为了探索东方亚种数量正在减少的原因和制定科学、有效的保护措施,利用微卫星DNA对大鸨东方亚种(Otistardadybowskii)的遗传多样性进行了研究。应用3个大鸨指名亚种的微卫星位点和13个波斑鸨的微卫星位点扩增了47个个体的基因组DNA,筛选出8个具有多态性的微卫星。其中有3个微卫星的多态性较低,其余5个微卫星的多态性较高。各位点的观察杂合度为0.0435-1.0000,平均杂合度(h)为0.6595;各位点多态信息含量(PIC)为0.0416-0.8520,平均为0.5497;有效等位基因数(E)为1.04-7.46,平均为3.61。4个位点符合HardyWeinberg平衡,4个位点偏离了HardyWeinberg平衡。多方面比较发现,大鸨东方亚种遗传多样性很低,且低于指名亚种,这可能由于其种群较小、历史遗传瓶颈作用、生境破碎化、分布地域紧缩等原因造成的。  相似文献

Dispersing individuals can use conspecifics as indicators of habitat quality and aggregate at traditionally occupied sites, leaving other favourable patches unoccupied. Here we test the predictions of the conspecific-based habitat selection hypothesis on a Spanish great bustard (Otis tarda) metapopulation, currently fragmented due to recent human-induced habitat changes. The number of birds had increased by 23% between 1988 and 1998, but not consistently among leks. Leks that were large in 1988 increased, while those that were small decreased, which suggests that dispersing individuals used the numbers of conspecifics as cues for breeding-site selection. Moreover, leks with high productivity increased, while those with low productivity decreased. Finally, lek distribution was markedly stable throughout the decade, with no establishment of new leks, and suitable habitat patches remained unoccupied, as predicted by the conspecific attraction hypothesis. These results were corroborated by a simulation model which incorporated natal dispersal rates between leks as obtained through radio-tracking of 15 birds that survived throughout their 4-year dispersal period. In conclusion, in spite of the apparent increase in total numbers throughout the decade, both conspecific attraction and local differences in reproductive success contributed to a more aggregated distribution, increasing the species' vulnerability to local catastrophes, and the risks of reduced genetic diversity and extinction of small leks.  相似文献

Recent advances in molecular biology have made it possible to use the trace amounts of DNA in faeces to non-invasively sample endangered species for genetic studies. Here we use faeces as a source of DNA and mtDNA sequence data to elucidate the relationship among Spanish and Moroccan populations of great bustards. 834 bp of combined control region and cytochrome-b mtDNA fragments revealed four variable sites that defined seven closely related haplotypes in 54 individuals. Morocco was fixed for a single mtDNA haplotype that occurs at moderate frequency (28%) in Spain. We could not differentiate among the sampled Spanish populations of Cáceres and Andalucía but these combined populations were differentiated from the Moroccan population. Estimates of gene flow (Nm = 0.82)are consistent with extensive observations on the southern Iberian peninsular indicating that few individuals fly across the Strait of Gibraltar. We demonstrate that both this sea barrier and mountain barriers in Spain limit dispersal among adjacent great bustard populations to a similar extent. The Moroccan population is of high ornithological significance as it holds the only population of great bustards in Africa. This population is critically small and genetic and observational data indicate that it is unlikely to be recolonised via immigration from Spain should it be extirpated. During a long-term study of individually marked, free-living male great bustards captured as chicks and radio-tracked through several years in Spain, we studied the development with age of two secondary sex traits, the moustachial feathers and the neck plumage pattern. Juvenile males acquired full adult plumage between their fourth and seventh years. The main changes occurred at the neck, coinciding with the onset of sexual maturity. The grey colour typical of immature males turned to ivory white around the fourth to fifth spring, and a gradual increase was appreciated in adults in the brightness of the white colour of the upper neck and in the contrast between this and a progressively more intense chestnut brown at the neck base. Based on these changes, we proposed four neck plumage patterns that can be used to differentiate male age classes during the mating period. The development of moustachial feathers showed more interindividual variability and was not as useful as the neck plumage to estimate male age.  相似文献

