Kinetics of ethanol inhibition in alcohol fermentation   总被引:3,自引:0,他引:3  
The inhibitory effect of ethanol on yeast growth and fermentation has been studied for the strain Saccharomyces cerevisiae ATCC No. 4126 under anaerobic batch conditions. The results obtained reveal that there is no striking difference between the response of growth and ethanol fermentation. The ethanol-producing capability of the cells was completely inhibited at 115 g/L ethanol. There have been few studies quantifying litterfall, standing litterstock and gross litter decomposition following forest conversion to plantation crops such as cocoa. Additionally, an assessment of changing processes occurring in forest floor litter systems with plantation age is lacking. We investigated litterfall production, standing litter changes and litter decomposition along a chronosequence of shaded cocoa farm fields (secondary forest, 3, 15 and 30-year-old) in the moist semi-deciduous forest belt in the Ashanti Region of Ghana in West Africa over 24 months. Mean annual litterfall production differed significantly among study sites and ranged from 5.0 to 10.4 Mg DM ha?1. Similarly, standing litter differed significantly between land-use /plot ages. The results showed significant differences in quality between litter from forest and litter from cocoa plantations. Litterfall from forests had higher concentrations of nitrogen and lower concentration of soluble polyphenols and lignin compared to litter from cocoa systems. Annual decomposition coefficients (k L ) were similar in cocoa systems (0.221–0.227) but higher under secondary forests (0.354). Correlations between litter quality parameters and the decomposition coefficient showed nitrogen and lignin concentrations as well as ratios that include nitrogen are the best predictors of decomposition for the litters studied. Our results confirm the hypothesis that decomposition decreases following forest conversion to shaded cocoa systems because of litter quality changes and that decomposition rates correlate to litter quality differences between forest and cocoa ecosystems. The study also showed that standing litter pools and litterfall production in recently converted cocoa plantations are low compared to secondary forests or mature cocoa systems. Management strategies involving the introduction of upper canopy species during plantation development with corresponding replacement of tree mortality with diverse fast growing species will provide high quality and quantity litter resources.  相似文献

