The alterations in mitochondrial bioenergetics during growth in a batch culture of Acanthamoeba castellanii were studied. The capacity of cytochrome pathway-dependent respiration measured in vitro decreased from the intermediary phase, when cell division slowed down. The pattern of the cytochrome pathway capacity changes was paralleled from the intermediary phase by alterations in the amount of total (and reducible) membranous ubiquinone. These changes were accompanied by a decrease in mitochondrial reactive oxygen species production in vitro (when no energy-dissipating system was active), and almost no change in superoxide dismutase activity and protein level, thus indicating an equivalent need for this enzyme in oxidative stress defence in A. castellanii culture. On the other hand, a decrease in the activity and protein level of alternative oxidase and uncoupling protein was observed in vitro, when cells shifted from the exponential growth phase to the stationary phase. It turned out that the contribution of both energy-dissipating systems in the prevention of mitochondrial reactive oxygen species generation in vivo could lead to its constant level throughout the growth cycle of A. castellanii batch culture. Hence, the observed functional plasticity insures survival of high quality cysts of A. castellanii cells.  相似文献

The aim of this work was to characterize nitric oxide (NO) generation during the growth of Chlorella sp. cells. The profile of NO production was compared with that observed in Chlorella from temperate climate and Antarctic Chlamydomonas sp. A distinctive electron paramagnetic resonance (EPR) signal for the adduct MGD-Fe-NO was detected in the Antarctic Chlorella sp. cells on days 6 and 7 of growth. An assay based upon the time-dependent detection of NO by EPR was developed to assess both nitrate reductase (NR) and NO synthase (NOS)-like activities. A significant increase in both enzymatic activities was observed on day 6 of growth of Antarctic Chlorella sp. cells. However, NOS-like activity proved undetectable on day 10 of growth, while NR activity was observed until day 22 of growth. When the cultures were supplemented with Nω-nitro- l -arginine methyl ester ( l -NAME) from day 0 to day 13, no growth was observed. After administration of l -NAME from day 5 to day 12, growth rate at log phase was inhibited from 0.18 ± 0.02 (control) to 0.11 ± 0.01 per day. Removal of l -NAME on day 12 increased the growth rate to 0.15 ± 0.02 per day. Supplementation with S-nitroso- N -acetyl-penicillamine on day 5, in the presence of l -NAME from day 0, restored the growth rate to control values. The same profile of NO generation was observed during the growth of Antarctic Chlamydomonas sp.; however, Chlorella cells from temperate climate did not show any difference in NO generation over the growth period. The data reported here are the first observations, by employing EPR, of NO and the activity of the enzymes responsible for its generation during the growth of Antarctic photosynthetic alga.  相似文献

In the last decade Ostreopsis cf. ovata blooms have been among the most intense along the entire Mediterranean coast, leading to ecological and human health problems, that are associated with the toxins (palytoxin-like compounds) produced by these algal cells. These compounds are secondary metabolites, whose rates of synthesis depend on the metabolism of their precursors. In general, growth dynamics and toxicity of dinoflagellates reflect the physiological status of the organism. The aim of the present study was to investigate the cellular production of the main biochemical compounds likely involved in the growth and toxicity dynamics of O. cf. ovata during exponential to the late stationary phase in batch cultures of an Adriatic strain. Removal of major nutrients from the medium was monitored along with concentration, biovolume and production of the main cellular components (e.g. polysaccharides, proteins, lipids and toxins). Nutrient uptake, as well as toxin production rates were calculated in the different growth periods. Nutrients (N and P) were completely depleted when cells entered stationary phase and the greatest net toxin production rate (RTOX) occurred during the first days of growth. The various palytoxins reported a relative abundance quite stable during the different growth phases, while the total toxin cellular amount increased along the growth curve. Total and extracellular released polysaccharides, as well as the lipid content increased greatly during the stationary phase, while proteins were mainly produced by cells during the exponential phase. The continuous release of polysaccharides could facilitate cell aggregation and the formation of the benthic community during algal blooms. The trend of production of the main cellular compounds in O. cf. ovata and the growth dynamics of this species lead us to hypothesize that the fast growth of this dinoflagellate, associated with the rapid use of environmental resources (nutrients, and phosphates in particular), may be an ecological/adaptive strategy which could favor this organism in competition with other species.  相似文献

