Frozen sections of formalin-fixed brains containing surgical lesions, were treated with 15% ethanol for 0.5 hr., soaked in 0.5% phosphomolybdic acid for 0.25-1.0 hr., and subsequently treated with 0.05% potassium permanganate for 4-10 min. (The duration of the latter treatment is critical and individually variable). Subsequent procedure is as follows: decolorize in a mixture of equal parts of 1% hydroquinone and 1% oxalic acid; wash thoroughly and soak sections in 1.5% silver nitrate for 20-30 min.; ammoniacal silver nitrate (silver nitrate 0.9 g., distilled water 20 ml., pure ethanol 10 ml., strong ammonia 1.8 ml., 2.5% sodium hydroxide 1.5 ml.) 0.5-1.0 min.; reduce in acidified formalin (distilled water 400 ml., pure ethanol 45 ml., 1% citric acid 13.5 ml., 10% formalin 13.5 ml.) 1 min.; wash, and pass section through 1 % sodium thiosulf ate (0.5-1.0 min.); wash thoroughly and pass sections through graded alcohols and xylene (3 changes); cover in neutral synthetic resin.

It is known that substantial boll weevil, Anthonomus grandis grandis Boheman, individuals can survive mild subtropical winters in some habitats, such as citrus orchards. Our study shows that endocarp of the fruit from prickly pear cactus, Opuntia engelmannii Salm-Dyck ex. Engel.; orange, Citrus sinensis L. Osbeck.; and grapefruit, Citrus paradisi Macfad., can sustain newly emerged adult boll weevils for >5 mo, which is the duration of the cotton-free season in the subtropical Lower Rio Grande Valley of Texas and other cotton-growing areas in the Western Hemisphere. Cotton, Gossypium hirsutum L., and the boll weevil occur in the same areas with one or all three plant species (or other citrus and Opuntia species that might also nourish boll weevils) from south Texas to Argentina. Although adult boll weevils did not produce eggs when fed exclusively on the endocarps of prickly pear, orange, or grapefruit, these plants make it possible for boll weevils to survive from one cotton growing season to the next, which could pose challenges to eradication efforts.

The O-specific polysaccharide of P. fluorescens IMV 2366 was studied by sugar and methylation analyses along with 1H and 13C NMR spectroscopy, including 2D gsCOSY, TOCSY, gsNOESY, H-detected 1H,(13)C gsHSQC, HMQC-TOCSY, and gsHMBC experiments. The polysaccharide contains L-rhamnose, 2-acetamido-2,6-dideoxy-D-galactose (D-FucNAc) and 3-acylamido-3,6-dideoxy-D-glucose (D-Qui3NAcyl, where Acyl is 3-hydroxy-2,3-dimethyl-5-oxoprolyl). The structure 1 of the polysaccharide was found to be similar to the structure 2 of a 6-deoxy-L-talose (L-6dTal)-containing O-specific polysaccharide of a non-classified P. fluorescens strain, 361, studied earlier [Khomenko, V. In the photoreaction of bacteriorhodopsin, the L intermediate shows an intense band at 3486 cm-1 which is unaffected by 2H2O (Maeda, A., Sasaki, J., Shichida, Y., & Yoshizawa, T. (1992) Biochemistry 31, 462-467]. This band is shifted to 3477 cm-1 by [indole-15N]tryptophan substitution and therefore is assigned to the N-H stretching vibration of the indole of tryptophan. Free indole in carbon tetrachloride shows its N-H stretching vibration at 3491 cm-1 [Fuson, N., Josien, M.-L., Powell, R. L., & Utterback, E. (1952) J. Chem. Phys. 20, 145-152]. Thus, it is suggested that at least one tryptophan residue in the L intermediate is not hydrogen bonded.

Ru(II)/Ru(III) polypyridyl complexes containing 2,6-(2'-benzimidazolyl)-pyridine or chalcone as co-ligands were synthesized and characterized previously (Mishra, L.; Sinha, R. Indian J. Chem., Sec. A 2001, in press. Mishra, L.; Sinha, R. Indian J. Chem., Sec. A, 39A, 2000, 1131). Their interaction with aqueous buffered calf thymus DNA was measured. (Novakova, O.; Kasparkova, J.; Vrana, O.; van Vliet, P. M., Reedijk, J.; Brabec, V., Biochem. 34, 1995, 12369 and these results prompted additional screening for anti-HIV (human immunodeficiency virus) activity against DNA replication in H9 lymphocytes and cytotoxic activity against eight tumor cell lines. The most active compounds were 17 in the former assay (EC(50) < 0.1 microg/mL and TI > 23.1) and 3, 8, 10, and 14 in the latter assay, especially selectively against the 1A9 ovarian cancer cell line (IC(50) = 4.1, 3.8, 3.6, and 2.5 microg/mL, respectively).

Helicoverpa armigera (Hiibner) (Lepidoptera: Noctuidae) is a well-known polyphagous insect pest. Mating compatibility among the insects occurring on different host plants is essential for free gene flow among populations. We tested the extent of crossability and fecundity of the insects that survived on Bacillus thuringiensis (Bt) cotton with those occurring on pigeon pea, Cajanus cajun (L.) Millsp., non-Bt cotton, Gossypium hirsutum L.; sunflower, Helianthus annuus; sorghum, Sorghum bicolor L. Moench.; okra, Abelmoschus moschatus Medikus; chickpea, Cicer arietinum L.; marigold, Tagetes spp.; and tomato, Lycopersicum esculentum L., crops. The insects from different crops were freely crossable with those collected from Bt cotton and among themselves. The average fecundity across different crosses ranged from 314.1 to 426.3 in direct and from 305.8 to 421.7 eggs per female in reciprocal crosses. In any given cross, a minimum of 85.89% egg hatch was recorded. Furthermore, the F1 individuals of different cross combinations were found to cross freely with their parents (BC1) and among themselves with similar fecundity and egg hatch. High crossability among H. armigera occurring on different host plants suggests that crop mosaics that may exist in countries such as India could play an important role as natural, nonstructured refugia and prolong the durability of the genes deployed for controlling this insect.
