A yogurt culture (Streptococcus thermophilus 15HA + Lactobacillus delbrueckii subsp. bulgaricus 2-11) was studied in conditions of aerobic batch fermentation (10–40% dissolved oxygen in milk). The growth and acidification of S. thermophilus 15HA were stimulated at 20% oxygen concentration and the lactic acid process in a mixed culture was shortened by 1 h (2.5 h for the aerobic culture and 3.5 h for the anaerobic mixed culture). Streptococcus thermophilus 15HA oxygen tolerance was significantly impaired at oxygen concentrations in the milk above 30%. Though S. thermophilus 15HA was able to overcome to some extent the impact of high oxygen concentration (40%), the lactic acid produced was insufficient to coagulate the milk casein (4.0 g lactic acid l−1 in the mixed culture and 3.8 g lactic acid l−1 in the pure culture). A dramatic decrease in the viable cell count of L. delbrueckii subsp. bulgaricus 2-11 in the pure and mixed cultures was recorded at 30% dissolved oxygen. Leaf pigments, such as chlorophyll and carotenoids, are essential plant molecules. They provide carbohydrates and energy during all plant life. Leaf pigments are also important parameters of decorative plants, such as floriculture items, cut foliage and flowers. Leaf yellowing is a form of senescence caused by an internal hormone imbalance, such as a lack of cytokinins. The aim of this study was to investigate the changes in total carotenoids and endogenous ABA in cut flower stock leaves during post-harvest life. The effect of pulse treatment with 5 or 10 M thidiazuron (TDZ), 150 mg l–1 8-hydroxyquinoline sulphate (8-HQS) and combinations of TDZ with 8-HQS on total carotenoids and ABA concentration was also investigated. Results showed that total carotenoids drastically decreased from 1548 g cm–2, until reaching 565 g cm–2 at the end of vase life. Endogenous ABA strongly increased at the same time, going from 167 ng g–1 DW at the beginning of the experiment to 1322 ng g–1 DW at the end of vase life. The TDZ inhibited carotenoid degradation, but did not affect the ABA concentration, while the 8-HQS did not prevent carotenoid degradation and the ABA concentration was only slightly affected. ABA seems to be a secondary senescence bio-product that may have a synergic effect with other senescence inducers dramatically accelerating leaf senescence.  相似文献

Exopolysaccharide (EPS) preparations from Lactobacillus delbrueckii ssp. bulgaricus (L. bulgaricus) strains LBB.B26 and LBB.B332 and Streptococcus thermophilus strains LBB.T54 and LBB.T6V were characterized using ion-exchange chromatography and gel filtration. All four preparations contained a neutral EPS with molecular mass in the range of 1.3−1.6 × 106 Da (HMM-EPS). The EPS preparations from the two L. bulgaricus strains also contained an acidic low molecular mass EPS fraction (LMM-EPS) comprising from 10% to 34% of the total EPS yield. HMM-EPS preparations were subjected to High Pressure Liquid Chromatography (HPLC) analysis of monomer sugars after complete hydrolysis. Glucose, galactose and/or rhamnose in different ratios proved to be the principal sugars building the HMM-EPS from all four strains. The chemical composition of HMM-EPS was strictly strain-specific. The LMM-EPS contained galactose. The viscosifying properties of the four different HMM-EPS varied greatly with intrinsic viscosity in the range from 0.26 (strain B26) to 2.38 (strain T6V). For 24 h the two L. bulgaricus strains accumulated more HMM-EPS in milk (>70 mg l−1) than S. thermophilus strains T54 and T6V (<30 mg l−1), but maximal yields were reached earlier with cocci (8 h) than with rods (16–24 h). The contribution of HMM-EPS production to increased viscosity of fermented milk was demonstrated for all of the tested strains grown as monocultures or as mixed yogurt starters compared to non-EPS producing S. thermophilus LBB.A and poor EPS-producer L. bulgaricus LBB.B5. The extent of increased viscosity was strongly dependent on the nature of the produced HMM-EPS, rather than simply on polymer yield.  相似文献

