Denitrification beds are a simple and relatively inexpensive technology for removing nitrate from point source discharges. To date, operational beds have used wood media as the carbon source, as it provides a sustained nitrate removal rate (2-10 g N m−3 of media d−1) while maintaining permeability. In pilot-scale (2.9 m−3) denitrification beds receiving municipal wastewater effluent dosed with KNO3, we looked at improving nitrate removal by using alternative carbon media (maize cobs) and increasing bed temperature through passive solar heating. The influence of flow regime (horizontal-point, horizontal-diffuse, downflow and upflow) on short-circuit flow was also investigated.The long-term nitrate removal rate (21.8 g N m−3 d−1) of the maize cob beds over the 15-month period of the trial was 2-11-fold higher than sustained removal rates reported by other researchers for wood-based beds. While passive solar heating raised the mean bed temperature by 3.4 °C, it did not cause a measurable increase in the nitrate removal rate due to the variability in the removal rate exceeding the expected increase due to temperature.Horizontal flow had more short-circuiting than vertical flow. Short-circuiting in the horizontal flow was attributed to flow being concentrated near the top surface due to the buoyancy effect of warmer water. Greater short-circuiting in the solar heated horizontal and upflow beds than in the corresponding unheated beds was attributed to the buoyancy effect being more pronounced in the solar heated beds.Overall, downflow was deemed the most effective of the four tested flow regimes. It provided the highest increase in bed temperature due to solar heating, had the highest nitrate removal rate in the latter part of the trial and had more plug-flow characteristics. While passive solar heating raised bed temperature, we were unable to demonstrate a significant difference (at 95% CL) in nitrate removal rate between the unheated and solar heated beds because of the high variability in nitrate removal rate and the increase in short-circuiting in the solar heated horizontal and upflow beds. 相似文献

The aim of this research was to design a solar heated reactor system to enhance the anaerobic treatment of wastewater or biological sludge at temperatures higher than the ambient air temperature. For the proposed reactor system, the solar energy absorbed by flat plate collectors was transferred to a heat storage tank, which continuously supplied an anaerobic-filter reactor with water at a maximum temperature of 35 degrees C. The packed reactor was a metallic cylindrical tank with a peripheral twin-wall enclosure. Inside this enclosure was circulated warm water from the heat storage tank. Furthermore, a mathematical model was developed for the prediction of the temperature distribution within the reactor under steady state conditions. Preliminary results based on model simulations performed with meteorological data from various geographical regions of the world suggested that the proposed solar reactor system could be a promising and environmentally friendly approach for anaerobic treatment of wastewater and biological sludge. 相似文献

AIM: To determine whether batch solar disinfection (SODIS) can be used to inactivate oocysts of Cryptosporidium parvum and cysts of Giardia muris in experimentally contaminated water. METHODS AND RESULTS: Suspensions of oocysts and cysts were exposed to simulated global solar irradiation of 830 W m(-2) for different exposure times at a constant temperature of 40 degrees C. Infectivity tests were carried out using CD-1 suckling mice in the Cryptosporidium experiments and newly weaned CD-1 mice in the Giardia experiments. Exposure times of > or =10 h (total optical dose c. 30 kJ) rendered C. parvum oocysts noninfective. Giardia muris cysts were rendered completely noninfective within 4 h (total optical dose >12 kJ). Scanning electron microscopy and viability (4',6-diamidino-2-phenylindole/propidium iodide fluorogenic dyes and excystation) studies on oocysts of C. parvum suggest that inactivation is caused by damage to the oocyst wall. CONCLUSIONS: Results show that cysts of G. muris and oocysts of C. parvum are rendered completely noninfective after batch SODIS exposures of 4 and 10 h (respectively) and is also likely to be effective against waterborne cysts of Giardia lamblia. SIGNIFICANCE AND IMPACT OF THE STUDY: These results demonstrate that SODIS is an appropriate household water treatment technology for use as an emergency intervention in aftermath of natural or man-made disasters against not only bacterial but also protozoan pathogens. 相似文献

Methods and Results: Known quantities of Giardia duodenalis and Entamoeba histolytica/dispar cysts in sterile water were exposed to the sun. Containers were made of polyethylene terephthalate, eight painted black on one side, one not painted and another cut open at the top and the last was a high density polypropylene container. Viability testing was performed using vital and fluorescent dyes. The same assays were conducted under cloudy conditions. Thermal control tests were also performed using heat without ultra violet light from the sun. Results show that 99·9% of parasites was inactivated when water temperatures reached 56°C after sunlight exposure.
Conclusion: Both solar radiation and heat produced by the sun have a synergistic effect in killing cysts of Giardia duodenalis and Entamoeba histolytica/dispar when temperatures rise above 50°C, with complete death at 56°C, using painted 2-l PET containers.

