Cysteine endopeptidases, SH-EP from Vigna mungo and EP-C1 from Phaseolus vulgaris, act to degrade seed storage protein during seed germination. Using transgenic tobacco plants, expression of SH-EP and promoter activity of the EP-C1 gene were analyzed in transgenic tobacco plants. The promoters of the two genes in tobacco seeds showed germination-specific activation, although post-translational processing of SH-EP and regulatory regions of promoter of the gene for EP-C1 were found to differ between leguminous seeds and transgenic tobacco seeds.  相似文献

Phytocystatins (PCs) are protein inhibitors of endogenous plant endopeptidases and exogenous pathogen proteinases. We have previously described the protein inhibitor TrcC-4, which is probably involved in the control of protein degradation during triticale seeds germination. The occurrence of the LARFAVXEHN motif supports the TrcC-4 designation as a PC. In this paper TrcC-4 was expressed in Escherichia coli using the pET28 expression vector. TrcC-4(6×His) was purified by affinity chromatography with a single step of purification. Western blot analysis showed the presence of TrcC-4 in both developing and germinating triticale seeds. TrcC-4 protein level was higher both in scutellum of germinating seeds and in developing grains of the triticale cultivar more resistant to pre-harvest sprouting (Zorro) than in a less resistant one (Disco). Furthermore it was demonstrated that the activity of EP8, cysteine endopeptidase responsible for the mass hydrolysis of prolamin during germination, is inhibited by TrcC-4(6×His), as confirmed by native PAGE with gliadin as a substrate. These results suggest that phytocystatin TrcC-4 controls the activity of cysteine endopeptidases involved in germination and, thus, is potentially involved in pre-harvest sprouting.  相似文献

Summary Amino acid, polyamine and protein concentrations in seeds and their evolution during seed germination of two dipterocarp species, Hopea odorata and Dipterocarpus alatus, were determined with the help of a multianalytical system. Glutamic acid and glutamine were the major amino compounds present. Hopea seeds also contain high levels of aspartic acid/asparagine, serine, threonine, arginine and alanine, while those of Dipterocarpus contain high levels of alanine, arginine and threonine. These species were quite different in their germination behavior and thus in their protein and amine metabolism rates. In Hopea, polyamines increased during the first 3 days of germination and reached a maximum by the 3rd day, 1 day before maximum germination rate. In Dipterocarpus polyamines reached their maximum at the 6th day while maximum germination rate is observed by the 7th day. This suggests that polyamine compounds could play a role in the early part of the germination process in Hopea and Dipterocarpus seeds. The possibility that control of polyamine biosynthesis could be used for the establishment of biochemical methods to improve seed storage and to control germination of these recalcitrant seeds is discussed.  相似文献

In this work, we investigated the inhibitory effects of water-soluble phenolic compounds (WSPCs) in the coat of after-ripening wheat (Triticum aestivum L.) seeds on the processes of germination and peroxidase reactivation. Wheat bran has a WSPC content of 862.5 μg gallic acid equivalent g−1 dry weight. When seeds were incubated in the water extract of bran, germination, peroxidase reactivation, and coleoptile and radicle growth were suppressed in a WSPC concentration-dependent manner. The inhibitory effects were significantly ameliorated by removing WSPCs from bran extract by treating with 1% insoluble polyvinylpolypyrrolidone. Pretreatment of seeds with 0.1% H2O2 reduced the WSPC content in the coat, which was confirmed using Fourier transform infrared microspectroscopy. With H2O2 pretreatment, seed germination, peroxidase reactivation, and post-germination seedling growth were significantly stimulated. Application of the known phenolics caffeic acid, feruic acid, or vanillin to the germination medium blocked seed germination and suppressed peroxidase reactivation. The results described here indicate that WSPCs act as endogenous inhibitors in the coat to control germination of Triticum aestivum seeds, and that inhibition of germination is at least partially caused by suppressing peroxidase reactivation.  相似文献

