Summary The sulphur content in leaves increased with increasing levels of sulphur and with the advancement of age and was maximum at 50 days stage and declined towards maturity. There was a significant positive correlation between sulphur content of leaf at various stages of growth and sulphur-containing amino acids viz. methionine, cystine, and cysteine in seed. The sulphur containing amino acids in seeds also increased with increasing levels of sulphur upto 90 ppm and with further increase in the dose it decreased.  相似文献

Summary Effect of S, P and Mo application on quality of soybean grain was studied at Haryana Agricultural University, Hissar (India), in pots. Sulphur was applied at 0, 40, 80 and 120 ppm, P at 0, 40, and 80 ppm and Mo at 0 and 1 ppm. Increasing levels of S upto 120 ppm increased crude protein, cystine, cysteine and methionine contents of grain. However, oil percentage significantly increased only with 40 ppm S and with higher levels of S a decreasing trend was observed. The reducing, non-reducing and total sugars increased with 40 and 80 ppm S and thereafter decreased. Application of 40 ppm P significantly increased reducing total sugar and methionine content of seed whereas Mo application depressed cystine and cysteine. The S concentration in leaves gave positive significant correlation with the percentage of crude protein, oil, total sugar and S containing aminoacids, whereas Mo concentration of leaves was found negatively correlated with cystine and methionine.  相似文献

In order to increase the concentration of the nutritionally essential sulphur amino acids in seed protein, a transgene encoding a methionine- and cysteine-rich protein, sunflower seed albumin (SSA), was transferred to chickpeas (Cicer arietinum L). Transgenic seeds that accumulated SSA contained more methionine and less oxidized sulphur than the controls, suggesting that additional demand for sulphur amino acids from the expression of the transgene stimulated sulphur assimilation. In addition, the activity of trypsin inhibitors, a known family of endogenous, sulphur-rich chickpea seed proteins, was diminished in transgenic, SSA-containing seeds compared with the non-transgenic controls. Together, these results indicate that the reduced sulphur sequestered into SSA was supplied partly by additional sulphur assimilation in the developing transgenic seeds, and partly by some diversion of sulphur amino acids from endogenous seed proteins. Growth of chickpeas on nutrient with a high sulphur-to-nitrogen ratio increased the total seed sulphur content and the accumulation of sulphur amino acids in the seeds, and partly mitigated the effect of SSA accumulation on the trypsin inhibitor amount. The results suggest that free methionine and O-acetylserine (OAS) acted as signals that modulated chickpea seed protein composition in response to the variation in sulphur demand, as well as in response to variation in the nitrogen and sulphur status of the plant.  相似文献

Summary In a pot experiment the effects were estimated of varying nitrogen and sulphur applications on the total and nitrate nitrogen, free amino acid and amide and total amino acid concentrations of barley plants at five early stages of growth.Variations with time in contents of total and nitrate nitrogen differed at low and high nitrogen levels. Concentrations of free amino acids and amides were influenced by light intensity preceding sampling, age of plant and by levels of nitrogen and sulphur. At the fifth harvest (55 days old plants), high nitrogen addition without sulphur caused a fivefold increase in asparagine content and lesser increases in glycine and serine. Alterations in total amino acid contents with time were largely dependent on variations in total nitrogen. Sulphur deficiency, as judged by contents of S-amino acids and amides, became more pronounced with time. A positive interaction was found between effects of nitrogen and sulphur on the yield and cystine and methionine content of grain. Absence of sulphur reduced the cystine content of grain much more than that of methionine, particularly at the high nitrogen level.  相似文献

