Traditional taxonomic methods of botanical identification that rely primarily on morphological observations cannot be used efficiently when only powdered plant materials are available. Thus, our objectives were to determine if we could apply a molecular approach to: a) produce unique DNA profiles that are characteristic of the species, and b) determine if the geographical area or time of collection influences these DNA profiles. Towards this end, random amplified polymorphic DNA (RAPD) analyses were performed on a number of botanicals currently used for women's health. The test materials included samples from three species each of the genera Cimicifuga (Actaea) and Trifolium, as well as samples of Vitex agnus-castus L., Glycyrrhiza glabra L., Gingko biloba L., Valeriana officinalis L., Angelica sinensis (Oliv.) Diels, Viburnum prunifolium L., Humulus lupulus L., Vaccinium macrocarpon Ait., Panax ginseng C.A. Mey. Cimicifuga racemosa (L.) Nutt. and Trifolium pratense L. are currently under clinical investigation in our basic research laboratories and medical clinic for the relief of post-menopausal symptoms. Characteristic profiles produced with the OPC-15 primer could distinguish the three Cimicifuga species: C. racemosa, C. americana and C. rubifolia. Similar results were obtained with the three Trifolium species: Trifolium pratense L., Trifolium incarnatum L., and Trifolium repens L. Accessions of cultivated T. pratense collected from the same field at different times, produced identical profiles. Accessions of Cimicifuga species collected from different geographical areas produced similar but not identical DNA profiles; however, species-specific DNA fragments were identified. These results demonstrate that RAPD analysis can be applied to distinguish species when only powdered material is available for testing. This methodology can be applied to identify species of commercial value regardless of collection time or geographic area.  相似文献

采用人工气候箱培养试验,初步研究了入侵杂草加拿大一枝黄花对本地植物的化感作用.测定了根系和根状茎的不同浓度浸提液对白三叶发芽率和幼苗生长的影响;不同播种密度下根系1:60提取浓度对白三叶种子萌发和幼苗生长的影响;根状茎1:60提取浓度对白三叶、红三叶、天蓝苜蓿、黑麦草、北美车前、鸡眼草、苇状羊茅、胜红蓟、马齿苋、碱蓬和刺苋的种子萌发和幼苗生长的影响.结果表明,加拿大一枝黄花根系和根状茎的化感抑制作用均随浓度的升高而增强,且根状茎提取物的抑制效应高于根系提取物的效应;在1:60低浓度根系浸提液作用下,不同密度的白三叶幼苗萌发率和幼根、幼芽长度差异不显著;1:60低浓度根状茎提取物对11个物种的种子萌发均有显著抑制作用,但对不同物种的抑制程度有差异,对禾本科植物的抑制作用大于非禾本科和豆科;1:60低浓度根状茎提取物对10个物种的幼根影响与对萌发率的影响相似,但在对幼芽的影响上,仅对鸡眼草、刺苋和苇状羊茅起抑制作用,而对其他植物无显著抑制(黑麦草、北美车前、胜红蓟、马齿苋和刺苋)或起促进作用(白三叶、红三叶、天蓝苜蓿).  相似文献

目的:以不同浓度乙醇和大孔吸附树脂法提取和纯化葛花的提取物,并用HPLC和UV进行异黄酮成分含量比较分析。方法:以不同浓度的乙醇回流提取葛花药材制备提取物,并采用大孔吸附树脂对其进行纯化;然后以HPLC法对提取物中的3种主要黄酮类成分进行定量分析,并以UV法测定其总黄酮含量。结果:葛花不同提取物的黄酮类成分含量差别较大;所建立的HPLC方法可以同时测定葛花提取物中鸢尾苷、6"-O-木糖鸢尾苷和染料木素的含量;UV法与HPLC法联合应用,可对不同葛花提取物中的异黄酮类成分进行评价。结论:不同工艺提取的葛花提取物中异黄酮类成分有较大区别,大孔吸附树脂纯化后可显著提高总黄酮的纯度;HPLC联合UV法可共同用于葛花的质量评价及提取工艺的研究。  相似文献

