Two experiments were conducted to investigate the effect of inoculating Vicia faba plants (broad beens) raised in clean and oily sand with nodule-forming rhizobia and plant-growth-promoting rhizobacteria (PGPR) on growth of these plants in sand and to test whether this can improve the phytoremediation potential of this crop for oily desert areas. It was found that crude oil in sand at concentrations < 1.0% (w/w) enhanced the plant heights, their fresh and dry weights, the total nodule weights per plant, and the nitrogen contents of shoots and fruits. Similar enhancing effects were recorded when roots of the young plants were inoculated with nodule bacteria alone, PGPR alone, or a mixture of one strain of nodule bacteria and one of the PGPR. Such plant growth effects were associated with a better phytoremediation potential of V. faba plants for oily sand. The total numbers of oil-utilizing bacteria increased in the rhizosphere and more hydrocarbons were eliminated in sand close to the roots. The nodule bacteria tested were two strains of Rhizobium leguminosarum and the PGPR were Pseudomonas aeruginosa and Serratia liquefaciens. The four strains were found to use crude oil, n-octadecane, and phenanthrene as sole sources of carbon and energy. It was concluded that coinoculation of V. faba plant roots in oily sand with nodule bacteria and PGPR enhances the phytoremediation potential of this plant for oily desert sand through improving plant growth and nitrogen fixation.  相似文献

The rhizospheres and phyllospheres of peas, beans, tomatoes, and squash raised in a desert sand soil mixed with 0.5% crude oil were rich in oil-utilizing bacteria and accommodated large numbers of free-living diazotrophic bacteria, with potential for hydrocarbon utilization. According to their 16S rRNA-sequences, the cultivable oil-utilizing bacteria were affiliated with the following genera, arranged in decreasing frequency: Bacillus, Ochrobactrum, Enterobacter, Rhodococcus, Arthrobacter, Pontola, Nocardia, and Pseudoxanthomonas. Diazotrophic isolates were affiliated with Rhizobium, Bacillus, Rhodococcus, Leifsonia, Cellulosimicrobium, Stenotrophomonas, Kocuria, Arthrobacter, and Brevibacillus. The crude oil–utilizing and diazotrophic isolates grew, with varying growth intensities, on individual aliphatic (C8 to C40) and aromatic hydrocarbons, as sole sources of carbon and energy. Quantitative gas liquid chromatographic measurements showed that representative bacterial isolates eliminated pure n-hexadecane, n-decosane, phenanthrene, and crude oil from the surrounding liquid media. Cultivation of oily sand–soil samples with any of the four tested crops led to enhanced oil degradation in that soil, as compared with the degradation in uncultivated oily sand–soil samples.  相似文献

The objective of this study is to determine the distribution factors and enrichment coefficients between soil and plant parts by studying the accumulation and distribution of selenium (Se) in the roots and shoots of different plants. The plants (9 samples of Euphorbia macroclada, 5 samples of Verbascum cheiranthifolium, 8 samples of Astragalus gummifer) and their associated soil samples were collected from the Keban mining area. The roots and the shoots of these plants, together with the associated soils, were analyzed by ICP-MS. The mean Se value of the contaminated surface soils was found to be two to five times higher than those of previously studied uncontaminated surface soils. Se concentrations of the plant parts were lower than those in their associated alkaline soils, where the plants were grown, except for in the shoots of A. gummifer. Mean Se concentrations in the roots of E. macroclada, V. cheiranthifolium, and A. gummifer were 0.82, 0.22, and 0.47 mg kg?1 on a dry weight basis, respectively, while Se concentrations were 0.29, 0.26, and 2.66 mg kg?1 in the shoots on a dry weight basis, respectively. The enrichment coefficients and the distribution factors of those plants were lower than 1, except for the distribution factors of V. cheiranthifolium and A. gummifer plants. Thus, it appeared that the shoots of these plants could make them efficient bioaccumulator plants for Se because of high distribution factors and enrichment coefficients. Due to such factors, they can also be used to clean or rehabilitate soils and areas contaminated with Se.  相似文献

