When serum-starved A431 cells were treated with 200 nM phorbol ester TPA for 15 min, the cellular activity of protein kinase FA/glycogen synthase kinase-3α (kinase FA/GSK-3α) could be decreased to ~25% of control. Conversely, when treated with 1 μM TPA for 24 hr, the activity could be reversibly increased to ~200% of Control. The naturally occurring protein kinase C (PKC) inhibitor sphingosine at a concentration of 27 μM could also induce activation of kinase FA/GSK-3α to ~200% of control within 60 min. Further, when cells were chronically treated with 1 μM TPA for 24 hr and then with 27 μM sphingosine for 60 min, the activity of kinase FA/GSK-3α could only be increased to ~200% of control. Furthermore, when cells were pretreated with sphingosine and then acutely treated with TPA, the acute TPA effect on kinase FA/GSK-3α activity could be abolished by genistein or tyrosine phosphorylation, which could be blocked by genistein or tyrosine phosphatase, but could be reversed by orthovanadate. Taken together, the results demonstrate that TPA/sphingosine induce tyrosine phosphorylation and concurrent activation of kinase FA/GSK-3α in a common signalling pathway. Since TPA and sphingosine are potent PKC modulators, the results further suggest a potential role of PKC in modulating tyrosine phosphorylation/activation of kinase FA/GSK-3α. Kinetic studies on seven subtypes of PKC further demonstrate a specific involvement of PKC∈ in this tyrosine phosphorylation/activation process. This provides a new mode of signal transduction between these two important serine/threonine kinases in cells.  相似文献

从外来水生植物大聚藻的耐寒性、再生能力和耐盐性等方面入手,研究比较了其与有害植物水花生对环境适应能力的差异。结果表明:大聚藻的越冬能力明显高于水花生;大聚藻断枝离体水培后复活快、发根与生长迅速,生长量与营养繁殖力也明显大于水花生;大聚藻具有很强的耐盐能力,其枝条在NaCl 盐浓度为0 g·L-1~3 g·L-1 的水体中能正常生长,4 g·L-1~5 g·L-1的浓度中可存活2~4 周。因此,大聚藻在自然生态环境中具有超强的生存能力和繁殖能力,存在一定的生态风险,对大聚藻的利用必须在有限范围内谨慎进行,应严格控制其群体在自然生态环境中的过量扩散与繁殖。  相似文献

A model of the use of the platinum group metals (PGMs) platinum, palladium, and rhodium in Europe has been developed and combined with a model of the environmental pressures related to PGM production. Compared to the base case presented in Part I of this pair of articles, potential changes in PGM production and use are quantified with regard to cumulative and yearly environmental impacts and PGM resource use, for the period 2005–2020. Reducing sulfur dioxide (SO2) emissions of PGM producer Norilsk Nickel could cut the cumulative SO2 emissions associated with the use of PGMs in Europe by 35%. Cleaner electricity generation in South Africa could reduce cumulative SO2 emissions by another 9%. Increasing the recycling rate of end-of-life catalytic converters to 70% in 2020 could save 15% of the cumulative primary PGM input into car catalysts and 10% of the SO2 emissions associated with PGM production. In 2020, PGM requirements and SO2 emissions would be, respectively, 40% and 22% lower than the base case.
Substituting palladium for part of the platinum in diesel catalysts, coupled with a probable palladium price increase, could imply 15% more cumulative SO2 emissions if recycling rates do not increase.
A future large-scale introduction of fuel cell vehicles would require technological improvements to significantly reduce the PGM content of the fuel cell stack. The basic design of such vehicles greatly influences the vehicle power, a key parameter in determining the total PGM requirement.  相似文献

