One of the most common marine dinophytes is a species known as Heterocapsa triquetra. When Stein introduced the taxon Heterocapsa, he formally based the type species H. triquetra on the basionym Glenodinium triquetrum. The latter was described by Ehrenberg and is most likely a species of Kryptoperidinium. In addition to that currently unresolved nomenclatural situation, the thecal plate composition of H. triquetra sensu Stein (1883) was controversial in the past. To clarify the debate, we collected material and established the strain UTKG7 from the Baltic Sea off Kiel (Germany, the same locality as Stein had studied), which was investigated using light and electron microscopy, and whose systematic position was inferred using molecular phylogenetics. The small motile cells (18–26 μm in length) had a biconical through fusiform shape and typically were characterized by a short asymmetrically shaped, horn‐like protuberance at the antapex. A large spherical nucleus was located in the episome, whereas a single pyrenoid laid in the lower cingular plane. The predominant plate pattern was identified as apical pore complex (Po, cp?, X), 4′, 2a, 6′′, 6c, 5s, 5′′′, 2′′′′. The triradiate body scales were 254–306 nm in diameter, had 6 ridges radiating from a central spine, 9 peripheral and 3 radiating spines, and 12 peripheral bars as well as a central depression in the basal plate. Our work provides a clarification of morphological characters and a new, validly published name for this important but yet formally undescribed species of Heterocapsa: H. steinii sp. nov.  相似文献

The diversity of extant calcareous dinophytes (Thoracosphaeraceae, Dinophyceae) is not fully recorded at present. The establishment of algal strains collected at multiple localities is necessary for a rigorous study of taxonomy, morphology and evolution in these unicellular organisms. We collected sediment and water tow samples from more than 60 localities in coastal areas of the eastern Mediterranean Sea and documented 15 morphospecies of calcareous dinophytes. Internal transcribed spacer (ITS) barcoding identified numerous species of the Scrippsiella trochoidea species complex that were genetically distinct, but indistinguishable in gross morphology (i.e. with the same tabulation patterns of the motile theca and similar spiny coccoid stages). We assessed a possible minimal number of cryptic species using ITS ribotype networks that indicated the existence of at least 21 species within the Scrippsiella trochoidea species complex. Species diversity of calcareous dinophytes appears higher in the Mediterranean Sea than in other parts of the world??s oceans such as the North Sea. Our data underline the importance of field work to record the diversity of calcareous dinophytes and other unicellular life forms.  相似文献

The diversity of extant calcareous dinophytes (Thoracosphaeraceae, Dinophyceae) is currently not sufficiently recorded. The majority of their coccoid stages are cryptotabulate or entirely atabulate, whereas relatively few forms exhibit at least some degree of tabulation more than the archeopyle. A survey of coastal surface sediment samples from the Mediterranean Sea resulted in the isolation and cultivation of several strains of calcareous dinophytes showing a prominent tabulation. We investigated the morphologies of the thecate and the coccoid cells and conducted phylogenetic analyses using Maximum Likelihood and Bayesian approaches. The coccoid cells showed a distinct reflection of the cingulum (and were thus cingulotabulate), whereas thecal morphology corresponded to the widely distributed and species‐rich Scrippsiella. As inferred from molecular sequence data (including 81 new GenBank entries), the strains belonged to the Scrippsiella sensu lato clade of the Thoracosphaeraceae and represented two distinct species. Morphological details likewise indicated two distinct species with previously unknown coccoid cells that we describe here as new, namely S. bicarinata spec. nov. and S. kirschiae spec. nov. Cingulotabulation results from the fusion of processes representing the pre‐ and postcingular plate series in S. bicarinata, whereas the ridges represent sutures between the cingulum and the pre‐ and postcingular series in S. kirschiae, respectively. Bicarinate cingulotabulation appears homoplasious among calcareous dinophytes, which is further supported by a comparison to similar, but only distantly related fossil forms.  相似文献

