We investigated variation in shell size and shape of marinesnail species of the Littorina saxatilis complex (L. saxatilis,L. compressa and L. arcana) using geometric morphometric methods.These morphologically similar periwinkle species that are commonin the European intertidal have presented many problems fordiagnosis based on morphology alone. A discriminant analysisdemonstrated that geometric morphometrics is very efficientfor diagnosing individuals to species among sympatric populations.We successfully diagnosed an average of 96% of the specimens(with 85.7–100% correct diagnosis for specific comparisons).The diagnosis capability of this method is absolute at the populationlevel. This makes the technique potentially useful for the designof manipulative field or laboratory experiments. Moreover, ageometric-morphometric analysis was also accomplished in twosnail ecotypes (H and M) of L. saxatilis from rocky shores ofNE England which are apparently adapted to different degreesof wave exposure. We found that the H (exposed) ecotype hasa relatively rounded shell shape with a bigger aperture, whereasthe M (protected) ecotype has a smaller aperture. Eight species of the genus Littorina were hitherto recognised in the north-western region of the Pacific Ocean: L. sitkana, L. brevicula, L. mandshurica, L. squalida, L. aleutica, L. naticoides, L. kasatka and L. subrotundata. Using allozyme electrophoresis it has been demonstrated that, in the Kurile Islands, three of these species (L. sitkana, L. subrotundata and L. kasatka) co-occur, together with a fourth, still undescribed species (L. sp.). These four species were compared at 16 loci coding for 13 enzymes. All species were easily distinguished by diagnostic enzyme markers. The mean genetic distances and ranges between species pairs are: L. sitkana and L. sp. D = 0.622 (0.561–0.741), L. sitkana and L. subrotundata D=0.981 (0.821–1.110), L. subrotundata and L. sp. D=0.975 (0.955–0.995). The genetic distance between L. kasatka and each of the other three species was greater than 1 (range 1.123–2.087). These data suggest that L. sitkana, L. subrotundata and L. sp could be members of a species complex; according to current classifications these three belong to the subgenus Neritrema. However, the genetic distance between L. kasatka and L. sitkana is much greater than between L. sitkana and other Neritrema species, and thus supports the classification of L. kasatka in the subgenus Littorina.  相似文献

As adults Littorina littorea and L. saxatilis have overlapping distributions, both on the shore and on a geographical scale, this implies that they are affected by similar selective forces to a large extent. The two species have, however, different reproductive strategies. Littorina littorea has a planktonic larval stage lasting 4–6 weeks, whereas L. saxatilis has direct development. This influences dispersal rates of the two species, and gene flow and effective population size are both assumed to be much smaller in L. saxatilis than in L. littorea. Intraspeciflc variation in morphological and allozyme characters differs between the two species. Littorina littorea is homogeneous for both types of characters, both within and between populations, while populations of L. saxatilis may show pronounced differences in shell characters with micro-environment changes, and allozyme variation is markedly higher in this species too. The polymorphism in shell characters of L. saxatilis and the monomorphism of L. littorea, may be expected from theory as a consequence of the different dispersal strategies. Also the lack of divergence between geographically separate populations of L. littorea, but the high degree of between population differentiation in L. saxatilis, could be attributed to the high and low dispersal rates, respectively. However, the larger allozyme variation within subpopulations of L. saxatilis compared to that of L. littorea, is surprising as the former species has a smaller effective population size than the latter. Furthermore, there is no correlation between dispersal ability and overall allozyme heterozygosity, when 10 additional species of Littorina are included in the comparison. It is suggested that different evolutionary backgrounds of different species will possibly explain this, as neither selectionist nor neutralist models alone account for the observation. That is, an earlier passage through a population bottleneck may, for example, influence the genetic variation for a long period of time.  相似文献

