The effect of brassinolide (BL) on cultured calluses of Spartina patens (Ait.) Muhl. (Poaceae), a halophyte monocot was studied. BL at 0.03–0.04 mg l–1 at fixed concentrations of IAA (0.2 mg l–1) and BA (3.0 mg l–1) added in MS medium increased the ratio for fresh weight (CIRFW) to dry weight (CIRDW) by 96–111% and 235–326%. Similarly, in callus regeneration capacity, BL at 0.03 mg l–1 was most effective, increasing the shoot regeneration ratio (SRR) by 425%. BL at 0.04 mg l–1 had not such an increasing effect as BL at 0.03 mg l–1, which increased SRR by 79%. However, BL at 0.005 mg l–1 promoted regenerated shoot growth most significantly, increasing the shoot height increasing ratio (SHIR) by 395% after a 40-day culture. BL at 0.05 mg l–1 was least effective in the callus regeneration and regenerated shoot growth, decreasing SRR by 27% and SHIR by 52%. Present results suggest that BL at 0.03 mg l–1 is suitable for the callus growth and shoot regeneration, while BL at 0.005 mg l–1 effectively enhanced the regenerated shoot growth.  相似文献

The biological control of water hyacinth is affected by water nitrogen and phosphorus content and this was investigated experimentally at five levels of nutrient supply by measuring plant photosynthetic and growth responses, and mirid reproduction and herbivory of nutrient treated plants. Low nitrogen (2–0.2 mg L−1) and phosphorus (0.2–0.01 mg L−1) supply decreased hyacinth photosynthesis, growth and biomass accumulation relative to plants supplied 200 mg L−1 N and 20 mg L−1 P. This effect depended more on nitrogen supply than phosphorus supply. Chlorophyll fluorescence showed that the photosynthetic light reactions of low nutrient plants were affected and leaves had decreased chlorophyll content, density of functional photosystems II and dissipated a greater proportion of absorbed energy as heat. Gas exchange parameters showed reduced carboxylation efficiency, rates of RuBP regeneration and light saturated photosynthetic rates, but not quantum yields. Effects on photosynthesis translated into lower plant dry biomass. Mirid herbivory exacerbated the effects of low nutrients noted for chlorophyll fluorescence, gas exchange parameters and biomass accumulation, however, these effects were not always significant and there was no obvious correlation between the level of nutrients supplied and the effect of mirid herbivory. Low nutrient supply did, however, affect mirid performance reducing the number of adult insects, nymphs and herbivory intensity suggesting that in the long-term mirid populations would be significantly affected by water nutrient status.  相似文献

The productivity of water hyacinth in the middle Paraná River was measured at three initial biomass levels by means of periodic harvesting of plants contained in 2 m2 floating baskets. The measuring period extended between August 1981 and July 1982. Initial biomass densities were 2, 5 and 10 kg (fw) m–2.Compared with inner island ponds, flowing waters connected to the main river showed better conditions for water hyacinth growth. The duration of the growth period was some 30% longer than in island ponds, probably due to the relatively warmer river waters. Productivity ranged between 108 and 164 g (fw) m –2 d–1 (annual average) for the lower and higher initial biomass values, respectively.  相似文献

S. mossambicus was exposed to toxic and sublethal concentrations of the fertilizer diammonium phosphate (0.2 to 1.0 g l–1). Mortality, food utilization and growth were studied. At a concentration of 0.6 g l–1 DAP, 100% mortality was observed within 96 h; no mortality occurred at 0.5 g l–1; LC50 was 0.55 g l–1. Rearing the fish in increasing sublethal concentrations of DAP, it was found that the feeding rate decreased from 25.4 mg g–1 fish–1 d–1 (fish reared in DAP-free water) to 10.1 mg g–1 d–1 at the highest sublethal concentration (0.5 g l–1). Growth rate was drastically reduced. At high sublethal concentrations of DAP, the fish lost reserve energy, in addition to the energy obtained from food intake for survival, as a result of increased swimming activity and opercular beats.  相似文献

