目的: 研究有氧运动和白藜芦醇对2型糖尿病大鼠肾脏Janus激酶2(JAK2)及转化生长因子-β1(TGF-β1)表达的影响,探讨运动与白藜芦醇改善糖尿病肾损伤的可能作用机制。方法: SD大鼠经5周高糖高脂饲料喂养加腹腔注射链脲佐菌素(STZ)建立糖尿病模型后,将糖尿病大鼠随机分为糖尿病安静组(DC组),糖尿病运动组(DE组),糖尿病药物组(DR组)和糖尿病运动药物组(DER组),每组各12只,另设正常对照组(NC组)。运动组大鼠进行8周的有氧运动(跑速为20 m/min),每天运动60 min,每周运动6 d;药物组大鼠进行8周的白藜芦醇灌胃(每天45 mg/kg,7天/周)。8周末,检测血糖、24 h尿白蛋白(24 h UA)、血肌酐(Scr)、血尿素氮(BUN)的变化;采用荧光定量PCR检测肾脏JAK2 mRNA的表达,免疫组织化学法和Western blot法检测肾脏JAK2和TGF-β1的表达。结果: 8周干预后,与NC组相比, DC组血糖浓度、24 h UA、Scr、BUN均显著上升(P<0.05),肾组织病理损伤加重,肾组织TGF-β1、JAK2和JAK2 mRNA的表达均明显增加(P<0.05)。与DC组相比,DE、DR和DER组血糖浓度、24 h UA、Scr、BUN均显著下降(P<0.05),肾组织病理损伤减轻,肾组织TGF-β1、JAK2和JAK2 mRNA的表达均明显减少(P<0.05),且DER组的降低更显著,与DE、DR组相比差异有显著性(P<0.05)。结论: 有氧运动、白藜芦醇及联合干预可能通过下调肾脏JAK2 mRNA表达,抑制JAK2蛋白的合成,使TGF-β1表达减少,从而改善糖尿病大鼠肾脏损伤的病理性变化。有氧运动联合白藜芦醇干预减轻肾脏病理损伤的效果优于单一的有氧运动或白藜芦醇干预。  相似文献

目的: 观察格列齐特对糖尿病大鼠心肌保护作用及其可能的机制。方法: 将60只健康SD大鼠随机分为正常组(NC,n=10),造模组(n=50)给予高糖高脂饲料4周后,腹腔注射STZ(45 mg/kg)建立糖尿病大鼠模型,随机抽取以FBG≥16.7 mmol/L作为糖尿病模型建立成功。将造模成功的38只糖尿病大鼠随机分为模型组(MC,n=9)、格列齐特组(Glic,80 mg/kg,n=10)、格列本脲组(Glib,2.5 mg/kg,n=10)、法舒地尔组(Fas,10 mg/kg,n=9);NC组和MC组灌胃等容积蒸馏水,Glic组和Glib组灌胃给药,Fas组采用腹腔注射。各组大鼠每天给药一次,每周记录体质量及空腹血糖(FBG),持续8周。实验结束时取血并测定心脏质量,计算心脏质量指数(HWI);测定各组糖化血红蛋白(HbA1c)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)含量以及血清丙二醛(MDA)水平和超氧化物歧化酶(SOD)活性;通过HE和Masson染色,观察心肌病理变化和组织胶原纤维水平;TUNEL染色观察并计算心肌细胞凋亡率;Western blot法检测心肌组织中RhoA、ROCK1、eNOS、Bcl-2和Bax蛋白表达。结果: 与NC组比较,MC组FBG、HWI、HbA1c、TC、TG、LDL-C、MDA水平,心肌组织胶原沉积和心肌细胞凋亡率以及心肌组织中RhoA、ROCK1、Bax蛋白明显升高,SOD活性及HDL-C、eNOS、Bcl-2和体重显著降低(P<0.01);与MC组相比,Glic组FBG、HWI、HbA1c、TC、TG、LDL-C和MDA等指标明显下降,心肌组织胶原沉积及心肌细胞凋亡减轻,心肌组织RhoA、ROCK1、Bax蛋白表达下调(P<0.01或P<0.05),大鼠体重和血清中SOD活性,HDL-C升高,eNOS、Bcl-2蛋白水平升高(P<0.01或P<0.05)。与Glic组相比,Glib组与Fas组体重、血脂、FBG、HWI、MDA以及心肌纤维化和心肌细胞凋亡水平升高,SOD和Bcl-2降低,Glib组心肌组织RhoA、ROCK1、Bax蛋白表达上调(P<0.01或P<0.05)。结论: 格列齐特可改善糖尿病大鼠心肌损伤并减轻心肌细胞凋亡水平,其机制可能与降低血糖,改善氧化应激状态,调控RhoA/ROCK1/eNOS信号通路有关。  相似文献

