Herbivore feeding elicits defense responses in infested plants, including the emission of volatile organic compounds that can serve as indirect defense signals. Until now, the contribution of plant tissue wounding during the feeding process in the elicitation of defense responses has not been clear. For example, in lima bean (Phaseolus lunatus), the composition of the volatiles induced by both the insect caterpillar Spodoptera littoralis and the snail Cepaea hortensis is very similar. Thus, a mechanical caterpillar, MecWorm, has been designed and used in this study, which very closely resembles the herbivore-caused tissue damage in terms of similar physical appearance and long-lasting wounding period on defined leaf areas. This mode of treatment was sufficient to induce the emission of a volatile organic compound blend qualitatively similar to that as known from real herbivore feeding, although there were significant quantitative differences for a number of compounds. Moreover, both the duration and the area that has been mechanically damaged contribute to the induction of the whole volatile response. Based on those two parameters, time and area, which can replace each other to some extent, a damage level can be defined. That damage level exhibits a close linear relationship with the accumulation of fatty acid-derived volatiles and monoterpenes, while other terpenoid volatiles and methyl salicylate respond in a nonlinear manner. The results strongly suggest that the impact of mechanical wounding on the induction of defense responses during herbivore feeding was until now underestimated. Controlled and reproducible mechanical damage that strongly resembles the insect's feeding process represents a valuable tool for analyzing the role of the various signals involved in the induction of plant defense reactions against herbivory.  相似文献

Plants are able to respond to herbivore damage with de novo biosynthesis of an herbivore-characteristic blend of volatiles. The signal transduction initiating volatile biosynthesis may involve the activation of the octadecanoid pathway, as exemplified by the transient increase of endogenous jasmonic acid (JA) in leaves of lima bean (Phaseolus lunatus) after treatment with the macromolecular elicitor cellulysin. Within this pathway lima bean possesses at least two different biologically active signals that trigger different biosynthetic activities. Early intermediates of the pathway, especially 12-oxo-phytodienoic acid (PDA), are able to induce the biosynthesis of the diterpenoid-derived 4,8, 12-trimethyltrideca-1,3,7,11-tetraene. High concentrations of PDA result in more complex patterns of additional volatiles. JA, the last compound in the sequence, lacks the ability to induce diterpenoid-derived compounds, but is highly effective at triggering the biosynthesis of other volatiles. The phytotoxin coronatine and amino acid conjugates of linolenic acid (e.g. linolenoyl-L-glutamine) mimic the action of PDA, but coronatine does not increase the level of endogenous JA. The structural analog of coronatine, the isoleucine conjugate of 1-oxo-indanoyl-4-carboxylic acid, effectively mimics the action of JA, but does not increase the level of endogenous JA. The differential induction of volatiles resembles previous findings on signal transduction in mechanically stimulated tendrils of Bryonia dioica.  相似文献

A comparative study on diurnal changes in metabolite levels associated with crassulacean acid metabolism (CAM) in the leaves of three CAM species, Ananas comosus (pineapple), a hexose-utilizing species, and Kalancho? daigremontiana and K. pinnata, two starch-utilizing species, were made. All three CAM species showed a typical feature of CAM with nocturnal malate increase. In the two Kalancho? species, isocitrate levels were higher than citrate levels; the reverse was the case in pineapple. In the two Kalancho? species, a small nocturnal citrate increase was found and K. daigremontiana showed a small nocturnal isocitrate increase. Glucose 6-phosphate (G-6-P), fructose 6-phosphate (F-6-P) and glucose 1-phosphate (G-1-P) levels in the three CAM species rose rapidly during the first part of the dark period and decreased during the latter part of the dark period. The levels of the metabolites also decreased during the first 3 h of the light period, then, remained little changed through the rest of the light period. Absolute levels of G-6-P, F-6-P and G-1-P were higher in pineapple than in the two Kalancho? species. Fructose 1,6-bisphosphate (F-1,6-P(2)) levels in the three CAM species increased during the dark period, then dramatically decreased during the first 3 h of the light period and remained unchanged through the rest of the light period. The extent of nocturnal F-1,6-P(2) increase was far greater in the two Kalancho? species than in pineapple. Absolute levels of F-1,6-P(2) were higher in the two Kalancho? species than in pineapple, especially during dark period. Diurnal changes in oxaloacetate (OAA), pyruvate (Pyr) and phosphoenolpyruvate (PEP) levels in the three CAM species were similar.  相似文献