Isozymes were used to study the putative ancestors of white clover ( Trifolium repens L.). Ten enzymes were examined, and 18 loci were resolved via starch-gel electrophoresis for accessions representing T. repens, Trifolium isthmocarpum, Trifolium nigrescens, Trifolium occidentale and Trifolium uniflorum, in addition to two more distantly related species, Trifolium alpinum and Trifolium purseglovei. Nei's genetic identities indicate that T. uniflorum and T. nigrescens are the closest relatives of T. repens. The isozyme data thus support a hypothesis that the two genomes of the tetraploid T. repens could have been derived from hybridization between T. nigrescens and T. uniflorum. This conclusion is further supported by shared alleles between T. repens, T. nigrescens and T. uniflorum. However, the origin of T. repens is somewhat obscured by the presence of shared alleles between T. repens and both T. occidentale and T. isthmocarpum, suggesting that introgression of genes from the latter two species into T. repens may also have taken place. High values of genetic identity are shared between T. occidentale with T. nigrescens and T. uniflorum, also indicative of introgression. Alternatively the presence of shared alleles among the five species may reflect their recent common ancestry.  相似文献

Rhodophyta produce a variety of chemically different mycosporine‐like amino acids (MAAs), compounds that are known as some of the strongest ultraviolet (UV) absorbing molecules in nature. Accordingly, they primarily act as photoprotectants against harmful levels of solar ultraviolet radiation in the UV‐A and UV‐B range. In order to get a deeper understanding of the chemical diversity of MAAs in red algae, pure standards of eleven mycosporine‐like amino acids were isolated from three different species (Agarophyton chilense, Pyropia plicata and Champia novae‐zelandiae) using various chromatographic methods. Their structures were confirmed by nuclear magnetic resonance and mass spectrometry. Four out of the eleven MAAs are reported for the first time in algae. In addition, a new high‐performance liquid chromatography method was developed for the separation of all isolated MAAs and successfully applied for the analysis of twenty‐three red algal species of marine origin. All of them contained MAAs, the most abundant compounds were shinorine, palythine, asterina‐330 and porphyra‐334. For some samples, the direct assignment of MAAs based on their UV spectra was not possible; therefore, the target analytes were enriched by a simple concentration step, followed by liquid chromatography‐mass spectrometry analysis of the extracts. This approach enabled a deeper insight into the MAA pattern of red algae, indicating that not only the four dominant ones are synthesized but also many others, which were often described as unknown compounds in previous studies.  相似文献

本文通过HPLC法首次对产于我国11省的红车轴草中7种主要异黄酮单体含量进行了测定,结果表明,7种异黄酮总量变化范围为0.105%~1.725%。7种异黄酮单体含量随产地变化较显著,德鸢尾素、红车轴草素、毛蕊异黄酮、芒柄花素、大豆黄素、染料木素和鸡豆黄素A的含量变化范围分别为:0~0.189%、0~0.066%、0.020%~0.076%、0.004%~0.948%、0~0.089%、0.020%~0.073%和0~0.424%。  相似文献

The effect of concurrent salinity (0-60 mM NaCl) and rootzonehypoxia (flooding for up to 15 d) on shoot and root growth andshoot ion concentrations of six species of Trifolium (T. subterraneumL., T. fragiferum L., T. michelianum Savi., T. isthmocarpumBot., T. purpureum Lois., and T. repens L.), was studied intwo greenhouse experiments. There was a significant salinityx flooding effect for shoot yield but no significant salinityx flooding x species interaction although individual speciesdiffered significantly (P < 0·001) in their growthresponse to the saline or flooded conditions separately. Concentrationsof Na and Cl in the shoots of all species increased with increasingperiods of saline flooding and there was a significant salinityx flooding interaction. Sodium and Cl concentrations were significantlyhigher (P < 0·001) in T. purpureum, the species inwhich shoot growth was most depressed by saline flooding, thanother species. In T. michelianum, T. fragiferum and T. repens,fresh and dry weight of roots increased with flooding underboth saline and non-saline conditions while in T. subterraneumroot growth decreased. A significant proportion of the increasedroot growth in the first three species occurred as new adventitiousroots. These roots had higher percentages of internal gas spaceswithin the root tissue even in the presence of salinity comparedwith roots from non-flooded conditions. There were also significantlymore gas spaces in the total root tissue in T. fragiferum andT. repens under saline-flooding than in roots of T. subterraneum.Electron micrographs of the root cross sections illustratedthe presence of these gas spaces or aerenchyma. This study investigated the ability of neonatal larvae of the root-feeding weevil, Sitona lepidus Gyllenhal, to locate white clover Trifolium repens L. (Fabaceae) roots growing in soil and to distinguish them from the roots of other species of clover and a co-occurring grass species. Choice experiments used a combination of invasive techniques and the novel technique of high resolution X-ray microtomography to non-invasively track larval movement in the soil towards plant roots. Burrowing distances towards roots of different plant species were also examined. Newly hatched S. lepidus recognized T. repens roots and moved preferentially towards them when given a choice of roots of subterranean clover, Trifolium subterraneum L. (Fabaceae), strawberry clover Trifolium fragiferum L. (Fabaceae), or perennial ryegrass Lolium perenneL. (Poaceae). Larvae recognized T. repens roots, whether released in groups of five or singly, when released 25 mm (meso-scale recognition) or 60 mm (macro-scale recognition) away from plant roots. There was no statistically significant difference in movement rates of larvae.  相似文献

