比较了鱼类养殖前后 ,地下水中正磷酸盐 (o P)浓度、碱性磷酸酶活性 (APA)在不同大小颗粒之间的分布、溶解态APA对pH、温度、CuSO4、ZnSO4、EDTA 2Na与表面活性剂 (CTAB与TritonX 10 0 )的应答方式及其动力学特征。养鱼之后 ,玻璃缸水中碱性磷酸酶表现出明显较高的活性 ,且以溶解态为主要存在形式 ,这种效应与鱼类的品种有关 ,溶解态APA的最大反应速度 (Vmax)与米氏常数 (Km)均明显提高 ,最适温度与pH值以及对于Zn2 的应答方式亦发生明显改变 ,颗粒结合态APA甚微或不可监测 ,藻类极少 ,故非溶解态APA的主要贡献者。因此 ,鱼类饲养与溶解态APA之间具有密切联系。养鱼缸底颗粒的蒸馏水提取液与同缸水中的溶解态APA对pH值的应答方式十分接近 ,这一结果暗示养殖产生的固体废物是溶解态APA的来源之一。  相似文献

文中综述了灵芝的抗氧化清除自由基作用。灵芝对各种因素引起的脑、心脏、胰腺、肝脏、胃肠道、肾脏和其他重要脏器的脂质过氧化损伤具有明显的保护作用。灵芝可显著减少脂质过氧化产物丙二醛(MDA)的含量,增强抗氧化酶如超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)以及其他抗氧化酶的活性。稹灵芝对体外培养的巨噬细胞(小鼠)、胰岛细胞(小鼠)、大脑皮层细胞(大鼠)、嗜铬细胞瘤细胞(大鼠)、血管内皮细胞(大鼠、人)和皮肤角质细胞(人)的氧化损伤具有明显保护作用。灵芝在体内外对不同动物模型和细胞模型的抗氧化清除自由基作用可能与其免疫调节、抗肿瘤、降血压、降血糖、保肝、心血管保护和抗衰老作用的机制有关。  相似文献

白首乌对超氧阴离子自由基清除作用的研究   总被引:10,自引:2,他引:10  
为深入探讨耳叶牛皮消块根(白首乌)的药理作用,指导其保健食品的研究与开发,采用化学发光法测定了去皮白首乌及白首乌栓皮对超氧阴离子自由基(O-2)的清除作用。结果表明,两者均能直接清除O-2,其IC50分别为6969μg/ml和2298μg/ml,而且后者对O-2的清除能力比前者高出30多倍,并同时测定了赤何首乌、西洋参、灵芝、当归、黄芪、菊花和竹叶。结果表明,去皮白首乌清除O-2的能力显著低于赤何首乌和灵芝,与西洋参和当归相差不大,显著高于黄芪、菊花和竹叶。而白首乌栓皮清除O-2的能力却高于其中任何一种中药。  相似文献

研究了地木耳(Nostoc commune)、葛仙米(N.sphaerohies)和发菜(N.flagelliforme)3种念珠藻多糖对自由基的清除作用。结果表明,3种念珠藻多糖对超氧阴离子自由基和羟自由基具有很强的清除作用,且呈量效关系,地木耳清除超氧自由基作用最强,最高清除率达72.3%,葛仙米和发菜分别为46.7%、35.5%;发菜清除羟自由基效果最强,最高清除率达74.3%,葛仙米和地木耳清除率分别为49.0%、46.7%;3种念珠藻多糖对DPPH自由基的清除作用不显著。  相似文献

用荧光染料FIuo-3标记人肝癌细胞株H_(7402)细胞内游离钙,在粘附式细胞仪观察检测单个细胞内游离钙水平的动态变化,细胞在无钙环境中,直接溶解因子(DLF)刺激下细胞内游离钙迅速升高,达到峰值后下降;在细胞培养皿中加入1mmol/L CaCl_2,DLF使胞浆游离钙持续升高;加入10mmol/L CaCl_2,DLF刺激后胞浆游离钙水平无明显变化,表明DLF能引起胞内Ca~(2 )释放和胞外Ca~(2 )内流,细胞外高浓度Ca~(2 )能阻断DLF升高细胞内Ca~(2 )浓度的 作用。  相似文献

