The lytic activity of plague phage II, serovar 3, with respect to 1,800 bacterial strains has been studied: 760 Yersinia pestis strains, 262 Y. pseudotuberculosis strains, 252 Y. enterocolitica strains, 166 Escherichia coli strains, 90 Shigella strains and 270 strains of other species. The phage has been found to lyse 81.8% of Y. pestis strains, 1 Y. pseudotuberculosis strain and 1 Y. enterocolitica strain. The representatives of other 19 bacterial species have proved to be resistant to the phage. Though having a wide range of action within Y. pestis, the phage does not lyse most of the strains of the causative agent of plague, isolated in certain natural foci. This fact offers promise for using the phage for the differentiation of Y. pestis.  相似文献

The results of a prolonged (more than 18 years), comprehensive study have revealed that stable natural foci of tularemia in backwater swamps are widely spread in the Leningrad region. These foci are located in the narrow swampy flood-plains of small watercourses with adjacent meadow areas among forests. Water from such small watercourses can often serve as the indicator of the epizootic process: during the above-mentioned period 346 Francicella tularensis strain have been isolated from water and 86 strains from small mammals. The water factor plays an important role in the circulation of the infective agent in natural foci.  相似文献

The typing of F. tularensis strains by four variable number of tandem repeats (VNTR) loci has been carried out. Among the strains isolated in the Stavropol and Krasnodar Territories seven genotypes have been detected and their spread in different natural foci has been analyzed. The data thus obtained suggest that the VNTR analysis may become an important instrument for studying the structure of the natural foci of tularemia and evolutionary relationships between individual areas of these foci.  相似文献

Strains of tick-borne encephalitis virus isolated in the natural foci of infection in the eastern part of the Russian Plain (the Kirov region) were examined for their biological properties. The strains examined were 69 strains isolated from ticks Ixodes persulcatus, 62 strains obtained from patients with the clinically manifest form of tick-borne encephalitis and 56 strains isolated from the blood of patients with the inapparent form of infection. Comparative studies on laboratory animals (albino mice, golden hamsters, suckling guinea pigs and other mammals) as well as comparative serologic studies provided evidence which suggested that all virus isolates from the Kirov region were antigenically identical with the strain "Sofin" isolated in the Far East and represent thus a single causative agent of the tick-borne encephalitis virus infection. This strain of virus is supposed to exist in two variants, in dependence on ecological conditions: one of these variants is the eastern variant (strain Sofin and strains from the Kirov region) and the other one is the western variant of tick-borne encephalitis virus.  相似文献

With a large collection of the strains of F. tularensis isolated it has been recently shown that cultures belonging to holarctica and mediaasiatica circulate in the endemic foci of the USSR. By their biological and genetic properties the natural strains of F. tularensis were homogeneous and represented type cultures of F. tularensis. Various ecological conditions in the natural environment did not change within the last 20 years the sensitivity of the tularemia microbe to the antibacterial drugs.  相似文献

Application of the methods developed by the author to the plague causative agent isolated in six natural plague foci provided detection of its ability to synthesize a factor inhibiting the growth of the producing strain and cultures isolated from the same focus. In the strains isolated from these foci the inhibitor presence was stable on the agar consisting of fermentative casein hydrolysate and 0.6 per cent of lithium chloride when the indicator strains were incubated under definite temperature conditions. The results of the present study and previous studies of the author, as well as the literature data suggested that the ability to synthesize the inhibitor is a stable feature of the plague causative agent.  相似文献