Nutrient and faecal contamination is an increasing problem to the shrimp productivity and wildlife conservation at the internationally important wetland ecosystems of the Mai Po Marshes (Hong Kong, P.R. China). The present study examined the nutrient status and faecal bacteria loading and potential retention capacity of contaminants of two wetland enclosures. Water in the wetland enclosures was eutrophicated with high concentrations of dissolved inorganic nitrogen (inorg–Ndiss= 15.0 mg l–1) and orthophosphate phosphorus (o-P = 1.89 mg l–1) and was loaded with high levels of faecal coliforms (172 ×103 cfu in 100 ml) and faecal streptococci (1.94 ×103 cfu in 100 ml). The pattern of nutrient enrichment of two wetland enclosures is related to a north-to-south pollution gradient from the Shenzhen River to the wetlands. By retaining tidal water in the wetland for an 8-day period, water quality was greatly improved; NH4–N was removed by 83%, o-P by 45% and faecal bacteria by 100%. This implies a self-purification capability of the wetland enclosures and a potentiality of using them as an alternative sewage treatment.  相似文献

Individuals of the fairy shrimp, Branchinecta longiantenna, were subjected to 5 concentrations (0.1 to 15 mg l–1) of Pb in diluted habitat water at 13 °C. Lead concentrations (mg kg–1 wet weight) in the animals were determined at 2-day intervals by digestion in nitric acid followed by atomic absorption analysis. The shrimp were also subjected to 0.1 mg l–1 media of Cd and Zn, separately.Uptake rates by the fairy shrimp for the three metal ions at 0.1 mg l–1 were: 0.111, 0.0885, and 0.0460 mg kg–1 day–1 for Zn, Pb, and Cd, respectively. After 2 days in 1.0 mg l–1 Cd or Zn, the animals expired; but they surviced for 8 days in a 10 mg l–1 Pb medium and for 2 days in 25 mg l–1 Pb. Lead uptake demonstrated a linear dependence on the Pb concentration in the media.Shrimp survived at much higher tissue accumulations of Pb compared to Zn and Cd. Estimated lethal doses were 20, 1.2–2.4, and 0.4–1.4 mg kg–1 wet weight for Pb, Zn, and Cd, respectively. Pb was found to be at much lower concentration than Cd or Zn in the natural pond water but between Cd and Zn levels in the sediment. The production of extracellular enzymes by the thermophilic fungus Thermomyces lanuginosus was studied in chemostat cultures at a dilution rate of 0.08 h–1 in relation to variation in the ammonium concentration in the feed medium. Under steady state conditions, three growth regimes were recognised and the production of several extracellular enzymes from T. lanuginosus was recorded under different nutrient limitations ranging from nitrogen limitation to carbon/energy limitation. The range and the production of carbohydrate hydrolysing enzymes and lipase increased from Regime I (NH4Cl 600 mg l–1) to Regime III (NH4CI 1200 mg l–1), whereas production of protease was highest in Regime II (600 mg l–1 < NH4Cl <1200 mg l–1).  相似文献

Evidence for the influence of food type and heavy metals on shell growth and fertility is presented for a freshwater population of the snail P. jenkinsi. When fed an excess of lettuce or lamb heart (protein source), growth rates were higher for lettuce. Highest growth rates occurred at a diet of lettuce plus lamb heart. Fertility was favoured by a diet of lamb heart. When fed an excess of lettuce, the EC50 growth values were 16 µg Cd l–1, 13 µg Cu l–1, and 103 µg Zn l–1 in lake water; snail fertility was inhibited at 25 µg Cd l–1 and 30 µg Cu l–1. A diet of lake detritus spiked with Cd or Cu resulted in a decrease of approximately 50% in growth rates, when compared with growth on non-spiked detritus. Spiked detritus lost metals into lake water. Food type positively interacted with metal stress, both for growth rate and fertility. The assessment of inhibitory effects of detritus contaminated either in the field or, notably, by spiking, and serving as food source for deposit feeders is hampered by sampling problems in the field and by redistribution processes of pollutants between particles and water in laboratory-scale experiments.  相似文献

Addition of 5 mg abscisic acid l–1 after 12 days' growth of Taxus chinensis suspension culture gave the greatest paclitaxel accumulation at 11 mg l–1, which was almost 5 times that of the control culture. The highest paclitaxel production, 18 mg l–1, was obtained using 5 mg abscisic acid l–1 and 20 mg methyl jasmonate l–1.  相似文献

