have been implicated in damage to plant organs and biopolymers such as chloroplasts, cell membranes, proteins, and DNA. The principal defenses against these reactive molecules and free radicals in plants include detoxifying enzymes (catalase, superoxide dismutase, etc.) and also lower molecular weight secondary products with antioxidant activity. These latter compounds include a great variety of phenolic compounds, carotenoids, nitrogenous, and sulfur-containing materials. Some of the more important mechanisms of action of the secondary compounds will be discussed, with emphasis on the use of structural and kinetic data to identify the most effective antioxidants against peroxy radical-induced damage, which is perhaps the most important of the oxidative stresses present in the usual environment of plants. © 1995 Wiley-Liss, Inc.  相似文献

Continuous depletion of the stratospheric ozone layer has resulted in an increase in ultraviolet-B (UV-B; 280-315 nm) radiation on the earth's surface which inhibits photochemical and photobiological processes. However, certain photosynthetic organisms have evolved mechanisms to counteract the toxicity of ultraviolet or high photosynthetically active radiation by synthesizing the UV-absorbing/screening compounds, such as mycosporine-like amino acids (MAAs) and scytonemin besides the repair of UV-induced damage of DNA and accumulation of carotenoids and detoxifying enzymes or radical quenchers and antioxidants. Chemical structure of various MAAs, their possible biochemical routes of synthesis and role as photoprotective compounds in various organisms are discussed.  相似文献

Summary The method of mapping the vegetation on scale 1: 200,000 and the starting points in relation to the potential natural vegetation and ecotopes, are discussed.In view of the planological background of this study, some restrictions have been added to the concept of potential natural vegetation, concerning the period of development and the human influence.The relationship between soil, ground water and vegetation was studied, which resulted in the map of the potential natural vegetation.Each type of potential natural vegetation stands for a series of vegetation types on the same site. Seven main series, with a number of sub-series are distinguished. Within each vegetation series the plant communities have been spread over five groups, according to their structure and naturalness.Ecotopes and ecotope complexes are considered as landscape ecological units. A list of ecotopes was obtained by interpreting topographical maps and by inventory data.The actual vegetation was mapped by estimating the size of the ecotopes within the separate areas. Carotenoids are plant pigments, some of which act as a vital source of vitamin A to all animals, that appear to have additional benefits to primates. They are potent antioxidants and photoprotectants and can additionally modulate gene activity resulting in protection from experimentally-induced inflammatory damage and neoplastic transformation. Anti-neoplastic properties appear tightly correlated to their ability to induce the gap junctional protein connexin 43 (Cx43). This when upregulated leads to decreased proliferation and decreased indices of neoplasia in animal and human cells. Delivery of natural carotenoids can be compromised by poor bioavailability. To overcome this, a synthetic water-dispersible derivative of astaxanthin has been synthesized and shown to be: highly bioavailable; a potent antioxidant; protective against experimental ischemia-reperfusion injury and capable of inducing Cx43, suggesting antineoplastic potential. The ability to deliver biologically active carotenoids at high concentration and with good reproducibility appears to have been achieved.  相似文献

Plant strategies to survive ozone stress include exclusion or tolerance of ozone. If these processes fail, past observations of ozone injury have indicated many physiological and metabolic changes then occur; most of these changes are likely to have been initiated at the level of gene expression, suggesting signal transduction. In the last decade considerable understanding of the biochemical process within plants has been developed. Currently there are several hypotheses regarding a response of plants to ozone fumigation: [1] membrane dysfunction and alteration of purpose; [2] stress ethylene interactions; [3] impairment of photosynthesis via changes in Rubisco levels and the guard cells so that the stomata do not track correctly the environment; [4] antioxidant protection through metabolites and enzyme systems to reduce the oxidant load; and [5] general impairment or disruption of metabolic pathways. Many believe that free radicals and other oxidative products, formed in plant leaves under ozone exposure, are responsible for much of the spread of the biochemical alterations. There are obvious chemicals that may account for the changes that are observed, such as hydrogen peroxide. Once the ozone enters the tissue, evidence suggests the first line of defense is a range of antioxidants, such as ascorbate, glutathione peroxidase, superoxide dismutase, and catalase. If overwhelmed, subsequent events occur which are highly suggestive of systemic acquired resistance. Furthermore, other defensive indicators, such as salicylic acid and jasmonic acid, tend to increase, but more slowly than ethylene, and spread their signaling effects more widely in the plant. The primary set of metabolic reactions that ozone triggers is thought to be "wounding" responses with a secondary response of senescence. The dramatic strides in understanding the genetic make-up of plants, gene control, and signal transduction/control over the last few years will only accelerate in the future. We need now to have an understanding of those events that can be translated into more detailed schemes of how ozone alters much of the basic metabolism of plants and how plants counteract or cope with ozone. What is now known about how varied biochemicals and their pathways are changed upon ozone exposure will be discussed.  相似文献

