Incubation in buffer of Bdellovibrio bacteriovorus 109J, B. stolpii UKi2, or B. starrii A3.12 with washed eucaryotic animal cells (mouse liver, hamster kidney, or bovine mammary gland) resulted in neither attachment nor growth of the bdellovibrios. When cells of these bdellovibrio strains were incubated with erythrocyte suspensions (bovine or rabbit) a very low level of bdellovibrio attachment and penetration occurred, but no growth could be detected. Using micurgical procedures, bdellovibrios were injected into the perivetelline space or the cytoplasm of rabbit ova. After 18–24h incubation, neither a significant loss nor increase of injected, intracellular bdellovibrios was observed. Limited axenic growth of bdellovibrios (109J or UKi2) occurred in media containing rabbit ova extracts and dilute nutrient broth. It is concluded that eucaryotic rabbit ova do not provide a suitable environment for intracellular bdellovibrio growth.  相似文献

This study is concerned with three aspects of the ionic requirements for cell attachment to collagen; namely (a) the divalent, (b) monovalent cation specificities and (c) pH optima for cell interaction with collagen. The divalent cation requirement for cell attachment to collagen can be sufficed by Ca2+, Mg2+ and certain transition group metals; whereas Ba2+, Sr2+, and polyamines are inactive. The pH optimum for cell attachment in this system occurs in the physiological range. The monovalent cation requirement for cell attachment to collagen is satisfied by isotonic NaCl, KCl, LiCl, NH4Cl, sucrose, and glucose. Pronounced inhibition of cell attachment occurs under both hypertonic and hypotonic conditions.  相似文献

Summary Leishmania donovani, the etiological agent for the disease visceral leishmaniasis, attach themselves to the macrophages for initiation of the disease. The attachment process has been found to be regulated by Ca2+ ions. Verapamil, a Ca2+-channel blocker inhibits Leishmania-macrophage attachment. The inhibitory effect is increased with time. Nifedipine, another Ca2+-channel blocker exhibits the same effect. The attachment process is stimulated by Ca2+-ionophore alone. The inhibitory effects of the calcium channel blockers are reversed by the ionophore.  相似文献

Various cations have been examined for their effects on phagocytosis. Media with high [Ca2+] and low [K+] favor phagocytosis, which is inhibited in media with high [K+], [Rb+], or [Ba2+] and low [Ca2+]. Microscopical observations of inhibited cells demonstrate that swimming behavior is not modified but they cannot perform the initial step of phagocytosis, attachment to food; when Ca2+ is added, cells attach to and ingest food, demonstrating rapid reversal of inhibition. Attachment is shown to be a linear function of the ratio [K+]/[Ca2+]1/2 or [Rb+]/[Ca2+]1/2 in the medium. The Ca2+ influx inhibitor Verapamil blocks attachment, as does La3+; the latter is believed to compete with Ca2+ for access to the Ca2+ channel. Likewise, treatment of cells with media containing no added Ca2+ inhibits attachment, and addition of 10 μM Ca2+ allows 90% of these cells to attach to and ingest food. The ionophore A23187, known to transport Ca2+ into a wide variety of cells, provokes lysosomal streaming movements typical of attachment. Based upon these observations, Ca2+ influx plays an essential role in attachment; K+ efflux also appears to be necessary since tetraethylammonium chloride blocks attachment. Treatment of cells with Tetrodotoxin, an inhibitor of Na+ transport, or suspending them in media containing no added Na+ does not affect attachment or ingestion, indicating that Na+ is not implicated in these processes. An hypothesis is presented which implicates Ca2+ in both direct adhesion and exocytosis phenomena during attachment.  相似文献

