We have previously shown (R.A. Raguso, E. Pichersky [1995] Plant Syst Evol 194: 55-67) that the strong, sweet fragrance of Clarkia breweri (Onagraceae), an annual plant native to California, consists of 8 to 12 volatile compounds, including 4 phenylpropanoids. Although some C. breweri plants emit all 4 phenylpropanoids (eugenol, isoeugenol, methyleugenol, and isomethyleugenol), other C. breweri plants do not emit the latter 2 compounds. Here we report that petal tissue was responsible for the bulk of the phenylpropanoid emission. The activity of S-adenosyl-L-methionine: (iso)eugenol O-methyltransferase (IEMT), a novel enzyme that catalyzes the methylation of the para-4'-hydroxyl of both eugenol and (iso)eugenol to methyleugenol and isomethyleugenol, respectively, was also highest in petal tissue. IEMT activity was absent from floral tissues of plants not emitting (iso)methyleugenol. A C. breweri cDNA clone encoding IEMT was isolated, and its sequence was shown to have 70% identity to S-adenosyl-L-methionine:caffeic acid O-methyltransferase. The protein encoded by this cDNA can use eugenol and isoeugenol as substrates, but not caffeic acid. Steady-state IEMT mRNA levels were positively correlated with levels of IEMT activity in the tissues, and no IEMT mRNA was observed in flowers that do not emit (iso)methyleugenol. Overall, the data show that the floral emission of (iso)methyleugenol is controlled at the site of emission, that a positive correlation exists between volatile emission and IEMT activity, and that control of the level of IEMT activity is exerted at a pretranslational step.  相似文献

The leaf essential oil of Laurus nobilis Linn., Lauraceae, which has been used as an antiepileptic remedy in Iranian traditional medicine, was evaluated for anticonvulsant activity against experimental seizures. The essential oil protected mice against tonic convulsions induced by maximal electroshock and especially by pentylenetetrazole. Components responsible for this effect may be methyleugenol, eugenol and pinene present in the essential oil. At anticonvulsant doses, the essential oil produced sedation and motor impairment. This effect seems to be related in part to cineol, eugenol and methyleugenol. Although the essential oil had an acceptable acute toxicity, further studies are required before any absolute conclusions can be drawn.  相似文献

Deletion of an integrated plasmid, a specific type of intrachromosomal recombination, was evaluated for inducibility with the phenylpropenes safrole, eugenol and methyleugenol in the yeast Saccharomyces cerevisiae. These phenylpropenes are found in food products, spices, pharmaceuticals and clove cigarettes. Safrole and eugenol are known carcinogens in animals and methyleugenol is a suspected carcinogen. These phenylpropenes are not detectable by the Ames assay and most other short-term tests used currently in predictive carcinogenesis. Like safrole, which has been shown to be nonmutagenic with the Ames assay, eugenol and methyleugenol were found to be nonmutagenic with the Ames assay. In contrast, with the yeast assays which screen for intra- and inter-chromosomal recombination in logarithmic phase cultures, all 3 compounds gave a positive dose-related response. These results demonstrate further that the yeast system can be modified easily to detect various genetic endpoints and that it deserves serious consideration as a test system for predictive carcinogenesis.  相似文献

Methyleugenol is a substituted alkenylbenzene found in a variety of foods, products, and essential oils. In a 2-year bioassay conducted by the National Toxicology Program, methyleugenol caused neoplastic lesions in the livers of Fischer 344 rats and B6C3F(1) mice. We were interested in the cytotoxicity and genotoxicity caused by methyleugenol and other alkenylbenzene compounds: safrole (a known hepatocarcinogen), eugenol, and isoeugenol. The endpoints were evaluated in cultured primary hepatocytes isolated from male Fischer 344 rats and female B6C3F(1) mice. Cytotoxicity was determined by measuring lactate dehydrogenase (LDH) release, while genotoxicity was determined by using the unscheduled DNA synthesis (UDS) assay. Rat and mouse hepatocytes showed similar patterns of toxicity for each chemical tested. Methyleugenol and safrole were relatively non-cytotoxic, but caused UDS at concentrations between 10 and 500 microM. In contrast, isoeugenol and eugenol produced cytotoxicity in hepatocytes with LC50s of approximately 200-300 microM, but did not cause UDS. Concurrent incubation of 2000 microM cyclohexane oxide (CHO), an epoxide hydrolase competitor, with a non-cytotoxic concentration of methyleugenol (10 microM) resulted in increased cytotoxicity but had no effect on genotoxicity. However, incubation of 15 microM pentacholorophenol, a sulfotransferase inhibitor, with 10 uM methyleugenol resulted in increased cytotoxicity but had a significant reduction of genotoxicity. These results suggest that methyleugenol is similar to safrole in its ability to cause cytotoxicity and genotoxicity in rodents. It appears that the bioactivation of methyleugenol to a DNA reactive electrophile is mediated by a sulfotransferase in rodents, but epoxide formation is not responsible for the observed genotoxicity.  相似文献