Summary Drosophila paulistorum Dobzhansky et Pavan is a complex of six races or incipient species. The races are mostly allopatric, but they are reproductively isolated sufficiently to permit them to exist also sympatrically in some places. The gene arrangements in the chromosomes of the races have been compared by means of examination of the giant chromosomes in the larval salivary glands; 28 strains of all races, and about an equal number of interracial hybrids have been studied.Chromosomal inversion polymorphism has been discovered in all races, even in the Guianan race of which only a single strain is available. Inversion heterozygotes are found in every one of the five chromosomal strands which the species has. Interracial hybrids tend to be heterozygous for more inversions than are present in the strains of the parental races. The Transitional race has however much the same gene arrangements as the widespread Andean — South Brazilian race.With the exception of the Transitional race, and of three other possible exceptions, each race has a collection of its own race-specific inversion polymorphs, not found in the other races. Thymine- and thymidine-dependent mutants of Y. pestis strain EV-76 have been isolated and characterized. Obtaining Y. pestis thymine-dependent mutants in trimethoprim-containing media with full nutritional value in the presence of thymine and thymidine and the capacity of natural strains from the foci of infection in Transcaucasia and Mongolia to grow in such media indicate that Y. pestis has gene tpp controlling thymidine phosphorylase, but this enzyme is strongly suppressed under normal conditions. The capacity for its suppression under definite conditions and the degree of the activation of thymidine phosphorylase determine the realization of Thy and Thyd phenotypes in Y. pestis mutants under study, though both types of these mutants have a mutation damage of gene thy A coding the synthesis of thymidylate synthetase.  相似文献

Bacteriocins formed by four strains of Lactococcus lactis subsp. lactis have been studied and compared: 729 (a natural strain isolated from milk), 1605 (a mutant of strain 729), F-116 (a recombinant obtained by fusing of protoplasts of the two related strain 729 and 1605), and a nisin-forming strain obtained by adaptive selection at Moscow State University. Antimicrobial activity studies revealed differences between the strains in the effects on individual groups of microorganisms; the activities of the strains were also distinct from that of Nisaplin (a commercial preparation of the bacteriocin nisin). Methods for isolation and purification of bacteriocins have been developed, making it possible to obtain individual components of antibiotic complexes as chromatographically pure preparations. Bacteriocins formed by the strains of Lactococcus lactis subsp. lactis have been identified and differences in their biological and physicochemical properties, established. A novel potent broad-spectrum antibiotic substance distinct from nisin has been isolated from the recombinant strain F-116.  相似文献

Biological properties of two strains of the Burnet rickettsia isolated in the natural foci of the Q fever in the northwest of the Ukrainian SSR are studied. Strain "Gishin" was characterized by high virulence for the laboratory animals and considerably exceeded the strain "Politsa" in the virulent properties. Antigen of phase I prepared from the strain "Politsa" was highly active in revealing the corresponding antibodies. The high-sensitive reaction of indirect immunofluorescence has shown the possibility to determine antibodies to phase I of the agent already from the 17th day after infection. Circulation of the Q fever agent with different virulent properties has indicated the necessity of purposeful diagnosis of this sickness both among the acute fever diseases and among flaccid course, subclinical and chronic ones not excluding the etiological role of the Burnet rickettsia.  相似文献

92 strains of Yersinia pestis isolated from different natural foci and stored for 3-40 years in the museum of live cultures have been studied. The strains having three typical plasmids, their different combinations, plasmidless strains or the strains carrying nontypical plasmids with the molecular masses 9, 15, 55, 80, 90 and 150 Md were found. The old museum strains are proposed to be used as a source of plasmids for the genetical research. The current control of plasmid contents in the museum strains is suggested by the plasmid changes in course of storage.  相似文献

Genome polymorphism by the locus (CAAA)n was studied in 69 strains of Yersinia pestis isolated from natural foci of the former Soviet Union. The polymorphism was found to be represented by ten alleles in chromosomes, which could be regarded as evidence of variability of this VNTR-locus (diversity index, DI = 0.86). The value of DI was found to vary substantially: from 0.24 in a group of vole strains from seven isolates from the Transcaucasian highlands to 0.77 in nine strains from the Central Asia desert focus. The allele polymorphism of the variable locus (CAAA)n in natural strains of Y. pestis was suggested to be used as a possible genetic marker of the strain. It was concluded that the oligonucleotide primers used in polymerase chain reaction should be upgraded to the genotyping accuracy.  相似文献

