The baculovirus expression vector system (BEVS) is used extensively for the production of proteins from exogenous cDNAs. However, BEVS is not ideal for pharmaceutical production of glycoproteins owing to the properties of the N-glycans in the expressed products and that insect cells lack several of the enzymes required for mammalian-type N-glycan synthesis. Racemic diisophorone (500 mg) was converted by Cephalosporium aphidicola and Neurospora crassa over 10 days at 25 °C to 8β-hydroxydiisophorone in yields of 10% (52 mg) and 20% (103 mg), respectively. N-(9-Fluorenylmethoxycarbonyl)-ω-aminoalkyl-, N-(9-fluorenylmethoxycarbonyl)-8-amino-3,6-dioxaoctyl, and N-[(9-fluorenylmethoxycarbonyl)-6-aminohexanoyl]-2-aminoethyl triphosphates were synthesized. All of them were shown to be the substrates of the calf thymus terminal deoxynucleotidyl transferase. Transgenic Laminaria japonica gametophytes producing a recombinant tissue-type plasminogen activator (rtPA) protein, which is an effective third-generation thrombolytic agent for acute myocardial infarction (AMI), were cultured in an illuminated bubble column bioreactor. A maximum final dry cell weight of 1120 mg l−1 was obtained in batch culture with an initial dry cell weight of 126 mg l−1 and with aeration rate of 1.2 l air min−1 l−1 culture, nitrate at 1.5 mM and phosphate at 0.17 mM. The yield of rtPA was 56 μg g−1 dry cell wt. Hydraulic redistribution (HR) in karité (Vitellaria paradoxa) and néré (Parkia biglobosa) tree species was studied by monitoring the soil water potential (ψs) using thermocouple psychrometers at four compass directions, various distances from trees and at different soil depths (max depth 80 cm) during the dry seasons of 2004 and 2005. A modified WaNuLCAS model was then used to infer the amount of water redistribued based on ψs values. Tree transpiration rate was also estimated from sap velocity using thermal dissipative probes (TDP) and sapwood area, and the contribution of hydraulically redistributed water in tree transpiration was determined. The results revealed on average that 46% of the psychrometer readings under karité and 33% under néré showed the occurrence of HR for the two years. Soil under néré displayed significantly lower fluctuations of ψs (0.16 MPa) compared to soil under karité (0.21 MPa). The results of this study indicated that the existence of HR leads to a higher ψs in the plant rhizosphere and hence is important for soil water dynamics and plant nutrition by making more accessible the soluble elements. The simulation showed that the amount of water redistributed would be approximately 73.0 L and 247.1 L per tree per day in 2005 for karité and néré, and would represent respectively 60% and 53% of the amount transpired a day. Even though the model has certainly overestimated the volume of water hydraulically redistributed by the two species, this water may play a key role in maintaining fine root viability and ensuring the well adaptation of these species to the dry areas. Therefore, knowledge of the extent of such transfers and of the seasonal patterns is required and is of paramount importance in parkland systems both for trees and associated crops.  相似文献

Cell volume distribution in Chlorella vulgaris cultures coming out of senescence was measured by flow cytometry every 6 h for 114 h in a full-factorial experiment with initial nitrate (420–4200 g NO3-N l–1), phosphate (9–186 g PO4-P l–1), and continuous light (50–330 E m–2 s–1) as treatments. The maxima in median and median absolute deviation (MAD) of cell volume were achieved within 6 h of each other and their timing was not affected by any treatment. HP (2-20) [derived from the N-terminal region of Helicobacter pylori Ribosomal Protein L1 (RPL1)], a 19-mer peptide, possesses broad-spectrum anti-microbial activity. As the N- (residues 2–3) and C-terminal (residues 14–20) residues can be deleted without affecting antimicrobial activity, we have now determined the minimum chain length necessary for the retention of antimicrobial activity, and its mode of action. The N- (residues 2–3) and C-terminal (residues 17–20) truncated fragments [HP (4–16)] induce increased antibiotic activity against several bacterial strains without hemolysis. Production of clavulanic acid (CA) by Streptomyces clavuligerus in a shake-flask culture increased from 92 to 180 mg l−1 with an increased O2 transfer efficiency (0.039 → 0.058 s−1), which maintained the redox potential values above −250 mV. Compared with traditional measures, such as dissolved O2 concentration and respiratory activity, the redox potential can easily be determined and correlates closely with CA production. It can therefore be used to monitor microbial activities during biosyntheses of secondary metabolites. The study of the reaction of p-nitrophenyl acetate (PNPA) with histidine and certain derivatives showed that the species in which the amino group is unprotonated (R(NH2)Im) react with second-order rate constants ( ) that are higher than predicted by a Brønsted relation for a series of neutral amino acids. The reason for this behavior was investigated through an analysis of the kinetics of the reaction of PNPA with these compounds in order to assess the reactivities of the amino and imidazole groups in the two species . The rate constant for the reaction with the imidazole group ( ) of Nπ-methyl histidine agrees with the value predicted by a Brønsted relation obtained from a series of model imidazole compounds. Nτ-Methyl histidine, however, is unreactive, indicating that Nτ is the reactive nitrogen in the imidazole ring of histidine. The values found for histidine, histidine methyl ester, and Nα-dimethyl histidine are lower than predicted by the Brønsted relation. This behavior was found to be due to low reactivity of the
. The evidence presented suggests that the lower reactivity of is due to an ion-dipole interaction between the protonated amino group and the unprotonated imidazole ring, which displaces the tautomeric equilibrium toward the unreactive Nτ-H form. The higher reactivity of the imidazole group in the species R(NH2)Im, relative to that in , is responsible for the observed high values for histidine, for histidine methyl ester, for Nτ-methyl histidine, and for Nα-dimethyl histidine, in contrast with the normal value found for Nτ-methyl histidine. The conclusions from this study of histidine and its derivatives support the proposal of an interaction between the protonated N-terminal amino group and the imidazole ring of His6 in the octapeptide hormone angiotensin.  相似文献