The purpose of this study was to investigate the protective effect of puerarin on retina pigment epithelial (RPE) cells of diabetic rats against apoptosis. One hundred and eight Sprague-Dawley (SD) rats were randomly divided into 3 groups: control group, streptozotocin (STZ) group and puerarin group. STZ and puerarin groups received 3 d of STZ injection (45 mg/kg per day, i.p.). Additionally, puerarin groups were treated with puerarin (140 mg/kg, i.p.) from the 4th day to the end of experiment. The rats from different groups were sacrificed on 20, 40 and 60 d after STZ injection for harvesting RPE cells. Western blot analysis, DNA laddering, RT-PCR and immunohistochemistry were used for determining the expression of nitrotyrosine (NT, the foot print of peroxynitrite), cell apoptosis, iNOS mRNA and Fas/Fas ligand (FasL) signal transduction in RPE cells, respectively. The results showed that control group maintained low apoptosis level and little NT, iNOS mRNA, Fas/FasL protein expressions, as well as normal blood glucose and body weight during 60 d of the experiment. Compared with control group, STZ group showed obvious apoptosis and higher NT, iNOS mRNA, Fas/FasL protein expressions from 20 d after STZ injection. Puerarin relieved apoptosis of RPE cells and decreased NT, iNOS mRNA, Fas/FasL protein expressions in puerarin group 20 or 40 d after STZ injection, compared with STZ group. These results suggest puerarin can decrease RPE cells apoptosis in diabetic rats by reducing peroxynitrite level and iNOS expression, thus being a potential therapeutic agent in controlling of diabetic retinopathy.  相似文献

This study examined the downstream signaling whereby hyperglycemia may lead to myocardial fibrosis and apoptosis in the left ventricle of diabetic rats. The effects of sulfurous mineral water or sodium hydrosulfide (NaHS) as possible modulators were also examined. Sulfurous mineral water (as drinking water) and NaHS (14 μmol/kg/day, IP) were administered for 7 week to rats with streptozotocin (STZ)-induced diabetes. Hyperglycemia, overproduction of glycated hemoglobin (HbA1C) and serum decline in insulin, C-peptide and insulin like growth factor-I (IGF-I) were observed in diabetic rats. Up-regulation of gene expressions of nuclear factor (NF-κB), profibrogenic growth factor such as transforming growth factor-β1 (TGF-β1), matrix metalloproteniase-2 (MMP-2), procollagen-1 and Fas ligand (Fas-L) were observed in the left ventricle of diabetic rats. A linear positive correlation between TGF-β1 and MMP-2 was also detected in diabetic group. An increase in hydroxyproline level and a disturbance in oxidative balance were detected in heart of diabetic rats. Sulfurous mineral water and NaHS treatment possibly, by improving cardiac GSH level, counteracted the enhanced expression of NF-κB, the profibrogenic and apoptotic parameters. Histopathological examination was in accordance with the biochemical and molecular findings of this study. We suggest a novel therapeutic approach of sulfurous mineral water and exogenous supplementation of H2S in diabetic cardiomyopathy.  相似文献