Sulphur amino acid requirement of juvenile Asian sea bass Lates calcarifer   总被引:1,自引:0,他引:1  
The dietary requirement of juvenile Asian sea bass Lates calcarifer Bloch for total sulphur amino acids was studied. Fish (average initial weight of 2.59 ± 0.08 g) were reared in twelve 500 L fibreglass tanks provided with flow-through seawater at 26°C and salinity of 31 p.p.t. for 12 weeks. They were fed semi-purified test diets containing 6.2, 7.2, 8.1, 9.0, 10.8, or 12.6 g methionine kg−1 dry diet and a basal level of 3.1 g cystine kg−1 dry diet. The mean crude protein of the diets (containing defatted Peruvian fishmeal, squid meal, soybean meal, and free amino acid mixture to simulate the pattern of hydrolysed sea bass protein) was 46.02%. The crude fat content of the diets was 10.51% from a 1 : 1 mixture of cod liver oil and soybean oil. Survival was 100% in all treatments. On the basis of the growth response, the total sulphur amino acid requirement of juvenile Asian sea bass was estimated to be 13.4 g kg−1 dry diet (2.9% of protein). Fish fed low levels of l -methionine had significantly lower weight gains and feed efficiency ratios as well as slightly higher hepatosomatic indices. No nutritional deficiency signs were observed other than growth depression in fish fed on diets that were low in methionine. This information is valuable in further refinement of formulations of practical diets for the Asian sea bass.  相似文献

Data collected on the feeding behavior, food intake, and chemical analyses of plant foods were used to document seasonal variation in diet and nutrition in Eulemur mongoz in northwestern Madagascar. E. mongoz conforms to the general Eulemur dietary pattern, with a predominantly frugivorous diet supplemented mainly by leaves, flowers, and nectar. Phytochemical analysis revealed high water contents in all the main plant foods; mature fruit and flowers contained the most water-soluble carbohydrates; immature leaves were richest in protein and essential amino acids; the limiting amino acids in all plant foods were methionine and cystine; ash (mineral) content was highest in petioles and mature leaves; crude lipid content was highest in seeds; and crude fiber content was indistinguishable between immature and mature fruit and leaves. High-fiber foods were eaten during both seasons; the wet season diet was dominated by high-energy foods (mature fruit, nectar, and seeds), while the dry season diet contained foods high in energy (mature fruit and flowers) and high in protein (immature leaves) and minerals (mature leaves and petioles). However, nutrient intake did not vary between seasons, implying that nutrient requirements are met throughout the year. These results suggest we draw more conservative conclusions when interpreting dietary variability in the absence of chemical analysis, and also draw into question the idea that nutritional stress is a factor in the timing of reproduction in lemurs and, by extension, is linked to the prevalence of female dominance and small group size in lemurs.  相似文献

DOF1 (DNA binding with one finger) plays an important role in regulating C/N metabolism in cereals. In order to validate its role in the regulation of nitrogen use efficiency (NUE) and photosynthetic efficiency in finger millet, 5′–3′ RACE PCR was performed to obtain and characterize full-length Dof1 genes of high and low grain protein finger millet genotypes. The full-length DOF1 ORFs were both 1,284 nt long and were 98.8 % similar over 427 amino acids containing the characteristic Dof domain. Comparison of both the EcDof1 protein sequences with the Dof1 of other cereals revealed high sequence similarity to the Dof1 of rice. Southern hybridization carried out using the probe developed from the region encoding the highly variable C-terminal region of EcDof1 showed the presence of four copies of the DOF1 gene in finger millet, which might explain the high NUE and photosynthetic performance of finger millet. Since the genes involved in C/N metabolism are regulated diurnally and play crucial roles in determining grain protein content during grain filling, the diurnal expression of EcDOF1 was assessed in two finger millet genotypes (GE 3885 and GE 1437) with differing grain protein content (13.8 % and 6.15 % respectively). It was found that EcDOF1 exhibited diurnal regulation and peak differential pattern expression with early phasing in GE3885 and late phasing in GE1437. Differential expression of DOF1 might alter the regulation of genes involved in C/N metabolism affecting grain protein composition of finger millet genotypes.  相似文献

