Results are presented from time-integrated measurements of soft X-ray emission from Z-pinches during the implosion of simple and nested wire arrays. The blackening density distribution obtained with the help of a pinhole camera is recalculated into the time-integrated Z-pinch radiance. It is found that, in the case of a simple wire array, up to 70% of the total SXR energy emitted during a discharge is radiated from the axial region, the rest of energy being radiated from plasma jets, whereas in the case of a nested wire array, more than 90% of the SXR energy is radiated from the axial region.  相似文献

Two heart muscarinic responses are compared, the reduction of cyclic AMP concentration and the hydrolysis of phosphoinositides. It is suggested that the former is more important physiologically, since the latter is a slow response requiring much higher agonist concentrations. Inositol trisphosphate released from phosphoinositides is unlikely to produce a positive inotropic effect by releasing calcium from the sarcoplasmic reticulum. However, long term muscarinic effects may involve activation of protein kinase C by diacylglycerol released from phosphoinositides.  相似文献

The lateral geniculate nucleus is the best understood thalamic relay and serves as a model for all thalamic relays. Only 5-10% of the input to geniculate relay cells derives from the retina, which is the driving input. The rest is modulatory and derives from local inhibitory inputs, descending inputs from layer 6 of the visual cortex, and ascending inputs from the brainstem. These modulatory inputs control many features of retinogeniculate transmission. One such feature is the response mode, burst or tonic, of relay cells, which relates to the attentional demands at the moment. This response mode depends on membrane potential, which is controlled effectively by the modulator inputs. The lateral geniculate nucleus is a first-order relay, because it relays subcortical (i.e. retinal) information to the cortex for the first time. By contrast, the other main thalamic relay of visual information, the pulvinar region, is largely a higher-order relay, since much of it relays information from layer 5 of one cortical area to another. All thalamic relays receive a layer-6 modulatory input from cortex, but higher-order relays in addition receive a layer-5 driver input. Corticocortical processing may involve these corticothalamocortical 're-entry' routes to a far greater extent than previously appreciated. If so, the thalamus sits at an indispensable position for the modulation of messages involved in corticocortical processing.  相似文献

The illness-death model is the simplest multistate model where the transition from the initial state 0 to the absorbing state 2 may involve an intermediate state 1 (e.g., disease relapse). The impact of the transition into state 1 on the subsequent transition hazard to state 2 enables insight to be gained into the disease evolution. The standard approach of analysis is modeling the transition hazards from 0 to 2 and from 1 to 2, including time to illness as a time-varying covariate and measuring time from origin even after transition into state 1. The hazard from 1 to 2 can be also modeled separately using only patients in state 1, measuring time from illness and including time to illness as a fixed covariate. A recently proposed approach is a model where time after the transition into state 1 is measured in both scales and time to illness is included as a time-varying covariate. Another possibility is a model where time after transition into state 1 is measured only from illness and time to illness is included as a fixed covariate. Through theoretical reasoning and simulation protocols, we discuss the use of these models and we develop a practical strategy aiming to (a) validate the properties of the illness-death process, (b) estimate the impact of time to illness on the hazard from state 1 to 2, and (c) quantify the impact that the transition into state 1 has on the hazard of the absorbing state. The strategy is also applied to a literature dataset on diabetes.  相似文献

Consciousness is expected to have a specific temporal dynamics. The COrollary Discharge of Attention Movement (CODAM) model of consciousness is deduced from an engineering approach to attention and motor attention. This model is briefly described, as is support arising from brain dynamics, especially that for the attentional blink. The understanding of known temporal dynamics in the brain associated with the emergence of consciousness is then developed from CODAM, and specifically related to the N2 ERP brain signal. How the pre-reflective self, as content-free, interacts with the content of experience is discussed in terms of the possibility that such experience arises from some proto-self generated by body signals; experiments are described which indicate that no pre-reflective self based on body signals is observable. Only a content-free pre-reflective self is consistent with this data, as CODAM suggests. How such a pre-reflective self can be further fused to give temporal continuity of a sense of self is considered in terms of various mechanisms which could be present for preserving the sense of self. The observation of the N2 signal in hippocampal encoding is proposed as providing a justification for the encoding of the N2–P3 sequence of brain signals. This would correspond to episodic encoding of the sequence of experiences of the pre-reflective self; this will thereby provide the necessary control signals in time so that ‘I’ is experienced as part of the retrieval of such memories.  相似文献