The intertidal snail Littorina saxatilis displays a range ofshell morphologies associated with a variety of habitats. Sincemorphology has an environmental and genetic basis, shell-basedtaxonomy may not accurately reflect genetic relationships. Weexplored genetic structure among adjacent populations of L.saxatilis (the robust open-shore type), L. neglecta (the tinybarnacle-dwelling type) and L. tenebrosa (the fragile brackish-watertype), at nine sites in Britain. Using single-strand conformationalpolymorphism analysis of a 375bp fragment of cytochrome-b wefound no evidence of species distinction. In AMOVA tests significantvariation was contained among populations (68%) and among individuals(32%, both P < 0.001), and insignificant variation was foundamong ecotypes. Understanding what determines range expansion or extinction is crucial to predict the success of biological invaders. We tackled this long-standing question from an unparalleled perspective using the failed expansions in Littorina saxatilis and investigated its present and past habitat suitability in Europe through Ecological Niche Modelling. This intertidal snail is a typically successful Atlantic colonizer and the earliest confirmed alien species in the Mediterranean Sea, where, however, it failed to thrive despite its high dispersal ability and adaptability. We explored the environmental constraints affecting its biogeography, identified potential glacial refugia in Europe that fuelled its post-glacial colonisations and tested whether the current gaps in its distribution are linked to local ecological features. Our results suggested that L. saxatilis is unlikely to be a glacial relict in the Mediterranean basin. Multiple Atlantic glacial refugia occurred in the Last Glacial Maximum, and abiotic environmental features such as salinity and water temperature have influenced the past and current distributions of this snail and limited its invasion of the Mediterranean Sea. The snail showed a significant overlap in geographic space and ecological niche with Carcinus maenas, the Atlantic predator, but distinct from Pachygrapsus marmoratus, the Mediterranean predator, further pointing to Atlantic-like habitat requirements for this species. Abiotic constrains during introduction rather than dispersal abilities have shaped the past and current range of L. saxatilis and help explaining why some invasions have not been successful. Our findings contribute to clarifying the processes constraining or facilitating shifts in species’ distributions and biological invasions.

Four Littorina species from the Pacific (L. sitkana, L. brevicula,L. mandshurica, L. squalida) and four from the Atlantic (L.mariae, L. obtusata, L. saxatilis and L. littorea) were comparedelectrophoretically using 17 loci. Analysis of genetic identities showed that there were threegroups. The first group comprises L. sitkana, L. mariae, L.obtusata and L. saxatilis The second group consists of L. breviculaand L. mandshurica and the third one includes L. squalida andL. littorea. Microsatellites are important genetic markers both in population genetics and for delimitation of closely related species. However, to develop microsatellites for each target organism is expensive and time consuming. In this study, we have therefore developed 65 new microsatellite primers for the species Draba nivalis and tested cross-species and cross-genus transfer success of these primers for two other genera in the Brassicaceae; Cardamine and Smelowskia. Furthermore, 15 previously developed microsatellites were tested for amplification in these three genera. The microsatellite markers that amplify across these genera may be useful for other genera in the Brassicaceae as well.  相似文献

Characterizing the patterns of hybridization between closely related species is crucial to understand the role of gene flow in speciation. In particular, systems comprising multiple contacts between sister species offer an outstanding opportunity to investigate how reproductive isolation varies with environmental conditions, demography and geographic contexts of divergence. The flat periwinkles, Littorina obtusata and L. fabalis (Gastropoda), are two intertidal sister species with marked ecological differences compatible with late stages of speciation. Although hybridization between the two was previously suggested, its extent across the Atlantic shores of Europe remained largely unknown. Here, we combined genetic (microsatellites and mtDNA) and morphological data (shell and male genital morphology) from multiple populations of flat periwinkles in north‐western Iberia to assess the extent of current and past hybridization between L. obtusata and L. fabalis under two contrasting geographic settings of divergence (sympatry and allopatry). Hybridization signatures based on both mtDNA and microsatellites were stronger in sympatric sites, although evidence for recent extensive admixture was found in a single location. Misidentification of individuals into species based on shell morphology was higher in sympatric than in allopatric sites. However, despite hybridization, species distinctiveness based on this phenotypic trait together with male genital morphology remained relatively high. The observed variation in the extent of hybridization among locations provides a rare opportunity for future studies on the consequences of different levels of gene flow for reinforcement, thus informing about the mechanisms underlying the completion of speciation.  相似文献

Three species in the Littorina saxatilis complex (the ovoviviparousL. saxatilis and the two oviparous species L. arcana and L.nigrolineata) were screened for the products of up to 22 lociusing gel elec-trophoresis. Analysis of allele frequencies andderived genetic distances showed that, with rare exceptionsin L. saxatilis and L. arcana, conspecific populations clusteredtogether. Sympatric pairs of populations showed significantdifferentiation at many polymorphic loci. The results confirmedthe biological reality of the three taxa, and did not suggestthe existence of any previously unrecognized species. Gross anatomical characters of all 18 species of Littorina are used to construct a phylogenetic hypothesis for the genus, by the method of cladistic analysis. The resulting cladogram suggests that of the four subgenera of Littorina, one (Littorina) is paraphyletic. It is uncertain whether the genus Mainwaringia should be included in Littorina. It is shown that the non-planktotrophic Littorina species in the northern Atlantic comprise a monophyletic group, with the sister-species L. kurila and/or L. subrotundata in the northern Pacific. Invasion of the Atlantic by a minimum of two Pacific species, across the Arctic migration route established during the late Cenozoic, is sufficient to account for the modern distribution of the subgenera Littorina and Neritrema. The importance of the cladogram as a basis for hypotheses of adaptation is illustrated by a discussion of spawn and development in Littorina.  相似文献