Summary This study was undertaken to evaluate water stress effects during vegetative, flowering, and podfilling stages of cowpea plants (Vigna unguiculata L.) grown under natural field conditions in southern California on seed yield and protein and free amino acid content of the cowpea seeds. The lowest concentration of N was found in the seeds of the control treatment plants while the seed yield from these treatments was the highest as compared with the N concentration and yield of seeds from plants subjected to water stress during flowering and podfilling stages. The concentration of N in the seeds was inversely related to the seed dry weight yield. Protein arginine,-threonine,-serine,-cystine,-valine,-methionine, and-isoleucine were significantly affected by water stress at the three growth stages. There was no consistent pattern in the effect of water stress on the individual amino acids. The sum of protein amino acids in the cowpea seeds was not significantly influenced by the various treatments since some of the protein amino acids increased and others decreased producing an averaging effect on the figures comprising the sums of the amino acids. Water stress during the flowering and pod-filling stages increased the free amino acid pool, and at the same time, inhibited incorporation of the amino acids into the protein chain-thus lowering the protein amino acid fraction simultaneously. With the exception of methionine plus cystine, the essential amino acids in the seeds were present at concentrations equal to or greater than recommended by the World Health Organization and FAO. It is of particular importance to note that the concentration of lysine in the cowpeas was substantially higher than that found in wheat grain. It is also important to note that the amount of essential amino acids per gram of protein was not measurably affected by the water stress treatments during any of the growth stages.  相似文献

The effects of exogenous methionine on growth of developingVicia faba seeds in vivo was studied. Methionine (51 or 102µmol in total) injected into the base of the pod overa period of 15 d (26–40 d after pollination) decreasedgrowth and protein accumulation in proximal seeds but increasedin distal seeds. Distal seeds of methionine injected pods accumulatedmore d. wt and protein than distal seeds of water-injected pods.However, on a pod basis methionine reduced overall seed growthand protein accumulation. Exogenous methionine caused a relative increase in legumin buta decrease in vicilin. Injected methionine also affected thecomposition of uncombined amino acids, especially those derivedfrom aspartic acid. In contrast, the amino acid compositionof the protein fraction did not change appreciably. The data suggest that seed growth is not limited by methioninebiosynthesis but seed protein composition is regulated by methionine. One hundred and twenty pigs, initially 4–5 kg liveweight, were fed on wheat-based diets supplemented with meat meal and amino acids in two experiments, each of 4 weeks duration.In the first experiment, the supplementation of a normal meat meal diet with lysine and methionine increased the feed intake and weight gains of the pigs by 15 and 18%, respectively. Nitrogen retention was increased by 30%. Tryptophane gave a similar response to lysine and methionine.In the second experiment, a 21% crude protein basal diet was similar to the basal diet of the first experiment but it contained 1.02% lysine and 0.50% methionine plus cystine, compared to 0.90 and 0.51%, respectively, in the first experiment. Lysine and methionine supplementation of the diet did not significantly improve the performance of the pigs in the second experiment, but the supplementation of the diets with 0.08% tryptophane and lysine and methionine increased weight gains and feed intakes and decreased urea content of the blood plasma. The 21% crude protein diet contained 0.15% tryptophane.Increasing the crude protein content of diets from 18 to 24% by increasing their meat meal content increased the daily weight gains from 190 to 276 g.  相似文献

Summary The effects of Se and P on the dry matter yield and chemical composition of Raya (Brassica juncea Cos. var. R.L. 18) were studied in the green-house. The dry matter yield in P treated and untreated pots increased with Se application upto 2.5 ppm. With further increase in Se dose upto 10 ppm, dry matter yield decreased. The increase in P dose from 0 to 50 ppm increased dry matter yield in all Se treated and untreated pots but 100 ppm P rather showed decrease in dry matter yield as compared to 50 ppm P.The Se concentration increased by about 100 fold with the application of 10 ppm Se over 0 ppm Se where P was not added whereas increase was 258 and 336 times when 50 ppm and 100 ppm P was added, respectively. Inorganic, organic and total P increased with increasing Se and P. The increase in inorganic P was more than organic and total P. S and N concentration decreased with increasing Se application and increased with P application.Crude protein showed the same behaviour as N. The total sulphur-containing amino acids (which ranged from 39 to 49% of crude protein) and individual sulphur-containing amino acids like methionine (16.9 tot 20%) cysteine (9.8 to 13%) and cystine (12.3 to 15.9%) decreased with the increasing selenium but increased with increasing phosphorus. N/S ratio in the plant showed significant negative correlation with total sulphur-containing amino acids (r=–0.940**), methionine (r=–0.951**), cysteine (r=–0.929**) and cystine (r=–0.920*) whereas total sulphur in the plant showed positive significant correlation with methionine (r=0.805**), cysteine (r=0.924**) and cystine (r=0.821**).  相似文献

