Feeding experiments on tillers of a non-flowering red cloversegregate indicated that the promotive effect of gibberellinupon stem extension and flowering occured only under inductivelong days (LD). Furthermore, floral initiation took place inLD only when free gibberellin exceeded an unspecified criticallevel. Various intermediates in fungal gibberellin synthesis were fedin a similar fashion but none exerted any effect upon reproductivegrowth. Studies using 3H-gibberellin A3 (GA3) showed that innormal (flowering) material GA3 was more rapidly metabolizedin short days than in long days. Qualitative differences ingibberellin metabolism between normal and non-flowering genotypeswere revealed by radio-isotope studies. Non-flowering materialrapidly degraded GA3 under LD conditions to a biologically inactivecompound chromatographically indentical with allo-gibberic acid,whereas normal plants metabolized GA3 more slowly producinga compund similar to gibberellenic acid. The implications ofthese results are discussed in terms of the mechanism of stemelongation and floral initiation.  相似文献

A mutant red clover is used to investigate changes in the carbohydratestatus of leaves and apical zones of red clover (Trifolium pratenseL.) during the transition to flowering. Mutant plants will notflower during normally inductive photoperiods unless suppliedwith exogenous gibberellin. Changes in carbohydrates are describedin normal-flowering plants following transfer to inductive long-daysas well as in mutant plants with and without gibberellin. Theeffects of gibberellin on mutants under short-days—whenstem elongation occurs but flowering does not ensue—arealso investigated. Carbohydrate concentrations are virtuallyidentical in the leaves of normal and mutant plants under short-daysalthough significant differences exist at the apex. Transferfrom short to long-days results in major alterations in thecarbohydrate status, but changes specifically associated withthe onset of flowering were not apparent. Relationships between gibberellins and floral initiation were investigated in a conditional non-flowering mutant of red clover, Trifolium pratense. Untreated mutant plants will not flower under long-days, but will do so when certain GAs are applied. Gibberellins, A3, A1, A7, and A5 all resulted in both stem elongation and flowering whilst GA4 produced the elongation only. Applications of GA20, GA8 and GA13 under long-days had no detectable effect. Thus, by combining the use of the mutant with the application of different GAs, the correlation between the processes of stem elongation and floral initiation, which is normally strongly expressed in this species, was broken. Endogenous gibberellins shown to be present in normal plants were also found in the mutant genotype. Gibberellins alone were not sufficient to initiate floral development in the mutant, there being an essential element of interaction with long-days. These results are discussed in relation to the nature of the lesion in the mutant and the signal provided by the applied gibberellin.  相似文献

The veg1 ( vegetative ) mutant in pea ( Pisum sativum L.) does not flower under any circumstances and gi ( gigas ) mutants remain vegetative under certain conditions. gi plants are deficient in production of floral stimulus, whereas veg1 plants lack a response to floral stimulus. During long days in particular, these non-flowering mutant plants eventually enter a stable compact phase characterised by a large reduction in internode length, small leaves and growth of lateral shoots from the upper-stem (aerial) nodes. The first-order laterals in turn produce second-order laterals and so on in a reiterative pattern. The apical bud is reduced in size but continues active growth. Endogenous hormone measurements and gibberellin application studies with gi-1 , gi-2 and veg1 plants indicate that a reduction in gibberellin and perhaps indole-3-acetic acid level may account, at least partially, for the compact aerial shoot phenotype. In the gi-1 mutant, the compact phenotype is rescued by transfer from a 24- to an 8-h photoperiod. We propose that in plants where flowering is prevented by a lack of floral stimulus or an inability to respond, the large reduction in photoperiod gene activity during long days may lead to a reduction in apical sink strength that is manifest in an altered hormone profile and weak apical dominance.  相似文献