Coastal vegetation has been widely recognized as a natural method to reduce the energy of tsunami waves. However, a vegetation barrier cannot completely stop a tsunami, and its effectiveness depends on the magnitude of the tsunami as well as the structure of the vegetation. For coastal rehabilitation, optimal planning of natural coastal systems, and their maintenance, we need to quantitatively elucidate the capacity of vegetation to reduce the energy of tsunami waves. The limitations of coastal forests in relation to the magnitude of a tsunami and the maintenance of forests as natural disaster buffer zones have to be understood correctly for effective coastal vegetation planning. Demerits of coastal forests have also been revealed: for example, an open gap in a forest (i.e., a road, river, difference in elevation, etc.) can channel and amplify a strong current by forcing it into the gap. Floating debris from broken trees also can damage surrounding buildings and hurt people. However, many studies have revealed that these demerits can be overcome with proper planning and management of mangroves and coastal forests, and that coastal vegetation has a significant potential to mitigate damage in constructed areas and save human lives by acting as buffer zones during extreme natural events. However, mangrove forests have been damaged by anthropogenic activities (i.e., tourism, shrimp farming, and industrial development), making coastal areas increasingly vulnerable to tsunamis and other natural disasters. The effectiveness of vegetation also changes with the age and structure of the forest. This highlights the fact that proper planning and management of vegetation are required to maintain the tsunami buffering function of coastal forests. Although many government and nongovernmental organizations have implemented coastal vegetation projects, many of them have been unsuccessful due to a lack of proper maintenance. A pilot project in Matara City, Sri Lanka, revealed that participation and support from local authorities and communities is essential to make the planting projects successful. An integrated coastal vegetation management system that includes utilization of the materials produced by the forest and a community participation and awareness program are proposed to achieve a sustainable and long-lasting vegetation bioshield.  相似文献

In this review all recent field studies on the effects of UV-B radiation on bryophytes are discussed. In most of the studies fluorescent UV-B tubes are used to expose the vegetation to enhanced levels of UV-B radiation to simulate stratospheric ozone depletion. Other studies use screens to filter the UV-B part of the solar spectrum, thereby comparing ambient levels of UV-B with reduced UV-B levels, or analyse effects of natural variations in UV-B arising from stratospheric ozone depletion. Nearly all studies show that mosses are well adapted to ambient levels of UV-B radiation since UV-B hardly affects growth parameters. In contrast with outdoor studies on higher plants, soluble UV-B absorbing compounds in bryophytes are typically not induced by enhanced levels of UV-B radiation. A few studies have demonstrated that UV-B radiation can influence plant morphology, photosynthetic capacity, photosynthetic pigments or levels of DNA damage. However, there is only a limited number of outdoor studies presented in the literature. More additional, especially long-term, experiments are needed to provide better data for statistical meta-analyses. A mini UV-B supplementation system is described, especially designed to study effects of UV-B radiation at remote field locations under harsh conditions, and which is therefore suited to perform long-term studies in the Arctic or Antarctic. The first results are presented from a long-term UV-B supplementation experiment at Signy Island in the Maritime Antarctic.  相似文献

Activated oxygen or oxygen free radical mediated damage to plants has been established or implicated in many plant stress situations. The extent of activated oxygen damage to potato (Solanum tuberosum L.) tubers during low temperature storage and long-term storage is not known. Quantitation of oxygen free radical mediated damage in plant tissues is difficult. However, it is comparatively easy to quantitate endogenous antioxidants, which detoxify potentially damaging forms of activated oxygen. Three tuber antioxidants, superoxide dismutase, catalase, and α-tocopherol were assayed from four potato cultivars stored at 3°C and 9°C for 40 weeks. Tubers stored at 3°C demonstrated increased superoxide dismutase activities (up to 72%) compared to tubers stored at 9°C. Time dependent increases in the levels of superoxide dismutase, catalase, and α-tocopherol occurred during the course of the 40 week storage. The possible relationship between these increases in antioxidants and the rate of activated oxygen production in the tubers is discussed.  相似文献