The esterase system has been studied using isoelectric focusingin two closely related rough periwinkles, Littorina saxatilisand L. arcana, in an investigation of possible differences betweenthe two species at this level of discrimination. Collectionsof both species were obtained from Yorkshire, north Cornwalland south Devon; L. saxatilis was also obtained from sites insouth Cornwall (in and near the estuary of the Tamar), Dorsetand Scotland, where L. arcana was not present. There is considerablevariation both within and between sites which tends to maskany overall differences between the two species. However, ingeneral there is a much greater degree of heterogeneity withinsamples of L. saxatilis than within samples of L: arcana whenboth species are found together; this is reflected in the ShannonWiener diversity index for bands. Also the number of esterasebands is greater for L. saxatilis than for L. arcana at eachof the five sites where both occurred. A dendrogram derivedfrom the percentage occurrence of each band present in eachsample produced a reasonable degree of separation of the twospecies (Received 8 January 1988; accepted 24 February 1988)  相似文献

Eight polymorphic microsatellite loci are described for Littorina littorea (Linnaeus, 1758). Data on allelic variation in Irish and Celtic Sea samples are reported. The average number of alleles per locus was 11 (range 4–29), and observed and expected heterozygosities ranged from 6.9 to 84.3% and from 9.4 to 95.2%, respectively. Loci did not deviate from Hardy–Weinberg equilibrium and no linkage disequilibrium between loci pairs was detected. Microsatellites were not highly conserved in the congeners, L. fabalis, L. saxatilis, L. compressa and L. obtusata as evidenced by a low rate of cross-amplification. These microsatellites should prove useful in population genetic studies.  相似文献

This paper details attempts to breed members of the L. saxatilis species complex in the laboratory and to construct interspecific hybrids between them. Success in reciprocal crosses between animals from the type localities of the species of Littorina saxatilis (Olivi) and Littorina rudis (Maton) indicates that these two taxa are synonymous. Six of the twelve possible reciprocal hybridisation crosses between the four species (L. saxatilis, L. arcana Hannaford Ellis, L. nigrolineata Gray and L. neglecta Bean) have been attempted, with only one proving successful, that between male L. saxatilis and female L. arcana. This hybrid cross produces viable offspring, although at a lower frequency than either of the parental crosses in the laboratory, while the reciprocal cross has, as yet, proved unsuccessful. Limited work on the F1 hybrids shows them to be inter-fertile and also capable of backcrossing with male L. saxatilis, but not with female L. saxatilis or L. arcana. Details are also given of attempts to find natural hybrids in sympatric populations of the species of the complex, using gel electrophoresis.  相似文献

Examination of eight Atlantic species of the genus Littorina by starch gel electrophoresis of purine nucleoside phosphorylase revealed extensive polymorphism within the L. saxatilis complex. In this group, four alleles have been identified. Heterozygotes are four banded, and thus, as in vertebrates, the enzyme is likely to be a trimer. Breeding experiments confirmed the genetic interpretation of the phenotype patterns. Where species of the saxatilis complex [L. saxatilis (=L. rudis), L. arcana, L. nigrolineata, L. neglecta] are sympatric, there are sometimes significant allele frequency differences between them. A fifth allele was present at a high frequency in L. obtusata and L. mariae, and L. littorea and L. neritoides each possessed unique alleles. A total of eight alleles was identified. Densitometric scanning of heterozygote patterns pointed to activity differences between alleles and also showed that, while the heterotrimeric bands were never less intense than the homotrimeric bands, the heterotrimeric bands were sometimes less intense than expected. It is not clear whether this represents nonrandom association of subunits, decreased stability of heterotrimers, or simply an artifact of the staining and quantifying process.This work was supported by NERC Grant GR3/5319.  相似文献

An Atlantic salmon (Salmo salar L.) expressed sequence tag (EST) database consisting of 58 146 ESTs was screened for microsatellite sequences. Subsequent development of 75 polymorphic EST‐associated microsatellite markers in this species is described together with cross‐species amplification results of 133 gene‐associated tandem repeat markers in five salmonid species (Salmo trutta, Oncorhynchus mykiss, Salvelinus aplinus, Thymallus thymallus, Coregonus lavaretus). The number of alleles among EST‐linked microsatellites in Atlantic salmon ranged from two to 41 with an average of 12 alleles per locus. Cross‐species amplification resulted in detection of a total of 111 polymorphic locus‐species combinations (12–32 loci per species).  相似文献

