Twenty-four axenic strains of planktonic Anabaena with straight trichomes, assigned to 7 species, were investigated by analyzing the pattern and content of their fatty acid composition and comparing their fatty acid composition with their morphological properties. In general, the fatty acids in planktonic Anabaena contained 14:0, 16:0, 16:1(cis-), 18:0, 18:1, 18:2, and 18:3(alpha) as their major components, and were classified as Type 2 according to the Kenyon-Murata system. These strains were further divided into 2 subtypes: 18 strains belonging to Type 2A, which contains 16:2 and 16:3, and 6 strains to Type 2B, which lacks 16:2 and 16:3. Fatty acid compositions of strains of A. solitaria, A. smithii, and A. kisseleviana closely corresponded to morphological properties; however, 10 strains of A. planctonica were divided into 4 clusters, and 3 strains of A. affinis into 2 clusters. These clusters should be taxonomically evaluated based on other aspects such as genetic characteristics.  相似文献

对毛壳属25个种50个菌株的最低生长温度、最适生长温度、最高生长温度、致死温度以及在最适生长温度下的菌落日生长速率进行了系统比较。结果表明,毛壳属不同的物种具有稳定的特征性最高生长温度,这一温度指标可作为该属物种鉴定,特别是区分形态相似种的重要生理性状;其他温度指标在种内变异大,在种间存在明显交叉,不具分类学价值。在此基础上,测定了42个种的最高生长温度,为毛壳属物种鉴定提供了一个重要的生理性状指标。  相似文献

DNA base compositions (GC content) ofSaccharomyces species are reported and discussed. Several amendments of the four groups given by van der Walt are suggested, viz. the transfer ofS. kluyveri to group 1, and ofS. eupagycus, S. cidri, S. montanus, S. microellipsodes andS. florentinus to group 2. The synonomy ofS. amurcae andS. cidri is suggested. The DNA base compositions revealed two possible pairs of sibling species:S. elegans andS. bailii, with a difference in GC content of 4.1%;S. dairensis andS. servazzii with a difference in GC content of ca. 3%.S. mrakii had a GC content of 47.3–48.5% the highest encountered in this genus and similar to that ofKluyveromyces thermotolerans.  相似文献

The nuclear DNa of 28 species (30 strains investigated) of yeasts classified currently or previously in the genus Trichosporon. was analysed for its molar percentage of guanine + cytosine (mol% G+C). This criterion, together with biochemical characteristics, suggested the separation of the organisms studied into two groups. The first group, which appears related to the Ascomycetes, includes thirteen species with a G+C content lower than 50 mol% (34.7–48.8), and lacks urease (except T. margaritiferum). The second group appears related to the Basidiomycetes and includes fifteen species with a G+C content higher than 50 mol% (57–64) and has the ability to hydrolyse urea.A DNA homology experiment with T. beigelii and twelve other species of the second group showed very low values of complementarity with T. beigelii-labeled DNA. All these species must be considered as taxa other than T. beigelii.  相似文献

Molybdenum deficiency and accumulation was examined in Anabaena oscillarioides (Cyanobacteria). Molybdenum deficiency was induced by: 1) culturing the cyanobacterium on modified Chu-10 (-N) medium containing 4–5 ng Mo · l−1, or 2) adding tungsten to reversibly inactivate dinitrogenase. Stimulation of dinitrogenase activity, while heterocyst frequencies were decreasing, occurred in the range of 5–40 ng · l−1 of added Mo. Molybdenum deficient A. oscillarioides was able to deplete Mo in the medium. This ability was rapidly lost at higher concentrations of added Mo when this cyanobacterium started to accumulate Mo. These results are of potential use in predicting potential Mo limitation in natural environments.  相似文献