Abstract Forty-nine bacterial strains representing five species known to interact with human plasminogen were tested for the ability to bind the two major human plasminogen activators, t-PA and urokinase. The bacterial species tested included Haemophilus influenzae, Neisseria meningitidis, Streptococcus pyogenes, Streptococcus equisimilis and human group G streptococci. All N. meningitidis and 11 of 14 H. influenzae strains displayed substantial binding of t-PA with values in the range of 20–46%. On the contrary, none of the streptococcal strains bound significant amounts of tPA. With urokinase no binding could be found for any of the bacterial species tested. Scatchard analysis with a selected H. influenzae strain (HI23354) demonstrated 10 000 receptors per bacterium for t-PA with a K d value of about 20 nmol l−1. The corresponding values with a selected N. meningitidis strain (Mo 52) was 8500 receptors per bacterium and 70 nmol l−1. t-PA binding could be reduced about 40% by the addition of 10 nmol l−1 of the lysine analogue ϵ-aminocaproic acd (EACA) whereas no inhibitory effect could be demonstrated with arginine. Addition of 2 μmol l−1 of plasminogen which is enough to occupy all bacterial sites for plasminogen did not interfere with the t-PA binding, suggesting that the receptors for t-PA and plasminogen are distinct. Using very high plasminogen concentrations however, t-PA binding could be reduced by about 50% possibly due to an interaction between t-PA and plasminogen in the fluid phase. Our results demonstrate the occurrence of a previously unknown type of bacterial receptor that is capable of specifically binding t-PA.  相似文献

Surface plasmon resonance (SPR) was used as an affinity biosensor to determine absolute heparin concentrations in human blood plasma samples. Protamine and polyethylene imine (PEI) were evaluated as heparin affinity surfaces. Heparin adsorption onto protamine in blood plasma was specific with a lowest detection limit of 0.2 U/ml and a linear window of 0.2–2 U/ml. Although heparin adsorption onto PEI in buffer solution had indicated superior sensitivity to that on protamine, in blood plasma it was not specific for heparin and adsorbed plasma species to a steady-state equilibrium. By reducing the incubation time and diluting the plasma samples with buffer to 50%, the non-specific adsorption of plasma could be controlled and a PEI pre-treated with blood plasma could be used successfully for heparin determination. Heparin adsorption in 50% plasma was linear between 0.05 and 1 U/ml so that heparin plasma levels of 0.1–2 U/ml could be determined within a relative error of 11% and an accuracy of 0.05 U/ml.  相似文献

Strontium (Sr) ions in aqueous solutions could be adsorbed by root tissue powders of Amaranthus spinosus, a common weed found in the fields. The adsorption isotherm could be fitted by either the Langmuir or the Freundlich model with the maximum adsorption capacity being 12.89 mg/g from the Langmuir isotherm. The maximum adsorption capacity of the biosorbent decreased with increasing temperature, whereas alkaline pretreatment enhanced the adsorption capacity 1.9 fold. Alginate gel beads (1 mm diameter) containing the root tissue powders were prepared and packed in a column for continuous adsorption/desorption of Sr in solution. Efficient desorption of Sr could be carried out with 0.1 CaCl2 to give a concentrated Sr solution with 94% recovery.  相似文献

Extracellular nucleotides cause neutrophil degranulation by activating the purinergic receptor subtype P2Y. However, the molecular mechanism involved in the signal pathway remains unknown. A hypothetical scheme suggesting that leukotriene(s) and leukotriene receptor(s) activation is required for extracellular nucleotide-mediated neutrophil degranulation is presented here. Subsequent to the extracellular nucleotide binding to its receptors, intracellular arachidonic acid (AA) levels are elevated. Although AA is a known substrate of the lipoxygenase pathway mediated by 5-lipoxygenase, excess AA could form a complex with S100A8/A9 for transport to the extracellular milieu. Extracellular availability of the S100A8/A9+AA complex could potentially be used for transcellular metabolism by resting and/or activated leukocytes (PMN, MN), vascular endothelium and smooth muscle cells at the inflammatory foci. Once imported into the resting and/or activated leukocytes, AA derived from the S100A8/A9+AA complex could serve as a substrate in the 5-lipoxygenase-mediated leukotriene pathway. Essentially, in addition to extracellular nucleotide-induced leukotrienes, AA derived from the S100A8/A9+AA complex could also be utilized for the synthesis of inflammatory mediators such as leukotriene B(4)(LTB(4)), which in turn could trigger leukocyte degranulation, as well as cellular damage to vascular endothelium and smooth muscle cells, thereby exacerbating inflammation.  相似文献