The results of the study of the species composition of the fecal microflora of 15 healthy rhesus macaques, from whom 204 strains of lactobacilli have been isolated and identified, are presented. The intestinal microflora has been shown to include different species, subspecies and biovars of homo- and heterofermentative lactobacilli and to possess definite individual properties. L. acidophilus, mannose-negative biovar I, and L. fermentum, mannose-negative biovars I and II, occur most frequently in feces. Certain differences in the species composition of lactoflora in monkeys and humans have been revealed: in the former, L. fermentum, mannitose-positive biovar III, and L. casei occur considerably more seldom. The inclusion of L. acidophilus strain I L. fermentum strains I and II isolated from rhesus macaques into eubiotics for these monkeys has been suggested.  相似文献

The nucleotide sequences of the Tc's insect toxin complex genes have been analyzed in 18 natural strains of the main and non-main subspecies of Yersinia pestis isolated in different natural foci in the Russian Federation, as well as neighboring and more remote countries, as compared to the data on Y. pestis and Y. pseudotuberculosis strains stored in the NCBI GenBank database. The nucleotide sequences of these genes in plague agent strains have been found to be highly conserved, in contrast to those of the pseudotuberculosis agent. The sequences of two genes, tcaC and tccC2, have been found to be almost identical in Y. pestis strains, whereas other three genes (tcaA, tcaB, and tccC1) contain a few mutations, which, however, are not common for all strains of the plague agent. Exceptions are only strains of the Y. pestis biovar orientalis, whose tcaB gene is in a nonfunctional state due to a nucleotide deletion. The results suggest that the formation of the species Y. pestis as an agent of a natural focal infection with a transmissive mechanism has not resulted in degradation of the Tc's complex genes. Instead, these genes are likely to have been altered as the plague agent have been adapting to the new environment.  相似文献

Reaction of the strains of the tularemia causative agent to erythromycin (100 gamma/ml) or oleandomycin (400 gamma/ml) is one of the taxonomic tests. The study of 82 strains of the holarctic race and 63 strains of the Central Asiatic race according to this test showed that the strains of the holarctic race were divided with respect to these macrolides into sensitive (biotype I) and resistant (biotype II). The strains of the both races were isolated at the territory of the Kazakh SSR. Such a reaction of the strains of the holarctic race is a stable feature and is not connected with virulence, the isolation source and the biochemical properties of the strains. Division of the holarctic race into the biotypes (I and II) with respect to erythromycin and oleandomycin may be of definite significance in epidemiological and epizootological examination for tularemia with a purpose of determining the source of the infection, as well as in defining the borders of the infection focus area with circulation of this or that biotype of the holarctic race of the tularemia causative agent.  相似文献

BackgroundPlague is a highly dangerous vector-borne infectious disease that has left a significant mark on history of humankind. There are 13 natural plague foci in the Caucasus, located on the territory of the Russian Federation, Azerbaijan, Armenia and Georgia. We performed whole-genome sequencing of Y. pestis strains, isolated in the natural foci of the Caucasus and Transcaucasia. Using the data of whole-genome SNP analysis and Bayesian phylogeny methods, we carried out an evolutionary-phylogeographic analysis of modern population of the plague pathogen in order to determine the phylogenetic relationships of Y. pestis strains from the Caucasus with the strains from other countries.ResultsWe used 345 Y. pestis genomes to construct a global evolutionary phylogenetic reconstruction of species based on whole-genome SNP analysis. The genomes of 16 isolates were sequenced in this study, the remaining 329 genomes were obtained from the GenBank database. Analysis of the core genome revealed 3315 SNPs that allow differentiation of strains. The evolutionary phylogeographic analysis showed that the studied Y. pestis strains belong to the genetic lineages 0.PE2, 2.MED0, and 2.MED1. It was shown that the Y. pestis strains isolated on the territory of the East Caucasian high-mountain, the Transcaucasian high-mountain and the Priaraksinsky low-mountain plague foci belong to the most ancient of all existing genetic lineages - 0.PE2.ConclusionsOn the basis of the whole-genome SNP analysis of 345 Y. pestis strains, we describe the modern population structure of the plague pathogen and specify the place of the strains isolated in the natural foci of the Caucasus and Transcaucasia in the structure of the global population of Y. pestis. As a result of the retrospective evolutionary-phylogeographic analysis of the current population of the pathogen, we determined the probable time frame of the divergence of the genetic lineages of Y. pestis, as well as suggested the possible paths of the historical spread of the plague pathogen.  相似文献

