The fine structure of the kidney is studied in the sexually mature male three-spined stickleback after administration of an antiandrogen, cyproterone acetate. Under these conditions, the dedifferentiation of renal tubules is characterized by the same involutive processes as those induced by castration, with the difference that cyproterone acetate only begins to act after 14 days whereas after castration the first signs of involution are visible after 7 days. The ultrastructural modifications affect the nucleoli, the rough endoplasmic reticulum and the Golgi apparatus. They reflect an inhibition of the secretory process. The results obtained demonstrate that administration of cyproterone acetate to male sticklebacks has an inhibitory effect on renal target cells, apparently indistinguishable from the changes induced by lack of male sex hormone, and that this drug may be a valid substitute for castration in fish.  相似文献

Epithelial cells from involuting rat ventral prostate (VP) express Matrilysin (MMP-7) mRNA. Herein, we investigated by immunohistochemistry the MMP-7 protein location and its association with tissue changes following castration in the VP. Normal and castrated adult male Wistar rats were sacrificed at different times after surgery. VP was examined by immunocytochemistry and immunoprecipitation. Castration promoted a shrinking of prostate ducts with an extensive stromal remodeling. In the VP from normal rats, MMP-7 immunoreactivity was found in epithelial secretory granules. Three days after castration, immunostaining for MMP-7 was found in both the epithelial secretory granules and in the stroma just below the epithelium, mainly at the distal ductal tips. At seven and 21 days after castration, the immunostaining for MMP-7 was found only in the stromal space. Immunoprecipitation confirmed the specificity of the primary antibody by rescuing a pro-enzyme form (28kDa) in the prostate extracts. The present results suggest that MMP-7 participates in the epithelial-stromal interface remodeling of the ventral prostate during the involution achieved by castration, probably in the degradation of components of the epithelial basement membrane.  相似文献

Summary Glandular cells in the prostate of the intact, adult dog contain numerous, large secretory granules that are released by exocytosis. Following hypophysectomy or castration, the glandular epithelium atrophies and the secretory granules degenerate and eventually disappear. Pharmacologic doses of estradiol-17 17-cyclopentylpropionate cause the regressed glandular cells to synthesize a new population of smaller granules that are also released by exocytosis, even though estrogen is known to inhibit fluid secretion by the canine prostate. Thus, the mechanisms involved in prostatic synthesis and exocytosis of secretory granules are independent of those regulating fluid secretion and are operative in the absence of androgen or pituitary hormones.  相似文献

Using electron microscopic techniques the prostate glands of male Beagle dogs were studied 3 days after castration. At this time marked differences in the extent of alterations of the glandular epithelium were observed: Whereas several acini showed only minor changes with reduction of epithelial height and diminution of secretory granules, many acini were severely affected with pronounced alteration of cellular structure and accumulation of large lipid droplets. A constant feature was the stimulation of the basal cells of the grandular epithelium. Additionally, in some areas of the gland aggregations of stimulated basal cells forming an acinus-like structure with a slit-like lumen were found. Our study shows that castration leads to marked alterations of prostatic epithelium within a short time. Androgen deprivation causes regressive changes of secretory epithelial cells, but clearly stimulates the basal cell population.  相似文献

The effects of thyroidectomy, adrenalectomy, and castration on the pars distalis of male Japanese quail, and of injection of LH-RH on sexually inactive females, were investigated by light and electron microscopy. Correlation between light and electron microscopy was attained by use of alternate thin and thick sections. Six types of secretory cells were identified and the ultrastructural characteristics described. Putative endocrine functions have been designated on the basis of responses to experimental interventions and on other criteria. The putative STH cells are characterized by the presence of large dense secretory granules (250-300 nm) that are stained with orange-G by the trichrome method. They occur only in the caudal lobe and appear to be unchanged by castration, thyroidectomy, adrenalectomy and LH-RH injection. The putative prolactin cells are characterized by large (400-600 nm), spherical or polmorphic, dense secretory granules stainable with acid fuchsin and aniline blue; prominent Golgi apparatus and well developed endoplasmic reticulum with densely packed, regularly parallel lamellae. They are found mainly in the cephalic lobe. The prolactin cells develop some vacuolization after adrenalectomy and undergo some degeneration after castration. The ACTH cells, which are restricted to the cephalic lobe, are identified by the dense, spherical granules (250-300 nm) that are stained with acid fuchsin. After adrenalectomy, they lose their secretory granules and are transformed into large, chromophobic adrenalectomy cells. TSH cells are so designated by their response to thyroidectomy including loss of their fine secretory granules and transformation to large, vacuolated thyroidectomy cells. We have found TSH cells and thyroidectomy cells only in the cephalic lobe. Basophilic cells, considered to be gonadotropes, occur in both the cephalic and caudal lobes. The gonadotropes of the cephalic lobe appear to have slightly larger (120-200 nm) granules than the caudal lobe (120-150 nm). However, after castration, the gonadotropes in both lobes become hypertrophied and vacuolated and are transformed into mutually indistinguishable castration cells. Twenty minutes after injection with LH-RH, the gonadotropes of both lobes increase in size and number, degranulate, develop vacuoles in the cytoplasm, and appear very similar to castration cells.  相似文献