Breakdown of gliadin during germination of xHaynaldoticum sardoum Meletti et Onnis seeds is correlated with the appearance in the endosperms of a proteinase activity, which is absent in the quiescent seed. This activity is optimal at pH 4 and has a maximum stability at pH 4–5. Gel filtration of proteinase activity extracted from quiescent seeds indicates a molecular weight of 60–100 kDa. The proteinase can hydrolyze hemoglobin but not gliadin and is inhibited by pepstatin A and, to a lower extent, by p -chloromercuribenzoic acid (p-CMB). Gel filtrations of crude extracts from germinating seeds reveal two peaks (molecular weight 66 and 21 kDa) of activity against hemoglobin and a shoulder and a peak (molecular weight 21 kDa) of activity on gliadin. The first peak of activity against hemoglobin is inhibited by pepstatin A and p-CMB; the second one is inhibited by p-CMB and leupeptin. As for the gliadin-eluted activity the shoulder is mainly inhibited by pepstatin A and p-CMB, whereas the peak is inhibited by p-CMB and leupeptin. Estimations of the ratios of total nitrogen to α-amino nitrogen, suggest that the enzyme preparations mainly contain proteinases. It is concluded that the proteinases present in the quiescent seeds of xH. sardoum , in particular aspartic proteinases (EC 3.4.23), could play a role as initiator endoproteases or participate in the digestion of modified proteins during the mobilization of reserve proteins. The cysteine proteinases (EC 3.4.22) appearing during the germination seem to account for the hydrolysis of the most abundant class of protein reserves, the prolamins.  相似文献

以野鸦椿(Euscaphis japonica)种壳和胚为材料,甲醇浸提得到种壳浸提液和胚浸提液,配制浸提液浓度梯度为原浸提液浓度的10%、20%、30%、40%,研究不同浓度的种壳和胚浸提液对白菜、小麦、绿豆种子底物酶活性、发芽率、幼苗根长和苗高的影响,萃取和分离种壳和胚甲醇浸提液中的内源抑制物质,探讨野鸦椿种子内源抑制物质的活性与成分。结果表明:随着种壳和胚浸提液浓度的增加,白菜种子酸性磷酸酶活性和发芽率均显著降低(P0.05),表现为抑制作用递增,种壳的抑制作用小于胚,而幼苗的根长和苗高则表现为低促高抑,在10%浸提液处理下根长和苗高达最大值,种壳的促进效果弱于胚;小麦种子淀粉酶活性及幼苗的根长和苗高递减(P0.05),表现为抑制作用递增,而发芽率则在浓度≤20%时差异不明显(P0.05),浓度为30%时显著降低(P0.05),40%时发芽率为0,种壳的抑制作用大于胚;绿豆种子蛋白酶活性、发芽率、幼苗根长和苗高均在浸提液浓度≥20%时显著下降(P0.05),且种壳的作用效果小于胚。种子内源抑制物萃取及分离表明,外壳中含酚酸类较胚多,含碱类较胚少。综上认为,野鸦椿种壳和胚中均含有较高活性的内源抑制物,但性质、成分及含量存在差异,外壳内源抑制物主要作用对象为淀粉类物质,胚乳内源抑制物主要作用于油脂类和蛋白类物质。  相似文献

We analysed changes in AMP, ADP, and ATP concentrations and adenylate energy charge in Norway maple (Acer platanoides L.) and European beech (Fagus sylvatica L.) seeds during dormancy breaking (at 3 °C) and in the control variant at 15 °C. Values of the studied indicators in stratified beech seeds were generally higher at 15 °C, at least until germination (+3 °C). By contrast, in maple seeds, the values recorded during dormancy breaking by cold stratification were much higher than at 15 °C. Three peaks (usually in weeks 3, 6, and 8) were observed in maple seeds at 3 °C, but not at 15 °C. Among adenine nucleotides, AMP reached the highest levels in both species in both variants of the experiment.  相似文献

Seed germination behavior in two types of dimorphic fruits inSalsola komarovii Iljin (Chenopodiaceae) was studied. The germination percentage was much higher in the long-winged type than in the short-winged type when the fruits were tested for germinability. If lignified perianths were removed from the fruits, seeds of both types germinated at higher percentages and the difference in the germination percentage between two types decreased. The extract of the perianths inhibited the seed germination. Abscisic acid was detected in the extract of the perianths by enzyme-linked immunosorbent assay and its level was higher in the short-winged type than in the long-winged type. Germinability in both types was lost one year after harvest when the fruits were stored at room temperature and the germination was tested with fruits. When the seeds were tested, however, the short-winged type germinated even two years after the harvest; the germinability of the short-winged type lasted one year longer than that of the long-winged type. Light irradiation was not required for the seed germination in either type regardless of presence or absence of the perianths. No clear difference between the two types was observed either in optimal temperature for the germination or in strength of the resistance to salinity.  相似文献