实验以小球藻为藻种, 构建室内模拟培养装置, 在选定的一组优化的光照、温度和营养盐浓度条件下,采用间歇培养+连续培养的方式对其进行单种群培养, 模拟藻类生物量积累的过程。将培养期划分为5个阶段, 通过分析各阶段的光合荧光参数以及藻细胞内C、N、P含量, 初探生物量积累过程中藻生理参数的变化特征, 为研究水华的发生、维持和消退提供一定的参考。结果显示: 调整期, 藻生长不受生境光热、营养盐限制, 小球藻光合活性较强; 对数生长期, 藻类开始出现光抑制, 热耗散增加, 胞内N、P含量及N/P值较高; 对数生长期和稳定期, 藻同时受光抑制和营养盐抑制, N和P含量、N/P值均下降; 连续培养期, 营养盐限制得到缓解, 小球藻光合活性开始升高, 且藻细胞对N、P营养物吸收能力增强。监控藻类生物量积累过程中细胞内氮磷下降的拐点和细胞内元素比例变化可为预测水华的消退提供一定的参考依据。  相似文献

Copper-zinc superoxide dismutases (CuZnSODs) are infrequently found in bacteria although widespread in eukaryotes. Legionella pneumophila, the causative organism of Legionnaires' disease, is one of a small number of bacterial species that contain a CuZnSOD, residing in the periplasm, in addition to an iron SOD (FeSOD) in their cytoplasm. To investigate CuZnSOD function, we purified the enzyme from wild-type L. pneumophila, obtained amino acid sequence data from isolated peptides, cloned and sequenced the gene from a L. pneumophila library, and then constructed and characterized a CuZnSOD null mutant. In contrast to the cytoplasmic FeSOD, the CuZnSOD of L. pneumophila is not essential for viability. However, CuZnSOD is critical for survival during the stationary phase of growth. The CuZnSOD null mutant survived 10(4)- to 10(6)-fold less than wild-type L. pneumophila. In wild-type L. pneumophila, the specific activity of CuZnSOD increased during the transition from exponential to stationary-phase growth while the FeSOD activity was constant. These data support a role of periplasmic CuZnSOD in survival of L. pneumophila during stationary phase. Since L. pneumophila survives extensive periods of dormancy between growth within hosts. CuZnSOD may contribute to the ability of this bacterium to be a pathogen. In exponential phase, wild-type and CuZnSOD null strains grew with comparable doubling times. In cultured HL-60 and THP-1 macrophage-like cell lines and in primary cultures of human monocytes, multiplication of the CuZnSOD null mutant was comparable to that of wild type. This indicated that CuZnSOD is not essential for intracellular growth within macrophages or for killing of macrophages in those systems.  相似文献

小球藻广泛分布于各种生境,特别是淡水环境中,适应性强。其同化产物主要是淀粉,但在环境胁迫条件下可显著积累中性脂,其脂肪酸类型主要为C16和C18,适合作为生物柴油的原料。我们从中国部分地区水体中分离纯化到若干株小球藻,通过薄层层析比较分析了21株产油小球藻的油脂含量,筛选到一株三酰基甘油含量较高的藻株Chlorella sp.NMX37N。其适宜生长温区为15—35℃,在25℃时生长速率最快,比生长速率为0.53/d,生长的最适光强为250μmol photons/(m2.s)。批量培养实验显示,藻细胞的三酰基甘油含量随培养时间延长而增加,并在培养的稳定期达到最大值,此时培养液中氮基本被耗尽。在批量培养条件下培养Chlorella sp.NMX37N约40d,藻细胞中总脂含量可达到33%左右,与此相比通过两步培养方式,将培养至对数后期(约20d)的藻细胞缺氮处理48h后,得到的总脂产率相当。通过两步培养方式可以大大缩短培养时间,使得该藻细胞快速有效积累油脂。另外,气相色谱分析显示,该藻的总脂和三酰基甘油的脂肪酸均以C16∶0和C18∶2为主,占总脂肪酸的70%以上,且不含C20以上的长链脂肪酸,可以作为优质的生物柴油原料。  相似文献

Summary The influence of the hydrogen-ion concentration on the growth and metabolism of a highly acid-resistant green alga, Chlorella ellipsoidea (strain Marburg St), was studied. Chlorella pyrenoidosa (Emerson strain) served as a normal control organism. Growth of Chlorella ellipsoidea occurs in the entire range from Ph 2.0 to Ph 10, whereas for Chlorella pyrenoidosa the limits were found to be Ph 3.5 and Ph 10. Respiration is much less sensitive to hydrogen-ion concentration in the acid-resistant as compared to the normal strain. Thus an increase in acidity from Ph 4.0 to Ph 2.0 increases the respiratory oxygen uptake by 120% in Chlorella pyrenoidosa and by 25% in Chlorella ellipsoidea. In addition, only the less resistant Chlorella pyrenoidosa shows an accumulation of nitrite in the dark in acid culture media, indicating a disturbance of the normal course of nitrate reduction under these conditions. On the other hand, the rate of photosynthesis of both organisms was found to be almost independent of acidity between Ph 4.0 and Ph 2.0. At the acid and alkaline limits of growth in both algae, an inhibition of cell division leads to an increase of cell size and dry weight per cell, frequently connected with the occurrence of bizarre giant cells. — In addition, adaptation phenomena were found to play a role in determining the acid limit of growth. Cells of Chlorella ellipsoidea, after inoculation from normal medium (Ph about 6) into a solution of Ph 2.0, begin growth at a high rate only after a lag of about two weeks. Cells grown previously in an acid medium, however, immediately resume growth upon inoculation into a medium of Ph 2.0. This adaptation involves a considerable reduction of cell size.  相似文献