The experiments were commenced in March 2003 and repeated in June 2003 at Sutton Bonington Campus, the University of Nottingham, UK, to investigate the effect of irradiance on plant growth and volatile oil content and composition in plants of basil. Four levels of irradiance were provided in the glasshouse, i.e. no shade (control), 25, 50 and 75% glasshouse irradiance. It suggested that basil grows well in full sun, however it can tolerate light shade. Heavy shading (75%) to provide a light integral of 5.3 moles m−2 d−1 resulted in shorter plants, lower weight, smaller leaf area, less shoots and higher specific leaf area, and also strongly reduced the rate of photosynthesis. There was no difference in CO2 assimilation rate between 24.9 moles m−2 d−1 light integrals (no shading) and 13.5 moles m−2 d−1 light integrals (25% shading). Shading effectively reduced leaf temperature when air temperature was less than 30 °C, but heavy shading (75%) could not reduce leaf temperature when air temperature was above 36 °C due to a limitation of free air convection. Consequently, leaf temperature increased. Heavy shading strongly reduced total volatile oil content in fresh leaves, especially in older plants (shading treatment applied at the 3 leaf-pair growth stage). There were three chemical compounds in basil leaves, namely linalool, eugenol and methyl eugenol, influenced by the shading treatments. Linalool and eugenol, which contribute to the characteristic taste of basil, were significantly increased by high daily light integrals, whereas methyleugenol was increased by lower daily light integrals. No differences in the relative content of 1,8-cineole, one of the key aromatic compounds of Ocimum species, were observed.  相似文献

Essential oil extracted from nutmeg seeds (Myristica fragrans Houtt .) by hydrodistillation was subjected to GC/MS and GC analysis. A total of 27 constituents were identified, of which eugenol (19.9%), methylisoeugenol (16.8%), methyleugenol (16.7%), sabinene (11.8%), and terpinen‐4‐ol (8.5%) were the major components. The essential oil was tested against Lasioderma serricorne for insecticidal and repellent activity, the LD50 value at the end of 24 h exposure period was 19.3 μg/adult. Six active compounds were isolated by bioassay‐guided fractionation. They were identified as eugenol ( 1 ), methyleugenol ( 2 ), methylisoeugenol ( 3 ), elemicin ( 4 ), myristicin ( 5 ), and safrole ( 6 ). Among these isolates, 4 showed the strongest contact toxicity against L. serricorne adults with an LD50 value of 9.8 μg/adult. Repellency of crude oil and active compounds were also determined. Compounds 1, 2, 4 , and 5 were strongly repellent against the cigarette beetle and exhibited the same level of repellency compared with the positive control, DEET. The results indicate that the essential oil of M. fragrans and its active constituents have potential for development as natural insecticides and repellents to control L. serricorne.  相似文献

对水葱(Scirpus validus Vahl)、香蒲(Typha orientalis Presl)、小香蒲(T.minima Funk.)、再力花(Thalia dealbata Fraser ex Roscoe)、黄菖蒲(Iris pseudacorus Linn.)、灯芯草(Juncus effusus Linn.)、梭鱼草(Pontederia cordata Linn.)和菖蒲(Acorus calamus Linn.)8种多年生湿地植物1年生和3年生植株的地上和地下部分干质量、株高(包括花序高和叶层高)、根长、根数和根粗进行了测定,并对各表型特征指标间以及苗龄与各表型特征指标间的相关性进行了分析。结果显示:8种湿地植物3年生植株的地上和地下部分干质量普遍高于1年生植株;从生长量分配情况看,除小香蒲外,其余种类3年生植株地下部分干质量所占比例均高于1年生植株。多数种类1年生和3年生植株的株高差异较小。从根系特征看,根数小于100、根粗1.0~2.0 cm的植株以1年生为主,而根数大于100、根粗1.0 cm以下和2.0~3.5 cm的植株以3年生为主;根长15~25 cm的植株以1年生所占比例较高(62.5%),而根长25~35 cm的植株1年生和3年生所占比例相等。相关性分析结果表明:1年生植株的地上部分干质量与株高、根长与根粗呈显著正相关(P<0.05),3年生植株的地下部分干质量与根粗也呈显著正相关;但不同苗龄植株的其他表型特征指标间的相关性均不显著。苗龄与植株地下部分干质量呈极显著正相关(P<0.01),但与地上部分干质量、株高、根长、根数和根粗的相关性不显著。总体而言,8种湿地植物3年生植株的表型特征优于1年生植株,更适用于污染水体及退化湿地生态系统的修复。  相似文献