The light-microscopical and ultrastructural differentiation of the disseminate prostate was studied in 12 boars and 8 barrows, ranging in age from 8 to 28 and 12 to 27 weeks, respectively. The barrows had been castrated at the age of 8 weeks. In young boars the prostate is organized in an inner zone and an outer zone. The glandular tubules of the latter are more differentiated than those in the inner zone. Sexual maturation in the prostate is characterized by almost complete reduction of the inner zone and concomitant enlargement of the outer zone. This differentiation of the inner zone occurs progressively and is manifested by a higher density of the glandular tubules, by a decrease in the number of basal cells and by an increase in the secretory tubular cells containing more microvilli, organelles and secretory vesicles. A few weeks after castration, the characteristic morphology of the undifferentiated inner zone is found throughout the entire glandular layer. The number of glandular tubules in barrows has decreased, and their cells show minimal secretory activity, nuclear indentations and a decrease in their organelles. A further castration effect is vacuolization of the perinuclear cytoplasm in numerous glandular cells.  相似文献

Summary The immunocytochemical reactivity of the glandular cells of the corpus cardiacum (CCG-cells) of Locusta migratoria and Schistocerca gregaria was investigated at the electron-microscopic level, using the protein A-gold method, with three antisera against fragments of the adipokinetic hormones AKH I and AKH II. This combination of antisera permitted discrimination between anti-AKH I and anti-AKH II immunoreactivity. Fixation in a mixture of 2% glutaraldehyde and 2% formaldehyde, in combination with low-temperature embedding in Lowicryl K4M, produced the highest and most consistent selective immunogold labelling of the secretory and ergastoplasmic granules. All secretory granules in all CCG-cells investigated possessed a distinct anti-AKH I-immunopositive reaction, whereas most secretory granules showed a weaker anti-AKH II immunoreaction. Ergastoplasmic granules reacted similar to the secretory granules. The average immunolabelling of the secretory granules was higher in the processes than in the cell bodies of the CCG-cells. The results in Schistocerca gregaria were essentially similar to those in Locusta migratoria. It is concluded that (i) the individual CCG-cells synthesize AKH I as well as AKH II; (ii) these hormones coexist in the same ergastoplasmic and secretory granules; and (iii) these granules contain a higher content of AKH I than AKH II.  相似文献

The endocrine regulation of the mucosa of the oviductal pars convoluta was analyzed by ultrastructural studies demonstrating that ovariectomy, together with a decrease in ovarian steroids circulating levels, caused a marked regression in this portion of Bufo arenarum oviduct. Twenty-five days after ovariectomy, a decrease in the depth of the epithelial and glandular layers was observed due to the notable loss of secretory cells, whose number was clearly smaller than in nonovariectomized females. The remaining secretory cells showed involution signs, with few secretory granules in their cytoplasm, little endoplasmic reticulum near poorly developed Golgi complexes and a large amount of lipid droplets. Cells in an advanced autolysis state were found in the lumen. These characteristics evidence a nonfunctional state of the pars convoluta. Treatment with 5alpha-dihydrotestosterone (DHT) completely reversed the ovariectomy effect, inducing pars convoluta growths and restoring the characteristics of epithelial and glandular secretory cells in the whole pars convoluta, with micrographs similar to the control. These same effects were observed after treatment with estradiol-17beta (E2), progesterone (P) o E(2)+P in the glandular layer of the whole pars convoluta, but only in the epithelial layer of the most anterior region of this duct. In the secretory cells of other segments these treatments induced the formation of granules of high electron density and homogeneous aspect. Each steroid had a particular effect on the pars convoluta. Although E2 and DHT induced the development of the organoids involved in the proteins biosynthesis, P and DHT acted as secretagogues.  相似文献

