ABSTRACT

Tree nuts comprise a category of food allergens that must be included in the food labels in several countries. We developed a polymerase chain reaction (PCR) method using eight specific primer pairs to detect eight representative tree nuts (almond, Brazil nut, cashew, hazelnut, macadamia nut, pecan, pistachio, and walnut) under the same experimental conditions. The specificity of the eight primer pairs was confirmed by PCR testing against a variety of plant and animal samples. The detection limit of the method ranged from 1 fg to 1 pg DNA of individual tree nuts. The method detected tree nut DNA in processed and unprocessed food. In addition, the primer pairs could be combined into two sets of tetraplex PCR system. The developed method is specific, sensitive, and efficient, making it useful for detecting trace amounts of eight species of tree nut in foods.  相似文献

Fungal flora of 20 samples of lentil seeds collected from Assiut governorate, Egypt, were studied. Seventeen genera and 13 species were isolated on glucose- (15 genera and 27 species), cellulose-(15 genera and 25 species)Czapek's agar media at 28 °C. The most common species were as follows: on glucose-Czapek's agar, Aspergillus fumigatus, A. niger, A. flavus, A. terreus, Penicillium notatum and Rhizopus oryzae and on cellulose agar, A. fumigatus, A. niger, A. flavus and P. notatum. Thin layer chromatographic (TLC) analysis and a biological test (Artemia salina) indicated the presence and the toxicity of aflatoxin in the extract of one sample (aflatoxins B1, B2, G1 and G2, at 20 mg/kg, total). This is the first report of the natural occurrence of aflatoxins in lentils.  相似文献

Thirty-nine species and 16 fungal genera were isolated from Egyptian cotton seeds, cotton seed meal and cotton seed cake on 1% glucose-Czapek's agar medium incubated at 28 °C. Aspergillus was the most frequent genus and it emerged in 87–100% of the samples contributing 70–98% of total fungi in the three substrates tested. The most common species were A. niger, A. flavus, A. fumigatus, A. terreus and Rhizopus stolonifer; A. niger, A. fumigatus and Penicillium corylophilum; and A. niger, A. flavus, A. terreus, A. nidulans and Rhizopus stolonifer, respectively. Cotton seeds and cotton seed products were naturally contaminated by aflatoxin B1 and B2. About 16% of the different substrates tested were positive for aflatoxin contamination. No citrinin, ochratoxin A, patulin, sterigmatocystin, diacetoxyscirpenol, T-2 toxin or zearalenone were detected in the samples assayed.  相似文献

A survey was carried out to obtain data on the occurrence of mycotoxins and the mycotoxin-producing potential of fungi isolated from nuts (almonds, peanuts, hazelnuts, pistachio nuts) and sunflower seeds in Spain. Thin-layer chromatography was used to separate the toxins. Aflatoxins were detected in one sample of almonds (95 ppb aflatoxin B1 and 15 ppb aflaxtoxin B2) and in one sample of peanuts at a level below 10 ppb of aflatoxin B1. 100% of samples showed variable incidence of fungal contamination. The predominant fungi present in samples were Penicillium spp, Aspergillus niger, A. flavus, A. glaucus and Rhizopus spp. The results showed that isolates of different species were able to produce aflatoxins B1, B2, G1, and G2, sterigmatocystin, ochratoxin A, patulin, citrinin, penicillic acid, zearalenone, and griseofulvin.  相似文献

The distribution in Turkey of filbert, pistachio, chestnut, almond and walnut is discussed in relation to their method of propagation and climatic parameters. The effect of temperature and rainfall severely limits the present production areas of filbert and pistachio, both of which are vegetatively propagated. In contrast, almonds and walnuts are mainly propagated from seed and are more widely distributed throughout the country. Their distribution is discussed in relation to the genetic diversity and the adaptability of local tree populations to varied climatic conditions. Data are presented on the range of variability of nut and kernel weight of almonds and walnuts.  相似文献