The population structure of the canopy liana Machaerium cuspidatum Kuhlm. & Hoehne (Fabaceae) was studied in ten transects covering a total of 2.5 ha of tropical rain forest in Yasuní National Park, eastern Ecuador. The aim was to investigate how trellis availability, topographic position and light availability affected the population density and structure of the most abundant liana species in the area. The environmental variation affected plants in different size classes differently. Large plants (diameter 1 cm) were almost exclusively found in areas with low canopy and dense undergrowth. These areas had the highest density of suitable host plants. Seedling-sized plants (height < 30 cm) and saplings (height 30 cm but diameter < 1 cm) occurred more frequently and withhigher density in steep upland areas than in the floodplains, presumably due to elevated seedling mortality in the periodically flooded areas. Seedling-sized plants and saplings did not grow in areas with elevated light levels, and although plants of all sizes were highly clumped, this clumping rarely coincided with patches with abundant canopy gaps. Machaerium cuspidatum reproduces clonally when branches re-root and break off from the parentplant. The density of clonally produced plants was high in areas where the density of sexually produced plants was low, whereas the total density varied little among habitat types. A higher proportion of the seedling-sized plants were of sexual origin in the upland areas. An environmentally induced increase in the production of independent ramets enables M. cuspidatum to persist in the floodplains. This shows that plants may expand theirrealised niches to habitats with a low potential for seedling survival if high seedling mortality can be compensated for by increased clonal reproduction.  相似文献

Soil communities associated with specific plant species affect individual plants' growth and competitive ability. Limited evidence suggests that unique soil communities can also differentially influence growth and competition at the ecotype level. Previous work with Arabidopsis thaliana has shown that accessions produce distinct and reproducible rhizosphere bacterial communities, with significant differences in both species composition and relative abundance. We tested the hypothesis that soil communities uniquely affect the growth and reproduction of the plant accessions with which they are associated. Specifically, we examined the growth of four accessions when exposed to their own soil communities and the communities generated by each of the other three accessions. To do this we planted focal accessions inside a ring of six plants that created a "background" soil community. We grew focal plants in this design in three separate soil treatments: non-sterile soil, sterilized soil, and "preconditioned" soil. We preconditioned soil by growing accessions in non-sterile soil for six weeks before the start of the experiment. The main experiment was harvested after seven weeks of growth and we recorded height, silique number, and dry weight of each focal plant. Plants grown in the preconditioned soil treatment showed less growth relative to the non-sterile and sterile soil treatments. In addition, plants in the sterile soil grew larger than those in non-sterile soil. However, we saw no interaction between soil treatment and background accession. We conclude that the soil communities have a negative net impact on Arabidopsis thaliana growth, and that the unique soil communities associated with each accession do not differentially affect growth and competition of study species.  相似文献

Plants only interact with neighbors over restricted distances, so local conditions are of great significance for plants. It is therefore important to consider spatial structure and neighborhood effects if we are to understand plants' strategies. We constructed a spatially-explicit, game theory model to explore optimal height growth at the individual-level. In the model, there is no ESS for height growth at the population level, because there is an “instantaneous” optimal height growth strategy for the individual plant that changes depending on the local light environment. The optimal strategy is plasticity in response to local conditions. Game-theoretic models for plant phenotypic traits should move from “mean-field approximations” towards explicit modeling of local interactions.  相似文献