The ultrastructural differentiation of the secretory cells and the nature of secretory proteins in the male accessory gland of Drosophila funebris have been studied by electron-microscopic and immunological methods. (1) In the pupae at 112 days before eclosion, secretory products can be detected in the lumen, even though most glandular cells are at the initial phase of differentiation. At the time of eclosion both main and secondary cells are fully differentiated, but the whole set of five immunologically active proteins are detectable only on the second to third day of adult life. (2) The secondary cells contain giant protein granules, the so-called filamentous bodies, which become partially fused and the filaments assume a twisted form. Randomly dispersed filaments and closely packed filament bundles are also visible in the gland lumen. Antigenic labelling of ultrathin sections and immunoreplica electrophoresis yielded no evidence for the microtubular nature of these filaments. The secretion stored in the lumen contains in addition a large quantity of flocculent proteins which have their origin in the main cells. (3) During the period of high secretory activity in the 7-day-old male flies no vacuolization and disintegration of either the main or secondary cells have been observed. We conclude that both types of cells have the merocrine secretory mechanism. (4) Ultrastructural alterations in the glandular cells confirmed our previous observation that copulation stimulates RNA and protein synthesis.  相似文献

The ultrastructure of male reproductive accessory glands was investigated in the scorpionfly Sinopanorpa tincta (Navás, 1931) (Mecoptera: Panorpidae) using light and transmission electron microscopy. The male accessory glands comprise one pair of mesodermal glands (mesadenia) and six pairs of ectodermal glands (ectadenia). The former opens into the vasa deferentia and the latter into the ejaculatory sac. The mesadenia consist of a mono-layered elongated columnar epithelium, the cells of which are highly microvillated and extrude secretory granules by means of merocrine mechanisms. The epithelium of ectadenia consists of two types of cells: the large secretory cells and the thin duct-forming cells. These two types of cells that join with a cuticular duct constitute a functional glandular unit, corresponding to the class III glandular cell type of Noirot and Quennedey. The cuticular duct consists of a receiving canal and a conducting canal. The secretory granules were taken up by the receiving canal and then plunged into the lumen through the conducting canal.  相似文献

A battery of horseradish peroxidase-conjugated lectins has been employed as a cytochemical tool for the labelling of specific cell types in amphibian epidermis. Among the lectins used, onlyUlex europaeus I (UEA I) showed specific reaction with the cytoplasm of flask cells. In addition, UEA I stained flask-shaped secretory cells in dermal glands and a reaction on glandular ductal cells was also observed. At the electron microscopic level, lect-in binding was found in granules distributed among mitochondria in the cytoplasm of flask cells and in larger mucous granules of flask-shaped glandular cells, which were released into the lumen. UEA I also stained the extracellular space above flask cells. The labelling was due mainlty to a glycoprotein of mol. wt. approx. 27 kDa. Structural and cytochemical similarities between flask cells and flask-shaped cells of dermal glands could be a consequence of a common secretory role of both cell types.  相似文献

Summary The histochemical (iron, lipopigments, acid phosphatase, leucine aminopeptidase) and cytologic (lysosomes) changes occuring during pregnancy, lactation and involution of mouse, rat, rabbit, guinea-pig mammary glands are studied by light microscopy and electron microscopy.In all the animals examined, the mammary epithelium has an intracellular digestive system which is adapted to subserve two functions. The first one is the segregation of cytoplasmic components which often precedes cellular involution. The second one is the regulation of secretory processes in the non lactating glands. This digestion of endogenous materials results in the formation of various lytic bodies: dense bodies sometimes containing ferritin, vacuolated dense bodies with membranous residues, autophagic vacuoles. The lysosomes can give large complex dense bodies like lipofuscin pigments with or without ferritin.Leucine aminopeptidase which always disappears in the mouse mammary epithelium during lactation is not present in rat, rabbit, guinea-pig mammary epithelium. In these species only the vascular tissue contains the enzyme. This observation indicates that leucine aminopeptidase does not take care of the overproduction of secretory products in the non-lactating glands.Acid phosphatase is concentrated in secretory granules and in lytic bodies: multivesicular bodies, dense bodies with ferritin, vacuolated dense bodies, lipopigments. This enzyme constitutes probably a mechanism for controlling and triggering the destruction of the secretory material with no active elimination.The iron of the mammary epithelium appears in virgin mice older than 30 weeks and in mice, rats, rabbits, guinea-pigs during glandular cells involution. This is a catabolic iron located in lysosomes. Its amount depends upon the iron content of the milk and upon the competitive secretory and catabolic activities of the glandular cells. An explanation of iron disappearance during a second pregnancy and lactation is discussed.  相似文献