Pistachio nut samples taken during various stages of development from orchards in Iran, showed that contamination with fungi occurred mainly during the later stages of nut development. Members of the genera Aspergillus and Penicillium occurred most frequently. Of the Aspergilli, the species A. niger, A. flavus and A. fischeri var. spinosus occurred most frequently, followed by A. terreus, A. tamarii and A. nidulans. Twenty-two isolates comprising 13 species were tested for toxicity to ducklings. Isolates of known toxic fungi included A. flavus, A. niger, A. parasiticus, A. ochraceus, A. versicolor, A. nidulans and A. terreus. The toxicity of A. fischeri var. spinosus is reported. Chemical analysis showed that all isolates of A. flavus and A. parasiticus produced aflatoxin B1, the isolates of A. versicolor and A. nidulans produced sterigmatocystin while the toxic isolate of A. ochraceus did not produce ochratoxins.Toxic fungi have been shown to occur in a variety of nuts (4), (5), (11), (12), (13), (15), (18), (20), (21). Aflatoxin contamination of pistachio nuts has been reported and has in the past led to the rejection of consignments of Iranian pistachio nuts (1). In Iran, pistachio nuts are produced mainly in the south eastern provinces (Kerman & Zahedan) and to a limited extent in the Northern part (Kazvin & Damghan). In 1975 it was estimated (7) that there were some 24 million pistachio trees in Iran, of which 60% were situated in Rafsanjan, Kerman (Table 1). Economic considerations as well as the potential health hazard posed by aflatoxin-contaminated nuts, prompted the University of Isfahan to initiate a study of various aspects of the mycotoxin problem in pistachio nuts.  相似文献

A shipment of South African corn (1989) exported to Taiwan, was analyzed for various ear-rot fungi andFusarium mycotoxins. Two sets of samples, one from the points of origin in South Africa prior to shipment, and the other from the end-point distributors in Taiwan, were studied. Surface-sterilized kernels were plated onto two different agar media and the fungal colonies identified. High Performance Liquid Chromatography was used to analyze mycotoxin levels. The predominant ear-rot fungi, in decreasing order of isolation frequency, wereFusarium subglutinans, F. moniliforme, Diplodia maydis andF. graminearum. Aspergillus flavus andA. parasiticus were not isolated from samples prior to export, but a small number ofA. flavus isolates were found after shipment. The predominant mycotoxins were fumonisins B1 (0–865 ng/g) and B2 (0–250 ng/g). Low levels of moniliformin (390 ng/g) were detected in some samples before shipment. Zearalenone (25 ng/g), and nivalenol (120 ng/g) were detected in two out of 32 samples taken in Taiwan. The enhancing effect of various concentrations of 18 oils and a silicon antifoam agent on erythromycin production by Saccharopolyspora erythraea was evaluated in a complex medium containing soybean flour and dextrin as the main substrates. The oils used consisted of sunflower, pistachio, cottonseed, melon seed, water melon seed, lard, corn, olive, soybean, hazelnut, rapeseed, sesame, shark, safflower, coconut, walnut, black cherry kernel and grape seed oils. The biomass, erythromycin, dextrin and oil concentrations and the pH value were measured. Also, the kinds and frequencies of fatty acids in the oils were determined. The productivity of erythromycin in the oil-containing media was higher than that of the control medium. However, oil was not suitable as a main carbon source for erythromycin production by S. erythraea. The highest titer of erythromycin was produced in medium containing 55 g/l black cherry kernel oil (4.5 g/l). The titers of erythromycin in the other media were also recorded, with this result: black cherry kernel > water melon seed > melon seed > walnut > rapeseed > soybean > (corn = sesame) > (olive = pistachio = lard = sunflower) > (hazelnut = cotton seed) > grape seed > (shark = safflower = coconut). In media containing various oils, the hyphae of S. erythraea were longer and remained in a vegetative form after 8 days, while in the control medium, spores were formed and hyphae were lysed.  相似文献