Measurements of the rate of oxygen uptake per cell in transplants of Bacterium coli from cultures of this organism in different phases of growth have given results in essential agreement with the observations of others. 2. Correlations of viable count, centrifugable nitrogen, and turbidity, with oxygen consumption, indicate that the increased metabolism during the early portion of the growth period is quantitatively referable to increased average size of cells. 3. DNA synthesis kinetics of P388 leukemic cells growing in ascites form in BDF1 hybrid mice were investigated during the periods of exponential growth and growth restriction. Incorporation of tritiated thymidine, and in some instances tritiated uridine, was studied by autoradiography in cells sorted from S-phase fractions during DNA flow cytometry. During exponential growth continuous labeling with tritiated thymidine indicated a growth fraction of unity, whereas the growth fraction was about 30% during growth restriction. At this growth phase the majority of cells with S phase DNA content remained unlabeled after pulse labeling with tritiated thymidine or uridine, indicating that both the "salvage" and the "de novo" DNA synthesis pathways were blocked in most S-phase cells. After pulse labeling with tritiated thymidine the DNA synthesis rate pattern was investigated by sorting of consecutive fractions of cells throughout the S phase followed by quantitative autoradiography. With exception of a reduced rate in the middle of S phase, the DNA synthesis rate increased as the cells progressed through S phase during exponential growth. In contrast, the DNA synthesis rate pattern had a relative peak in the middle of S phase during growth restriction, which is otherwise characterized by a low mean DNA synthesis rate.  相似文献

A method was developed to provide a real-time measurement of intracellular adenosine 5'-triphosophate (ATP) concentrations in growing Escherichia coli. The bacteria to be monitored must first be modified by inserting the cDNA for firefly luciferase expressed from a constitutive promoter. Such a construct leads to constant specific activity of firefly luciferase during both the lag phase and exponential growth. When the luciferase substrate, D-luciferin, is added to the medium, ATP within the cells is utilized in the luciferase-catalyzed reaction that produces light. The light is carried from the bioreactor to a computer-based detector by an optical fiber. The detected per cell light emission varies during exponential growth. Analysis of cytoplasm extracts shows that this variance is related to changes in the ATP concentration, which ranges from 1 to 6 times the literature value for K(M). Experimental analyses demonstrated that inner filter effects are not a significant factor affecting the use of this system. The method was tested in a benchtop fermentor at cell densities above 13 g/L dry cell weight. A correction factor based on the accumulated light data is calculated and used in real time to account for consumption of luciferin from the culture broth by the light producing reaction. Dissolved oxygen concentrations must be kept above 15% of air saturation to ensure constant light output, but no detectable increase in oxygen demand is seen. Simultaneous washouts of He, N2, and SF6 were monitored during jet ventilation with tidal volumes of 50-200 ml and rates of 1-2 Hz. Gas concentrations were measured from the trachea and from a lower lobe bronchus in six baboons by mass spectrometry. Washouts using large tidal volumes approximated single exponential decays with the relative exponential rates of decay being He fastest, SF4 slowest, and N2 intermediate. Washouts using smaller tidal volumes demonstrated a two-phase exponential decay pattern. During the fast phase, the relative exponential rates of decay were He slowest, SF6 fastest, and N2 intermediate, the reverse order seen during large-volume washouts. During the slow phase, the relative exponential rates of decay were He fastest, SF4 slowest, and N2 intermediate, the same order seen during large-volume washouts. The magnitude of the first phase observed from the lower lobe bronchus was less than that observed from the trachea. These data are consistent with a serial two-compartment transport model incorporating a limitation of molecular diffusion between the peripheral and proximal compartments. The more rapid clearance of less diffusible gases from the central airways during the first phase of washout was due to slower transport from the alveoli to the central airways rather than faster transport from the central airways to the airway opening.  相似文献