The Tibetan folk medicine Qinjiao is traditionally used to treat various conditions, and its main active constituents comprise four iridoid glycosides, i.e., loganic acid, swertiamarin, gentiopicroside, and sweroside. The traditional crude medicine Qinjiao is derived from the dried roots of three species belonging to Gentiana sect. Cruciata (Gentianaceae) growing in the Qinghai‐Tibetan Plateau (QTP). In this study, we determined by HPLC the contents of the four main active constituents in the dried roots collected from 83 localities at different altitudes across the QTP. The material was classified under the seven taxonomic species G. straminea, G. dahurica, G. crassicaulis, G. waltonii, G. officinalis, G. ihassica, and G. macrophylla. Our results suggested that the four constituents were present in the roots of all seven species for all localities, but their concentrations varied greatly within and between species. The level of gentiopicroside revealed to be the most dominant for all examined localities (2.1–12.4 mg/g), and G. macrophylla Pall . contained the highest concentration of all the four constituents at the species level. Except for loganic acid in G. officinalis, there was no significant correlation between the contents of these constituents and the altitude of the sampling localities. These results suggest that all species of all origins can be used as reliable resource for the crude medicine Qinjiao. However, a few species contain higher concentrations of the main active constituents, irrespective of their origin.  相似文献

Summary A peridinin-chlorophyll a-protein complex (PCP) was obtained in large quantity from the marine dinoflagellates, Glenodinium sp. and Gonyaulax polyedra. The chromoproteins have similar molecular weights, 35,500 for Glenodinium sp. and 34,500 for G. polyedra. The proteins from the PCP complex of Glenodinium sp. dissociated from the chromophore on treatment with 1% sodium dodecyl sulfate (SDS) at room temperature. The protein component was a single subunit with a molecular weight of 15,500. Proteins from the PCP complex of G. polyedra were composed of a single polypeptide with a molecular weight of about 32,000. Two peridinin-chlorophyll a-proteins from Glenodinium sp. accounted for 70% of the PCP complex and had isoelectric points of 7.4 and 7.3. The PCP complex from G. polyedra was dominated by a single chromoprotein with an isoelectric point of 7.2 Chromophore analysis indicated the presence of only peridinin and chlorophyll a in a molar ratio approaching 4:1. Other pigments characteristically found in dinoflagellates were absent. Fluorescence excitation spectra of purified PCP indicated an efficient energy transfer from peridinin to chlorophyll a, an observation that lends support to the reported role of peridinin as an accessory pigment in photosynthetic oxygen evolution. In several other brown colored dinoflagellates examined, PCP representtd less than 20% of the total peridinin. However, no PCP could be isolated from cultures of Amphidinium carterae (PY-1). This study provides further evidence that PCP is a normal component of most peridinin-containing dinoflagellates, and functions as a light-harvesting component of the dinoflagellate chloroplast. Feeding in Peranema trichophorum (Ehrenberg 1838) Stein 1878 was first observed over a century ago, yet there is still contention over how the feeding apparatus is used in feeding. Using video microscopy and scanning microscopy, this study documents two types of feeding. Peranema may engulf prey cells whole. Immotile cells are preferred, but moving cells occasionally are engulfed. Details of the early stages of engulfment are presented using scanning electron microscopy. A second method of feeding begins with the attachment of Peranema to a prey cell. The rods of the feeding apparatus then are repeatedly scraped over the surface of the prey until a tear occurs in the cell. The feeding apparatus is inserted into the opening and the internal cell contents are sucked out. The anterior flagellum is inserted into the prey to help remove the cell's contents.  相似文献

Hybridisation is considered an important evolutionary phenomenon in Gnaphalieae, but contemporary hybridisation has been little explored within the tribe. Here, hybridisation between Helichrysum orientale and Helichrysum stoechas is studied at two different localities in the islands of Crete and Rhodes (Greece). Using three different types of molecular data (AFLP, nrDNA ITS sequences and cpDNA ndhF sequences) and morphological data, the aim is to provide simultaneous and direct comparisons between molecular and morphological variation among the parental species and the studied hybrid populations. AFLP profiles, ITS sequences and morphological data support the existence of hybrids at the two localities studied, shown as morphological and genetic intermediates between the parental species. Chloroplast DNA sequences show that both parental species can act either as pollen donor or as maternal parent. Fertility of hybrids is demonstrated by the viability of seeds produced by hybrids from both localities, and the detection of a backcross specimen to H. orientale. Although there is general congruence of morphological and molecular data, the analysis of morphology and ITS sequences can fail to detect backcross hybrids.  相似文献