We compared volatiles from lima bean leaves (Phaseolus lunatus) infested by either beet armyworm (Spodoptera exigua), common armyworm [Mythimna (Pseudaletia) separata], or two-spotted spider mite (Tetranychus urticae). We also analyzed volatiles from the leaves treated with jasmonic acid (JA) and/or methyl salicylate (MeSA). The volatiles induced by aqueous JA treatment were qualitatively and quantitatively similar to those induced by S. exigua or M. separata damage. Furthermore, both S. exigua and aqueous JA treatment induced the expression of the same basic PR genes. In contrast, gaseous MeSA treatment, and aqueous JA treatment followed by gaseous MeSA treatment, induced volatiles that was qualitatively and quantitatively more similar to the T. urticae-induced volatiles than those induced by aqueous JA treatment. In addition, T. urticae damage resulted in the expression of the acidic and basic PR genes that were induced by gaseous MeSA treatment and by aqueous JA treatment, respectively. Based on these data, we suggest that in lima bean leaves, the JA-related signaling pathway is involved in the production of caterpillar-induced volatiles, while both the SA-related signaling pathway and the JA-related signaling pathway are involved in the production of T. urticae-induced volatiles.  相似文献

Leaf-cutting ants (LCAs) are polyphagous, yet highly selective herbivores. The factors that govern their selection of food plants, however, remain poorly understood. We hypothesized that the induction of anti-herbivore defences by attacked food plants, which are toxic to either ants or their mutualistic fungus, should significantly affect the ants' foraging behaviour. To test this "induced defence hypothesis," we used lima bean (Phaseolus lunatus), a plant that emits many volatile organic compounds (VOCs) upon herbivore attack with known anti-fungal or ant-repellent effects. Our results provide three important insights into the foraging ecology of LCAs. First, leaf-cutting by Atta ants can induce plant defences: Lima bean plants that were repeatedly exposed to foraging workers of Atta colombica over a period of three days emitted significantly more VOCs than undamaged control plants. Second, the level to which a plant has induced its anti-herbivore defences can affect the LCAs' foraging behaviour: In dual choice bioassays, foragers discriminated control plants from plants that have been damaged mechanically or by LCAs 24 h ago. In contrast, strong induction levels of plants after treatment with the plant hormone jasmonic acid or three days of LCA feeding strongly repelled LCA foragers relative to undamaged control plants. Third, the LCA-specific mode of damaging leaves allows them to remove larger quantities of leaf material before being recognized by the plant: While leaf loss of approximately 15% due to a chewing herbivore (coccinelid beetle) was sufficient to significantly increase VOC emission levels after 24 h, the removal of even 20% of a plant's leaf area within 20 min by LCAs did not affect its VOC emission rate after 24 h. Taken together, our results support the "induced defence hypothesis" and provide first empirical evidence that the foraging behaviour of LCAs is affected by the induction of plant defence responses.  相似文献

Plants produce species-specific herbivore-induced plant volatiles (HIPVs) after damage. We tested the hypothesis that herbivore-specific HIPVs prime neighboring plants to induce defenses specific to the priming herbivore. Since Manduca sexta (specialist) and Heliothis virescens (generalist) herbivory induced unique HIPV profiles in Nicotiana benthamiana, we used these HIPVs to prime receiver plants for defense responses to simulated herbivory (mechanical wounding and herbivore regurgitant application). Jasmonic acid (JA) accumulations and emitted volatile profiles were monitored as representative defense responses since JA is the major plant hormone involved in wound and defense signaling and HIPVs have been implicated as signals in tritrophic interactions. Herbivore species-specific HIPVs primed neighboring plants, which produced 2 to 4 times more volatiles and JA after simulated herbivory when compared to similarly treated constitutive volatile-exposed plants. However, HIPV-exposed plants accumulated similar amounts of volatiles and JA independent of the combination of priming or challenging herbivore. Furthermore, volatile profiles emitted by primed plants depended only on the challenging herbivore species but not on the species-specific HIPV profile of damaged emitter plants. This suggests that feeding by either herbivore species primed neighboring plants for increased HIPV emissions specific to the subsequently attacking herbivore and is probably controlled by JA.  相似文献

By altering foliage quality, exposure to elevated levels of atmospheric CO(2) potentially affects the amount of herbivore damage experienced by plants. Here, we quantified foliar carbon (C) and nitrogen (N) content, C : N ratio, phenolic levels, specific leaf area (SLA) and the amount of leaf tissue damaged by chewing insects for 12 hardwood tree species grown in plots exposed to elevated CO(2) (ambient plus 200 microl l(-1)) using free-air CO(2) enrichment (FACE) over 3 yr. The effects of elevated CO(2) varied considerably by year and across species. Elevated CO(2) decreased herbivore damage across 12 species in 1 yr but had no detectable effect in others. Decreased damage may have been related to lower average foliar N concentration and SLA and increased C : N ratio and phenolic content for some species under elevated compared with ambient CO(2). It remains unclear how these changes in leaf properties affect herbivory. Damage to the leaves of hardwood trees by herbivorous insects may be reduced in the future as the concentration of CO(2) continues to increase, perhaps altering the trophic structure of forest ecosystems.  相似文献