The effects of eight microelements (I, BO3 3–, MoO4 2–, Co2+, Cu2+, Mn2+, Fe2+, Zn2+) on the biosynthesis of camptothecin and the growth of suspension cultures of Camptotheca acuminata were studied. The increase of I to 25 M l–1, Cu2+ to 1 M l–1, Co2+ to 2 M l–1 and MoO4 2– to 10 M l–1 in Murashige and Skoog (MS) medium resulted in 1.66, 2.84, 2.53 and 2.04 times higher of camptothecin yield than that in standard MS medium respectively. Combined treatment of I (25 M l–1), Cu2+ (1 M l–1), Co2+ (2 M l–1) and MoO4 2– (10 M l–1) lead to improve cell dry weight, camptothecin content, and camptothecin yield to 30.56 g l–1, 0.0299%, and 9.15 mg l–1, respectively, which were 20.2, 208.9 and 273.8% increment respectively when compared with those of control.  相似文献

Summary The principles of a new process for the continuous manufacture of yoghurt withLactobacillus bulgaricus andStreptococcus thermophilus entrapped in Ca-alginate beads were given. Characteristics of non-optimized pH-stat continuous stirred tank reactors withL. bulgaricus andS. thermophilus entrapped in the same or separate particles were examined. In these reactors the highest production rate were 9.4 g.l–1.h–1 for lactic acid and 3.4×1011 C.F.U. 1–1.h–1 for inoculation when microorganisms were entrapped separately. A stable balance of the yoghurt bacteria liberated in continuously prefermented milk was observed.  相似文献

Streptococcus thermophilusand Lactobacillus bulgaricus were co-immobilized in different systems with varying calcium (0.1–1.5M) and alginate (1–2<><>, w/v) concentrations. Highest lactic acid production was 35 g l1 when both bacteria were in high viscosity beads (1<><>, w/v alginate) hardened in 0.1 M CaCl2 .The gel bead composition affected size and distribution of entrapped lactic acid bacteria.  相似文献

Catabolites related to tricarboxylic acid cycle affected growth and carotenogenesis in Phaffia rhodozyma. Glutamate, glutamine, aspartate, asparagine and proline at 75 mM of N increased biomass from 2 g l–1 to 2.9–4.7 g l–1 but decreased carotenoid from 420 g g–1 yeast to 200–260 g g–1 yeast in strain 67-385. However, simple nitrogen sources did not decrease carotenoid formation. Tricarboxylic acid intermediates repressed carotenogenesis to a less degree than the corresponding amino acids. Carotenoid hyper-producing mutants were impaired in nitrogen utilization. These results indicated that nitrogen assimilation and the concentrations of tricarboxylic acid cycle intermediates are involved in regulation of carotenoid biosynthesis.  相似文献

Excised root cultures of Gloriosa superba reached 7.5 g dry wt l–1 and accumulated 240±40 g colchicine g–1 cell dry wt after 4 weeks growth. While all precursors (except trans-cinnamic acid) enhanced colchicine content of root cultures without adversely affecting root growth, treatment with p-coumaric acid + tyramine (each at 20 mg l–1) increased colchicine content to 1.9 mg g–1 cell dry wt.  相似文献

Loblolly pine (Pinus taeda L.) somatic embryogenesis initiation was improved by supplementing the initiation medium with the pH buffer agent 2(n-morpholino)ethanesulphonic acid (MES) at 250 mg l–1, folic acid at 0.5 mg l1, and biotin at 0.05 mg l–1. MES and vitamins increased the percentage of explants with extruded tissue that continued the initiation process to form embryogenic tissue. The increase in initiation was about 12%. Initiation of 12 open-pollinated families averaged 38.5%, which is 16% higher than initiation on medium without these additives. When tested with 18 control-pollinated families, initiation averaged 26.3%. Basal medium contained a combination of modified 1/2 P6 salts, activated carbon (AC) at 50 mg –1, Cu and Zn adjusted to compensate for adsorption by AC, 1.5% maltose, 2% myo-inositol, 500 mg l–1 casamino acids, 450 mg l–1 glutamine, 2 mg l–1 NAA, 0.63 mg l–1 BAP, 0.61 mg l–1 kinetin, 3.4 mg l–1 silver nitrate, 10 M 8-Br-cGMP, 0.1 M brassinolide, and 2 g l–1 Gelrite. Early-stage embryo growth and initiation in Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) were also improved in the presence of these additives.  相似文献