目的观察Wnt信号通路中β-catenin及其下游靶基因WISP~1在糖尿病大鼠心肌组织中的表达,分析Wnt/β-catenin信号通路在糖尿病大鼠心肌损伤中的作用。方法雄性SD大鼠随机分为正常对照组(Control Group)和糖尿病模型组(DMGroup),腹腔注射STZ55mg/kg诱导糖尿病大鼠模型,喂养至8周。测定大鼠的空腹血糖、心体比和心肌羟脯氨酸含量;电镜观察心肌超微结构变化,免疫组化法观察心肌组织β-catenin和WISP-1的表达。结果与对照组相比,糖尿病组大鼠空腹血糖水平明显增加,心体比增加,心肌羟脯氨酸含量增高。心肌超微结构显示肌纤维断裂,线粒体呈空泡样改变。心肌β-catenin和WISP-1蛋白表达增加。结论糖尿病大鼠心肌组织β-catenin和其下游靶基因WISP-1表达增加,提示Wnt/β-catenin信号通路的激活参与糖尿病所致的心肌损伤。  相似文献

目的:研究淫羊藿总黄酮(TFE)对链脲佐菌素(STZ)致糖尿病大鼠肾脏损伤的影响,并初步探讨其可能的作用机制。方法:健康雄性SD大鼠一次性尾静脉注射STZ(40 mg/kg)建立糖尿病模型。动物随机分成3组(n=10):对照组、模型组和TFE组(100 mg/kg,i.g.)。12周后,处死大鼠。测定空腹血糖,肾脏脏器系数,血清尿素氮(BUN)、肌酐(Cr)含量;测定肾组织中超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;Masson染色观察肾组织胶原纤维增生;免疫组化测定转化生长因子β1(TGF-β1)蛋白的表达。结果:与对照组比较,模型组肾脏脏器系数增大、肾功能下降、肾组织抗氧化能力降低;病理学可见肾小球、肾小管间质纤维化;同时TGF-β1蛋白表达水平上调。TFE组明显改善上述指标。结论:TFE对STZ致糖尿病大鼠肾脏损伤有明显的改善作用,其作用机制可能与抗氧化作用和抑制TGF-β1蛋白表达有关。  相似文献

葛根素对糖尿病心肌细胞的保护及其机制研究   总被引:1,自引:0,他引:1  
观察葛根素(Puerarin)对链脲佐菌素(streptozotocin,STZ)诱导的糖尿病大鼠心肌细胞的保护作用,并探讨血小板反应素1(Thrombospondin-1,TSP-1)的表达改变及其作用。雄性SD大鼠45只随机分为三组(n=15):糖尿病组和葛根素治疗组采用一次腹腔注射链脲佐菌素(STZ)65mg/kg制备糖尿病模型,其中葛根素治疗组于造模后葛根素腹腔注射4周(100mg/kg/day),正常对照组仅腹腔注射等量生理盐水(6ml/kg),同样喂养4周。四周后各组大鼠处死。H—E染色及透射电子显微镜观察三组大鼠心肌细胞纤维显微结构和超微结构的病理改变.免疫组化和实时荧光定量PCR法观察大鼠心肌细胞中TSP-1蛋白和mRNA表达的变化.同时利用Langendorff离体心脏灌流法测定各组大鼠心室肌细胞功能。结果发现葛根素治疗组较糖尿病组大鼠的体重增加明显,同时血糖下降,有显著性差异(P〈0.01)。H—E染色显示糖尿病大鼠多处心肌肌丝紊乱伴少量炎症细胞浸润,电镜下发现有线粒体嵴消失溶解,肌丝排列紊乱等病理改变,而葛根素治疗组大鼠偶见上述病理变化。免疫组化显示葛根素治疗组心肌内TSP-1阳性细胞密度小于糖尿病大鼠,TSP-1 mRNA表达也比糖尿病大鼠要低。此外葛根素治疗组大鼠的左室收缩末压(LVSEP)、左心室舒张末期压(LVEDP)等心功能指标均明显低于正常组(P〈0.01),但较糖尿病组有显著改善(P〈0.01)。上述结果显示葛根素能保护糖尿病大鼠心肌细胞的高糖损伤和维持心室肌细胞的功能,而该机制可能与抑制心肌细胞TSP-1表达的水平有关。  相似文献