This paper reports on the mycorrhizal status of 82 plant species growing in traditionally managed grasslands in three different locations in the boreal and boreo-nemoral vegetation zone in the eastern part of Norway. Seventy-four species were found to have arbuscular mycorrhiza (AM). To our knowledge, we report AM for the first time in Achillea ptarmica, Ajuga pyramidalis, Alchemilla glaucescens, Carex brunnescens, Carex pallescens, Crepis praemorsa, Hieracium lactucella, Rumex longifolius, Scorzonera humilis, Trifolium aureum and Trifolium spadiceum. The rare and threatened species Arnica montana, S. humilis, C. praemorsa, Gentianella campestris, Parnassia palustris, T. aureum and T. spadiceum, all confined to grasslands, were found to possess AM fungi.  相似文献

A simple method involving polyamide column chromatography in combination with HPLC-PAD and HPLC-ESI/MS for isolating and identifying two kinds of lignans, arctiin and arctigenin, in the leaves of burdock (Arctium lappa L.) has been established. After extraction of burdock leaves with 80% methanol, the aqueous phase of crude extracts was partitioned between water and chloroform and the aqueous phase was fractionated on a polyamide glass column. The fraction, eluting with 100% methanol, was concentrated and gave a white precipitate at 4 degrees C from which two main compounds were purified by semi-preparative HPLC. In comparison with the UV and ESI-MS spectra and the HPLC retention time of authentic standards, the compounds were determined to be arctiin and arctigenin. The extraction/separation technique was validated using an internal standard method.  相似文献

Introduction – Cistaceae is a large family of shrubs widely spread over the Mediterranean area. It includes Helianthemum, Halimium and Cistus genus. Cistus genus contains approximately 20 species distributed in three subgenus. The essential oil of Cistus species has been thoroughly studied, but the polyphenolic composition of the aerial parts of the different Cistus species needs further characterisation. Objective – To perform a comparative analysis of the qualitative and quantitative polyphenolic composition of the aerial parts of the most commonly distributed Spanish Cistus species in order to find a relationship between chemotype and subgenus. Methodology – Thirteen aqueous extracts derived from 10 different Cistus species were analysed by using HPLC with diode array‐detection coupled to electrospray ion trap mass spectrometry technique (HPLC‐DAD‐ESI‐MS/MS). Their major compounds were identified and ellagitannins were quantified. Principal component analysis (PCA) was performed on the most relevant compounds to find out the statistical association between chemotype and variety. Results – Three main groups of compounds were found, i.e. ellagitannins, flavonoids and phenolic acids derivatives. The polyphenolic profile was specific for each species, although the abundance of some compounds also varied depending on the soil type. Whereas C. ladanifer, C. salviifolius, C. populifolius and C. libanotis were specially rich in ellagitannins, C. clusii, C. laurifolius and C. monspeliensis contained significant amounts of flavonoids and much less ellagitannins. In contrast, C. crispus, C. incanus and C. albidus showed a polyphenolic profile mostly based on flavonoids. PCA analysis showed a strong relationship between Cistus subgenus and its chemotype based on the most relevant water‐soluble polyphenolic compounds. Conclusions – Chemical composition of the leaves' aqueous extracts from plants belonging to the Cistus genus is strongly related to their subgenus, in agreement to previous taxonomical and phylogenetic divisions. In contrast, soil and climate are less influencing factors. Leucocistus and Halimioides subgenus showed a higher content in ellagitannins. However, Cistus subgenus had higher flavonoid content. 通过测定不同的温度、水分、盐分和光照等条件下,杂三叶和地三叶(考拉)在萌发期和幼苗期的发芽率、发芽势、生长速度、叶绿素含量、组织含水量、脯氨酸含量和细胞膜透性等指标,对两者抗性进行了比较研究。结果表明,在不同盐分浓度处理下,三叶草的叶绿素含量随盐浓度的增加而降低,组织含水量变化不明显,游离脯氨酸含量随盐浓度的增加而增加,细胞膜透性随盐浓度的增加而增强;在不同水分条件处理下,三叶草的游离脯氨酸含量和细胞膜透性随水分的增加而增加,叶绿素含量和组织含水量均变化不明显;在不同光照时间处理下,三叶草的叶绿素含量和游离脯氨酸含量均变化不明显,其细胞膜透性也无较大改变,而组织含水量则随光照时间的增加而增加;在不同温度条件下处理,三叶草的叶绿素含量、组织含水量和游离脯氨酸含量均变化不明显,细胞膜透性随温度的升高而降低。另外,从各项测定结果可以看出,杂三叶的各项性状要强于地三叶。  相似文献