A plant–bacterium association of the cardon cactus (Pachycereus pringlei) and endophytic bacteria promotes establishment of seedlings and growth on igneous rocks without soil. These bacteria weather several rock types and minerals, unbind significant amounts of useful minerals for plants from the rocks, fix in vitro N2, produce volatile and non-volatile organic acids, and reduce rock particle size to form mineral soil. This study revealed the presence of large populations of culturable endophytic bacteria inside the seeds extracted from wild plants, from seeds extracted from the guano of bats feeding on cactus fruit, in seedlings growing from these seeds, in the pulp of fruit, and in small, mature wild plants, and are comparable in size to populations of endophytic populations in some agricultural crops. The dominant culturable endophytes were isolates of the genera Bacillus spp., Klebsiella spp., Staphylococcus spp., and Pseudomonas spp. Based on partial sequencing of the 16s rRNA gene, the isolated strains had low similarity to known strains in these genera. However, these strains have higher molecular similarity among endophytes obtained from seeds, endophytes from roots, and some bacterial strains from the rhizoplane. Seedlings developed from seeds with endophytes contain the similar species of endophytes in their shoots, possibly derived from the seeds. This study shows the involvement of endophytic bacteria in rock weathering by cacti in a hot, subtropical desert and their possible contribution to primary colonization of barren rock. This study proposes that cacti capable of acquiring diverse populations of endophytes may give them an evolutionary advantage to gain a foothold on highly uncompromising terrain.  相似文献

Fresh leaves of American grass and broad beans grown in pristine soil were naturally colonized with cultivable volatile oil hydrocarbon-utilizing bacteria, whose numbers increased significantly in plants grown in oily soil. According to their 16S rRNA gene sequences those bacteria were affiliated to various species of the genera Rhodococcus and Pseudomonas. Qualitative growth studies revealed that pure cultures of these phyllospheric bacteria could grow successfully on a solid mineral medium containing individual alkanes with chain lengths of C9 through C40 and the aromatics phenanthrene, naphthalene, and biphenyl as sole sources of carbon and energy. Quantitative measurements showed that the individual pure bacterial isolates degraded between about 20 and 30% of crude oil, n-hexadecane, or phenanthrene in batch culture after a one-week incubation. These results reflect the high hydrocarbon degradation potential of those bacteria. The isolates were diazotrophic (nitrogen fixers), meaning that they were self-dependent in covering their nitrogen requirements. Incubating fresh leaves in closed microcosms containing volatile oil hydrocarbons resulted in up to more than 80% attenuation of these compounds after two weeks. Experimental evidence was provided that the leaf tissues did not contribute to this attenuation, which was exclusively due to the bacterial activity.  相似文献

The rhizosphere of two turf cover sorts; Bermuda grass and American grass contained high numbers, 8.1 to 16.8 x 10(6) g(-1) of cultivable oil-utilizing and diazotrophic bacteria belonging predominantly to the genera Agrobacterium, Arthrobacter, Pseudomonas, Gordonia, and Rhodococcus. Those bacteria also grew on a nitrogen-free medium and demonstrated the ability to reduce acetylene to ethylene. These isolates grew on a wide range of n-alkanes (C9 to C40) and aromatic hydrocarbons, as sole sources of carbon. Quantitative determinations revealed that predominant bacteria consumed crude oil and representative aliphatic (n-octadecane) and aromatic (phenanthrene) hydrocarbons efficiently. The fact that those organisms had the combined activities of hydrocarbon-utilization and nitrogen-fixation makes them suitable tools for bioremediating oily desert areas that are normally poor in nitrogenous compounds. Phytoremediation experiments showed that spreading turf cover on oily desert soil inhibited oil volatilization and enhanced oil loss in soil by about 15%. Oil loss was also enhanced in turf free soil samples fertilized with NH4NO3. In conclusion, covering this oil-polluted soil with turf cover minimized atmospheric pollution, increased the numbers of the oil-utilizing/nitrogen-fixing bacteria by about 20 to 46% thus, encouraging oil attenuation.  相似文献