The use of a thermosensitive genic male sterility (TGMS) system in two-line hybrid rice breeding is affected greatly by the sterility instability of TGMS lines caused by temperature fluctuation beyond their critical temperatures for fertility reversion. To prevent seed production from self contamination, we have developed a system to secure seed purity using a herbicide-sensitive TGMS mutant, M8077S, obtained by radiation. Genetic analysis, using the F1, F2 and F3 populations derived from this mutant and other normal varieties, revealed that bentazon lethality/sensitivity was controlled by a single recessive gene, which was named bel. The mutant can be killed at the seedling stage by bentazon at 300 mg/l or higher, a dosage that is safe for its F1 hybrids and all other normal varieties. This mutant is also sensitive to all the tested sulfonylurea herbicides. Response of segregating plants to these two types of herbicide indicated that sulfonylurea sensitivity was also controlled by bel. By crossing this mutant with Pei-Ai 64S, an F2 population was developed for genetic mapping. Surveying the two DNA pools from sensitive and non-sensitive F2 plants identified four markers that were polymorphic between the pools. The putative linked markers were then confirmed with the F2 population. The bel locus was located on chromosome 3, 7.1 cM from the closest microsatellite marker RM168. Phenotypic analysis indicated that the bel gene had no negative effect on agronomic traits in either a homozygous or heterozygous status. The mutant M8077S is valuable in the development of a TGMS breeding system for preventing impurity resulting from temperature fluctuation of the TGMS. Dwarfing and sensitivity to the duration of a single inductive dark period for flowering ofPharbitis nil in F2 progeny of a cross between the tall strain Tendan, and the dwarf, Kidachi appear to be controlled by the alleles at two independent loci. Progeny of a similar cross between the tall strain Violet and the dwarf Kidachi at F2 and F3 also showed single locus segregation for tall: dwarf plants. In this cross, differences in photoperiodic response could be identified in F3 families but they were not simply inherited. There was some evidence of difficulties with classification of the F2 plants, but also, the flowering of the F1 between the two less sensitive strains Tendan and Violet indicated complex inheritance of their photoperiodic response. Complementary dominant alleles at three independent loci may be necessary for flowering in even shorter dark periods with the sensitive strain Kidachi. The dwarf strain Kidachi has a reduced gibberellin (GA) content (Barendse and Lang 1972), it flowers in a short dark period without terminal flowering, and it responds positively to GA application both for flowering and growth. However, since control of dwarfing and photoperiodic sensitivity can be separated genetically, there is no strick link between the gibberellin responsiveness of Kidachi for its growth and flowering. Despite the complexity of flowering genetics in Violet×Kidachi, a short-dark-period-sensitive, terminal flowering and tall F7 line was obtained in a pedigree previously held heterozygous for the dwarf: tall character but not selected for flowering time. Thus, flowering in a short dark period can also be obtained in the presence of the non-dwarfing allele from strain Violet, again demonstrating genetic independence.  相似文献

A model is presented as an explanation for an anomaly observed in germination and establishment and isozyme segregation patterns in Allium cepa x A. fistulosum F2BC3 populations generated in an introgression-breeding program. The F1BC3 parent of these populations was selected for its heterozygous PGI phenotype, Pgi-1(2/3); Pgi-1(2) was inherited from an A. cepa (Ac) seed parent and Pgi-1(3) from an A. fistulosum (Af) pollen parent. Germination and establishment was recorded for the F2BC3 progeny population. Segregation of Ac and Af Pgi-1 alleles was investigated in F2BC3 seeds and embryo and endosperm tissue was isolated and tested for isozyme expression. A pooled goodness-of-fit test of the segregation of Pgi-1 alleles in the populations to the expected Mendelian 1:2:1 ratio using the chi-square statistic gave a chi2 = 185.9, well beyond the accepted limits at 2 degrees of freedom. The 1:2:1 ratio expected for simple Mendelian inheritance was rejected, while a pooled chi-square goodness-of-fit test of the segregation of Pgi-1 alleles in the populations fit a 1:1 ratio with a chi2 = 0.203, based on the incongruity model. We present here the central cell nuclear-cytoplasmic incongruity hypothesis to explain the observed anomalies.  相似文献

During vegetative growth in controlled environments, the patternof distribution of 14C-labelled assimilates to shoot and root,and to the meristems of the shoot, was measured in red and whiteclover plants either wholly dependent on N2 fixation in rootnodules or receiving abundant nitrate nitrogen but lacking nodules. In experiments where single leaves on the primary shoot wereexposed to 14CO2, nodulated plants of both clovers generallyexported more of their labelled assimilates to root (+nodules),than equivalent plants utilizing nitrate nitrogen, and thiswas offset by reduced export to branches (red clover) or stolons(white clover). The intensity of these effects varied with experiment.The export of labelled assimilate to growing leaves at the terminalmeristem of the donor shoot was not influenced by source ofnitrogen. Internode elongation in the donor shoot utilized nolabelled assimilate. Whole plants of white clover exposed to 14CO2 on seven occasionsover 32 days exhibited the same effect on export to root (+nodules),which increased slightly in intensity with increasing plantage. Nodulated plants had larger root: shoot ratios than theirequivalents utilizing nitrate nitrogen. Exogenous gibberellin A3(GA3) reduced the number of leaf nodesat flowering and time to flowering and increased the stem heightat flowering in three genotypes of spring rape (Brassica napusvar.annua L.). The responses to GA3were similar to those forlong days (LD) and low-temperature treatments, suggesting thatthe effect of photoperiod and the vernalization response areprobably mediated through gibberellins. The response to exogenousGA3was greatest in non-cold-treated plants in short days (SD)suggesting that endogenous GAs are limiting in these conditions.CCC, an inhibitor of gibberellin biosynthesis, caused a smallincrease in the number of leaf nodes at flowering and time toflowering and a small decrease in the stem height at flowering,but unexpectedly, its effect was hardly influenced by the applicationof exogenous GA3. Genotypes that showed the clearest responsesto the treatments with regard to the number of leaf nodes atflowering and time to flowering did not show the clearest responseswith regard to the stem height at flowering; the pattern ofresponses of the number of leaf nodes at flowering and timeto flowering was distinct from that of stem height at flowering.This indicates that flower formation and stem elongation areseparable developmental processes which may be controlled bydifferent endogenous gibberellins, different levels of a specificendogenous gibberellin, or different responses to gibberellin.Copyright 1999 Annals of Botany Company Brassica napus var. annua, gibberellin, photoperiod, spring rape, vernalization.  相似文献