Diabetic nephropathy is one of the most serious complications of diabetes and the major cause of end-stage renal failure. Consequences of diabetic nephropathy include increased kidney size and glomerular volume, thickening of basement membranes and progressive accumulation of extracellular matrix. Reports in the literature support an association between increased secretion of inflammatory molecules, such as cytokines, growth factors and metalloproteinases, and development of diabetic nephropathy. We investigated the potential of granulocyte colony- stimulating factor (G-CSF) as a therapeutic candidate for preventing diabetic nephropathy. We used 21 8-week-old male rats; 14 were administered a single dose of 60 mg/kg streptozotocin (STZ) to induce diabetes. The rats were divided into three groups of seven: group 1, control; group 2, diabetic; group 3, diabetic plus G-CSF treatment. After 4 weeks, immunoexpressions of transforming growth factor β1 (TGF-β1), Akt and CD34 levels were measured in the kidney tissue. Blood glucose, urine protein and the glomerular area also were measured for each group. We found that G-CSF treatment decreased TGF-β1 immunoexpression, urine protein and glomerular area in kidneys of diabetic rats, and increased CD 34 and Akt immunoexpression in kidneys of diabetic rats. The effects of G-CSF were independent of blood glucose levels. G-CSF may be a useful therapeutic agent for preventing diabetic nephropathy.  相似文献

目的:观察彩色蚕茧提取物—丝胶对糖尿病肾病大鼠肾脏转化生长因子-β1(TGF-β1)和Smad3蛋白表达的影响。方法:60只雄性SD大鼠随机分为5组(n=12):正常对照组、糖尿病肾病模型组、丝胶治疗组、二甲双胍组和丝胶预防组。模型组、丝胶治疗组、丝胶预防组和二甲双胍组大鼠均建立链脲佐菌素(STZ)致动物模型,以血糖≥16.7 mmol/L作为成模标准;待模型成功建立后,丝胶治疗组大鼠给予丝胶灌胃(2.4 g/(kg.d),35 d)、二甲双胍组大鼠给予二甲双胍灌胃(55.33 mg/(kg.d),35 d),丝胶预防组大鼠于注射STZ前给予同等剂量丝胶灌胃35天。分别检测各组大鼠血糖和肾重/体重;免疫组化染色观察肾脏TGF-β1蛋白的表达;Western blot法观察肾脏Smad3蛋白的表达。结果:与正常对照组大鼠相比,模型组大鼠血糖、肾重/体重和肾脏TGF-β1、Smad3蛋白的表达均明显升高(P〈0.01)。丝胶治疗组、丝胶预防组和二甲双胍组大鼠的血糖和肾脏TGF-β1、Smad3蛋白的表达明显低于模型组(P〈0.01),且丝胶治疗组、丝胶预防组与二甲双胍组比较无明显差别(P〉0.05);丝胶治疗组、丝胶预防组和二甲双胍组大鼠的肾重/体重明显低于模型组(P〈0.01),且丝胶治疗组、丝胶预防组大鼠的肾重/体重明显低于二甲双胍组(P〈0.05)。结论:丝胶可抑制糖尿病肾病大鼠肾脏TGF-β1/Smda3信号通路的激活,减轻肾小球硬化和肾间质纤维化,发挥对糖尿病肾病肾脏损伤的保护和预防保护作用,且作用与二甲双胍相当。  相似文献