Listeria monocytogenes and Listeria innocua are able, under certain conditions, to produce chemiluminescence (CL), which is amplified by luminol. Kinetic studies of CL by L. monocytogenes and L. innocua show a close parallelism between CL and growth curves during the exponential phase, with a maximum of CL reached just before entrance of bacteria into the stationary phase. CL is tightly correlated with the release of oxygen compounds. The reactive oxygen species scavengers tryptophan, mannitol, and tiron, as well as cellobiose and high temperature, were assessed with regard to CL in the two Listeria species. Only tiron strongly reduced the CL emitted by L. monocytogenes and L. innocua. On the other hand, charcoal pretreatment of the growth medium inhibited the CL, whereas ferric citrate strongly increased the CL of L. monocytogenes and L. innocua. These data suggest that iron and superoxide radical are implicated in the CL produced by these bacteria, but this phenomenon is not correlated to virulence.  相似文献

Cultures of Bordetella pertussis from phases of exponential growth, retarded growth and from stationary phase were obtained during periodic dynamic cultivation. Preparations for intravenous immunization of rabbits were made from these cultures. Levels of IgG to pertussis toxin, cell walls preparations from 12 bacterial species, 4 organo-specific antigens, and 7 organospecific human antigens were measured in obtained sera. It was shown that higher levels of IgG to pertussis toxin were found in sera of rabbits immunized with cultures from exponential growth phase whereas decrease of this level in 8 times was observed in sera of rabbits immunized with cultures from retarded growth phase or end of stationary phase. After immunization with culture from exponential growth phase increase of IgG levels to cross-reactive antigens was not observed compared to levels of these antibodies in control sera obtained before immunization. After immunization with cultures from retarded growth phase or end of stationary phase increase of IgG levels to preparations of cell walls of Staphylococcus aureus, S. epidermidis, Pseudomonas aeruginosa, Klebsiella pneumoniae, to denaturated DNA, elastin, and renal and liver microsomal fractions was detected compared to control sera. Described data can substantiate usefulness of obtaining the most specific diagnostic sera and test-systems using cultures of B. pertussis from the phase of exponential growth.  相似文献

The sensitivity of Lactobacillus fermenti ATCC 9338 to lysozyme has its peak during the exponential phase of growth, after the autolytic activity of the organism has begun to decline. Cells from the stationary growth phase are resistant to lysozyme. The two lytic activities require different ionic conditions for their functioning; they appear mutually exclusive. Incubation with trypsin renders cells from all growth phases sensitive to lysozyme. The effect of trypsin is independent of the presence of lysozyme and vice versa, as demonstrated by use of trypsin inhibitors. Cells from early stationary phase of growth require higher temperature for optimum lysozyme action than do those from the exponential growth phase.  相似文献

During the first entering into hibernation in the ground squirrels there are periodic cessations of the process, especially during the transition to moderate and deep torpidity ("critical" periods) which significantly increase energy expenditure in animals during the phase of entering. During this period, high amplitude near-the-hour fluctuations are observed in total duration of wakefulness and sleep, contractile muscle activity, oxygen consumption, heart rate, brain temperature, and thyroid hormone concentration in the blood; at the end of this period, as well as during the exiting from it the duration of deep slow wave sleep significantly increases, paradoxical sleep is not identified. The data obtained confirm a hypothesis of "kindling" homeostasis as neuro-hormonal mechanism of the increase in thermal range of the activity of functional systems of the organism during the entering into torpidity in homoiothermic animals.  相似文献

A numerical analysis of experimental growth curves obtained for Nitrobacter by observing changes in cell numbers, substrate concentration and rate of heat evolution has allowed the calculation of the growth rate constants during the phase of balanced growth. The molar growth yield was smaller during that phase than during the phase preceding it. On the other hand, the rate of heat evolution was larger during exponential growth by a factor of about 1.5 than during the stages up to and including this phase. The two observations being in agreement since, if less efficient synthesis occurs during exponential growth, more free energy must be dissipated as heat.  相似文献

Chlorella pyrenoidosa (Chick) was grown heterotrophically in batch culture on defined medium with glucose. Morphometric analysis of cells in the exponential growth phase showed that starch accounted for 57% of the volume of the chloroplast and 36% of the total cell volume. During the stationary growth phase, the amount of starch accounted for only 36% of the chloroplast volume and 13% of the total cell volume. This represented a 36% decrease in the amount of starch/cell between the exponential and stationary phase. Determination of starch as grams/cell using quantitative techniques on cell extracts showed a comparable decrease in the amount of starch during this same transition. Based on these results, morphometric techniques provided an accurate assay of starch and have the added advantage of visualization of cellular structures not available when quantitative techniques are used.  相似文献