The productivity of a fermentation is proportional to the biomass concentration. The productivity can therefore be increased by retention of the cells in the fermentor. In this study microfiltration was used for cell retention in a fermentation of glucose to ethanol by baker's yeast. Compared to a system without cell retention the productivity could be increased 12-fold to 55 kg/m3 h at a biomass concentration of 135 kg/m3. Maximal ethanol concentrations of 76 kg/m3 were obtained at conditions of growth. At zero growth conditions in the integrated system the ethanol concentration could be increased to about 115 kg/m3, and could be produced for at least 10 hours. The fermentation results in the integrated system could be described reasonably well with a mathematical model based on a different linear inhibition kinetics for growth and substrate consumption.  相似文献

The synproportionation reaction between ferryl leghemoglobin and oxyleghemoglobin does not occur, at least under conditions where this process could be clearly demonstrated with myoglobin and hemoglobin. In contrast, a cross synproportionation can occur between oxyleghemoglobin and ferryl myoglobin or between ferryl leghemoglobin and oxymyoglobin. The non-exposure, at the surface of the leghemoglobin molecule, of the nearest tyrosine residue to the heme group could explain this behaviour. Thus leghemoglobin per se does not appear to be able to act as an antioxidant in removing H2O2 by synproportionation. However, in the presence of ascorbate and/or glutathione which can reduce ferryl leghemoglobin, this hemoprotein could act as an H2O2-removing antioxidant, in a process similar to that described for myoglobin. This could also explain why, despite the absence of synproportionation, ferryl leghemoglobin is not detected in nodule extracts.  相似文献

以活血丹为材料,应用组织培养和快速繁殖技术,对适于活血丹增殖分化的培养基、培养方式进行了系统的研究。用活血丹叶片作为外植体,在MS添加生长素2,4-D和细胞分裂素BA的培养基上成功诱导出愈伤组织,并对其继代培养条件进行研究,分析了继代培养中褐化的原因。在MS添加NAA和BA的培养基中,活血丹的茎尖和带腋芽茎段能直接诱导出大量丛生芽,随后将不定芽转入MS添加IBA和KT的培养基中,可生成不定根,完成快速繁殖技术体系。结果表明:活血丹愈伤组织诱导的最佳培养基为MS+2,4-D(1.5mg/L)+BA(1.0mg/L),诱导率高达91.38%。丛生芽诱导的适宜培养基为MS+NAA(0.1mg/L)+BA(1·0mg/L),在此培养基上,出芽率达100%,芽增殖系数接近于10,有利于生物量的积累。而根的诱导则在MS+IBA(1.0mg/L)+KT(1.0mg/L)培养基上进行最好,此基础上能诱导出健康、粗壮的根。试管苗炼苗后移栽,成活率达100%。  相似文献

The role of reactive oxygen species (ROS) generated by polymorphonuclear leucocytes (PMNs) in the host response against malaria was investigated. Non-activated human PMNs were added to cultures of P. falciparum in microtitre cells. Parasite viability was evaluated by the incorporation of radioactive hypoxanthine. Using PMN/RBC = 1/150 (starting parasitemia was 1+) the incorporation on the second day in culture was only 61+ of the control cultures. An effect could be observed already after two hours of incubation (30+ reduction at a 1/50 PMN/RBC ratio). A direct contact between the effector and target cells was obligatory for the expression of the damage.

Parasites within G6PD-deficient erythrocytes were more sensitive to the PMNs than normal parasitized erythrocytes. This difference could be attributed to the production of reactive oxygen intermediates in the experimental system, since G6PD-deficient erythrocytes are generally more sensitive to oxidant stress.