The lobster (Homarus americanus) hepato-pancreatic epithelial baso-lateral cell membrane possesses three transport proteins that transfer calcium between the cytoplasm and hemolymph: an ATP-dependent calcium ATPase, a sodium-calcium exchanger, and a verapamil-sensitive cation channel. We used standard centrifugation methods to prepare purified hepato-pancreatic baso-lateral membrane vesicles and a rapid filtration procedure to investigate whether 65Zn2+ transfer across this epithelial cell border occurs by any of these previously described transporters for calcium. Baso-lateral membrane vesicles were osmotically reactive and exhibited a time course of uptake that was linear for 10–15 s and approached equilibrium by 120 s. In the absence of sodium, 65Zn2+ influx was a hyperbolic function of external zinc concentration and followed the Michaelis-Menten equation for carrier transport. This carrier transport was stimulated by the addition of 150 M ATP (increase in Km and Jmax) and inhibited by the simultaneous presence of 150 mol l–1 ATP+250 mol l–1 vanadate (decrease in both Km and Jmax). In the absence of ATP, 65Zn2+ influx was a sigmoidal function of preloaded vesicular sodium concentration (0, 5, 10, 20, 30, 45, and 75 mmol l–1) and exhibited a Hill Coefficient of 4.03±1.14, consistent with the exchange of 3 Na+/1Zn2+. Using Dixon analysis, calcium was shown to be a competitive inhibitor of baso-lateral membrane vesicle 65Zn2+ influx by both the ATP-dependent (Ki=205 nmol l–1 Ca2+) and sodium-dependent (Ki=2.47 mol l–1 Ca2+) transport processes. These results suggest that zinc transport across the lobster hepato-pancreatic baso-lateral membrane largely occurred by the ATP-dependent calcium ATPase and sodium-calcium exchanger carrier proteins.Communicated by: I.D. Certain yeast cells on solid nutrient medium produced colonies surrounded by a light zone of selenite absorption. This screening procedure resulted in the selection of 22 strains out of 200 isolates with different Se4+-absorbing capacity ranging from 16 to 98.8 g Se4+ g–1 l–1 h–1. The highest rate of Se4+ elimination from the Na2SeO3 solution was observed with an oval shaped, cream pigmented fermentative yeast, tentatively called Candida sp. strain MS4. This strain was isolated from wastewater and found to accumulate selenium oxyanions. Se4+ uptake involved both inactive and active phenomena. The amounts of selenium (initial concentration 2 mg Se4+ l–1) removed from aqueous solution by inactive and active phenomena were 667 g Se4+ g–1 l–1, and 1580 g Se4+ g–1 l–1, respectively. Cell suspension cultures of Agave amaniensis and Costus speciosus were grown in media containing Cd2 + up to 25 and 20 mg l–1, respectively, and Pb2+ up to 40 mg l–1. The cultures hyper-accumulated Cd2+ up to 900 and 530 g g–1 and Pb2+ up to 1390 and 1170 g g–1 dry wt. in their respective biomasses. Increasing Pb2+ up to 30 mg l–1 increased the biomass production and total sitosterol content of Costus speciosus by up to 1.7- and 1.3-fold, respectively.  相似文献