Thirty new Bdellovibrio strains were isolated from an agricultural soil and from the rhizosphere of plants grown in that soil. Using a combined molecular and culture-based approach, we found that the soil bdellovibrios included subpopulations of organisms that differed from rhizosphere bdellovibrios. Thirteen soil and seven common bean rhizosphere Bdellovibrio strains were isolated when Pseudomonas corrugata was used as prey; seven and two soil strains were isolated when Erwinia carotovora subsp. carotovora and Agrobacterium tumefaciens, respectively, were used as prey; and one tomato rhizosphere strain was isolated when A. tumefaciens was used as prey. In soil and in the rhizosphere, depending on the prey cells used, the concentrations of bdellovibrios were between 3 × 102 to 6 × 103 and 2.8 × 102 to 2.3 × 104 PFU g−1. A prey range analysis of five soil and rhizosphere Bdellovibrio isolates performed with 22 substrate species, most of which were plant-pathogenic and plant growth-enhancing bacteria, revealed unique utilization patterns and differences between closely related prey cells. An approximately 830-bp fragment of the 16S rRNA genes of all of the Bdellovibrio strains used was obtained by PCR amplification by using a Bdellovibrio-specific primer combination. Soil and common bean rhizosphere strains produced two and one restriction patterns for this PCR product, respectively. The 16S rRNA genes of three soil isolates and three root-associated isolates were sequenced. One soil isolate belonged to the Bdellovibrio stolpii-Bdellovibrio starrii clade, while all of the other isolates clustered with Bdellovibrio bacteriovorus and formed two distantly related, heterogeneous groups.  相似文献

The paper deals with a comparative study of the growth of free-living and immobilized predatory bacteria of the genus Bdellovibrio in the presence of toxic concentrations of urea and phenol. It was found that the cell wall of bdelloplasts plays a protective role in the adaptation of bdellovibrios to xenobiotics. The attachment of bdellovibrios to solid surfaces allows them to survive under unfavorable environmental conditions.  相似文献

The predatory activity of bdellovibrios is affected by various environmental pollutants such as detergents, heavy metals, and pesticides. This was shown in a two-membered system ofBdellovibrio andPhotobacterium, in which the effect of the predator on the bioluminescence of the prey indicated the activity of the former. The high sensitivity of the bdellovibrios toward certain chemicals (e.g., CdCl2) indicates the possibility of using the system for biological monitoring of those chemicals.  相似文献

The synergistic stimulation of ethylene production by kinetin and Ca2+ in hypocotyl segments of mung bean (Phaseolus aureus Roxb.) seedling was further studied. The requirement for Ca2+ in this system was specific. Except for Sr2+, which mimicked the effect of Ca2+, none of the following divalent cations, including Ba2+, Mg6+, Cu2+, Hg2+, Co2+, Ni2+, Sn2+, and Zn2+, showed synergism with kinetin on ethylene production. Fe2+, however, showed a slight synergism with kinetin. Some of them (Hg2+, Co2+, and Ni2+) had a strong inhibitory effect, while others (Zn2+, Mg2+, Sn2+, and Ba2+) had a slight or no inhibitory effect on ethylene production in the absence or presence of kinetin.  相似文献

The membrane potential of the frog egg undergoes a transient positive shift at fertilization which is a block to polyspermy. This paper addresses the question of how a sperm elicits this “fertilization potential.” Iontophoretic injection of Ca2+ activates Rana pipiens eggs to develop and initiates a transient, positive-going shift in the membrane potential (the activation potential) which is like the sperm-induced fertilization potential in amplitude, duration, and Cl? dependence. Activation potentials are elicited by Ca2 injection into both animal and vegetal regions of the egg, but the rate of the initial depolarization is much less when Ca2+ is injected into the vegetal region. Injections of K+, Na+, Cl?, or Mg2+ do not result in activation potentials, but the Ca2+ analogs, Sr2+ and Ba2+, can substitute for Ca2+. Treatment of eggs with the divalent cation ionophore, A23187, also initiates a transient, positive-going depolarization. Because injection of Ca2+ is sufficient to elicit a response almost identical to a fertilization potential, the ion transport mechanisms necessary to produce a fertilization potential must preexist in the unfertilized eggs; the sperm contributes only the stimulus to activate these mechanisms. The results reported here suggest that the stimulus may be a rise in free Ca2+.  相似文献