Ginsenoside Rg1 has been demonstrated to have cardiovascular protective effects. However, whether the cardioprotective effects of ginsenoside Rg1 are mediated by endoplasmic reticulum (ER) stress‐induced apoptosis remain unclear. In this study, among 80 male Wistar rats, 15 rats were randomly selected as controls; the remaining 65 rats received a diet rich in fat and sugar content for 4 weeks, followed by intraperitoneal injection of streptozotocin (STZ, 40 mg/kg) to establish a diabetes model. Seven days after STZ injection, 10 rats were randomly selected as diabetic model (DM) controls, 45 eligible diabetic rats were randomized to three treatment groups and administered ginsenoside Rg1 in a dosage of 10, 15 or 20 mg/kg/day, respectively. After 12 weeks of treatment, rats were killed and serum samples obtained to determine cardiac troponin (cTn)‐I. Myocardial tissues were harvested for morphological analysis to detect myocardial cell apoptosis, and to analyse protein expression of glucose‐regulated protein 78 (GRP78), C/EBP homologous protein (CHOP), and Caspase‐12. Treatment with ginsenoside Rg1 (10–20 mg/kg) significantly reduced serum cTnI levels compared with DM control group (all P < 0.01). Ginsenoside Rg1 (15 and 20 mg/kg) significantly reduced the percentage of apoptotic myocardial cells and improved the parameters of cardiac function. Haematoxylin and eosin and Masson staining indicated that ginsenoside Rg1 could attenuate myocardial lesions and myocardial collagen volume fraction. Additionally, ginsenoside Rg1 significantly reduced GRP78, CHOP, and cleaved Caspase‐12 protein expression in a dose‐dependent manner. These findings suggest that ginsenoside Rg1 appeared to ameliorate diabetic cardiomyopathy by inhibiting ER stress‐induced apoptosis in diabetic rats.  相似文献

目的:研究Iuteolin对链脲佐菌素诱导的Ⅰ型糖尿病大鼠心功能及心肌线粒体氧化应激的影响。方法:雄性SD大鼠,随机分成正常对照组,Iuteolin对照纽,糖尿病模型组,低剂量Iuteolin(10ms/(kg·d))灌胃治疗组,高剂量Iuteolin(100ms/(kg·d))灌胃治疗组。各组大鼠饲养8周后,测体重、血糖、心功能、左心室重量、心肌胶原含量及活性氧自由基(ROS)水平,分离心肌线粒体检测ROS水平、超氧化物歧化酶(SOD)活性及线粒体肿胀程度。结果:Iuteolin处理对糖尿病大鼠血糖无明显影响,但可减少糖尿病引起的体重下降。高剂量Iuteolin可显著减小糖尿病大鼠心室与体重比值,提高左室发展压,降低左室舒张末压。高剂量Iuteolin治疗后,糖尿病大鼠心肌ROS及胶原含量。心肌线粒体ROS水平与肿胀程度均明显下降,心肌线粒体SOD活性明显增加。结论:Iuteolin处理可显著改善糖尿病大鼠心功能.其机制可能与减轻心肌线粒体氧化应激及抑制线粒体肿胀有关。  相似文献

目的: 探讨西格列汀对糖尿病小鼠心肌重构和自噬的影响和可能的机制。方法: 10周龄的C57小鼠腹腔注射STZ 50 mg/(kg·d),连续注射5 d,7 d测血糖浓度>16.7 mmol/L视为糖尿病小鼠造模成功,造模成功4周后给与药物干预。本实验分四组,对照组(control, 腹腔注射等体积的缓冲液, n=10)、模型组(Streptozocin, STZ腹腔注射诱导糖尿病模型,n=8)、处理组(在模型组基础上给与西格列汀灌胃10 mg/(kg·d),n=8)、抑制剂组(在处理组的基础上给与腹腔注射Compound C (AMPK通路抑制剂,10 mg/(kg·d),n=8),对照组腹腔注射等体积缓冲液,6周后称体重,处死,取小鼠心脏并分离心室称重,计算心室/体重比,HE染色观察心肌细胞形态,Masson染色观察纤维化程度,Western blot 检测心肌脑钠肽(BNP)、转化生长因子β(TGF-β)、缝隙连接蛋白43(Cx43)、AMP依赖的蛋白激酶(AMPK)、LC3B蛋白表达。结果: 给药6周后,与对照组相比,模型组小鼠体重没有明显改变,心室/体重比明显增加(P<0.05),苏木素-伊红(HE)染色显示细胞增大,Masson染色显示心肌间隙纤维化增多,BNP、TGF-β蛋白明显升高,Cx43、LC3B、AMPK蛋白下降(P<0.05)。与模型组相比,西格列汀组BNP、TGF-β蛋白明显下降,Cx43、LC3B、AMPK蛋白增多(P<0.05)。然而Compound C会抑制Cx43、LC3B、AMPK蛋白表达的上调(P< 0.05)。结论: 西格列汀可以改善糖尿病小鼠心肌肥厚和纤维化,并且可以通过AMPK相关通路调节Cx43和自噬。  相似文献