Wheat from two cultivars with contrasting characteristics were harvested in ten experimental plots located in wheat producing areas of the State of São Paulo, Brazil. The samples (10 of each cultivar) were analyzed by a gaschromatographic method for deoxynivalenol (DON), nivalenol (NIV), diacetoxyscirpenol (DAS), toxins T-2 (T-2) and HT-2, T-2 tetraol, T-2 triol, and by a thin-layer chromatographic method for zearalenone (ZEN), aflatoxins B1, B2, G1, G2, ochratoxin A and sterigmatocystin. No mycotoxins were detected in 13 samples. DON was found in four samples (0.47–0.59 µg/g), NIV in three samples (0.16–0.40 µg/g), T-2 in two samples (0.40, 0.80 µg/g), DAS in one sample (0.60 µg/g), and ZEN in three samples (0.04–0.21 µg/g). The wheat samples were also examined for the incidence of fungi.Alternaria, Drechslera, Epicoccum andCladosporium were the prevailing genera. Among theFusarium spp.,F. semitectum was present in 19 samples andF. moniliforme in 18 samples. The aim of the study presented here was to determine if there is a correlation between the presence of specific protein domains within tree nut allergens or tree nut allergen epitopes and the frequency of bioactive fragments and the predicted susceptibility to enzymatic digestion in allergenic proteins from tree nuts of cashew (Anacardium occidentale), pecan (Carya illinoinensis), English walnut (Juglans regia) and pistachio (Pistacia vera) plants. These bioactive peptides are distributed along the length of the protein and are not enriched in IgE epitope sequences. Classification of proteins as bioactive peptide precursors based on the presence of specific protein domains may be a promising approach. Proteins possessing a vicilin, N-terminal family domain, or napin domain contain a relatively low occurrence of bioactive fragments. In contrast, proteins possessing the cupin 1 domain without the vicilin N-terminal family domain contain a relatively high total frequency of bioactive fragments and predicted release of bioactive fragments by the joint action of pepsin, trypsin, and chymotrypsin. This approach could be utilized in food science to simplify the selection of protein domains enriched for bioactive peptides.  相似文献

Thirteen seed samples of lentil (Lens esculenta) were incubated on agar plate and moist filter papers (Moist Chambers) at 28 ± 2 °C for determination of the incidence of seed-borne fungi. Aflatoxins content of the seeds was measured using the bright greenish-yellow fluorescence test (BGYF) and thin-layer chromatography (TLC). Sixty-nine species and seven varieties, which belong to 24 genera of fungi, were isolated from this crop. Of these fungi, 51 species and two varieties are considered new for this crop, whereas seven genera and 13 species are new to the mycoflora of the Sudan. The genus Aspergillus (13 species and 6 varieties) which comprising 44% of the total colony count was the most prevalent genus followed by Rhizopus (2 species, 19%), Penicillium (6 species) and Fusarium (8 species) (12%), Chaetomium (3 species) and Cladosporium (5 species) (6%), where the 18 genera (1–4 species) showed very low level of incidence (19%). Of the possible pathogens of lentil plants, F. oxysporum the main cause of vascular wilt was recovered from seeds of this crop. Thin layer chromatographic analysis of chloroform extracts of 13 seed samples showed that only one samples was naturally contaminated with aflatoxins B1, B2, G1 and G2 (14.3 μg/kg). 该研究以目前主栽的平榛、毛榛及25个平欧杂种榛品种(系)的坚果为研究对象,测定了不同品种(系)榛子坚果的17个表型性状指标,用方差分析、相关性分析、聚类分析和主成分分析方法对其进行差异性分析和综合评价,为国内榛树品种(系)的选优、开发及加工利用提供科学依据。结果表明:(1)不同品种(系)榛子坚果的17个表型性状指标存在显著差异,其中果壳质量、坚果体积和坚果质量差异最为明显,变异系数分别为31.49%、30.44%、27.50%,横纵径比、果形指数、伸长率、圆球度的变异系数均小于10%。(2)平欧杂种榛坚果的纵径、横径、侧径及坚果质量、果仁质量、出仁率等指标显著优于毛榛和平榛,其中杂种榛的平均果仁质量(0.70~1.46 g)和出仁率(31.48%~56.12%)显著高于平榛(0.51 g、35.13%)和毛榛(0.37 g、38.12%)。(3)榛子坚果表型性状指标间的相关性分析表明,榛子坚果体积越大,其质量和果仁质量也越大,果壳越薄则出仁率更高。(4)主成分分析结果发现,影响榛子坚果综合得分的主要指标为坚果的纵径、横径、侧径、质量和果仁质量,其次是果形指数、横纵径比、出仁率、果仁饱满...  相似文献