With recent advances in molecular biology, it is now possible to use the trace amounts of DNA in faeces to non-invasively sample endangered species for genetic studies. A highly vulnerable population of approximately 100 great bustards (Otis tarda) exists in Morocco necessitating the use of non-invasive protocols to study their genetic structure. Here we report a reliable silica-based method to extract DNA from great bustard faeces. We found that successful extraction and amplification correlated strongly with faeces freshness and composition. We could not extract amplifiable DNA from 30% of our samples as they were dry or contained insect material. However 100% of our fresh faecal samples containing no obvious insect material worked, allowing us to assess the levels of genetic variation among 25 individuals using a 542 bp control region sequence. We were able to extract DNA from four out of five other avian species, demonstrating that faeces represents a suitable source of DNA for population genetics studies in a broad range of species. Early anthropogenic impacts on the abundance and distribution of wild species are difficult to document, but can help us to understand the causes and relative importance of current declines. Genetic data can be of use in inferring historical demographic events, but the accuracy of these inferences depends on the availability and precision of demographic parameters that are difficult to obtain in the field. Here, we use demographic data on Iberian populations of the threatened great bustard (Aves: Otis tarda), obtained from an intensive population monitoring programme over the last 20 years, to estimate critical population parameters (population size and generation time), which are then used in a Bayesian Skyline Plot (BSP) analysis of mitochondrial DNA sequence data to assess changes in population size over the last several thousand years. BSP showed a sudden and sharp great bustard population decline coinciding with human expansion in Iberia, and the associated agricultural and urban development and increased hunting pressure. These results illustrate the importance of human population size as a possible ultimate cause of an environmental impact that occurred in the historical past, a fact that has often been neglected. Our results also suggest the role of human activities in driving historical population declines in great bustards, and underscore the importance of precise, long‐term field data to infer past demographic trends from parameters of extant populations. © 2013 The Linnean Society of London, Biological Journal of the Linnean Society, 2013, 110 , 518–527.  相似文献

Individual display rates and attractiveness of 17 radio-tagged, free-living great bustard males in north-western Spain were studied in relation to the degree of development of secondary sex traits, body condition, and age. Variance in display rate was significantly explained by body condition and length of moustache feathers, and neck development and display rate significantly accounted for the variance in male attractiveness. Age did not present any clear association with display rate or attractiveness, and it was significantly correlated only with neck development. These results suggest a total viability scenario of female mate choice in which the advertisement of global fitness is based on condition-dependent phenotypic and behavioural traits. This paper describes at a regional scale the distribution pattern, density, productivity and sex ratio of great bustards in northwestern Spain and explores the role played by habitat type, terrain characteristics and human disturbance on variation in its demographic parameters. Data from 136 plots covering an area of 7314 km2 were obtained in two censuses carried out in the spring and summer of 1998. The density of the great bustards was 1.39 individuals/km2 in the pre-breeding period, decreasing by 22% in the post-breeding period. Density was significantly higher in central plots than in peripheral plots. Mean productivity was 0.24 chicks per female and showed a high variability among plots, being significantly lower in the densest plots. The overall sex ratio was 1.35 females per male during the pre-breeding period. Productivity related positively to areas holding small fields and a high interspersion of land uses. Density and productivity were negatively affected by river density and altitude, and seasonal density variation was positively correlated with human population density.  相似文献