In the current study, the effect of soy protein and genistein, one of the main isoflavones in soybeans, on blood glucose, lipid profile, and antioxidant enzyme activities in streptozotocin (STZ)-induced diabetic rats was investigated. Male Sprague-Dawley rats were divided into nondiabetic control, STZ, STZ-genistein supplemented group (STZ-G; 600 mg/kg diet), and STZ-isolated soy protein supplemented group (STZ-ISP; 200 g/kg diet). Diabetes was induced by a single injection of STZ (50 mg/kg BW) freshly dissolved in 0.1 mol/L citrate buffer (pH 4.5) into the intraperitonium. Diabetes was confirmed by measuring the fasting blood glucose concentration 48-h post-injection. The rats with blood glucose level above 350 mg/dL were considered to be diabetic. Genistein and ISP were supplemented in the diet for 3 weeks. The supplementation of genistein and ISP increased the plasma insulin level but decreased the HbA(IC) level of the STZ-induced diabetic rats. The supplementation of genistein and ISP increased the glucokinase level of the STZ-induced diabetic rats. A significant reduction in glucose-6-phosphatase was observed in the groups treated with genistein and ISP in comparison with the diabetic control group. Hepatic superoxide dismutase, catalase, and glutathione peroxidase activities of the STZ-induced diabetic rats were significantly decreased in comparison with the control rats. Administering genistein and ISP to the STZ-induced diabetic rats significantly increased those enzyme activities. The concentration of thiobarbituric acid reactive substances in the STZ-induced diabetic rats was significantly elevated, while the genistein and ISP supplement decreased it to the control concentration. Genistein and ISP supplements seem to be beneficial for correcting the hyperglycemia and preventing diabetic complications.  相似文献

Diabetic nephropathy (DN) is the leading cause of death in diabetic patients and the current treatment options available have a limited significance. The insect galls of Quercus infectoria are traditionally important in the treatment of numerous diseases including diabetes. Hence, the present study was undertaken to evaluate the effect of Q. infectoria gall extract (QIGE) against experimental DN. Type 2 diabetes was induced by feeding rats with a high-fat diet (HFD) initially for 5 weeks, followed by a single intraperitoneal injection of streptozotocin (STZ, 35?mg/kg?bw/day). QIGE was administered to the rats orally at doses of 100 and 200?mg/kg?bw/day, respectively. At the end of the experimental period, various glycemic and renal function parameters were evaluated in the serum, urine and kidney tissues. The QIGE treatment significantly (p?p?via the inhibition of hyperglycemia-induced oxidative stress and renal TGF-β expression and is, therefore, a potential therapeutic agent in the treatment of diabetic complications, especially DN.  相似文献