In well-characterised species of the Rhodobacter (Rba.) genus of purple photosynthetic bacteria it is known that the photochemical reaction centre (RC) is intimately-associated with an encircling LH1 antenna pigment protein, and this LH1 antenna is prevented from completely surrounding the RC by a single copy of the PufX protein. In Rba. veldkampii only monomeric RC-LH1 complexes are assembled in the photosynthetic membrane, whereas in Rba. sphaeroides and Rba. blasticus a dimeric form is also assembled in which two RCs are surrounded by an S-shaped LH1 antenna. The present work established that dimeric RC-LH1 complexes can also be isolated from Rba. azotoformans and Rba. changlensis, but not from Rba. capsulatus or Rba. vinaykumarii. The compositions of the monomers and dimers isolated from these four species of Rhodobacter were similar to those of the well-characterised RC-LH1 complexes present in Rba. sphaeroides. Pigment proteins were also isolated from strains of Rba. sphaeroides expressing chimeric RC-LH1 complexes. Replacement of either the Rba. sphaeroides LH1 antenna or PufX with its counterpart from Rba. capsulatus led to a loss of the dimeric form of the RC-LH1 complex, but the monomeric form had a largely unaltered composition, even in strains in which the expression level of LH1 relative to the RC was reduced. The chimeric RC-LH1 complexes were also functional, supporting bacterial growth under photosynthetic conditions. The findings help to tease apart the different functions of PufX in different species of Rhodobacter, and a specific protein structural arrangement that allows PufX to fulfil these three functions is proposed.  相似文献

1. Incubation of intact epididymal adipose tissue from fed rats at 37 degrees in an albumin solution at pH7.4 in vitro results in rapid loss of clearing-factor lipase activity until a low activity, stable to prolonged incubation, is attained. The clearing-factor lipase activity of intact tissue from starved rats, which is initially much less than that of tissue from fed rats, is mainly stable to incubation at 37 degrees . 2. Much of the clearing-factor lipase activity of intact epididymal adipose tissue from fed rats is inactivated by collagenase. The enzyme activity of intact tissue from starved rats is not inactivated by collagenase. 3. The clearing-factor lipase activity of fat cells isolated from the epididymal adipose tissue of fed rats is stable to prolonged incubation at 37 degrees . It represents only a small proportion of the total activity of the intact tissue. In starved rats, the isolated fat cells contain a much higher proportion of the activity of the intact tissue. Their activity is also stable at 37 degrees . 4. Incubation of isolated fat cells in a serum-based medium leads to a progressive rise in clearing-factor lipase activity. Actinomycin increases the extent of this rise in activity. No rise in clearing-factor lipase activity occurs when stromal-vascular cells isolated from epididymal adipose tissue are incubated in the medium. 5. The findings indicate that less than 20% of the activity of intact adipose tissue from fed rats is retained when fat cells are isolated from the tissue by collagenase treatment. The activity that is lost could be that which normally functions in the uptake of triglyceride fatty acids by the tissue.  相似文献

Fragments of the anterior part of a mandibular rostrum and a maxillary bone of the plesiosaur Polycotylidae indet. from the Karyakino and Beloe Ozero localities of the Saratov Region (Upper Cretaceous, Campanian) are described. The specimens belong to a relatively long-snouted polycotylid with a mandibular symphysis incorporating 13 pairs of teeth. A unique feature of this plesiosaur is the fact that, in the symphysis, the splenial extends to the very anterior end of the rostrum. The polycotylid from the Campanian of the Saratov Region is probably closely related or identical to Georgiasaurus from the Santonian of the Penza Region, in which structural details of the mandibular symphysis remain uncertain.  相似文献