The absorption of Cu2+, Zn2+, and Mn2+ by leaf tissue of 4-month old sugarcane plants (Saccharum officinarum L., var. H53-263) has been investigated. After the “apparent free space” fraction was desorbed, the absorption of Cu2+, Mn2+, and Zn2+ yielded a curve typical of many ion uptake processes when measured as a function of the external concentration. However, only 1 absorption mechanism was evident for each cation. The pH optimum for Cu2+ and Zn2+ uptake was 5.0 to 6.0, whereas that for Mn2+ absorption was 4.5 to 6.0. Absorption was competitively inhibited by H+, and this inhibition was reversible when 0.5 mm Ca2+ was present. Cu2+ and Zn2+ were absorbed through the same carrier sites, as concluded from their mutually competitive activities. Mn2+ was absorbed through a second, independent mechanism. Uptake of each cation was strongly inhibited by uncouplers of oxidative phosphorylation, by Amytal and Nembutal2, by 5 × 10−2m succinate, and by ADP and Pi. Absorption of Cu2+, Zn2+, and Mn2+ was concluded to be coupled to oxidative phosphorylation, and specifically to energy-conservation Site I.  相似文献

Purinergic receptor expression and involvement in steroidogenesis were examined in NCI-H295R (H295R), a human adrenal cortex cell line which expresses all the key enzymes necessary for steroidogenesis. mRNA/protein for multiple P1 (A2A and A2B), P2X (P2X5 and P2X7), and P2Y (P2Y1, P2Y2, P2Y6, P2Y12, P2Y13, and P2Y14) purinergic receptors were detected in H295R. 2MeS-ATP (10–1000 µM), a P2Y1 agonist, induced glucocorticoid (GC) secretion in a dose-dependent manner, while other extracellular purine/pyrimidine agonists (1–1000 µM) had no distinct effect on GC secretion. Extracellular purines, even non-steroidogenic ones, induced Ca2+-mobilization in the cells, independently of the extracellular Ca2+ concentration. Increases in intracellular Ca2+ concentration induced by extracellular purine agonists were transient, except when induced by ATP or 2MeS-ATP. Angiotensin II (AngII: 100 nM) and dibutyryl-cyclic AMP (db-cAMP: 500 µM) induced both GC secretion and Ca2+-mobilization in the presence of extracellular Ca2+ (1.2 mM). GC secretion by AngII was reduced by nifedipine (10–100 µM); whereas the Ca2+ channel blocker did not inhibit GC secretion by 2MeS-ATP. Thapsigargin followed by extracellular Ca2+ exposure induced Ca2+-influx in H295R, and the cells expressed mRNA/protein of the component molecules for store-operated calcium entry (SOCE): transient receptor C (TRPC) channels, calcium release-activated calcium channel protein 1 (Orai-1), and the stromal interaction molecule 1 (STIM1). In P2Y1-knockdown, 2MeS-ATP-induced GC secretion was significantly inhibited. These results suggest that H295R expresses a functional P2Y1 purinergic receptor for intracellular Ca2+-mobilization, and that P2Y1 is linked to SOCE-activation, leading to Ca2+-influx which might be necessary for glucocorticoid secretion.  相似文献

The first step in attachment of Rhizobiaceae cells to plant root hair tips is mediated by a Ca2+-dependent, Ca2+-binding protein, rhicadhesin. The possible role of Ca2+ in synthesis, anchoring and activity of rhicadhesin was investigated. Growth of Rhizobium leguminosarum biovar viciae cells under Ca2+-limitation was found to result in loss of attachment ability. Under these conditions, rhicadhesin could not be usolated from the bacterial cell surface, but was found to be excreted in the growth medium. Divalent ions appeared to be essential for the ability of purified rhicadhesin to inhibit attachment of R. leguminosarum biovar viciae cells to pea root hair tips. Calcium ions were found not to be involved in binding of rhicadhesin to the plant surface, but appeared to be involved in anchoring of the adhesin to the bacterial cell surface. A model for the role of Ca2+ in activity of rhicadhesin is presented.  相似文献