  1. Reliable determination of organisms is a prerequisite to explore their spatial and temporal occurrence and to study their evolution, ecology, and dispersal. In Europe, Bavaria (Germany) provides an excellent study system for research on the origin and diversification of freshwater organisms including dinophytes, due to the presence of extensive lake districts and ice age river valleys. Bavarian freshwater environments are ecologically diverse and range from deep nutrient-poor mountain lakes to shallow nutrient-rich lakes and ponds.
  2. We obtained amplicon sequence data (V4 region of small subunit-rRNA, c. 410 bp long) from environmental samples collected at 11 sites in Upper Bavaria. We found 186 operational taxonomic units (OTUs) associated with Dinophyceae that were further classified by means of a phylogenetic placement approach.

The maximum likelihood tree inferred from a well-curated reference alignment comprised a systematically representative set of 251 dinophytes, covering the currently known molecular diversity and OTUs linked to type material if possible. Environmental OTUs were scattered across the reference tree, but accumulated mostly in freshwater lineages, with 79% of OTUs placed in either Apocalathium, Ceratium, or Peridinium, the most frequently encountered taxa in Bavaria based on morphology.
  4. Twenty-one Bavarian OTUs showed identical sequences to already known and vouchered accessions, two of which are linked to type material, namely Palatinus apiculatus and Theleodinium calcisporum. Particularly within Peridiniaceae, delimitation of Peridinium species was based on the intraspecific sequence variation.
  5. Our approach indicates that high-throughput sequencing of environmental samples is effective for reliable determination of dinophyte species in Bavarian lakes. We further discuss the importance of well-curated reference databases that remain to be developed in the future.

Busseola fusca (Fuller) (Lepidoptera: Noctuidae) is one of the major cereal pests in sub-Saharan Africa. Previous phylogeographic investigations on samples collected in Kenya, Cameroon and West-Africa showed the presence of three main clades (W, KI, KII) originated from populations isolated in West and East Africa around one million years ago. Demographic and phylogenetic analyses suggested that this event was followed by local demographic expansion and isolation by distance. These hypotheses were tested by a more comprehensive sampling across B. fusca’s geographic range in Africa. Comparisons of sequences of partial mitochondrial DNA gene (cytochrome b) from 489 individuals of 98 localities in southern, central, eastern and western African countries confirmed the presence of the three main clades. Phylogenetic, F-statistics, demographic parameters and nested clade phylogeographic analyses confirmed that the clades experienced geographic and demographic expansion with isolation by distance after their isolation in three refuge areas. The geographic range of clade KII, already known from East to Central sub-Saharan Africa was extended to Southern Africa. Mismatch distribution analysis and the negative values of Tajima’s D index are consistent with a demographic expansion hypothesis for these three clades. Significant genetic differentiations were revealed at various hierarchical levels by analysis of molecular variance (AMOVA). Hypotheses about the geographic origin of the three main clades are detailed.  相似文献

Powdery mildew, caused by Blumeria graminis is an important disease of cereals in many production regions. Until end of the last century triticale had been regarded as a species characterized by high level of resistance for this disease. However, after several years of intensive production on a big area in Poland, Germany and other European countries it start to be susceptible for many pathogens including B. graminis. Because of this, virulence structure of this pathogen population on triticale in Poland was evaluated across 2008–2010. Leaf samples with symptoms of the powdery mildew disease were collected randomly from nineteen localities. As a total, 1402 B. graminis isolates were collected: 23–25 isolates per locality in each year. Standard differential set of 28 genotypes was used: twenty‐one wheat with known resistance genes and seven triticale. Based on the obtained results it was possible to observe significant differences in virulence structure between years and localities. No virulence's against Pm21 (Yangmai5), and Pm3d + 4b (Kadett) were found in any year. All tested isolates were virulent on Moreno and Lamberto cultivars. In a total, 36% of tested isolates possessed 9, 11 or 12 virulence's per genotypes. Twenty five percent of tested isolates were virulent to 5 triticale cultivars. Correlation between pathotypes frequency and sampling region were not found what suggest that local epidemics play the most important role in triticale growing regions in Poland.  相似文献