We examined transepithelial transport of Ca2+ across the isolated opercular epithelium of the euryhaline killifish adapted to fresh water. The opercular epithelium, mounted in vitro with saline on the serosal side and fresh water (0.1 mmol·l–1 Ca2+) bathing the mucosal side, actively transported Ca2+ in the uptake direction; net flux averaged 20–30 nmol·cm–2·h–1. The rate of Ca2+ uptake varied linearly with the density of mitochondria-rich cells in the preparations. Ca2+ uptake was saturable, apparent K 1/2 of 0.348 mmol·l–1, indicative of a multistep transcellular pathway. Ca2+ uptake was inhibited partially by apically added 0.1 mmol·l–1 La3+ and 1.0 mmol·l–1 Mg2+. Addition of dibutyryl-cyclic adenosine monophosphate (0.5 mmol·l–1)+0.1 mmol·l–1 3-isobutyl-l-methylxanthine inhibited Ca2+ uptake by 54%, but epinephrine, clonidine and isoproterenol were without effect. Agents that increase intracellular Ca2+, thapsigargin (1.0 mol·l–1, serosal side), ionomycin (1.0 mol·l–1, serosal side) and the calmodulin blocker trifluoperazine (50 mol·l–1, mucosal side) all partially inhibited Ca2+ uptake. In contrast, apically added ionomycin increased mucosal to serosal unidirectional Ca2+ flux, indicating Ca2+ entry across the apical membrane is rate limiting in the transport. Verapamil (10–100 mol·l–1, mucosal side), a Ca2+ channel blocker, had no effect. Results are consistent with a model of Ca2+ uptake by mitochondria rich cells that involves passive Ca2+ entry across the apical membrane via verapamil-insensitive Ca2+ channels, intracellular complexing of Ca2+ by calmodulin and basolateral exit via an active transport process. When grown photoautotrophically, Chlorella zofingiensis strain CCAP 211/14 accumulates a significant amount of valuable carotenoids, namely astaxanthin and lutein, of increasing demand for use as feed additives in fish and poultry farming, as colorants in food, and in health care products. Under standard batch-culture conditions, this microalgal strain exhibits high values of both growth rate (about 0.04 h–1) and standing cell population (over 1011 cells l–1, or 7 g dry weight l–1). Lutein, in a free (unesterified) form, was the prevalent carotenoid during early stages of cultivation (over 0.3 pg cell–1, equal to 4 mg g–1 dry weight, or 20 mg l–1 culture), whereas esterified astaxanthin accumulated progressively, to reach a maximum (over 0.1 pg cell–1, equal to 1.5 mg g–1 dry weight, or 15 mg l–1 culture) in the late stationary phase. A differential response of lutein and astaxanthin accumulation was also recorded with regard to the action of some environmental and nutritional factors. C. zofingiensis CCAP 211/14 represents a unique model system for analyzing the differential regulation of the levels of primary (lutein) and secondary (astaxanthin) carotenoids. Relevant also from the biotechnological viewpoint, this photosynthetic organism, with outstanding attributes for fast photosynthetic growth and carotenoid accumulation, might prove most valuable for its application to the mass production of either or both lutein and astaxanthin.  相似文献

Isolated embryos ofKarwinskia humboldtiana were cultured in vitro. The growth of embryos and development to plantlets on woody plant medium supplemented with indole-3-acetic acid 6.10-2 mol l–1, gibberellic acid (GA3) 3.10-2 mol l–1, and 6-benzylaminopurine (BA) 2 mol l–1 was obtained. Multiplication of shoots and rooting of excised shoots has been achieved. Sodium concentrations as low as 2 mM exerted a significant protective effect on the high-pressure inactivation (160–210 MPa) of Rhodotorula rubra at pH 6.5, but not on two other yeasts tested (Shizosaccharomyces pombe and Saccharomyces cerevisiae). A piezoprotective effect of similar magnitude was observed with Li+ (2 and 10 mM), and at elevated pH (8.0–9.0), but no effect was seen with K+, Ca2+, Mg2+, Mn2+, or NH4 +. Intracellular Na+ levels in cells exposed to low concentrations of Na+ or to pH 8.0–9.0 provided evidence for the involvement of a plasma membrane Na+/H+ antiporter and a correlation between intracellular Na+ levels and pressure resistance. Summary From acetylene reduction assays over a 10-month period starting in April 1979, nodule activities averaged 18.78 (se 4.67) moles C2H4 g nodule dw–1 h–1 forAlnus rubra and 59.95 (se 12.14) moles C2H4 g nodule dw–1 h–1 forCytisus scorparius. Plant rates were 1.91 (se. 47) moles C2H4 plant–1 h–1 forA. rubra and 0.55 (se. 17) moles C2H4 plant–1 h–1 forC. Scoparius. Plant activity and total leaf N were strongly correlated with the dw of other plant parts, but nodule activity and percent leaf N were not. Plant and nodule activities were not associated with temperature, moisture stress, precipitation events or percent light for either species over the growing season nor for 54A. rubra sampled in mid-season 1979 on one replication. After 5 to 6 growing seasons, 14A. rubra on the same site ranged from 30 to 332 cm in height and showed strong correlation between nodule dw, leaf dw, plant size and total leaf N. Results from this study and others indicate logistic equations may be modified to predict the effect of adding a N2 fixing plant to a population of non N2 fixing trees.  相似文献