The accessory glands of the male reproductive system of Campodea remyi (Apterygota : Diplura : Campodeidea) consist of similar glandular units, each of which is made of a long secretory cell with a sieve at the tip of its extracellular cavity and a cell forming the excretory canalicule. During the annual cycle of sexual activity, they show morphological differences in terms of their size and the number of secretory granules. When ecdysis occurs, the cuticle of the ejaculatory duct and canalicules, the sieves, and the apical part of the glandular cells disappear. In Bufo arenarum the oviduct exhibits conspicuous changes throughout the sexual cycle. In the present study, we analyzed the optical and ultrastructural characteristics of the oviductal pars convoluta mucosa, the portion responsible for jelly secretion, during both the preovulatory and postovulatory periods. Secretory epithelial cells, ciliated cells, basal cells, and glandular cells are described. Secretory epithelial cells are characterized by the presence of secretory granules, the size, shape and electron density of which vary markedly. Their contents are mainly released by exocytosis into the oviductal lumen. Moreover, in the preovulatory period, apocrine, and holocrine secretion processes frequently occur. During the postovulatory period, these cells exhibit a marked diminution of secretory granules. Ciliated cells show a typical ultrastructural organization. Basal cells are distinguished in the lower part of the epithelium by their heterochromatic nuclei and electron‐lucent cytoplasm. These cells, to the best of our knowledge, are reported for the first time in Amphibia. Glandular cells exhibit oval, round, or polyhedric granules, most of them with one or more cores. Our results indicate that the contents of epithelial and glandular secretory cells are partially secreted during the preovulatory period. The localization of aminopeptidase M (APM), dipeptidyl peptidase I (DAP I), II (DAP II) and IV (DAP IV) in the renal section was investigated histochemically, and their activities were determined fluorometrically in renal homogenate of normal, castrated and testosteron treated male rats.--After castration the activities of the lysosomal DAP II (pars convoluta of the proximal tubule), DAP I (distal and proximal tubule) and of the mainly membrane-bound DAP IV (glomeruli, brush border of the proximal tubule) increase in comparison to normal males, whereas the activities of the brush border-bound APM decrease. After testosteron treatment of castrated animals (0.1, 0.5 and 1.0 mg testosterone proprionate/100 g BW and day; 5-day treatment) the activities of DAP I, II and IV decrease again, so that after treatment with 0.1 mg testosterone proprionate, the activities of DAP I and II approach those in normal males.--The additionally determined urinary protein excretion shows that there is a significant decrease in proteinuria after castration, whereas testosterone treatment of castrated animals is accompanied by an increase of proteinuria.--Our results would suggest that the protein catabolism in the proximal tubule and the proteinuria are interrelated, and that testosterone influences (decreases) the protein catabolism in the proximal tubule. This means that high activities of lysosomal proteinases in the proximal tubule (castrates) are accompanied by a low proteinuria, and low activities of those proteinases (testosterone treated castrated or normal males) by a high proteinuria.  相似文献

(1) alpha-amylase was extracted and purified from the stomach/digestive gland complex of the scallop Pecten maximus and an anti-serum was induced against the purified amylase by rabbit immunization. (2) The anti scallop amylase was used to localize the amylase-secreting cells in the stomach of Pecten maximus by immunofluorescence and immunogold labelling. The amylase-secreting cells are glandular cells particularly numerous in the main sorting area of the stomach. Their secretory granules were found strongly positive for anti-amylase. Three types of glandular cells were observed, actually corresponding to the three stages of the glandular-cell activity, synthesis, secretion and excretion. (3) The synthesizing cell shows the characteristic features of a protein-synthesizing cell: a conspicuous nucleolus and abundant granular endoplasmic reticulum. In the secretory cell, the secretory granules are formed by the Golgi apparatus and accumulate in the apical part of the cell. The secretory cell is filled with two types of secretory granules which are released in the stomach lumen by apocrine excretion. (4) The present study brings the first demonstration of the synthesis and extracellular release of amylase by glandular cells of the stomach epithelium of a bivalve.  相似文献