HutP is an RNA-binding protein that regulates the expression of the histidine utilization (hut) operon in Bacillus subtilis, by binding to cis-acting regulatory sequences on hut mRNA. It requires L-histidine and an Mg2+ ion for binding to the specific sequence within the hut mRNA. In the present study, we show that several divalent cations can mediate the HutP–RNA interactions. The best divalent cations were Mn2+, Zn2+ and Cd2+, followed by Mg2+, Co2+ and Ni2+, while Cu2+, Yb2+ and Hg2+ were ineffective. In the HutP–RNA interactions, divalent cations cannot be replaced by monovalent cations, suggesting that a divalent metal ion is required for mediating the protein–RNA interactions. To clarify their importance, we have crystallized HutP in the presence of three different metal ions (Mg2+, Mn2+ and Ba2+), which revealed the importance of the metal ion binding site. Furthermore, these analyses clearly demonstrated how the metal ions cause the structural rearrangements that are required for the hut mRNA recognition.  相似文献

Two BODIPY derivative sensors for metal ion recognition containing 10-(4-hydroxyphenyl) (L1) and 10-(3,4-dihydroxyphenyl) (L2) were synthesized in a one-pot reaction of benzaldehyde derivative and 2,4-dimethylpyrrole in the presence of trifluoroacetic acid as catalyst. The binding abilities between these sensors and 50 equivalents of Na+, K+, Ag+, Ca2+, Fe2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+, Pb2+, Al3+ and Cr3+ ions were studied using UV–vis and fluorescent spectroscopic methods. Of all the metal ions tested, Al3+ ion showed the greatest decrease in intensity in the spectra of the sensors, and therefore Al3+ ion forms the strongest complex. The binding abilities of BODIPY receptors with Na+, Ag+, Ca2+, Co2+, Ni2+, Cu2+, Zn2+ and Al3+ ions were also investigated using density functional theory (DFT) calculations at B3LYP/LanL2DZ theoretical level. The calculated results point to the same conclusion. DFT calculations also provided the HOMO–LUMO energy levels, which can explain the spectrum change upon complexation.

In this study, dried and humid fruiting bodies of Tremella fuciformis and Auricularia polytricha were examined as cost-effective biosorbents in treatment of heavy metals (Cd2+, Cu2+, Pb2+, and Zn2+) in aqueous solution. The humid T. fuciformis showed the highest capacity to adsorb the four metals in the multi-metal solutions. The Pb2+ adsorption rates were 85.5%, 97.8%, 84.8%, and 91.0% by dried T. fuciformis, humid T. fuciformis, dried A. polytricha, and humid A. polytricha, respectively. The adsorption amount of Pb2+ by dried and humid T. fuciformis in Cd2+ + Pb2+, Cu2+ + Pb2+, Pb2+ + Zn2+, Cd2+ + Cu2+ + Pb2+, and Cd2+ + Zn2+ + Pb2+ solutions were not lower than that in Pb2+ solutions. The results suggested that in humid T. fuciformis, Cd2+, Cu2+, and Zn2+ promoted the Pb2+ adsorption by the biomass. In the multi-metal solutions of Cd2+ + Cu2+ + Pb2+ + Zn2+, the adsorption amount and rates of the metals by all the test biosorbents were in the order of Pb2+ > Cu2+ > Zn2+ > Cd2+. Compared with the pseudo first-order model, the pseudo second-order model described the adsorption kinetics much better, indicating a two-step biosorption process. The present study confirmed that fruiting bodies of the jelly fungi should be useful for the treatment of wastewater containing Cd2+, Cu2+, Pb2+, and Zn2+.  相似文献

Summary In an artificial system, the ionophore A23187, which transports Ca2+ but not Na+, is able to mediate the uphill translocation of Ca2+ from one aqueous medium to another across an organic immiscible phase, provided that a Na+, Li+ or H+ gradient is imposed on the system. Therefore, in the process known as Na-Ca countertransport, the downhill influx of Na+ may not be necessary for causing Ca2+ extrusion against its electrochemical gradient.  相似文献