Pyricularia grisea, the fungus that causes blast disease in finger millet [Eleusine coracana (L) Gaertn] produced maximum quantity of crude toxins. Crude toxins were partially purified and characterized by UV and mass spectra. Pyrichalasin H, a phytotoxic metabolite, has been identified and found to be toxic when tested for inhibition of seed germination and seedling growth of blast resistant and susceptible finger millet varieties.  相似文献

A field experiment with groundnut as test crop was conducted to evaluate the manurial potential of three distillery effluents: raw spent wash (RSW), biomethanated spent wash (BSW) and lagoon sludge (LS) vis-à-vis recommended fertilizers (NPK + farm yard manure (FYM)) and a control (no fertilizer or distillery effluent). It was found that all the three distillery effluents increased total chlorophyll content, crop growth rate (CGR), total dry matter, nutrient uptake (N, P and K) and finally seed yield compared to the control but inhibited nodulation and decreased nitrogen fixation. Among the three distillery effluents, BSW produced the highest seed yield (619 kg ha(-1)) twice that of control (3.10 kg ha(-1)), followed by RSW (557 kg ha(-1)) and LS (472 kg ha(-1)). However, the distillery effluents did not influence protein and oil contents. It was concluded that these distillery effluents because of their high manurial potential could supply nutrients, particularly potassium, nitrogen and sulphur, to the crops and thus reduce the fertilizer requirement of crops. Nevertheless, the crop performance and yield with three distillery effluents were overall less than that produced by recommended NPK + FYM probably on account of failure of the effluents to supply balanced nutrition to the plants for achieving their potential growth capacity.  相似文献

The effect of crude proteinase inhibitor extracts from seeds of different crop plants (black gram, chickpea, chickling vetch, finger millet, French bean, green gram, horse gram, lentil, pea and soybean) on the insecticidal activity of B. thuringiensis var. kurstaki HD-1 was investigated against neonate larvae of H. armigera by diet incorporation method. The larval mortality due to crude proteinase inhibitors alone (5% seed weight equivalent) ranged from 4.1 to 19.1%; the maximum mortality with finger millet and the minimum with pea var. DDR-23. A mixture of B. thuringiensis var. kurstaki HD-1 (10 ppm) and proteinase inhibitor (5% seed weight equivalent) was synergistic in larval mortality with respect to proteinase inhibitors of pea var. DMR-16, chickling vetch var. RLK-1098 and B101-212, lentil var. ILL-8095 and L-4076, soybean var. PK-1042, PK-416 and Pusa-22, chickpea var. Pusa-413, French bean (Chitra) and black gram; and antagonistic with respect to those of finger millet, horse gram and kidney bean. The larval growth reduction with crude proteinase inhibitors alone ranged from 17.9 to 53.1%; the maximum growth reduction with soybean var. PK-1042 and minimum with lentil var. L-4076. A mixture of B. thuringiensis var. kurstaki and proteinase inhibitor was synergistic in growth reduction with respect to proteinase inhibitors of lentil var. ILL-8095, and L-4626 and antagonistic with respect to that of finger millet. The midgut proteinase inhibition with crude seed extracts (3.3% seed weight equivalent) ranged from 9.3 to 60.9% and was negatively correlated with larval mortality. These results showed that interactive effect of B. thuringiensis var. kurstaki HD-1 and proteinase inhibitors in the larvae of H. armigera depended upon the quality and quantity of proteinase inhibitors, which vary widely in different plants.  相似文献