Several plant species defend themselves indirectly from herbivores by producing herbivore-induced volatile compounds that attract the natural enemies of herbivores. Here we tested the effects of elevated atmospheric CO(2) (720 micromol mol(-1)) concentration on this indirect defense, physiological properties, and constitutive and induced emissions of white cabbage (Brassica oleracea ssp. capitata, cvs Lennox and Rinda). We monitored the orientation behavior of the generalist predator Podisus maculiventris (Heteroptera: Pentatomidae) and the specialist parasitoid Cotesia plutellae (Hymenoptera: Braconidae) to plants damaged by Plutella xylostella (Lepidoptera: Plutellidae) in the Y-tube olfactometer. Elevated CO(2) levels did not affect stomatal densities but reduced specific leaf area and increased leaf thickness in cv Lennox. In addition to enhanced constitutive monoterpene emission, P. xylostella-damaged cabbages emitted homoterpene (E)-4,8-dimethyl-1,3,7-nonatriene, sesquiterpene (E,E)-alpha-farnesene, and (Z)-3-hexenyl acetate. Growth at elevated CO(2) had no significant effect on the emissions expressed per leaf area, while minor reduction in the emission of homoterpene (E)-4,8-dimethyl-1,3,7-nonatriene and (E,E)-alpha-farnesene was observed at elevated CO(2) in one of two experiments. The generalist predator P. maculiventris discriminated only between the odors of intact and P. xylostella-damaged cv Rinda plants grown at ambient CO(2) concentration, preferring the odor of the damaged plants. The specialist parasitoid C. plutellae preferred the odor of damaged plants of both cultivars grown at ambient CO(2) but did not detect damaged cv Lennox plants grown at elevated CO(2). The results suggest that elevated atmospheric CO(2) concentration could weaken the plant response induced by insect herbivore feeding and thereby lead to a disturbance of signaling to the third trophic level.  相似文献

Nitrogen-fixing rhizobia can substantially influence plant–herbivore interactions by altering plant chemical composition and food quality. However, the effects of rhizobia on plant volatiles, which serve as indirect and direct defenses against arthropod herbivores and as signals in defense-associated plant–plant and within-plant signaling, are still unstudied. We measured the release of jasmonic acid (JA)-induced volatiles of rhizobia-colonized and rhizobia-free lima bean plants (Fabaceae: Phaseolus lunatus L.) and tested effects of their respective bouquets of volatile organic compounds (VOCs) on a specialist insect herbivore (Mexican bean beetle; Coccinellidae: Epilachna varivestis Mulsant) in olfactometer choice trials. In a further experiment, we showed that VOC induction by JA reflects the plant responses to mechanical wounding and insect herbivory. Following induction with JA, rhizobia-colonized plants released significantly higher amounts of the shikimic acid-derived compounds, whereas the emission of compounds produced via the octadecanoid, mevalonate and non-mevalonate pathways was reduced. These changes affected the choice behavior of beetles as the preference of non-induced plants was much more pronounced for plants that were colonized by rhizobia. We showed that indole likely represents the causing agent for the observed repellent effects of jasmonic acid-induced VOCs of rhizobia-colonized lima bean plants. Our study demonstrates a rhizobia-triggered efficacy of induced plant defense via volatiles. Due to these findings, we interpret rhizobia as an integral part of legume defenses against herbivores.  相似文献