Mimosa bimucronata is a pioneering tree that occurs predominantly in moist lowlands, floodplains and on margins of rivers and lakes in Latin America. The effect of submergence on seed germination in M. bimucronata was firstly studied. Patterns of water absorption by M. bimucronata seeds were investigated thereafter to assess the imbibition phases of scarified and unscarified seeds. The germination percentage was significantly higher in scarified than in unscarified seeds, and the velocity of seed germination also increased considerably in scarified seeds. Submergence duration did not significantly affect germination percentages of scarified and unscarified seeds. Therefore, seed viability after submersion suggests that M. bimucronata may display hydrochorous dispersal and also that seeds are able to germinate successfully in areas with frequent seasonal flooding. With respect to imbibition phases, phase II was very short or even absent for scarified and unscarified seeds; therefore, a plateau, where water absorption by seeds is established, was not observed. Finally, we verified that the passage from phase I to III was very tenuous and took a long time in seeds without scarification.  相似文献

以西南地区具有代表性的16种绿肥植物为受体材料,采用培养皿药膜法研究了铁核桃(Juglans sigillata)根水浸提液对受体种子发芽率及幼苗鲜重、干重的化感效应;并进一步研究了铁核桃根、叶水浸提液和胡桃醌对化感效应存在明显差异的4种绿肥植物(绿豆、红三叶、白三叶、花生)种子萌发与幼苗生长以及抗氧化酶特性的影响,以筛选适宜中国西南地区核桃园种植的绿肥植物,探讨核桃根和凋落物对绿肥作物的化感作用机制。结果表明:(1)铁核桃根水浸提液对绿豆的发芽率没有影响,但对绿豆幼苗鲜重和干重有显著抑制作用,而对其他15种绿肥的发芽率和鲜重、干重均有抑制作用。(2)胡桃醌显著抑制绿豆种子萌发,而铁核桃根或叶水浸提液对绿豆种子萌发没有影响。(3)铁核桃根或叶水浸提液以及胡桃醌对绿肥植物幼苗生长的化感效应趋势一致,但核桃根或叶水浸提液的化感效应强于胡桃醌。(4)绿豆幼苗在铁核桃根或叶水浸提液以及胡桃醌处理下,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)的活性均高于其他3种(红三叶、白三叶、花生)受体幼苗,表明绿豆清除活性氧能力高,细胞受损害程度较低,受化感作用影响最弱。研究认为,绿豆为适宜中国西南地区幼龄核桃园种植的间作绿肥植物。  相似文献

Intercropping of Vigna mungo (black gram) isprevalent for varied crop regimes in subtropical northern plains of India. Insugarcane, the negative impacts of a black gram intercrop on crop yields,rhizosphere soil properties and benefit: cost ratio have been reported. Thepossible allelopathic potential of black gram seeds on the germination,seedlingemergence and root elongation of other commercial crops is reported here. Theeffects of imbibed seeds of black gram, their aqueous leachate and seedextractson the germination and root elongation of seeds of four test crops viz. wheat,maize, gram and lentil, both under aseptic and soil conditions, wereinvestigated. Inhibitory effects were evident in all the bioassays of seeds ofthese crops. The black gram seeds directly and leachates were highly inhibitorybut the aqueous seed extract was less so. The observations indicate a releaseofinhibitory substances from black gram seeds leading to the observed inhibitoryeffects.  相似文献

Seed dispersal by animals is one of the most important plant-animal mutualisms, but saurochory, the dispersal of seeds by reptiles, has received little attention. We investigated the role of the Florida box turtle (Terrapene carolina bauri) as a seed dispersal agent in pine rockland forests of the lower Florida Keys and examined the effect of turtle digestion on seed germination. We obtained seeds of 11 species with fleshy fruits and 2 species with non-fleshy fruits (a grass and legume) from the feces of 145 box turtles collected on Key Deer National Wildlife Refuge from 1999 to 2000. We planted the seeds of nine species and germination percentage (percentage of seeds that germinated during the experiment) varied from 10% to 80%. Comparative germination experiments were conducted with Thrinax morrissii, Serenoa repens, and Byrsonima lucida. We compared the germination percentage and germination rate (number of days from planting to seedling emergence) of seeds from three treatments (seeds recovered from feces, control seeds with pulp, and control seeds without pulp) and continued these experiments for up to 2 years. Passage through the box turtle digestive tract greatly enhanced the germination percentage and germination rate of S. repens, but decreased the germination percentage of B. lucida and T. morrissii, and decreased germination rate for T. morrissii. Subsequent destructive seed viability tests revealed that many ungerminated T. morrissii seeds remained viable, suggesting long-term seed dormancy may occur, even after passage through the turtle digestive system. In addition, the proportion of ungerminated seeds which remained viable was greater for seeds recovered from turtle feces than from control seeds with pulp. Furthermore, removal of fleshy pulp either manually or by the turtle digestive system may allow T. morrissii to escape insect predation.  相似文献