Aim: To investigate the influence of reducing beef surface water activity (aw) on the survival of Salmonella Typhimurium DT104 during heating. Methods and Results: Beef discs were surface inoculated with S. Typhimurium DT104 and either untreated or dried to achieve surface aw values of 0·95, 0·85 and 0·70. The samples were vacuum packed, heat‐treated at 60°C and removed at predetermined times. The inactivation curves were influenced by aw and treatment time. Biphasic inactivation curves were observed for S. Typhimurium DT104 heat‐treated on beef samples with altered aw values, which were characterized by an initial decline in cell numbers at commencement of heating followed by a much slower rate of inactivation during the remaining treatment period. Point estimates of the heating time required to achieve a 1 log reduction on beef surfaces with aw of 0·99, 0·95, 0·85 and 0·70 were 0·5, 1·55, 11·25 and 17·79 min, respectively. Conclusions: A decrease in beef surface aw can substantially enhance the survival of S. Typhimurium DT104 after heating. Significance and Impact of the Study: Caution needs to be taken using dry air as a decontamination method as this may rapidly decrease product surface and pathogen aw values resulting in enhanced survival. 相似文献

In the literature on hydrolase-catalysed synthetic reactions in aqueous-organic biphasic systems, it has been stated that low water concentrations contribute to favourable shifts in equilibrium. It is argued here that this is not a sufficient condition for mass action effects of water. A simple method of treating such equilibria is suggested, using the thermodynamic activity of water. Only when its activity is significantly reduced below 1 can water contribute to a shift of equilibrium in favour of synthetic products. Such a reduction is not necessarily obtained by creating a biphasic system, however low the water content, but requires that the aqueous phase becomes a very concentrated solution/dispersion of hydrophilic species. 相似文献

Aims: To determine Legionella pollution prevalence, describe the amount of Legionellae with respect to temperature in Shanghai cooling tower water (CTWs) in various types of public sites.
Methods and Results: Six urban districts were selected as the study fields, adopting multiple-phase sampling methods. Routine culture was used to identify Legionellae. Of the samples, 58·9% (189/321) were observed to be positive, 19·9% were isolated over 100 CFU ml−1 . Legionella pneumophila serogroup 1 was the most frequently isolated species (155/189, 82·0%), followed by Leg. micdadei that was at the second place (44/189, 23·3%). The mean CFU ml−1 of Legionellae in CTWs reached its peak from July to September. Over all 15·4% of the samples exceeding 100 CFU ml−1 were observed in a hospital setting.
Conclusions: The prevalence of Legionella pollution in CTWs, especially in CTWs of subway stations and hospitals, is worrying, and the positive rate and CFU ml−1 of Legionellae in CTWs have a close relationship with air temperature.
Significance and Impact of the Study: The study demonstrates pollution prevalence rates in different types of sites and various seasons, and provides a proportion of different serogroups of Legionellae . Aims: In vitro experiments were undertaken to evaluate biocide formulations commonly used in cooling water systems against protozoa previously isolated from cooling towers. The investigations evaluated the efficacy of these formulations against amoebic cysts and trophozoites.
Methods and Results: Laboratory challenges against protozoa isolated from cooling towers using chlorine, bromine and isothiazolinone biocides showed that all were effective after 4 h. The presence of molybdate and organic phosphates resulted in longer kill times for bromine and isothiazolinones. All treatments resulted in no detectable viable protozoa after 4 h of exposure.
Conclusions: The chemical disinfection of planktonic protozoa in cooling water systems is strongly influenced by the residence time of the formulation and less so by its active constituent. Bromine and isothiazolinone formulations may require higher dosage of concentrations than currently practiced if used in conjunction with molybdate- and phosphate-based scale/corrosion inhibitors.
Significance and Impact of the Study: Cooling water systems are complex microbial ecosystems in which predator–prey relationships play a key role in the dissemination of Legionella . Preliminary results from an energy analysis of the coal fuel cycle in an Appalachian coal county have provided a systematic assessment of hidden energy subsidies in extraction, transport, processing, and combustion. Current results indicate that the system operates at an annual energy deficit of approximately 350 × 1010
kcal. A major loss is depletion of the coal resource base by use of inefficient mining techniques. Although of smaller magnitude, reductions in work force and community productivity from occupational accidents, disease, and road maintenance requirements for transport also appear to be significant. Aim: To examine the efficacy of tetra-sodium EDTA in controlling microbial contamination of dental unit water systems (DUWS).
Methods and Results: Ten dental units were treated once a week with either 4% or 8% tetra-sodium EDTA for four or two consecutive weeks, respectively. Before treatment, 43% and 60% of the water samples from the air/water triple syringe and high-speed hand-pieces, respectively, exceeded the American Dental Association (ADA) guidelines of 200 CFU ml−1 water during a 6-week baseline period. After each weekend treatment, the levels of microbial contamination in all DUWS fell significantly ( P < 0·001) to below the ADA guideline. By the end of the week, microbial counts in the outflowing water had returned to baseline levels indicating a transient effect of single doses of tetra-sodium EDTA, and the need for multiple applications. The biofilms were virtually eliminated after a single weekend treatment.
Conclusions: Tetra-sodium EDTA is effective in controlling microbial contamination in DUWS.
Significance and Impact of the Study: Inexpensive, effective and safe products for reducing the microbial load of water from DUWS are needed to meet ADA and other national guidelines. The developed primer sets were effective in directly amplifying deh II genes from enriched consortia samples, and the DNA extracted from tap water provided that an additional nested PCR step for detection of the deh II gene was used.