The influence of maltose and growth regulators on microspore culture response was investigated in japonica rice. High frequency of callus induction of isolated microspores was obtained with liquid medium containing MS salts, 100 mg l–1 myo-inositol, 1 mg l–1 thiamine-HCl, 500 mg l–1 glutamine, 60 g l–1 maltose, and several growth regulators. The effect of maltose on promoting callus formation was associated with keeping a high proportion of swollen microspores after 5 day preculture and increasing the microspore division rate on the 3rd day after culture initiation. No significant effect of maltose in place of sucrose on plantlet regeneration was seen in regeneration medium. Among the growth regulators tested, the combination of auxin 2,4-dichlorophenoxyacetic acid (1 mg l–1), naphthaleneacetic acid (1 mg l–1), and cytokinin (6-benzyl-aminopurine 1 mg l–1) in the medium proved to be much better for callus formation than in the other media, and the percentage of callusing microspores of that medium reached 0.86%. Indole-3-acetic acid (0.5 mg l–1) and kinetin (2 mg l–1) in regeneration medium were beneficial for green plantlet differentiation. The results also showed that the frequencies of microspores initial division, callus formation and green plant regeneration varied among genotypes no matter what kind of growth regulator and sugar were used. Xiushui 117 was the best variety for callusing followed by 02428 & Taipei 309. To investigate the production potential of eicosapentaenoic acid (EPA) by the diatom Nitzschia laevis, the growth characteristics and fatty acid composition of the cells were studied under photoautotrophic, mixotrophic and heterotrophic conditions of growth. The specific growth rate and maximum biomass concentration were respectively 0.466 d–1 and 2.27 g l–1 for mixotrophic culture, 0.344 d–1 and 2.04 g l–1 for heterotrophic culture, and 0.167 d–1 and 0.5 g l–1 for photoautotrophic culture, respectively. As for EPA production, the yield and productivity were respectively 52.32 mg l–1 and 10.46 mg l–1 d for mixotrophic culture, 35.08 mg l–1 and 6.37 mg l–1 d for heterotrophic culture, and 6.78 mg l–1 and 3.39 mg l–1 d for photoautotrophic culture, respectively. Results suggest that mixotrophic culture is the most suitable growth mode for the production of EPA by the diatom Nitzschia laevis. The results are useful for the development of a cost-effective fermentation process for EPA production by Nitzschia laevis.  相似文献

South Africa has some of the most eutrophic aquatic systems in the world, as a result of the adoption of an unnecessarily high 1 mg l−1 phosphorus (P) standard for all water treatment works in the 1970 s. The floating aquatic macrophyte, water hyacinth (Eichhornia crassipes (Mart.) Solms (Pontederiaceae)), has taken advantage of these nutrient rich systems, becoming highly invasive and damaging. Despite the implementation of a biological control programme in South Africa, water hyacinth remains the worst aquatic weed. A meta-analysis of published and unpublished laboratory studies that investigated the combined effect of P and nitrogen (N) water nutrient concentration and control agent herbivory showed that water nutrient status was more important than herbivory in water hyacinth growth. Analysis of long-term field data collected monthly from 14 sites around South Africa between 2004 and 2005 supported these findings. Therefore the first step in any water hyacinth control programme should be to reduce the nutrient status of the water body.  相似文献

Loblolly pine (Pinus taeda L.) somatic embryogenesis initiation was improved by supplementing the initiation medium with the pH buffer agent 2(n-morpholino)ethanesulphonic acid (MES) at 250 mg l–1, folic acid at 0.5 mg l1, and biotin at 0.05 mg l–1. MES and vitamins increased the percentage of explants with extruded tissue that continued the initiation process to form embryogenic tissue. The increase in initiation was about 12%. Initiation of 12 open-pollinated families averaged 38.5%, which is 16% higher than initiation on medium without these additives. When tested with 18 control-pollinated families, initiation averaged 26.3%. Basal medium contained a combination of modified 1/2 P6 salts, activated carbon (AC) at 50 mg –1, Cu and Zn adjusted to compensate for adsorption by AC, 1.5% maltose, 2% myo-inositol, 500 mg l–1 casamino acids, 450 mg l–1 glutamine, 2 mg l–1 NAA, 0.63 mg l–1 BAP, 0.61 mg l–1 kinetin, 3.4 mg l–1 silver nitrate, 10 M 8-Br-cGMP, 0.1 M brassinolide, and 2 g l–1 Gelrite. Early-stage embryo growth and initiation in Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) were also improved in the presence of these additives.  相似文献