Between 1991 and 1997 we studied the offspring independenceand juvenile dispersal in a wild population of great bustards(Otis tarda). Young males were independent and began their juveniledispersal at an earlier age (6–11 months) than young females(8–15 months). The juvenile dispersal period was longerand the distances reached farther in males than in females.Natal dispersal distances were also longer in males, all ofwhich dispersed from their natal areas and established as adultsat 5–65 km from their natal nests. In contrast, most femaleswere strongly philopatric, settling at 0.5–5 km from theirnatal nests. These marked sex differences in offspring independenceand dispersal may have evolved originally to maintain geneticdiversity and are probably reinforced through male competitionfor mates. Young males that had fed at higher rates and receivedmore feedings from their mothers during the early maternal dependenceperiod became indepthdent and tended to disperse earlier. Theyalso integrated earlier into adult male flocks and settled earlierat their definitive leks, which were closer to their natal sites,in areas of higher adult male density. None of these correlationswas found among young females. These results suggest that enhancedfood intake and maternal care of male offspring are vitallyimportant in increasing their competitive ability during theimmature period and probably also in their fitness as breedingadults. These results are in accordance with the selective valueof large size in males and suggest how this species might havereached such a marked sexual dimorphism in size.  相似文献

Genetic variation of mitochondrial DNA (mtDNA) in 18 great tits (Parus major) from three neighboring localities in Sweden was investigated with eight tetranucleotide restriction endonucleases. The 18 individuals could be separated into 13 different maternal lineages. The high number of female lineages present in this regional population contrasts with a low level of sequence divergence between the different mtDNA clones, with a mean of 0.19% sequence divergence between all individuals. There was no obvious spatial structuring of mtDNA clones among the three localities. The presence of a high number of different clones with a low degree of sequence divergence could be explained by the effects of a large long-term effective population size, with the mtDNA clones having diverged about 25,000–200,000 years ago.This study was supported by the Swedish Natural Science Research Council, the Erik Philip-Sörensen Foundation, and the Nilsson-Ehle Foundation.  相似文献

Malus sieversii (Lebed.) M. Roem. is a wild progenitor species of the domesticated apple. It is found across a mountainous region of central Asia and has been the focus of several collection expeditions by the USDA-ARS-National Plant Germplasm System. This study used microsatellite variation at seven loci to estimate diversity and differentiation within M. sieversii using several complimentary approaches. Multilocus genotypes were amplified from 949 individuals representing seedling trees from 88 half-sib families from eight M. sieversii populations collected in Kazakhstan. Apportioning of genetic variation was estimated at both the family and site level. Analyses using a hierarchical model to estimate F st showed that differentiation among individual families is more than three times greater than differentiation among sites. In addition, average gene diversity and allelic richness varied significantly among sites. A rendering of a genetic network among all sites showed that differentiation is largely congruent with geographical location. In addition, nonhierarchical Bayesian assignment methods were used to infer genetic clusters across the collection area. We detected four genetic clusters in the data set. The quality of these assignments was evaluated over multiple Markov Chain Monte Carlo runs using both posterior likelihood and stability of the assignments. The spatial pattern of genetic assignments among the eight collection sites shows two broadly distributed and two narrowly distributed clusters. These data indicate that the southwestern collection sites are more admixed and more diverse than the northern sites.  相似文献

为了掌握云南省各地中华按蚊种群间的遗传变异和种群结构特征,测序并分析了采自云南9个样本点5个种群组的89头中华按蚊的线粒体COII基因。结果表明这些中华按蚊种群的COII基因序列平均单倍型多样性指数和核苷酸多样性指数分别为h=0.933,π=0.00406,共有51个变异位点,占分析的739个碱基总数的6.9%;定义了39个单倍型,有2个频率最高的单倍型H1和H9,分别占个体序列数的20.2%和12.4%;系统发育分析表明单倍型与地理位置没有明显的对应关系,单倍型网络图显示大部分单倍型分布没有明显的亲缘地理格局,主要以单倍型H1、H9、H4、H33和H2为中心呈星状分布,但元江和元阳构成的种群组(YU)单倍型存在明显地域分布特征;AMOVA结果表明种群组间遗传变异为12.58%,达到显著水平(P=0.04888),地理种群组间具有明显种群遗传结构。不同地区两两种群组间的Fst值和Nm值显示大部分种群组间存在基因交流,没有形成明显的遗传分化,但YU种群组和其他种群组间缺乏明显的基因交流,这主要是因为哀牢山的阻隔,使云南东西部形成两种不同的气候,产生了明显的遗传分化;歧点分布图显示为明显单峰分布,中性检测结果均为显著负值,说明云南省的中华按蚊种群在近期经历过复杂的种群扩张事件。掌握中华按蚊遗传多样性及分化特征,对中华按蚊及疟疾控制具有重要的作用。  相似文献