AIMS: To investigate treated water distribution systems in England as a source of Helicobacter pylori. METHODS AND RESULTS: Water and biofilms were obtained from 11 domestic and seven educational properties and from hydrants, reservoirs and water meters supplied by three water utilities. Samples were cultured on nonselective and antibiotic containing media combined with immunomagnetic separation concentration. Viable helicobacters were not detected in any of the 151 samples but Helicobacter-specific PCR assays detected DNA in 26% of samples from domestic properties, schools and hydrants with the highest frequency in biofilms (42%). Direct sequencing of six selected amplicons confirmed >95% sequence homology to H. pylori. CONCLUSIONS: While viable helicobacters were not isolated, evidence was obtained for the presence of Helicobacter DNA, including that of H. pylori. Biofilms on surfaces within water distribution systems may act either as sites for the passive accumulation of helicobacters or as potentially important reservoirs of infection. SIGNIFICANCE AND IMPACT OF THE STUDY: Our findings strengthen evidence that H. pylori may be transmitted through drinking water. However, there is currently no evidence that viable cells can survive the disinfection levels used in UK mains supplies and the health risk from this source remains unclear. 相似文献

Aims: To investigate the prevalence of culturable and nonculturable Legionella species in hot water systems of public buildings in Japan and assess the risk factors associated with Legionella contamination in hot water systems. Methods and Results: Legionella species were detected by conventional culture and molecular methods in 130 water samples collected from 40 buildings. A total of 26 (20·0%) water samples from 17 (42·5%) buildings were positive by culture, qualitative PCR or both methods: Legionella pneumophila and Leg. anisa were detected in four samples by a culture method, whereas 23 samples were positive by qualitative PCR, with the presence of various Legionella species confirmed by sequencing. Of these 23 samples, bacterial counts were quantifiable in 21 by real‐time PCR (from 1·7 × 105 to 2·6 × 1011 cells per litre). Phylogenetic analysis of amplified partial 16S rRNA gene showed close relations to various species of Legionella, including Leg. anisa and Leg. micdadei, all of which have been associated with respiratory diseases or increased antibody titres in human sera. Assessment of risk factors showed that turbidity, free chlorine concentration, iron concentration and heterotrophic plate count (HPC) were significantly associated with Legionella contamination (P < 0·05). Conclusions: Contamination of hot water systems of public buildings with culturable and nonculturable Legionella species may be a potential risk factor for Legionella infection in Japan. Adequate levels of chlorine, low levels of iron and HPC are important maintenance measures in the reduction of Legionella contamination in hot water systems. Significance and Impact of the Study: More than 40% of hot water systems in the Japanese public buildings examined were contaminated by not only culturable Leg. pneumophila and Leg. anisa but also by nonculturable pathogenic species. We investigated the effects of the lyophilisation medium (enzyme plus buffer salt and additives) and of water activity (a(w)) on the catalytic properties of lipase from Chromobacterium viscosum (lipase CV) in organic solvents; catalysis of ester and lactone synthesis were compared and, despite the similarities of the reactive groups involved in these reactions, some interesting differences were observed. Including 2-[N-morpholino]ethanesulfonic acid (MES) buffer in the lyophilisation medium of lipase CV increased its catalytic activity in transesterification and lactonisation, although the buffer salt requirement for maximal activity differed between the two reactions. Sorbitol, glucose, lactose, 18-crown-6 (crown ether 18-C-6), beta-cyclodextrin and bovine serum albumin were employed as alternative additives in the transesterification reaction, but were not as effective as MES buffer. Salt hydrates were used to investigate the effect of a(w) on esterification and lactonisation reactions catalysed by lipase CV. The maximum rate of hexadecanolide synthesis in toluene occurred at a(w) = 0.48. The optimum a(w) for the transesterification reaction in heptane/alcohol mixtures depended on the alcohol substrate employed (1-heptanol, 2-heptanol, or 3-methyl-3-hexanol) but not on the acyl donor (p-NP acetate or caprylate). The optimum a(w) values for both reactions were unchanged when a common solvent system (toluene/1-heptanol) was employed, indicating that the dependence of enzyme activity on a(w) is an intrinsic property of the enzyme-catalysed reaction and not a function of the solvent or other additives. 相似文献