Callus of Orthosiphon stamineus could be induced successfully from petiole, leaf and stem tissues but not roots when cultured on MS medium containing different concentration of NAA (0–4.0 mg l–1) and 2,4-D (0–2.0 mg l–1). Highest fresh weight callus production was obtained from leaf explants and those with best friability were obtained on MS medium plus 1.0 mg l–1 2,4-D plus 1.0 mg l–1 NAA. Cell suspension cultures were established from these cultures. The appropriate cell inoculum size for the best cell growth was 0.75 g of cells in 20 ml culture medium. Cell suspension culture using MS medium supplemented with 1.0 mg l–1 2,4-D promoted the best cell growth with maximum biomass of 8.609 g fresh weight and 0.309 g dry weight 24 days after inoculation. Cells that grew in MS medium supplemented with 1.0 mg l–1 2,4-D reached the stationary growth phase in 15 days as compared to the cells that grew in MS medium supplemented with 1.0 mg l–1 2,4-D + 1.0 mg l–1 NAA reached the stationary phase in 24 days. MS medium supplemented with 1.0 mg l–1 2,4-D was considered as the maintenance medium for maintaining the optimum cell growth of O. stamineus in the cell suspension cultures with 2-week interval subculture.  相似文献

Bacterioplankton abundance, biomass and production were studied at a central station (35 m depth) from April 1987 to September 1988 in a mesotrophic reservoir. Bacterial production was calculated by the (3H) thymidine method.For the water column, integrated estimates of bacterioplankton abundance ranged from 2.3 109 to 4.6 109 cells l–1, and carbon biomass from 0.037 to 0.068 mg C l–1; the thymidine incorporation rates ranged from 0.8 to 17.2 picomoles l–1 h–1, leading to net bacterial production estimates of less than 0.7 µg C l–1 d–1 in winter to 18 µg C l–1 d–1 in summer. About 55% of the production occurred in the euphotic layers.Over the year, the bacterial carbon requirement represented 90% of the autotrophic production for the whole lake. It was five times lower than autotrophic production in spring, but twice as high in summer. This important temporal lack of balance suggests that not all the spring primary production products are consumed immediately and/or that other carbon sources probably support bacterial growth in summer.  相似文献

Water reuse is a proposed strategy for utilizing or disposing of poor quality drainage water produced in the westside of central California. This 2-year field study evaluated the ability of two potential forage species to tolerate irrigation with water high in salinity, boron (B), and selenium (Se). The species used were: Sporobulus airoides var. salado (alkali sacaton) and Medicago sativa var. salado (alfalfa). After first year establishment with good quality water (<1 dS m–1), the two species were furrow-irrigated with drainage effluent that had an average composition of sulfate-dominated salinity ((electrical conductivity (EC) of 6.2 dS m–1)) B (5 mg l–1), and Se (0.245 mg l–1). Both crops were clipped monthly from June to October of each year. Total dry matter yields averaged between 11 and 12 mg ha–1 for both crops irrigated with effluent for two growing seasons. Plant concentrations of Se ranged from a low of 1.3 mg kg–1 in alkali sacaton to a high of 2.5 mg kg–1 in alfalfa, while B concentrations ranged from a low of 60 mg kg–1 in alkali sacaton to a high of 170 mg kg–1 in alfalfa. Chemical composition of the soil changed as follows from preplant to post-irrigation after two seasons with drainage effluent: EC from 2.78 to 6.5 dS m–1, extractable B from 1.9 to 5.6 mg l–1, and no change in extractable Se at 0.012 mg l–1 between 0 and 45 cm. Between 45 and 90 cm, EC values increased from 4.95 to 6.79 dS m–1, extractable B from 2.5 to 4.8 mg l–1, and no change in extractable Se at 0.016 mg l–1. Increased salinity and extractable B levels in the soil indicate that management of soil salinity and B will be necessary over time to sustain long term reuse with poor quality water.  相似文献