Summary The use of several S1 individuals to represent an S0 individual permits the use of a Design II mating scheme for plants with only one pistillate flower per plant. Estimates of additive (V A ) and dominance (V D ) variance from this mating scheme will be biased upwards, when a small number (10) of individuals of each S1 line are used. This bias can be computed, and the additive and dominance estimates can be corrected. Of particular interest is the observation that the additive genetic variance contributes to bias in estimates of V D . When S0 plants are non inbred and their selfedprogeny (S1 lines) are used to represent them in developing families for use in the Design II, where m1 is the number of individuals used to represent an S1 line in developing half sib-families and m2 is the number of individuals used to represent the S1 line in making up full sib-families. For example, in a 3×3 Design II, with about 10 individuals used to represent each S1 line in each cross, m2 = 10 and m1 = 30. In T-10 experiments, attempts were made to significantly exceed the Greenwald limit $\bar n_{Gr} $ during high-power (P ab=750 kW) electron-cyclotron resonance heating (ECRH) and gas puffing. Formally, the density limit $(\bar n_e )_{\lim } $ exceeding the Greenwald limit $({{(\bar n_e )_{\lim } } \mathord{\left/ {\vphantom {{(\bar n_e )_{\lim } } {\bar n_{Gr} }}} \right. \kern-0em} {\bar n_{Gr} }} = 1.8)$ was achieved for q L=8.2. However, as q L decreased, the ratio ${{(\bar n_e )_{\lim } } \mathord{\left/ {\vphantom {{(\bar n_e )_{\lim } } {\bar n_{Gr} }}} \right. \kern-0em} {\bar n_{Gr} }}$ also decreased, approaching unity at q L≈3. It was suggested that the “current radius” (i.e., the radius of the magnetic surface enclosing the bulk of the plasma current I p), rather than the limiter radius, was the parameter governing the value of $(\bar n_e )_{\lim } $ . In the ECRH experiments, no substantial degradation of plasma confinement was observed up to $\bar n_e \sim 0.9(\bar n_e )_{\lim } $ regardless of the ratio ${{(\bar n_e )_{\lim } } \mathord{\left/ {\vphantom {{(\bar n_e )_{\lim } } {\bar n_{Gr} }}} \right. \kern-0em} {\bar n_{Gr} }}$ . The objective of this study was to evaluate whether lead (Pb) and arsenic (As) levels in biological fluids were associated to the body composition in a group of reproductive-age women. Voluntary childbearing-age women (n = 107) were divided into three groups according to their body mass index (BMI: weight/height2 (kg/m2): low weight (BMI<18.5 kg/m2), normal $ \left( {{\text{BMI}} > 19\kern1.5pt<\kern1.5pt24.9\,{{\text{kg}} \mathord{\left/{\vphantom {{\text{kg}} {{{\text{m}}^{\text{2}}}}}} \right.} {{{\text{m}}^{\text{2}}}}}} \right) $ \left( {{\text{BMI}} > 19\kern1.5pt<\kern1.5pt24.9\,{{\text{kg}} \mathord{\left/{\vphantom {{\text{kg}} {{{\text{m}}^{\text{2}}}}}} \right.} {{{\text{m}}^{\text{2}}}}}} \right) , and overweight (BMI>25 kg/m2). Body composition and fat mass percentage were determined by the isotopic dilution method utilizing deuterated water. Blood lead concentrations were determined by graphite furnace atomic absorption spectrometry and urinary arsenic (AsU) concentrations by inductively coupled plasma mass spectrometry. The type and frequency of food consumption and lifestyle-related factors were also registered. Most women had $ {\text{PbB}}\,{\text{levels}} > 2\kern1.5pt<\kern1.5pt10\,{\mu{{\text{ g}}} \mathord{\left/{\vphantom {\mu{{\text{ g}}} {\text{dL}}}} \right.} {\text{dL}}} $ {\text{PbB}}\,{\text{levels}} > 2\kern1.5pt<\kern1.5pt10\,{\mu{{\text{ g}}} \mathord{\left/{\vphantom {\mu{{\text{ g}}} {\text{dL}}}} \right.} {\text{dL}}} , and only 2.6% had AsU concentrations above 50 μg/L. Four intense and separate localized surface plasmon resonance (LSPR) absorption peaks have been obtained in the gold-dielectric–gold–silver multilayer nanoshells. The silver coating on the gold shell results in a new LSPR peak at about 400 nm corresponding to the $ {{\left| {\omega_{+}^{-}} \right\rangle}_{Ag }} $ mode. The intense local electric field concentrated in the silver shell at the wavelength of 400 nm indicates that this new plasmonic band is coming from the symmetric coupling between the antibonding silver shell plasmon mode and the inner sphere plasmon. Increasing the silver shell thickness also leads to the intensity increasing of the $ {{\left| {\omega_{+}^{-}} \right\rangle}_{Au }} $ mode and blue shift of $ \left| {\omega_{-}^{+}} \right\rangle $ and $ \left| {\omega_{-}^{-}} \right\rangle $ modes. Therefore, quadruple intense plasmonic resonances in the visible region could be achieved in gold-dielectric–gold–silver multilayer nanoshells by tuning the geometrical parameters. And the quadruple intense plasmonic resonances in the visible region provide well potential for multiplex biosensing based on LSPR.  相似文献

The ion current to a spherical probe is considered with allowance for volume ionization, ion-neutral collisions, and the ion orbital moment. Experimental kinetic data (initial rate and high conversion) on the hydrolysis of cellobiose by 1,4-β-glucosidace (Gliocladium sp.) have been analysed and a competitive inhibition by glucose has been proposed. The determination of kinetic parameters from integral data is based upon algorithms for non-linear optimization and numerical integration. The values of kinetic constants \documentclass{article}\pagestyle{empty}\begin{document}$(v_{\max } = 1.02\frac{{\mu {\rm M}_{{\rm glucose}} }}{{{\rm mg}_{{\rm protein}} \cdot \min }},K_M = 2.6{\rm mM/l, and }K_P = 1.2{\rm mM/l)}$\end{document} agree well with the initialrate results. An important distinction is the confidence limit of parameters. Linear regression analysis shows a virtual accuracy and can lead to wrong conclusions.  相似文献

The data processing method of the turbidimetric bioassay of nisin was modified to facilitate its industrial application. The influence of the initial indicator concentration was minimized by a redefined specific dose of the bacteriocin as the quotient between the titer of the added bacteriocin and the initial population density of the indicator in the suspension. It was found that d c = 0.125 μg ml−1 was the critical dose of nisin that can cause a complete inhibition of the indicator, Pediococcus acidilactici UL5, with an initial OD of 0.135. To eliminate the interference of the cell debris, an equation, , exploiting d c, was formulated to obtain the intrinsic survival proportion. The use of the specific dose of the bacteriocin and the intrinsic survival proportion as parameters of the dose/response curve greatly enhanced its repeatability and feasibility. A dual-dosage approach was developed to further simplify the conventional standard dose/response curve method.  相似文献