A lower molar of Sulestes sp., a deltatheridiid from the Coniacian of Uzbekistan, is described. The Deltatheroididae new fam. is erected. The Deltatheroididae and Deltatheridiidae are assigned to the order Deltatheroida Kielan-Jaworowska 1982 and it is argued that the Deltatheroida are metatherians, a sister group of Marsupialia. The Deltatheroida have at least an incipient alisphenoid bulla, three premolars sharply differentiated morphologically from four molars, a very large stylar area and the protoconid higher than the metaconid. They differ from known Cretaceous marsupials in having a tendency to lose the last molar, in having a small protocone, a narrow and short talonid, and a hypoconulid and entoconid not approximated. Known deltatheroidan taxa are compared. Relationships of the Deltatheroida and Aegialodontia are discussed. The gam gene of bacteriophage Mu encodes a protein which protects linear double stranded DNA from exonuclease degradation in vitro and in vivo. We purified the Mu gam gene product to apparent homogeneity from cells in which it is over-produced from a plasmid clone. The purified protein is a dimer of identical subunits of 18.9 kd. It can aggregate DNA into large, rapidly sedimenting complexes and is a potent exonuclease inhibitor when bound to DNA. The N-terminal amino acid sequence of the purified protein was determined by automated degradation and the nucleotide sequence of the Mu gam gene is presented to accurately map its position in the Mu genome.  相似文献

This study determined the optimum number of tubules to be counted per testis cross section, and the number of animals per treatment group, when changes in stage frequencies in the cycle of the seminiferous epithelium are criteria for assessing effects of treatment on spermatogenesis. A data base of 9,672 observed and staged tubules was collected from testicular cross sections of 15 Sprague-Dawley rats. A significant variation between animals was found for the frequencies of Stages I, II, IV, VI, VIII, and XIII. Computer simulation was used to randomly select different combinations of animal and tubule numbers from the observed data. Stage frequency means from each simulation experiment were compared statistically to observed mean frequencies. A model that used data from all 14 stages was analyzed. The following conclusions were made: a) a minimum of 200 tubule cross sections/testis is recommended for estimating stage frequencies; b) for a fixed number of tubules scored, the number of animals sampled is more important than the number of tubules per animal in reducing variance; c) to detect a difference of 2 standard deviations from the mean with a 2% error rate and examining 200 tubules/testis, at least 12 animals must be used per group when assessing all 14 stages; d) when individual stages are examined using 10 animals per group, only Stage VII has 80% or greater power of test (alpha = 0.05) to detect a frequency difference; e) pooling stages into 3-4 groups is recommended to improve the power of detecting a treatment difference.  相似文献

Zoogeography of the coral reef fishes of the Socotra Archipelago   总被引:1,自引:0,他引:1  
Fish communities and habitats were studied at the Socotra archipelago (Gulf of Aden, ≈12°N 54°E). Extensive and unexpected hermatypic coral communities were recorded, at the centre of a 2200 km gap in knowledge of species and habitat distributions which coincides with a change from a western Indian Ocean coral reef fauna to an Arabian one. The fish assemblage associated with the Socotra archipelago corals is predominantly south Arabian. An east African influence, minimal on the mainland coasts of Arabia, is more evident here, and results in previously unrecorded sympatry between Arabian endemic species and their Indian Ocean sister taxa. A study of distributions of Chaetodontidae (butterflyfishes) in the north-western Indian Ocean reveals a number of distinct patterns, with a trend for species replacement along a track from the northern Red Sea to the Indian Ocean. A major feature of the reef fish zoogeography of the region is found to be a distinct south Arabian area, characterized by a 'pseudo-high latitude effect' which results from seasonal cold water upwelling along the Arabian sea coasts of Yemen and Oman and the Indian Ocean coast of Somalia. This south Arabian feature is consistent across a wide range of fish families. It is most pronounced in Oman and Yemen, and although it is the dominant influence at Socotra it is slightly 'diluted' here by the east African influence. The south Arabian area wholly or partly accounts for most of the major marine zoogeographic features around Arabia, and is the principal feature fragmenting Arabian coastal fish assemblages, and separating them from those of the wider Indo-west Pacific.  相似文献