In recent years there has been an intensification of efforts to make use of microbial fermentation for the production of nutrients and feeds containing protein. Therefore we have investigated some yeasts and a bacterium produced on unconventional nutrient substrates such as methanol, crude oil, n-alkanes and whey, comparing them with yeasts produced by normal techniques, using sulphite liquors, molasses and extract of barley-malt as a carbon source.The crude protein content of the dry matter of all the years was between 39 and 68% and that of the bacterium examined was about 82%. As regards the amounts of essential amino acids in SCP, the high concentration of lysine, with 6.6–8.4 g/16 g N in the yeasts, and 5.9 g/16 g N in the bacterium, deserves special mention. By contrast, the amount of amino acids containing sulphur in the yeasts examined is not satisfactory at 1.5–3.3 g/16 g N, whereas the amount in the bacterium was more suitable at 3.6 g/16 g N. Methionine and cystine are thus the first limiting amino acids. Also, the arginine content in some products might be inadequate for the growth of fowl.Besides the protein, the crude fat fraction has to be considered, firstly because of its considerable order of magnitude, and secondly because of its very variable composition.In an animal experiment, a satisfactory to good biological value of the protein of 61–80%, and an NPU of 55–76%, were found. By adding 0.2% DL-methionine to the dry feed, it was possible to improve considerably these criteria. On the basis of their very high lysine content, these materials have a good to very good value as supplements to our basic feeds for pigs.Metabolic trials with pigs yielded among other things, a protein digestibility of 93% for 2 yeasts, and 84% for the bacterium studied. In fattening trials with pigs, an equally good feeding effect was achieved through equal-protein replacement of fish meal and extracted soya bean meal by yeasts and the bacterium.For the feeding of calves, it is advisable at first, to replace only small amounts of the milk protein in the milk-substitute feed with SCP.  相似文献

Summary The reserve protein composition of 22 Phaseolus species has been studied. The non-sulphur-containing amino acids were present at values higher than those suggested for animal (and human) nutrition, but the sulphur-containing amino acids, with some exceptions, were under the minimum accepted requirement. However, taking into account the variability in the percentages of methionine and cystine, as well as the cystine/methionine ratio, it is concluded that the genus Phaseolus has a theoretical possibility for synthesizing a reserve protein with a balanced sulphur-containing amino acid content. An accession from Mexico of the species Ph. phyllanthus possesses a protein characterized by a high sulphur-containing amino acid content (3,84%). The possible utilization of this species in breeding for legume protein quality is suggested. Some observations about the percentage of arginine in wild and cultivated forms of Ph. Vulgaris are also presented.  相似文献

The effect of the supplementation of sulfur amino acids to a low casein or soy protein isolate diet on tissue lipid metabolism was investigated. Supplementation of methionine to a 8% casein diet produced a fatty liver in rats, however, supplementation of methionine to a 8.8% soy protein diet (corresponding to a 8% casein diet as to sulfur amino acids content) did not produce a fatty liver. At the level of 8% or less of soy protein in the diet, the accumulation of liver lipids and serum triglyceride was observed. An amino acid mixture simulating the composition of soy protein isolate caused significant accumulation of liver lipids, but serum triglyceride was not changed. Serum cholesterol in rats fed the soy protein diet was lower than that in rats fed the casein diet, but on feeding the amino acid mixtures simulating these protein diets, there was no difference between the two groups. The small differences between soy protein isolate and casein as to lipid metabolism might be due to the small differences in the contents of sulfur amino acids or the specific nature of the soy protein or casein. The supplemental effect of methionine and cystine was also studied. About 60% of total sulfur amino acids could be substituted by cystine for maximum growth.  相似文献