Symbiotic algae of the ciliate Paramecium bursaria (Ehrenberg) Focker are key species in the fields of virology and molecular evolutionary biology as well as in the biology of symbiotic relationships. These symbiotic algae were once identified as Zoochlorella conductrix Brandt by the Dutch microbiologist, Beijerinck 120 years ago. However, after many twists and turns, the algae are today treated as nameless organisms. Recent molecular analyses have revealed several different algal partners depending on P. bursaria strains, but nearly all P. bursaria contains a symbiont belonging to either the so‐called ‘American’ or ‘European’ group. The absence of proper names for these algae is beginning to provoke ill effects in the above‐mentioned study areas. In the present study, we confirmed the genetic autonomy of the ‘American’ and ‘European’ groups and described the symbionts as Chlorella variabilis Shihira et Krauss and Micractinium reisseri Hoshina, Iwataki et Imamura sp. nov., respectively (Chlorellaceae, Trebouxiophyceae).  相似文献

Main aspects of biology and ecology of Bassus tumidulus (Nees), a parasitoid of Gypsonoma aceriana (Dup.), were studied during the period 1989–2000. Poplar tender shoots with G. aceriana larvae were collected at four localities in Bulgaria (Sofia, Svoge, Vardim and Pazardzhik) and examined in laboratory conditions. B. tumidulus was recovered in the Sofia and Svoge localities. It is a solitary internal parasitoid that attacks young larvae of G. aceriana (first–second instars) and kills host pre‐pupae. It develops two generations and overwinters as a larva in the host. Adult emergence of both overwintering and summer generation of B. tumidulus coincides with adult emergence of the host. In 1997, the first generation emergence of B. tumidulus was in relatively good synchrony with the first larval population of G. aceriana. However, the second generation B. tumidulus was not very well synchronized with the life cycle of G. aceriana because adult parasitoids appeared mostly in the beginning of the host larval population. The average mortality of G. aceriana, caused by this parasitoid in 1997 in Sofia, was 15.7 and 23.3% for the overwintering and summer generation, respectively. The highest level of parasitism by B. tumidulus, observed in individual study, was 61.5%, occurring during the summer generation in 1997.  相似文献

Abstract. The mealybug Trabutina mannipara (Hemprich & Ehrenberg, 1829) is described and illustrated, and a neotype female selected from topotypic material collected in the Sinai Peninsula. Gyrodactylus anguillae is known from four continents, and intraspecific variation in the morphology of specimens from each continent is negligible. However, morphological characters alone may not always be adequate for recognizing populations that have been isolated geographically over evolutionary time. We were unable to obtain specimens from Northeast Asia, but G. anguillae collected from North America, Europe and Australia had identical ribosomal DNA sequences (ITS1-5.8SrRNA-ITS2 region). This lack of even minor genetic variation in populations from three remote continents leads us to conclude that they share a contemporary common ancestor. This is one of at least five pests of eels to have invaded new continents as a result of the eel trade in the last four decades; these other known eel pests include Pseudodactylogyrus spp. (Monogenea) and Anguillicola spp. (Nematoda). Their dispersal has probably been secondarily augmented by longshore migration of infected eels, and perhaps also by transportation in ballast waters. The only other known gyrodactylid from eel gills, G. nipponensis from Japanese eel, is not closely related to G. anguillae, because the ITS1 region of the former is short (370 bases) and that of the latter long (685 bases). G. anguillae probably originated in Europe, because it was recorded there over 40 years ago, before the development of the international eel trade. We predict that the worm has a high potential to invade eel populations in East Africa, Southeast Asia, and New Zealand.  相似文献