Disorganized cells of A. dracunculus have been demonstratedto be competent to synthesize the four phenylpropene compoundscharacteristic of tarragon essential oil. Allylanisole and methyleugenol, the major components, are accumulated at particularlylow levels and are inhibited largely by the presence of exogenousNAA. Volatilization, active metabolism, and autotoxicity ofthese compounds are involved in limiting their accumulation.The data reported here suggest that both limited expressionof specific synthetic pathways, and the absence of suitable(inert) accumulation sites can restrict accumulation of phenylpropenesin disorganized cultures of A. dracunculus. Freezing tolerance and freezing avoidance were studied, during the growing season, in plant species from two different elevations (3200 m and 3700 m) in a desert region of the high Andes (29° 45S, 69° 59W) in order to determine whether there was a relationship between plant height and cold resistance mechanisms. Freezing injury and supercooling capacity were determined in plants of different height, from ground-level (<20 cm tall) to tall shrubs (27–90 cm). All ground-level plants showed freezing tolerance as the main mechanism for resistance to freezing temperatures. Tall shrubs avoided freezing temperatures, mainly through supercooling. Supercooling was only present in plants occupying the lower elevation (i.e., 3200 m). Both avoidance and tolerance mechanisms are present in a single genus (i.e., Adesmia).  相似文献

Two aspects of abiotic transport of nucleopolyhedrovirus from soil to cotton plants were examined in greenhouse experiments: the distance from the plants and depth in soil from which the virus could be transported under controlled conditions of soil type and moisture, wind, and precipitation. Transport distance and depth were tested separately under relatively conducive (precipitation/sandy soil and wind/clay soil) and non-conducive (precipitation/clay soil and wind/sandy soil) conditions, as determined in previous research. The amount of virus transported by precipitation generally decreased as distance from the plant increased, but in wind the amounts of virus transported were best described by polynomial models, with transport efficiency usually peaking at a distance of 60 cm. Depending on plant height and tissue, the farthest distances that virus was transported ranged from 30 to 60 cm by precipitation from clay soil, 60-75 cm in precipitation/sand, 60-80 cm in wind/clay, and 60-80 cm in wind/sand. In the depth experiments, transport by precipitation and wind generally decreased as the depth of virus in soil increased. The greatest depth from which NPV was transported ranged from 0 to 0.5 cm by precipitation from clay soil, 0.5-1.0 cm in precipitation/sand, 1.0-2.0 cm in wind/clay, and 0.5-1.0 cm in wind/sand. All of the experimental parameters (distance or depth, soil type, plant height, plant tissue) and all two-way interactions significantly (P<0.05) affected transport in all four experiments, except for the "soilxplant tissue" interaction in the depth/wind experiment. In all of the experiments, transport was significantly greater (P<0.05) to lower than to upper portions of plants and to leaves than to buds and squares. Transport was significantly greater from sandy soil than from clay in precipitation, and it was greater from clay than from sandy soil in wind. The results will contribute to NPV epizootiology, microbial control, and risk assessment.  相似文献

Background

Since loss of height may indicate vertebral fracture, the accuracy of the information on height is relevant for clinical practice. We undertook this study to compare reported and measured loss of height among post-menopausal women in a primary care setting. We also analyzed the determinants of this height loss.

Methods

In an observational study conducted between December 2007 and May 2008, we asked 1779 randomly selected general practitioners to recruit the first five female patients who were more than 60 years of age, regardless of the reason for the consultation. Using a questionnaire, physicians collected data on demographic and clinical variables, history of osteoporosis and current anti-osteoporotic treatment. We used three assessments of height: tallest height in early adulthood recalled by the patient, estimated current height reported by the patient at the visit and current measured height. We defined loss of height as the difference between the patient’s tallest recalled height and her current measured height.

Results

A total of 8610 patients were included in the analysis; the mean age was 70.9 (standard deviation [SD] 7.2) years. The mean loss of height was 4.5 cm. The mean current reported height was 2.1 (SD 2.5) cm lower than the tallest recalled height and 2.4 (SD 2.6) cm lower than the measured current height. The best predictors of a loss of height of 3 cm or more were age (odds ratio [OR] 1.09, 95% confidence interval [CI] 1.08–1.10), previous vertebral fracture (OR 1.49, 95% CI 1.16–1.91), previous nonvertebral fracture (OR 1.26, 95% CI 1.06–1.51), thoracic kyphosis (OR 2.07, 95% CI 1.69–2.55), scoliosis (OR 1.35, 95% CI 1.12–1.63), back pain (OR 1.22, 95% CI 1.07–1.39) and osteoporosis (OR 1.39, 95% CI 1.20–1.60).