Summary The morphogenetic potential of the shoot tip explants ofEnsete superbum (Roxb.) Cheesman, a wild relative of the cultivated bananas, was investigated and an effective clonal propagation method devised. Shoot tip explants grown in modified MS medium containing 1.5 mg l–1 BAP and 1 mg l–1 KIN developed corms which on transfer to medium containing 3 mg l–1 IBA and 1.5 mg l–1 BAP, regenerated a large number of shoots from the surface of the corm, the origin of which was traced to single hypodermal cells. Shoots were rooted on a half-strength MS medium salts containing 3 mg l–1 IBA and 0.1 mg l–1 BAP. The rooted plantlets were hardened and planted in the field where the plants looked normal.  相似文献

Organic material transport in the New River, Virginia, was investigated over a 12 month period. Collections were made using drift nets and grab water samples from bridges at two sites about 210 km apart. About midway between the two sampling sites is a 1920 ha impoundment used for flood control and power generation. Dissolved organic matter (DOM) ranged 1–50 mg l–1 at Site 1, upstream from the impoundment, and 11–19 mg l–1 at Site 2 and was the most abundant form of organic matter at both sites during most periods of the year. Fine particulate organic matter (FPOM) ranged 1–45 mg l–1 at Site 1 and 1–9 mg l–1 at Site 2. Concentration of coarse particulate organic matter (CPOM) ranged 0.1–0.7 mg l–1 at Site 1 and 0.1–0.2 mg l–1 at Site 2. On an annual basis, the organic matter loads at Site 1 and Site 2 were computed to be 67 000 and 76 800 T y–1, respectively, suggesting that the impoundment trapped and processed POM, and that municipal and industrial treatment facilities between the study sites supplemented DOM in the river.  相似文献

High percentages of micro-calli and micro-derived embryos were produced from isolated asparagus microspores at late uninucleate stage on MS liquid medium supplemented with 1.0 mg l–1 2,4-D and 0.5 mg l–1 BA. Two types of calli, namely compact callus (CC) and loose callus (LC), were found. Plantlets were regenerated via organogenesis, when these calli were transferred onto MS solid medium supplemented with 1.0 mg l–1 BA and 0.2 mg l–1 IBA 6 weeks. Embryos were produced from liquid cultured microspores, or from solid cultured micro-calli. The frequencies of haploid plant production from organogenesis and embryogenesis were compared. Effects of plant growth regulators on callus production, plantlet regeneration, and haploid plant production were tested. The effect of an enhanced nutrient supply to coastal waters of a landlocked bay, Hopavågen in Central Norway, on the phytoplankton production and biomass, and on growth of scallops (Pecten maximus) was studied in 1997–1999. Nitrogen, silicon and phosphorous (N:Si:P = 16:8:1, atomic) were added daily between May and October in 1998 at a level of 0.4 mg P m–3 day–1. The concentration of nutrient addition was doubled in 1999 during the same period. High addition of nutrients (1999) resulted in a significantly higher phytoplankton biomass in the summer period, expressed as chlorophyll a content, than without nutrient (1997) and low nutrient (1998). The respective mean chlorophyll a levels were 2.4 in 1999, 1.6 in 1998 and 1.2 g l–1 in 1997. The mean primary production during the summers generally increased with the addition of nutrients from an average level of 320 mg carbon m–2 day–1 in 1997 to 1200 mg carbon m–2 day–1 in 1999. Scallops placed at 10 m depth in Hopavågen showed an increase in growth rate of the outer scallop shell in the period July–September from 0.16% day–1 in 1997 to 0.53% day–1 in 1998. Scallops grown in an unfertilised control station in the fjord outside Hopavågen had a significantly lower growth rate than those grown in the fertilised water of Hopavågen. The results showed decreased growth rate with increasing shell sizes. However, for all size groups studied a higher growth rate of the scallops was observed when nutrients were added to the bay. The tissue dry weight content of scallops grown in Hopavågen was 2–4 times higher than in the control scallops.  相似文献