Liriomyza sativae Blanchard and Liriomyza huidobrensis (Blanchard) (Diptera: Agromyzidae) are two invasive flies in China that have caused economical damage on vegetables and ornamental plants. In this article, we report the profiles of emitted volatiles from healthy, mechanically damaged, and leafminer-damaged bean, Phaseolus vulgaris L., plants. Among 25 emitted volatiles identified, (E)-2-hexen-1-al, (3E)-4,8-dimethyl-1,3,7-nonatriene (DMNT), (Z)-3-hexenyl acetate, (Z)-3-hexen-1-ol, (syn)- and (anti)-2-methylpropanal oxime, (syn)-2-methylbutanal oxime, linalool, and (E,E)-α-farnesene were consistently released from damaged bean plants. Combined amounts of these nine compounds made up more than 70% of the total volatiles emitted from each treatment. No qualitative differences in volatile emission were found between bean plants damaged by the two fly species; however, amounts of several major compounds induced by L. huidobrensis damage were significantly higher than those from plants damaged by L. sativae. The mechanically damaged plants released a higher proportion of green leaf volatiles than plants in the other treatments, whereas leafminer-damaged plants produced more terpenoids and oximes. Furthermore, the volatile profiles emitted from plants, damaged by adult leafminers, by second instar larvae, and even the plants with empty mines left by leafminer larvae (the pupal stage) were significantly different. Abstract.  The Indian meal moth Plodia interpunctella Hubner (Lepidoptera: Pyralidae) measures night length and enters diapause as a last-instar larva. To examine the role of photophase on dark-time measurement, the main LD 7 : 17 h photoperiod is disrupted by various lengths of darkness at 25 °C. When the light phase is not disrupted, the incidence of diapause is 76%. As the dark pulse disrupting a 7-h photophase becomes longer, the incidence of diapause decreases. To detect the dynamic kinetics of the time-measuring process, the main scotophase of 17 h is scanned by a 2-h light pulse. When the dark pulse in a 7-h photophase is fixed at 1 h after dawn and its duration is varied systematically from 1 to 3 h, or when the end of the dark pulse is fixed at 1 h before dusk, diapause is prevented completely by a 2-h light pulse inserted in the middle of 17-h darkness. These results are compared with those of a single night interruption of a 17-h scotophase with a 2-h light pulse but with an intact 7-h photophase. The disruption of a 7-h photophase by a dark pulse shifts the descending and ascending slopes of the response curve to some extent toward dawn and dusk, respectively, indicating that the dark pulse tends to shorten the critical length of dark time for diapause induction. When the main photophase (7 h) is interrupted by a 1-h dark pulse at 3–4 h after dawn, the 2-h scanning light pulse in the main scotophase (17 h) appears to act effectively as a dusk signal in the early scotophase. However, those in the mid- and late scotophase do not define the critical night length from dusk as sharply as for the critical night length from a 2-h light pulse to dawn. The results indicate the importance of photophase in the dark-time measurement.  相似文献

Diel courses of net CO2 exchange of leaves were studied in Clusia uvitana (Clusiaceae), a tropical Crassulacean acid metabolism (CAM) hemiepiphyte, growing in the crown of a 47-m tall kapok tree on Barro Colorado Island, Panama. Measurements on days without precipitation showed that net uptake of atmospheric CO2 occurred at night, a feature of CAM, as well as in the early morning and late afternoon. During 36 h of almost continuous rainfall, nocturnal net CO2 uptake was abolished and the diel pattern of net CO2 exchange became similar to that of a C3 plant. Exposing well-watered, potted plants of Clusia in the laboratory to temperatures and photosynthetic photon flux densities similar to those during the tropical rainstorm also abolished nocturnal net CO2 uptake. In contrast, Kalanchoe pinnata (Crassulaceae), an obligate CAM plant, still showed net CO2 dark fixation following the same low-light and moderate-temperature conditions, albeit at decreased rates. During these 12-h photoperiods, titratable acidity in Clusia increased slightly above its high level measured at the end of the previous dark period, whereas in Kalanchoe, the acid content decreased by about 40%. A survey among outer canopy leaves of Clusia on Barro Colorado Island showed that leaves that exhibited little or no nocturnal acidification maintained high levels of H+ at dawn and dusk. Progressively lower levels of H+ at dusk were accompanied by progressively higher nocturnal increases in H+. The data suggest that in C. uvitana the rapid switching between CAM- and C3-type carbon fixation that may occur within 24 h in response to environmental changes is controlled by the acidity status of the leaves in the light. Nocturnal CO2 fixation is enhanced by conditions that decrease the organic acid content during the light period.  相似文献