本文采用三种分光光度法,测定了中药地榆提取物不同极性溶剂萃取所得各部分对羟自由基和超氧阴离子自由基的清除作用,以BHT为参照.结果表明:中药地榆提取物各萃取部分除石油醚层外,其它各萃取部分对·OH自由基都有清除作用,且随着提取物浓度的增加,清除率逐渐增高;对O2-自由基没有明显的清除作用.其中乙酸乙酯萃取部分对·OH自由基的清除作用最强.  相似文献

The radical scavenging activity of β,β-dimethylacrylalkannin derivatives has been studied by using density functional theory. The hydrogen bond property of the studied structures was investigated using the atoms in molecules theory. It turned out that the hydrogen bond is important for good radical scavenging activity. The hydrogen atom transfer for β,β-dimethylacrylalkannin derivatives is difficult to occur compared with the zero compound phenol. However, β,β-dimethylacrylalkannin derivatives appear to be good candidates for the one-electron transfer, particularly for β,β-dimethylacrylalkannin derivatives with electron-donating groups. Their naphthoquinone planar conformation and the extended electronic delocalisation between adjacent substituent groups determine low adiabatic ionisation potential (IPa) values. The IPa values of β,β-dimethylacrylalkannin derivatives with –NHPh, –N(CH3)Ph, –N(CH2CH2)O and –N(CH3)2 groups are lower than that of the parent compound β,β-dimethylacrylalkannin, suggesting that these derivatives are expected to be the promising candidates for radical scavenging activity compounds. Taking this system as an example, we present an efficient method for the investigation of radical scavenging activity from theoretical point of view.  相似文献

Monthly observations performed on a typical harmonic-type lake, Lake Biwa (latitude 35°15′ N, longitude 136°05′ E, Japan), showed that the particulate aluminum concentration varied around a mean value of 0.8 μM, with occasional extraordinarily high values, whereas the dissolved aluminum concentration varied, in the surface layer of the whole lake, between a minimum of 0.01 μM and a maximum of 0.30 μM, depending on the season. Although the variation in dissolved aluminum paralleled the variation in the pH of the lake water, the variation in dissolved aluminum lagged behind the variation in pH by approximately 1 month. A series of laboratory incubation experiments suggested that the supply of dissolved aluminum from, and its removal by, the suspended particulate matter involved a slow, pH-dependent reaction. The stoichiometry and the apparent equilibrium constant of this reaction were evaluated by adopting a zeolite-like structure for the surfaces of the suspended particulates. Viable diatom and dinoflagellate resting stages were recovered from sediments in Koljö Fjord on the west coast of Sweden. To determine the maximum survival time of buried resting stages, samples from sediment depths down to 50?cm were incubated at temperatures of 3, 10 and 18?°C. Sediment cores were dated by 210Pb and the age of samples containing viable resting stages was determined using the constant rate of supply model. Dilution cultures of surface sediments allowed semiquantitative estimates of the potential seed bank. Dinoflagellate cysts from species such as Diplopsalis sp., Gymnodinium nolleri, Oblea rotunda and Protoceratium reticulatum were viable down to 15?cm depth, or 37 years old. Spores and resting cells of the diatoms Chaetoceros spp., Detonula confervacea and Skeletonema costatum were viable to over 40?cm depth, and may have been buried for many decades. The seed bank of living resting stages in surficial sediments was found to be rich (c. 57000 diatom resting stages g?1 wet weight and c. 200 dinoflagellate cysts g?1 wet weight), and the percentage of viable resting stages was higher for spore- and cyst-forming species. The oxygen-deficient sediments in Koljö Fjord appear to be a natural conservator of cell viability, a condition not easily simulated in laboratory studies. These results are ecologically important since spores and cysts are a repository of genetic material able to repopulate waters if resuspended and exposed to suitable light, temperature and nutrients.  相似文献