The aim of this study was to test the effect of oxygen partialpressure as a possible limiting factor of nitrogen fixationfollowing defoliation. The response of nitrogenase activity(C2H2-reduction) of defoliated and undefoliated white and redclover plants (Trifolium repens L. and Trifolium pratense L.)to either 19 kPa oxygen or 55 kPa oxygen was investigated. Priorto defoliation, white clover plants were grown for five weeksin a growth chamber, and red clover plants for 7 or 11 weeksin a glasshouse. The results included measurements of 16N2-uptake. Increasing oxygen partial pressure from 19 to 55 kPa severelyrestricted nitrogenase activity of undefoliated white cloverplants; however, 2 h after complete defoliation, the same treatmentcaused a significant increase. A fivefold increase in nitrogenaseactivity upon exposure to the elevated oxygen partial pressurewas found at the end of a 24 h period. This beneficial effectdecreased gradually from 1 to 5 d after defoliation. The responseof recently defoliated red clover plants to 55 kPa oxygen partialpressure was similar to that of white clover, independentlyof plant age. The gradual recovery of nitrogenase activity duringthree weeks of regrowth was associated with a simultaneous changein the response to increased oxygen partial pressure, leadingagain to the response of undefoliated plants. These data suggested that lack of oxygen at the site of nitrogenfixation, resulting from a dramatic increase in oxygen-diffusionresistance, is the main factor limiting nitrogenase activityfollowing defoliation. Summary Co-segregation studies of isozyme markers and male fertility restoration showed that a restorer gene from radish was introduced into rapeseed along with an isozyme marker (Pgi-2). The radish chromosome segment carrying these genes was introgressed into rapeseed through homoeologous recombination, substituting for some of the rapeseed alleles. By crossing heterozygous restored plants to male-sterile lines and to maintainers, tight linkage was found between the restorer gene and the marker. The recombination fraction was estimated at 0.25 ± 0.02%. Although few restored plants lacked the radish isozyme marker, it was still possible to distinguish male-fertile from male-sterile plants by their PGI-2 patterns. Furthermore, homozygous and heterozygous restored plants could be separated by specific PGI-2 phenotypes. Thus, the Pgi-2 marker is now currently used in restorer breeding programs.  相似文献

摘要: 以海岛棉海1为供体亲本, 中221、中棉所36(中36)两个陆地棉品种(系)为轮回亲本配制了两套杂交回交的组合: 中36×海1组合和中221×海1组合。通过对这2套组合的不同世代群体杂交铃、自交铃纤维品质性状的分析, 结果: 以分离群体BC1F1为父本, 轮回亲本中221、中36分别为母本时, 母体植株上的杂交铃(BC2F0)的纤维品质性状与父本BC1F1群体植株上自交铃的纤维品质性状之间在平均值、极差和变异系数上存在显著差异, 相关性不显著, 而与母本(轮回亲本)值接近; 又以分离群体BC2F1为母本, 轮回亲本中221、中36分别为父本, 母体植株上的杂交铃(BC3F0)纤维品质与母本BC2F1群体植株上自交铃的纤维品质性状在平均值、极差和变异系数上接近, 除整齐度以外, 纤维长度、比强度、麦克隆值和伸长率都达到了极显著的正相关。这些结果表明, 杂交铃的纤维品质偏向母体植株上的自交铃的纤维品质, 父本花粉的基因型对杂交当代种子上的纤维品质没有显著的影响, 杂交铃种子上的纤维品质主要由当代种子的表皮细胞基因型即母体植株基因型决定, 而当代种子胚的基因型作用不明显。  相似文献