The changes in α-galactosidase activity in guar (Cyamopsis tetragonolobus) seeds was followed during seven days of germination. The enzyme activity was maximal on the first day of germination and gradually decreased during subsequent days. On the second day of germination the partially purified enzyme upon ion-exchange chromatography on CM-Sephadex C-50 was resolved into α-galactosidase-A (anionic), α-galactosidase-C1 (cationic) and α-galactosidase-C2 (cationic) and their relative proportions were 28,12 and 60%, respectively. The combined α-galactosidase C1 and C2 activities increased in the first two days of germination followed by significant decrease after the 3rd day onwards, whereas α-galactosidase-A remained fairly constant throughout the germination period, α-Galactosidase-A and C2 had differentK m and Vmax values withp-nitrophenyl α-D-galactopyranoside, raffinose and melibiose as substrates and also differed in their thermal stabilities  相似文献

The endopeptidase of the highest electrophoretic mobility was the main endopeptidase hydrolyzing gliadin in the endosperm of germinated triticale (X Triticosecale Wittm.) grains after three days of imbibition. Activity of this endopeptidase, named EP8 starts to be detectable after two days of imbibition. The appearance of its activity in the endosperm on a second day of imbibition may suggest that EP8 is synthesized in aleurone during germination and/or secreted into the starchy endosperm as an inactive polypeptide during grains development and then activated. EP8 was isolated from the endosperm of germinating triticale seeds and purified 257-fold using ammonium sulphate, ion exchange chromatography on DEAE Sepharose CL-6B and gel filtration on Sephadex G-100. The enzyme was totally inhibited by E-64—class-specific cysteine proteinases inhibitor and activated by thiol compounds. Molecular weight estimated by SDS-PAGE was 39.5 kDa. The optimum pH for the hydrolysis of gliadin was 4.2 and for hemoglobin 5.2. High activity of EP8 against wheat gliadin in vitro suggests that this cysteine endopeptidase plays a major role in the mobilization of storage proteins in the endosperm of germinating triticale grains.  相似文献

The effects of an aqueous plant-derived smoke extract, octanoic acid and ethylene on germination of light-sensitive Grand Rapids lettuce seeds were investigated. The smoke extract brought about a concentration dependent increase in germination and a complete inhibition of germination at high concentrations. Octanoic acid could not induce germination. Ethylene at concentrations over 5 L L–1 increased lettuce seed germination, but not to the same degree as smoke. Aqueous smoke in combination with ethylene showed a synergistic effect on germination at suboptimal smoke concentrations. At high smoke concentrations the effect of ethylene was almost completely inhibited. Octanoic acid in combination with ethylene brought about a higher level of germination than with ethylene alone, but only at the highest concentration of octanoic acid tested (1 mM). Standardized hexane and dichloromethane-partitioned smoke extracts and octanoic acid were subjected to TLC separation. The R f -fractions in the smoke lanes showing activity in the lettuce seed bioassay did not correspond to the R f -value of octanoic acid. As aqueous smoke can withstand autoclaving and can be separated by TLC and HPLC without loosing activity it is unlikely that the activity of aqueous smoke is linked to ethylene. The spermine, spermidine, and the total protein content of embryos or embryonic axes from Triticum durum, Helianthus annuus, and Glycine max seeds at different times of early germination was evaluated. Mitotic activity of root-tip meristems from germinating seeds was also determined. Summary Seed set and fruit development in cucumber (Cucumis sativus L.) were studied in relation to female flower receptivity from day — 2 before anthesis to day + 2 after anthesis. The female cucumber flower is protogynous. The pistil was receptive 2 days before anthesis. The iso-electric focusing (IEF) patterns of the stigma/style proteins, were identical from day -5 to day +2. In pollinated flowers in vivo germination and pollen-tube growth in the ovary were affected by pistil age from day -2 to day +2. In addition, differences in sectorial filling in full seeds were observed within the fruits. A negative correlation was observed between the frequency of fertilized ovules in the pedoncular part of the fruit and ovary length at the time of pollination. In the whole fruit, significant differences in the number of full seeds and fruit size at maturity were found, and these were observed to be correlated with the various stages of female flower maturation at pollination. The day -2 and day +2 stages yielded the smallest fruits with few full seeds compared to the day -1, day 0 and day +1 stages, which had the biggest fruits and a large number of full seeds. A strong positive correlation was found between total seed number (including full and empty seeds), fruit length and weight at maturity. All these results suggest that both seed set in the different parts of the fruit and fruit development are controlled by ovular receptivity rather than by stigma/style receptivity.  相似文献