Root growth inhibition is a well-known symptom of aluminum (Al) toxicity in intact plants, mainly because the mechanisms of Al exclusion or resistance that operate outside the root endodermis prevent the ascent of this metal from roots into shoots. This work presents a new method to better understand the direct effects of this metal on rice leaves. For this, Al-sensitive and tolerant rice genotypes, having had their root apices removed, were incubated in AlCl3 solutions to evaluate unblocked metal ascension toward the leaf cells. To avoid regrowth and closing of roots, apices were removed daily and also verified for a lack of tyloses production and consequent obstruction in tracheal elements. Thus, seedlings of both cultivars, which were root apex-free, accumulated differentially high amounts of Al in the leaves, highlighting the importance of mechanisms of Al exclusion or resistance in roots of intact plants. Also, Al moved freely toward leaf cells, clearly inducing necrosis-like mesophyll alterations in both genotypes. Added ultrastructural analyses revealed significant cytoplasmatic damage, mainly in chloroplasts. These results suggest that differential responses to Al sensitivity/tolerance preserved in roots between the genotypes studied are also expressed in leaves. Therefore, this method allowed for development of a possible biological model suitable for investigating the direct effect of Al on cells and, alternatively, other compounds in plant leaf cell physiology.  相似文献

In the past decade, research on long‐term persistence of phytoplankton resting stages has intensified. Simultaneously, insight into life‐cycle variability in the diverse groups of phytoplankton has also increased. Aquatic ‘seed banks’ have tremendous significance and show many interesting parallels to terrestrial seed beds of vascular plants, but are much less studied. It is therefore timely to review the phenomenon of long‐term persistence of aquatic resting stages in sediment seed banks. Herein we compare function, morphology and physiology of phytoplankton resting stages to factors central for persistence of terrestrial seeds. We review the types of resting stages found in different groups of phytoplankton and focus on the groups for which long‐term (multi‐decadal) persistence has been shown: dinoflagellates, diatoms, green algae and cyanobacteria. We discuss the metabolism of long‐term dormancy in phytoplankton resting stages and the ecological, evolutionary and management implications of this important trait. Phytoplankton resting stages exhibiting long‐term viability are characterized by thick, often multi‐layered walls and accumulation vesicles containing starch, lipids or other materials such as pigments, cyanophycin or unidentified granular materials. They are reported to play central roles in evolutionary resilience and survival of catastrophic events. Promising areas for future research include the role of hormones in mediating dormancy, elucidating the mechanisms behind metabolic shut‐down and testing bet‐hedging hypotheses.  相似文献

Scavenging can have important consequences for food web dynamics, for example, it may support additional consumer species and affect predation on live prey. Still, few food web models include scavenging. We develop a dynamic model that includes two facultative scavenger species, which we refer to as the predator or scavenger species according to their natural scavenging propensity, as well as live prey, and a carrion pool to show ramifications of scavenging for predation in simple food webs. Our modeling suggests that the presence of scavengers can both increase and decrease predator kill rates and overall predation in model food webs and the impact varies (in magnitude and direction) with context. In particular, we explore the impact of the amount of dynamics (exploitative competition) allowed in the predator, scavenger, and prey populations as well as the direction and magnitude of interference competition between predators and scavengers. One fundamental prediction is that scavengers most likely increase predator kill rates, especially if there are exploitative feedback effects on the prey or carrion resources like is normally observed in natural systems. Scavengers only have minimal effects on predator kill rate when predator, scavenger, and prey abundances are kept constant by management. In such controlled systems, interference competition can greatly affect the interactions in contrast to more natural systems, with an increase in interference competition leading to a decrease in predator kill rate. Our study adds to studies that show that the presence of predators affects scavenger behavior, vital rates, and food web structure, by showing that scavengers impact predator kill rates through multiple mechanisms, and therefore indicating that scavenging and predation patterns are tightly intertwined. We provide a road map to the different theoretical outcomes and their support from different empirical studies on vertebrate guilds to provide guidance in wildlife management.  相似文献