Activity of gibberellin-like substances was lacking in leavesof strawberry plants at time of flower initiation, but peduncleelongation was accompanied by significant activity of two substancesprobably related to GA3 and GA7. Exogenous gibberellin (GAS),applied to strawberry plants at the flower-differentiation stage,strongly promoted peduncle elongation, while similar applicationof the growth-retardant (2 chloro-ethyl) trimethylammonium chloride(CCC), an inhibitor of gibberellin biosynthesis, inhibited peduncleelongation. These treatments also affected subsequent runnering,GA increasing the number of runners, as well as their mean individuallength, while CCC caused the opposite effect. Application of gibberellin immediately after flower-bud initiation,but before peduncle emergence, affected the distribution offruit yield. GA-treated plants produced more than untreatedones in the first half of the fruiting season, but the situationwas reversed in the second half of the season—so thattotal yields were unaffected. It was suggested that the exogenousgibberellin hastened yield by anticipating the endogenous gibberellinwhich is normally formed at a somewhat later stage in the plant'sdevelopment. The parallel responses with regard to elongationof vegetative runners and flower peduncles together with thedata of the other experiments seem to indicate that peduncleelongation is a typical vegetative growth response enhancedby gibberellin and checked by substances which inhibit biosynthesisof endogenous gibberellin.  相似文献

With the aim of establishing the basic knowledge and resourcesneeded for applied genetics, we investigated the genome structureof red clover Trifolium pratense L. by a combination of cytological,genomic and genetic approaches. The deduced genome size was440 Mb, as estimated by measuring the nuclear DNA content byflow cytometry. Seven chromosomes could be distinguished bymicroscopic observation of DAPI stained prometaphase chromosomesand fluorescence in situ hybridization using 28S and 5S rDNAprobes and bacterial artificial chromosome probes containingmicrosatellite markers with known positions on a genetic linkagemap. The average GC content of the genomes of chloroplast, mitochondrionand nucleus were shown to be 33.8, 42.9 and 34.2%, respectively,by the analysis of 1.4 Mb of random genomic sequences. A totalof 26 356 expressed sequence tags (ESTs) that were grouped into9339 non-redundant sequences were collected, and 78% of theESTs showed sequence similarity to registered genes, mainlyof Arabidopsis thaliana and rice. To facilitate basic and appliedgenetics in red clover, we generated a high-density geneticlinkage map with gene-associated microsatellite markers. A totalof 7159 primer pairs were designed to amplify simple sequencerepeats (SSRs) identified in four different types of libraries.Based on sequence similarity, 82% of the SSRs were likely tobe associated with genes. Polymorphism was examined using twoparent plants, HR and R130, and 10 F1 progeny by agarose gelelectrophoresis, followed by genotyping for the primer pairsshowing polymorphisms using 188 F1 plants from the mapping population.The selected 1305 microsatellite markers as well as the previouslydeveloped 167 restriction fragment length polymorphism markerswere subjected to linkage analysis. A total of 1434 loci detectedby 1399 markers were successfully mapped onto seven linkagegroups totaling 868.7 cM in length; 405 loci (28%) were bi-parental,611 (43%) were specific to HR and 418 (29%) were specific toR130. Each genetic linkage group was linked to a correspondingchromosome by FISH analysis using seven microsatellite markersspecific to each of the linkage groups as probes. Transferabilityof the developed microsatellite markers to other germplasmswas confirmed by testing 268 selected markers on 88 red clovergermplasms. Macrosynteny at the segmental level was observedbetween the genomes of red clover and two model legumes, Lotusjaponicus and Medicago truncatula, strongly suggesting thatthe genome information for the model legumes is transferableto red clover for genetic investigations and experimental breeding.  相似文献