Urease fromAnabaena doliolum andAnacystic nidulans showed maximum activity at pH 7.0–7.4 at 40°C when measured in cell-free, phosphate-buffered extracts. It is a soluble enzyme located in cytoplasm. The apparent Km forA. doliolum urease was 120 M. Anacystis nidulans urease exhibited biphasic kinetics (Km=250 M and 1.66 mM). Enzyme, fully expressed in cells grown with urea, nitrate, or N2, was repressed in ammonia-grown cells, but ammonia did not inhibit the activity in vitro. Incubation of algal cells in N2 medium with chloramphenicol for 12 h caused degradation of urease. The DNA base compositions of 60 strains ofScenedesmus were determined and found to be a valuable indicator for the differentiation within this genus (except for the very closely related species of the subsect.Desmodesmus). The range for allScenedesmus species was 49.9–69.3 mol% GC for nuclear DNA and 36.8–39.9 mol% GC for chloroplast DNA. The separation of the genusTetradesmus cannot be verified by GC values, becauseS. obliquus andS. (Tetradesmus)wisconsinensis have a similar GC content.S. (Chlorella)ultrasquamata, S. costato-granulatus (sect.Costato-granulati) andS. lunatus are separated clearly from all other species of the genus because of their high GC content.  相似文献

The taxonomic study of 14 strains of Geitlerinema amphibium (Ag. ex Gom.) Anagnostidis and Geitlerinema unigranulatum (R.N. Singh) Komárek and Azevedo, coming from several localities was undertaken. Use was made of morphological data and molecular data were obtained by means of the DNA fingerprinting technique using highly iterated palindrome (HIP1) sequences. The employed morphological characteristics were those used for species taxonomic identification belonging to the Geitlerinema genus, namely, cell dimensions, shape of the apical cell, motility, number and localization of cyanophycin granules in the cell. The two species revealed as polymorphic were discriminated only by means of the average cellular diameters. In spite of this, minima and maxima values of the cellular diameters overlapped. It was found from molecular analysis that a high genetic diversity and the formation of two clusters consisted of G. amphibium and G. unigranulatum, plus a sole strain keeping itself isolated from the remaining. Also, these clusters were not related to the geographic location; they encompassed strains from water bodies distant from each other by as much as 3500 km, or Brazilian and Spanish strains. Molecular and morphological data support the possibility that G. unigranulatum could be considered a synonym for G. amphibium. HIP1 fingerprinting is a powerful tool for the study of genetic of cyanobacteria closely related taxa. This study points to the necessity of using other than morphological data in the taxonomic revision of cyanobacteria, as well as in the proposition of new taxons.  相似文献

Questions of biogeography of freshwater cyanobacteria and their ability to colonize new areas have been recently discussed in connection with increasing occurrence of some formerly rare morphospecies in temperate zones. Nevertheless, the general knowledge about the distribution of cyanobacterial species is still fragmentary, and any new findings on cyanobacterial biogeography and spread are valuable. In this study, we provide updated information on the occurrence of Anabaena bergii, Raphidiopsis mediterranea, and Sphaerospermopsis aphanizomenoides in the Czech Republic. In addition, more nostocacean morphospecies are newly reported from the Czech Republic (A. fusca, A. tenericaulis, Dolichospermum curvum, D. mucosum, and S. reniformis). All of these morphospecies were characterized from a morphological point of view, and their phylogenetic affiliations were assessed on the basis of their 16S rRNA gene sequences. Based on these results, Anabaena bergii was reclassified into Chrysosporum gen. nov., and D. tenericaule comb. nova was established.  相似文献

The taxonomic status ofSedum Telephium and its allied species (Crassulaceae-Sedoideae) is discussed, and a new genus,Hylotelephium, is recognized. Under the genus, 27 new combinations are proposed and one new species,H. callichromum, from Central Asia is described. Bioactive products of the multicellular filamentous benthic cyanobacterium, Pseudanabaena species, isolated from wastewater stabilization ponds at Marrakech, were tested against some pathogenic micro-organisms. Extracellular and intracellular products released by this blue-green alga in the stationary, growth phase, reduced the survival of Escherichia coli, Salmonella sp., Staphylococcus aureus and Candida albicans, and stimulated the growth of non-O1 V. cholerae, whereas no significant effect was noted on the growth of Candida tropicalis. Intracellular substances (methanolic extract) had a stronger inhibitory effect on these micro-organisms than that produced by extracellular substances. These antibacterial and antifungal substances may have a pharmacological value. They may have an important ecological effect on the composition of the bacterial community in Marrakech stabilization ponds when blooms of Pseudanabaena species occurred in this sewage treatment ecosystem during hot periods.  相似文献