Four new strains of Botryococcus braunii were isolated from Japanese waters and cultured under defined conditions. Their hydrocarbon content and composition were analyzed and compared with those of the Darwin and Berkeley strains. The Yamanaka strain produced only alkadienes characteristic of the A race, whereas the others, the Yayoi, Kawaguchi-1 and -2 strains as well as the Darwin and Berkeley strains, produced botryococcenes peculiar to the B race. The hydrocarbon content of the Yamanaka strain was 16.1 % dry weight and that of the B race strains ranged from 9.7 to 37.9%. Botryococcene composition of the Japanese strains differed from each other as well as from the Darwin and Berkeley strains. More than 50% of the hydrocarbons in the Yayoi, Darwin, and Berkeley strains were composed of C34H58, but the main components were different from one another as isomers. The Kawaguchi-1 and -2 strains did not have a high level of C34 botryococcenes, C32 ones being the main components. In these strains significant amounts of squalene-related compounds were detected.  相似文献

The partial reactions of photosynthesis shown by strain F208, a non-photosynthetic mutant strain of Scenedesmus obliquus, have been compared with those performed by other mutant strains which lacked; Photosystem II activity (strains 11 and F131), cytochrome f (strain 50), P-700 and cytochrome f (strain F 119), and P-700 (strains F139 and 199). In this respect the properties of strain F208 were those that would be expected if Photosystem II activity and cytochrome f were not present in this strain. Examination of the composition of strain F208 has shown the absence of cytochrome f in both the soluble and the membrane-bound form. The considerably lower level of plastoquinone compared to that found in the wild type is characteristic of the strains which lack Photosystem II activities. Fraction 1 protein could not be detected in extracts of strain F208 by sedimentation velocity experiments in the ultracentrifuge, and only 7% of the wild type ribulose diphosphate carboxylase activity was found after chromatography of these extracts on DEAE-cellulose. The properties of strain F208 are compared with those of the ac-20 and cr-1 strains of Chlamydomanas rheinhardi, both of which have a deficiency of ribulose diphosphate carboxylase which is considered to result from a deficiency of chloroplast ribosomes. Strain F208 resembles these strains in its abnormal chloroplast ultrastructure and its decreased levels of the RNA forms derived from the chloroplast ribosomes when compared with the wild type. Chloroplast fragments isolated from strains of S. obliquus which lacked cytochrome f (strains 50 and F208) were able to use diaminodurene and ascorbate as an electron donor to Photosynstem I. Since this reaction was inhibited by mercuric salts it would appear that plastocyanin, but not cytochrome f, was involved in this electron transfer.  相似文献

1. The soluble cytochromes c-556 from three strains of Agrobacterium tumefaciens, B6, II Chrys and Apple 185 have been purified to homogeneity. The strains are representative members of the three main genetic races of Agrobacterium. The purity of the final preparations was established by electrophoresis with an without sodium dodecyl sulphate, by analytical isoelectric focusing and ultracentrifugation, and by N-terminal analysis. 2. Properties of these cytochromes were compared wih those of cytochrome c-556 from A. tumefaciens, strain B2a, a member of the same genetic race as strain B6. The four cytochromes are monohaem proteins with molecular weights of about 12300 (determined by four different methods). The isoelectric points of those from strains B6 and B2a are identical at pH 5.5, but they differ from the cytochromes of the other genetic races: cytochrome c-556 from strain Apple 185 is more acidic (ph 5.2) and that from strain II Chrys more basic (pH 6.2). The cytochromes from strains b6 and B2a have very similar but not identical amino acid compositions; both of them differ more from Apple 185 than from II Chrys c-556. 3. Comparison of the tryptic, chymotryptic and thermolytic fingerprints of cytochrome c-556 from strains B2a and II Chrys reveals strong homology between the primary structures of these cytochromes. Therefore and because of the sequence identity of the first eight residues, the cytochromes c-556 from strains II Chrys, B6 and B2a are most likely C-terminal haem-bound, of the same type as the cytochrome c' from photosynthetic bacteria.  相似文献