Genetic parameters for growth, stem straightness, pilodyn penetration, relative bark thickness and survival were estimated in a base-population of five open-pollinated provenance/progeny trials of Eucalyptus viminalis. The trials, located in northern, central and southern Buenos Aires Province, Argentina, comprised 148 open-pollinated families from 13 Australian native provenances and eight local Argentinean seedlots. The Australian native provenances come from a limited range of the natural distribution. Overall survival, based on the latest assessment of each trial, was 62.4%. Single-site analyses showed that statistically significant provenances differences (p?<?0.05) for at least one of the studied traits in three out of the five trials analyzed. The local land race performed inconsistently in this study. The average narrow-sense individual-tree heritability estimate $ \left( {{{\hat{h}}^2}} \right) $ was 0.27 for diameter and 0.17 for total height. Values of $ {\hat{h}^2} $ also increased with age. Pilodyn penetration, assessed at only one site, was more heritable $ \left( {{{\hat{h}}^2} = 0.32} \right) $ than the average of growth traits. Estimated individual-tree heritabilities were moderate to low for stem straightness (average of 0.20) and relative bark thickness (0.16). The estimated additive genetic correlations $ \left( {{{{r}}_{{A}}}} \right) $ between diameter and height were consistently high and positive ( $ {{r}_{^A}} $ average of 0.90). High additive genetic correlations were observed between growth variables and pilodyn penetration ( $ {{r}_{^A}} $ average of 0.58). Relative bark thickness showed a negative correlation with diameter $ \left( {{{{r}}_{^A}} = - 0.39} \right) $ and height $ \left( {{{{r}}_{^A}} = - 0.51} \right) $ . The average estimated additive genetic correlation between sites was high for diameter (0.67). The implications of all these parameter estimates for genetic improvement of E. viminalis in Argentina are discussed.  相似文献

Ten color transparencies were presented 30 times each to ten normal adults in response to changes in their occipital-parietal EEG's. By means of a feedback paradigm, detected alphas caused each slide to flash and stay on as long as alpha persisted, and then to turn off when alpha attenuated, according to four different contingency conditions. For half of the feedback trials, stimulus presentation depended on alpha detection in only one hemisphere and was not influenced by changes in the simultaneously recorded contralateral EEG. For the other feedback trials, stimulus presentation was bilaterally contingent. These contingency configurations were compared with sham feedback, a noncontingent condition during which slide presentation was controlled by a prerecorded tape. For both alpha and no-alpha, the ratio of mean duration over standard error was used as a quantification of the EEG response to visual stimulation. It was assumed that larger ratios indicated increased control of the EEG. Compared with the sham condition, all feedback contingencies produced greater ratios, and hence, improved control of the EEG. The highest ratios were obtained during unilateral feedback from the EEG in which the occurrence of alpha elicited the visual stimulus. The results show that contingency between a visual stimulus and the EEG is an important parameter with regard to experimental control of the EEG.  相似文献

Methanobacterium thermoautotrophicum was grown in continuous culture in a fermenter gassed with H2 and CO2 as sole carbon and energy sources, and in a medium which contained either NH4Cl or gaseous N2 as nitrogen source. The phosphate removal in small, completely mixed storage reservoirs (preimpoundment basins) mainly is a function of the production of biomass by the phytoplankton. The knowledge of the critical detention time of the water is the most important premise to the prediction. The analyses of the Mulliken charge transfer, natural bond orbital (NBO), atoms in molecules (AIM) theory and electron density shifts reveal that the nature of the ??-halogen bond interaction in the complexes of ClF, BrF and BrCl might partly be charge transfer from the delocalized ??-HOMO orbital of $ \left( {\hbox{BNN}} \right)_3^{+} $ to X1X2. This result suggests that the positive aromatic ring $ \left( {\hbox{BNN}} \right)_3^{+} $ might act as a ??-electron donor to form the ??-halogen bond.

The effects of these salts on the helix–coil transition of rA–rU and rI–rC helices were examined. Increasing the size of the hydrophobic substituents, R1, R2, and R3 lowers the degree of stabilization of the helical structure. The disproportionation reaction, 2rA–rU→rA–rU2 + rA occurs readily with salts II and III, especially when the substituents, R1, R2, and R3 are small, i.e., H or Me. Spermine is found to stabilize the rA–rU2 and rI–rC helices to approximately the same extent; however, large differences between the degree of stabilization of rA–rU2 and rI-rC helices are observed when the substituents R1, R2, and R3 are large hydrophobic groups. Similar results are also obtained for the spermidine series.