The coding region of the 2S albumin gene of Brazil nut (Bertholletia excelsa H.B.K.) was completely synthesized, placed under control of the cauliflower mosaic virus (CaMV) 35S promoter and inserted into the binary vector plasmid pGSGLUC1, thus giving rise to pGSGLUC1-2S. This was used for transformation of tobacco (Nicotiana tabacum L. cv. Petit Havanna) and of the grain legume Vicia narbonensis L., mediated by the supervirulent Agrobacterium tumefaciens strain EHA 101. Putative transformants were selected by screening for neomycin phosphotransferase (NPT II) and -glucuronidase (GUS) activities. Transgenic plants were grown until flowering and fruiting occurred. The presence of the foreign gene was confirmed by Southern analysis. GUS activity was found in all organs of the regenerated transgenic tobacco and legume plants, including the seeds. In the legume, the highest expression levels of the CaMV 35S promoter-controlled 2S albumin gene were observed in leaves and roots. 2S albumin was localized in the vacuoles of leaf mesophyll cells of transgenic tobacco. The Brazil nut protein was present in the 2S fraction after gel filtration chromatography of the legume seed proteins and could be clearly identified by immunoblotting. Analysis of seeds from the R2 progenies of the legume and of transgenic tobacco plants revealed Mendelian inheritance of the foreign gene. Agrobacterium rhizogenes strain RifR 15834 harbouring the binary vector pGSGLUCl2S was also used to transform Pisum sativum L. and Vicia faba L. Hairy roots expressed the 2S albumin-specific gene. Several shoots were raised but they never completely rooted and no fertile plants were obtained from these transformants.  相似文献

Nowadays, public concern relating to ecological deleterious effects of heavy metals is on the rise. To evaluate the potential of Rapistrum rugosum and Sinapis arvensis in lead- contaminate phytoremediate, a pot culture experiment was conducted. The pots were filled by soil treated with different rates of leadoxide (PbO) including 0 (control), 100, 200, 300, 400, and 500 mg Pb per 1 kg soil. Germinated seeds were sown. Surprisingly, with increasing concentration of Pb, dry weight of R. rugosum and S. arvensis did not decrease significantly. In both of species, the concentration of Pb was higher in roots than shoots. In general, S.arvensis was absorbed more Pb compared to R. rugosum. The results revealed high potential of R. rugosum and S. arvensis in withdrawing Pb from contaminated soil. For both species, a positive linear relation was observed between Pb concentration in soil and roots. However, linear relationship was not observed between Pb concentration in the soil and shoots. Although both species test had low ability in translocation Pb from roots to shoots but they showed high ability in uptake soil Pb by roots. Apparently, these plants are proper species for using in phytoremediation technology.  相似文献

The pp150 gene of human cytomegalovirus (HCMV) was transferred into Vicia faba plants by Agrobacterium tumefaciens-mediated transformation. Three of five hygromycin resistant V. faba plants were identified as positive by PCR and dot-blot hybridization. The ELISA results indicated that pp150 protein from three plants of transformed V. faba leaves and seeds made up 0.005–0.015% of the total soluble protein. The results of detection by immunoblot and inhibition of immunofluorescent assay (IFA) showed that pp150 soluble protein had immunity activity. HCMV pp150-specific antibody (IgG, IgA) and IFN-γ producing T cells were detected in 100% of the mice immunized with pp150 transgenic V. faba seeds by ELISA and intracellular staining and flow cytometry analysis, respectively. The transgenic V. faba plants will provide new material for the development of edible vaccination against HCMV infection.  相似文献

Plant regeneration of Prunus salicina (Japanese plum) using mature seeds was studied and evaluated. Shoots were effectively induced from hypocotyl slices of mature seeds on media containing cytokinins. Among three plant growth regulators evaluated, thidiazuron (TDZ) was the most effective for shoot induction overall. Shoots were also induced using 6-benzylaminopurine (BA), but the effectiveness was reduced at low concentrations. Low regeneration was induced using kinetin. Three plum varieties were evaluated and the regeneration appeared to be genotype dependent. Induced shoots elongated, roots formed, and plantlets developed upon transfer of the shoots to the rooting medium. Primary shoots, when sub-cultured on fresh induction medium, produced multiple shoots, and such multiplication could continue for more cycles. The plantlets were transferred to soil, and the full plants were readily recovered in a greenhouse. The regeneration process was relatively fast as plants could be recovered in 4 to 5 mo. after the culture initiation.  相似文献