A monoclonal antibody against cis-3-hexen-1-ol was prepared and used to separate and/or concentrate Δ17-6-keto-prostaglandin F1α (PGF1α) in the human sera. cis-3-Hexen-1-ol was conjugated with the human serum albumin (HSA) according to the N-succinimidylester method and hyperimmunized to BALB/c mouse. The monoclonal afntibodies were obtained from hybridoma clones established by a fusion between SP2/0-Ag14-k13 mouse myeloma cells and splenocytes of a mouse. A monoclonal antibody, named 4G9-12B, recognized the epitope characteristic for ω3-olefin structure. The 4G9-12B antibody became more specific for Δ17-6-keto-PGF1α than 6-keto-PGF1α by applying inhibition ELISA using amino-residue coating plates. Using the prepared immunoaffinity columns of this antibody, Δ17-6-keto-PGF1α was clearly detected in 6 pg/ml of the human blood sera by GC/MS analysis. These results suggest that the monoclonal antibody to the partial structure of trienoic prostanoid, ω3-olefin unit, and that its immunoaffinity columns are useful in separating and concentrating Δ17-6-keto-PGF1α in the human blood or urine.  相似文献

Root cultures of Gloriosa superba were treated with 5 mm methyl jasmonate and 125 μm AlCl3 which enhanced the intracellular colchicine content of the roots by 50-fold and 63-fold, respectively. Ten millimolar of CaCl2 and 1 mm CdCl2 enhanced biomass significantly (7- to 8.6-fold, respectively) while maximum release of colchicine into the medium was obtained with 10 mm CdCl2. Casein hydrolysate, yeast extract and silver nitrate had no significant effect on growth and colchicine accumulation in root cultures. Heat stress adversely affects plant growth and development, while glycinebetaine (GB) plays a protective role under stressful conditions. The objective of this study was to assess the optimum level of GB for use as a presowing seed treatment and the subsequent effect on the heat tolerance of barley (Hordeum vulgare L. cv. Haider-93) seedlings. Among a range of GB levels, the 20 mM concentration emerged as the most effective in enhancing seed germination, shoot fresh and dry weight and shoot water content under heat stress, and this level was selected for further studies. Time course changes revealed that the seedlings developing from 20 mM GB treated seeds had greater shoot dry weight, net photosynthetic rate (PN), leaf water potential (ψw) and reduced relative membrane permeability (RMP), compared to no-GB treated plants under heat stress. Correlations between dry weight and high PN (r = 0.881), low ψw (r = −0.938) and RMP (r = −0.860) of shoots suggested the involvement of GB in heat stress tolerance. Leakage of Ca2+ and NO3 was the greatest followed by K+ and PO43− under no-GB seed treatment, and GB application under heat stress appreciably reduced the leakage of all these ions, particularly Ca2+, K+ and NO3. In conclusion GB absorbed by seeds, after translocation to the seedlings, enhanced their capacity to maintain greater water content, and higher seedling vigor by virtue of increased PN, reduced RMP and leakage of important ions under heat stress. These results have implications for final field stand under the conditions where the ambient temperature is supra-optimal for barley growth.  相似文献