Extracts of the brown seaweed Ascophyllum nodosum have been used as a biostimulant to promote growth and productivity in a number of agricultural production systems. Although the extracts have been shown to improve seedling emergence and vigor in a variety of plants, including barley, the mechanism(s) of this growth-promoting effect is(are) largely unknown. In our study, A. nodosum extract induced amylase activity in barley seed-halves; a significant difference in amylase activity was observed in seeds without an embryo. The addition of activated charcoal to the treatment media negated the bioactivity of the extracts suggesting the organic nature of bioactive compounds in A. nodosum extracts. The extracts induced amylase activity in a gibberellic acid (GA)-deficient barley mutant (grd2). LC-MS-MS analysis failed to detect the presence of GA3 in the extracts. ABA supplementation of the medium caused a significant reduction of amylase activity in GA-treated seeds compared with those treated with the A. nodosum extract. Taken together, our results suggest that the organic components of A. nodosum extract induce amylase activity independent of GA3 and might act in concert with GA-dependent amylase production leading to enhanced germination and seedling vigor in barley. Being derived from a renewable resource, the bioactive compounds from A. nodosum could be used to improve crop productivity in sustainable agricultural systems.  相似文献

In addition to effects mediated by resource competition, some invasive plants may impact surrounding vegetation by secreting compounds that are directly inhibitory to growth. Lonicera maackii, an invasive Asian shrub of forests and open areas in eastern and midwestern North America, has devastating effects on understory vegetation, some of which persist even after this shrub is removed. In this study, we explored the potential of aqueous extracts of the leaves and roots of this plant to inhibit seed germination of Impatiens capensis, Alliaria petiolata, Arabidopsis thaliana, and L. maackii in Petri dish bioassays. Both L. maackii root and leaf extracts significantly decreased germination in the three herb species. This inhibitory effect generally increased with increasing extract concentration and was more pronounced with application of leaf extract than root extract. However, when the same extracts were applied to seeds of L. maackii itself, germination was delayed in some cases, but was not significantly reduced by the end of the experiment. Germination of L. maackii seeds even reached significantly higher levels in some extract treatments than in no-extract controls. This implies that L. maackii can successfully inhibit the germination of other plants with few autotoxic effects and may even promote the germination of its own seeds.  相似文献

Biological soil crusts dominated by drought-tolerant mosses are commonly found through arid and semiarid steppe communities of the northern Great Basin of North America. We conducted growth chamber experiments to investigate the effects of these crusts on the germination of four grasses: Festuca idahoensis, Festuca ovina, Elymus wawawaiensis and Bromus tectorum. For each of these species, we recorded germination time courses on bare soil and two types of biological soil crusts; one composed predominantly of the tall moss Tortula ruralis and the other dominated by the short moss Bryum argenteum. On the short-moss crust, the final germination percentage was about half of that on bare soil. Also, the mean germination time was 4 days longer on short-mosses than on bare soil. In contrast to the short-moss crust, the tall-moss crust did not reduce the final germination percentage but increased the mean germination time. Similar results were observed in the four grasses studied. To investigate the mechanism by which moss crusts affected germination, we analyzed the water status of seeds on bare soil and moss crusts. Six days after seeding, the water content of seeds on bare soil was approximately twice that of seeds on tall- or short-moss crust. Analysis of the time course of changes in seed weight and water potential in Bromus tectorum revealed that overtime seeds on tall mosses reached higher water content than those on short mosses. The increase in the water content of seeds on tall mosses occurred as the seeds gradually fell through the moss canopy. Taken together, our results indicate that biological soil crusts with distinct structural characteristics can have different effects on seed germination. Furthermore, this study revealed that a biological soil crust dominated by short mosses had a negative effect on seed water status and significantly reduced seed germination.  相似文献