This paper describes multiple shoot regeneration from leaf and nodal segments of a medicinally important herb Centella asiatica L. on Murashige and Skoog’s (MS) medium supplemented with a range of growth regulators. The highest number of multiple shoots was observed on MS augmented with 3.0 mg dm−3 N6-benzylaminopurine (BAP) and 0.05 mg dm−3 α-naphthaleneacetic acid (NAA). Leaf explant showed maximum percentage of cultures regenerating shoots (81.6 %), with the highest shoot number (8.3 shoots per explant) and the shoot length (2.1 cm) whereas, nodal explant showed less number of shoots with callus formation at the base cut end. Successive shoot cultures were established by repeatedly sub-culturing the original explant on a fresh medium. Rooting of in vitro raised shoots was best induced on half strength MS supplemented with 0.5 mg dm−3 indole-3-butyric acid (IBA) with highest percentage of shoot regenerating roots (76.8 %) with 3–4 roots per shoot. Plantlets were acclimated in Vermi-compost and eventually established in soil. Contents of chlorophyll, total sugars, reducing sugars and proteins were estimated in leaf tissue from both in vivo and in vitro raised plants. Chlorophyll content was higher in in vivo plants, whereas other three components were higher in in vitro plants.  相似文献

In this study, the ability of Lysinibacillus sphaericus to degrade aromatic hydrocarbons as well as complex hydrocarbon mixtures, such as diesel oil and oily sludge, was evaluated. L. sphaericus was able to grow when toluene, naphthalene, or phenanthrene were used as a sole carbon source in minimal salt medium. Removal efficiencies of up to 95% were found for C10-C28 hydrocarbons in the biodegradation assays of diesel oil. The biodegradation of oily sludge was evaluated in landfarming-like experiments in the open air and in completely covered containers in the field. After 50 days of treatment, the removal efficiency of total petroleum hydrocarbons in open-air and closed assays was of 84.1% and 60.1%, respectively. Furthermore, L. sphaericus was able to degrade volatile hydrocarbons (benzene, toluene, ethylbenzene, and phenol) in the headspace of closed containers, preventing the emission of these compounds to the atmosphere. L. sphaericus was herein proposed as a promising candidate to be used in bioremediation strategies of petroleum hydrocarbons.  相似文献

Penetration of very strong soils by seedling roots of different plant species   总被引:19,自引:2,他引:17  
The abilities of seedling roots of twenty-two plant species to penetrate a strong growth medium were compared under controlled conditions. Seedlings were grown for 10 days in compression chambers filled with siliceous sandy soil at 0.2 kg kg–1 water content and mean penetrometer resistance of 4.2 MPa. Root elongation and thickening were measured after growth. The results show that soil strength reduced the elongation of roots of all plant species by over 90% and caused the diameters of the roots to increase compared with control plants grown in vermiculite (0 MPa resistance).Differences in both root elongation and root diameter were observed among plant species. Generally, the roots of dicotyledons (with large diameters) penetrated the strong medium more than graminaceous monocotyledons (with smaller diameters). There was a significant positive correlation (r=0.78, p<0.05) between root diameter and elongation over all the species in the stressed plants. The species were ranked according to the relative root elongation and relative root thickening. Based on this ranking, lupin (Lupinus angustifolius), medic (Medicago scutelata) and faba bean (Vicia faba) were the species with the greatest thickening and elongation while wheat (Triticum aestivum), rhodesgrass (Chloris gayana) and barley (Hordeum vulgare) had the least. The weight of the seeds did not seem to influence either the thickening or elongation of the roots.  相似文献