Salicylic acid was used as a scavenger and reporter molecule for hydroxyl radical fluxes. It is converted to the corresponding dihydroxybenzoic acid derivatives, which could be detected by HPLC. Uninfected NRBC or parasitized erythrocytes containing young ring forms could trigger the PMNs to produce much less ROS than the mature forms of the parasites. Other factors associated with PMNs may inactivate the parasites, such as phagocytosis, lysosomal enzymes or degradation toxic products of the PMNs. However our results indicate that increased oxidative stress induced by PMNs interfere with the growth of P. falciparum and could play a role in human evolution of abnormal erythrocytes.  相似文献

Electroporation methods for introduction of plasmid DNA into the ruminal bacteria Butyrivibrio fibrisolvens and Streptococcus bovis were developed. Electroporation of the strictly anaerobic B. fibrisolvens was carried out in an anaerobic glovebox with a buffer of 10% (v/v) glycerol and 1 mM MgCl2 in distilled water. Streptococcus bovis electroporation could be carried out aerobically with a buffer of 10% (v/v) glycerol in distilled water. The Escherichia coli/Bacillus subtilis shuttle vector pBS42 could be transformed into B. fibrisolvens strain H17c, selecting for chloramphenicol resistance. The Streptococcus sanguis/E. coli shuttle vector pVA838 could replicate and express erythromycin resistance in Strep. bovis. Both vectors were stable in each organism in the absence of antibiotic selection. While the efficiency was low (<102/μg DNA), the results demonstrate a means to introduce cloned genes into these organisms.  相似文献

以红根草试管苗为材料,研究了不同培养基(MS、1/2MS、1/4MS)、蔗糖浓度和植物生长抑制剂(CCC、PP333、ABA、MH)在红根草试管苗保存中的作用。结果表明:培养基1/2MS对红根草保存最好,保存270d后存活率最高。培养基中不添加蔗糖较添加一定浓度蔗糖时植株的形态和色泽差,但保存时间更长,转继代后能正常恢复生长。添加生长抑制剂能减缓生长速度,延长保存时间,最佳浓度分别为:CCC1.2~1.6mg/L;PP3331.6mg/L;ABA0.5~4.0mg/L;MH0.5mg/L。其中,ABA0.5~4.0mg/L对植株的生长最好,CCC浓度为1.2mg/L和1.6mg/L时,保存时间长,360d时,存活率达90%。  相似文献

A dual laser flow system has been proved and used with a novel staining method for simultaneous quantitative DNA and protein analysis. A diamidinophenylindole compound (DAPI) for DNA has been employed in combination with sulforhodamine (SR 101) for protein. With this dye mixture various cell types of the cervical smear could be identified. Despite some overlapping, clusters of leukocytes, endocervicals, (para)basals, intermediates, superficials and dys/neoplastic cells together with artificial events could be discriminated. Up to now the problem of negative cells which create positive signals (false alarms) in the dys/neoplastic region could not be sufficiently solved.  相似文献

Reliable estimates of feedstock resources are a prerequisite to the establishment of a biomass based-industry for energy and non food products. Field trials in the European Union (EU) show that Miscanthus spp. can produce high yields. Here we use a model (MISCANMOD) coupled with a GIS environment to estimate the contribution that Miscanthus could make to projected national electricity consumption. We describe the integration of different data sets, transformation procedures, and spatial analyses using GIS to produce energy statistics for the EU-25. Overall, Miscanthus grown on the 10% of arable land which is currently in set-aside could generate 282 TWh yr−1 electricity. This would meet 39% of the EU-25 target of 723 TWh yr−1 of electricity from renewable energy sources (RES) by 2010. As RES targets rise, land available for energy crops is also expected to increase. We consider three additional scenarios where Miscanthus could be grown on 10%, 20% and 35% of all agricultural land and we estimate it could generate respectively 345, 691 and 1209 TWh yr−1 of electrical energy. At a national scale France, Poland and Germany have the highest potentials for Miscanthus production based on agricultural land area (respectively 83, 52, 49 TWh yr−1 when 10% agricultural land is used). Finally, we reduced the scale to the EU NUTS2 (Nomenclature of Territorial Units for Statistics) regions to examine regional generation capacities. Key regions have been identified where national RES targets are exceeded. These regions could become net exporters of renewable energy.  相似文献