Direct regeneration from explants without an intervening callus phase has several advantages, including production of true type progenies. Axillary bud explants from 6-month-old sugarcane cultivars Co92061 and Co671 were co-cultivated with Agrobacterium strains LBA4404 and EHA105 that harboured a binary vector pGA492 carrying neomycin phosphotransferase II, phosphinothricin acetyltransferase (bar) and an intron containing -glucuronidase (gus-intron) genes in the T-DNA region. A comparison of kanamycin, geneticin and phosphinothricin (PPT) selection showed that PPT (5.0 mg l–1) was the most effective selection agent for axillary bud transformation. Repeated proliferation of shoots in the selection medium eliminated chimeric transformants. Transgenic plants were generated in three different steps: (1) production of putative primary transgenic shoots in Murashige-Skoog (MS) liquid medium with 3.0 mg l–1 6-benzyladenine (BA) and 5.0 mg l–1 PPT, (2) production of secondary transgenic shoots from the primary transgenic shoots by growing them in MS liquid medium with 2.0 mg l–1 BA, 1.0 mg l–1 kinetin (Kin), 0.5 mg l–1 -napthaleneacetic acid (NAA) and 5.0 mg l–1 PPT for 3 weeks, followed by five more cycles of shoot proliferation and selection under same conditions, and (3) rooting of transgenic shoots on half-strength MS liquid medium with 0.5 mg l–1 NAA and 5.0 mg l–1 PPT. About 90% of the regenerated shoots rooted and 80% of them survived during acclimatisation in greenhouse. Transformation was confirmed by a histochemical -glucuronidase (GUS) assay and PCR amplification of the bar gene. Southern blot analysis indicated integration of the bar gene in two genomic locations in the majority of transformants. Transformation efficiency was influenced by the co-cultivation period, addition of the phenolic compound acetosyringone and the Agrobacterium strain. A 3-day co-cultivation with 50 M acetosyringone considerably increased the transformation efficiency. Agrobacterium strain EHA105 was more effective, producing twice the number of transgenic shoots than strain LBA4404 in both Co92061 and Co671 cultivars. Depending on the variety, 50–60% of the transgenic plants sprayed with BASTA (60 g l–1 glufosinate) grew without any herbicide damage under greenhouse conditions. The production of extracellular enzymes by the thermophilic fungus Thermomyces lanuginosus was studied in chemostat cultures at a dilution rate of 0.08 h–1 in relation to variation in the ammonium concentration in the feed medium. Under steady state conditions, three growth regimes were recognised and the production of several extracellular enzymes from T. lanuginosus was recorded under different nutrient limitations ranging from nitrogen limitation to carbon/energy limitation. The range and the production of carbohydrate hydrolysing enzymes and lipase increased from Regime I (NH4Cl 600 mg l–1) to Regime III (NH4CI 1200 mg l–1), whereas production of protease was highest in Regime II (600 mg l–1 < NH4Cl <1200 mg l–1).  相似文献

An in vitro plant regeneration protocol of Cymbidium faberi from immature seeds was established. The immature seeds of 50 days old started to form rhizomes 4 months after they were cultured on hormone free medium. The rhizomes multiplied 5 times when subcultured on the medium containing 1.0 mg l–1 -naphthalene acetic acid (NAA) for 40 days and more than 90% of the rhizomes initiated shoots within 60 days on the media containing 0.5 or 1.0 mg l–1 NAA plus 2.0 or 5.0 mg l–1N6-benzylaminopurine (BA). Plantlets were regenerated when the shoots were planted on the basal medium amended with 1 g l–1 activated charcoal for 50 days and the plantlets grew normally after transplanting.  相似文献