As nitrogen is known to be a limiting factor for plant growth, we were interested in the relationship between soil microbial activity and the nitrogen assimilation of 5 different halophytes from 4 saline sites near the lake “Neusiedlersee”, Austria. The following were studied between May and October 1985: nitrogen fixation (15N2 and acetylene reduction): N-mineralization; several soil characteristics and in vivo nitrate reductase activity of roots and shoots of these plants. NO?3, org. N- and carboxylate contents of both roots and shoots, as well as the effect of NO?3-fertilization on the amounts of these substances, were determined on plants growing in the field during a 3-day period in September 1985. Fertilization led to a decrease in acetylene reduction activity at most sites, and an increase in the nitrate reductase activity of the shoots of all plants. Overall, carboxylate and organic nitrogen contents of these halophytes did not change in response to fertilization. Only in the roots of Aster tripolium and Atriplex hastata was there a marked increase in the nitrate reductase activity in response to fertilization. Species growing at the same site, such as Plantago maritima and Lepidium crassifolium showed contrasting levels of assimilatory activity. Apparent low rates of ammonification and nitrification were detected in soils from the 4 sites. The results are discussed in relation to the nitrogen and carbon economies of the microorganisms and plants.  相似文献

Single-node stem segments of Rosmarinus officinalis L. var. genuina forma erectus proved better explants than shoot tips (ca. 2 cm long) for extablishment of field-grown plants in aseptic cultures. Benzylaminopurine was far more effective than kinetin for shoot induction in shoot tips excised from aseptically-grown plants. Maximum numbers of shoot buds (ca. 14) were formed per explant at 0.2 mgl-1 benzylaminopurine in 30 days. After further growth of isolated shoots and treatment with 0.25 mgl-1 indolepropionic acid for 7 days, 80% shoots produced roots. In vitro raised plantlets were successfully grown in soil to plants. Abstract

The present work aims to investigate the attractive ability of the newly isolated bacterium Serratia plymuthica BMA1, to release phosphorus (P) from rock phosphate (RP) and also to assess its beneficial effect in promoting the growth of Vicia faba. This strain exhibited the highest RP-solubilization activity ever reported, with 450?mg l?1 of soluble P at 30?°C. At 10 and 20?°C, its RP-solubilization was estimated at 100 and 200?mg l?1, respectively. HPLC analysis revealed that RP-solubilizing activity was associated with the release of gluconic acid. The hydroxyapatite (HA) solubilization activity concomitantly occurred with the secretion of gluconic acid and lactic acid. Under greenhouse conditions, application of BMA1 strain as an inoculant in presence of RP as the sole P source, considerably increased phosphorus uptake by V. faba L. and upgraded its overall growth in terms of dry weight and length by 76% and 39%, respectively. This growth promoting effect was accompanied by a substantial increase in chlorophyll contents. Additionally, phosphorus levels within roots and shoots of S. plymuthica BMA1-treated plants were approximately three times greater, compared to the non-inoculated control plants. When HA was used instead of RP, bacterization with BMA1 strain also enhanced the plant growth parameters and P contents, but significantly less than RP. These findings revealed that S. plymuthica BMA1 could be a potential candidate to improve the agronomic effectiveness of RP, toward a clean P-nutrition through the formulation of bio-phosphate fertilizers for plant growth promotion.  相似文献

The aim of this study was to assess the impact of soil inoculation with the Rhodococcus erythropolis CD 106 strain on the effectiveness of the phytoremediation of an aged hydrocarbon-contaminated [approx. 1% total petroleum hydrocarbon (TPH)] soil using ryegrass (Lolium perenne). The introduction of CD 106 into the soil significantly increased the biomass of ryegrass and the removal of hydrocarbons in planted soil. The fresh weight of the shoots and roots of plants inoculated with CD 106 increased by 49% and 30%, respectively. After 210 days of the experiment, the concentration of TPH was reduced by 31.2%, whereas in the planted, non-inoculated soil, it was reduced by 16.8%. By contrast, the concentration of petroleum hydrocarbon decreased by 18.7% in non-planted soil bioaugmented with the CD 106 strain. The rifampicin-resistant CD 106 strain survived after inoculation into soil and was detected in the soil during the entire experimental period, but the number of CD 106 cells decreased constantly during the enhanced phytoremediation and bioaugmentation experiments.