Vitex rotundifolia L.f. is a woody perennial and has sexual and asexual modes of reproduction. Allozyme study was conducted on 550 plants in 13 Korean populations. The levels of genetic variability and divergence within and among populations, respectively, are considerably lower and higher than the mean values for woody plants with similar life history tralts. Mean percentage of polymorphic loci (P P), mean number of alleles per locus (A P), and mean genetic diversity (He P) within populations ofV. rotundifolia were: 16.7%, 1.21, and 0.047. On average, about 79% of the total variation inV. rotundifolia was common to all populations (meanG ST=0.208). In addition, significant differences in allele frequencies among populations were found in all polymorphic loci examined (P<0.001). On the other hand, levels of genotypic diversity within and among populations were moderate. About 44% (18/41) of multilocus genotypes were “local genotypes” (genotypes occurring in only one population), whereas only one “widespread genotype” (genotypes occurring in more than 75% of the populations) were detected. The mean number of multilocus genotypes per population (G) and mean genotypic diversity index (D G) were 8.4 and 0.74, respectively. Most common multilocus genotypes found in populations were homozygous for five polymorphic loci. The abundance of ramets of these genets is responsible for the low levels of expected heterozygosity within populations. The results indicate that clonal reproduction may act as an enhancer of genetic drift by reducing effective size of local populations ofV. rotundifolia.  相似文献

Three subspecies of sandhill crane (Grus canadensis) are recognized in the Midcontinental population, the lesser (Grus c. canadensis), Canadian (G. c. rowani), and greater (G. c. tabida). Blood samples collected on the population's primary spring staging area in Nebraska, U.S.A., were used to resolve the genetic relationship among these subspecies. Phylogenetic analysis of 27 G. canadensis, by DNA sequencing of a 675 bp region of the mtDNA, supports the subspecies designations of G. c. canadensis and G. c. tabida. G. c. rowani individuals were intermediate with each of the other two subspecies. Genetic divergence ranged from 6.5 to 14.5% between G. c. canadensis and G. c. tabida, 0.5 to 6.6% within G. c. canadensis, and 0.1 to 6.0% within G. c. tabida. Sufficient DNA for analysis was obtained from shed feathers indicating a source of genetic material that does not require the capture or sacrifice of the birds. Other genetic markers and methods, including satellite telemetry, are required for obtaining detailed information on crane distributions as needed to establish effective management units for the MCP.  相似文献

Larval dispersal may have an important impact on genetic structure of benthic fishes. To examine population genetic structure of spottedtail goby Synechogobius ommaturus, samples from five different locations of China and Kunsan population in Korea were analyzed by using amplified fragment length polymorphism (AFLP) technology. A total of 253 bands were identified from 91 individuals by 5 primer combinations and the percentage of polymorphic bands was 43.87%. The average gene diversity was 0.0794 ± 0.1470 and Shannon’s information index was 0.1279 ± 0.2138. The pairwise Fst values ranged from 0.022 to 0.201. The results of AMOVA analysis indicated that 90.54% of the genetic variation contained within populations and 9.46% occurred among populations. The gene flow estimates (Nm) demonstrated that different gene flow existed among populations from 0.994 to 11.114. No significant genealogical branches or clusters were recognized on the UPGMA tree. The results support the hypothesis that larval dispersal ability can be responsible for the genetic diversity and population structuring.  相似文献

