Antifreeze proteins (AFPs), characterized by their ability to separate the melting and growth temperatures of ice and to inhibit ice recrystallization, play an important role in cold adaptation of several polar and cold-tolerant organisms. Recently, a multigene family of AFP genes was found in the diatom Fragilariopsis cylindrus, a dominant species within polar sea ice assemblages. This study presents the AFP from F. cylindrus set in a molecular and crystallographic frame. Differential protein expression after exposure of the diatoms to environmentally relevant conditions underlined the importance of certain AFP isoforms in response to cold. Analyses of the recombinant AFP showed freezing point depression comparable to the activity of other moderate AFPs and further enhanced by salt (up to 0.9 °C in low salinity buffer, 2.5 °C at high salinity). However, unlike other moderate AFPs, its fastest growth direction is perpendicular to the c-axis. The protein also caused strong inhibition of recrystallization at concentrations of 1.2 and 0.12 μM at low and high salinity, respectively. Observations of crystal habit modifications and pitting activity suggested binding of AFPs to multiple faces of the ice crystals. Further analyses showed striations caused by AFPs, interpreted as inclusion in the ice. We suggest that the influence on ice microstructure is the main characteristic of these AFPs in sea ice. 相似文献

Home range size estimates are often used to assess the amount of space required for animals to perform the activities essential for their survival and reproduction. However, in moving environments, traditional home range estimates may be ill suited to this task. In particular, traditional home range estimates are inaccurate representations of the space required by polar bears Ursus maritimus. The sea ice is the prime foraging platform of polar bears, and estimating the amount of ice encountered by bears may provide a better approximation of space use. We develop a technique to make these estimates. Our results confirm that polar bears use more space than terrestrial carnivores to find the resources and conditions they require. We also show that the traditional geographic home range can underestimate both the movement of bears and the amount of space encountered. Moreover, area of ice encountered increased with ice drift, indicating that bears living on highly mobile ice might be exposed to higher energetic costs, and potentially larger energetic gains, than bears inhabiting more stable ice. The methods and concepts presented here can serve as a foundation for new approaches to study the space use of the many species living in moving environments. 相似文献

Antifreeze proteins (AFPs) protect many plants and organisms from freezing in low temperatures. Of the different AFPs, the most studied AFP Type I from winter flounder is used in the current computational studies to gain molecular insight into its adsorption at the ice/water interface. Employing molecular dynamics simulations, we calculate the free energy difference between the hydrophilic and hydrophobic faces of the protein interacting with ice. Furthermore, we identify three properties of Type I "antifreeze" proteins that discriminate among these two orientations of the protein at the ice/water interface. The three properties are: the "surface area" of the protein; a measure of the interaction of the protein with neighboring water molecules as determined by the number of hydrogen bond count, for example; and the side-chain orientation angles of the threonine residues. All three discriminants are consistent with our free energy results, which clearly show that the hydrophilic protein face orientations toward the ice/water interface, as hypothesized from experimental and ice/vacuum simulations, are incorrect and support the hypothesis that the hydrophobic face is oriented toward the ice/water interface. While global climate change in polar regions is expected to cause significant warming, the annual cycle of light and dark
will remain unchanged. Cultures of three species of Antarctic sea ice diatoms, Fragilariopsis cylindrus (Grunow) Krieger, Thalassiosira antarctica Comber and Entomoneis kjellmanii (P.T. Cleve) Poulin and Cardinal, were incubated in the dark and exposed to differing temperatures. Maximum dark survival times varied between 30 and 60 days.
Photosynthetic parameters, photosynthetic efficiency (α), maximum quantum yield (Fv/Fm), maximum relative electron transport
rate (rETRmax) and non-photochemical quenching (NPQ), showed that dark exposure had a significant impact on photoacclimation.
In contrast, elevated temperatures had a relatively minor impact on photosynthetic functioning during the dark exposure period
but had a considerable impact on dark survival with minimal dark survival times reduced to only 7 days when exposed to 10°C.
Recovery of maximum quantum yield of fluorescence (Fv/Fm) was not significantly impacted by temperature, species or dark exposure
length. Recovery rates of Fv/Fm ranged from −5.06E−7 ± 2.71E−7 s−1 to 1.36E−5 ± 1.53E−5 s−1 for monthly experiments and from −9.63E−7 ± 7.71E−7 s−1 to 2.65E−5 ± 2.97E−5 s−1 for weekly experiments. NPQ recovery was greater and more consistent than Fv/Fm recovery, ranging between 5.74E−7 ± 8.11E−7 s−1 to 7.50E−3 ± 7.1E−4 s−1. The concentration of chl-a and monosaccharides remained relatively constant in both experiments. These results suggest that there will probably be little