Similar DNA homology values were recorded when a modified S1 nuclease technique and a standard nitrocellulose membrane filter method were applied to representative strains of Rhodococcus. The DNA homology data showed that R. globerulus, R. luteus and R. sputi form distinct genomic species. The congruence between the DNA homology and earlier numerical phenetic data was good, but there was evidence that some strains had been misclassified in the previous studies. In particular, the type strains of R. obuensis and R. sputi belong to a single genomic species. The former name is thus a later, subjective synonym of the latter. The guanine plus cytosine content of the DNA of the rhodococci fell within the range 61 to 72 mol%.  相似文献

The restriction site and size variation of five PCR amplified fragments of noncoding chloroplast DNA (cpDNA) was examined in material from 13 populations ofPanax from Nepal and China. Fourteen restriction endonucleases produced 81 restriction site and length variations from the large single-copy region of cpDNA, 27 of which are polymorphic. The cpDNA dataset suggests two distinct groups ofPanax from Nepal (clades I and II). Clade I consists of two populations ofP. pseudoginseng subsp.pseudoginseng, and clade II is composed of material referable toP. pseudogingeng subsp.himalaicus (vars.himalaicus, angustifolius, andbipinnatifidus). The three accessions ofP. pseudoginseng subsp.japonicus andP. ginseng studied from China had cpDNA characters that differed from the HimalayanPanax. The highly distinctive cpDNA profile and morphology ofP. pseudoginseng subsp.pseudoginseng sensu Hara (1970) from central Nepal support its status as a separate species, which has an extremely restricted distribution.  相似文献

The acetolysed pollen of 33 species of the genus Justicia in Thailand is investigated using both light and scanning electron microscopy. The pollen of the genus is characterised as being isopolar, bilaterally or radially symmetrical, with mostly prolate or prolate spheroidal shape. Based on characters with high factor loading in the cluster analysis and principal components analysis, the studied species are divided into two major groups; one with 3-colporate with six pseudocolpi and the other with 2-porate or 2–3-colporate with aperture areas. The pollen morphology of each group is described and illustrated. Pollen characters are useful for taxonomic delimitation and relationships among Thai species.  相似文献

The quinacrine banding patterns of chromosomes of Dipodomys ordii and Mus musculus are described. Satellite and mainband DNA fractions from D. ordii and M. musculus were tested for their ability to quench or enhance the fluorescence of quinacrine dihydrochloride in solution. The relationship between the base composition of a particular DNA fraction, its effect on the fluorescence of quinacrine in solution and its location in chromosomes relative to the quinacrine banding pattern is discussed.  相似文献

The interaction of the alkanesulphonate, methylene dimethanesulphonate (MDMS) with DNA has been studied. Thermal denaturation studies on mixtures of MDMS and DNA showed a dose-dependent decrease of the melting temperature midpoint (Tm) of the DNA. In addition, an irreversible decrease in ultraviolet absorption (hypochromism) preceded the hyperchromic shift, the magnitude of the former being linearly related to both the relative concentration of MDMS and the G-C content of the DNA used. Neither the reduction in melting temperature nor the initial UV absorption decrease occurred after dialysis of the reaction mixture. Equimolar proportions of the hydrolysis products of MDMS did not give the same effects as observed with the unhydrolysed agent. A similar hypochromism followed by strand separation occurs when DNA is allowed to stand with MDMS at room temperature, the time of subsequent strand separation being related to the treatment level of the drug. A weak association of MDMS with DNA is considered to be involved resulting in a local compression of the helical structure in the vicinity of the G-C pairs. It is suggested that this conformational change may act as a substrate for repair enzymes in vivo.  相似文献