The normal ventral and dorsal prostatic lobes of the young adult Syrian hamster were examined at the light and electron microscopic levels. Each lobe is composed of branched tubular secretory units separated from each other by loose interacinar connective tissue and draining into the urethra. The lumen of each acinus is lined by a simple epithelium composed of columnar secretory cells with occasional small basal cells. The epithelial layer, with the thin underlying lamina propria, forms a mucosa that is often highly folded. The whole acinus is bounded by a thick muscular stroma. In each of the ventral lobes, there are three main ducts, each one formed of tubular branched tributary secretory units. The walls of the secretory acini are moderately folded. Microvilli dominate the lumenal surface of the secretory epithelial cells. The Golgi complex is very extensive and shows dilated cisternae and secretory vesicles and vacuoles of various sizes. Membrane-bounded secretory granules populate the Golgi and apical areas and are released into the acinar lumen by exocytosis. The rough endoplasmic reticulum is dispersed throughout the cytoplasm, except in the region of the Golgi apparatus. In each of the dorsal lobes, there are several main tubular ducts that open into the urethra. Both proximal (ductal) and distal portions of the glandular tree are secretory in nature. Microvilli and cytoplasmic bulges and blebs dominate the lumenal surface of the secretory cells. The cells are also characterized by highly dilated cisternae of rough endoplasmic reticulum. The secretory cells show heterogeneity in the degree of dilation and distribution of rough endoplasmic reticulum, and this heterogeneity may reflect location in the glandular tree.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The aim of the present work was to study the sexual differences in secretory mechanisms and intracellular calcium ion dynamics in the Harderian gland of the golden hamster. In both sexes the Harderian gland consisted of small and large lobes. In the intact control male glands the secretory portions of both lobes showed wide lumina that contained secretory material and cytoplasmic fragments, suggestive of the occurrence of exocytosis and apocrine secretion. After perfusion with HEPES-buffered Ringer's solution containing 10 microM carbamylcholine (CCh), the glandular cells showed features of enhanced secretion and a rise in intracellular calcium concentration ([Ca2+]i). In the intact control female gland the lumina of most secretory portions in the large lobe contained porphyrin accretions, and exocytosis was the sole secretory mechanism. Stimulation of the large lobe with 10 microM CCh did not raise [Ca2+]i or cause enhanced secretion. The small lobe in females resembled the male gland in secretory functions, and CCh administration caused enhanced secretion and a rise in [Ca2+]i. Castration in males abolished apocrine secretion; exocytosis became the sole secretory mechanism, and stimulation of the glandular cells with CCh did not cause enhanced secretion or induce a rise in [Ca2+]i. To the contrary, in females, castration restored apocrine secretion and CCh administration caused enhanced secretion and a rise in [Ca2+]i. Castration did not affect the secretory mechanisms and the effect of CCh on the glandular cells in the small lobes of both male and female glands. The present study points to the possibility that sex hormones may control the functioning or expression of muscarinic receptors in the Harderian gland of the golden hamster.  相似文献

Kidney cells of the marine stickleback Spinachia have been studied with histochemical methods for the demonstration of glycoconjugates. The fine structure of epithelial cells is described. Mucus threads in the nephronic tubule of sexually mature males consist of neutral glycoprotein which corresponds with the secretory granules in proximal tubule segment II cells. Large lysosome-like inclusions, which also react with PAS, are present in many P II cells. All cells of the collecting duct epithelium differentiate into mucous cells in male Spinachia. The nature of their secretory products, which are well preserved by freeze-drying, is discussed. Sialylated glycoprotein is present in mucus granules and sulphated glycoprotein can be demonstrated at the apex of collecting duct cells. Collecting duct cell mucus can be digested with testicular hyaluronidase indicating that proteoglycans may be involved in the structure of macromolecules. The observations are compared with studies of mucus production in the urinary apparatus of several other vertebrates.  相似文献