Recent reductions in thickness and extent have increased drift rates of Arctic sea ice. Increased ice drift could significantly affect the movements and the energy balance of polar bears (Ursus maritimus) which forage, nearly exclusively, on this substrate. We used radio‐tracking and ice drift data to quantify the influence of increased drift on bear movements, and we modeled the consequences for energy demands of adult females in the Beaufort and Chukchi seas during two periods with different sea ice characteristics. Westward and northward drift of the sea ice used by polar bears in both regions increased between 1987–1998 and 1999–2013. To remain within their home ranges, polar bears responded to the higher westward ice drift with greater eastward movements, while their movements north in the spring and south in fall were frequently aided by ice motion. To compensate for more rapid westward ice drift in recent years, polar bears covered greater daily distances either by increasing their time spent active (7.6%–9.6%) or by increasing their travel speed (8.5%–8.9%). This increased their calculated annual energy expenditure by 1.8%–3.6% (depending on region and reproductive status), a cost that could be met by capturing an additional 1–3 seals/year. Polar bears selected similar habitats in both periods, indicating that faster drift did not alter habitat preferences. Compounding reduced foraging opportunities that result from habitat loss; changes in ice drift, and associated activity increases, likely exacerbate the physiological stress experienced by polar bears in a warming Arctic. 相似文献

Low photosynthetic active radiation is a strong determinant in the development and growth of sea ice algae. The algae appear to have universal mechanisms to overcome light limitation. One important process, which is induced under light limitation, is the desaturation of chloroplast membrane lipids. In order to discover whether this process is universally valid in sea ice diatoms, we investigated three species coexisting in chemostats illuminated with 15 and 2 micromol photons m(-2) s(-1) at -1 degrees C. Growth under 2 micromol photons m(-2) s(-1) caused a 50% increase in monogalactosyldiacylglycerols (MGDG) thylakoid membrane related 20:5 n-3 fatty acids. This fatty acid supports the fluidity of the thylakoid membrane and therefore the velocity of electron flow, which is indicated by increasing rate constants for the electron transport between Q(A) (first stable electron acceptor) and bound Q(B) (second stable electron acceptor) (11.16 +/- 1.34 to 23.24 +/- 1.35 relative units). Two micromol photons m(-2) s(-1) furthermore resulted in higher amounts of non-lipid bilayer forming MGDG in relation to other bilayer forming lipids, especially digalactosydiacylglycerol (DGDG). The ratio of MGDG:DGDG increased from 3.4 +/- 0.3 to 5.7 +/- 0.3. The existence of bilayer thylakoid membranes with high proportions of non. bilayer forming lipids is only possible when sufficient thylakoid pigment-protein complexes are present. If more thylakoid pigment-protein complexes are present in membranes, as found under extreme light limitation, less bilayer forming lipids such as DGDG are required to stabilize the bilayer structure. Differences in protein contents between both light intensities were not found. Consequently pigment contents which nearly doubled under 2 micromol photons m(-2) s(-1) must be responsible in balancing the potential stability loss resulting from an increase in MGDG:DGDG ratio. 相似文献

Sexual reproduction is commonly assumed to occur in the vast majority of diatoms due to the intimate association of this process with cell size control. Surprisingly, however, little is known about the impact of sexual events on diatom population dynamics. The Sig1 gene is strongly upregulated during sexual reproduction in the centric diatom Thalassiosira weissflogii and has been hypothesized to encode a protein involved in gamete recognition. In the present study, degenerate PCR primers were designed and used to amplify a portion of Sig1 from three closely related species in the cosmopolitan genus Thalassiosira, Thalassiosira oceanica, Thalassiosira guillardii, and Thalassiosira pseudonana. Identification of Sig1 in these three additional species facilitated development of this gene as a molecular marker for diatom sexual events. Examination of the new sequences indicated that multiple copies of Sig1 are probably present in the genome. Moreover, compared to the housekeeping gene beta-tubulin, the Sig1 genes of isolates of T. weissflogii collected from different regions of the Atlantic and Pacific oceans displayed high levels of divergence. The Sig1 genes of the four closely related Thalassiosira species also displayed high levels of sequence divergence compared to the levels observed with a second gene, Fcp, probably explaining why Sig1 could not be amplified from more distantly related species. The high levels of sequence divergence both within and between species suggest that Sig1 is rapidly evolving in a manner reminiscent of the manner observed in other genes that encode gamete recognition proteins. A simple model is presented for Sig1 evolution and the implications of such a rapidly evolving sexual reproduction gene for diatom speciation and population dynamics. 相似文献