摘要 目的:探讨5岁以下哮喘儿童与血清特异性过敏原(specific IgE,sIgE)的分布情况。方法:本研究采用免疫印迹法对 2019 年1月至 2019 年12月在西安交通大学第二附属医院住院的5岁以下62例哮喘患儿和49例喘息患儿的行血清特异性过敏原检测,对比分析5岁以下哮喘和喘息儿童过敏原分布情况及与哮喘的发病关系。结果:户尘螨、猫毛皮屑、狗毛皮屑、蒿草、葎草、桤杨柳山毛榉橡胡桃、烟曲霉、念珠菌点青霉分枝孢霉交链孢霉黑曲霉吸入过敏原和花生黄豆、腰果开心果榛子杏仁核桃、虾蟹、桃苹果芒果荔枝草莓食物过敏原这12类过敏原在哮喘组与喘息组有显著差异(P<0.05),与哮喘发病有关。多因素logistic回归分析结果显示户尘螨、猫毛皮屑、坚果类、霉菌、水果类是哮喘发病的危险因素(P<0.05)。户尘螨、猫毛皮屑和虾蟹是男性哮喘患儿发病的危险因素,念珠菌点青霉分枝孢霉交链孢霉黑曲霉是女性哮喘患儿发病的危险因素(P<0.05)。结论:血清特异性(sIgE)过敏原在哮喘与喘息患儿中分布不同,同时发现过敏原在哮喘患儿中存在性别差异,故对哮喘患儿进行过敏性检测可以作为回避过敏原的依据。  相似文献

Brewing and consumption of traditional beer have social–economic significance in most African countries including Botswana. Traditional sorghum malt, wort, and beer samples were collected from three villages around Gaborone, Botswana. Forty-six malt samples were analyzed for fungi on three different media and developing colonies were subcultured for identification. Rhizopus, Fusarium, Mucor, and Aspergillus were the most common genera isolated. Out of the 46 malt samples, 72% contained Rhizopus stolonifer, 63% Fusarium verticillioides (syn. Fusarium moniliforme), and 37% Aspergillus flavus. Although Aspergillus flavus was isolated from malt samples, aflatoxins (B1, B2, G1, and G2) were not detected in any of the samples analyzed. When the malt, wort, and beer samples were analyzed for fumonisin Bl and zearalenone, fumonisin B1 was detected in 3 malt samples, with concentrations ranging from 47 to 1316 μg/kg, while zearalenone was detected in 56%, 48% and 48% of the malt, wort and beer samples, respectively. Zearalenone concentration in samples ranged from 102 to 2213 μg/kg in malt, 26 to 285 μg/l in wort and 20 to 201 μg/l, in beer. Zearalenone carry-over from wort to beer ranged from 23 to 403%. Therefore, although aflatoxins and fumonisin B1 do not appear to be major contaminants, zearalenone is common and could pose a potential problem in traditional beer in Botswana.  相似文献

The nut weevil Curculio nucum (Coleoptera: Curculionidae) is responsible for high yield losses in several hazelnut‐growing areas of Europe and Turkey, if not regularly controlled using pesticides. The resistance of hazelnut varieties to the nut weevil is known but poorly investigated so far. Thus a 2‐year study was carried out to investigate factors affecting C. nucum attacks on two cultivars [Tonda Gentile delle Langhe (TGL), Ennis] and four selections (Daria, 101, B6, L35) in a 15‐year‐old orchard of northwestern Italy. Nut developmental stages and shell characteristics, such as hardness and thickness, were measured and correlated to the biological cycle of the nut weevil and to the damage by its larvae. During the 2‐year sampling, nut weevil adults were collected on all cultivars and selections, but in greater quantities on TGL and Daria, and in lower quantities on 101 and B6. In 2005, by female dissection, mature eggs were observed from late June, when the mean temperature exceeded 18°C. Differences between cultivars and selections were found in nut and kernel development, and in shell hardening; in particular, the kernel development was directly related with the shell hardness. At harvest, the damage varied on average from 2.57% in cultivar TGL to 51.84% in selection 101 of total nuts harvested, so independently of the numbers of the collected weevil adults. These two varieties were the earliest and the latest in kernel development and shell hardening, respectively. Therefore, the susceptibility to nut weevil attacks was strictly related to shell hardening; in fact, a rapid hardening of the shell can hamper the oviposition of C. nucum females. By contrast, no correlation was found between shell thickness and damage at harvest.  相似文献

Fungi of 19 genera, 30 species, and one variety were isolated from 25 samples of sheep-, cattle- and camel feedstuffs collected from different farms in the Beida Governorate, Libya.Aspergillus, Penicillium andFusarium were the most common genera in the three substrates tested. TLC was used to establish the identity of aflatoxins in the chloroform extract of all samples and the ability to produce aflatoxins byAspergillus flavus in a synthetic liquid medium. Twenty samples out of 25 tested were naturally contaminated and 21 isolates ofA. flavus out of 30 produced at least one of the following aflatoxins: B1; B1, G1; and B1, B2, G1, G2. This is the first report about the natural occurrence of aflatoxins and aflatoxin-producers of the genusAspergillus in Libya.  相似文献