The Pleistocene glacial cycles resulted in significant changes in species distributions, and it has been discussed whether this caused increased rates of population divergence and speciation. One species that is likely to have evolved during the Pleistocene is the Norwegian lemming (Lemmus lemmus). However, the origin of this species, both in terms of when and from what ancestral taxon it evolved, has been difficult to ascertain. Here, we use ancient DNA recovered from lemming remains from a series of Late Pleistocene and Holocene sites to explore the species' evolutionary history. The results revealed considerable genetic differentiation between glacial and contemporary samples. Moreover, the analyses provided strong support for a divergence time prior to the Last Glacial Maximum (LGM), therefore likely ruling out a postglacial colonization of Scandinavia. Consequently, it appears that the Norwegian lemming evolved from a small population that survived the LGM in an ice‐free Scandinavian refugium.  相似文献

Contemporary pollen flow in forest plant species is measured by the probability of paternal identity (PPI) for two randomly sampled offspring, drawn from a single female, and contrasting that with PPI for two random offspring, drawn from different females. Two different estimation strategies have emerged: (a) an indirect approach, using the 'genetic structure' of the pollen received by different mothers and (b) a direct approach, based on parentage analysis. The indirect strategy is somewhat limited by the assumptions, but is widely useful. The direct approach is most appropriate where a large majority of the true fathers can be identified exactly, which is sometimes possible with high-resolution SSR markers. Using the parentage approach, we develop estimates of PPI, showing that the obvious estimates are severely biased, and providing an unbiased alternative. We then illustrate the methods with SSR data from a 36-tree isolated population of Pinus sylvestris from the Meseta region of Spain, for which categorical paternity assignment was available for over 95% of offspring. For all the females combined, we estimate that PPI=0.0425, indicating uneven male reproductive contributions. Different (but overlapping) arrays of males pollinate different females, and for the average female, PPI=0.317, indicating substantial 'pollen structure' for the population. We also relate the direct measures of PPI to those available from indirect approaches, and show that they are generally comparable.  相似文献

Following a blood meal that initiates oöcyte development, the host-seeking behaviour of Aedes aegypti mosquitoes is inhibited by a haemolymph-borne factor that is released in response to a humoral signal from a vitellogenic ovary. This inhibition is accompanied by a decrease in the sensitivity of the peripheral lactic acid receptors. Implantation of corpora allata, medial neurosecretory cells, or terminal abdominal ganglia from blood-fed donors could not induce the inhibition in sugar-fed recipients. However, fat body transplanted from blood-fed into sugar-fed females suppressed host-seeking behaviour as well as the sensitivity of lactic acid receptors, suggesting that the source of the behavioural inhibitor is the fat body. Resting-stage ovaries from other mosquito species inhibited host-seeking after the A. aegypti host was fed on blood only if the fat body was activated by the donor ovary.  相似文献