利用构建的重组菌株Pichia pastoris GS115/GLP-1/HSA,在10L发酵罐中表达了胰高血糖素样肽-1与人血清白蛋白融合蛋白(GLP-1/HSA),表达量为63.6mg/L。发酵液经中空纤维柱浓缩、疏水层析、阴离子交换层析和凝胶过滤分离纯化,获得了较高纯度的GLP-1/HSA,经HPLC分析纯度达95.8%。进一步的体内活性分析结果表明,GLP-1/HSA不仅具有天然GLP-1的生物活性,而且在给药后4h仍能发挥显著性降血糖作用。以上结果表明,利用Pichia pastoris分泌型表达系统和建立的分离纯化方法,能获得大量较高纯度的GLP-1/HSA,为进一步研究和开发能够用于糖尿病临床治疗的长效GLP-1类似物奠定了基础。  相似文献

通过盆栽试验,评价栎属植物在铅锌尾矿中的生长响应及植被恢复前景.分析比较了覆瓦栎、猩红栎、樱皮栎、舒玛栎和白栎5种栎树幼苗在铅锌矿砂中生长30个月后的生物量、根系形态及其对营养元素和重金属的吸收及转移特征.结果表明: 5种栎树在矿砂中均能生长,其中,猩红栎和白栎的生物量较对照有下降趋势,其他3种栎树的生长与对照相比无显著差异;栎树根系生物量均较对照有不同程度增加(猩红栎除外),且仅猩红栎侧根形态学参数较对照有所减少.重金属胁迫下,栎树根系和茎中营养元素浓度较对照无显著变化.5种栎树体内重金属浓度均较低,且其生物富集系数和转移系数均小于1.但樱皮栎叶片和茎中Cd浓度分别为22.4和15.1 mg·kg-1,转移系数为2.3,显著高于其他4种栎树.除猩红栎以外,其他参试栎树均可作为有潜力的污染土壤修复树种.其中舒玛栎的耐性较高、生物富集系数和转移系数较低,是适合在尾矿区造林和生态修复的优选树种.  相似文献

An ability to predict the likelihood of cellular response towards particular chemotherapeutic agents based upon protein expression patterns could facilitate the identification of biological molecules with previously undefined roles in the process of chemoresistance/chemosensitivity, and if robust enough these patterns might also be exploited towards the development of novel predictive assays. To ascertain whether proteomic based molecular profiling in conjunction with artificial neural network (ANN) algorithms could be applied towards the specific recognition of phenotypic patterns between either control or drug treated and chemosensitive or chemoresistant cellular populations, a combined approach involving MALDI-TOF matrix-assisted laser desorption/ionization-time of flight mass spectrometry, Ciphergen protein chip technology and ANN algorithms have been applied to specifically identify proteomic 'fingerprints' indicative of treatment regimen for chemosensitive (MCF-7, T47D) and chemoresistant (MCF-7/ADR) breast cancer cell lines following exposure to Doxorubicin or Paclitaxel. The results indicate that proteomic patterns can be identified by ANN algorithms to correctly assign 'class' for treatment regimen (e.g. control/drug treated or chemosensitive/chemoresistant) with a high degree of accuracy using boot-strap statistical validation techniques and that biomarker ion patterns indicative of response/non-response phenotypes are associated with MCF-7 and MCF-7/ADR cells exposed to Doxorubicin. We have also examined the predictive capability of this approach towards MCF-7 and T47D cells to ascertain whether prediction could be made based upon treatment regimen irrespective of cell lineage. Models were identified that could correctly assign class (control or Paclitaxel treatment) for 35/38 samples of an independent dataset. A similar level of predictive capability was also found (> 92%; n = 28) when proteomic patterns derived from the drug resistant cell line MCF-7/ADR were compared against those derived from MCF-7 and T47D as a model system of drug resistant and drug sensitive phenotypes. This approach might offer a potential methodology for predicting the biological behaviour of cancer cells towards particular chemotherapeutics and through protein isolation and sequence identification could result in the identification of biological molecules associated with chemosensitive/chemoresistance tumour phenotypes.  相似文献