Abstract

The genus Baldellia Parl. has always been a complex taxon. Three species and three subspecies have been proposed for the Iberian Peninsula. The morphological variation of this genus in the Iberian Peninsula, Balearic islands and northern Morocco, and its thermo-pluviometric and altitudinal correlation are the focus of this contribution. Twenty-eight morphological variables were examined and subjected to a multivariate statistical analysis. The three morphotypes observed (Form 1, which includes B. ranunculoides subsp. repens and B. alpestris; Form 2, which includes B. ranunculoides and Form 3 – for the new combination B. ranunculoides subsp. ranunculoides var. tangerina (Pau) J. Rocha, A. Crespí, M. García-Barriuso, R. Almeida, J. Honrado, comb. nova proposed here for the first time) seem to represent different reproductive strategies. Morphological variables related with the presence or absence of stolons and the architecture of the inflorescence; the size and number of fruits are the most discriminating variables. Form 1 represents the asexual morphotype; Forms 2 and 3 correspond to morphotypes for which sexual reproduction is preferential. The environmental approach revealed that the asexual form (Form 1) grows in temperate, in more humid conditions, and within a broad altitudinal range. In contrast, the two sexual forms are more common in warmer and drier conditions, and occur over a narrower interval of altitudes.  相似文献

The houbara bustard (Chlamydotisundulata), the main quarry for Arab falconry,is currently threatened by excessive huntingand poaching as well as by habitat loss andfragmentation. We have investigated the geneticdiversity, population structure and demographichistory of houbara bustards across theirgeographical range by analyzing mitochondrial(mt)DNA sequences (a 370 bp fragment of controlregion I and a 264 bp fragment of thecytochrome b gene) and 4 microsatelliteloci in 74 individuals sampled from the CanaryIslands to China. Both markers revealed low tomoderate diversity that could be partitionedinto two monophyletic groups or evolutionarysignificant units belonging to the NorthAfrican (C. u. undulata) and Asian (C. u. macqueenii) subspecies. A history ofrelatively recent population growth (35,000years ago) accompanied by range expansion isthe most likely demographic scenario for theAsian subspecies. In addition, Macqueen'sbustards are able to disperse efficiently overbroad areas, which is consistent with ourinference of weak phylogeographic structure(global F ST = 0.20 and 0.04 formtDNA and microsatellites, respectively) andhigh levels of homogenizing gene flow on widegeographical scales. We therefore suggest thatmanagement actions should focus on maintainingmigratory connectivity between breeding andnon-breeding areas.  相似文献

This article presents the distribution and status of bustards, which are listed as first-category protected animals according to the survey results during 1990–2002 in China. The Chinese populations of Otis tarda dybowskii are breeding in south-west of Heilongjiang Province, western Jilin Province, east and middle Inner Mongolia, north Ningxia Hui Autonomous Region, and Gansu Province. A few can winter in the south breeding-range. Its winter-range lies from the south to the Yellow River, as far as to Guizhou Province and Jiangxi Province. Its population number is about 200–300 or 500–800. The Chinese populations of O. t. tarda are breeding in the north and west of Xinjiang. It is unclear about its winter-range, which is presumed to be in south Asia. Recently we found individuals wintering in Chabuchaer and west Xinjiang. The population number is about 2000–3000. The habitat in breeding range includes steppe, grassland, desert grassland, and farmland. The habitat in winter range is the beach of rivers and lakes, meadows, meadow-grassland, and wheatland. The Chinese populations of Chlamydotis undulata macqueeni are breeding in the fringe of the Jungar Basin, the banks of the Ulungur River, Balikun and south Turpan Basin in Xinjiang, west Inner Mongolia, and west Gansu. NortheastMulei in eastern Jungar Basin of Xinjiang is the main breeding-range in the world. The bird uses desert and desert grassland as its habitat. Its winter-range is west Asia and south Asia. Its population number is about 2000. The Chinese populations of Tetrax tetrax are breeding in north Xinjiang, and China is located on the east border of its breeding-range. Its habitat is grassland and semi-desert, and its winter-range lies in south Asia. Its population in China is very scarce.