Fragilariopsis species composition and abundance from the Argentine Sea and Antarctic waters were analyzed using light and electron microscopy.
Twelve species (F. curta, F. cylindrus, F. kerguelensis, F. nana, F. obliquecostata, F. peragallii, F. pseudonana, F. rhombica, F. ritscheri, F. separanda, F. sublinearis and F. vanheurckii) are described and compared with samples from the Frenguelli Collection, Museo de La Plata, Argentina. F. peragallii was examined for the first time using electron microscopy, and F. pseudonana was recorded for the first time in Argentinean shelf waters. New information on the girdle view is included, except for the
species F. curta, F. cylindrus and F. nana, for which information already existed. In the Argentine Sea, F. pseudonana was the most abundant Fragilariopsis species, and in Antarctic waters, F. curta was most abundant. Of the twelve species of Fragilariopsis documented, four occurred in the Argentine Sea, nine in the Drake Passage and twelve in the Weddell Sea. Future sea ice conditions in Western Hudson Bay and consequences for polar bears in the 21st century

The primary habitat of polar bears is sea ice, but in Western Hudson Bay (WH), the seasonal ice cycle forces polar bears ashore each summer. Survival of bears on land in WH is correlated with breakup and the ice‐free season length, and studies suggest that exceeding thresholds in these variables will lead to large declines in the WH population. To estimate when anthropogenic warming may have progressed sufficiently to threaten the persistence of polar bears in WH, we predict changes in the ice cycle and the sea ice concentration (SIC) in spring (the primary feeding period of polar bears) with a high‐resolution sea ice‐ocean model and warming forced with 21st century IPCC greenhouse gas (GHG) emission scenarios: B1 (low), A1B (medium), and A2 (high). We define critical years for polar bears based on proposed thresholds in breakup and ice‐free season and we assess when ice‐cycle conditions cross these thresholds. In the three scenarios, critical years occur more commonly after 2050. From 2001 to 2050, 2 critical years occur under B1 and A2, and 4 under A1B; from 2051 to 2100, 8 critical years occur under B1, 35 under A1B and 41 under A2. Spring SIC in WH is high (>90%) in all three scenarios between 2001 and 2050, but declines rapidly after 2050 in A1B and A2. From 2090 to 2100, the mean spring SIC is 84 (±7)% in B1, 56 (±26)% in A1B and 20 (±13)% in A2. Our predictions suggest that the habitat of polar bears in WH will deteriorate in the 21st century. Ice predictions in A1B and A2 suggest that the polar bear population may struggle to persist after ca. 2050. Predictions under B1 suggest that reducing GHG emissions could allow polar bears to persist in WH throughout the 21st century. 相似文献

Prior to this study, antifreeze proteins (AFPs) had not been identified in terrestrial arthropods from the Arctic or anywhere in Alaska. The hemolymph of 75 species of insects and six spiders from interior and arctic Alaska were screened for thermal hysteresis (a difference between the freezing and melting points), characteristic of the presence of AFPs. Eighteen species of insects and three spiders were shown to have AFPs. Ten of the insects with AFPs were beetles including the first species from the families Chrysomelidae, Pythidae, Silphidae and Carabidae. In addition, the first Neuropteran to have AFPs was identified, the lacewing Hemerobius simulans together with the second and third Diptera (the first Tipulids) and the second and third Hemiptera, the stinkbug Elasmostethus interstinctus (the first Pentatomid), and the water strider Limnoporus dissortis (the first Gerrid). Prior to this study, 33 species of insects and three spiders had been reported to have AFPs. Most AFP-producing terrestrial arthropods are freeze avoiding, and the AFPs function to prevent freezing. However, some of the AFP- producing insects identified in this study are known to be freeze tolerant (able to survive freezing) to very low temperatures (-40 to -70 degrees C). 相似文献