目的:探讨激动乙醛脱氢酶2(ALDH2)在糖尿病大鼠心肌损伤中的作用。方法:腹腔注射55 mg/kg链脲佐菌素复制糖尿病大鼠模型,分为糖尿病组和乙醇+糖尿病组(n=8)。8周后行离体心肌缺血/再灌注(I/R),测定心室动力学指标和复灌期间冠脉流出液中乳酸脱氢酶(LDH)含量。测定空腹血糖、糖化血红蛋白(HbA1c)水平。RT-PCR和Western blot测定左心室前壁心尖组织线粒体ALDH2 mRNA和蛋白表达。结果:与正常大鼠心肌I/R相比,糖尿病大鼠左室发展压、左心室最大上升和下降速率、左室做功进一步下降,左室舒张末压抬高,复灌期冠脉流出液中LDH释放增多,心室ALDH2 mRNA和蛋白表达降低;与糖尿病大鼠心肌I/R相比,ALDH2激动剂乙醇明显促进左室发展压、左心室最大上升和下降速率、左室做功的恢复,降低左室舒张末压,同时降低HbA1c水平和LDH的释放,ALDH2 mRNA和蛋白表达增高。结论:糖尿病大鼠心肌缺血/再灌注时,心肌ALDH2表达降低;增强ALDH2在糖尿病大鼠心肌中的表达可发挥保护作用。  相似文献

目的动态观察链脲佐菌素(STZ)诱导的糖尿病大鼠血糖控制前后肾小管上皮细胞(TEC)中血管内皮生长因子(VEGF)、转化生长因子β1(TGF-β1)、Smad2/3、Smad4的表达情况,探讨四者在糖尿病大鼠TEC表型转变和肾间质纤维化中可能发挥的作用及相互关系。方法实验动物随机分为5组,依病程长短分为①A组(2周组),②B组(4周组),③C组(8周组),④D组(16周组),⑤E组(24周组),每组分别设有正常对照组(N组)和糖尿病组(a组);另外,16周、24周两组加设胰岛素治疗组(b组)。采用尾静脉注射STZ法复制糖尿病大鼠模型;免疫组织化学方法检测肾小管VEGF、TGF-β1、Smad2/3、Smad4及α-平滑肌肌动蛋白(-αSMA)和纤连蛋白(FN)的表达;Western blot检测肾皮质VEGF和TGF-β1蛋白;PAS染色光镜观察肾小管基底膜变化及细胞外基质沉积情况等形态学改变;生化方法测定血糖、血肌酐及24小时尿蛋白量。结果正常对照组VEGF、TGF-β1及Smad2/3、Smad4在肾小管均有少量表达,-αSMA在肾小管无表达;糖尿病组肾小管前述四者的表达均显著高于正常对照组,且从16周开始肾小管上皮细胞可见α-SMA蛋白阳性表达;糖尿病16周时肾小管VEGF、TGF-β1、Smad2/3、Smad4两两之间呈正相关;随糖尿病进展,α-SMA及FN在肾小管表达增多,24h尿蛋白增多,肾脏肥大指数增大,而VEGF、TGF-β1二者都分别和-αSMA、FN、24h尿蛋白及肾脏肥大指数呈正相关性;胰岛素治疗后,VEGF、TGF-β1、Smad2/3、Smad4及FN的表达都比糖尿病组明显下降,且各指标之间的正相关性依然存在,-αSMA蛋白则呈阴性表达。结论糖尿病肾病大鼠肾小管上皮细胞表达的VEGF、TGF-β1及Smad2/3、Smad4参与了TEC表型转变和肾间质纤维化的发生,并且VEGF和TGF-β1相互作用,共同促进了肾脏损害。胰岛素对DN大鼠TEMT和肾间质纤维化的影响可能部分是通过间接阻断VEGF、TGF-β1和Smad2/3、Smad4在TEC中的合成来实现的。  相似文献