Interpretation

Our study showed that the patients’ estimated current height was not correct, with a mean difference of −2.5 cm from the current measured height. The mean height loss was 4.5 cm. Previous vertebral fracture and thoracic kyphosis were strong determinants of the height loss.Loss of height is common with advancing age.1,2 Causes include changes in curvature of the spine, narrowing of intervertebral discs and vertebral fractures. Height loss is associated with back pain and thoracic hyperkyphosis.3,4 Two-thirds of adults have back pain at any time. Controversies exist about the need for radiographs of the spine: Does the benefit of detecting treatable disorders of the spine such as vertebral fracture outweigh the harm of unnecessary radiographs? Loss of height is usually recorded as one of the clinical signs to help identify postmenopausal women with vertebral fractures.5 The use of this parameter to decide whether radiography is needed depends on the threshold for height loss associated with a strong risk of vertebral fracture. The thresholds useful in clinical practice to detect prevalent vertebral fracture range from 3 cm to 6 cm,69 with the risk of prevalent fracture increasing with the magnitude of the height loss. Thus, the accuracy of the information on height is relevant for clinical practice.We conducted this study to compare reported and measured loss of height in a large population of women more than 60 years old in a primary care setting and to analyze the determinants of this height loss.  相似文献

In order to investigate broad patterns of variation of the foliar mineral nutrient concentrations of herbaceous plant communities in the ground layer of W Europe forests, correlations were examined between Ellenberg's indices (N-index: mineral nitrogen availability, R-index: pH, F-index: soil moisture and L-index: light intensity) and literature values of macronutrient concentrations for 84 forbs and 39 graminoid species. Significant, positive correlations were found between the R-index and the plants' concentration of Ca and K (forbs only) and between the N-index and the plants' concentration of K, P (forbs only) and N (forbs and graminoids). Multiple regressions showed that the N-index was the best predictor of the plants' concentration of N (forbs and graminoids), P (forbs) and Ca (graminoids) and the R-index of the plants' concentration of Ca and K (forbs). The mineral nutrient concentrations of graminoids were lower and less strongly correlated with Ellenberg's indices than those of forbs. It is argued that the mineral nutrient concentrations in the plants match the availability of mineral nutrients in the soil for N, P (N-index) and Ca (R-index), but not for K and Mg. Significant, positive correlations were found between potential relative growth rate and the concentration of some elements (N, P, K and Ca in forbs, Ca and Mg in graminoids). This suggests that the increase in the concentration of these elements in plants along fertility gradients is due, at least partly, to genetically controlled alterations of leaf anatomy associated with increasing potential relative growth rate.  相似文献

? In crowded stands, height is often similar among dominant plants, as plants adjust their height to that of their neighbours (height convergence). We investigated which of the factors, light quality, light quantity and mechanical stimuli, is primarily responsible for stem elongation and height convergence in crowded stands. ? We established stands of potted Chenopodium album plants. In one stand, target plants were surrounded by artificial plants that were painted black to ensure that the light quality was not modified by their neighbours. In a second stand, target plants were surrounded by real plants. In both stands, one-half of the target plants were anchored to stakes to prevent flexing by wind. The target plants were lifted or lowered by 10?cm to test whether height convergence was affected by the different treatments. ? Stem length was affected by being surrounded by artificial plants, anchoring and pot elevation, indicating that light quality, light quantity and mechanical stimuli all influenced stem elongation. Height convergence did not occur in the stand with artificial plants or in anchored plants. ? We conclude that light quality and mechanical stimuli are important factors for the regulation of stem growth and height convergence in crowded stands.  相似文献

本研究的目的是将甜菜碱合成关键酶CMO与BADH基因构建到同一表达载体中,利用转基因方法将该表达载体导入植物体内,完善植物体内的甜菜碱合成途径,提高植物的抗旱性和耐盐性。以pC1303质粒为基础,构建了均由35S启动子驱动的CMO基因和BADH基因的植物双基因表达载体pC35SC35SB1303。利用冻融法将其导入农杆菌LBA4404中,通过农杆菌介导法分别将CMO基因、BADH基因以及该双基因表达载体导入烟草中,PCR检测和Northern杂交分析表明,外源基因已整合到受体植物基因组中并正常表达。对转基因植株及对照植株甜菜碱含量的检测结果表明,转双基因植株的甜菜碱含量明显高于转BADH基因植株、转CMO基因植株及对照植株。  相似文献