A patent has been granted for the formulation of baculoviruses with stilbene-derived optical brighteners, a group of compounds that absorb ultraviolet (UV) radiation and emit visible blue wavelengths. These compounds are being extensively tested for control of forest-feeding lepidopterous insects in North America; optical brighteners may thus become a common ingredient in commercial baculovirus formulations in the near future. Many flower species use UV signals to attract insects and to direct them to the nectaries. We examined a possible consequence of field applications of optical brighteners: their effects on the ability of pollinators to find and handle flowers. In field studies carried out in Mexico and the United Kingdom on three different flower species, application of dilute (0.1% or 1%) concentrations of the optical brightener Tinopal CBS reduced recruitment of bees to flowers. Bees that approached flowers were less likely to land and feed on flowers treated with Tinopal than on controls. On one plant species, Trifolium repens, the time taken for bees to handle inflorescences was longer following applications of Tinopal. It seems that this optical brightener may both reduce recruitment of insects to flowers and interfere with their ability to locate rewards. Field-scale applications could reduce pollination of crops, weeds, and wildflowers and adversely affect bee populations. These possibilities should be examined in more detail before widespread applications of these compounds to the environment are made.  相似文献

Clovers (Trifolium) may possess a significant therapeutic potential, but the effects of compounds from these plants on blood platelets and haemostasis have been poorly recognized. The present study was designed to evaluate the antioxidative action of extracts from three species of clovers: Trifolium pratense, Trifolium pallidum and Trifolium scabrum in the protection of human blood platelets in vitro. Platelet suspensions were pre-incubated with crude extract and phenolic fraction of T. pratense or phenolic fractions of T. scabrum and T. pallidum, at the final concentrations of 0.5–50 μg/ml. Then, for the induction of oxidative stress, 100 μM peroxynitrite was added. The antioxidative activity of plant extracts was assessed by measurements of the level of 3-nitrotyrosine, thiol groups and lipid peroxidation products (hydroperoxides and thiobarbituric acid-reactive substances). Despite the significant differences in the composition of the investigated extracts, we observed antioxidative effects of all used mixtures. The presence of Trifolium extracts considerably reduced the peroxynitrite-mediated modifications of proteins and diminished peroxidation of lipids in platelets. Our results indicate on a strong antioxidative activity of the tested extracts-statistically significant effects were found even for the lowest concentrations (0.5 μg/ml) of all extracts. This action may be useful in the protection of blood components, very susceptible to oxidative modifications. The obtained results suggest that the examined clovers are a promising source of compounds, valuable for the protection against oxidative stress-induced damage to blood platelets.  相似文献