A wide range of field and storage fungi were isolated from black pepper, white pepper and Brazil nut kernels from Amazonia. A total of 42 species were isolated from both peppers. Aspergillus flavus and A. niger were isolated more frequently from black than from white pepper. Other potential mycotoxigenic species isolated included: A. ochraceus, A. tamarii, A. versicolor, Emericella nidulans and Chaetomium globosum, Penicillium brevicompactum, P. citrinum, P. islandicum and P. glabrum. Species isolated from pepper for the first time were Acrogenospora sphaerocephala, Cylindrocarpon lichenicola, Lacellinopsis sacchari, Microascus cinereus, Petriella setifera and Sporormiella minima. Seventeen species were isolated from Brazil nut kernels. A. flavus was the dominant species followed by A. niger. P. citrinum and P. glabrum were the only penicillia isolated. Species isolated for the first time included Acremonium curvulum, Cunninghamella elegans, Exophiala sp., Fusarium oxysporum, Pseudoallescheria boydii, Rhizopusoryzae, Scopulariopsis sp., Thielavia terricola and Trichoderma citrinoviride. Considerably more metabolites were detected from black than white pepper in qualitative analyses. Chaetocin, penitrem A, and xanthocillin were identified only from black pepper, and tenuazonic acid was identified from both black and white pepper. Aflatoxin G2, chaetoglobosin C, and spinulosin were identified from poor quality brazil nuts. Aflatoxin B1 and B2 were also only detected in poor quality brazil nuts at concentrations of 27.1 g kg–1 and 2.1 g kg–1 respectively (total 29.2 g kg–1).This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Lipid-rich and energy-dense diets can have significant effects on the reproductive physiology, including the ovarian function and fertility. The aim of this study was to assess the effect of cashew nut bran supplementation as a lipid source on follicle development, plasma and intrafollicular concentrations of cholesterol, and developmental competence of in vitro–matured goat oocytes. The inclusion of cashew nut bran as 24% of the goats' diet for 28 days increased the percentage and number of degenerated oocytes compared with the control (P < 0.05), and also the plasma cholesterol levels and the proportion of grade IV oocytes compared with all other treatments (P < 0.05). Moreover, a significant reduction was observed in the proportion of viable oocytes compared with the control and in the percentage of grade II oocytes compared with all other treatments (P < 0.05). Oocyte maturation, cleavage, and blastocyst rates after parthenogenetic activation of viable oocytes were not affected by the type of diet. In conclusion, the inclusion of cashew nut bran as 24% of the diet of adult goats for 28 days changed plasma cholesterol levels and reduced the proportion of viable immature oocytes; however, the 12% and 24% diet supplementations with cashew nut bran did not interfere with competence of resulting viable oocytes to reach the metaphase II stage after IVM, and to develop after parthenogenetic activation.  相似文献

该研究以贵州省内的3个山脉(乌蒙山、苗岭山和武陵山)、19个核桃和泡核桃实生居群、245棵实生单株为研究材料,对坚果表型性状29个指标的多样性、相关性、聚类及变异进行分析,以揭示贵州核桃资源的表型遗传多样性水平,为贵州核桃资源的保存与利用及核心种质构建提供依据。结果显示:(1)核桃和泡核桃实生居群245份种质资源的18个表型质量性状Simpson指数为0.26~0.82,Shannon-Wiener指数为0.12~1.79;核桃小叶形状多样,易发生顶叶退化,坚果核壳表面特征、坚果形状及核仁皮色多样性丰富。(2)坚果表型变异系数为3.32%~47.67%,平均为21.28%且差异显著(P<0.05),居群间变异系数为9.42%~31.61%且差异显著(P<0.05);居群内和居群间变异均是核桃表型多样性的来源,但坚果性状在居群间差异显著性均高于居群内,说明居群间变异是该区域坚果表型变异的主要来源。(3)3个山脉区域核桃表型均有不同程度变异,其中,苗岭山核桃表型变异程度低,乌蒙山核桃表型变异程度高;UPGMA聚类结果显示,19个核桃实生居群依据贵州山脉区域划分为三类,表明贵州核...  相似文献