A new molecular fluorescent sensor (L) for Cu2+ has been synthesized by derivatizing the lower rim of calix[4]arene with benzothiazole moiety, through amide linkage to result in 1,3-di-derivative. The receptor molecule, L exhibited fluorescence quenching towards Cu2+ among eleven divalent ions, viz., Mn2+, Fe2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+, Hg2+, Ca2+, Mg2+ and Pb2+, studied. The 1:1 stoichiometry of the complex formed between L and Cu2+ has been demonstrated by electronic absorption and ESI-MS. The role of calix[4]arene for the selective sensing of Cu2+ has been established by comparing the data with that obtained for an appropriate control molecule. The minimum concentration at which L can detect Cu2+ has been found to be 403 ppb. The computations carried out at DFT level have provided the coordination and structural features of the Cu2+ complex of L as species of recognition. The Cu2+ complex thus formed recognizes iodide by bringing change in the color, among the 14 anions studied.  相似文献

A metallothionein-like gene, ThMT3, encoding a type 3 metallothionein, was isolated from a Tamarix hispida leaf cDNA library. Expression analysis revealed that mRNA of ThMT3 was upregulated by high salinity as well as by heavy metal ions, and that ThMT3 was predominantly expressed in the leaf. Transgenic yeast (Saccharomyces cerevisiae) expressing ThMT3 showed increased tolerance to Cd2+, Zn2+, Cu2+, and NaCl stress. Transgenic yeast also accumulated more Cd2+, Zn2+, and NaCl, but not Cu2+. Analysis of the expression of four genes (GLR1, GTT2, GSH1, and YCF1) that aid in transporting heavy metal (Cd2+) from the cytoplasm to the vacuole demonstrated that none of these genes were induced under Cd2+, Zn2+, Cu2+, and NaCl stress in ThMT3-transgenic yeast. H2O2 levels in transgenic yeast under such stress conditions were less than half those in control yeast under the same conditions. Three antioxidant genes (SOD1, CAT1, and GPX1) were specifically expressed under Cd2+, Zn2+, Cu2+, and NaCl stress in the transgenic yeast. Cd2+, Zn2+, and Cu2+ increased the expression levels of SOD1, CAT1, and GPX1, respectively, whereas NaCl induced the expression of SOD1 and GPX1.  相似文献

The binding of α-naphthaleneacetic acid (14C-NAA) to pelletable particulates from corn (Zea mays) coleoptiles was found to be influenced by inorganic solutes. La3+, Ca2+, and Mg2+ increased the binding whereas monovalent cations did not. The concentrations of CaCl2 which increased auxin binding were similar to those which inhibited coleoptile elongation in the presence of auxin. These results are interpreted as suggesting that the alteration of hormonal effectiveness by some inorganic solutes involves alterations in the attachment of the hormone to binding sites in the cell.  相似文献

AlCl3, MnCl2, and CdCl2 inhibited the rates of accumulation of 14C] L-glutamate and 3H] gammaaminobutyrate (GABA) in purified rat forebrain nerve-ending particles in a dose-dependent fashion. The concentrations that would give 50% inhibition (IC50) of GABA transport were 316 μM, 7.4 mM, and 1.4 mM, respectively. Ca2+ (1 mM) enhanced the inhibitory effect of Al3+ (IC50 decreased to 149 μM) but antagonized that of Mn2+ (IC50 = 10 mM) and Cd2+ (IC50 = 2.1 mM). For glutamate transport 1 mM Ca2+ changed the IC50 values from 299 to 224 μm for Al3+, 7.1 to 10 mM for Mn2+, and 2 to 3 mM for Cd2+. In contrast, the rates of accumulation of 14C] 2-deoxy-glucose and 3H] L-phenylalanine were mostly unaffected by these metal ions. The results indicate that Al3+, Mn2+, and Cd2+ exerted selective and differential effects on the transport systems of neurotransmitter substances in the synaptosomal membrane.  相似文献