A β-1,3-glucanase was detected, using laminarin as substrate, in the culture broth of Chaetomium sp. Major activity was associated with a 70 kDa protein band visualized on a polyacrylamide gel. β-1,3-Glucanase was purified by a one-step, native gel purification procedure. Optimal activity was observed at pH 6.0 and 30 °C (over 30 min). It could degrade cell walls of plant pathogens including Rhizoctonia solani, Gibberella zeae, Fusarium sp., Colletotrichum gloeosporioides and Phoma sp. The N-terminal amino acid residues of the purified β-1,3-glucanase are PYQLQTP, which do not exhibit homology to other fungal β-1,3-glucanases suggesting it may be a novel enzyme. Prostaglandin E2 (PGE2) secreted by Day-6, Day-7, Day-8 and Day-9 equine embryos (ovulation = Day 0) during in vitro incubation was measured by radioimmunoassay. Embryonic PGE2 secretion (ng/embryo/24 hr) was detectable on Day 6 (0.27±0.39), tended to increase (P <0.1) on Day 7 (0.57±0.88), and increased significantly (P <0.05) on Day 8 (2.23±0.86) and Day 9 (4.13±0.71). Several synthetic adeonosine analogs: 8-fluoro-, 8-azido-, 8-iodo-, 8-methylthioadenosine; 8-bromo-2′-deoxyadenosine, 8-bromoxylofuranosyladenine, 5′-benzoly-8-bromoadenosine; 8,2′-S-, 8,2′-O-, 8,2′-NH-, 8,2′-N-CH3-, 8,3′,-S-, 8,3′-O-, 8,5′-S- and 8,5′O-cycloadenosine; 1-deaza- and 3-deazaadenosine, as well as tubercidine (7-deazaadenosine), were tested as substrates of calf intestine adenosine deaminase.It was found that the adenine base of adenosine should be in the range φrmCN = 0–120° (anti to syn-anti) and 8-fluoroadenosine was hydroylzed very slowly. The purine base should have N1, N3 or N7 atoms for the hydrolysis and only 1-deazaadenosine revealed an inhibitory effect toward the hydrolysis of adenosine.5′-OH group should be in the position of S-configuration and must not be substituted.  相似文献

Factors influencing the release and anchorage of cytochrome c to the inner membrane of brain mitochondria have been investigated. Metabolic activity of mitochondria caused a decrease in the membrane potential Δψm, accompanied by detachment of the protein from the inner membrane. In a model system of cytochrome c reconstituted in cardiolipin (CL) liposomes, phosphate was used to breach the hydrophilic lipid-protein interactions. About 44% cytochrome c was removable when heart CL (80% 18:2n-6) was employed, whereas the remaining protein accounted for the tightly bound conformation characterized by hydrophobic lipid-protein interactions. Cytochrome c release from brain CL liposomes was higher compared to heart CL, consistent with lower polyunsaturated fatty acid content. The release was even higher with CL extracted from metabolically stressed mitochondria, exhibiting more saturated fatty acid profile compared to control (30% vs.17%). Therefore, weakening of the hydrophobic interactions due to saturation of CL may account for the observed cytochrome c release from mitochondria following metabolic stress. Moreover, mitochondria enriched with polyunsaturated CL exhibited higher Δψm, compared to less unsaturated species, suggesting that CL fatty acid composition influences Δψm. Mitochondria incorporated exogenous cytochrome c without protease-sensitive factors or Δψm. The internalized protein anchored to the inner membrane without producing swelling, as monitored by forward and side light scattering, but produced Δψm consumption, suggesting recovery of respiratory activity. Two isoforms of laccase produced from the culture supernatant of Pycnoporus sanguineus were partially purified by phenyl-Sepharose chromatography. Molecular masses of the enzymes were 80 kDa (Lac I) and 68 kDa (Lac II). Optimum activity of Lac I was at pH 4.8 and 30 °C, and Lac II was at pH 4.2 and 50 °C over 5 min reaction. The Km values of enzymes toward syringaldazine were 10 μm (Lac I) and 8 μm (Lac II). Sodium azide inhibited Lac I (85%) and Lac II (75%) activities. To clarify the diversity and function of isozymes of ascorbate peroxidase (APX) in plants, a method of producing large quantities of these proteins is needed. Here, we describe an Escherichia coli expression system for the rapid and economic expression of two rice APX genes, APXa and APXb (GeneBank accession Nos. D45423 and AB053297, respectively). The two genes were cloned into the pGEX-6p-3 vector to allow expression of APX as a glutathione-S-transferase (GST) fusion protein. The GST-APXa and GST-APXb fusion proteins were purified by affinity chromatography using a glutathione-Sepharose 4B column, with final yields of 40 and 73 mg g–1 dry cells, respectively. Specific activities were 15 and 20 mM ascorbate min–1 mg–1 protein, respectively. The Km values for ascorbate were 4 and 1 mM, respectively, and those for H2O2 were 0.3 and 0.7 mM, respectively indicating that the two rice isoenzymes have different properties.Revisions requested 27 September 2004; Revisions received 12 November 2004  相似文献

To quantify photosynthetic response of wheat to the combination of a fungal brown rust infection and a post-infection drought, four treatments were compared: no stress (control), fungal stress (FS), water stress (WS), and twofold stress (WS×FS). Predawn leaf water potential (wp) was similar in FS and WS treatments over a 3-week period. In the WS treatment, net photosynthetic rate (P N) and stomata CO2 conductance (g s) diminished concomitantly with a constant intercellular CO2 concentration (C i) close to 200 µmol mol–1. In the FS treatment, a reduction of P N occurred with an increase in respiration rate (doubling of the CO2 compensation concentration) and in C i but with no water loss modification. Healthy leaves of infected plants (FS) showed a reduction of P N as well, with constant g s and increased C i. In the twofold stress treatment (WS×FS), leaves showed reduced P N in relation to the lower wp. Deleterious effects of both drought and fungal infection on the final area of leaves and dry matter were additive.  相似文献