Transgenic Bt (expressing the cry1Ac endotoxin gene) and conventional oilseed rape plants grown in different soils were used to study nutrient uptake and emission of volatiles after herbivore damage. All plants were greenhouse-grown in soils representing low-, medium- and high-nutrient levels. The concentrations of N, P, K, Mg and Zn were significantly affected by the transgene, while the main effect of soil type appeared in N, P, Ca, Mg, B, Mn and Zn concentrations in the plants. Plants with four to five leaves were infested with the third instar larvae of Bt-susceptible Plutella xylostella for 48 h, and samples of volatiles were collected and analysed. In the first experiment, the soil nutrient level had a significant effect on the emissions of (Z)-3-hexen-1-ol, (Z)-3-hexenyl acetate, hexyl acetate, (E)-4,8-dimethyl-1,3,7-non-atriene (DMNT), beta-elemene, gamma-bisabolene, alpha-bisabolene and (E)-nerolidol. The induction of these volatiles was significantly higher in infested conventional plants grown at a high-soil nutrient level compared to infested conventional plants at a low-soil nutrient level. In the second experiment, the soil nutrient level had a significant effect on the emissions of (Z)-3-hexen-1-ol, (Z)-3-hexenyl acetate and beta-elemene and, again, this was significantly higher in infested conventional plants grown at high-soil nutrient levels in comparison with infested plants at a low-soil nutrient level. In both experiments, the transgene effect was significant on the emissions of DMNT and (E,E)-alpha-farnesene. The differences in emissions between the two separate experiments suggest that growth conditions (particularly daylength) and sampling procedure may affect the ratio of compounds detected in the emission blend, even though the response to herbivory, nutrient availability and the transgene were similar.  相似文献

In the present study, Y-tube olfactometric assays demonstrated that headspace volatile extracts collected from leaf miner-damaged, or artificially damaged, bean plants were more attractive to naive females of the parasitoid insect Opius dissitus than those collected from healthy plants. Headspace extracts from both Liriomyza huidobrensis and Liriomyza sativae second-instar larvae-damaged beans were analyzed by coupled gas chromatography-electroantennographic detection (GC-EAD). Of nine EAD-active volatiles identified, (3E)-4,8-dimethyl-1,3,7-nonatriene, (3Z)-hexenyl acetate, (syn)-2-methylpropanal oxime, and (syn)-2-methylbutanal oxime were the most abundant compounds that evoked significant electroantennogram (EAG) responses. Compounds (3Z)-hexen-1-ol, (anti)-2-methylbutanal oxime, linalool, beta-caryophyllene, and (3E,7E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene also elicited clear EAG responses but were present in smaller amounts. Choice experiments in a Y-tube olfactometer indicated that synthetic versions of (3Z)-hexen-1-ol, 2-methylpropanal oxime, 2-methylbutanal oxime, 3-methylbutanal oxime, linalool, (E,E)-alpha-farnesene, and (3E,7E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene were attractive individually, while (3Z)-hexenyl acetate and (3E)-4,8-dimethyl-1,3,7-nonatriene were unattractive at concentrations similar to those obtained from the headspace collection. Moreover, a blend of nine EAD-active volatiles was significantly more attractive relative to hexane (solvent). A mixture of oximes, tereponids, and green leaf volatiles may facilitate host location by female O. dissitus.  相似文献

为探讨菜豆对西花蓟马Frankliniella occidentalis取食的防御反应分子机制, 本实验利用荧光定量PCR(RT-qPCR)研究了西花蓟马取食、 机械损伤及外源水杨酸甲酯(MeSA)和茉莉酸(JA)处理对菜豆叶片防御相关基因(LOX, AOS, PAL及PR-2)相对表达量的影响。结果表明: 脂氧合酶基因(LOX)的相对表达量在外源JA和机械损伤诱导后分别在24 h和48 h时达到最大值, 但与对照相比差异不显著(P>0.05), 外源MeSA处理后LOX几乎没有表达, 西花蓟马取食后LOX的相对表达量在24 h显著升高, 约为对照的41.9倍, 且明显高于其余3个处理(P<0.05)。不同诱导处理后丙二烯氧化物合成酶基因(AOS)的相对表达量较低, 其中外源MeSA处理后在整个诱导时间内几乎没有表达。苯丙氨酸解氨酶基因(PAL)在机械损伤和外源JA处理后在整个诱导时间内几乎没有表达, 外源MeSA处理后PAL的表达量在24 h时最高, 为对照的1.9倍左右。PAL的相对表达量在遭受蓟马取食诱导后迅速升高, 在24 h时约为CK的4.3倍, 显著高于其他3种处理诱导的(P<0.05)。β-1, 3-葡聚糖酶基因(PR-2)的相对表达量在蓟马取食和外源JA处理的整个诱导时间内受到抑制, 机械损伤诱导后PR-2的表达水平在24 h时有所升高, 之后诱导期间内受到抑制。外源MeSA诱导后菜豆叶片中PR-2的相对表达量在24 h就急剧升高, 达到对照的6.63倍, 且显著高于其余3种处理诱导的(P<0.05), 但48 h后却几乎检测不到。研究结果提示, 西花蓟马取食不仅能够诱导SA和JA介导的信号传导途径, 且两通路间存在交互作用。  相似文献