The net production of dissolved organic matter (DOM) and dissolved combined and free amino acids (DCAA and DFAA, respectively) by the hermatypic coral Acropora pulchra was measured in the submerged condition, and the production rates were normalized to the coral surface area, tissue biomass, and net photosynthetic rates by zooxanthellae. When normalized to the unit surface area, the production rates of dissolved organic carbon and nitrogen (DOC and DON, respectively) were 37 and 4.4 nmol cm− 2 h− 1, respectively. Comparing with the photosynthetic rate by zooxanthellae, which was measured by 13C-tracer accumulation in the soft tissue of the coral colony, the release rate of DOC corresponded to 5.4% of the daily net photosynthetic production. The tissue biomass of the coral colony was 178 µmol C cm− 2 and 23 µmol N cm− 2, indicating that the release of DOC and DON accounted for 0.021% h− 1 and 0.019% h− 1 of the tissue C and N, respectively. The C:N ratios of the released DOM (average 8.4) were not significantly different from those of the soft tissue of the coral colonies (average 7.7). While DFAA did almost not accumulate in the incubated seawater, DCAA was considerably released by the coral colonies at the rate of 2.1 nmol cm− 2 h− 1 on average. Calculating C and N contents of the hydrolyzable DCAA, it was revealed that about 20% and 50%–60% of the released bulk DOC and DON, respectively, were composed of DCAA.  相似文献

Aluminum inhibition of root growth is a major world agricultural problem where the cause of toxicity has been linked to changes in cellular calcium homeostasis. Therefore, the effect of aluminum ions (Al) on changes in cytoplasmic free calcium concentration ([Ca2+]c) was followed in root hairs of wild-type, Al-sensitive and Al-resistant mutants of Arabidopsis thaliana (L.) Heynh. Generally, Al exposure resulted in prolonged elevations in tip-localized [Ca2+]c in both wild-type and Al-sensitive root hairs. However, these Al-induced increases in [Ca2+]c were not tightly correlated with growth inhibition, occurring up to 15 min after Al had induced growth to stop. Also, in 32% of root hairs examined growth stopped without a detectable change in [Ca2+]c. In contrast, Al-resistant mutants showed little growth inhibition in response to AlCl3 exposure and in no case was a change in [Ca2+]c observed. Of the other externally applied stresses tested (oxidative and mechanical stress), both were found to inhibit root hair growth, but only oxidative stress (H2O2, 10 μM) caused a prolonged rise in [Ca2+]c similar to that induced by Al. Again this increase occurred after growth had been inhibited. The lack of a tight correlation between Al exposure, growth inhibition and altered [Ca2+]c dynamics suggests that although exposure of root hairs to toxic levels of Al causes an alteration in cellular Ca2+ homeostasis, this may not be a required event for Al toxicity. The elevation in [Ca2+]c induced by Al also strongly suggests that the phytotoxic action of Al in root hairs is not through blockage of Ca2+-permeable channels required for Ca2+ influx into the cytoplasm. 用化学发光法研究了天竺葵花粗担液对超氧阴离子自由基(O2)^=和羟自由基(OH)的清除作用,用分光不镀法研究了它对1,1-二苯基-2-苦肼基自由基(DPPH0的清除作用。结果表明,天竺葵花具有很强的清除自由基活性。其粗提物清除O2^-分别为3.5μg/ml(红花),3.0μg/ml(粉花),1.6μg/ml(浅粉花),与茶多酚(IC50=1.1μg/ml)相近,清除OH的IC50分别为5.7μg/  相似文献