Berberine (BBR) is one of the main constituents in Rhizoma coptidis and it has widely been used for the treatment of diabetic nephropathy. The aims of the study were to investigate the effects and mechanism of action of berberine on renal damage in diabetic rats. Diabetes and hyperglycaemia were induced in rats by a high-fat diet and intraperitoneal injection of 40 mg/kg streptozotocin (STZ). Rats were randomly divided into 5 groups, such as i) control rats, ii) untreated diabetic rats iii) 250 mg/kg metformin-treated, iv and v) 100 and 200 mg/kg berberine-treated diabetic rats and treated separately for 8 weeks. The fasting blood glucose, insulin, total cholesterol, triglyceride, glycosylated hemoglobin were measured in rats. Kidneys were isolated at the end of the treatment for histology, Western blot analysis and estimation of malonaldehyde (MDA), superoxide dismutase (SOD) and renal advanced glycation endproducts (AGEs). The results revealed that berberine significantly decreased fasting blood glucose, insulin levels, total cholesterol, triglyceride levels, urinary protein excretion, serum creatinine (Scr) and blood urea nitrogen (BUN) in diabetic rats. The histological examinations revealed amelioration of diabetes-induced glomerular pathological changes following treatment with berberine. In addition, the protein expressions of nephrin and podocin were significantly increased. It seems likely that in rats berberine exerts an ameliorative effect on renal damage in diabetes induced by high-fat diet and streptozotocin. The possible mechanisms for the renoprotective effects of berberine may be related to inhibition of glycosylation and improvement of antioxidation that in turn upregulate the expressions of renal nephrin and podocin.  相似文献

Clinical and preclinical studies have shown that diabetic individuals present more depressive behaviors than non-diabetic individuals. Taurine, one of the most abundant free amino acids in the central nervous system, modulates a variety of biological functions and acts as an agonist at GABAA receptors. Our objective was to assess the antidepressant effect of taurine in diabetic rats. Additionally, we studied the effect of taurine on weight gain, water and food intake, and blood glucose levels in diabetic and non-diabetic rats. Male Wistar rats were divided into control (CTR) and streptozotocin-induced diabetic (STZ) groups and were administered daily 0, 25, 50 or 100?mg/kg of taurine (n?=?10 per subgroup) intraperitoneally. After 28?days of treatment, the animals were exposed to the forced swimming test, and their behaviors were recorded. Weight gain, water and food intake, and blood glucose levels were measured weekly. Our results showed that STZ rats had a higher immobility duration than CTR rats, and taurine decreased this depressive-like behavior in STZ rats at doses of 25 and 100?mg/kg. Both of these doses of taurine also decreased water intake and improved weight gain in STZ rats. All doses of taurine decreased the water intake in CTR rats. Taurine, at a dose of 100?mg/kg, decreased food intake and blood glucose levels in STZ rats. Because taurine is a GABA agonist and both amino acids are lower in the plasma of diabetic and depressive individuals, we hypothesize that taurine may represent a new adjuvant drug for the treatment of depression in diabetic individuals.  相似文献

建立糖尿病性心肌病(DCM)大鼠模型,观察不同剂量链脲佐菌素(STZ)单次腹腔注射后大鼠心肌和胰腺的病理学变化。用STZ 50 mg/kg、55 mg/kg、60 mg/kg 3种剂量单次腹腔注射,制备糖尿病大鼠模型;以柠檬酸三钠-柠檬酸缓冲液腹腔注射,作为对照。72 h后,测空腹血糖及做口服葡萄糖耐量实验(OGTT);3周后,HE染色观察各组大鼠胰腺和心肌形态学变化,Masson三色染色观察心肌纤维化改变。OGTT和空腹血糖显示3组存活大鼠糖尿病均成模;3周末,50 mg/kg和55 mg/kg剂量死亡率为25%;60 mg/kg剂量高,达到75%;HE染色显示55 mg/kg剂量组大鼠胰岛明显萎缩,轮廓不清晰,胰岛细胞数量少,心肌细胞肥大、排列紊乱,细胞间隙增大,并有炎症细胞浸润;50 mg/kg组胰岛和心肌也有变化,但无55 mg/kg组明显。心肌Masson染色显示55 mg/kg组心肌内胶原组织明显增多,排列紊乱,分布不均。55 mg/kg剂量的STZ单次注射大鼠腹腔,造模3周可以建立较明显的DCM模型,可为DCM的组织病理学和实验研究提供一个较好的动物模型。  相似文献