The plant growth-promoting and hydrocarbon-degrading properties of CD 106, which are connected with its long-term survival and limited impact on autochthonous microflora, make this strain a good candidate for improving the phytoremediation efficiency of soil contaminated with hydrocarbons.  相似文献

外来入侵植物可以通过淋溶、自然挥发、根系分泌和植株凋落物分解等途径向周围环境释放化感物质,抑制伴生植物的生长、发育。该研究以不同浓度紫茎泽兰(Eupatorium adenophorum)叶片水浸液处理蚕豆(Vicia faba)种子,研究紫茎泽兰叶片水浸液对蚕豆根尖细胞微核、染色体畸变、细胞凋亡、蚕豆幼苗叶片叶绿素和N含量、光合生理特性、生物量的影响。结果表明:(1)紫茎泽兰叶片水浸液处理显著抑制蚕豆根尖的伸长和细胞的有丝分裂,并诱导蚕豆根尖细胞染色体畸变和细胞微核的产生,有丝分裂指数随着叶片水浸液浓度增加而减小,根尖细胞微核率随叶片水浸液浓度增加而增大,高浓度叶片水浸液处理对蚕豆根尖细胞的凋亡及坏死有明显影响。(2)紫茎泽兰叶片水浸液处理引起蚕豆幼苗叶片的叶绿素和N含量显著降低,并导致蚕豆幼苗最大净光合速率和生物量的显著下降。总之,紫茎泽兰叶片水浸液可能引起蚕豆根尖的氧化损伤和抑制根尖的伸长,且叶片水浸液的抑制作用呈现一定的剂量效应。紫茎泽兰叶片水浸液对蚕豆根尖的损伤和抑制作用可能影响了植株对氮素的吸收,进而对蚕豆幼苗光合生理表现以及生物量积累产生显著负面效应。  相似文献

Phytoremediation is a promising approach for the cleanup of soil contaminated with petroleum hydrocarbons. This study aimed to develop plant-bacterial synergism for the successful remediation of crude oil-contaminated soil. A consortia of three endophytic bacteria was augmented to two grasses, Leptochloa fusca and Brachiaria mutica, grown in oil-contaminated soil (46.8 g oil kg?1 soil) in the vicinity of an oil exploration and production company. Endophytes augmentation improved plant growth, crude oil degradation, and soil health. Maximum oil degradation (80%) was achieved with B. mutica plants augmented with the endophytes and it was significantly (P < 0.05) higher than the use of plants or bacteria individually. Moreover, endophytes showed more persistence, the abundance and expression of alkB gene in the rhizosphere as well as in the endosphere of the tested plants than in unvegetated soil. A positive relationship (r = 0.70) observed between gene expression and crude oil reduction indicates that catabolic gene expression is important for hydrocarbon mineralization. This investigation showed that the use of endophytes with appropriate plant is an effective strategy for the cleanup of oil-contaminated soil under field conditions.  相似文献

The aim of this study was to degrade total petroleum hydrocarbon (TPH) in a petroleum sludge contaminated site (initial TPH concentration of 65,000–75,000 mg.kg–1) with two native sedge species namely Cyperus rotundus (Linn.) and Cyperus brevifolius (Rottb.) Hassk. Fertilized and unfertilized treatments were maintained separately to record the influence of fertilizer in TPH degradation. The average biomass production (twenty plants from each treatment) of C. rotundus was 345.5 g and that of C. brevifolius was 250.6 g in fertilized soil during 360 days. Decrease in soil TPH concentration was higher in fertilized soil (75% for C. rotundus and 64% for C. brevifolius) than in unfertilized soil (36% for C. rotundus and 32% for C. brevifolius). In unvegetated treatments, decrease in soil TPH concentration in fertilized (12%) and unfertilized soil (8%) can be attributed to natural attenuation and microbial degradation. TPH accumulation in roots and shoots was significantly higher in fertilized soil in comparison to unfertilized soils (p < 0.05). Most probable number (MPN) in planted treatments was significantly higher than in unplanted treatments (p < 0.05).  相似文献