Microsatellite markers were developed in Acacia mangium Willd. to provide highly variable co-dominant markers for linkage mapping and studies of the breeding system. After an enrichment procedure 40% of colonies contained microsatellites in contrast with less than 1% from a non-enriched library. The majority of microsatellite sequences were AC repeats. Co-dominant segregation of alleles in two full-sib crosses of A. mangium was demonstrated at 33 microsatellite loci. The markers were highly variable relative to restriction fragment lengths polymorphisms (RFLPs). In the two pedigrees 53% of microsatellite loci were fully informative compared with 15% of RFLPs. Based on alleles detected among four parental genotypes, the microsatellites consisting of dinucleotide repeats were more polymorphic than those with tri- and tetra-nucleotide repeats. The microsatellite markers were not as transferable across species in the genus Acacia as RFLPs. Two thirds of the primers developed in A. mangium (subgenus Phyllodineae, section Juliflorae) amplified DNA from other species within the same section but failed to amplify in species from the subgenus Acacia. The availability of multiallelic, PCR-based, co-dominant microsatellite loci makes possible efficient studies of gene flow and breeding systems in A. mangium, a species with low allozyme variation. Divergence of ecological niches in phylogenetically closely related species indicates the importance of ecology in speciation, especially for sympatric species are considered. Such ecological diversification provides an advantage of alleviating interspecies competition and promotes more efficient exploitation of environmental resources, thus being a basis for ecological speciation. We analyzed a group of closely related species from the subgenus Neritrema (genus Littorina, Caenogastropoda) from the gravel‐bouldery shores. In two distant sites at the Barents and Norwegian Sea, we examined the patterns of snail distribution during low tide (quantitative sampling stratified by intertidal level, presence of macrophytes, macrophyte species, and position on them), shell shape and its variability (geometric morphometrics), and metabolic characteristics (metabolomic profiling). The studied species diversified microbiotopes, which imply an important role of ecological specification in the recent evolution of this group. The only exception to this trend was the species pair L. arcana / L. saxatilis, which is specifically discussed. The ecological divergence was accompanied by differences in shell shape and metabolomic characteristics. Significant differences were found between L. obtusata versus L. fabalis and L. saxatilis / L. arcana versus L. compressa both in shell morphology and in metabolomes. L. saxatilis demonstrated a clear variability depending on intertidal level which corresponds to a shift in conditions within the occupied microhabitat. Interestingly, the differences between L. arcana (inhabiting the upper intertidal level) and L. compressa (inhabiting the lower one) were analogous to those between the upper and lower fractions of L. saxatilis. No significant level‐dependent changes were found between the upper and lower fractions of L. obtusata, most probably due to habitat amelioration by fucoid macroalgae. All these results are discussed in the contexts of the role of ecology in speciation, ecological niche dynamics and conservatism, and evolutionary history of the Neritrema species.  相似文献

Three sister species of rough periwinkles, viz. Littorina saxatilis (Olivi 1792), L. arcana (Hannaford Ellis 1978) and L. compressa (Jeffreys 1865) from the Barents Sea (Russia), the White Sea (Russia) and the Norwegian Sea (Norway) were studied. The identification of two sibling species L. saxatilis and L. arcana is often difficult as both species have extremely similar shell morphology and reproductive systems. Only mature females can be unambiguously distinguished, with a jelly gland present in female L. arcana, but which is replaced by a brood pouch containing developing embryos in L. saxatilis. No clear-cut diagnostic features have been found to discriminate between males or juveniles of the two species. The very first diagnostic DNA marker (DNA fragment A2.8, 271 bp length) for L. arcana and L. saxatilis separation was developed. The marker was derived from apparently species-specific L. arcana DNA fragments obtained via Random Amplified Polymorphic DNA (RAPD) analysis. This fragment was cloned and sequenced, whereupon specific primers were designed and the amplification was surveyed in a large number of morphologically well-identified females of both species. Subsequently, the specific DNA marker was used for the identification of male L. arcana and partners in copulating pairs. In this way, we obtained evidence of possible interspecific hybridization between the sibling species L. arcana and L. saxatilis living in sympatry in natural populations: the presence of A2.8 fragment in 12% of morphologically well identified L. saxatilis females and its absence in 14% of morphologically well identified L. arcana females. The A2.8 fragment never amplified in L. saxatilis from sites without L. arcana. The A2.8 fragment did not amplify in L. compressa, not even in microsympatric populations, and we did not observe interspecific copulations between L. arcana and L. compressa.  相似文献