Phenotypic variability and mixing of material due to massive cultivation for commercial purposes has contributed to the taxonomic confusion ofGracilaria in Chile. At least four species with cylindrical thalli and similar morphology have been recorded. However, since establishment ofG. chilensis, most of the collected thalli have been attributed to this species despite the lack of diagnostic features. In an attempt to resolve whetherGracilaria from 3 localities where it grows in natural and artificial populations belongs to the same species, gametophytic samples were compared by applying RAPD-PCR to their total DNA. This was analysed using 25 different 10-mer primers from which 21 revealed polymorphism within and between populations. Similarity matrices and cluster analyses were performed based on the presence/absence of bands representing fragments of DNA generated by random amplification. Similarity values between two of the populations were equivalent to those detected within a third, indicating the mixing of genetic material due to transplant between the two former localities. Similarities between samples of ChileanGracilaria andG. tenuistipitata from Sweden are considerably lower (0.45–0.53) than those between populations from Chile (0.74–0.88), confirming the existence of a single specific taxon,G. chilensis, in these three localities.  相似文献

Morphological studies at the light microscopy and scanning electron microscopy levels of selected fragilarioid diatoms occurring in North American streams and lakes are presented herein. The majority of the samples studied were collected by the US Geological Survey's National Water Quality Assessment Program, which concentrates on stream water quality monitoring throughout the continental USA and Hawaii. Two new species (Staurosirella confusa Morales and Punctastriata mimetica Morales) and a new forma (Pseudostaurosiropsis geocollegarum f. triradiatum Morales) are described and two new combinations (Pseudostaurosira subsalina (Hustedt) Morales and Staurosirella olden‐burgiana (Hustedt) Morales) are provided. Morphological details of an additional taxon, Staurosira construens var. binodis (Ehrenberg) Hamilton in Hamilton et al. are also presented. The taxonomic affinities of all these taxa, as well as some evolutionary aspects and ecologic characteristics, are discussed in the light of published material.  相似文献

Dinophytes acquired chloroplasts obviously early in evolution and later lost them multiple times. Most families and genera contain both photosynthetic and heterotrophic species. Chloroplasts enveloped by three membranes with thylakoids in stacks of three, containing peridinin as the main pigment, are regarded as the original dinophyte plastids. Pyrenoids are generally present. Stigmata, if present, are usually parts of the chloroplast or are modified original plastids. The form II type RUBISCO found in the dinophytes is unique for eukaryotes, otherwise known only in some anaerobic bacteria. It is disputed whether the original dinophyte chloroplasts are derived from a prokaryotic or an eukaryotic endosymbiosis. Various dinoflagellates contain aberrant chloroplasts. Glenodinium foliaceum and Peridinium balticum have a single complete endosymbiont, originally a pcnnate diatom. Podolampas bipes houses several dictyophycean symbiont cells. The “symbionts” of Lepidodiniurn viride and Gymnodinium chlorophorum are highly reduced prasinophyte cells. The chloroplasts of Gymnodinium mikimotoi have aberrant pigments (fucoxanthin derivatives, no peridinin) and fine structure. The dinoflagellate hosts do not seem to contain any parts of the former endosymbiont except the chloroplasts. Photosynthetic Dinophysis species have cryptophycean-like chloroplasts, whereas symbiotic cyanobacteria are found in other members of the Dinophysiales, e.g., Ornithocercus. Various dinophytes, e.g. Gymnodinium aeruginosum, use kleptochloroplasts from ingested cryptophytes transiently for photosynthesis. Original or secondarily acquired chloroplasts can only be used for phylogenetic considerations in exceptionally cases: it seems unlikely that the Prorocentrales have evolved from the Dinophysiales because all Prorocentrales possess original dinoflagellate chloroplasts, whereas no member of the Dinophysiales has such chloroplasts.  相似文献