124 sections of water courses from Northwest Germany and 154 sections of water courses from Southern Germany were classified into ten groups with regard to water quality and physico-chemical water type. Vegetation data were transformed into a two-step scale. Using the 39 most frequent water plant species as predictor variables a stepwise multiple discriminant analysis was carried out.29 species proved to be suitable for optimal discrimination of the groups. Most of the species correspond to those to which a certain indicator value had already been adjudged, using other methods. Species number, which was also tested, was not taken into the analyses. The score of right predictions ranges between 70 and 85 per cent depending on the technique of analysis and the number of data. Totaly wrong predictions with regard to water quality, as well as to water type were not frequent. Some possibilities for an explanation of mispredictions were suggested with respect to the fact that a statical model will never immediately indicate changes in the investigated systems.Finally, an ordination of the groups was carried out. The position of the groups in the canonical discriminant space could partly be interpreted in terms of succession.
Zusammenfassung 124 Gewässerabschnitte aus Nordwestdeutschland (eigene Aufnahmen) sowie 154 Gewässerabschnitte aus Süddeutschland (nach Veröffentlichungen der Arbeitsgruppe Kohler) wurden nach der Gewässergüte und dem physiko-chemischen Gewässertyp in 10 Gruppen eingeteilt. Die Vegetation wurde in einer 2-teiligen Skala geschätz bzw. umgerechnet. Mit den 39 häufigsten Wasserpflanzenarten as Prädiktoren wurde eine schrittweise Multiple Diskriminanzanalyse durchgeführt.Es zeigt sich, daß 29 Arten geeignet sind, die Gruppen optimal zu trennen. Einigen dieser Arten war auch schon vorher mit Hilfe anderer Methoden ein Zeigerwert zugesprochen worden. Die Artenzahl wird nicht in die Trennung der Gruppen einbezogen. Die Trefferquote für die richtige Zuordnung schwankt zwischen 70 und 85% je nach Methode und Datenmenge. Absolute Falschzuordnungen (sowohl nach Gewässergüte als auch Gewässertyp) sind selten. Einige Möglichkeiten der Erklärung von Falschzuordungen werden angedeutet, unter Bezug auf die Tatsache, daß ein statisches Modell Veränderungen im System nicht sofort widerspiegeln kann.Abschließend wird eine Ordination der Gruppen durchgeführt. Die Lage der Gruppen im kanonischen Diskriminanzraum kann teilweise im Sinne einer Sukzession interpretiert werden.相似文献

ABSTRACTHot water bathing – a Japanese traditional practice – has not been evaluated for its association with night- and sleep-time blood pressure (BP) in large population. In this longitudinal analysis, bathing parameters and ambulatory BP were repeatedly measured for 2 nights in 758 Japanese elderly individuals. Participants were divided into three groups according to tertile values of time soaked in the bathtub (Duration: tertile value, 11 and 15 min), time from bathing-end to bedtime (Time before bedtime: tertile value, 42 and 106 min), and temperature of hot water in the bathtub (Water temp: tertile value, 40.3 and 41.2 °C). Participants’ mean age was 70.9 years, and mean night- and sleep-time systolic BP (SBP) and dipping were 115.1 ± 16.1, 114.2 ± 16.2 mmHg, and 14.2 ± 8.8%, respectively. Multivariable mixed-effect linear regression models adjusted for potential confounding factors suggested that nighttime SBP was significantly lower in the intermediate Time before bedtime group by 1.7 mmHg (95% CI, 0.2–3.1) and in the short group by 1.9 mmHg (95% CI, 0.1–3.7) than that in the long group. Dipping was significantly greater in the intermediate Time before bedtime group by 1.8% (95% CI, 0.7–2.9) and in the short group by 1.8% (95% CI, 0.6–3.1) than that in the long group. These associations were consistent regarding sleep-time SBP. Conversely, Water temp and Duration did not significantly associate with any ambulatory BP parameter. Remarkably, Time before bedtime significantly prolonged with increases in tertiles of Water temp (P for trend = 0.006). In conclusion, the findings of this study revealed that Japanese hot water bathing, especially the short time from bathing-end to bedtime, was associated with lower night- and sleep-time BP and greater dipping in an elderly population. 相似文献