Using a deletion approach on the alpha subunit of DNA polymerase III from Escherichia coli, we show that there is an N-proximal polymerase domain which is distinct from a more C-proximal tau and beta binding domain. Although deletion of 60 residues from the alpha N terminus abolishes polymerase activity, deletions of 48, 169, and 342 amino acids from the C terminus progressively impair its catalytic efficiency but preserve an active site. Deletion of 342 C-terminal residues reduces k(cat) 46-fold, increases the Km for gapped DNA 5.5-fold, and increases the Km for deoxynucleoside triphosphates (dNTPs) twofold. The 818-residue protein with polymerase activity displays typical Michaelis-Menten behavior, catalyzing a polymerase reaction that is saturable with substrate and linear with time. With the aid of newly acquired sequences of the polymerase III alpha subunit from a variety of organisms, candidates for two key aspartate residues in the active site are identified at amino acids 401 and 403 of the E. coli sequence by inspection of conserved acidic amino acids. The motif Pro-Asp-X-Asp, where X is a hydrophobic amino acid, is shown to be conserved among all known DnaE proteins, including those from Bacillaceae, cyanobacteria, Mycoplasma, and mycobacteria. The E. coli DnaE deletion protein with only the N-terminal 366 amino acids does not have polymerase activity, consistent with the proposed position of the active-site residues.  相似文献

The iron-sulphur cluster-free hydrogenase (Hmd, EC 1.12.98.2) from methanogenic archaea is a novel type of hydrogenase that tightly binds an iron-containing cofactor. The iron is coordinated by two CO molecules, one sulphur and a pyridone derivative, which is linked via a phosphodiester bond to a guanosine base. We report here on the crystal structure of the Hmd apoenzyme from Methanocaldococcus jannaschii at 1.75 A and from Methanopyrus kandleri at 2.4 A resolution. Homodimeric Hmd reveals a unique architecture composed of one central and two identical peripheral globular units. The central unit is composed of the intertwined C-terminal segments of both subunits, forming a novel intersubunit fold. The two peripheral units consist of the N-terminal domain of each subunit. The Rossmann fold-like structure of the N-terminal domain contains a mononucleotide-binding site, which could harbour the GMP moiety of the cofactor. Another binding site for the iron-containing cofactor is most probably Cys176, which is located at the bottom of a deep intersubunit cleft and which has been shown to be essential for enzyme activity. Adjacent to the iron of the cofactor modelled as a ligand to Cys176, an extended U-shaped extra electron density, interpreted as a polyethyleneglycol fragment, suggests a binding site for the substrate methenyltetrahydromethanopterin.  相似文献

The Eurasian bullfinch spermatozoon differs from typical passeridan spermatozoa in several major respects. The mature acrosome consists of a concavo‐convex vesicle differing from the typical passeridan acrosome, which is a helical structure, is usually longer than the nucleus and has a prominent helical keel. The nucleus differs from that of other oscines in not showing a twisted cylindrical form, in being shorter, and in tending to be an elongate ellipsoid in shape. The chromatin often appears in an uncondensed form reminiscent of a spermatid and consists of discrete fascicles. A small proportion of the mature sperm population however, is characterized by marked chromatin condensation. The midpiece comprises a small group of mitochondria clustered around the nuclear–axonemal junction in contrast to the single, long mitochondrion wound helically around the axoneme that is found in typical Passerida. The presence of a proximal centriole (in addition to the distal one) is a notable difference from all other oscine passerines. We suggest that the unusual morphology of the Eurasian bullfinch spermatozoon, resembling that of a spermatid, is the result of the progressive suppression of the final stages of spermiogenesis and is associated with the likelihood that sperm competition is infrequent in this species.  相似文献

In striated muscle the mechanism of contraction involves the cooperative movement of contractile and elastic components. This review emphasizes a structural approach that describes the cellular and extracellular components with known anatomical, biochemical, and physical properties that make them candidates for these contractile and elastic components. Classical models of contractile and elastic elements and their underlying assumptions are presented. Mechanical properties of cardiac and skeletal muscle are compared and contrasted and then related to ultrastructure. Information from these approaches leads to the conclusion that the Z band is essential for muscle contraction. Our review of Z band structure shows the Z band at the interface where extracellular components meet the cell surface. The Z band is also the interface from cell surface to myofibril, from extra-myofibrillar to myofibril, and finally from sarcomere to sarcomere. Our studies of Z band in defined physiologic states show that this lattice is an integral part of the contractile elements and can function as an elastic component. The Z band is a complex dynamic lattice uniquely suited to play several roles in muscle contraction.  相似文献