Water extracts of the compost produced from activated sludge and coffee residue were found to be selectively inhibitory to seed germination of some legumes. Germination rate of white clover (Trifolium repens L.), red clover (Trifolium pratense L.) and alfalfa (Medicago sativa L.) seeds were reduced to 2, 29 and 73% of the control, respectively, by water extracts of the compost (20 g l–1). However, the extracts did not show any inhibition to seed germination of sorghum (Sorghum bicolor Moench), African millet (Eleusine coracana Gaertn.), and Komatsuna (Brassica rapa L.) at the same concentration. The inhibitors in the compost extracts were separated by ion-exchange chromatography and reverse-phase high performance liquid chromatography (HPLC) and the inhibitory activities of seed germination were tested with white clover seeds. Five inhibitors were isolated and identified as 3,4-dichlorophenylacetic acid (3,4-DCP), 3,4-dichlorobenzoic acid (3,4-DCB), 3,4,5-trichlorophenylacetic acid, 3,4,5-trichlorobenzoic acid and mono-2-ethylhexylphthalate by 1H-, 13C-NMR spectroscopy and mass spectrometry. The inhibitory activities of some authentic chemicals of the inhibitors and the related compounds were compared. The results indicated that the main inhibitor in the compost could be 3,4-DCB, which was contained at the concentration of 6.58 mg kg–1 compost and showed the strongest inhibitory effect on seed germination of white clover among the tested compounds.  相似文献

采用蚕豆根尖细胞微核试验和染色体畸变试验,研究了温度胁迫下白车轴草水浸提液对蚕豆根尖细胞的化感作用。结果表明:不同温度(5℃、25℃和35℃)处理后的白车轴草的水浸提液对蚕豆根尖细胞有丝分裂指数的影响为低浓度促进,高浓度抑制。在白车轴草水浸提液的作用下,蚕豆根尖细胞内出现了多种染色体畸变,如微核、染色体断片、染色体桥、落后染色体等,对微核率和染色体畸变率的诱导效应表现为随水浸提液浓度的增大而增大。温度胁迫后的化感作用明显大于胁迫前,其化感效应表现为低温(5℃)下白车轴草水浸提液高温(35℃)常温(25℃)白车轴草水浸提液。推测原因可能是温度胁迫改变和增加了化感物质的释放。  相似文献

Introduction – There are numerous Ligularia species in the eastern Qinghai–Tibet Plateau and adjacent areas. L. virgaurea has been used as a traditional folk medicine for the treatment of stomachache and nausea. Objective – To analyse chemical constituents of L. virgaurea, grown in Yunnan and Sichuan provinces of China. Methodology – The direct MS measurement of the crude extract of plant samples was used for grouping of this species. As the main compounds were available in pure form, the main peaks were analysed by LCMS. Results – An easy and speedy method of analysis using MS spectra was developed. On the basis of the findings, L. virgaurea could be divided into two groups. The genetic studies also supported this grouping. Type 1 mainly includes virgaurenones and virgaurenolides. The MS of type 2 is quite different because it includes mainly ligularol and its congeners. Both MS were easily distinguished. Conclusion – The crude extracts of 11 L. virgaurea samples already collected in recent years were analysed and it was possible to identify them as type 1 or 2. Citrus peels were successively extracted with hexane, chloroform and acetone using a soxhlet extractor. The hexane and chloroform extracts were fractionated into alcohol-soluble and alcohol-insoluble fractions. These fractions were tested against different gram positive and gram negative bacteria. The EtOH-soluble fraction was found to be most effective. Fractionation of EtOH-soluble fraction on silica gel column yielded three polymethoxylated flavones, namely desmethylnobiletin, nobiletin and tangeretin. Their structures were confirmed by UV, 1H, 13C NMR and mass spectral studies. The findings indicated a potential of these natural compounds as biopreservatives in food applications.  相似文献

通过对影响甜菜碱提取和测定的各种因素进行研究,建立了一种准确、快速的测定长期盐胁迫下盐爪爪和盐穗木中甜菜碱的HPLC方法。样品采用超声波甲醇提取,并依次通过强阴阳离子交换树脂(Dowex 50×8和Dowex 50×2)进行纯化,得到的样品采用反相C18柱进行分离,色谱条件:流动相为50 mmol/L、pH 4.5的KH2PO4溶液;波长200 nm下紫外测定,流速0.7 mL/min;此方法的检出限为0.02μg/mL,平均回收率为97.2%。通过对0~500 mmol/L NaCl胁迫下的盐爪爪和盐穗木中甜菜碱进行测定,甜菜碱的含量均呈现先增加后减少的趋势,盐爪爪在NaCl浓度为200 mmol/L时达到最大,而盐穗木在NaCl浓度为300 mmol/L时达到最大。  相似文献