Dwarf cultivar Progress No. 9 and normal cultivar Alaska ofPisum sativum L. were grown under conditions of darkness orred light. Red light decreased the stem elongation rate of bothcultivars. Gibberellins present during the linear phase of stemelongation were isolated and the two main components were tentativelyidentified by gas chromato-chromatography and mass spectrometryas Gibberellin A1 and A5. Both gibberellins varied quantitativelywithin and between cultivar and treatment groups, and the amountspresent were inversely related to stem elongation rate. Althoughthe stem elongation rate of plants grown under red light wasrepressed, endogenous gibberellin was not limiting and was asmuch as two-fold higher in red light-grown plants than in dark-grownplants. Signals produced in leaves are transported to the shoot apex where they cause flowering. Protein of the gene FLOWERING LOCUS T (FT) is probably a long day (LD) signal in Arabidopsis. In the companion paper, rapid LD increases in FT expression associated with flowering driven photosynthetically in red light were documented. In a far red (FR)-rich LD, along with FT there was a potential role for gibberellin (GA). Here, with the GA biosynthesis dwarf mutant ga1-3, GA(4)-treated plants flowered after 26 d in short days (SD) but untreated plants were still vegetative after 6 months. Not only was FT expression low in SD but applied GA bypassed some of the block to flowering in ft-1. On transfer to LD, ga1-3 only flowered when treated simultaneously with GA, and FT expression increased rapidly (<19.5 h) and dramatically (15-fold). In contrast, in the wild type in LD there was little requirement for GA for FT increase and flowering so its endogenous GA content was near to saturating. Despite this permissive role for endogenous GA in Columbia, RNA interference (RNAi) silencing of the GA biosynthesis gene, GA 20-OXIDASE2, revealed an additional, direct role for GA in LD. Flowering took twice as long after silencing the LD-regulated gene, GA 20-OXIDASE2. Such independent LD input by FT and GA reflects their non-sympatric expression (FT in the leaf blade and GA 20-OXIDASE2 in the petiole). Overall, FT acts as the main LD floral signal in Columbia and GA acts on flowering both via and independently of FT.  相似文献

以孝顺竹为材料,利用HPLC和MSAP技术在其未开花至开花的生长过程中进行甲基化水平的检测,分析竹子开花过程中基因组DNA甲基化的动态,以揭示DNA甲基化水平与竹子开花现象的相关性。结果显示:(1)孝顺竹基因组DNA甲基化率在不同的时间处于动态变化中,进入开花状态的孝顺竹植株其甲基化水平极显著低于开花竹丛中未开花植株和未开花竹丛;开花和未开花植株的总甲基化率分别为9.00%和12.42%,全甲基化率分别为5.06%和7.53%。(2)MSAP位点中有66.83%的位点在开花和未开花材料中甲基化状态保持一致,33.17%的位点在开花和未开花植株中发生甲基化变化:其中22.28%的位点在开花植株中发生完全的去甲基化,1.98%的谱带在开花植株中发生甲基化,8.91%的位点在开花和未开花材料中甲基化水平呈现上升或降低的趋势。研究表明,开花的孝顺竹同时发生甲基化和去甲基化,但发生去甲基化的概率明显大于发生甲基化的概率,最终导致其甲基化水平极显著降低。  相似文献

AbA, ethephon and gibberellin were applied to cucumber plantsof monoecious, gynoecious, andromoneocious and hermaphroditeinbred lines, as well as to F1 (gynoecious?monoecious) plants.Exogenous AbA enhanced the male tendency in monoecious cucumberplants and the female tendency in gynoecious plants, irrespectiveof light regime. Exogenous ethephon treatments increased thefemale tendency in monoecious plants, and decreased it in gynoeciousones. These effects were influenced by day length. ExogenousAbA counteracted the effect of gibberellin (A4+7) treatmentin gynoecious plants, but had no such effect in monoecious ones. In addition to its differential effect on sexual differentiation,AbA stimulated flower development in gynoecious plants and inhibitedit in monoecious plants. These responses to AbA are discussedin the light of previously reported effects of plant growthregulators on various sex types of cucumber. Abstract. The influence of leaf age, total leaf area and its dispersion in space on canopy photosynthesis were studied using microswards of red clover ( Trifolium pratense L.) which were established in the greenhouse. Two varieties, Renova (flowering) and Molstad (non-flowering), were sown in separate plastic boxes at densities of 225, 400 and 625 plants per m2.
Vertical distribution of photosynthetically active radiation (PAR), leaf area, leaf age and 14CO2-fixation were determined periodically. Net photosynthesis and dark respiration of canopies were measured. Maximum photosynthetic capacity of individual leaves was measured on plants taken from the intact canopy or from plants where shading of the growing leaves had been prevented.
Net photosynthetic rate of canopies increased linearly with leaf area index (LAI) up to an LAI of 3.5 and then declined at higher LAI, independent of variety and sowing density. Below the optimum LAI, net photosynthesis depended mainly on interception of PAR. Decrease in canopy photosynthesis above the optimum LAI was due to a higher proportion of old leaves with decreased photosynthetic capacity, and not to an increase in respiring plant parts. It is concluded that LAI and position of leaf age categories in the canopy are more important than vertical distribution of leaf area in determining canopy photosynthesis of red clover.  相似文献
